Abstract Objective Breast cancer is one of the most common cancers in women worldwide. This study aims is to investigate the burden of breast cancer in Iran and its relationship with the Human Development Index (HDI) during 2009 to 2019. Study design The present study is an observational study in Iran during the years 2009 to 2019. Data related to the incidence, mortality, Years of Life with Disability (YLD), Years of Life Lost (YLL) and Disability-Adjusted Life-Years (DALY) of breast cancer in Iran were extracted from the Global Burden of Disease 2019 (GBD-2019) website. Correlation tests are used to check the relationship between these indicators and the human development index. Conclusions: Considering that a positive correlation was observed between the incidence and burden of breast cancer and the human development index in the country, it is suggested to implement preventive measures such as public education programs to reduce the incidence and burden of breast cancer and the necessity of screening programs in areas with low human development index. We conducted an EMS mutagenesis screen on chromosome arm 2L to identify recessive suppressors of GMR-hid-induced apoptosis in the Drosophila eye. Through this screen, we recovered three alleles of the lysine demethylase gene Kdm5. Kdm5, a member of the JmjC-domain-containing protein family, possesses histone demethylase activity towards H3K4me3. Our data suggest that Kdm5 specifically regulates Hid-induced cell death during development, as we did not observe control of Reaper- or Grim-induced cell death by Kdm5. Interestingly, GMR-hid-induced apoptosis is suppressed independently of Kdm5’s demethylase activity. Our findings indicate that Rbf and dMyc are necessary for Kdm5 mosaics to suppress GMR-hid-induced cell death. Moreover, Kdm5 mosaics failed to suppress apoptosis induced by a mutant form of Hid that is resistant to inhibition by Erk-type MAPK activity. Additionally, Kdm5 dominantly enhances the wing phenotype of an activated MAPK mutant. These results collectively suggest that Kdm5 controls Hid-induced apoptosis by regulating the Rbf, dMyc, and MAPK pathways.

Badly burned skeletal remains are commonly submitted to forensic laboratories for victim identification via DNA analysis methods. Burned skeletal remains present many challenges for DNA analysis as they can contain low amounts of DNA which can also be damaged and degraded, resulting in partial or no STR profiles. Therefore, a simple, but effective screening method that identifies which samples may provide the most successful STR or mtDNA typing results for identification would enable forensic laboratories to save time, money, and resources. One metric that can be used and a screening method is the color of burned bone, as bone color changes with exposure to fire as temperature and length of exposure increase. This research developed a quantitative screening method based on the surface color of burned bone. The different visual bone colors (light brown, dark brown, black, gray, and white) were quantified using the Commission on Illumination L*a*b color space. These values were then compared to DNA yield, STR, and mtDNA profile completeness to identify whether the L*a*b values can predict genotyping success. A Bayesian network was constructed to determine the probability of STR typing success, given a set of L*a*b values. Results demonstrated that samples with an a* value greater than or equal to one and b* value greater than eight (light brown and dark brown burned samples) were the most predictive of STR typing success and mtDNA typing success. This paper reports the results of experiments and studies involving different types of devices that can implement a BadUSB scenario, for example, BadUSB, Rubber Ducky, which, when connected to a computer, impersonate a device with a Human Interface Device, emulating other devices such as a keyboard and mouse. Given the problem of the lack of management tools for detecting preliminary modifications of USB devices against attacks based on the seizure of computer control, a software and hardware system is proposed as an object of study. It is implemented programmatically in the Arduino IDE environment, and physically it is made on the Arduino Mega board with Shield, which reads the parameters of the devices. It monitors the startup of USB devices and checks each device for pre-retrofitting by passing HID descriptors from the connected hardware. Having parsed the data using Python, the data are represented in the appropriate form for analysis, on the basis of which a decision is made by the system on the possible preliminary modification of the USB drive from which these data came. This is due to the detailed consideration and thorough analysis of data, data types, temporal characteristics of data transmitted along different channels. The technical characteristics and functionality of USB devices were investigated; the parameters transmitted at the moment when they are supplied with power were determined. The system can draw a conclusion based on the analysis according to its algorithm and block a suspicious USB device that has been connected and that can intercept control over the computer. The results of the study could be used in the field of protection of information systems from attacks based on the seizure of control from external media. The designed solution increases the level of security of the system, making it possible to recognize a possibly pre-modified device at the connection stage. [ABSTRACT FROM AUTHOR]

Worldwide, road-users complain about glare from oncoming cars' headlights, often blaming new light sources such as light-emitting diodes (LEDs). However, drivers' own behavior and attitudes might contribute to the issue of glare. The aim of this research was to establish the prevalence of on-road glare and its associations with drivers' attitudes in a sample of Czech drivers. We used an online survey and gained responses from 539 passenger-car-drivers. The majority experienced glare at least once a week or almost daily, identifying mainly white- or bluish-colored headlights as the source of glare. However, about 60% of participants would prefer the view of the road that is provided by these sources, and an association was found between the preferred part of the lighting color spectrum and the perceived glare source color. A large proportion of drivers were also not aware of the specifics of the lighting sources that they used. Nowadays, information obtained from mobile phones is often the subject of evidence in front of a court. Forensic analysts often come across smartphones about which they have no prior information. However, they need to extract data from them. The main prerequisite to extract the data is to bypass Android lock screen protection. The HID attack is a promising method to break Android lock screen protection. In many cases, this is the only way how to break the smartphone's nonbiometric lock screen protections on newer Android OS versions. The article contains examples of three non-biometric types of Android smartphone lock screen protections and their computational complexity. Together with sickle (skl), the Drosophila paralogs reaper (rpr), head involution defective (hid), and grim (RHG) control a critical switch in the induction of programmed cell death. RHG homologs have been identified in other dipteran and lepidopteran species but not beyond. Revisiting this issue with a “taxon hopping” BLAST search strategy in current genome and transcriptome resources, I detected high confidence RHG homologs in Coleoptera, Hymenoptera, Hemiptera, and Dictyoptera. Analyses of gene structure and protein sequence conservation revealed aconserved splicing pattern and highly conserved amino acid residues at both the N- and C-terminal ends that identify hid as the most ancestrally organized RHG gene family member in Drosophila. hid-like RHG homologs were also detected in mosquitoes, redefining their michelob_x (mx) genes as an expansion of derived RHG homologs. Only singleton homologs were detected in the large majority of other insect clades. Lepidopteran RHG homologs, however, stand out by producing an evolutionarily-derived splice isoform, identified in previous work, in addition to the newly detected hid-like isoform. Exceptional sequence diversification of select RHG homologs at the family- and genus-level explain their previous elusiveness in important insect genome model species like the red flour beetle Tribolium castaneum and the pea aphid Acyrthosiphon pisum. Combined, these findings expand the minimal age of the RHG gene family by about 100 million years and open new avenues for molecular cell death studies in insects.

Cyberattacks exploiting Universal Serial Bus (USB) interfaces may have a high impact on individual and corporate systems. The BadUSB is an attack where a USB device’s firmware is spoofed and, once mounted, allows attackers to execute a set of malicious actions in a target system. The countermeasures against this type of attack can be grouped into two strategies: phyiscal blocking of USB ports and software blocking. This paper proposes a distributed architecture that uses software blocking to enhance system protection against BadUSB attacks. This architecture is composed of multiple agents and external databases, and it is designed for personal or corporate computers using Microsoft Windows Operating System. When a USB device is connected, the agent inspects the device, provides filtered information about its functionality and presents a threat assessment to the user, based on all previous user choices stored in external databases. By providing valuable information to the user, and also threat assessments from multiple users, the proposed distributed architecture improves system protection.

This paper presents a low-cost microcontroller-based data acquisition device. The key component of the presented solution is a configurable microcontroller-based device with an integrated USB transceiver and a 12-bit analogue-to-digital converter (ADC). The presented embedded DAQ device contains a preloaded program (firmware) that enables easy acquisition and generation of analogue and digital signals and data transfer between the device and the application running on a PC via USB bus. This device has been developed as a USB human interface device (HID). This USB class is natively supported by most of the operating systems and therefore any installation of additional USB drivers is unnecessary. The input/output peripheral of the presented device is not static but rather flexible, and could be easily configured to customised needs without changing the firmware. When using the developed configuration utility, a majority of chip pins can be configured as analogue input, digital input/output, PWM output or one of the SPI lines. In addition, LabVIEW drivers have been developed for this device. When using the developed drivers, data acquisition and signal processing algorithms as well as graphical user interface (GUI), can easily be developed using a well-known, industry proven, block oriented LabVIEW programming environment.

Almost 6 million people died in Poland during the Nazi occupation and about 570 thousand during the Soviet occupation. But the end of the war was not the end of the trauma. Historians estimate that at least 30 thousand people were killed during the Stalinist regime in Poland. In 2012 the Institute of National Remembrance started to search for hidden burials of victims of communism. Many exhumations were carried out under the project. One of them took place in Białystok, eastern Poland. According to information gathered by local historians, a detention centre in the heart of city was the place of secret burials of victims of the communist regime. During the exhumation work a burial pit with the remains of 24 victims was found. It's characteristics supported the hypothesis that these people were shot on the spot, in a mass execution during the Nazi occupation. Historians knew of only one such execution, but its victims - according to the available records - were supposed to have been exhumed at the end of the war. Exhumation works and the discovery of the discussed mass grave put in question the events of 1944, which would have been impossible without the field work. The first identifications confirmed the doubts of historians, since both the results of genetic profiling and the conducted anthropological analysis revealed that at the end of the war a mistake was made, and bodies other than those suspected had been exhumed. Although the mechanisms of apoptotic cell death have been well studied in the fruit fly, Drosophila melanogaster , it is unclear whether such mechanisms are conserved in other distantly related species. Using degenerate primers and PCR, we cloned a proapoptotic gene homologous to Head involution defective ( Hid ) from the Scuttle fly, Megaselia scalaris ( MsHid ). MsHid cDNA encodes a 197-amino acid-long polypeptide, which so far is the smallest HID protein. PCR analyses revealed that the MsHid gene consists of four exons and three introns. Ectopic expression of MsHid in various peptidergic neurons and non-neuronal tissues in Drosophila effectively induced apoptosis of these cells. However, deletion of either conserved domain, N-terminal IBM or C-terminal MTS, abolished the apoptogenic activity of MsHID, indicating that these two domains are indispensable. Expression of MsHid was found in all life stages, but more prominently in embryos and pupae. MsHid is actively expressed in the central nervous system (CNS), indicating its important role in CNS development. Together MsHID is likely to be an important cell death inducer during embryonic and post-embryonic development in this species. In addition, we found 2-fold induction of MsHid expression in UV-irradiated embryos, indicating a possible role for MsHid in UV-induced apoptosis. [ABSTRACT FROM AUTHOR]

Las características ácido-base de la γ-alúmina, soporte de catalizadores sulfurados CoMo/g-Al2O3, fueron modificadas por la adición de boro (2, 3, 5, y 8% B2O3) y potasio (1, 3 y 5% K). Estos catalizadores fueron ensayados en reacciones simultáneas de hidrogenación (HID) de olefinas (2,4,4 trimetil-1-penteno y 2,4,4 trimetil-2-penteno; 3:1) e hidrodesulfuración (HDS) de 2-metiltiofeno, simulando el hidrotratamiento (HDT) de una nafta de craqueo o de FCC (Fluidized Catalytic Cracking). Los resultados mostraron que aunque los cambios inducidos en las características ácido-base del soporte de alúmina disminuyeron la actividad para la HID y la HDS de los catalizadores, también dieron lugar a cambios apreciables en la selectividad hacia la HDS de tiofenos relativa a la HID de olefinas, lo que los convierte en buenos candidatos para el HDT de la nafta de FCC. Por otro lado, el craqueo, la alquilación y la isomerización de metilos se favorecieron cuando se hicieron las modificaciones con B, lo mismo que la migración del doble enlace de las olefinas ramificadas cuando se hizo la modificación con K. Los resultados de este trabajo permiten concluir que la actividad y selectividad del catalizador CoMo/γ-Al2O3 para la HDS de la nafta de FCC, puede ser controlada por una adecuada modificación de las propiedades ácido-base del soporte de alúmina mediante la adición de pequeñas cantidades de B y de K.

As the result of the communist terror in Poland, during years 1944–1956 more than 50,000 people died. Their bodies were buried secretly, and most places are still unknown. The research presents the results of identification of people buried in one of many mass graves, which were found at the cemetery Powązki Military in Warsaw, Poland. Exhumation revealed the remains of eight people, among which seven were identified genetically. Well‐preserved molars were used for the study. Reference material was collected from the closest living relatives. In one case, an exhumation of victim's parents had to be performed. DNA from swabs was extracted with a PrepFiler®BTA Forensic DNA Extraction Kit and organic method. Autosomal, Y‐STR amplification, and mtDNA sequencing were performed. The biostatistical calculations resulted in LR values from 1608 to 928 × 1018. So far, remains of more than 50 victims were identified. [ABSTRACT FROM AUTHOR]

Dendritic arbor morphology is a key determinant of neuronal function. Once established, dendrite branching patterns must be maintained as the animal develops to ensure receptive field coverage. The translational repressors Nanos (Nos) and Pumilio (Pum) are required to maintain dendrite growth and branching of Drosophila larval class IV dendritic arborization (da) neurons, but their specific regulatory role remains unknown. We show that Nos-Pum-mediated repression of the pro-apoptotic gene head involution defective (hid) is required to maintain a balance of dendritic growth and retraction in class IV da neurons and that upregulation of hid results in decreased branching because of an increase in caspase activity. The temporal requirement for nos correlates with an ecdysone-triggered switch in sensitivity to apoptotic stimuli that occurs during the mid-L3 transition. We find that hid is required during pupariation for caspase-dependent pruning of class IV da neurons and that Nos and Pum delay pruning. Together, these results suggest that Nos and Pum provide a crucial neuroprotective regulatory layer to ensure that neurons behave appropriately in response to developmental cues. The identification of unknown victims is one of the most challenging tasks faced by forensic medicine. This is due to the rapid decomposition of tissues, beginning at the moment of death and caused by released enzymes and microbial activity. Decay is directly associated with the decomposition of soft tissues and also the degradation of genetic material inside cells. Decomposition rates vary depending on a number of environmental factors, including temperature, humidity, season, and soil properties. Decomposition also differs between bodies left in the open air or buried. To date, forensic medicine has identified mainly people who were the victims of various types of criminal offences. However, with advances in identification methods, increasingly frequent attempts are made to identify the victims of armed conflicts, crimes of totalitarian regimes, or genocide. The aim of the study was to compare three different methods for the extraction of nuclear DNA from material considered in forensic medicine as difficult to handle, i.e. fragments of bones and teeth, and to determine the performance of these methods and their suitability for identification procedures. Histone modulations have been implicated in various cellular and developmental processes where in Drosophila Mof is involved in acetylation of H4K16. Reduction in the size of larval imaginal discs is observed in the null mutants of mof with increased apoptosis. Deficiency involving Hid, Reaper and Grim [H99] alleviated mof induced apoptosis in the eye discs. mof induced apoptosis leads to activation of caspases which is suppressed by over expression of caspase inhibitors like P35 and Diap1clearly depicting the role of caspases in programmed cell death. Also apoptosis induced by knockdown of mof is rescued by JNK mutants of bsk and tak1 indicating the role of JNK in mof induced apoptosis. The adult eye ablation phenotype produced by ectopic expression of Hid, Rpr and Grim, was restored by over expression of Mof. Accumulation of Mof at the Diap1 promoter 800 bp upstream of the transcription start site in wild type larvae is significantly higher (up to twofolds) compared to mof mutants. This enrichment coincides with modification of histone H4K16Ac indicating an induction of direct transcriptional up regulation of Diap1 by Mof. Based on these results we propose that apoptosis triggered by mof proceeds through a caspase-dependent and JNK mediated pathway. [ABSTRACT FROM AUTHOR]

Allele distribution and forensic parameters were estimated for 15 STR loci (AmpF l STR Identifiler kit) in 251 Mexican-Mestizos from the state of Guerrero (South, Mexico). Genotype distribution was in agreement with Hardy–Weinberg expectations for all 15 STRs. Similarly, linkage disequilibrium test demonstrated no association between pair of loci. The power of exclusion and power of discrimination values were 99.999634444% and >99.99999999%, respectively. Genetic relationship analysis regarding Mestizo populations from the main geographic regions of Mexico suggests that the Center and the present South regions conform one population cluster, separated from the Southeast and Northwest regions. Eiger, the sole Drosophila TNF-alpha homolog, causes ectopic apoptosis through JNK pathway activation. Yet, its role in developmental apoptosis remains unclear. eiger mutant flies are viable and fertile but display compromised elimination of oncogenic cells and extracellular bacteria. Here we show that Eiger, specifically expressed in embryonic neurons and glia, is not involved in developmental neuronal apoptosis or in apoptotic cell clearance. Instead, we provide evidence that Eiger is required for damage-induced apoptosis in the embryonic CNS through regulation of the pro-apoptotic gene hid independently of the JNK pathway. High voltage gain dc-dc converters are required in many industrial applications such as photovoltaic and fuel cell energy systems, high-intensity discharge lamp (HID), dc back-up energy systems, and electric vehicles. This paper presents a novel input-parallel output-series boost converter with dual coupled inductors and a voltage multiplier module. On the one hand, the primary windings of two coupled inductors are connected in parallel to share the input current and reduce the current ripple at the input. On the other hand, the proposed converter inherits the merits of interleaved series-connected output capacitors for high voltage gain, low output voltage ripple, and low switch voltage stress. Moreover, the secondary sides of two coupled inductors are connected in series to a regenerative capacitor by a diode for extending the voltage gain and balancing the primary-parallel currents. In addition, the active switches are turned on at zero current and the reverse recovery problem of diodes is alleviated by reasonable leakage inductances of the coupled inductors. Besides, the energy of leakage inductances can be recycled. A prototype circuit rated 500-W output power is implemented in the laboratory, and the experimental results shows satisfactory agreement with the theoretical analysis. [ABSTRACT FROM AUTHOR]

Industrial petrochemical facilities have predominantly used high-intensity discharge light sources and installation practices unchanged for several decades. Today, new light-emitting diode (LED)-based products are being developed, and there appear to be many advantages to this technology; however, caution is advised in its application. This paper examines LED lighting technology, standards, and available products along with design and application considerations. Moreover, presented are case studies of LED luminaire retrofits in existing facilities. This paper discusses the results of the studies as they relate to safety, environment impacts, capital and operating costs, design implications, and installation improvements. Drosophila lin52 (dlin52) is a member of Myb transcription regulator complex and it shows a dynamic pattern of expression in all Drosophila tissues. Myb complex functions to activate or repress transcription in a site-specific manner; however, the detailed mechanism is yet to be clearly understood. Members of the Drosophila melanogaster Myb-MuvB/dREAM complex have been known to regulate expression of a wide range of genes including those involved in regulating apoptosis. E2F and its corepressor RBF also belong to this complex and together they regulate expression of genes involved in cell cycle progression, apoptosis, differentiation, and development. In the present study, we examined whether the depletion of dlin52 in developing photoreceptor neurons results in enhanced apoptosis and disorganisation of the ommatidia. Strikingly, we found that dLin52 is essential for transcriptional repression of the pro-apoptotic gene, hid; decrease in dlin52 levels led to dramatic induction of hid and apoptosis in eye-antennal discs. Reduction of Rpd3 (HDAC1), another member of the dREAM complex, also led to marginal upregulation of Hid. In addition, we also demonstrated that an optimum level of dLin52 is needed for dE2F1/2 activity on the hid promoter. dlin52 cooperates with dRBF and dE2F1/2 for recruitment of repressor complex on the hid promoter. Preliminary data indicate that Rpd3/HDAC1 also contributes to hid repression. Based on the findings, we conclude that dLin52 functions as a co-factor and modulates activity of members of dMyb/dREAM complex at hid promoter, thus regulating apoptosis by repressing this pro-apoptotic gene in the developing Drosophila eye. The input/output peripheral of the presented device is not static but rather flexible, and could be easily configured to customised needs without changing the firmware. When using the developed configuration utility, a majority of chip pins can be configured as analogue input, digital input/output, PWM output or one of the SPI lines. In addition, LabVIEW drivers have been developed for this device. When using the developed drivers, data acquisition and signal processing algorithms as well as graphical user interface (GUI), can easily be developed using a well-known, industry proven, block oriented LabVIEW programming environment. Abstract: The development of the Drosophila leg is a good model to study processes of pattern formation, cell death and segmentation. Such processes require the coordinate activity of different genes and signaling pathways that progressively subdivide the leg territory into smaller domains. One of the main pathways needed for leg development is the Notch pathway, required for determining the proximo-distal axis of the leg and for the formation of the joints that separate different leg segments. The mechanisms required to coordinate such events are largely unknown. We describe here that the zinc finger homeodomain-2 (zfh-2) gene is highly expressed in cells that will form the leg joints and needed to establish a correct size and pattern in the distal leg. There is an early requirement of zfh-2 to establish the correct proximo-distal axis, but zfh-2 is also needed at late third instar to form the joint between the fourth and fifth tarsal segments. The expression of zfh-2 requires Notch activity but zfh-2 is necessary, in turn, to activate Notch targets such as Enhancer of split and big brain. zfh-2 is controlled by the Drosophila activator protein 2 gene and regulates the late expression of tarsal-less. In the absence of zfh-2 many cells ectopically express the pro-apoptotic gene head involution defective, activate caspase-3 and are positive for acridine orange, indicating they undergo apoptosis. Organizing and annotating biomedical data in structured ways has gained much interest and focus in the last 30 years. Driven by decreases in digital storage costs and advances in genetics sequencing, imaging, electronic data collection, and microarray technologies, data is being collected at an alarming rate. The need to store and exchange data in meaningful ways in support of data analysis, hypothesis testing and future collaborative use is pervasive. Because trans-disciplinary projects rely on effective use of data from many domains, there is a genuine interest in informatics community on how best to store and combine this data while maintaining a high level of data quality and. The difficulties in sharing and combining raw data become amplified after post-processing and/or data analysis in which the new dataset of interest is a function of the original data and may have been collected by multiple collaborating sites. Simple meta-data, documenting which subject and version of data were used for a particular analysis, becomes complicated by the heterogeneity of the collecting sites yet is critically important to the interpretation and reuse of derived results. This manuscript will present a case study of using the XML-Based Clinical Experiment Data Exchange (XCEDE) schema and the Human Imaging Database (HID) in the Biomedical Informatics Research Network’s (BIRN) distributed environment to document and exchange derived data. The discussion includes an overview of the data structures used in both the XML and the database representations, insight into the design considerations, and the extensibility of the design to support additional analysis streams.

Drosophila Toll-1 and all mammalian Toll-like receptors regulate innate immunity. However, the functions of the remaining eight Toll-related proteins in Drosophila are not fully understood. Here, we show that Drosophila Toll-9 is necessary and sufficient for a special form of compensatory proliferation after apoptotic cell loss (undead apoptosis-induced proliferation [AiP]). Mechanistically, for AiP, Toll-9 interacts with Toll-1 to activate the intracellular Toll-1 pathway for nuclear translocation of the NF-κB-like transcription factor Dorsal, which induces expression of the pro-apoptotic genes reaper and hid. This activity contributes to the feedback amplification loop that operates in undead cells. Given that Toll-9 also functions in loser cells during cell competition, we define a general role of Toll-9 in cellular stress situations leading to the expression of pro-apoptotic genes that trigger apoptosis and apoptosis-induced processes such as AiP. This work identifies conceptual similarities between cell competition and AiP. The analysis of plasma temperatures by self-reversed mercury lines is well established in the diagnostics of HID lamps. In mercury-free HID lamps or mercury containing lamps including a complex fill, however, the analysis of mercury lines could be impossible or difficult because additives may disturb the spectral line shape. This work analyzes the suitability of self-reversed scandium lines for temperature determination using Bartels method. Temperatures obtained by the three scandium lines analyzed here were found to agree very well with results from mercury lines, demonstrating their applicability as a diagnostic tool in multicomponent HID lamps. Pro-apoptotic proteins from the reaper, hid, grim (RHG) family are primary regulators of programmed cell death in Drosophila due to their antagonistic effect on inhibitor of apoptosis (IAP) proteins, thereby releasing IAP-inhibition of caspases that effect apoptosis. Using a degenerate PCR approach to conserved domains from the 12 Drosophila species, we have identified the first reaper and hid orthologs from a tephritid, the Caribfly Anastrepha suspensa. As- hid is the first identified non-drosophilid homolog of hid, and As- rpr is the second non-drosophilid rpr homolog. Both genes share more than 50% amino acid sequence identity with their Drosophila homologs, suggesting that insect pro-apoptotic peptides may be more conserved than previously anticipated. Importantly, both genes encode the conserved IBM and GH3 motifs that are key for IAP-inhibition and mitochondrial localization. Functional verification of both genes as cell death effectors was demonstrated by cell death assays in A. suspensa embryonic cell culture, as well as in heterologous Drosophila melanogaster S2 cells. Notably, heterologous cell death activity was found to be higher for Anastrepha genes than their Drosophila counterparts. In common with the Drosophila cognates, As- hid and As- rpr negatively regulated the Drosophila inhibitor of apoptosis (DIAP1) gene to promote apoptosis, and both genes when used together effected increased cell death activity, indicating a co-operative function for As- hid and As- rpr. We show that these tephritid cell death genes are functional and potent as cell death effectors, and could be used to design improved transgenic lethality systems for insect population control. Although much has been learned in recent years about the apoptotic machinery, the mechanisms underlying survival and death choices during development of metazoans remain less clearly understood. During early oogenesis in Drosophila, a small excess in the number of specialized somatic cells, called polar cells (PCs), produced at follicle extremities is reduced to exactly two cells through apoptosis by mid-oogenesis. We have found that PCs destined to die first lose their apical contacts and then round up and shrink progressively until they disappear. Caspases are activated only once the cells have begun to shrink, suggesting that they are implicated in this part of the process, but not in the initial loss of cell polarity. Loss-of-function analyses based on mutant, clonal and RNAi approaches show that among the RHG family of pro-apoptotic factors, Hid is specifically necessary for PC apoptosis, as well as the initiator caspase Dronc and its adaptor Dark/Apaf-1, and likely several effector caspases, in particular Drice. In addition, we show that Hid protein and transcripts accumulate specifically in PCs destined to die, while the anti-apoptotic factor Diap1 is downregulated in these cells in a hid-dependent manner. Abstract: The present paper deals with hydrogen embrittlement (HE) susceptibility of a high strength steel grade (X80). The respective implication of different hydrogen populations, i.e. adsorbed, dissolved in interstitial sites, trapped on dislocations and/or microstructural elements on the associated embrittlement mechanisms has been addressed through mechanical testing in high pressure of hydrogen gas at room temperature. Tensile tests at various strain rates and hydrogen pressures have been carried out. Moreover, changes of gas (hydrogen or nitrogen) during loading have been imposed in order to get critical experiments able to discriminate among the potential hydrogen embrittlement mechanisms already proposed in the literature. The results of these tests have shown that hydrogen induces several kind of damages including decohesion along ferrite/pearlite interfaces and microcracks initiations on the specimens external surface. It is shown that decohesion is not critical under the loading paths used in the present study. On the contrary, it appears that the external microcracks initiation, followed by a quasi-cleavage fracture, is responsible for the premature failure of the material in high pressure of hydrogen gas. These experimental results have been further discussed by modeling hydrogen diffusion in order to identify hydrogen populations (adsorbed, diffusible or trapped) involved in HE. It was then demonstrated that adsorbed and near surface diffusible hydrogen are mainly responsible for embrittlement. [ABSTRACT FROM AUTHOR]

Key decisions one makes in a lifetime include whether and how often to reproduce, what role to play in the community and, under certain conditions, whether to live or die. Similar decisions are also made at the level of cells: whether to divide, what fate to assume in the multicellular context of metazoan development and, under certain conditions, whether to live or to die. The pro-apoptotic gene hid plays an important role in the execution of cell death in Drosophila. Abstract: Inactivation of the retinoblastoma gene Rb leads to defects in cell proliferation, differentiation, or apoptosis, depending on specific cell or tissue types. To gain insights into the genes that can modulate the consequences of Rb inactivation, we carried out a genetic screen in Drosophila to identify mutations that affected apoptosis induced by inactivation of the Retinoblastoma-family protein (rbf) and identified a mutation that blocked apoptosis induced by rbf. We found this mutation to be a new allele of head involution defective (hid) and showed that hid expression is deregulated in rbf mutant cells in larval imaginal discs. We identified an enhancer that regulates hid expression in response to developmental cues as well as to radiation and demonstrated that this hid enhancer is directly repressed by RBF through an E2F binding site. These observations indicate that apoptosis of rbf mutant cells is mediated by an upregulation of hid. Finally, we showed that bantam, a miRNA that regulates hid translation, is expressed in the interommatidial cells in the larval eye discs and modulates the survival of rbf mutant cells. Abstract: In Drosophila, heterozygosity in the pro-apoptotic gene hid significantly reduces apoptosis that is induced by ionizing radiation (IR). Therefore, mechanisms that regulate Hid levels can potentially contribute to life-or-death decision of an irradiated cell. 3′UTR of hid mRNA contains 5 potential binding sites for bantam microRNA. Ectopic expression of ban attenuated apoptosis that results from ectopic expression of hid but the significance of this regulation under physiological conditions remained to be investigated. We report here that ban is needed to limit IR-induced apoptosis in larval imaginal discs. Using tubulin–EGFP ban sensors with ban consensus sequences in the 3′UTR, we find that EGFP decreases following IR, indicating that IR activates ban. Likewise, a tubulin–EGFP reporter with hid-3′UTR is repressed in irradiated discs and this repression requires ban consensus sites in the hid 3′UTR. ban mutant larvae show increased sensitivity to killing by IR, which is suppressed by a mutation in hid. These results can fit into a model in which IR activates ban and ban represses hid to limit IR-induced apoptosis. miRNAs have been shown previously to be induced by radiation but this is the first report that a miRNA is functionally important for radiation responses.
