- Abstract
Objective To study the internal mechanisms of HSPA2 expression reduction due to heat stress. Methods The heat stress model of the reproductive system of rats was constructed, and MSP and BSP were used to detect methylation of CpG Island in the Hspa2 gene promoter region in the testicular tissue of rats. Results MSP: Whether using methylation primers or non-methylation primers, the heat stress group and the control group were able to successfully amplify the target DNA fragments, and in the area studied, the DNA samples of the heat stress group and the control group were present in both methylation and non-methylation. BSP: The heat stress group and the control group each contained 20 effective monoclonal, 440 CpG sites, of which the total number of CpG sites where methylation occurred in the heat stress group was 6, the total number of CpG sites with methylation in the control group was 7, and the methylation rate of the heat stress group and the control group was not significantly different in the target DNA fragments amplified by BSP (P> 0.05). Conclusion Heat stress leads to a reduction in the expression of HSPA2 in a short period of time, which may not be achieved by methylation modification of DNA. this conclusion needs further study to confirm.
