Grapevine seeds contain a large amount of antioxidant components, and are therefore recommended in the prevention and treatment of many diseases. For this research, we studied the antioxidant properties of grapevine seeds from the Marlen variety, as evidence suggests that these types have higher resistance against fungal diseases. Through high-performance liquid chromatography with UV/VIS detection, a total of 10 antioxidant components were selected for further investigation, specifically: catechin, epicatechin, rutin, quercitrin, quercetin, caftaric acid, caffeic acid, p-coumaric acid, ferulic acid, and gallic acid. The antioxidant activity was determinated spectrophotometrically through the adoption of three fundamentally different methods (the DPPH assay, the ABTS method, and the FRAP method). Using the Folin-Ciocalteu method, it was possible to determine the content of all the polyphenolic compounds. The results of the assessment antioxidant activity and the content of polyphenolic compounds were recalculated to gallic acid equivalents (GAE). The values of the antioxidant activity as determinated by the DPPH test were 6643 (±154) mg of GAE; 1984 (±88) mg of GAE when using the FRAP method; and 812 (±31) mg of GAE when the ABTS method was utilised. The content of the total polyphenolic compounds came to 6982 (±221) mg of GAE. The most abundant antioxidant was catechin, with a content of 115 mg.L-1, whilst the least represented compound was ferulic acid (0.139 mg.L-1). Overall, this study showed a high antioxidant potential of grapevine seeds.

Antioxidant activity was determined by three principally different methods (DPPH, ABTS and FRAP), the content of the total polyphenolic compounds was determined by the Folin ciocalteu method, and the content of the total flavanols was determined by DMACA reagent (p-dimethylaminocinnamaldehyde). Results are presented as an equivalent of gallic acid in g.L-1, respectively were expressed as g.L-1 of catechin equivalents (DMACA method). The highest values of antioxidant activity were measured in the cultivar Nativa (DPPH - 7.75 g.L-1, ABTS - 4.888 g.L-1, FRAP - 4.25 g.L-1). Conversely, the lowest values of antioxidant activity were detected in the cultivar Kofranka (DPPH - 7.08 g.L-1, ABTS - 4.17 g.L-1, FRAP - 4.55 g.L-1). Cultivar Nativa also reached the highest content of flavonols (3.77 g.L-1). The highest measured values of the content of total polyphenolic compounds were identified in the cultivar Cabernet Moravia (15.2 g.L-1 of GAE). Conversely, the lowest values of the content of total polyphenolic compounds were detected in the cultivar Nativa (8.04 g.L-1). Pearson correlation coefficients were calculated for the existing values between antioxidant activity (DPPH, ABTS, FRAP), contents of flavonols, and contents of total polyphenols. The highest correlation coefficient was found between the DPPH and ABTS methods; specifically, it was 0.857.

In our experiment, we studied the antimicrobial and antioxidative effect of phytogenic additives. Three additives (grapevine seeds, grape and rosehip pressings) were selected to be monitored. The extracts about concentrations of 1:3 and 1:5 were prepared from them. The monitoring of antimicrobial properties was focused on the pathogenic bacteria Clostridium perfringens and Escherichia coli causing a serious disease in avian species. The bacteria were prepared in the dilutions of 102, 104 and 106. The antimicrobial effect was observed in the inhibition zones. The antioxidant activity was determined using DPPH method within the antioxidant analysis. Furthermore, the content of flavanols, hydroxycinnamic acids and the total content of polyphenolic compounds was also determined. In the monitoring of the antimicrobial effect of grapevine seeds, grape and rosehip pressings at E. coli, a reduced growth of KTJ (colony forming units) was observed in the disk area during the dilution of 106 and 104. Reduced growth of C. perfringens at a dilution of 106 was noticed using the extracts of grapevine seeds and grape pressings. Low reduced growth of C. perfringens at a dilution of 106 was found out using rosehip pressings. In a dilution of 102 and 104 in C. perfringens and 102 in E. Coli, a very low increase of KTJ was observed therefore the zones of inhibition were not possible to measure. In all monitored additives, the antimicrobial effect was proved. The additives reduced the growth of pathogenic E. coli and C. perfringens. Within the antioxidant analysis, the highest antioxidant activity was found out in grapevine seeds (7.021 g.L-1 GAE), which also contained the highest content of flavanols (3000 times higher than the rosehip pressings and 300 times higher than grapevine seeds pressings), hydroxycinnamic acids (1000 times higher than in grape pressings and 7600 times higher than in rosehip pressings) and the total content of polyphenolic compounds (580 times higher than grape pressings and 2000 times higher than the rosehip pressings) of the monitored additives.

Phenolic compounds were extracted from European and Japanese grapevine species (Vitis vinifera and V. coignetiae) seeds using 80% methanol or 80% acetone. The total content of phenolic compounds was determined utilizing Folin-Ciocalteu’s phenol reagent, while the content of tannins was assayed by the vanillin and BSA precipitation methods. Additionally, the DPPH free radical and ABTS cation radical scavenging activities and the reduction power of the extracts were measured. The HPLC method was applied to determine the phenolic compounds, such as phenolic acids and catechins. The seeds contained large amounts of tannins and gallic acid and observable quantities of catechins, p-coumaric, ferulic and caffeic acids. The dominant form of phenolic acids in the extracts was the ester-bound form. The content of total phenolics was higher in the European grape V. vinifera seeds, which also contained more tannins, catechins and phenolic acids, except for caffeic acid. Extracts from V. vinifera seeds showed better radical scavenger properties and stronger reducing power. The total contents of phenolic compounds and tannins in acetone extracts were higher than in methanolic extracts. Acetone extracts also exhibited stronger antiradical properties as well as stronger reducing power.

Phenolic compounds were extracted from three wild grapevine species: Vitis californica, V. riparia and V. amurensis seeds using 80% methanol or 80% acetone. The total content of phenolic compounds was determined utilizing the Folin-Ciocalteu's phenol reagent while the content of tannins was assayed with the vanillin and BSA precipitation methods. Additionally, the DPPH free radical scavenging activity and the reduction power of the extracts were measured. The RP-HPLC method was applied to identify the phenolic compounds in the extracts, such as phenolic acids and catechins. The seeds contained large amounts of tannins, catechins and gallic acid and observable quantities of p-coumaric acid. The total content of phenolic compounds and tannins was similar in the extracts from V. californica and V. riparia seeds. However, the total content of total phenolic compounds and tannins in the extracts from V. californica and V. riperia seeds were about two-fold higher than that in the extracts from V. amurensis seeds. Extracts from seeds of the American species (V. californica and V. riparia) contained similarly high concentrations of tannins, whereas extracts from seeds of V. amurensis had approximately half that amount of these compounds. The content of catechin and epicatechin was similar in all extracts. The highest DPPH· anti-radical scavenging activity was observed in the acetonic and methanolic extracts of V. californica and V. riparia seeds--while the acetonic extract from the V. californica seeds was the strongest reducing agent.
