3. Crossings with Jatra: Bengali Folk-theatre Elements in a Transcultural Representation of Lady Macbeth

This paper examines a transcultural dance-theatre focusing on Lady Macbeth, through the lens of eastern Indian Bengali folk-theatre tradition, jatra. The wide range of experimentation with Shakespeare notwithstanding, the idea of an all-female representation is often considered a travesty. Only a few such explorations have earned recognition in contemporary times. One such is the Indian theatre-dance production Crossings: Exploring the facets of Lady Macbeth by Vikram Iyenger, first performed in 2004. Four women representing four facets of Lady Macbeth explore the layered nuances that constitute her through the medium of Indian classical dance and music juxtaposed with Shakespearean dialogues from Macbeth. This paper will argue the possibilities posited by this transgressive re-reading of a major Shakespearean tragedy by concentrating on a possible understanding through a Hindu religious sect —Vaishnavism, as embodied through the medium of jatra. To form a radically new stage narrative in order to bring into focus the dilemma and claustrophobia of Lady Macbeth is perhaps the beginning of a new generation of Shakespeare explorations. Iyenger’s production not only dramatizes the tragedy of Lady Macbeth through folk dramatic tradition, dance and music, but also Indianises it with associations drawn from Indian mythological women like Putana (demoness) and Shakti (sacred feminine).

4. Stanovení jaterní steatózy MR spektroskopickými metodami PRESS, STEAM a HISTO.

Aim: To compare MR spectroscopic methods for steatosis determination by PRESS, STEAM and HISTO sequences on 3T TRIO and VIDA tomographs. Method: 1H MR spectra of the liver of volunteers were measured on 3T tomographs Siemens TRIO and VIDA (45/25 subjects). The sequences PRESS with TE = 30/33 ms, STEAM with TE = 20/33 ms and the HISTO method with TE = 12-72 ms were used. The same sequences were used to determine T2 of lipids and water. The spectra were processed using the LCModel method. Fractional (FF) and volume (HFC) fractions (%) were corrected for individual and mean T2 values. Correlation, regression and Bland-Altman data analyses were performed. Results: Relaxation times T2 reduce FF and HFC values by up to approx. 47%. Regression analysis and Bland-Altman graphs show that the parameters obtained by PRESS, STEAM and HISTO techniques with short TE are comparable on VIDA. Mean T2 values of lipids between 45 and 53 ms and T2 of water between 24 and 31 ms were measured. HISTO FF cut-off values for the degree of steatosis in transplant patients were set in the following ranges:S0: < 0.8%; S1: 0.81-6.2%; S2: 6.21-16.5%; S3: > 16.5%. Conclusion: The obtained FF and HFC values are sensitive to T2 corrections. Bland-Altman's graphs indicate good agreement among STEAM, PRESS and HISTO methods. Limit values for the steatosis degree in transplant patients from the result of the HISTO method agree with literature values. [ABSTRACT FROM AUTHOR]

Tato bakalářská práce pojednává o hepatotoxicitě těžkých kovů. V první části práce je charakterizována in vitro buněčná kultivace. Na tuto část volně navazuje kapitola, ve které jsou charakterizovány buněčné kultury pro in vitro testování hepatotoxicity lidského i zvířecího původu. Nejpoužívanější buněčnou linií je HepG2, která je odvozená z buněk lidského hepatomu. Déle jsou zde charakterizovány primární lidské hepatocyty a lidské jaterní buněčné linie HepaRG a Huh-7. V práci je uveden i výčet zvířecích jaterních buněčných kultur. V hlavní části práce jsou definovány a charakterizovány těžké kovy a jejich působení na lidský organizmus. V další kapitole jsou podrobně popsány mechanizmy hepatotoxicity těžkých kovů se zaměřením na kadmium. Kadmium narušuje mitochondriální funkce, indukuje autofagii, apoptózu, ferroptózu a zánět. Kadmium významně ovlivňuje homeostázu lipidů v játrech., This bachelor thesis discusses the hepatotoxicity of heavy metals. In vitro cell culture is characterized in the first part of the thesis. This part freely links the chapter in which cell cultures for in vitro testing of hepatotoxicity of human and animal origin are characterized. The most common cell line derived from human hepatoma cells is HepG2. Primary human hepatocytes and the human liver cell lines HepaRG and Huh-7 are further described. This thesis also includes a list of animal liver cell cultures. The main part of the thesis is devoted to definiton and characterization of heavy metals. In the next chapter, the mechanisms of hepatotoxicity of heavy metals are described in detail, focusing on cadmium. Jatra is known for its distinctive presentational styles comprising song-dance sequences, larger than life physicalization, enthralling dialogues, and the anthropomorphizing of the "conscience," commonly blown as the bibek. This article focuses on an interview with Shekh Madhusudan, the 87-year old Jatra artist who reminiscences about his relationship with this form of theatre. This interview exposes how religion, and his journey as a bibek in relation to the dramatic and theatrical theories of postcolonialism and hybridity inform the nature of contemporary Jatra in West Bengal. Liver cell regeneration is an important biological process that allows mammals to maintain liver function while recovering from liver damage. Liver cell proliferation serves as the primary mode of liver regeneration, which in hepatocytes is activated by the transition from the G0 to G1 phase of the cell cycle. Proliferation is also promoted by non-parenchymal liver cells among which include Ito cells, Kupffer cells, and endothelial cells of hepatic sinusoids. In a comprehensive analysis of key signaling pathways, it was clearly demonstrated that the Wnt/β catenin, Notch, Hippo, NF-κB, and Hedgehog signaling pathways play a key role in the regulation of liver cell proliferation and differentiation during regeneration. The regenerative potential of the liver is influenced by various factors such as age, extent of damage and health conditions. Additionally, the remarkable regenerative capacity of the liver has clinical implications in the context of liver transplantation, partial hepatectomy and the treatment of liver diseases such as cirrhosis, hepatitis and hepatocellular or cholangiocellular carcinoma. Methods: This bachelor's thesis is divided into two parts. The liver and large intestine's anatomy, physiology, and pathophysiology are discussed in the first general section. Following chapters are devoted to the main treatment options for advanced liver diseases, i.e. liver transplantation, and indication and possibilities of colon resection. Physiotherapeutic approaches used in the mentioned procedures and further options of treatment for these patients are mentioned in the later chapters. The circadian system influences almost all physiological processes in the mammalian body. Among other things, detoxication processes in the liver are under circadian control. Both central and peripheral clock in the liver regulate the expression of genes involved in the detoxication of xenobiotic substances and drugs. The first part of this thesis summarizes the main characteristics of the central and peripheral circadian clocks, including their molecular basis. The second part focuses on the main functions of liver tissue with a focus on detoxification processes. The emphasis of the third section is on circadian rhythms in the expression and activity of enzymes called cytochromes P450, which are the most important system catalyzing phase I detoxication, and also briefly discusses the influence of the circadian system on the expression of nuclear receptors and PAR bZIP transcription factors involved in the regulation of cytochrome P450 transcription. The last section describes the influence of the circadian system on the efficacy and toxicity of selected drugs, on paracetamol metabolism with the main purpose of describing how paracetamol-induced hepatotoxicity is affected by time of administration. The present thesis investigated the impact of a ten-week oral exposure to BPS (50 ?g/kg live weight/day) in female outbred Wistar Han rats. The effect of BPS exposure on live weight, absolute and relative kidney and liver weights, morphometric parameters of kidney and renal corpuscles, histopathological changes of kidney and liver were evaluated. In addition, the concentration of total protein, triacylglycerols, cholesterol, urea and alanine aminotransferase activity were determined in blood serum.

Objectives: To compare speed and accuracy between both the automatic and manual CT liver volumetry and between two radiologist with different long of their medical practice. Methods: 66 patients with liver metastases of colorectal cancer were included to the study. The analysis of total liver volume and FLRV (future liver remnant volume) was performed using automatic and manual CT volumetry by two different experienced radiologist. Results: Statistically significantly faster procedure was observed by the automatic CT volumetry in comparison to the manual method (p < 0.0001). No statistically significantly different value of detected liver volumes were found by manual and automatic method (p = 0.552) neither the two performing radiologist (p = 0.612 from time and p = 0.909 from volumes). Conclusion: The automatic CT volumetry is optimal method for the measurement of the liver. The results are accurate and the speed of analysis is significantly higher in comparison to the manual CT volumetry. [ABSTRACT FROM AUTHOR]

- Abstract
Aim: To present a rare case report about 38-year-old man with recurrence of the epithelioid hemangioendothelioma in the liver which was detected by means of dual-time 18F-FDG PET/CT. Methods: The patient was examined for a few weeks lasting low back pain. The abdomen ultrasound and computed tomography detected multifocal lesions in the liver. The epithelioid hemangioendothelioma was proven by biopsy and the patient underwent chemotherapy. The finding had been stable without changes on the repeated follow-up radiology imaging for 12 years. A new lesion in the right liver lobe was detected on the next follow-up imaging, which was suspected for the recurrence. The PET/CT scan from base of skull to mid femur was obtained 60 minutes after intravenous injection of 18F- -FDG. The PET scan acquisition time was 2.5 minutes per bed position and seven bed positions were necessary. Contrast-enhanced CT (venous phase) provided both full CT evaluation (including intravenous and oral contrast) and PET attenuation correction. Results: No pathological 18F-FDG uptake was detected on early PET/CT scan and so the next delayed PET/CT imaging was performed 3 hours after 18F-FDG injection. The increased focal 18F-FDG uptake was demonstrated in the incriminated lesion in the right liver lobe on the delayed PET/CT images, this lesion showed a significant increase of tumor lesion-to-liver background ratio. The next distant or locoregional focus of the increased 18F-FDG uptake were excluded. The hepatic recurrence of epithelioid hemangioendothelioma was proven by biopsy. Conclusion: In this report dual-time- -point 18F-FDG PET/CT imaging correctly detected recurrent epithelioid hemangioendothelioma in a liver. The delayed scan significantly improved sensitivity of the PET/CT examination and detected the hypermetabolic lesion in a liver. [ABSTRACT FROM AUTHOR]

- Abstract
1HMR spectroscopy is a suitable method of fat evaluation in human body. It is possible to determine the level of liver steatosis using the measurement of water and fatty acids signal intensities. The results from spectroscopy correlate well with the results of histology. The method is useful for long-term studies due to its non-invasive character. A similar examination procedure can be used for fat measurement in myocardium. Editing 1H MRS techniques with different echo times are used to measure intra- and extracellular fat in muscles. Advanced editing techniques such as oMEGA PRESS enable measuring the content of omega fatty acids. Editing techniques are also applied for the measurement of metabolite concentrations in prostate. The concentration of citrate significantly increases with the age of healthy males. The age dependency of metabolite concentrations has to be considered in long-term studies as well as in the case of patients' examinations. [ABSTRACT FROM AUTHOR]

Title: A Case Study of Physiotherapy on a Patient after Liver Transplantation Author: Lukas Hettiaratchi Objectives: The purpose of this work is to review in-depth theory concerning liver transplantations along with the rehabilitation process that entails it. Moreover, create an adequate case study of a patient after liver transplantation. Methods: This work is divided into two sections, the theoretical part, and the special part. The first part consists of information gathered through scientific literature, it includes the anatomy, physiology and pathophysiology of the liver. Furthermore, it dives into the specifics of liver transplantations and recommended physiotherapy. The second section exhibits a complete case study of early post-operative physiotherapy on a patient after liver transplantation. The case study was conducted during a month of clinical practice at Institut Klinické a Experimentální Medicíny (IKEM) from the 10/01/2022 - 04/02/2022. Results: The patient significantly progressed throughout the physiotherapeutic care. Their overall physical condition improved, that led to the ability of independent verticalization and gait. 19. Evaluation of antibody quality for Endoglin detection in liver

The diploma thesis deals with endoglin, a transmembrane glycoprotein that can be cleaved to form so-called soluble endoglin, which then circulates in the blood. The theoretical part of the thesis is devoted to cardiovascular and liver diseases related to changes in the expression or level of endoglin in the blood and summarizes current knowledge about endoglin and its relation to various diseases. The experimental part of the thesis describes the selection of a suitable antibody against endoglin intended for western blot (WB). We have four anti-endoglin primary antibodies available from three manufacturers. The purpose was to test the antibodies and determine, which one had the best ability to detect endoglin in mouse tissue samples such as liver or aorta on WB. Affiblot was used to verify their ability to bind endoglin and also to compare their affinities to this protein. Newly developed patented dot-blot-based device. The binding of the used antibodies to endoglin was verified using the WB method and then the results of both methods were compared. The obtained results showed that the Abcam antibody has the most suitable parameters regarding specificity and sensitivity for the identification of endoglin in mouse tissue on WB. The liver is a very important parenchymatous organ for humans. Their function can be impaired by various influences. Common noxins include hepatotropic viruses and other microorganisms, alcohol and other toxins, including drugs, metabolic disorders, biliary obstruction, tumors, genetic abnormalities, and others. The work focuses on the comparison of histological images and clinical diagnosis of healthy individuals with steatosis and cirrhosis. The theoretical part describes the anatomy, physiology and histology of the liver with the perihepatic area. The next chapters focus on the clinic of steatosis and cirrhosis. The practical part describes the procedures for the preparation of healthy and affected liver preparations, as well as the histological methods used for staining individual cell structures. The result of comparing diagnoses of histological specimens and clinical diagnoses of individuals is 94.23 % agreement. Although the found discrepancy of diagnoses is small (5.77 %), it is a reason to think about the development and use of new or modified methods, including the creation of various calculators, including clinical data. Amphibians are undergoing large population declines in many regions around the world. As environmental pollution from both agricultural and urban sources has been implicated in such declines, there is a need for a biomonitoring approach to study potential impacts on this vulnerable class of organism. This study assessed the use of infrared (IR) spectroscopy as a tool to detect changes in several tissues (liver, muscle, kidney, heart and skin) of late-stage common frog (Rana temporaria) tadpoles collected from ponds with differing water quality. Small differences in spectral signatures were revealed between a rural agricultural pond and an urban pond receiving wastewater and landfill run-off; these were limited to the liver and heart, although large differences in body size were apparent, surprisingly with tadpoles from the urban site larger than those from the rural site. Large differences in liver spectra were found between tadpoles from the pesticide and nutrient impacted pond compared to the rural agricultural pond, particularly in regions associated with lipids. Liver mass and hepatosomatic indices were found to be significantly increased in tadpoles from the site impacted by pesticides and trace organic chemicals, suggestive of exposure to environmental contamination. Significant alterations were also found in muscle tissue between tadpoles from these two ponds in regions associated with glycogen, potentially indicative of a stress response. This study highlights the use of IR spectroscopy, a low-cost, rapid and reagent-free technique in the biomonitoring of a class of organisms susceptible to environmental degradation., V mnoha oblastech světa dochází k velkému poklesu populací obojživelníků. Jelikož se za příčinu tohoto poklesu považuje znečištění životního prostředí jak ze zemědělských, tak z městských zdrojů, existuje potřeba biomonitoringu sloužícího studiu potenciálních dopadů na tuto zranitelnou skupinu organismů. Tato práce posuzuje užití IČ spektroskopie jako nástroje detekce změn ve tkáních (játra, svaly, ledviny, srdce a kůže) pulců skokana hnědého (Rana temporaria) v pozdních fázích vývoje, kteří byly odebráni z vodních nádrží s různou kvalitou vody. Malé rozdíly byly shledány mezi nádržemi venkovského zemědělství a městskými nádržemi, do kterých přitékají odpadní vody a odtoky ze skládek; tyto rozdíly byly omezeny na játra a srdce, ačkoliv byly zjevné velké rozdíly ve velikosti pulců, překvapivě pulci z městských nádrží byli větší než pulci z venkovských nádrží. Větší rozdíly ve spektrech jater byly nalezeny mezi pulci z nádrží zatížených pesticidy a živinami ve srovnání s venkovskými nádržemi. Hmotnost jater a hepatosomatické indikátory byly významně zvýšené u pulců z nádrží znečištěných pesticidy a stopovými organickými chemikáliemi, což naznačuje vystavení kontaminaci životního prostředí.Významné změny byly též nalezeny ve svalových tkáních u pulců z těchto dvou nádrží, v částech spojených s glykogenem, což potenciálně indikuje reakci na stress. Tato práce vyzdvihuje využití IČ spektroskopie, levné a rychlé analytické techniky biomonitoringu třídy organismů ovlivněných degradací životního prostředí.

During the swadeshi agitation in Bengal the books presented in the performative space of Bengal became a source of instructing the public and for putting their mind and their imagination in order. The intellectual world of the Bengalis was in an uproar during this period due to the presence of a well-developed public sphere in Bengal. To legitimise these ideas as the social, moral and cultural authority of the popular bonding (the non-intellectuals of Gramscian analysis) attempts were made to create a homogeneous body of literature for the people. In this connection, the texts of popular performances like that of theatre, jatra and songs gained heightened importance. Texts of these popular media did not remain the mere framework of performance; they assumed the role of collecting and putting together of the stories and experiences of the people, establishing a degree of uniformity in it. In this article, I will look closely at the texts of the popular performative media of the age like theatre, jatra and song to gauge the process in which such media ordered the imagination of the people of Bengal in a homogeneous and seamless network of memory, laying the first step towards imagining a national community. Aims: This thesis aims to record physiotherapy care for the patient after undergoing ALPPS treatment for leiomyosarcoma, determination, management and recording of individual therapeutic units, and evaluation of their effect. Methods: The thesis consist of a theoretical and practical part. The theoretical part consist of general information about the composition and physiology of the liver. Leiomyosarcoma and other liver diseases are also described. Lastly, the ALPPS method is mentioned. All information is cited from academic literature and other professional sources. The practical part contains a case report of a patient, who has undergone ALPPS treatment for leiomyosarcoma. It also contains the record of individual therapies, that have been performed based on a kinesiological analysis. The analysis has been done as a part of a physiotherapy intership at the Institute of clinical and experimental medicine in Prague from 18th January to 12th February 2021. Bisphenol S (BPS) is an organic compound widely used in industrial production as a substitute for bisphenol A (BPA) for which negative impacts on the human body have been demonstrated and have therefore been significantly regulated. However, as time progresses, the negative properties of BPS are also shown, especially for the endocrine and reproductive systems. The liver is an organ in which xenobiotics are detoxified and thus BPS and BPA. The aim of the bachelor thesis was to verify whether 10-week oral exposure to BPS (4 g, 50 g, 100 mg/kg bw/day) and BPA (100 mg/kg bw/day) will lead to a negative impact on body weight, weight gain, weight and morphological liver structure and on the concentration of triglycerides, cholesterol, total white-vinas, urea and alkaline phosphatase activity in blood serum. The experiment involved 24 rats. BPA and BPS were administered daily by gastric tube in sunflower oil. The results of our research clearly did not show a negative effect of BPS and BPA in rats on body weight, absolute gain, absolute and relative liver weight in rats. There was also no evidence of histopathological changes in rat liver. In contrast, bio-chemical parameters were altered compared to the control group. Decreases in trigly-cerides, total protein content, urea, alkaline phosphatase activity and increases in serum cholesterol were noted. In the 50 g/kg body weight/day group of rats, the highest and lowest decreases in biochemical parameters were observed, respectively. The exposure dose of 100 mg BPS and BPA in our experiment did not cause liver histopathological changes or changes in biochemical parameters according to the hy-pothesis. A possible explanation could be given by the carrier for which sunflower oil with natural antioxidant content was used.

Methods: The bachelor thesis consists of two parts. The first part is the theoretical part, which summarizes basic information about anatomy, physiology and patology of the liver. The next chapter is about one of the main treatment options in advanced liver disease, what is the liver transplantation. Physiotherapeutic procedures that can improve patient's condition are mentioned in the last chapter of the theoretical part. The special part describes the case study of the patient after liver transplantation. It also includes the record of individual therapies, which was applied during the practice in IKEM in January 2020. Results: The patient improved after the therapies. "Vitamin D blood level, factors that affect it, and its relationship to reccurent infections" is topic repeatedly discussed in domestic and foreign literature. This topic is a multidisciplinary issue, but it mainly relates to nutrition and sun exposure. The aim of this study is to map relationship of vitamin blood levels in relation to diseases of the respiratory system, vitamin D suplementation, recurrent infections, usage of sunscreen with a protective factor, nutrition and outdoor stay in winter months. The study was conducted on a research sample of preschool children. Two sets of data were used to evaluate objectives and research questions. The first research group consisted of 48 children and had 2 groups. The first group of 21 children was established in the Waldorf kindergarten in Rudolfov and the second group of 17 children was created in a non-state medical facility in Trebon. The first research consisted of 48 children who were tested on vitamin D blood level and filled survey questionnaire. The second research consisted of 61 children who were tested on vitamin D blood level in a non-state medical facility. Some results of this study are contradictory because of inadequate methodology. Positive results have been shown with vitamin D supplementation, higher consumption of liver, eggs and daily intake of vegetable oils.

Use the contrast-enhanced ultrasound in the monitoring of the effect of liver tumors treatment by radiofrequency ablation. Summary High quality imaging is crucial for the treatment of liver tumors by the percutaneous radiofrequency ablation. Imaging methods are used for planning, navigation of the intervention and monitoring after treatment. An early detection of residual tumor tissue or recurrence affect significantly the quality of life and life expectancy of the patients. Contrast enhanced ultrasound (CEUS) seems to be a suitable method for monitoring the locoregional treatment mainly because zero radiation impact on patient and allows the highly accurate real-time assessment of vascularization. The aim of my work was to verify the ability of CEUS to detect residual tumor tissue or tumor recurrence in the field of changes after radiofrequency ablation. We evaluated the group of 73 patients, who were treated by radiofrequency ablation (RFA) for liver cancer. CEUS reached in our group the overal sensitivity 77.27 %, and in the group of colorectal cancer metastases 83.33 %. We compared CEUS with results of computed tomography (CT), which is the most commonly used method for monitoring the treatment. CEUS and CT results are similar. They are known as inherited hyperbilirubinemia characterized by an impairment in bilirubin metabolism. These genetic diseases are very rare with exception of Gilbert's syndrome. However, despite low prevalence and incidence it is necessary not to prolong their diagnosis. Most of them do not have any complications and do not require any treatment. 1 Abstract In this thesis I have focused on the role of lipids in the pathogenesis of liver diseases, specifically on cholestasis and non-alcoholic fatty liver disease (NAFLD). The first major aim was to clarify the changes in liver ganglioside metabolism in various types of cholestasis and to elucidate the role of heme oxygenase-1 (HMOX1) and associated oxidative stress. The second objective was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFA) administration on NAFLD development in a rodent dietary model of NAFLD and in patients with metabolic syndrome and NAFLD. Our results suggest that increased ganglioside biosynthesis and their re-distribution might represent a general protective mechanism of hepatocytes under cholestatic conditions (both estrogen-induced and obstructive aetiology). These changes are closely related to oxidative stress and might protect hepatocytes against deleterious effect of accumulated bile acids. The lack of HMOX1 activity and subsequent oxidative stress potentiate pathological changes in the liver and resulted in tissue-specific modulation of synthesis and re-distribution of gangliosides (in vivo and in vitro). The aim of this thesis was to evaluate the examined samples of liver tissue to find out the amount of aflatoxin in deer game. A total of 221 samples were examined for analysis purposes using the RIA method. Samples were obtained from dead and captured heads of deer game coming from private game reserves and game reserves belonging to the Forests of the Czech Republic. Samples of deer game were divided into groups according to their types, sex, and age. Sex was determined visually, and age via the development and grinding of the teeth. The aim of this thesis is to evaluate the influence of the seeds cakes of milk thistle (Silybum marianum L.) into feed fodder of laboratory rats. Milk thistle is one of the most famous and most frequently used herb positively influencing liver function. The aim was to verify the hypothesis that the seeds cakes of milk thistle in the feed fodder have influence on the biochemical parameters of blood. There were 24 male laboratory rats of Wistar albino species separated into 2 groups (A, B). Rats in the group A were fed with feed fodder with additional 10% of milk thistle seeds cakes. The control group (B) was fed with feed fodder without any additional milk thistle seeds cakes. The content of silimarin in the seeds cakes was 26,5mg/g. During the 30 days experiment average weight increase and biochemical values of blood was observed. Weight was measured on the 0. - 7. - 14. - 21. – 30. day of the experiment. Blood measurements were conducted by heart punction after killing 3 laboratory rats by inhalative anesthesia of Isofuran on 0. - 10. - 20. - 30. day of the experiment. The full blood sample was collected to determine biochemical blood parameters. There was no significant influence of milk thistle seeds cakes on the biochemical parameters of blood. Group fed with milk thistle seeds cakes showed higher activity of AST enzymes as well as higher concentration of HDL laboratory rats fed with milk thistle seeds cakes. Laboratory rats fed with milk thistle seeds cakes showed higher increase in daily weight measurements than the group of control. Results show that milk thistle seeds cakes have no significant influence on health indicators taken from the biochemical parameters of the blood.

Aims: The aim of this bachelor thesis is to summarize the theoretical knowledge about the issue of liver transplantation and to present an example of physiotherapeutic care on specific case of a patient after liver transplantation. Methodology: This thesis is divided into theoretical and professional part. The theoretical part contains information about the anatomy, physiology and pathologic physiology of the liver, liver transplantation and physiotherapy treatment after liver transplantation. These information were obtained from the scientific sources. Professional part was created as a research with case study during my professional experience in the Institute of Clinical and Experimental Medicine in Prague between 8 January 2018 and 2 February 2018, where I worked with a patient hospitalized after liver transplantation. Cílem je navrhnout počítačovou podporu, která pomůže chirurgům zvolit správnou léčebnou proceduru. Navrhnuté rozhraní bude realizováno formou webové stránky a bude zobrazitelné na různých platformách.

The term "biologically active compounds" covers a wide range of substances originating from natural or synthetic origins. These compounds can enter the aquatic environment through wastewater treatment plants, manufactural discharge or they are naturally present in the aquatic plants or microorganisms. Their adverse effects on fish has been widely studied and accepted. This thesis is focussed on the adverse effect of several bioactive compounds (i.e. pharmaceuticals, phytochemicals, or complex mixture of pollutants) on fish using different experimental design as in vitro, in vivo and in situ. In the first part of this thesis, in vitro effects of pharmaceuticals and phytochemicals or their combinations on fish CYP system were investigated. Moreover, effects of standard carrier solvents used in enzyme activity assays were also investigated. An important finding in this study was the lack of effects of either dexamethasone, quercetin, or indole-3-carbinol on EROD activity; however, when these agents were combined, EROD activity was strongly inhibited. This demonstrates that combination of compounds might exert different effects than single compounds, and the effects of mixture compounds cannot be predicted from the effects of individual compounds. In the second part of the thesis, the effects of chronic exposures of rainbow trout to dexamethasone and clotrimazole were investigated. The results regarding dexamethasone showed similarities with in vitro studies, and both in vitro and in vivo exposure of dexamethasone did not alter the CYP enzyme activities. On the other hand, in vivo exposure of clotrimazole yielded conflicting results with findings from the in vitro studies. Clotrimazole induced EROD activity in chronically exposed rainbow trout, and BFCOD activity showed biphasic pattern in which it was inhibited at environmentally relevant concentrations and induced at high concentrations. Thus, the observed effects suggest that clotrimazole could negatively affect fish CYPs at environmentally relevant concentrations. However, in the environment, the effects of clotrimazole and dexamethasone on fish CYPs system might be modified because of unknown compounds in these mixtures. Therefore, further investigations were done to identify the effects of mixture compounds using an in situ model. The last part of the dissertation addresses the effects of cocktail PPCPs on common carp under natural conditions. In situ studies provide valuable information on both hepatic and intestinal CYP activities. Both EROD and BFCOD activities were affected by the PPCPs that are present in the exposed fish. Moreover, changes in intestinal CYP activities suggest that fish can ingest some of these contaminants through their feed. Therefore, the intestines might be responsible for elimination of some of these pollutants and act as the first barrier of pollutant entry in fish. Despite the extensive studies concerning aquatic pollution, further studies are necessary. Development of new pharmaceuticals, their occurrence in the aquatic environment, and their effects on non-target organisms should be continuously monitored.

49. Stanovení termotolerantních druhů Campylobacter spp. v potravinách

Campylobacteriosis is according to European Food Safety Authority one of the most advanced worldwide foodborne disease causing gastroenteritis. The Czech republic ranks at number one in the highest incidence of campylobacteriosis. This thesis deals with the determination of thermotolerant Campylobacter spp. in samples of poultry and pork liver, in chicken and raw milk. From total number of 50 samples were 31 samples positive for Campylobacter spp. For poultry liver and chicken at the back of thighs were 92 % of positive samples. For pig liver was lower incidence, only 46 %. For fresh raw milk was the lowest incidence of just 17 %. Higher incidence in the Czech republic reflects good way of searching their subsequent reporting infections.

The consumption of dietary supplements such as flavonoids may reduce risk of many civilization diseases. Flavonoids are able to modulate the activity of cytochromes P450 (CYPs), xenobiotic-metabolising phase I enzymes of biotransformation that are involved in the activation and detoxification of food-derived carcinogens. Inhibition of CYP activities by flavonoids has been extensively studied because of their potential use as agents blocking the initiation stage of carcinogenesis. On the other hand, flavonoids have been shown to enhance the activation of carcinogens and/or influence their metabolism via induction of specific CYPs. In the first part of this study, flavonoids dihydromyricetin and α-napthoflavone were explored for their possible effects on CYP1A1 expression and activity when administered in combination with carcinogen benzo[a]pyrene (BaP). For this purpose, liver, small intestine and colon microsomal fractions were isolated from treated rats and induction of CYP1A1 was evaluated by immunodetection and EROD activity measurements. In liver and small intestine, all combinations of BaP and flavonoids led to strong induction of CYP1A1 expression.
