Invasive pulmonary aspergillosis (IPA) is a fatal fungal infection with a high mortality rate. Voriconazole (VCZ) is considered a first-line therapy for IPA and shows efficacy in patients for whom other antifungal treatments have been unsuccessful. The objective of this study was to develop a high-potency VCZ-loaded liposomal system in the form of a dry-powder inhaler (DPI) using the spray-drying technique to convert liposomes into a nanocomposite microparticle (NCMP) DPI, formulated using a thin-film hydration technique. The physicochemical properties, including size, morphology, entrapment efficiency, and loading efficiency, of the formulated liposomes were evaluated. The NCMPs were then examined to determine their drug content, production yield, and aerodynamic size. The L3NCMP was formulated using a 1:1 lipid/L-leucine ratio and was selected for in vitro studies of cell viability, antifungal activity, and stability. The kinetics of Os(VIII) accelerated L-Leucine (L-Leu) oxidation in cetyltrimethylammonium bromide (CTAB) by hexacyanoferrate(III) [HCF(III)] was investigated by registering the decline in absorbance at 431 nm. Employing the pseudo-first-order condition, the reaction's advancement has been examined as an indicator of [CTAB], [Os(VIII)], [OH−], [L-Leu], [HCF(III)], temperature, and ionic strength. The results show that [OH−], [CTAB], and [L-Leu] are the critical parameters with a discernible influence on reaction rate. L-Leu interacts with HCF(III) in a 1:2 ratio. The rate constant is independent of the [HCF(III)] and is merely used up to regenerate the Os(VIII) during the reaction. In the investigated concentration ranges of Os(VIII), [OH−], and [L-Leu], the reaction demonstrates first-order kinetics, but follows less than unit order at higher concentrations of L-Leu (more than 6.0 × 10−3 mole dm−3) and alkali (more than 0.4 mole dm−3). A positive salt effect is indicated by the linear rise in reaction rate with additional electrolytes. CTAB catalyzes the process substantially, and once at its peak at 7.0 × 10−4 mole dm−3, the rate remains constant as [CTAB] grows up to 8.5 × 10−4 mole dm−3. With anionic surfactant sodium dodecyl sulfate, the reaction rate was significantly reduced even in the presence of Os(VIII). Reduced repulsion between surfactant molecule's positive charge heads brought on by the negative-charged HCF(III), OH−, and [OsO5(OH)]3− molecules may be responsible for the witnessed drop in CTAB critical micellar concentration (CMC). Dry-powder inhalers (DPIs) are valued for their stability but formulating them is challenging due to powder aggregation and limited flowability, which affects drug delivery and uniformity. In this study, the incorporation of L-leucine (LEU) into hot-melt extrusion (HME) was proposed to enhance dispersibility while simultaneously maintaining the high aerodynamic performance of inhalable microparticles. This study explored using LEU in HME to improve dispersibility and maintain the high aerodynamic performance of inhalable microparticles. Formulations with crystalline itraconazole (ITZ) and LEU were made via co-jet milling and HME followed by jet milling. The LEU ratio varied, comparing solubility, homogenization, and aerodynamic performance enhancements. In HME, ITZ solubility increased, and crystallinity decreased. Higher LEU ratios in HME formulations reduced the contact angle, enhancing mass median aerodynamic diameter (MMAD) size and aerodynamic performance synergistically. Achieving a maximum extra fine particle fraction of 33.68 ± 1.31% enabled stable deep lung delivery. The proposed study aimed to explore the kinetics of L-leucine (Leu) oxidation in a micellar media of cetyltrimethylammonium bromide (CTAB) by hexacyanoferrate(III) {HCF(III)}. The reduction in absorbance at 420 nm, which is indicative of the concentration of HCF(III), was measured to determine the reaction rate. Applying the pseudo-first-order state, The reaction's progression was analyzed as an indicator of [HCF(III)], temperature, [Leu], [Cu(II)], [CTAB], ionic strength, and [OH–]. The findings indicate that the [CTAB] is the crucial factor that significantly affects the rate of the reaction. The HCF(III) undergoes a 2 : 1 stoichiometric interaction with Leu and has a first-order reliance on [HCF(III)]. The observed reaction exhibits fractional-first-order kinetics with regards to [Leu], [OH–], and Cu(II), within the range of concentrations investigated. The observed linear augmentation in reaction rate upon electrolyte's introduction is suggestive of a positive salt effect. The inclusion of CTAB substantially enhances the catalytic activity of the process. After reaching its maximum rate, the reaction exhibits a very steady behavior even when the [CTAB] is further increased. The observed decrease in CTAB CMC could potentially be attributed to the diminished electrostatic repulsion among the cationic surfactant head groups, which is caused by the anionic [Cu(OH)4]2–, OH–, and HCF(III) species. This study was conducted to compare the effects of rumen-protected (RP-Leu) and unprotected L-leucine (RU-Leu) on the fermentation parameters, bacterial composition, and amino acid metabolism in vitro rumen batch incubation. The 5.00  g RP-Leu or RU-Leu products were incubated in situ in the rumen of four beef cattle (Bos taurus) and removed after 0, 2, 4, 6, 12, 16, and 24  h to determine the rumen protection rate. In in vitro incubation, both RP-Leu and RU-Leu were supplemented 1.5  mmol/bottle (L-leucine HCl), and incubated after 0, 2, 4, 6, 8, 12, and 16  h to measure gas production (GP), nutrient degradability, fermentation parameters, bacterial composition, and amino acids metabolism. Results from both in vitro and in situ experiments confirmed that the rumen protection rate was greater (p  <  0.01) in RP-Leu than in RU-Leu, whereas the latter was slow (p  <  0.05) degraded within incubation 8  h. Free leucine from RP-Leu and RU-Leu reached a peak at incubation 6  h (p  <  0.01). RU-Leu supplementation increased (p  <  0.05) gas production, microbial crude protein, branched-chain AAs, propionate and branched-chain VFAs concentrations, and Shannon and Sobs index in comparison to the control and RP-Leu supplementation. RU-Leu and RP-Leu supplementation decreased (p  <  0.05) the relative abundance of Bacteroidota, which Firmicutes increased (p  <  0.05). Correlation analysis indicated that there are 5 bacteria at the genus level that may be positively correlated with MCP and propionate (p  <  0.05). Based on the result, we found that RP-Leu was more stable than RU-Leu in rumen fluid, but RU-Leu also does not exhibit rapid degradation by ruminal microbes for a short time. The RU-Leu was more beneficial in terms of regulating rumen fermentation pattern, microbial crude protein synthesis, and branched-chain VFAs production than RP-Leu in vitro rumen conditions. The extract of mulberry leaf and its active ingredients have already been reported to have anti-diabetic effects; however, further studies are required to obtain better quality extracts and higher yields of active ingredients. Reducose® is a commercially available aqueous extract of mulberry leaves with a high content of active ingredients. In this study, the biological activities of Reducose®, 1-deoxynojirimycin, and l-leucine were evaluated using a glucose-stimulated insulin secretion (GSIS) assay. The GSIS assay results were expressed as the glucose-stimulated index (GSI). Considering the pharmacological safety in pancreatic β-cells, the appropriate non-toxic concentrations were selected by screening for cytotoxicity of Reducose®, 1-deoxynojirimycin, and l-leucine before the GSIS assay. The effect of Reducose®, 1-deoxynojirimycin, and l-leucine on glucose-stimulated insulin secretion in INS-1 cells was compared. Reducose®, 1-deoxynojirimycin, and l-leucine increased the GSI values more effectively than gliclazide (positive control). This was associated with an increase in protein expression, such as peroxisome proliferator-activated receptor-γ, insulin receptor substrate-2, activated pancreatic and duodenal homeobox-1, which are related to the regulation of pancreatic β-cell function and survival. In order to elucidate the effect of Reducose® in anti-diabetic effects, blood glucose levels, insulin levels, and liver and lipid concentrations were investigated in a Sprague-Dawley rat model of high-fat diet/streptozotocin-induced diabetes. We observed that administration of Reducose® can decrease fasting blood glucose levels and reduce the production of AST, ALT, TG, and TC to a similar extent as metformin (positive control). These results suggested that Reducose® play a role in promoting GSIS but not enough to show that the content and proportion of 1-deoxynojirimycin and l-leucine play an important role in the GSIS activity of Reducose®.

Abstract Background l-Leucine is a high-value amino acid with promising applications in the medicine and feed industries. However, the complex metabolic network and intracellular redox imbalance in fermentative microbes limit their efficient biosynthesis of l-leucine. Results In this study, we applied rational metabolic engineering and a dynamic regulation strategy to construct a plasmid-free, non-auxotrophic Escherichia coli strain that overproduces l-leucine. First, the l-leucine biosynthesis pathway was strengthened through multi-step rational metabolic engineering. Then, a cooperative cofactor utilization strategy was designed to ensure redox balance for l-leucine production. Finally, to further improve the l-leucine yield, a toggle switch for dynamically controlling sucAB expression was applied to accurately regulate the tricarboxylic acid cycle and the carbon flux toward l-leucine biosynthesis. Strain LEU27 produced up to 55 g/L of l-leucine, with a yield of 0.23 g/g glucose. Conclusions The combination of strategies can be applied to the development of microbial platforms that produce l-leucine and its derivatives.

The human body is a remarkably aggressive medium and applied materials must be extremely resistant to corrosion and destruction, so copper, which is a biological material and is widely used in medicine, is being considered. Copper corrosion may be associated with inflammation in body because it releases ions that are toxic to humans. In aggressive solutions, biomolecules called amino acids act as corrosion inhibitors. In the present research, the purpose is to study the inhibitory action of l-alanine (L-Ala) and l-Leucine (L-Leu) amino acids on the corrosion of Cu. By density functional theory (DFT), inhibition behavior of l-Ala-and l-Leu-and their conformers against Cu corrosion have been investigated. According to values of back-donation of electrons (Eback-d), fraction of electron transferred (ΔN), electrophilicity (ω), electronegativity (χ), softness (σ), hardness (η), energy gap (Eg), ELUMO, and EHOMO computed reactivity factors between copper surface and inhibitors are studied. l-Leu's theoretical indicators confirm its tendency towards adsorption. l-Leu-activity against corrosion has been investigated and favorable results have been obtained.

The enzyme α-isopropylmalate synthase (α-IPMS) catalyzes the first step of L-leucine biosynthesis and is regulated via feedback inhibition by L-leucine. The gene of α-IPMS variant from strain ARTP-L04 was cloned and sequenced. Interestingly, amino acid mutations of Gly92Asp, Ile162Val, Arg494His and Gly526Asp occurred in the α-IPMS variant. The enzyme of α-IPMS variant was characterized, and exhibited higher resistance to feedback inhibition by L-leucine. In the presence of 20 mM L-leucine, the activity of the α-IPMS variant was still retained at over 50%. The α-IPMS variant almost removed feedback inhibition by L-leucine, while there was no significant difference in specific activities of the α-IPMS variant and wild-type α-IPMS. For the α-IPMS variant, the capacity to overproduce L-leucine was evaluated by recombinant Corynebacterium glutamicum strains. Interestingly, expression of the α-IPMS variant could significantly enhance L-leucine production, especially co-expressed with acetohydroxyacid synthase (accumulating 7.79 g/L L-leucine). Collectively, our findings provided valuable insights into further development in genetic engineering for L-leucine production.

Supplements and diets containing L-leucine, a branched-chain amino acid, have been considered beneficial for controlling oxidative stress and maintaining cardiac tissue in toxicity models using doxorubicin, a drug widely used in cancer treatment. However, there is a lack of studies in the literature that assess the effects of this diet on other organs and tissues, such as the liver and kidneys. Therefore, this study aimed to evaluate the effects of a leucine-rich diet on the liver and kidneys of healthy rats submitted to the doxorubicin toxicity model by analyzing biomarkers of oxidative stress and histological parameters. The animals were divided into four groups: naive, doxorubicin, L-leucine, and doxorubicin + L-leucine, and the diet was standardized with 5% L-leucine and a dose of 7.5 mg/kg of doxorubicin. We evaluated tissue injury parameters and biomarkers of oxidative stress, including enzymes, antioxidant profile, and oxidized molecules, in the liver and kidneys. Although some studies have indicated benefits of a diet rich in L-leucine for the muscle tissue of animals that received doxorubicin, our results showed that the liver was the most affected organ by the L-leucine-rich diet since the diet reduced its antioxidant defenses and increased the deposit of collagen and fat in the hepatic tissue. In the kidneys, the main alteration was the reduction in the number of glomeruli. These results contribute to the scientific literature and encourage further studies to evaluate the effects of an L-leucine-rich diet or its supplementation, alone or combined with doxorubicin using an animal model of cancer. Therefore, our study concludes that the leucine-rich diet itself was harmful and, when co-administered with doxorubicin, was not able to maintain the antioxidant defenses and tissue structure of the evaluated organs. L-leucine is an essential amino acid widely used in food and pharmaceutical industries. However, the relatively low production efficiency limits its large-scale application. In this study, we rationally developed an efficient L-leucine-producing Escherichia coli strain. Initially, the L-leucine synthesis pathway was enhanced by overexpressing feedback-resistant 2-isopropylmalate synthase and acetohydroxy acid synthase both derived from Corynebacterium glutamicum , along with two other native enzymes. Next, the pyruvate and acetyl-CoA pools were enriched by deleting competitive pathways, employing the nonoxidative glycolysis pathway, and dynamically modulating the citrate synthase activity, which significantly promoted the L-leucine production and yield to 40.69 g/L and 0.30 g/g glucose, respectively. Then, the redox flux was improved by substituting the native NADPH-dependent acetohydroxy acid isomeroreductase, branched chain amino acid transaminase, and glutamate dehydrogenase with their NADH-dependent equivalents. Finally, L-leucine efflux was accelerated by precise overexpression of the exporter and deletion of the transporter. Under fed-batch conditions, the final strain LXH-21 produced 63.29 g/L of L-leucine, with a yield and productivity of 0.37 g/g glucose and 2.64 g/(L h), respectively. To our knowledge, this study achieved the highest production efficiency of L-leucine to date. Abstract: The age-related change in the importance of nutrition for muscle health starts at the age of 50. Considering its effects on the mobility and physical independence of older people, the aging of the musculoskeletal system represents one of the greatest public health challenges and tasks for a demographically aging Switzerland. In particular sarcopenia, a pathological decrease in muscle strength, muscle mass and muscle function beyond the physiological age-related changes, correlates with a significantly increased risk of falls as well as increasing morbidity and mortality. Common chronic diseases related to old age not only promote additional muscle loss but also frailty, leading to an additional decline of the quality of life. General practitioners play a crucial role in the initial assessment of changing life circumstances and activity profiles of older people. Thanks to their medical care over many years they are able to identify functional impairments of their aging patients at an early stage and address them in time. This is important because the combination of a high-protein diet and exercise may be extremely effective for improving muscle health and function. Eating more proteins (taking into account the newly revised and increased daily protein requirement for healthy seniors of 1,0–1,2g/kg body weight (bw)) can significantly slow down age-related muscle loss. Depending on age and comorbidities, the daily protein requirement might be even higher (1,5 to 2,0g/kg bw). According to current studies, a minimal protein amount of 25–35g per main dish is recommended for optimal muscle growth stimulation among older individiuals. Thanks to their highly potent boosting power on myofibrillar protein synthesis rates the amino acid L-leucine and L-leucine-rich foods play an important role in elderly people's diet. Introduction: The overuse of antibiotics has made public health and safety face a serious cisis. It is urgent to develop new clinical treatment methods to combat drug resistant bacteria to alleviate the health crisis. The efficiency of antibiotics is closely related to the metabolic state of bacteria. However, studies on fluoroquinolone resistant Salmonella are relatively rare. Methods: CICC21484 were passaged in medium with and without sarafloxacin and obtain sarafloxacin-susceptible Salmonella Typhimurium (SAR-S) and sarafloxacin resistant Salmonella Typhimurium (SAR-R), respectively. Non-targeted metabolomics was used to analyze the metabolic difference between SAR-S and SAR-R. Then we verified that exogenous L-leucine promoted the killing effect of sarafloxacin in vitro, and measured the intracellular ATP, NADH and reactive oxygen species levels of bacteria. Gene expression was determined using Real Time quantitative PCR. Results: We confirmed that exogenous L-leucine increased the killing effect of sarafloxacin on SAR-R and other clinically resistant Salmonella serotypes. Exogenous L-leucine stimulated the metabolic state of bacteria, especially the TCA cycle, which increased the working efficiency of the electron transfer chain and increased the intracellular NADH, ATP concentration, and reactive oxygen species level. Our results suggest that when the metabolism of drug-resistant bacteria is reprogrammed, the bactericidal effect of antibiotics improves. Discussion: This study further enhances research in the anti-drug resistance field at the metabolic level and provides theoretical support for solving the current problem of sarafloxacin drug resistance, a unique fluoroquinolone drug for animals and indicating the potential of L-leucine as a new antibiotic adjuvant. Particulate amorphous solid dispersions (ASDs) have been recognised for their potential to enhance the performance of various solid dose forms, especially oral bioavailability and macromolecule stability. However, the inherent nature of spray-dried ASDs leads to their surface cohesion/adhesion, including hygroscopicity, which hinders their bulk flow and affects their utility and viability in terms of powder production, processing, and function. This study explores the effectiveness of L-leucine (L-leu) coprocessing in modifying the particle surface of ASD-forming materials. Various contrasting prototype coprocessed ASD excipients from both the food and pharmaceutical industries were examined for their effective coformulation with L-leu. The model/prototype materials included maltodextrin, polyvinylpyrrolidone (PVP K10 and K90), trehalose, gum arabic, and hydroxypropyl methylcellulose (HPMC E5LV and K100M). The spray-drying conditions were set such that the particle size difference was minimised, so that it did not play a substantial role in influencing powder cohesion. Scanning electron microscopy was used to evaluate the morphology of each formulation. A combination of previously reported morphological progression typical of L-leu surface modification and previously unreported physical characteristics was observed. The bulk characteristics of these powders were assessed using a powder rheometer to evaluate their flowability under confined and unconfined stresses, flow rate sensitivities, and compactability. The data showed a general improvement in maltodextrin, PVP K10, trehalose and gum arabic flowability measures as L-leu concentrations increased. In contrast, PVP K90 and HPMC formulations experienced unique challenges that provided insight into the mechanistic behaviour of L-leu. Therefore, this study recommends further investigations into the interplay between L-leu and the physico-chemical properties of coformulated excipients in future amorphous powder design. Trehalose is commonly used as a protein stabilizer in spray dried protein formulations delivered via the pulmonary route. Spray dried trehalose formulations are highly hygroscopic, which makes them prone to deliquescence and recrystallization when exposed to moisture, leading to impairment in aerosolization performance. The main aim of this study was to investigate and compare the effect of hydrophobic amino acids (i.e. L-leucine and L-isoleucine) in enhancing aerosolization performance and in mitigating moisture-induced changes in spray dried trehalose formulations. Trehalose was spray dried with 20-60% w/w of amino acid (i.e. L-leucine or L-isoleucine). The spray dried formulations were stored at 25 °C/50% RH for 28 days. Solid state characterization and in vitro aerosolization performance studies were performed on the spray dried formulations before and after storage. The addition of 20-60% w/w of amino acid (i.e. L-leucine or L-isoleucine) improved the emitted fractions of spray dried trehalose formulations from a dry powder inhaler. However, ≥ 40% w/w of L-leucine/L-isoleucine was needed to prevent recrystallization of trehalose in the formulations when exposed to 25 °C/50% RH for 28 days. X-ray photoelectron spectroscopy (XPS) demonstrated that samples with 40-60% w/w L-isoleucine had more amino acid on the surfaces of the particles compared to their L-leucine counterparts. This may explain the greater ability of the L-isoleucine (40-60% w/w) samples to cope with elevated humidity compared to L-leucine samples of the same concentrations, as observed in the dynamic vapour sorption (DVS) studies. In conclusion, this study demonstrated that both L-leucine and L-isoleucine were effective in enhancing aerosolization performance and mitigating moisture-induced reduction in aerosolization performance in spray dried trehalose formulations. L-isoleucine proved to be superior to L-leucine in terms of its moisture protectant effect when incorporated at the same concentration in the formulations.

The eggshell colour is related to the biological functions of birds, and the colour of poultry eggshells can affect consumers' choices. This study explored the difference in the metabolite composition of duck eggshells to screen the key substances that affect the eggshell colours. The green and white duck eggshells were selected for non-targeted metabolomics analysis. We screened 402 and 512 differentially expressed metabolites in the negative and positive ion modes, respectively. Among them, 40 differentially expressed metabolites were annotated with specific names and related functions, of which the expression levels of 8 metabolites showed extremely significant differences. They were 2-heptanone, 12-hydroxydodecanoic acid, D-fructose, dodecanedioic acid, L-leucine, methyl jasmonate, palmitoleic acid, and styrene oxide. Additionally, the annotated differentially expressed metabolites were enriched in 33 metabolic pathways, including aminoacyl-tRNA biosynthesis, amino acid metabolism, galactose metabolism, etc. The results showed that the expression of metabolites between green and white eggshells differed in ducks; among metabolites with extremely significant differences in expression, the expression level of L-leucine in green eggshells was higher than that in ducks white eggshells. Therefore, we speculated that the increased expression of L-leucine promoted the response of related metabolic pathways, enhanced the expression of antioxidants, and changed the eggshell colours.Highlights The metabolites of green and white shells were different. 40 DEMs were annotated with specific names and related functions. Amorphous and crystalline active pharmaceutical ingredients (APIs) are both widely studied for pulmonary delivery. The past research mainly studied the impact of solid-state properties on pharmacokinetic attributes; however, the influence of solid-state properties on aerosolization performance was much less studied. This study aimed to investigate the different aerosolization performances of amorphous and crystalline curcumin (Cur) stabilized with L-leucine. Cur was spray-dried with different concentrations of L-leucine (0, 5, 20, 35, and 50%, w/w) as both solution-based and suspension-based formulations to acquire amorphous and crystalline Cur powders. The physicochemical properties of the spray-dried powders, including particle size, morphology, and solid-state characteristics, were studied. The aerosolization performance as well as dissolution properties were evaluated. It was found that 35% (w/w) L-leucine or above led to the formation of amorphous Cur in the spray-dried powders, and the amorphous Cur powders exhibited higher FPF (70.8%, with 50% L-leucine, w/w) than the crystalline Cur formulations with an FPF at 56.3% (with 50% L-leucine, w/w). In conclusion, with a high concentration of L-leucine (35% or above) in the formulations, amorphous Cur would exhibit higher aerosolization efficiency than crystalline Cur. Carrier-free spray-dried dispersions for pulmonary delivery, for which the demand is growing, frequently require the incorporation of dispersibility-enhancing excipients into the formulations to improve the efficacy of the dosage form. One of the most promising of such excipients, L-leucine, is expected to be approved for inhalation soon and has been studied exhaustively. However, during stability, small fibers protruding from the particles of leucine-containing powders have occasionally been observed. To clarify the origin of these fibers and assess their potential influence on the performance of the powders, three different classes of spray-dried leucine-containing formulation systems were studied over an 8-month accelerated stability program. These systems consisted of a large molecule biologic (bevacizumab) in conjunction with a glass former (trehalose), an amorphous small-molecular mass active (moxidectin), and a crystallizing active (mannitol). It was determined that the appearance of the fibers was due to the presence of small quantities of leucine in higher energy states, either because these were amorphous or present as a less stable crystalline polymorph. It was further shown that the growth of these leucine fibers caused no significant physicochemical instability in the powders. Cytosolic pyruvate is an essential metabolite in lactic acid production during microbial fermentation. However, under aerobiosis, pyruvate is transported to the mitochondrial matrix by the mitochondrial pyruvate carrier (MPC) and oxidized in cell respiration. Previous reports using Saccharomyces cerevisiae or Aspergillus oryzae have shown that the production of pyruvate-derived chemicals is improved by deleting the MPC1 gene. A previous lactate-producing K. phaffii strain engineered by our group was used as a host for the deletion of the MPC1 gene. In addition, the expression of a bacterial hemoglobin gene under the alcohol dehydrogenase 2 promoter from Scheffersomyces stipitis, known to work as a hypoxia sensor, was used to evaluate whether aeration would supply enough oxygen to meet the metabolic needs during lactic acid production. However, unlike S. cerevisiae and A. oryzae, the deletion of Mpc1 had no significant impact on lactic acid production but negatively affected cell growth in K. phaffii strains. Furthermore, the relative quantification of the VHb gene revealed that the expression of hemoglobin was detected even in aerobic cultivation, which indicates that the demand for oxygen in the bioreactor could result in functional hypoxia. Overall, the results add to our previously published ones and show that blocking cell respiration using hypoxia is more suitable than deleting Mpc for producing lactic acid in K. phaffii. Simple Summary: The Mongolian horse, one of the unique breeds in China, has excellent endurance quality formed by the half-grazing and half-wild breeding feeding mode on the grassland. However, the muscle of horses is prone to mechanical damage in the process of exercise, which can reduce the performance of horses and even cause a series of diseases. Mammalian skeletal muscle satellite cells (SCs) are located between the sarcolemma and the basal lamina of the muscle fiber. The SCs will be activated once the muscles are injured. L-leucine, as an important essential amino acid, plays an important role in the synthesis and metabolism of muscle proteins and the composition of muscle fibers. Therefore, the study on the mechanism of L-leucine regulating the proliferation of equine SCs can not only improve the exercise performance of horses but also reduce muscle injury of horses during exercises. The results of this study reveal the mechanism of L-leucine promoting the proliferation of equine skeletal muscle satellite cells. In response to muscle injury, muscle stem cells are stimulated by environmental signals to integrate into damaged tissue to mediate regeneration. L-leucine (L-leu), a branched-chain amino acid (BCAA) that belongs to the essential amino acids (AAs) of the animal, has gained global interest on account of its muscle-building and regenerating effects. The present study was designed to investigate the impact of L-leu exposure to promote the proliferation of equine skeletal muscle satellite cells (SCs) on the regulation of RNA networks, including mRNA, long non-coding RNA (lncRNA), covalently closed circular RNA (circRNA), and microRNA (miRNA) in skeletal muscles. Equine SCs were used as a cell model and cultured in different concentrations of L-leu medium. The cell proliferation assay found that the optimal concentration of L-leu was 2 mM, so we selected cells cultured with L-leu concentrations of 0 mM and 2 mM for whole-transcriptiome sequencing, respectively. By high-throughput sequencing analysis, 2470 differentially expressed mRNAs (dif-mRNAs), 363 differentially expressed lncRNAs (dif-lncRNAs), 634 differentially expressed circRNAs (dif-circRNAs), and 49 differentially expressed miRNAs (dif-miRNAs) were significantly altered in equine SCs treated with L-leu. To identify the function of autoimmunity and anti-inflammatory responses after L-leu exposure, enrichment analysis was conducted on those differentially expressed genes (DEGs) related to lncRNA, circRNA, and miRNA. The hub genes were selected from PPI Network, including ACACB, HMGCR, IDI1, HAO1, SHMT2, PSPH, PSAT1, ASS1, PHGDH, MTHFD2, and DPYD, and were further identified as candidate biomarkers to regulate the L-leu-induced proliferation of equine SCs. The up-regulated novel 699_star, down-regulated novel 170_star, and novel 360_mature were significantly involved in the competing endogenous RNA (ceRNA) complex network. The hub genes involved in cell metabolism and dif-miRNAs may play fundamental roles in the L-leu-induced proliferation of equine SCs. Our findings suggested that the potential network regulation of miRNAs, circ-RNAs, lncRNAs, and mRNAs plays an important role in the proliferation of equine SCs, so as to build up new perspectives on improving equine performance and treatment strategies for the muscle injuries of horses. In the present study, micellization behaviour of gemini surfactant ethylene-1,2-bis(N,N-dimethyl-N-dodecylammonium bromide) (12-2-12) has been investigated in the presence of three different amino acids (glycine, L-leucine, and L-methionine) at several concentrations (0.01, 0.05, 0.1 mol/kg). Measurements via conductometry at 293.15–298.15 K and surface tension measurements at 298.15 K have been employed to study surfactant-amino-acid interactions. Several parameters viz. degree of ionization (α) as well as critical micelle concentration (CMC) plus thermodynamic parameters of micellization viz. standard entropy of micellization ( Δ S m 0 ), Gibbs free energy of micellization ( Δ G m 0 ), and standard enthalpy of micellization ( Δ H m 0 ) were computed by means of conductivity measurements. Objectives: The aim of this study was to improve the aerodynamic behavior and redispersibility of a lyophilized dry powder inhaler (DPI) formulation containing nanoparticles. Methods: Paclitaxel (PTX)-human serum albumin (HSA) nanoparticles were used as a model, and DPIs containing the nanoparticles were produced by lyophilization using different carriers and carrier ratios. A central composite design was employed to optimize the formulation. L-leucine and mannitol were chosen as independent variables, and mass median aerodynamic diameter (MMAD), emitted fraction, fine particle fraction (FPF), nanoparticle size, polydispersity index (PDI), zeta potential were selected as dependent variables. Results: The water content of DPIs was less than 5% for all DPIs. The cytotoxicity of the DPIs, determined using A549 cells, was due to PTX alone. Particle sizes of 204.3 ± 1.65 nm and 94.3–1353.0 nm were obtained before and after lyophilization, respectively. The developed method resulted in a reduction in the MMAD from 8.148 µm to 5.274 µm, an increase in the FPF from 17.63% to 33.60%, and an increase in the emitted fraction from 77.68% to 97.03%. The physico-chemical characteristics of the optimized formulation were also assessed. Conclusions: In conclusion, this study demonstrates that lyophilization can be used to produce nanoparticle-containing DPI formulations with improved redispersibility and aerodynamic properties. The enzyme α-isopropylmalate synthase (α-IPMS) catalyzes the first step of L-leucine biosynthesis and is regulated via feedback inhibition by L-leucine. The gene of α-IPMS variant from strain ARTP-L04 was cloned and sequenced. Interestingly, amino acid mutations of Gly92Asp, Ile162Val, Arg494His and Gly526Asp occurred in the α-IPMS variant. The enzyme of α-IPMS variant was characterized, and exhibited higher resistance to feedback inhibition by L-leucine. In the presence of 20mM L-leucine, the activity of the α-IPMS variant was still retained at over 50%. The α-IPMS variant almost removed feedback inhibition by L-leucine, while there was no significant difference in specific activities of the α-IPMS variant and wild-type α-IPMS. For the α-IPMS variant, the capacity to overproduce L-leucine was evaluated by recombinant Corynebacterium glutamicum strains. Interestingly, expression of the α-IPMS variant could significantly enhance L-leucine production, especially co-expressed with acetohydroxyacid synthase (accumulating 7.79 g/L L-leucine). Some amino acid systems are known to exhibit solid solution and/or co-crystal behavior upon crystallization, which significantly affects their phase diagrams and complicates the design of their purification processes. Such behaviors are observed in the l-valine/l-leucine system. In this work, the formation and stability of a 3:1 co-crystal of the two amino acids (designated as V3L) is further investigated. To accomplish the formation, liquid-assisted grinding, slurry equilibration, and sublimation experiments were performed and analyzed via HPLC and PXRD. Additionally, periodic DFT calculations were used to calculate lattice energies and determine the thermodynamics of possible solid phases. Experimental results show a clear metastability of the investigated V3L co-crystals when compared to its stable solid solution. The calculations underline the metastability and the possible formation of continuous solid solutions between l-valine and l-leucine since lattice energy differences between pure amino acids and mixed compositions are negligible. This previously unknown phase behavior can be used to assess the influence of V3L on the amino acid purification process and provides a basis for investigating similar systems with small energy differences between pure and mixed compositions in future studies. In addition, it demonstrates the particular variability of solid phases and their relationships in such simple but biologically important amino acid systems.

Detection of foodborne pathogen infection by detecting key metabolic markers is an effective measure to maintain food hygiene and safety. Therefore, the rapid, efficient, sensitive and convenient method of high-performance liquid chromatography-photodiode array (HPLC-PDA) was established and optimized to determine four key metabolic biomarkers in chicken meat infected with Salmonella Enteritidis, including L-proline (Pro), L-valine (Val), L-methionine (Met), L-leucine (Leu). The limits of detection and quantification (LOD, LOQ) for four target metabolic biomarkers were 4.50–6.5 μg/kg and 25 μg/kg, respectively. The calibration curves show a good linear relationship (R2 ≥ 0.9996). The recoveries were in the range of 80–120 %, and the relative standard deviations (RSD) were all lower than 6 %. The experimental results can be used for rapid detection of chicken meat contaminated by Salmonella Enteritidis to ensure food safety. Furthermore, the HPLC-PDA method is well applied in practice. Supplements and diets containing L-leucine, a branched-chain amino acid, have been considered beneficial for controlling oxidative stress and maintaining cardiac tissue in toxicity models using doxorubicin, a drug widely used in cancer treatment. However, there is a lack of studies in the literature that assess the effects of this diet on other organs and tissues, such as the liver and kidneys. Therefore, this study aimed to evaluate the effects of a leucine-rich diet on the liver and kidneys of healthy rats submitted to the doxorubicin toxicity model by analyzing biomarkers of oxidative stress and histological parameters. The animals were divided into four groups: naive, doxorubicin, L-leucine, and doxorubicin + L-leucine, and the diet was standardized with 5% L-leucine and a dose of 7.5 mg/kg of doxorubicin. We evaluated tissue injury parameters and biomarkers of oxidative stress, including enzymes, antioxidant profile, and oxidized molecules, in the liver and kidneys. Although some studies have indicated benefits of a diet rich in L-leucine for the muscle tissue of animals that received doxorubicin, our results showed that the liver was the most affected organ by the L-leucine-rich diet since the diet reduced its antioxidant defenses and increased the deposit of collagen and fat in the hepatic tissue. In the kidneys, the main alteration was the reduction in the number of glomeruli. These results contribute to the scientific literature and encourage further studies to evaluate the effects of an L-leucine-rich diet or its supplementation, alone or combined with doxorubicin using an animal model of cancer. Therefore, our study concludes that the leucine-rich diet itself was harmful and, when co-administered with doxorubicin, was not able to maintain the antioxidant defenses and tissue structure of the evaluated organs.

Graphene known as the wonder material and finds its utilization in various applications including sensors.This work reports the solvothermal route and the chemical reflux method synthesis of graphene oxide copper oxide nanocomposite material (GO-CuO). The hitherto prepared GO-CuO nanocomposite was well characterized using the usual spectroscopic and microscopic technique such as Fourier transform infrared (FTIR) spectroscopy, Scanning electron microscopy (SEM), X-ray diffraction (XRD), UV-Vis Spectroscopy (UV), Photoluminescence Spectroscopy(PL) etc. The prepared GO-CuO nanocomposite was found to selectively bind to L-Leucine without any interference from the other naturally occurring L-amino acids as a potential fluorescent chemosensor. The interference studies, effect of time, effect of pH and various equivalence studies were also performed for the GO-CuO nanocomposite with L-Leucine. The antibacterial activity of GO-CuO nanocomposite revealed a minimum inhibitory zone concentration for Methicillin Resistant Staphylococcus aureus to be 200 μg/ml, whereas for A. baumannii it was found to be 100 μg/ml. L-Leucine (L-Leu) in ovo administration was demonstrated to afford thermotolerance and modified amino acids metabolism in post-hatched broiler chicks under heat stress. This study aimed to investigate the changes in embryonic growth and amino acid metabolism after in ovo injection of L-Leu. Fertilized broiler eggs were subjected to in ovo injection of sterile water or L-Leu on embryonic day (ED) 7. The weight of embryos and yolk sacs were measured on ED 12, 14, 16, and 18. Plasma and livers were collected on ED 14 and 18 for free amino acid analysis. The weight and relative weight of embryos were significantly lowered by in ovo administration of L-Leu, but those of yolk sacs were not altered. Moreover, L-Leu in ovo injection significantly reduced the plasma proline concentration during embryogenesis and increased the plasma concentrations of tyrosine (Tyr) and lysine (Lys) in ED 18. Hepatic Lys concentration was also significantly increased by L-Leu in ovo injection. Interestingly, Leu concentrations in the plasma and liver were not affected by L-Leu administration. These results indicated that in ovo administered L-Leu was metabolized before ED 14 and affected embryonic growth and amino acid metabolism during embryogenesis.

Hydroxyl radical (HO·) formation initiated by the Fenton-type reactions of Fe and Cu complexes of l-leucine (Leu) amino acid as well as its oxidation reaction by HO· was computationally investigated by using the density functional theory method at the M05-2X/6-311++G(3df,2pd)//M05-2X/6-311++G(d,p) level of theory in the aqueous phase. The results showed that dipole-salt is the main form of Leu in the physiological condition. Leu exhibits high chelating potential towards both Fe(III)/Fe(II) and Cu(II)/Cu(I) ions with the most favourable coordinating positions at two oxygen atoms of the –COO functional group. Furthermore, the Leu-ions complexes show a high risk of HO· formation via Fenton-like reactions, especially when ascorbate anion exists in the environment as a reducing agent. Finally, the oxidation reaction of l-leucine by HO· demonstrated a relatively high overall apparent reaction rate, koverall, being 1.18 × 109 M−1 s−1, in which formal hydrogen transfer reactions of the dipole-salt form occur as the primary mechanism. Consequently, the Leu oxidation by HO· radical can be promoted by the Fenton reaction enhancement of its transition metal complexes.

Particulate amorphous solid dispersions (ASDs) have been recognised for their potential to enhance the performance of various solid dose forms, especially oral bioavailability and macromolecule stability. However, the inherent nature of spray-dried ASDs leads to their surface cohesion/adhesion, including hygroscopicity, which hinders their bulk flow and affects their utility and viability in terms of powder production, processing, and function. This study explores the effectiveness of L-leucine (L-leu) coprocessing in modifying the particle surface of ASD-forming materials. Various contrasting prototype coprocessed ASD excipients from both the food and pharmaceutical industries were examined for their effective coformulation with L-leu. The model/prototype materials included maltodextrin, polyvinylpyrrolidone (PVP K10 and K90), trehalose, gum arabic, and hydroxypropyl methylcellulose (HPMC E5LV and K100M). The spray-drying conditions were set such that the particle size difference was minimised, so that it did not play a substantial role in influencing powder cohesion. Scanning electron microscopy was used to evaluate the morphology of each formulation. A combination of previously reported morphological progression typical of L-leu surface modification and previously unreported physical characteristics was observed. The bulk characteristics of these powders were assessed using a powder rheometer to evaluate their flowability under confined and unconfined stresses, flow rate sensitivities, and compactability. The data showed a general improvement in maltodextrin, PVP K10, trehalose and gum arabic flowability measures as L-leu concentrations increased. In contrast, PVP K90 and HPMC formulations experienced unique challenges that provided insight into the mechanistic behaviour of L-leu. Therefore, this study recommends further investigations into the interplay between L-leu and the physico-chemical properties of coformulated excipients in future amorphous powder design. This also revealed the need to enhance bulk characterisation tools to unpack the multifactorial impact of L-leu surface modification.

Isoamyl alcohol (i-AmOH) is produced from a-ketoisocaproate in the l -leucine biosynthetic pathway in yeast and controlled by the negative feedback regulation of a-isopropylmalate synthase (IPMS), which senses the accumulation of l -leucine. It is known that i-AmOH production increases when mutations in the regulatory domain reduce the susceptibility to feedback inhibition. However, the impact of mutations in this domain on the IPMS activity has not been examined. In this study, we obtained 5 IPMS mutants, encoding the LEU4 gene, N515D/S520P/S542F/A551D/A551V, that are tolerant to 5,5,5-trifluoro-dl-leucine. All mutant proteins were purified and examined for both IPMS activity and negative feedback activity by in vitro experiments. The results showed that not only the negative-feedback regulation by L-leucine was almost lost in all mutants, but also the IPMS activity was greatly decreased and the difference in IPMS activity among Leu4 mutants in the presence of l -leucine was significantly correlated with i-AmOH production. The solute carrier L-type amino acid transporter 1 (LAT-1/SLC7A5) is a viable target for drug delivery to the central nervous system (CNS) and tumors due to its high abundance at the blood–brain barrier and in tumor tissue. LAT-1 is only localized on the cell surface as a heterodimer with CD98, which is not required for transporter function. To support future CNS drug-delivery development based on LAT-1 targeting, we established an ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) assay for stable isotopically labeled leucine ([13C6, 15N]-L-leucine), with a dynamic range of 0.1–1000 ng/mL that can be applied for the functional testing of LAT-1 activity when combined with specific inhibitors and, consequently, the LAT-1 inhibition capacity of new compounds. The assay was established in a 96-well format, facilitating high-throughput experiments, and, hence, can support the screening for novel inhibitors. Applicable recommendations of the US Food and Drug Administration and European Medicines Agency for bioanalytical method validation were followed to validate the assay. The assay was applied to investigate the IC50 of two well-known LAT-1 inhibitors on hCMEC/D3 cells: the highly specific LAT-1 inhibitor JPH203, which was also used to demonstrate LAT-1 specific uptake, and the general system L inhibitor BCH. In addition, the [13C6, 15N]-L-leucine uptake was determined on two human brain capillary endothelial cell lines (NKIM-6 and hCMEC/D3), which were characterized for their expressional differences of LAT-1 at the protein and mRNA level and the surface amount of CD98. The IC50 values of the inhibitors were in concordance with previously reported values. Furthermore, the [13C6, 15N]-L-leucine uptake was significantly higher in hCMEC/D3 cells compared to NKIM-6 cells, which correlated with higher expression of LAT-1 and a higher surface amount of CD98. [Display omitted] Both latent and multidrug-resistant tuberculosis (TB) have been causing significant concern worldwide. A novel drug, pretomanid (PA-824), has shown a potent bactericidal effect against both active and latent forms of Mycobacterium tuberculosis (MTb) and a synergistic effect when combined with pyrazinamide and moxifloxacin. This study aimed to develop triple combination spray dried inhalable formulations composed of antitubercular drugs, pretomanid, moxifloxacin, and pyrazinamide (1:2:8 w/w/w), alone (PaMP) and in combination with an aerosolization enhancer, L-leucine (20 % w/w, PaMPL). The formulation PaMPL consisted of hollow, spherical, dimpled particles (<5 μm) and showed good aerosolization behaviour with a fine particle fraction of 70 %. Solid-state characterization of formulations with and without L-leucine confirmed the amorphous nature of moxifloxacin and pretomanid and the crystalline nature of pyrazinamide with polymorphic transformation after the spray drying process. Further, the X-ray photoelectron spectroscopic analysis revealed the predominant surface composition of L-leucine on PaMPL dry powder particles. The dose–response cytotoxicity results showed pyrazinamide and moxifloxacin were non-toxic in both A549 and Calu-3 cell lines up to 150 µg/mL. However, the cell viability gradually decreased to 50 % when the pretomanid concentration increased to 150 µg/mL. The in vitro efficacy studies demonstrated that the triple combination formulation had more prominent antibacterial activity with a minimum inhibitory concentration (MIC) of 1 µg/mL against the MTb H37Rv strain as compared to individual drugs. In conclusion, the triple combination of pretomanid, moxifloxacin, and pyrazinamide as an inhalable dry powder formulation will potentially improve treatment efficacy with fewer systemic side effects in patients suffering from latent and multidrug-resistant TB. Cytosolic pyruvate is an essential metabolite in lactic acid production during microbial fermentation. However, under aerobiosis, pyruvate is transported to the mitochondrial matrix by the mitochondrial pyruvate carrier (MPC) and oxidized in cell respiration. Previous reports using Saccharomyces cerevisiae or Aspergillus oryzae have shown that the production of pyruvate-derived chemicals is improved by deleting the MPC1 gene. A previous lactate-producing K. phaffii strain engineered by our group was used as a host for the deletion of the MPC1 gene. In addition, the expression of a bacterial hemoglobin gene under the alcohol dehydrogenase 2 promoter from Scheffersomyces stipitis, known to work as a hypoxia sensor, was used to evaluate whether aeration would supply enough oxygen to meet the metabolic needs during lactic acid production. However, unlike S. cerevisiae and A. oryzae, the deletion of Mpc1 had no significant impact on lactic acid production but negatively affected cell growth in K. phaffii strains. Furthermore, the relative quantification of the VHb gene revealed that the expression of hemoglobin was detected even in aerobic cultivation, which indicates that the demand for oxygen in the bioreactor could result in functional hypoxia. Overall, the results add to our previously published ones and show that blocking cell respiration using hypoxia is more suitable than deleting Mpc for producing lactic acid in K. phaffii.

Carrier-free spray-dried dispersions for pulmonary delivery, for which the demand is growing, frequently require the incorporation of dispersibility-enhancing excipients into the formulations to improve the efficacy of the dosage form. One of the most promising of such excipients, L-leucine, is expected to be approved for inhalation soon and has been studied exhaustively. However, during stability, small fibers protruding from the particles of leucine-containing powders have occasionally been observed. To clarify the origin of these fibers and assess their potential influence on the performance of the powders, three different classes of spray-dried leucine-containing formulation systems were studied over an 8-month accelerated stability program. These systems consisted of a large molecule biologic (bevacizumab) in conjunction with a glass former (trehalose), an amorphous small-molecular mass active (moxidectin), and a crystallizing active (mannitol). It was determined that the appearance of the fibers was due to the presence of small quantities of leucine in higher energy states, either because these were amorphous or present as a less stable crystalline polymorph. It was further shown that the growth of these leucine fibers caused no significant physicochemical instability in the powders. Nor, more importantly, did it decrease their aerosol performance in a dry powder inhaler or reduce the concentration of their active pharmaceutical ingredients.

In response to muscle injury, muscle stem cells are stimulated by environmental signals to integrate into damaged tissue to mediate regeneration. L-leucine (L-leu), a branched-chain amino acid (BCAA) that belongs to the essential amino acids (AAs) of the animal, has gained global interest on account of its muscle-building and regenerating effects. The present study was designed to investigate the impact of L-leu exposure to promote the proliferation of equine skeletal muscle satellite cells (SCs) on the regulation of RNA networks, including mRNA, long non-coding RNA (lncRNA), covalently closed circular RNA (circRNA), and microRNA (miRNA) in skeletal muscles. Equine SCs were used as a cell model and cultured in different concentrations of L-leu medium. The cell proliferation assay found that the optimal concentration of L-leu was 2 mM, so we selected cells cultured with L-leu concentrations of 0 mM and 2 mM for whole-transcriptiome sequencing, respectively. By high-throughput sequencing analysis, 2470 differentially expressed mRNAs (dif-mRNAs), 363 differentially expressed lncRNAs (dif-lncRNAs), 634 differentially expressed circRNAs (dif-circRNAs), and 49 differentially expressed miRNAs (dif-miRNAs) were significantly altered in equine SCs treated with L-leu. To identify the function of autoimmunity and anti-inflammatory responses after L-leu exposure, enrichment analysis was conducted on those differentially expressed genes (DEGs) related to lncRNA, circRNA, and miRNA. The hub genes were selected from PPI Network, including ACACB, HMGCR, IDI1, HAO1, SHMT2, PSPH, PSAT1, ASS1, PHGDH, MTHFD2, and DPYD, and were further identified as candidate biomarkers to regulate the L-leu-induced proliferation of equine SCs. The up-regulated novel 699_star, down-regulated novel 170_star, and novel 360_mature were significantly involved in the competing endogenous RNA (ceRNA) complex network. The hub genes involved in cell metabolism and dif-miRNAs may play fundamental roles in the L-leu-induced proliferation of equine SCs. Our findings suggested that the potential network regulation of miRNAs, circ-RNAs, lncRNAs, and mRNAs plays an important role in the proliferation of equine SCs, so as to build up new perspectives on improving equine performance and treatment strategies for the muscle injuries of horses.

Tea is a very important source of the terepenoid α -ionone, which is much appreciated as a medicinal beverage. Schiff bases are a very important class of organic compounds usually formed by the condensation of carbonyl compounds with amines. α -ionone is a carbonyl terpenoid obtained from Schiff bases on condensation with nucleobases like adenine and cytosine and the amino acid l -leucine. We synthesized these three Schiff bases and characterized them using UV, FTIR, and H1 and C13-NMR spectra. The molecules were optimized using B3LYP/6-311+G(2d,p) level followed by the simulation of FT-IR spectra, of which the simulated and experimental spectra showed complete agreement. The UV spectra were simulated using TD-DFT, and the electronic excitations were carefully analyzed. Natural bond orbitals provided an analysis of the stability of the compound, which is supplemented by the data from frontier molecular orbital analysis. Detailed wavefunction analysis is reported which predicts the active centers, reactivity profile, and the extent of non-covalent interactions. PASS indicated that compounds show antieczemic properties and antiarthritic properties, which is confirmed with the help of molecular docking results. It has been widely reported that the addition of trimethylglycine (betaine) decreases osmotic pressure inhibition for cell growth, leading to increased production of amino acids. However, the underlying mechanism is unclear. To determine the global metabolic differences that occur under the addition of trimethylglycine, transcriptome analysis was performed. Transcriptome analysis of Corynebacterium glutamicum JL1211 revealed that 272 genes exhibited significant changes under trimethylglycine addition. We performed Gene Ontology (GO) and KEGG enrichment pathway analyses on these differentially expressed genes (DEGs). Significantly upregulated genes were mainly involved in the regulation of ABC transporters, especially phosphate transporters and sulfur metabolism. The three phosphate transporter genes pstC, pstA and pstB were upregulated by 13.06-fold, 29.80-fold and 30.49-fold, respectively. Notably, the transcriptional levels of the cysD, cysN, cysH and sir genes were upregulated by 81.5-fold, 57.3-fold, 77.6-fold and 125.4-fold, respectively, consistent with assimilatory sulfate reduction under the addition of trimethylglycine. The upregulation of ilvBN and leuD genes might result in increased l-leucine formation. The data indicated changes in the transcriptome of C. glutamicum with trimethylglycine treatment, thus providing a mechanism supporting the application of trimethylglycine in the production of l-leucine and other amino acids by C. glutamicum strains. Medicinal plants and their combinations due to the wide range of biologically active substances can influence on various links of the pathogenetic mechanism of development of DM type 2 and its complications. One of such combinations is an antidiabetic herbal mixture (Urticae folia, Rosae fructus, Myrtilli folia, Menthae folia and Taraxaci radices) with established hypoglycaemic, hypolipidemic, antioxidant, hepatoprotective, pancreatoprotective activity in previous pharmacological studies in vivo and in vitro and defined phytochemical composition. Thus, the aim of this study was to identify and establish the content of amino acids in the plant components of antidiabetic herbal mixture. The amino acids were separated by GC-MS method with pre-column derivatization. The calibration curves of twenty CRS of amino acids were linear (R² > 0.98) over the range of 1–100 μg/mL, the LODs and the LOQs were in the range of 0.01–0.07 μg/mL and 0.02–0.20 μg/mL, respectively. The results of analysis showed that the predominant essential amino acid was L-proline in Taraxaci radices, Urticae folia, Rosae fructus and Menthae folia, its total content was 101.46 mg/g, 25.31 mg/g, 23.04 mg/g and 19.30 mg/g, respectively. In addition, it was established total content of essential amino acid – L-leucine that can stimulate insulin secretion in β-cells of the pancreas. Its total content was 58.51 mg/g in Taraxaci radices, 9.58 mg/g in Myrtilli folia, 4.68 mg/g in Rosae fructus, 2.99 mg/g in Urticae folia and 0.79 mg/g in Menthae folia. Chromatographic examination also revealed L-phenylalanine, an essential amino acid important for antidiabetic therapy that can increase insulin secretion, stimulate proliferation and neogenesis of β-cells of the pancreas and reduce insulin resistance. Its total content was 13.42 mg/g in Myrtilli folia, 2.23 mg/g in Rosae fructus, 1.478 mg/g in Urticae folia, 1.46 mg/g in Taraxaci radices and 0.52 mg/g in Menthae folia. This phytochemical study shows, which plant material forms the amino acid composition and content in the finished herbal mixture and due to which biologically active substances the antidiabetic activity of this phytocomposition is manifested. Statins are essential naturally occurring β-hydroxy-γ-amino acids and (3S,4S)-Statine, derived from L-Leucine, is found in important aspartic proteases inhibitors like Pepstatin A. We developed a new synthesis of (3S,4S)-Statine based on the highly diastereoselective reduction of N,N-Boc/Benzyl protected β-ketoester intermediate. This Boc/Benzyl protection strategy gives access to either N-Fmoc or N-Benzyl protected (3S,4S)-Statine on multigram scale quantities.

Here, L-leucine based electrochemical sensor was designed for the simultaneous analysis of paracetamol (PA) and folic acid (FA). Electropolymerization of L-leucine was carried out on the surface of the carbon paste electrode (CPE). The electrochemical deeds of the biomolecules were explored through cyclic voltammetry (CV) and differential pulse voltammetry (DPV) at the fabricated sensor. The invented poly (L-leucine) modified carbon paste electrode (PLCN-MCPE) showed very good interaction with the analyte molecules which results in the significant determination of PA and FA in terms of sensitivity and selectivity. The electrochemical reaction mechanism between the surface of the developed sensor and the analytes was carried out in terms of effects of scan rate, concentration, and the electrode process was found to be adsorption-controlled. The kinetics such as the number of electron transfer (n) and the heterogeneous rate constant (ko) were defined for PA and FA at MCPE. The lower limit of detection for PA and FA were assessed to be 0.44 and 2.4 µM correspondingly. As well, the availability of the sensor in real-world sample analysis was also investigated with satisfactory results.
