Polyphenols (PP) found in brown algae are known for their wide range of biological activities including noteworthy antitumor properties. This article presents a method for obtaining an active polyphenolic extract from the Arctic alga Fucus vesiculosus with 98% purity and radical scavenging activity equivalent to 862 mg of ascorbic acid per gram of extract. Immunostimulant effects of polyphenols were assessed in vitro using venous blood from two groups of people: healthy people (HP) and people with chronic undifferentiated lymphocytic leukemia (LP). Polyphenols activated the surface properties of immunocompetent cells. Specifically, polyphenols dose-dependently increased the percentage of cells' spreading and adhesion by 2–3 times. Additionally, polyphenols increased the number of activated lymphocytes in the LP blood to levels characteristic of HP. Given their natural origin, high activity, non-toxicity, and straightforward production process, these studied polyphenols exhibit immense potential for use as new pharmaceuticals or as active components with immunostimulatory effects. Accumulating evidence suggests that high consumption of natural antioxidants promotes health by reducing oxidative stress and, thus, the risk of developing cardiovascular diseases. Similarly, fermentation of natural compounds with lactic acid bacteria (LAB), such as Lactiplantibacillus plantarum, enhances their beneficial properties as regulators of the immune, digestive, and cardiovascular system. We investigated the effects of fermentation with Lactiplantibacillus plantarum on the antioxidant and immunomodulatory effects of Pushgay berries (Vaccinium floribundum, Ericaceae family) in human umbilical vein endothelial cells (HUVECs) and macrophage cell line RAW264.7. Polyphenol content was assayed by Folin–Ciocalteu and HPLC-MS/MS analysis. The effects of berries solutions on cell viability or proliferation were assessed by WST8 (2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium, monosodium salt and Lactate dehydrogenase (LDH) release, Trypan blue exclusion test, and Alamar blue assay. Antioxidant activity was evaluated by a cell-based chemiluminescent probe for the detection of intracellular H2O2 production in HUVECs. Heme oxygenase-1 (HO-1) expression levels were investigated by RT-qPCR. Glutathione reductase (GR), glutathione peroxidase (Gpx), superoxide dismutase (SOD), and catalase (CAT) activities, as markers of intracellular antioxidant defense, were evaluated by spectrophotometric analysis. The immunomodulatory activity was examined in RAW 264.7 by quantification of inducible nitric oxide synthase (iNOS) and Tumor Necrosis Factor—alpha (TNFα) by RT-qPCR. Data showed that fermentation of Pushgay berries (i) enhances the content of quercetin aglycone, and (ii) increases their intracellular antioxidant activity, as indicated by the reduction in H2O2-induced cell death and the decrease in H2O2-induced HO-1 gene expression in HUVECs treated for 24 h with fermented berries solution (10 µg/mL). Moreover, treatment with Pushgay berries for 72 h (10 µg/mL) promotes cells growth in RAW 264.7, and only fermented Pushgay berries increase the expression of iNOS in the same cell line. Muramyl dipeptide (MDP) is the smallest peptidoglycan fragment able to trigger the immune response. Structural modification of MDP can lead to the preparation of analogs with improved immunostimulant properties, including desmuramyl peptides (DMPs). The aim of this work was to prepare the desmuramyl peptide (L-Ala-D-Glu)-containing adamantyl-triazole moiety and its mannosylated derivative in order to study their immunomodulatory activities in vivo. The adjuvant activity of the prepared compounds was evaluated in a murine model using ovalbumin as an antigen, and compared to the reference adjuvant ManAdDMP. The results showed that the introduction of the lipophilic adamantyl-triazole moiety at the C-terminus of L-Ala-D-Glu contributes to the immunostimulant activity of DMP, and that mannosylation of DMP modified with adamantyl-triazole causes the amplification of its immunostimulant activity.

Background: Bergenia species contain various therapeutically important compounds such as phenolic compounds-arbutin, bergenin, tannins, gallic acid, flavonoids, minerals, and many others. Bergenia plants show antibacterial, antiviral, anticancer, antidiabetic, diuretic and immunostimulant activities. Materials and Methods: Bergenia leaf extracts from Bergenia crassifolia, Bergenia ciliata, and Bergenia x ornata on lymphocyte activation using flow cytometry method were investigated. Their activation was then monitored with the help of the increasing fluorescence intensity. Results: By activating of cells, there has been an increase in the amount of the established antibodies. Bergenia extracts significantly stimulated the expression of CD69 on lymphocytes in the final concentration of 3.13 and 6.25 mg/ml. The values of stimulation indices of B. crassifolia and B. x ornata extract significantly statistically did not differ. Purpose: To investigate the immunomodulating activity of Mangifera indica. (MI) and Curcuma domestica (CD) extracts in mice. Methods: Mice were randomly divided into 4 groups, namely, vehicle, untreated, MI and CD groups (n = 6). They were treated with MI (160 mg/kg) or CD (200 mg/kg) extracts, and vehicle for control group, for 14 days and sacrificed on day 15. In innate immunity test, the mice were challenged with sheep red blood cells (SRBCs) antigen on day 8, while in adaptive immunity test, mice were immunized and challenged on days 7 and 14, respectively, with SRBC. White blood cells count (WBC), spleen index (SI), and haemagglutination (HA) titer, and delayed type hypersensitivity (DTH) responses were determined. Results: For both plant extracts, adaptive immunity groups showed the highest response compared to innate groups. In adaptive immunity, the WBC count of MI and CD treated animals was significantly higher than in the untreated and vehicle treated groups (p < 0.001). Moreover, the SI of mice from MI and CD treated groups differed significantly that of the untreated group (p < 0.01 and p < 0.05, respectively). HA titer in CD (both non-challenged and challenged) groups was significantly higher than in the non-challenged vehicle group (p < 0.001). HA titer in MI group (non-challenged) was significantly lower than in non-challenged CD and challenged groups (p < 0.01 and p < 0.001, respectively). Conclusion: MI and CD extracts, in appropriate doses, exerted immunostimulant effects in mice by enhancing both innate and adaptive immune systems via increase in WBC, SI, HA titer and DTH responses. Background: Dietary supplements of Carica papaya Linn fermented with yeast using a biotechnological process have well recognized positive effects on immunological, hematological, inflammatory, and oxidative stress parameters, utilized as biomarkers of chronic and degenerative diseases. Although many natural products fermented with lactic acid bacteria (LAB) have shown beneficial effects on the immune system and on antioxidant defenses, formulations of papaya fermented with LAB have not yet been studied. Aims: The aims of this study were to investigate the immunomodulatory activity linked to the type of papaya fermentation (yeast vs LAB) in macrophages and to evaluate whether the type of fermentation differently modulates oxidative stress both in cell free system and in a model of Methods: Cytotoxicity was evaluated through cell proliferation kinetic and lactate dehydrogenase release assays; immunomodulatory activity through the transcriptional activation of inducible nitric oxide synthase (iNOS) and Tumor Necrosis Factor α (TNFα) by qRT-PCR in RAW 264.7 macrophages; antioxidant capacity was assessed, in cell free system and in pheochromocytoma cells embryonic brain cells, by measuring the intracellular ROS levels through a fluorescent dye. Results: Our data showed that all the formulations studied are safe at low concentrations (3-6 mg/ml); the LAB-fermented formulations promoted the expression of iNOS and TNFα more efficiently than yeast-fermented papaya preparation (p <0.001). In a cell free system, the LABfermented formulation acted as mild scavengers of ROS while, in cells, both formulations didn't show any significant effect. Conclusions: This study corroborates previous reports showing the efficacy of yeast fermented papaya as a potent immunostimulant and highlights the beneficial contribution of lactic bacteria fermentation. Filifolinone is a semi-synthetic terpenoid derivative obtained from Heliotropium filifolium that increases the expression level of pro-inflammatory and anti-inflammatory cytokines in kidney cells of salmon. Because cytokines are produced in response to a foreign organism and by distinct other signals modulating immune responses, we further studied the potential immunomodulatory effects of a group of structural related terpenoid derivatives from H. filifolium on salmonids to determine the relationship between the chemical structure of the derivatives and their ability to modify cytokine expression and the lymphoid content. The resin and four 3H-spiro 1-benzofuran-2,1′-cyclohexane derivatives were tested in vivo in rainbow trout ( Oncorhynchus mykiss ) by quantifying the transcript levels of antiviral and T helper-type cytokines and T and B cells in the kidney. Three of the four terpenoids differ only in the C-7′substituent of the cyclohexane and the presence of the ketone group at this position in Filifolinone appeared responsible of an important up-regulation of IFN-α1, IFN-γ, IL-4/13A and IL-17D in the kidney of the treated trout. In addition, the absence of a methoxy group in carbon 7 of the benzene ring, found in all compounds but not in Folifolinoic acid, produced a significant reduction of IFN-γ, IL-12 and IL-4/13A transcripts. B cells were not affected by the compound treatment but Filifolinoic acid and the resin induced a significant reduction of T cells. Altogether, results showed that immunomodulating responses observed in the trout by effect of 3H-spiro 1-benzofuran-2,1′-cyclohexane derivatives is related to the presence of the ketone group in the carbon 7′ and the methoxy group in carbon 7 of the benzene ring, being Filifolinone the most active immunostimulatory compound identified. Abstract: Ethnopharmacological relevance: Barleria prionitis Linn. (Family: Acanthaceae), one of the important Ayurvedic medicinal plant in India, has long been used to treat variety of ailments including swellings, gout, arthritic and rheumatic disorders, nervine and skin diseases, and also acts as immunorestorative. Aim of the study: The present study was aimed to explore in vitro and in vivo immunomodulatory activities of the iridoids fraction i.e. n-butanol fraction of methanol extract from Barleria prionitis aerial parts (IFBp). Materials and methods: IFBp was studied for in vitro [nitroblue tetrazolium (NBT) test and neutrophils candidacidal assay] and in vivo immunomodulatory activity on cellular and humoral immune responses to the antigenic challenge by sheep red blood cells (SRBCs) and by neutrophil adhesion test, phagocytic activity and cyclophosphamide-induced myelosuppression. The study comprised the preliminary phytochemical screening, HPTLC standardization and maximum tolerable dose determination of IFBp. Results: IFBp (50, 100 and 200μg/ml) significantly (P ≤0.01) increased the intracellular killing activity of stimulated neutrophils assayed by in vitro NBT reduction test and neutrophils candidacidal assay. Pretreatment of IFBp (100 and 200mg/kg; p.o.) evoked a significant increase in percent neutrophils and neutrophils adhesion to nylon fibres. Oral administration of IFBp augmented the humoral immune response to SRBCs, evidenced by increase in antibody titres and dose dependently potentiated the delayed-type hypersensitivity reaction induced by SRBCs in mice. IFBp potentiated significantly (P ≤0.01) the macrophage phagocytic activity and ameliorated the red blood cells, total white blood cells and platelets count and haemoglobin concentration, and also restored the myelosuppressive effects induced by cyclophosphamide. The content (% w/w; mean±SD, n =3) of main iridoids i.e. shanzhiside methyl ester and barlerin was found to be 21.55±2.40 and 10.03±1.69 in IFBp of BP, respectively. Conclusion: The present investigation reveals that IFBp is a potent immunostimulant, stimulating both the specific and non-specific immune mechanisms. [Copyright &y& Elsevier]

Immunostimulant activities from methanol extract or fractions and isolated compounds of Zingiber cassumunar rhizome were evaluated. The test was performed by in vitro assay of Microscopic Smear Test using macrophage cells from mouse peritoneum and Staphylococcus epidermidis as antigen. The result was measured on the phagocytosis potential and percentage activity of the phagocytosis. The extract ethyl acetate fraction showed the greatest immonostimulat activity, produced by three phenylbutanoids. One of them was a significant immunostimulant [(E)-4-(3',4'-dimethoxyiphenyl) but-3-en-l-ol (1)], the other two compounds [(E)-4-(2',4', 5'-trimethoxyphenyl)-3-en-l-ol (2) and (E)-4-13',4'-dimethoxyphenyl)-3-en-l-methoxy-l-ol (3)] were less effective. Abstract: This study examined the effects of oral administration of an enzymatic protein hydrolysate from green microalga Chlorella vulgaris (Cv-PH) on the recovery of both innate and specific immune responses of undernourished Balb/c mice after a 3-day fasting period. Cv-PH was prepared by hydrolysis of ethanol-extracted cell biomass with pancreatin (20AU/g) at pH 7.5 and 45°C for 4h. The treatment with Cv-PH (500mg/kg) for 8 days provided benefits in terms of haemopoiesis, as judged by the recovery of bone marrow cellularity and the leukocyte counts in peripheral blood, particularly the lymphocyte pool, which increased up to 128% compared to control animals. Starved mice treated with Cv-PH showed a higher number of peritoneal exudate cells and the macrophage activation was demonstrated by the enhancement in glucose consumption and acid phosphatase activities relative to non-supplemented mice. The increased carbon clearance in peripheral blood suggested the stimulation of mononuclear phagocytic system. Cv-PH also stimulated both humoral and cell mediated immune functions positively, such as T-dependent antibody response and the reconstitution of delayed-type hypersensitivity response (DTH). These findings indicate that Chlorella protein hydrolysate can be used for developing physiologically functional foods with immunopotentiating activity. [Copyright &y& Elsevier]
