Insects are incredibly diverse, ubiquitous and have successfully flourished out of the dynamic and often unpredictable nature of evolutionary processes. The resident microbiome has accompanied the physical and biological adaptations that enable their continued survival and proliferation in a wide array of environments. The host insect and microbiome's bidirectional relationship exhibits their capability to influence each other's physiology, behavior and characteristics. Insects are reported to rely directly on the microbial community to break down complex food, adapt to nutrient-deficit environments, protect themselves from natural adversaries and control the expression of social behavior. High-throughput metagenomic approaches have enhanced the potential for determining the abundance, composition, diversity and functional activities of microbial fauna associated with insect hosts, enabling in-depth investigation into insect-microbe interactions. We undertook a review of some of the major advances in the field of metagenomics, focusing on insect-microbe interaction, diversity and composition of resident microbiota, the functional capability of endosymbionts and discussions on different symbiotic relationships. The review aims to be a valuable resource on insect gut symbiotic microbiota by providing a comprehensive understanding of how insect gut symbionts systematically perform a range of functions, viz., insecticide degradation, nutritional support and immune fitness. A thorough understanding of manipulating specific gut symbionts may aid in developing advanced insect-associated research to attain health and design strategies for pest management. Abstract Endosymbionts are known to have significant effects on their insect hosts, including nutrition, reproduction, and immunity. Insects gut microbiota is a critical component that affects their physiological and behavioral characteristics. The black cutworm (BCW), Agrotis ipsilon, is an economically important lepidopteran pest that has a diverse gut microbiome composed of nine species belonging to three phyla: Proteobacteria, Actinobacteria, and Firmicutes. This study was conducted to investigate the diversity of gut bacteria isolated from BCW larvae and moths and their effects on metabolism and pesticide degradation. The bacterial isolates were identified using the 16 S rRNA gene. The study showed that the gut microbiome composition significantly affected the metabolism of BCW larvae. Based on the screening results of synthesis of digestive enzymes and pesticide degradation, Brachybacterium conglomeratum and Glutamicibacter sp were selected to perform the remaining experiments as single isolates and consortium. The consortium-fed larvae showed high metabolic indices compared to antibiotic-fed larvae and the control. The gut bacteria were also shown to degrade three pesticide groups. Concerns regarding the health risk of chlorpyrifos have been raised due to its extensive use in agriculture. The isolated B. conglomeratum was more effective in chlorpyrifos degradation than the consortium. Furthermore, the study also examined the presence of sex related endosymbionts (Wolbachia, Spiroplasma, and Rickettsia) in the reproductive tissues of adults. The outcomes demonstrated that none of the examined endosymbionts existed. In conclusion, the study highlights the importance of the gut microbiome in insect physiology and behavior and its potential applications in biotechnology. Development of resistance to insecticides in insects is a global concern. Understanding the mechanisms is critical for effective plant protection and human health. Challenge in dealing the matter is that, resistance can occur via direct organism response (metabolic, physiological, and target-site changes) or via gut microbiome. Insects are constantly evolving like any other organisms; they are adopting various measures to overcome the chemicals sprayed to control them. Increasing evidence suggest that the gut microbiome can promote pesticide resistance in pests. Possible mechanisms by which gut bacteria play role in insecticide resistance are, direct acquisition of pesticide degrading microbes from the environment, difference in gut bacterial composition and diversity, difference in xenobiotic degrading enzymes and presence of microbial xenobiotic degradation pathways and cross-acclimatization to related insecticides. An investigation was carried out to isolate and characterize the culturable gut bacterial fauna of three stem borers attacking rice (Scirpophaga incertulus, Sesamia inferens and Chilo suppressalis) morphologically, biochemically and genetically (based on 16S rDNA sequences). Their ability to degrade two commonly used insecticides, i.e., chlorpyrifos and chlorantraniliprole was quantified in vitro through minimal media study using GC–MS and HPLC, respectively. There was a great variation in the gut bacterial composition of these three borers in spite of belonging to same guild and being collected from the same host (Varshadhan: CRLC-899) at the same time from ICAR-NRRI farm, Cuttack, India. Bacillus was prominent in S. incertulus and C. suppressalis whereas Stenotrophomonas was dominant in S. inferens. It is the first report of encountering B. pumilus, B. cereus and Acinetobacter calcoaceticus from the gut of S. incertulus and Paraclostridium from the gut of S. inferens. Chlorpyrifos and chlorantraniliprole was degraded to the tune of 44.87–92.02 % and 10.52 to 74.38 %, respectively. B. subtilis most efficiently degraded chlorpyrifos, while Klebsiella sp. and A. calcoaceticus were superior in degrading Chlorantraniliprole. The gut bacterial consortium of S. inferens was the most efficient in degrading the insecticides. The differential abilities of the gut bacterial isolates to degrade the insecticides provide insights for their possible involvement in xenobiotic detoxification in their host and further study on this aspect may improve management strategies of these pests. The cotton bollworm Helicoverpa armigera occurs as a major pest in many economically important crops, including cotton, pigeon pea, chickpea, pea, cowpea, sunflower, tomato, sorghum, pearl millet and other crops. Intestinal microorganisms play important role in the degradation of diet components of insects. In order to know the role of gut bacteria in insecticide resistance five insecticides Chlorpyriphos (20% EC), Cypermethrin (25% EC), Malathion (50% EC), Quinalphos (25% EC), Triazophos (40% EC), were selected for the insecticide degradation studies. All the bacterial isolates from the gut of lab and field populations of H. armigera were identified using 16S rRNA gene-based identification and tested for their growth on minimal salt medium (MSM) along with the selected insecticides. A total of 11 bacterial isolates were tested and among them, isolate CL4 (Rhodococcus sp.) was found to grow on minimal salt medium (MSM) and with chlorpyriphos and isolate CL2 (Enterococcus casseliflavus) was able to grow in MSM with chloropyriphos (C22H19Cl2NO3) and malathion (C10H19O6PS2) and no growth was seen in MSM without insecticide (control). Gas Chromatography analysis of the positive bacterial isolate cultures in MSM showed that the isolate CL4 (Rhodococcus sp.) was able to utilize 43.9% of chlorpyriphos and isolate CL2 (E.casseliflavus) was able to utilize 26% of chlorpyriphos and 57.1% of malathion in MSM broth cultures with comparison with the respective control cultures. In this study, a novel cold-adapted family Ⅳ esterase EstDR4 was identified and obtained from extremophile Deinococcus radiodurans (D. radiodurans). EstDR4 displayed significant substrate preference towards short and medium chain monoesters (C2–C12). It also showed regioselectivity, enantioselectivity and degradation effects on four insecticides. The optimum temperature and pH for EstDR4 activity were 30 °C and pH 8, respectively. Additionally, EstDR4 exhibited relatively high catalytic activity at 0 °C and high stability from 10–40 °C, with over 80% of its initial activity retained after 1 h of incubation. Moreover, EstDR4 activity was stimulated by Tween 80 and Triton X-100, and inhibited by metal ions such as Co2+, Cu2+ and Zn2+ and several organic solvents. A novel application of maghemite nanoparticles embedded activated carbon (γ-Fe 2 O 3 @AC), as a mesoporous heterojunction composite, coupled with UV light was used for enhanced activation of peroxymonosulfate (PMS) for degradation of cyfluthrin insecticide. The performance of γ-Fe 2 O 3 @AC/UV/PMS system was assessed as affected by different concentrations of PMS, catalyst and pollutant, pH of aqueous media, reaction time, co-exiting water matrix components and trapping agents. A tentative mechanism was proposed for the PMS decomposition and reactive species production in both solid and liquid phases. Catalytic activity of γ-Fe 2 O 3 @AC in the PMS decomposition was enhanced significantly when it coupled with UV light. In optimal conditions (pH 4.0, PMS 4.0 mM, 0.4 g/L catalyst, C 0 = 60 mg/L and time 80 min), the efficiency rates of 88.5 and 52.4% were achieved for degradation and mineralization of cyfluthrin, respectively. The degradation rate affected by matrix anions was reduced in order of HCO 3 − > NO 3 − > Cl− > SO 4 2−. Cycling tests displayed the negligible iron leaching and high recyclability for γ-Fe 2 O 3 @AC within 5 times use. A strong synergistic effect was observed between applied techniquess in the activation of PMS. Pseudo-first-order kinetic model represented a significant correlation with the experimental data of degradation process. Among detected free radicals, SO 4 •− species had a conquer role in catalytic oxidative degradation process. γ-Fe 2 O 3 @AC/UV/PMS system showed an excellent performance in treatment of simulated real water and wastewater samples. Generally, coupling of γ-Fe 2 O 3 @AC and UV for improving PMS activation can be introduced as a successful and efficient approach to catalytic degradation of organic matters owing to tremendous catalytic activity, easy recovery, high stability and reusability potential and the generation of various reactive free species simultaneously. Gamma irradiation (60Co) in postharvest of common beans (Phaseolus vulgaris L.), has already proven beneficial, regarding the prolongation of shelf life and physical, chemical, and organoleptic aspects of grains. However, few prior studies have investigated the effect of gamma irradiation on the dissipation of pesticides in foods, especially insecticides in beans. This study aimed to evaluate the waste dissipation of two insecticides, imidacloprid and thiamethoxam, used in the cultivation of common bean, a variety of Carioca, using gamma irradiation. Ground bean samples were spiked each insecticide in the laboratory at 3, 4, and 5 mg/kg, and were then irradiated at 0, 1, and 2 kGy per irradiator with a 60Co source. Liquid chromatography‐tandem mass spectrometry was used to quantify the insecticides. The irradiation promoted dissipation of thiamethoxam at 1 and 2 kGy doses, with the highest dissipation verified at 2 kGy. In general, at this higher dose of irradiation the reduction dissipation of thiamethoxam was 66% of the mean value found in the treatment without irradiation and ∼44% relative to the 1 kGy dose. Conversely, imidacloprid showed no effect of irradiation on waste dissipation, except in the treatment of 5 mg/kg at 2 kGy dose. From the results, it was concluded that the gamma irradiation might have applicability in dissipating thiamethoxam in real bean samples. Practical Application: Pesticide residues remaining from bean cultivation, sensitive to optimal doses of gamma irradiation, can be transformed into other less harmful or nonharmful substances, increasing the quality of the beans before the food arrives at the consumer's table. Irradiation of the grains after harvest also eliminates the need for application of other pesticides to preserve the grains during storage, as the technique is capable of eradicating deteriorating agents, such as microorganisms and insects. The presence of insecticide residues in cereals represents a risk for the consumer, because these substances are used in large scale to protect stored grains from the attack of pests. The effects of three processing methods for corn and wheat samples on the stability of stored-grain protective insecticides were here evaluated. Fenitrothion and esfenvalerate were applied so as to produce theoretical concentrations of 10 and 0.5 mg kg-1, respectively. Two hours after treatment, the grains were processed and deposition was analyzed by gas chromatography. Grain species did not influence insecticide stability. This stability was only dependent upon the processing method and insecticide type. Grains processed together with dry ice provided the greatest percentage of recovery for both insecticides. Regardless of the processing method, more esfenvalerate than fenitrothion was recovered, thus demonstrating the greater stability of the pyrethroid during this operation.A presença de resíduos de inseticida em amostras de cereais representa um risco ao consumidor, pois estas substâncias são empregadas em larga escala na preservação de grãos armazenados. Neste sentido, o efeito de três métodos de processamento de amostras de milho e trigo na estabilidade de inseticidas protetores de grãos armazenados foram avaliados. O fenitrotion e o esfenvalerato foram aplicados de modo a produzir concentração teórica de 10 e 0,5 mg kg-1, respectivamente. Duas horas após o tratamento, os grãos foram processados e os depósitos analisados mediante cromatografia gasosa. A espécie de grão não teve influência na estabilidade dos inseticidas, sendo que essa estabilidade apenas dependeu do método de processamento e do inseticida. Os grãos processados junto com gelo seco proporcionaram a maior porcentagem de recuperação de ambos os inseticidas. Independente do método de processamento utilizado, conseguiu-se recuperar mais esfenvalerado do que fenitrotion, demonstrando-se assim, a maior estabilidade do piretróide durante esta operação.

BACKGROUND: Neonicotinoid insecticides are generally efficacious against many turfgrass pests, including several important phloem-feeding insects. However, inconsistencies in control of western chinch bugs, Blissus occiduus, have been documented in field efficacy studies. This research investigated the efficacy of three neonicotinoid insecticides (clothianidin, imidacloprid and thiamethoxam) against B. occiduus in buffalograss under field conditions and detected statistically significant differences in B. occiduus numbers among treatments. A subsequent study documented the relative quantity and degradation rate of these insecticides in buffalograss systemic leaf tissues, using HPLC. RESULTS: Neonicotinoid insecticides initially provided significant reductions in B. occiduus numbers, but mortality diminished over the course of the field studies. Furthermore, while all three neonicotinoids were present in the assayed buffalograss leaf tissues, imidacloprid concentrations were significantly higher than those of clothianidin and thiamethoxam. Over the course of the 28 day study, thiamethoxam concentrations declined 700-fold, whereas imidacloprid and clothianidin declined only 70-fold and 60-fold respectively. CONCLUSIONS: Field studies continued to verify inconsistencies in B. occiduus control with neonicotinoid insecticides. This is the first study to document the relative concentrations of topically applied neonicotinoid insecticides in buffalograss systemic leaf tissues. The effect of environmental parameters (temperature and relative humidity) on the degradation rate of azinphos methyl and parathion methyl was studied. Proprietary emulsifiable concentrates were diluted and added to each of 90 glass Petri dishes for each pesticide and were left overnight to dry. Petri dishes were placed in 18 air-tight containers (9 for each pesticide) in which were created environments with relative humidity (RH) of 60, 82, and 96%. The containers were stored at 0, 20, and 40°C. From the experimental results best fit curves, kinetic equations, rate constants, and half-lives were calculated. Half-lives of azinphos methyl for the RH studied were, from 124 to 267 days at 0°C, from 89 to 231 days at 20°C, and from 25 to 71 days at 40°C. Corresponding half-lives for parathion methyl were from 48 to 57 days at 0°C, from 9.2 to 10.5 days at 20°C and from 1.3 to 1.5 days at 40°C. The results were correlated with relevant results from the decomposition of the same or similar pesticides on apples both, on the trees and during refrigerated storage. These correlations are suggesting that biological factors strongly affected the decomposition rate of azinphos methyl. Degradation of parathion methyl on lemon and apple on the trees, during storage of these fruits in refrigerated rooms, and during juice production was studied. The pesticide was applied to the trees according to the recommended application procedures. Apples and lemons received a single application of parathion methyl at 40 g active ingredient/100 litres. Residues were determined with a simple gas-chromatographic method; the recovery rate of parathion methyl from apples and lemons was 85-108% and the limit of determination was 0.004 mg kg[SUP-1]. The half-lives of parathion methyl on the trees were 7 days for apples and 5 days for lemons. The duration of storage of the fruits in a refrigerated room have been largely extended to 65 and 63 days, respectively. The high acidity of lemons was found not to affect the degradation rate of parathion methyl. One of the four glutathione- S-transferases (GST) that is overproduced in the insecticide-resistant Cornell-R strain of the housefly ( Musca domestica) produces an activity that degrades the insecticide dimethyl parathion and conjugates glutathione to lindane. In earlier work, it was shown that the resistant Cornell-R carries an amplification, probably a duplication, of one or more of its GST loci and that this amplification is directly related to resistance. Using polymerase chain reaction (PCR) amplification with genomic DNA, multiple copies of the gene encoding the parathion-degrading activity (called MdGst-3) were subcloned from both the ancestral, insecticide-susceptible strain BPM and from the insecticide-resistant Cornell-R. In BPM, three different MdGst-3 genes were identified while in Cornell-R, 12 different MdGst-3 sequences were found that, though closely related to ancestral genes, had diverged by a few nucleotides. This diversity in MdGst-3 genomic sequences in Cornell-R is reflected in the expressed sequences, as sampled through a cDNA bank. Population heterozygosity cannot account for these multiple GST genes. We suggest that selection for resistance to insecticides has resulted in not only amplification of the MdGst-3 genes but also in the divergence of sequence between the amplified copies. The improvement of a semiconductor thin film preparation technique is considered to be the primary strategy for photoelectrocatalytic technique development. This research study's focus was to develop an electrodeposition technique for tungsten oxide (WO 3) thin film fabrication to improve photoelectrocatalytic activities for water oxidation and insecticide degradation application. WO 3 thin film deposition on conductive glass was developed using the electrochemical amperometric and cyclic voltammetric methods to optimize the essential parameters that affect the characteristics and photoelectrocatalytic properties of the fabricated WO 3 electrode. Characteristics such as the visible light absorption, morphology, crystalline structure, and photoelectrocatalytic oxidation activities of the fabricated WO 3 electrode were studied and compared with both electrodeposition methods. The fabricated WO 3 electrode was found to improve photoelectrocatalytic water oxidation more with the amperometric method than with the cyclic voltammetric method and the traditional spin coating method, up to 4 times and 60 times, respectively. Photoelectrocatalytic enhancement is clarified by the effect of visible light absorption, roughness morphology, crystallinity, and charge transfer rate improvement. We can approve the efficacy of the developed WO 3 electrode for insecticide removal under the photoelectrocatalytic mechanism. Standing water from carbofuran-treated Azolla plots showing accelerated degradation was further enriched by five repeated transfers to carbofuran-supplemented mineral salts medium. This enrichment culture developed from standing water of carbofuran-treated Azolla plot can utilise carbofuran as sole source of carbon and nitrogen. The enrichment culture was able to hydrolyse nearly 100% of [ring-14C]carbofuran to carbofuran phenol in five days, which accumulated in themedium, while the carbamate side-chain in [carbonyl-14C]carbofuran was readily mineralized to [14C]carbon dioxide.
