Polyphagous aphids often consist of host‐specialized lineages, which have greater fitness on their native hosts than on others. The underlying causes are important for understanding of the evolution of diet breadth and host shift of aphids. The cotton‐melon aphid Aphis gossypii Glover is extremely polyphagous with many strict host‐specialized lineages. Whether and how the lineage specialized on the primary host hibiscus shifts to the secondary host cucumber remains elusive. We found that the hibiscus‐specialized lineage suffered high mortality and gave birth to very few nymphs developing into yellow dwarfs on fresh cucumber leaves, and did not inflict any damage symptoms on cucumber plants. The poor performance did not improve with prolonged exposure to cucumber; however, it did significantly improve when the cucumber leaves were pre‐infected with a biotrophic phytopathogen Pseudoperonospora cubensis. More importantly, the hibiscus‐specialized lineage with two‐generation feeding experience on pre‐infected cucumber leaves performed as well as the cucumber‐specialized lineage did on fresh cucumber leaves, and inflicted typical damage symptoms on intact cucumber plants. Electrical penetration graph (EPG) indicated that the hibiscus‐specialized lineage did not ingest phloem sap from fresh cucumber leaves but succeeded in ingesting phloem sap from pre‐infected cucumber leaves, which explained the performance improvement of the hibiscus‐specialized lineage on pre‐infected cucumber leaves. This study revealed a new pathway for the hibiscus‐specialized lineage to quickly acclimate to cucumber under the assistance of the phytopathogen. We considered that the short feeding experience on pre‐infected cucumber may activate expression of effector genes that are related to specific host utilization. We suggest to identify host‐specific effectors by comparing proteomes or/and transcriptomes of the hibiscus‐specialized lineage before and after acclimating to cucumber., The hibiscus‐specialized Aphis gossypii cannot use cucumber as host plant but performed well on cucumber that has been pre‐infected with the phytopathogen Pseudoperonospora cubensis; a short feeding experience on the pre‐infected cucumber enables the hibiscus‐specialized A. gossypii to fully adapt to fresh cucumber.

Microbial symbionts enable many phytophagous insects to specialize on plant-based diets through a range of metabolic services. Pollen comprises one-plant tissue consumed by such herbivores. While rich in lipids and proteins, its nutrient content is often imbalanced and difficult-to-access due to a digestibly recalcitrant cell wall. Pollen quality can be further degraded by harmful allelochemicals. To identify microbes that may aid in palynivory, we performed cDNA-based 16S rRNA metabarcoding on three related pollen beetles (Nitidulidae: Meligethinae) exhibiting different dietary breadths: Brassicogethes aeneus, B. matronalis, and Meligethes atratus. Nine bacterial symbionts (i.e., 97% OTUs) exhibited high metabolic activity during active feeding. Subsequent PCR surveys revealed varying prevalence of those from three Rickettsialles genera— Lariskella, Rickettsia, and Wolbachia—within beetle populations. Our findings lay the groundwork for future studies on the influence of phylogeny and diet on palynivorous insect microbiomes, and roles of symbionts in the use of challenging diets.

The comma butterfly (Polygonia c-album, Nymphalidae, Lepidoptera) is a model insect species, most notably in the study of phenotypic plasticity and plant-insect coevolutionary interactions. In order to facilitate the integration of genomic tools with a diverse body of ecological and evolutionary research, we assembled the genome of a Swedish comma using 10X sequencing, scaffolding with matepair data, genome polishing, and assignment to linkage groups using a high-density linkage map. The resulting genome is 373 Mb in size, with a scaffold N50 of 11.7 Mb and contig N50 of 11,2Mb. The genome contained 90.1% of single-copy Lepidopteran orthologs in a BUSCO analysis of 5,286 genes. A total of 21,004 gene-models were annotated on the genome using RNA-Seq data from larval and adult tissue in combination with proteins from the Arthropoda database, resulting in a high-quality annotation for which functional annotations were generated. We further documented the quality of the chromosomal assembly via synteny assessment with Melitaea cinxia. The resulting annotated, chromosome-level genome will provide an important resource for investigating coevolutionary dynamics and comparative analyses in Lepidoptera.

During the last two decades, ecological speciation has been a major research theme in evolutionary biology. Ecological speciation occurs when reproductive isolation between populations evolves as a result of niche differentiation. Phytophagous insects represent model systems for the study of this evolutionary process. The host-plants on which these insects feed and often spend parts of their life cycle constitute ideal agents of divergent selection for these organisms. Adaptation to feeding on different host-plant species can potentially lead to ecological specialization of populations and subsequent speciation. This process is thought to have given birth to the astonishing diversity of phytophagous insects and is often put forward in macroevolutionary scenarios of insect diversification. Consequently, numerous phylogenetic studies on phytophagous insects have aimed at testing whether speciation driven by host-plant adaptation is the main pathway for the diversification of the groups under investigation. The increasing availability of comprehensive and well-resolved phylogenies and the recent developments in phylogenetic comparative methods are offering an unprecedented opportunity to test hypotheses on insect diversification at a macroevolutionary scale, in a robust phylogenetic framework. Our purpose here is to review the contribution of phylogenetic analyses to investigate the importance of plant-mediated speciation in the diversification of phytophagous insects and to present suggestions for future developments in this field., 35 pages, 3 figures, 2 tables. Using a total of 14,043 trap-nests (potential nest for bees), we documented the nest architecture, host plants, seasonality,and associated organisms of the following six native species of Anthidium (Megachilidae: Anthidiini) from the CentralMonte Desert in Mendoza, Argentina: A. andinum Jo¨ rgensen; A. chubuti Cockerell; A. decaspilum Moure; A. friesei Cockerell;A. rubripes Friese; and A. vigintipunctatum Friese. Each species exhibited unique features in the arrangement of thefibers used to make their cells, and in the type of material used in the nest plug. Bees used a total of 41 plant species,but relied heavily on Prosopis flexuosa (Fabaceae) and Larrea spp. (Zygophyllaceae). Most bee species are univoltine andexhibited delayed emergence. The oil beetle Nemognatha sp. (Coleoptera: Meloidae) and the cuckoo wasp Chrysis striatulaBohart (Hymenoptera: Chrysididae) are recorded for the first time for Anthidium. Studying bee nests can enlighten our understanding of feeding specialization and phylogenetic relationships of bees. We studied the nesting and feeding habits of Trichothurgus laticeps in the Monte desert ecosystem. Our results show that T. laticeps is attracted to pre-existing cavities in wood (trap nests), which were further excavated for nest construction; nest cells are unlined and provisioned almost exclusively with pollen of the cactus Opuntia sulphurea. The species overwinters inside a hard cocoon. Our results reinforce the hypothesis previously proposed by other authors that the unworked pollen mass and oligolecty are plesiomorphic characters in the Megachilidae, and that the wood-boring behavior for nest construction indicates convergent evolution with the Xylocopinae. Understanding the process of adaptation of parasite populations to their host-plant is a key issue in evolutionary ecology. It is also a major subject in applied research that has implications for crop protection. The oomycete Plasmopara viticola, the causal agent of downy mildew, attacks the species of the Vitis genus. In a context where the main concern of the breeding programs is the durability of resistance, new knowledge about the ecology and evolution of the interaction between parasite and host is needed in order to evaluate the potential of downy mildew to overcome the resistance. In my thesis, I addressed the role of the host-plant as an evolutionary factor for downy mildew populations, by asking this question at two different evolutionary scales: (i) in the pathogen region of origin (North America) I assessed the degree of specialization of the parasite on its wild and cultivated host range (ii) in Europe, where downy mildew has been introduced recently, I studied the evolution of downy mildew populations subject to the selection pressure imposed by resistant grapevine varieties. To understand the host-plant specialization in this pathosystem, where several cryptic species have been identified, we performed cross inoculations between different host (Vitis spp.) and pathogen (P. viticola) species. Morphological and phenotypic data provide evidence of host-plant specialization in P. viticola populations: downy mildew species A and D are specialized on their host-plant, while the specialization process is ongoing for species B and C. Although no genetic differentiation has been shown inside species C, there are two distinct groups within species B. Isolates from the cultivated compartment are on average more aggressive than isolates from wild vines, indicating an adaptation of isolates growing on cultivated host-plants. Finally, a large-scale study of the distribution of downy mildew species on both their wild and cultivated host-plants resulted in the identification of a new cryptic species and confirmed the host-plant specialization. In Europe, our results show that the limited deployment of resistant varieties has led to changes in downy mildew populations: emergence of virulent isolates (i.e. breakdown of a major QTL for resistance), and increased aggressiveness on Vitis vinifera. In order to understand the mechanisms at the origin of specialization and resistance breakdown, we examined the parasite’s effector repertoire. Over one hundred effector candidates were identified using available data on the P. viticola genome. The polymorphism of 32 candidate genes revealed that three of them evolve under positive selection. Our results reveal the strong ability of downy mildew to adapt to its host plant and to plant resistance. They should be taken into account when devising strategies for the deployment of grapevine resistances in order to guarantee their durability.; La compréhension du processus d’adaptation des populations de parasites à leur plante-hôte est une question fondamentale en écologie évolutive. C’est également un enjeu majeur de recherche finalisée qui a des retombées pour la protection des cultures. L’oomycète Plasmopara viticola, agent causal du mildiou de la vigne, attaque les espèces du genre Vitis. Dans un contexte où l’enjeu principal des programmes d’amélioration est la durabilité des résistances, des connaissances nouvelles sur l’écologie et l’évolution de l'interaction entre le parasite et son hôte sont nécessaires afin d’évaluer le potentiel du mildiou à surmonter ces résistances. Dans ma thèse, je me suis intéressée au rôle de la plante-hôte comme facteur d’évolution des populations de mildiou, en posant cette question à différentes échelles évolutives : (i) dans le bassin d’origine du pathogène (Amérique du Nord), j’ai cherché à évaluer le degré de spécialisation du parasite sur sa gamme d’hôtes sauvages et cultivés; (ii) en Europe, où le mildiou de la vigne a été introduit récemment, j’ai étudié l’évolution des populations de mildiou soumis à la pression de sélection des résistances des nouvelles variétés de vigne. Pour comprendre la spécialisation plante-hôte dans ce pathosystème où plusieurs espèces cryptiques ont été identifiées, nous avons réalisé des tests d’inoculations croisées entre espèces hôtes (Vitis spp.) et agent pathogène (P. viticola). Les données phénotypiques et morphologiques apportent les preuves d’une spécialisation plante-hôte au sein des populations de P. viticola : les espèces A et D de mildiou sont spécialisées sur leur plante-hôte, tandis que le processus de spécialisation est en cours pour les espèces B et C. Même si aucune différenciation génétique n’a été montrée au sein de l’espèce C, il existe deux groupes distincts au sein de l’espèce B. Les isolats du compartiment cultivé sont en moyenne plus agressifs que les isolats issus des vignes sauvages, indiquant une adaptation des isolats cultivés sur leur plante hôte. A partir d’un large échantillonnage, nous avons étudié la distribution des espèces de mildiou sur leurs plantes-hôtes sauvages et cultivées. Ce travail a permis d’identifier une nouvelle espèce cryptique et a confirmé la spécialisation plante-hôte. En Europe, nos résultats montrent que le déploiement limité de variétés à résistantes partielles a conduit à des modifications des populations de mildiou: apparition d’isolats virulents (i.e. contournant un QTL majeur de résistance), et augmentation de l’agressivité sur Vitis vinifera. Dans le but de comprendre les mécanismes à l’origine de la spécialisation et du contournement des résistances, nous nous sommes intéressés au répertoire d’effecteurs du parasite. Une centaine d’effecteurs candidats ont été identifiés en utilisant les données disponibles sur le génome de P. viticola. L’analyse du polymorphisme de 32 candidats sur une sélection d’isolats montre que trois d’entre eux évoluent sous sélection positive. Ces résultats soulignent l’importance de la plante-hôte comme facteur de diversification des populations de l’agent pathogène et révèlent que le mildiou s’adapte rapidement aux résistances de la vigne. Il est désormais nécessaire de mieux appréhender le déploiement des résistances de la vigne afin qu’elles puissent être durables.

Divergent adaptation to host plant species may be the major mechanism driving speciation and adaptive radiations in phytophagous insects. Host plants can differ intrinsically in a number of attributes, but the role of natural enemies in host plant specialization is often underappreciated. Here, we report behavioural divergence between the European corn borer (ECB, Ostrinia nubilalis ) and its sibling species Ostrinia scapulalis , in relation to a major enemy: humans. Harvesting maize imposes selective mortality on Ostrinia larvae: those located above the cut-off line of the stalk face almost certain death. We show that ECB larvae diapause closer to the ground than those of O. scapulalis , which is sympatric but feeds mainly on weeds. The difference in diapause height results from genetically determined differences in geotactic behaviour. ECB larvae descend towards the ground specifically at harvest time, increasing their chances of surviving harvesting by about 50 per cent over O. scapulalis larvae. Natural enemies appear as a major driver of host-plant specialization in this example, stressing the need to consider ‘tri-trophic’ ecological niches to understand insect diversification. Our results also strongly suggest that geotaxis evolved as a singular instance of behavioural resistance in a major agricultural pest.

The study of intraspecific variation with respect to host plant utilization in polyphagous insects is crucial for understanding evolutionary patterns of insect-plant interactions. Aphis gossypii (Glover) is a cosmopolitan and extremely polyphagous aphid species. If host plant species or families constitute selective regimes to these aphids, genetic differentiation and host associated adaptation may occur. In this study, we describe the genetic structure of A. gossypii collected in six localities in Tunisia on different vegetable crops, on citrus trees and on Hibiscus. The aim was to determine if the aphid populations are structured in relation to the host plants and if such differentiation is consistent among localities. The genetic variability of A. gossypii samples was examined at eight microsatellite loci. We identified only 11 multilocus genotypes among 559 individuals. Significant deviations from Hardy-Weinberg equilibrium, linkage disequilibria and absence of recombinant genotypes, confirmed that A. gossypii reproduces by continuous apomictic parthenogenesis.
