Doktora Tezi, Prostat kanseri erkeklerde en sık görülen kanserdir. Akciğer kanserinden sonra erkeklerde kansere bağlı ölümlerde ikinci sırayı almaktadır ve kanser ölümlerinin %10'undan sorumludur. Prostat kanserlerinin oluşmasında yaş ve genetik faktörler oldukça önemlidir. Kinetokor proteinlerinin karsinogenezle bağlantıları yapılan çalışmalarda gösterilmiştir. Bunlardan birisi olan sentromer protein H'ın bazı kanser türlerinin gelişimi ile olan bağlantıları tespit edilmiştir. Bu çalışmada sentromer protein H (CENP-H) ekspresyon düzeyinin prostat karsinomunda (PCa) değişip-değişmediğinin saptanması ve karsinogenez sürecindeki muhtemel rolünün belirlenmesi amaçlanmıştır. Bu amaçla, prostat kanseri tanısı almış hastalardan çıkarılan total prostat dokularından elde edilen parafine gömülmüş örneklerde CENP-H geninin transkripsiyonel analizi gerçekleştirilmiştir. Ekspresyon analizleri aynı hastanın prostat dokusundaki tümöral ve non-tümöral alanlardaki ekspresyonun karşılaştırılması ile elde edilmiştir. Çalışmaya 40 adet radikal prostatektomi materyali alınmış, daha sonra yüksek Gleason skorlu 9 doku materyali; normal prostat dokusu alanı tümör dokusu alanı ile çok içiçe olduğu için; 3 doku materyali de RNA elde edilemediği için çalışmadan çıkarılmıştır. CENP-H gen ekspresyon analizi Real Time PCR (Gerçek zamanlı Polimeraz Zincir Reaksiyonu) (PZR) ile yapılmıştır. Kalan 28 olgunun tümöre ait doku örnekleri ve tümörsüz normal alana ait doku örneklerinden ayrı ayrı RNA izolasyonları gerçekleştirildi. Daha sonra gerçekleştirilen qRT-PZR analizleri üçer defa tekrarlanmış ve elde edilen verilerden Ct değerlerinin kalite kontrolleri yapılmıştır. House keeping gen GAPDH ve hedef gen CENP-H'a ait Ct değerleri doku (tümör ve normal) ve teknik tekrar (tekrar -1, -2, -3) gruplarında karşılaştırılmıştır. GADPH Ct değerleri tümör dokularında (25,62 ±0,3182) kontrol dokularına göre (26,61±o,3182) anlamlı düzeyde düşük bulunmuştur (p=0,029). CENP-H geni için gruplar ve teknik tekrarlar arsında farklılık gözlenmemiştir. Çalışmada, prostat kanserinde normal doku ile tümör dokusu arasında CENP-H ekspresyonu bakımından herhangi bir farklılık tespit edilememiştir. Ayrıca ölüm nedenlerine bakıldığında, hastaların hiç birisinde PCa nedenli mlüm saptanmamıştır., Prostate cancer is the most frequent cancer type in men. After lung cancer, it is the second most common cause of morbidity in men responsible for 10 % of deaths. Age and genetic factors have a high significance in the development of prostate cancer. The relation of kinetochore proteins with carcinogenesis have been shown in the studies made. The association of centromere protein H in the development cancer types has been determined. In the present study, the aim is to determine whether centromere protein H (CENP -H) expression levels change in prostate carcinoma (PCa) and the possible role of CENP -H in the carcinogenesis process. Hence, transcriptional analysis of the CENP –H gene was conducted in the total prostate tissues extracted from patients diagnosed with prostate cancer and embedded in paraffin. The expression analyses were obtained from the comparison of the expressions within the tumoral and non-tumoral areas in the prostate tissue of the same patient. For the present study, 40 radical prostatectomy samples were recruited; yet 9 tissue material with a high Gleason score were excluded as the normal prostate tissue area was to closely intertwined with tumor tissue and 3 due to the inability of obtaining RNA. CENP -H gene expression analysis was made using Real time PCR (real-time polymerase chain reaction). RNA isolations were carried out for tumor and non-tumor tissue areas separately in the remaining 28 samples. Subsequently, qRT - PCR analyzes were repeated three times and from the data obtained the quality of Ct values controlled. Ct values of the house keeping gene GAPDH and of the target CENP -H gene were compared among the tissue (tumor and normal) and the technical repetition (repetition -1, -2, -3) groups. In the tumor tissues GADPH Ct values (25.62 ± 0.3182) were significantly lower (p = 0.029) than the control tissues (26.61 ± 0.3182). No statistically significant difference for the CENP -H gene was obtained from the group and technical repetition comparisons. In the present study, no statistically significant in the CENP -H expression was determined between tumor and normal tissue in prostate cancer. Moreover, on investigating the causes of death, in none of the patients PCa related death was determined.

Background: Kinetochores are large multi-protein structures that assemble on centromeric DNA (CEN DNA) and mediate the binding of chromosomes to microtubules. Comprising 125 base-pairs of CEN DNA and 70 or more protein components, Saccharomyces cerevisiae kinetochores are among the best understood. In contrast, most fungal, plant and animal cells assemble kinetochores on CENs that are longer and more complex, raising the question of whether kinetochore architecture has been conserved through evolution, despite considerable divergence in CEN sequence. Results: Using computational approaches, ranging from sequence similarity searches to hidden Markov model-based modeling, we show that organisms with CENs resembling those in S. cerevisiae (point CENs) are very closely related and that all contain a set of 11 kinetochore proteins not found in organisms with complex CENs. Conversely, organisms with complex CENs (regional CENs) contain proteins seemingly absent from point-CEN organisms. However, at least three quarters of known kinetochore proteins are present in all fungi regardless of CEN organization. At least six of these proteins have previously unidentified human orthologs. When fungi and metazoa are compared, almost all have kinetochores constructed around Spc105 and three conserved multi-protein linker complexes (MIND, COMA, and the NDC80 complex). Conclusion: Our data suggest that critical structural features of kinetochores have been well conserved from yeast to man. Surprisingly, phylogenetic analysis reveals that human kinetochore proteins are as similar in sequence to their yeast counterparts as to presumptive Drosophila melanogaster or Caenorhabditis elegans orthologs.
