Background: X-linked acrogigantism (X-LAG; MIM: 300942) is a severe form of pituitary gigantism caused by chromosome Xq26.3 duplications involving GPR101. X-LAG-associated duplications disrupt the integrity of the topologically associating domain (TAD) containing GPR101 and lead to the formation of a neo-TAD that drives pituitary GPR101 misexpression and gigantism. As X-LAG is fully penetrant and heritable, duplications involving GPR101 identified on prenatal screening studies, like amniocentesis, can pose an interpretation challenge for medical geneticists and raise important concerns for patients and families. Therefore, providing robust information on the functional genomic impact of such duplications has important research and clinical value with respect to gene regulation and triplosensitivity traits. Methods: We employed 4C/HiC-seq as a clinical tool to determine the functional impact of incidentally discovered GPR101 duplications on TAD integrity in three families. After defining duplications and breakpoints around GPR101 by clinical-grade and high-density aCGH, we constructed 4C/HiC chromatin contact maps for our study population and compared them with normal and active (X-LAG) controls. Results: We showed that duplications involving GPR101 that preserved the centromeric invariant TAD boundary did not generate a pathogenic neo-TAD and that ectopic enhancers were not adopted. This allowed us to discount presumptive/suspected X-LAG diagnoses and GPR101 misexpression, obviating the need for intensive clinical follow-up. Conclusions: This study highlights the importance of TAD boundaries and chromatin interactions in determining the functional impact of copy number variants and provides proof-of-concept for using 4C/HiC-seq as a clinical tool to acquire crucial information for genetic counseling and to support clinical decision-making in cases of suspected TADopathies. The design of a deformation element or crash box that meets a given injury criterion based on deceleration requires careful consideration of physical properties and space requirements. Variations in material yield stress or geometry can result in statistical variations in the injury criterion output. Optimizing the crash box to fulfil two different injury criteria and two different energy levels may require more space than initially specified. In this study, we propose a protocol where the crash box is collapsed, and force–displacement is fitted to an equation. This fit is carried out with just two simulations and compared to 30 possible scenarios, obtaining a maximum error of 38.9%. With this initial fit, the appropriate thickness and yield stress can be chosen to perform crashes with two energy levels and monitor four injury values. With the ideal yield stress and sheet metal thickness, we introduce real statistical distributions using Monte Carlo design to perform 200 simulations and obtain 400 injury values for each design proposal. This technique ensures that the design will meet injury requirements for any possible combination of thickness and yield stress accepted by quality inspection. If only one simulation is performed, all designs meet the requirements, but only the last proposed design decreased the average injury to 9.2 g with a standard deviation of 2.68 g and a maximum value of 14.4 g, which is less than the required 15 g. Road restraint systems are used to protect vehicle occupants if the vehicle runs off the road and potentially collides with a dangerous obstacle. These road restraint systems must successfully pass the tests defined in EN 1317, or the Manual for Assessing Safety Hardware (MASH) before they are allowed to be installed. The safety assessment is carried out according to the criteria of ASI (Acceleration Severity Index), THIV (Theoretical Head Impact Velocity), OIV (Occupant Impact Velocity), ORA (Occupant Ridedown Acceleration), and PHD (Post-Impact Head Deceleration). Usually very old vehicles are used for these tests, and there is no assessment of occupant criteria such as HIC (Head Injury Criteria), chest deflection, etc. The objective of the study was to compare the occupant safety of vehicles that are commonly used in EN 1317 with vehicles that have improved safety equipment. Test results from two different vehicles (a commonly used vehicle in EN 1317 and a vehicle with improved safety equipment) and two different impact conditions (full overlap and an overlap of 50%) were compared. Measurement data from a Hybrid HIII 50th percentile anthropomorphic test device (ATD) (Denton ATD, INC.) was recorded during the tests to assess occupant safety. The tests have shown that vehicles with improved safety equipment perform better than vehicles that are commonly used in EN 1317-3 tests. The values for the occupant safety criteria assessed were well below the Euro NCAP (New Car Assessment Programme) or Federal Motor Vehicle Safety Standard (FMVSS) limits. However, the limits of the road safety criteria were in some cases considerably exceeded regardless of the vehicle. This has been observed in particular for the offset impact condition. THIV and OIV were supposed to be able to assess the risk of head injuries. However, these two criteria correlated negatively with the head criteria, HIC or a3ms. However, a positive correlation was found for the ASI with the HIC and the a3ms head acceleration. Even if some of the criteria for road safety correlate with the criteria for occupant safety, it is doubtful whether the criteria for road safety are suitable for assessing the risk of injury to vehicle occupants. In this research, the efficacy of specific heat treatment procedures in mitigating hydrogen embrittlement in X100 pipeline steels is discussed. The studied steels underwent a one-step austenitization process at temperatures of 780 °C (HT1), 830 °(HT2), and 880 °C (HT3) for 90 min, followed by oil quenching, tempering at 600 °C, and air cooling. The susceptibility of both the heat-treated and as-received (REF) specimens to HIC was assessed using an electrochemical hydrogen charging, tensile testing, micro-hardness testing, and electron backscatter diffraction (EBSD) techniques. The results showed that an increase in annealing temperature from 780 to 880 °C considerably decreases hydrogen embrittlement susceptibility. The hardness value at the centerline of the as-received (REF) specimen was found to be higher than that of the heat-treated specimens. Additionally, calculations of the hydrogen embrittlement (HE) index for all specimens were conducted. The results show that the REF specimen, which demonstrated the highest hardness and HE index values, has greater susceptibility to HIC. Conversely, the HT3 specimen, which was annealed at 880 °C and displayed the lowest hardness and HE index values, showed commendable resistance to HIC. The EBSD analysis revealed a decrease in Kernel average misorientation (KAM) values in heat-treated specimens, with the lowest values found in HT3. Dominant textures in all studied steels, {011} and {111}, were considered HIC-resistant and recrystallization mapping indicated a peak area of recrystallized grains in the REF specimen. We present a high-quality assembly and annotation of the periodical cicada species, Magicicada septendecula (Hemiptera: Auchenorrhyncha: Cicadidae). Periodical cicadas have a significant ecological impact, serving as a food source for many mammals, reptiles, and birds. Magicicada are well known for their massive emergences of 1 to 3 species that appear in different locations in the eastern United States nearly every year. These year classes ("broods") emerge dependably every 13 or 17 yr in a given location. Recently, it has become clear that 4-yr early or late emergences of a sizeable portion of a population are an important part of the history of brood formation; however, the biological mechanisms by which they track the passage of time remain a mystery. Using PacBio HiFi reads in conjunction with Hi-C proximity ligation data, we have assembled and annotated the first whole genome for a periodical cicada, an important resource for future phylogenetic and comparative genomic analysis. This also represents the first quality genome assembly and annotation for the Hemipteran superfamily Cicadoidea. With a scaffold N50 of 518.9 Mb and a complete BUSCO score of 96.7%, we are confident that this assembly will serve as a vital resource toward uncovering the genomic basis of periodical cicadas' long, synchronized life cycles and will provide a robust framework for further investigations into these insects. Rhabdomyosarcoma (RMS) is a pediatric malignancy of the muscle with characteristics of cells blocked in differentiation. NOTCH1 is an oncogene that promotes self-renewal and blocks differentiation in the fusion negative-RMS sub-type. However, how NOTCH1 expression is transcriptionally maintained in tumors is unknown. Analyses of SNAI2 and CTCF chromatin binding and HiC analyses revealed a conserved SNAI2/CTCF overlapping peak downstream of the NOTCH1 locus marking a sub-topologically associating domain (TAD) boundary. Deletion of the SNAI2-CTCF peak showed that it is essential for NOTCH1 expression and viability of FN-RMS cells. Reintroducing constitutively activated NOTCH1-ΔE in cells with the SNAI2-CTCF peak deleted restored cell-viability. Ablation of SNAI2 using CRISPR/Cas9 reagents resulted in the loss of majority of RD and SMS-CTR FN-RMS cells. However, the few surviving clones that repopulate cultures have recovered NOTCH1. Cells that re-establish NOTCH1 expression after SNAI2 ablation are unable to differentiate robustly as SNAI2 shRNA knockdown cells; yet, SNAI2-ablated cells continued to be exquisitely sensitive to ionizing radiation. Thus, we have uncovered a novel mechanism by which SNAI2 and CTCF maintenance of a sub-TAD boundary promotes rather than represses NOTCH1 expression. Deep learning frameworks have been applied to hyperspectral image classification (HIC) problems more and more in recent years, with excellent results. Existing network models, on the other hand, have a higher model complexity and take more time. The link between local spatial features is often overlooked in traditional HIC algorithms. This article presents a novel Lightweight Cascaded Deep Convolutional Neural Network (LC-DCNN) that describes the spatial as well as spectral characteristics of the hyperspectral images. The performance of the proposed LC-DCNN is validated for different spectral band reductions techniques to minimize the computational complexity of HIC such as Principle Component Analysis (PCA) and Linear Discriminant Analysis (LDA). The effectiveness of the proposed algorithm is validated on the Indian Pines and Salinas dataset for the vegetation and agriculture detection field based on accuracy, recall, precision and F1-score. The proposed approach provides 99% and 99.63% accuracy over Indian Pines and Salinas datasets, respectively, over the traditional state of arts employed previously for the HIC. The performance of the suggested LC-DCNN decreases for a larger number of classes. Background: All over the world, how health services can be provided in better quality and more economical way has been continuously investigated. In this regard, the implementation of rapid recovery procedures called ERAS (Enhanced Recovery After Surgery) is becoming more widespread recently. In this study, we aimed to investigate the effect of the ERAS protocol on reducing health expenditures. Materials and methods: Patients who underwent elective colorectal surgery in a public hospital between 2008 and 2018 were retrospectively reviewed following the approval of the Ethics Committee. The patients were divided into two groups as those who were applied ERAS protocol and treated with traditional methods. Patient billing information was requested from hospital management for analysis of treatment costs. Results: It was observed that invoices of all patients were arranged according to the common price tariff (Healthcare Implementation Communique (HIC)) which public institutions are subject to. There was no difference between the ERAS group and the control group in terms of demographic characteristics, smoking and alcohol use history, body mass index (BMI) and ASA (American Society of Anesthesiologists) score. All costs examined were statistically lower in the ERAS group (p < 0.001). Duration of hospitalization, time to return to daily activities, readmission rates within 30 days after discharge, rate of complication and mortality were similar in both groups. In the postoperative period, the rate of admission to the ICU (Intensive Care Unit) was considerably higher in the control group (p < 0.001). Conclusion: The implementation of the ERAS protocol provided both clinically positive effects and significant cost savings. Purpose: This research aimed to evaluate the biomechanical impact on a 3-year-old child's head during collisions with unmanned aerial vehicles (UAVs), focusing on the effects of UAV mass, impact velocity, and impact direction, using the Head Injury Criterion (HIC) for assessment. Methods: Experiments simulated impacts with UAVs of varying masses (249, 500 and 900 g) and velocities (19.0, 24.0 and 29.0 m/s) from different directions. HIC values were measured for each scenario and analyzed in relation to the Abbreviated Injury Scale to determine potential injury severity. Results: The findings showed that both the UAV's mass and impact velocity have a significant influence on the HIC value, with higher figures indicating a greater risk of serious injury. For the UAVs weighing 249 g and 500 g, frontal impacts resulted in the highest HIC values; however, for the UAV weighing 900 g, the highest HIC value occurred for the back hit. Moreover, injury risk was found to escalate non-linearly with increased velocity, especially for heavier UAVs. Conclusions: The study emphasizes the critical influence of UAV mass and impact velocity on the severity of head injuries in children. Increased mass and velocity correlated with higher HIC values, indicating a greater likelihood of severe injury. Frontal impacts were particularly hazardous for lighter UAVs, while rear impacts were more dangerous for heavier UAVs. These findings support the need for stringent regulations on UAV operational parameters, focusing on speed and mass limitations, to mitigate the risk of severe head injuries in children. In recent years, Mexico has suffereda series of explosive accidents linked to oil drilling and fuel pipeline operations. These explosions are often triggered by the rapid vapor expansion due to abrupt pressure changes, known as Boiling Liquid Expanding Vapor Explosions (BLEVEs). Tragically, in 2019, a devastating explosion occurred in Tlahuelilpan, claiming the lives of at least 100 people and injuring many others. This research aims to investigate brain injuries resulting from such explosions. Despite the potential for severe injuries, there is a significant lack of comprehensive injury data, with most research focusing on military contexts. In this study, we employed the Baker method in three configurations, utilizing a finite element model, Hybrid III 50Th Percentile Male Standing, in LS-Dyna®. Head Injury Criteria (HIC) is a measure for acceleration behavior in head. Our findings indicate that this value for acceleration, and BLAST propagation radio decrease with greater analysis distance. Brain injuries include edemas, contusions, lacerations, and skull compression, predominantly affecting individuals within a 6-meter radius of the detonation point.

Abstract Background Bacteria and their viruses, bacteriophages, are the most abundant entities of the gut microbiota, a complex community of microorganisms associated with human health and disease. In this ecosystem, the interactions between these two key components are still largely unknown. In particular, the impact of the gut environment on bacteria and their associated prophages is yet to be deciphered. Results To gain insight into the activity of lysogenic bacteriophages within the context of their host genomes, we performed proximity ligation-based sequencing (Hi-C) in both in vitro and in vivo conditions on the 12 bacterial strains of the OMM12 synthetic bacterial community stably associated within mice gut (gnotobiotic mouse line OMM12). High-resolution contact maps of the chromosome 3D organization of the bacterial genomes revealed a wide diversity of architectures, differences between environments, and an overall stability over time in the gut of mice. The DNA contacts pointed at 3D signatures of prophages leading to 16 of them being predicted as functional. We also identified circularization signals and observed different 3D patterns between in vitro and in vivo conditions. Concurrent virome analysis showed that 11 of these prophages produced viral particles and that OMM12 mice do not carry other intestinal viruses. Conclusions The precise identification by Hi-C of functional and active prophages within bacterial communities will unlock the study of interactions between bacteriophages and bacteria across conditions (healthy vs disease). Protein methylation is mainly observed in lysine, arginine and histidine residues. Histidine methylation occurs at one of two different nitrogen atoms of the imidazole ring, producing N τ-methylhistidine and N π-methylhistidine, and it has recently attracted attention with the identification of SETD3, METTL18 and METTL9 as catalytic enzymes in mammals. Although accumulating evidence had suggested the presence of more than 100 proteins containing methylated histidine residues in cells, much less information has been known regarding histidine-methylated proteins than lysine- and arginine-methylated ones, because no method has been developed to identify substrates for histidine methylation. Here, we established a method to screen novel target proteins for histidine methylation, using biochemical protein fractionation combined with the quantification of methylhistidine by LC-MS/MS. Interestingly, the differential distribution pattern of N τ-methylated proteins was found between the brain and skeletal muscle, and identified γ-enolase where the His-190 at the N τ position is methylated in mouse brain. Finally, in silico structural prediction and biochemical analysis showed that the His-190 in γ-enolase is involved in the intermolecular homodimeric formation and enzymatic activity. No‐salt flowthrough hydrophobic interaction chromatography (HIC) has been shown to effectively remove process and product‐related impurities from bioprocess streams. In this publication, a panel of six antibodies has been used to demonstrate operating principles for the application of no‐salt flowthrough HIC in antibody purification processes. The results indicate that no‐salt flowthrough HIC provides robust aggregate clearance across operating conditions including flow rate, and variations in resin ligand density. Additionally, HMW reduction has an optimal pH range relative to the isoelectric point of each molecule and high molecular weight (HMW) reduction can be improved by altering the total protein load and/or HMW concentration to drive binding of high molecular weight species to the resin. Background: Most studies assessing pathophysiological heterogeneity in asthma have been conducted in high-income countries (HICs), with little known about the prevalence and characteristics of different asthma inflammatory phenotypes in low-and middle-income countries (LMICs). This study assessed sputum inflammatory phenotypes in five centres, in Brazil, Ecuador, Uganda, New Zealand (NZ) and the United Kingdom (UK).Methods: We conducted a cross-sectional study of 998 asthmatics and 356 non-asthmatics in 2016-20. All centres studied children and adolescents (age range 8-20 years), except the UK centre which involved 26-27 year-olds. Information was collected using questionnaires, clinical characterization, blood and induced sputum.Results: Of 623 asthmatics with sputum results, 39% (243) were classified as eosinophilic or mixed granulocytic, i.e. eosinophilic asthma (EA). Adjusted for age and sex, with NZ as baseline, the UK showed similar odds of EA (odds ratio 1.04, 95% confidence interval 0.37-2.94) with lower odds in the LMICs: Brazil (0.73, 0.42-1.27), Ecuador (0.40, 0.24-0.66) and Uganda (0.62, 0.37-1.04). Despite the low prevalence of neutrophilic asthma in most centres, sputum neutrophilia was increased in asthmatics and non-asthmatics in Uganda.Conclusions: This is the first time that sputum induction has been used to compare asthma inflammatory phenotypes in HICs and LMICs. Most cases were non-eosinophilic, including in settings where corticosteroid use was low. A lower prevalence of EA was observed in the LMICs than in the HICs. This has major implications for asthma prevention and management, and suggests that novel prevention strategies and therapies specifically targeting non-eosinophilic asthma are required globally. North American sunfishes (Family Centrarchidae) are among the most popular sportfish throughout the United States and Canada. Despite the popularity of sunfishes, their ecological importance, and their extensive stocking and aquacultural history, few molecular studies have examined the evolutionary relationships and species boundaries among members of this group, many of which are known to hybridize. Here, we describe a chromosome-scale genome assembly representing Bluegill (Lepomis macrochirus), one of the most widespread centrarchid species. By combining long-read, Oxford Nanopore sequencing data with short-insert, whole-genome and HiC sequence reads, we produced an assembly (Lm_LA_1.1) having a total length of 889 Mb including 1,841 scaffolds and having a scaffold N50 of 36 Mb, L50 of 12, N90 of 29 Mb, and L90 of 22. We detected 99% (eukaryota_odb10) and 98% (actinopterygii_odb10) universal single-copy orthologs (BUSCOs), and ab initio gene prediction performed using this new assembly identified a set of 17,233 genes that were supported by external (OrthoDB v10) data. This new assembly provides an important addition to the growing set of assemblies already available for spinyrayed fishes (Acanthomorpha), and it will serve as a resource for future studies that focus on the complex evolutionary history of centrarchids. This study aimed to examine the prevalence of physical activity (PA), screen time (ST), sleep, and fruit and vegetable intake of children with low-and-middle-income countries (LMIC) parents, high-income countries (HIC), and Australian children, and whether these behaviours are associated with their immigrant status. Data for this study were from wave 7 of the Birth cohort (B cohort) of the Longitudinal Study of Australian Children. We used generalized estimating equations (GEE) to examine associations between health behaviours and immigrant status. The models were adjusted for a number of covariates. Children with LMIC parents had lower odds of meeting PA and sleep recommendations and higher odds ratio (OR) of meeting fruit and vegetable intake, and ST recommendations than Australian or HIC children. Children with LMIC parents had one-third the odds of meeting the PA recommendations (OR 0.39 [95%CI 0.22–0.70]) than Australian children. No significant differences were observed in lifestyle behaviours among children with HIC parents and Australian children. Lifestyle behaviours of children with LMIC parents differed from those of HIC and Australian children. However, children with HIC parents and Australian children had comparable lifestyle behaviours. Identified disparities in lifestyle behaviours among immigrant children can inform strategies to bring equity in Australian children's lifestyles. Our study underscores the importance of culturally appropriate targeted interventions to promote PA and sleep of children with LMIC parents.

Soccer is regarded as the most popular sport in the world, with millions of people actively involved in the game. Being a contact sport in nature, soccer players are susceptible to various kinds of injuries, such as lower extremities muscle injury. In addition to those familiar injuries that soccer players sustain during the game, traumatic brain injury is also a possibility. Head impacts in soccer could be a result of head-to-head impact with an opponent player, a headto-elbow impact, an impact with the goal post, an impact with the ground, as well as an impact with the soccer ball, which occurs during a heading manoeuvre. Soccer allows the players to use their heads to hit the ball to pass it to a teammate or even perform heading to score a goal. Although soccer heading is perceived as less harmful as compared to head impacts with other hard objects, many studies have shown compelling evidence that this repetitive heading might harm the brain, thereby leading to traumatic brain injury. Protective headgears designed especially for soccer players have been commercially available in the market for some years. However, the effectiveness of these headgears in reducing the impact due to soccer heading has not been well studied. This paper investigates the effectiveness of two commercially available headgears, the Full90 and the ForceField headgear by means of a heading experiment. An anthropometric test device known as Hybrid III head and neck dummy instrumented with an inertial sensor that consists of a triaxial accelerometer and gyroscope installed at the centre of gravity of the head was used in the experiment. A soccer ball launching machine was used to propel the ball at several inbound velocities. Peak linear acceleration (PLA) and peak angular acceleration (PRA) were recorded, and the head injury criterion (HIC) and the rotational injury criterion (RIC) were calculated. It was found that both headgears failed to reduce the linear components of head acceleration but instead increased the HIC (13 - 66% increment) depending on the inbound ball velocity. With respect to the rotational component of head injury, the Full90 headgear was found to reduce the RIC up to 29%, but the ForceField failed to provide a significant reduction of RIC. Overall, both headgears were found to be ineffective in reducing linear and rotational components of head injury, which could be attributed to the headgear design. This study conducted the pedestrian protection head impact test to determine the fracture mode of the windshield, acquire the acceleration curve of the headform impactor, and accurately obtain the peak acceleration and Head Injury Criterion (HIC). Subsequently, to quickly predict HIC and facilitate structural selection in designing automobiles, a theoretical analysis model for predicting peak acceleration was established and validated against the test results. To analyze the damage mechanism in detail, finite element analysis software LS-DYNA was employed to simulate the crack propagation in the windshield based on the peridynamics theory. Through a comparison between the numerical simulation and experimental results, the accuracy of the bond-based peridynamics model was verified, accurately revealing its failure mode and mechanical response to impact. A meta-analysis was conducted to compare: 1) time from traumatic brain injury (TBI) to the hospital, and 2) time within the hospital to intervention or surgery, by country-level income, World Health Organization region, and healthcare payment system. A comprehensive literature search was conducted and followed by a meta-analysis comparing duration of delays (prehospital and intrahospital) in TBI management. Means and standard deviations were pooled using a random effects model and subgroup analysis was performed using R software. Our analysis comprised 95,554 TBI patients from 45 countries. From 23 low- and middle-income countries, a longer mean time from injury to surgery (862.53 minutes, confidence interval [CI]: 107.42–1617.63), prehospital (217.46 minutes, CI: −27.34–462.25), and intrahospital (166.36 minutes, 95% CI: 96.12–236.60) durations were found compared to 22 high-income countries. African Region had the greatest total (1062.3 minutes, CI: −1072.23–3196.62), prehospital (256.57 minutes [CI: −202.36–715.51]), and intrahospital durations (593.22 minutes, CI: −3546.45–4732.89). Multiple-Payer Health Systems had a greater prehospital duration (132.62 minutes, CI: 54.55–210.68) but greater intrahospital delays were found in Single-Payer Health Systems (309.37 minutes, CI: −21.95–640.69). Our study concludes that TBI patients in low- and middle-income countries within African Region countries face prolonged delays in both prehospital and intrahospital management compared to high-income countries. Additionally, patients within Single-Payer Health System experienced prolonged intrahospital delays. An urgent need to address global disparities in neurotrauma care has been highlighted. To enhance the enzymatic digestibility of polyethylene terephthalate (PET), which is highly oriented and crystallized, a polyethylene glycol (PEG) surfactant of varying molecular weights was utilized to improve the stability of mutant cutinase from Humicola insolens (HiC) and to increase the accessibility of the enzyme to the substrate. Leveraging the optimal conditions for HiC hydrolysis of PET, the introduction of 1 % w / v PEG significantly increased the yield of PET hydrolysis products. PEG600 was particularly effective, increasing the yield by 64.58 % compared to using HiC alone. Moreover, the mechanisms by which PEG600 and PEG6000 enhance enzyme digestion were extensively examined using circular dichroism and fluorescence spectroscopy. The results from CD and fluorescence analyses indicated that PEG alters the protein conformation, thereby affecting the catalytic effect of the enzyme. Moreover, PEG improved the affinity between HiC and PET by lowering the surface tension of the solution, substantially enhancing PET hydrolysis. This study suggests that PEG holds considerable promise as an enzyme protector, significantly aiding in the hydrophilic modification and degradation of PET in an environmentally friendly and sustainable manner. Mangrove forest ecosystems support a diverse flora and fauna of marine and terrestrial species and have important direct and indirect economic, ecological and social values to mankind. Yellow mangrove (Bruguiera parviflora) belongs to the Rhizophoraceae family and is widely distributed in the intertidal zones along sheltered coastal areas in tropical latitudes. Here, we present a high‐quality, chromosome‐level assembly of the B. parviflora genome. We employed the 10x Genomics linked‐read technology to obtain a preliminary assembly, which was subsequently scaffolded using the long‐range chromatin contact mapping technique (HiC) to obtain a final assembly containing 213,026,782 bases in 10,045 scaffolds with an N50 length of 10,906,948 bases. Our gene prediction recovered 96.5% of the highly conserved orthologues in the Embryophyta lineage based on the Benchmarking Universal Single‐Copy Orthologues (BUSCO) analysis. We analysed the transversion rate at fourfold‐degenerate sites from orthologous gene pairs and discovered evidence supporting a recent whole‐genome duplication event in B. parviflora and other Rhizophoreae members. Comparative studies based on single‐copy orthologous genes indicated that B. parviflora and Bruguiera gymnorrhiza diverged approximately 24.1 million years ago. The population structure analysis revealed that 63 B. parviflora accessions from different geographical regions in Thailand were an admixture of two subpopulations. The examination of alternative splicing events in B. parviflora showed that the most prevalent splicing mechanism was intron retention. This high‐quality genome assembly together with the genetic diversity information obtained from the germplasm provide useful genomic resources for future studies on comparative phylogenetics and evolution of adaptive traits in mangrove species. Identifying cell type-specific enhancers in the brain is critical to building genetic tools for investigating the mammalian brain. Computational methods for functional enhancer prediction have been proposed and validated in the fruit fly and not yet the mammalian brain. We organized the 'Brain Initiative Cell Census Network (BICCN) Challenge: Predicting Functional Cell Type-Specific Enhancers from Cross-Species Multi-Omics' to assess machine learning and feature-based methods designed to nominate enhancer DNA sequences to target cell types in the mouse cortex. Methods were evaluated based on in vivo validation data from hundreds of cortical cell type-specific enhancers that were previously packaged into individual AAV vectors and retro-orbitally injected into mice. We find that open chromatin was a key predictor of functional enhancers, and sequence models improved prediction of non-functional enhancers that can be deprioritized as opposed to pursued for in vivo testing. Sequence models also identified cell type-specific transcription factor codes that can guide designs of in silico enhancers. This community challenge establishes a benchmark for enhancer prioritization algorithms and reveals computational approaches and molecular information that are crucial for the identification of functional enhancers for mammalian cortical cell types. Hydrogen-Induced Cracking (HIC) is a primary failure mechanism of pipeline-welded joints in the absence of external loading in the oil and gas exploration industries. Three different cooling rates after austenitization were used to simulate in the laboratory different regions of the heat-affected zone (HAZ) formed when welding an API X80 pipeline steel specially designed to enhance the HIC resistance. The samples were characterized with regard to microstructure and crystallography as well as HIC resistance. The HIC resistance test used NACE TM0284-2011 methodology. The microstructure and its homogeneity varied as a function of cooling rates. Samples containing inclusions and segregation zone from the segregation bands of specimens showed reduced HIC resistance, while specimens containing only acicular ferrite and granular bainite coupled with the absence of segregation zone showed significant improvement in HIC resistance. The best HIC resistance results came from samples presenting fine acicular ferrite consisting of fine interlocking plates, with divergent crystallographic orientations, preventing the formation of localized strain distribution inside the grain and at grain boundaries. It was also found that a large proportion of medium-angle boundaries prevent microcrack initiation and the transgranular mode of crack propagation.

Pedestrians suffer significant injuries from ground contact, but attempts to reduce them have been limited. A recent study using multi-body simulations showed ground contact injury may be improved by controlling vehicle braking. The aim of the study is to assesses whether controlled braking would be beneficial in real-world collisions. PC-Crash was used to reconstruct 150 freely available real-world vehicle pedestrian video collisions. Pairwise comparison of actual versus controlled braking was then performed for each case. Pedestrian injury evaluation metrics included head injury criterion HIC15 and pelvis contact force, and another metric named separation distance between the vehicle and pedestrian at the instant of ground contact was used to test the feasibility of external airbags for mitigating ground contact. Substantial head and pelvis injury reductions may be possible through the application of controlled braking. Full vehicle braking after first vehicle-pedestrian contact has little influence on overall pedestrian injury outcome. In controlled braking, an external semi-elliptic airbag with a range 2 m can prevent 83.1% ground contact (only 46.6% in actual cases) and there is a high probability (50%) that the available space to control braking is limited if the vehicle deviates from its original lane. Various kinematic explanations are presented, consistent with the principle that the vertical component of the impact on the ground can be reduced by controlling the vehicle braking. Systems based on spread spectrum technique are inherently interference resistant. However, asynchronous transmission and multipath propagation causes manifestation of multiuser interference. Multiuser interference in a multiple access system limits the capacity of the system. Hence, it becomes imperative to mitigate the interference. This work presents the design and mathematical analysis of Hybrid Interference Cancellation (HIC) detector for direct-sequence code division multiple access (DS-CDMA). The detector is analysed for single path as well as multipath propagation over a Rayleigh fading channel. Simulation results are compared with the results of Successive Interference Canceller (SIC). Furthermore, the SIC and HIC detectors are tested for convergence. An HIC detector is proposed for MC-CDMA system. The interference canceller discussed in this paper dispenses with the RAKE receiver for diversity combining and data estimation, which averts the likelihood of errors because of incorrect data estimates.

Background: Literature suggests that the presence of the current Bacillus Calmette Guerin (BCG) policy appears to mitigate COVID-19 disease burden but no information exists on the nature of the BCG strain and disease burden. Objectives: To study the association between type of BCG strain, BCG coverage (%), and COVID-19 disease burden. Methodology: An audit of global data on strains and disease burden was done. Country-specific data for COVID-19 cases and deaths, BCG-related data, and income level were obtained from the online databases, and the association was analyzed using linear regression. Results: Data of 139 countries were studied and 117 (84%) had a current BCG policy. Data on BCG strains were available for 51 countries and 18/51 (35%) used the Danish strain. While the choice of strain did not impact COVID-19-related disease burden, the presence of a current BCG policy was significantly associated with lower COVID-19 mortality. Conclusion: The presence of current BCG policy is associated with decreased COVID-19-related disease burden, but the type of strain used by a country in its vaccination program does not impact disease burden.

This work proposes an improved straw structure (ISS) sandwich hood (ISSH) to reduce the serious child head injuries caused by vehicle hood. The ISS sample is manufactured using the selective laser sintering (SLS) technique with nylon 11 material. A quasi-static compression experiment verifies the accuracy of the ISS in finite element analysis (FEA). The head injury criterion (HIC 15) with the original hood and ISSH are then compared. The obtained results show that ISSH reduced the HIC 15 by an average of 46.2 %. The ISSH has a regular deformation diffusion mode (RDDM), which helps to expand the energy absorption area. The ISS deformation mode is top-down layer-by-layer folding of structural layers. In addition, the impact of ISS geometric parameters on head injuries is studied. The thinner upper conical structures, larger layer heights, and material nylon 11 of the ISS contribute to the further improvement of protective effectiveness. The protective effectiveness of small-sized ISSs with connecting plates (SISC) against head injuries decreases with the increase of ISS quantity. In summary, this work provides new insights into the design of the sandwich hood for pedestrian protection, and the RDDM mechanism provides a novel direction for energy absorption. In order to focus on the injury mechanism of electric cyclists in the collision accident between right turn of truck and electric bicycle, Pc-Crash is used to value the injury of the electric cyclist in this paper. Through repeated tests in Pc-Crash, it was found that two dynamic response process were existed. The electric bicycle and cyclist collided with truck, and the electric bicycle and cyclist were thrown to the ground due to the impact force. Finally, they were crushed under certain conditions. The multi-rigid-body truck model is built on the theories of multi-rigid-body modeling and multi-rigid-body collision to get the value of the crushed part which the single rigid model can't calculate. After that, we analyze HIC, continuous 3 ms resultant acceleration of chest, the shear force of femur and leg, based on the electric bicycle speed, the mass of electric bicycle and cyclist, the height of cyclist's seat. We found that when the electric cyclist is not crushed by the truck, the value of each damage index is below the threshold. But when the cyclist is crushed by the truck, the damage index of the part where the cyclist is crushed exceeds the threshold, and causes fatal injury to the cyclist. The gas produced from an oil field is used to inject into the well for enhanced recovery of oil and gas. The injected gas flows through a pipeline called a header to individual wells for gas injection. One such 6" gas injection header at an offshore process complex experienced a leakage during operation just after 2 years of service. The operating pressure of the pipeline was normal, and there was no process disturbance at the time of the failure. There was no localized corrosion and thinning of the header. The material of construction of the header sample was reported to be ASTM A106 Gr B specification. Laboratory investigations revealed that the material did not conform to ASTM A106 Grade B standard. In the present case, the H2S partial pressure was 0.09 psi; this comes under sulfide stress cracking (SSC) region 2 as per ISO 15156-2:2015; therefore, the environment is prone to SSC. During metallography and SEM analysis, cracks filled with corrosion product typical of SSC were observed which propagated intergranularly. Zigzag cracks typical of hydrogen-induced cracking that branched out from the main SSC crack were also observed. As the metal was too brittle in nature, the gas injection header cracked at many sites. When the crack grew to the extent that the pipe could not withstand the operating pressure anymore, it failed in a brittle manner and the header was shattered into pieces. [ABSTRACT FROM AUTHOR]

The need to develop armour systems to protect against attacks from various sources is increasingly a matter of personal, social and national security. To develop innovative armour systems it is necessary to monitor developments being made on the type, technology and performance of the threats (weapons, projectiles, explosives, etc.) Specifically, the use of high protection level helmets on the battlefield is essential. The development of evaluation methods that can predict injuries and trauma is therefore of major importance. However, the risk of injuries or trauma that can arise from induced accelerations is an additional consideration. To develop new materials and layouts for helmets it is necessary to study the effects caused by ballistic impacts in the human head on various scenarios. The use of numerical simulation is a fundamental tool in this process. The work here presented focuses on the use of numerical simulation (finite elements analysis) to predict the consequences of bullet impacts on military helmets on human injuries. The main objectives are to assess the level and probability of head trauma using the Head Injury Criterion, caused by the impact of a 9 mm NATO projectile on a PASGT helmet and to quantify the relevance of projectile plasticity on the whole modelling process. The accelerations derived from the impact phenomenon and the deformations caused on the helmet are evaluated using fully three-dimensional models of the helmet, head, neck and projectile. Impact studies are done at impact angles ranging from 0 to 75°. Results are presented and discussed in terms of HIC and probability of acceleration induced trauma levels. Thorough comparison analyses are done using a rigid and a deformable projectile and it is observed that plastic deformation of the projectile is a significant energy dissipation mechanism in the whole impact process. Falls within children's playgrounds result in long bone and serious injuries. To lower the likelihood and severity of injury, impact attenuating surfaces (IAS) are installed within the impact area (fall zone). There are three primary IAS materials used, namely: granulated rubber products, wood fibre products, and sand. There is a deficiency with existing IAS test methods in that they do not take account of sand degradation over time. When children use the playground, sand degradation can occur when sand produces fines and smaller particles with low sphericity and angular which fill the voids between the sand particles. These fines and smaller particles tend to bind the sand and lower its impact attenuating performance. This paper proposes an additional IAS test to eliminate sands that degrade above an established threshold rate after installation due to normal usage. IAS degradation properties of fifteen IAS sands were tested including sand particle shape, sand particle distribution, percentage fines and sand particle degradation. This accelerated ageing test method is applicable only to sands and not rubber or wood fibre IAS products. The best IAS sands were sourced from quarries located on rivers that had eroded volcanic outcrops. These sands were shown to degrade the least and had little to no fines, and their particle shape was rounded to well-rounded. The most reliable source for good quality IAS sands on these rivers was on specific bends. The sand mined at these locations consistently had a tight particle size distribution. The haptic system has two key performance issues: stability and transparency. A haptic interface controller (HIC) is designed to address these issues. Addressing these issues becomes a complex problem as both are complementary to each other. Here, when transparency of the system is increased, its stability degrades and vice-versa. To overcome this problem, intelligent optimized solutions are used in this paper to design a HIC controller for the haptic system. SVM and NN techniques have been employed to identify the performance of the controller, ensuring stability and transparency both. The disadvantages of NN in terms of the number of neurons and hidden layers are overcome by SVM. Further, the performance of SVM is highly dependent upon the selection of free parameters. So, further, a modified PSO technique is employed for the optimal selection of these parameters to enhance the performance of SVM. Hence, this novel proposed hybrid technique of m-PSO optimized SVM is applied for the optimal design of the HIC to find out an optimal solution between trade-off the transparency and stability of the haptic device simultaneously. To appreciate the efficacy of the proposed technique, the result obtained with this is compared with HIC design using neural network and conventional ZN method also. This designed controller ensures stability as well as transparency, even under the presence of uncertainty, delay, and quantization error.

Abstract Background Enhancers play a fundamental role in orchestrating cell state and development. Although several methods have been developed to identify enhancers, linking them to their target genes is still an open problem. Several theories have been proposed on the functional mechanisms of enhancers, which triggered the development of various methods to infer promoter–enhancer interactions (PEIs). The advancement of high-throughput techniques describing the three-dimensional organization of the chromatin, paved the way to pinpoint long-range PEIs. Here we investigated whether including PEIs in computational models for the prediction of gene expression improves performance and interpretability. Results We have extended our $$\textsc{TEPIC}$$ TEPIC framework to include DNA contacts deduced from chromatin conformation capture experiments and compared various methods to determine PEIs using predictive modelling of gene expression from chromatin accessibility data and predicted transcription factor (TF) motif data. We designed a novel machine learning approach that allows the prioritization of TFs binding to distal loop and promoter regions with respect to their importance for gene expression regulation. Our analysis revealed a set of core TFs that are part of enhancer–promoter loops involving YY1 in different cell lines. Conclusion We present a novel approach that can be used to prioritize TFs involved in distal and promoter-proximal regulatory events by integrating chromatin accessibility, conformation, and gene expression data. We show that the integration of chromatin conformation data can improve gene expression prediction and aids model interpretability.

Northern bobwhites (Colinus virginianus) are small quails in the New World Quail family (Odontophoridae) and are one of the most phenotypically diverse avian species. Despite extensive research on bobwhite ecology, genomic studies investigating the evolution of phenotypic diversity in this species are lacking. Here, we present a new, highly contiguous assembly for bobwhites using tissue samples from a vouchered, wild, female bird collected in Louisiana. By performing a de novo assembly and scaffolding the assembly with Dovetail Chicago and HiC libraries and the HiRise pipeline, we produced an 866.8 Mb assembly including 1,512 scaffolds with a scaffold N50 of 66.8 Mb, a scaffold L90 of 17, and a BUSCO completeness score of 90.8%. This new assembly represents approximately 96% of the non-repetitive and 84% of the entire bobwhite genome size, greatly improves scaffold lengths and contiguity compared to an existing draft bobwhite genome, and provides an important tool for future studies of evolutionary and functional genomics in bobwhites.

Background: In high-income countries (HICs), between 65% and 70% of community-dwelling adults aged 65 and older are at high nutrition risk. Nutrition risk is the risk of poor dietary intake and nutritional status. Consequences of high nutrition risk include frailty, hospitalization, death, and reduced quality of life. Social factors (such as social support and commensality) are known to influence eating behavior in later life; however, to the authors' knowledge, no reviews have been conducted examining how these social factors are associated with nutrition risk specifically., Objective: The objective of this scoping review is to understand the extent and type of evidence concerning the relationship between social factors and nutrition risk among community-dwelling older adults in HICs and to identify social interventions that address nutrition risk in community-dwelling older adults in HICs., Methods: This review will follow the scoping review methodology as outlined by the JBI Manual for Evidence Synthesis and the PRISMA-ScR (Preferred Reporting Items for Systematic Reviews and Meta-Analyses extension for Scoping Reviews) guidelines. The search will include MEDLINE (Ovid), CINAHL, PsycINFO, and Web of Science. There will be no date limits placed on the search. However, only resources available in English will be included. EndNote (Clarivate Analytics) and Covidence (Veritas Health Innovation Ltd) will be used for reference management and removal of duplicate studies. Articles will be screened, and data will be extracted by at least 2 independent reviewers using Covidence. Data to be extracted will include study characteristics (country, methods, aims, design, and dates), participant characteristics (population description, inclusion and exclusion criteria, recruitment method, total number of participants, and demographics), how nutrition risk was measured (including the tool used to measure nutrition risk), social factors or interventions examined (including how these were measured or determined), the relationship between nutrition risk and the social factors examined, and the details of social interventions designed to address nutrition risk., Results: The scoping review was started in October 2023 and will be finalized by August 2024. The findings will describe the social factors commonly examined in the nutrition risk literature, the relationship between these social factors and nutrition risk, the social factors that have an impact on nutrition risk, and social interventions designed to address nutrition risk. The results of the extracted data will be presented in the form of a narrative summary with accompanying tables., Conclusions: Given the high prevalence of nutrition risk in community-dwelling older adults in HICs and the negative consequences of nutrition risk, it is essential to understand the social factors associated with nutrition risk.
