UBE3A is an E3 ubiquitin ligase encoded by an imprinted gene whose maternal deletion or duplication leads to distinct neurodevelopment disorders Angelman and Dup15q syndromes. Despite the known genetic basis of disease, how changes in copy number of a ubiquitin ligase gene can have widespread impact in early brain development is poorly understood. Previous studies have identified a wide array of UBE3A functions, interaction partners, and ubiquitin targets, but no central pathway fully explains its critical role in neurodevelopment. Here, we review recent UBE3A studies that have begun to investigate mechanistic, cellular pathways and the genome-wide impacts of alterations in UBE3A expression levels to gain broader insight into how UBE3A affects the developing brain. These studies have revealed that UBE3A is a multifunctional protein with important nuclear and cytoplasmic regulatory functions that impact proteasome function, Wnt signaling, circadian rhythms, imprinted gene networks, and chromatin. Synaptic functions of UBE3A interact with light exposures and mTOR signaling and are most critical in GABAergic neurons. Understanding the genome-wide influences of UBE3A will help uncover its role in early brain development and ultimately lead to identification of key therapeutic targets for UBE3A-related neurodevelopmental disorders.

Phosphoinositides, the seven phosphorylated derivatives of phosphatidylinositol have emerged as regulators of key sub-cellular processes such as membrane transport, cytoskeletal function and plasma membrane signaling in eukaryotic cells. All of these processes are also present in the cells that constitute the nervous system of animals and in this setting too, these are likely to tune key aspects of cell biology in relation to the unique structure and function of neurons. Phosphoinositides metabolism and function are mediated by enzymes and proteins that are conserved in evolution, and analysis of knockouts of these in animal models implicate this signaling system in neural function. Most recently, with the advent of human genome analysis, mutations in genes encoding components of the phosphoinositide signaling pathway have been implicated in human diseases although the cell biological basis of disease phenotypes in many cases remains unclear. In this review we evaluate existing evidence for the involvement of phosphoinositide signaling in human nervous system diseases and discuss ways of enhancing our understanding of the role of this pathway in the human nervous system’s function in health and disease.

This article describes my experiences over more than 50 years in initiating and maintaining research on human genetics and genomics at the National Research Centre in Cairo, Egypt, from its beginnings in a small unit of human genetics to the creation of the Center of Excellence for Human Genetics. This was also the subject of a lecture I gave at the 10th Conference of the African Society of Human Genetics, held in Cairo in November 2017, after which Professor Michèle Ramsay, president of the society, suggested that I write an autobiographical article for the Annual Review of Genomics and Human Genetics. I hope that I succeeded in the difficult assignment of summarizing the efforts of a researcher from a developing country to initiate and maintain the rapidly advancing science of human genetics and genomics in my own country and make contributions to the worldwide scientific community. Highlights from the article: I In each family, it is always the same mtDNA haplotype that segregates across generations and contributes to the observed mtDNA heteroplasmy i . The long-range PCR amplification employed by the authors would successfully amplify from both genomes, resulting in sequence reads from both the mtDNA molecules as well as the postulated full length mtDNA concatemer (Figure 1). Moreover, catenated mtDNA structures have been reported in normal human cardiac tissue (Pohjoismäki et al., [16]), and mtDNA concatemers that were presumed to be a product of rolling circle amplification have been observed in human fibroblasts exposed to reactive oxygen species (Ling et al., [8]).

Genomics and molecular biology has added substantial methods and knowledge to nearly all fields of biology and medicine. In this review we try to demonstrate how genomics and molecular biology is also on the way to have a profound impact on behavioral anthropology, evolutionary psychology, evolutionary sociology and bio-sociology. We propose that particularly studies on selection and adaptation will be influenced profoundly by genomics, for instance via identification of the partially genetic basis of human behavior by candidate gene studies and by genome wide association studies. In addition, epigenetics will lead to a deeper study of the interaction of the genetic basis of a behavior and its environmental regulation. We argue that the genomic revolution is much more than merely a new methodological approach, but will change our concepts of human behavior and its development in the evolution of homo.
