Centella asiatica, widely known as Gotu kola, is a traditional herb celebrated for its benefits in skin health and wound healing. Recent research has provided new insights into its efficacy, particularly through topical applications. This review highlights the plant's mechanisms, focusing on its active compounds such as asiaticoside, madecassoside, asiatic acid, and madecassic acid, which enhance collagen synthesis, modulate inflammation, and offer antioxidant protection. Clinical trials have been collected and summarized that innovative delivery systems, such as hydrogels, nanostructures or microneedles, can accelerate wound healing, reduce wound size, and improve recovery times in various wound types, including diabetic ulcers and burns. Future research will likely refine these technologies and explore new applications, reinforcing the role of C. asiatica in contemporary wound care. Two groups of specialized metabolites found in CA and purported to contribute to its beneficial effects are triterpenes (TTs) and caffeoylquinic acids (CQAs). In order to evaluate the role and interactions of TTs and CQAs in the effects of CA, we examined the neurotrophic effects of a water extract of CA (CAW) and combinations of its TT and CQA components in mouse primary hippocampal neurons in vitro and in Drosophila melanogaster flies in vivo. Methods: Primary hippocampal neurons were isolated from mouse embryos and exposed in vitro for 5 days toCAW(50 µg/mL), mixtures of TTs, CQAs or TT +CQA components or to 4 TTs or 8 individual CQA compounds of CAW. Dendritic arborization was evaluated using Sholl analysis. Drosophila flies were aged to 28 days and treated for 2 weeks with CAW (10 mg/mL) in the food, mixtures of TTs, CQAs or TT + CQA and individual TT and CQA compounds. TTs and CQAs were tested at concentrations matching their levels in the CAW treatment used. After 2 weeks of treatment, Drosophila aged 42 days were evaluated for phototaxis responses. Results: In mouse primary hippocampal neurons, CAW (50 µg/mL), the TT mix, CQA mix, all individual TTs and most CQAs significantly increased dendritic arborization to greater than control levels. However, the TT + CQA combination significantly decreased dendritic arborization. In Drosophila, a marked age-related decline in fast phototaxis response was observed in both males and females over a 60 days period. However, resilience to this decline was afforded in both male and female flies by treatment from 28 days onwards with CAW (10 mg/mL), or equivalent concentrations of mixed TTs, mixed CQAs and a TT + CQA mix. Of all the individual compounds, only 1,5-diCQA slowed age-related decline in phototaxis in male and female flies. Discussion: This study confirmed the ability of CAW to increase mouse neuronal dendritic arborization, and to provide resilience to age-related neurological decline in Drosophila. Gotu kola is one of the medicinal plants that is widely used because of its various nutritional content. In general, the cultivation only uses inorganic fertilizers, which have long-term effects that are not good for the environment. The use of organic fertilizers is an effort to produce high-standard products. Rabbit urine liquid organic fertilizer and goat manure are alternative types of fertilizer that can be given. Therefore, this study aimed to determine the response of giving liquid organic fertilizer of rabbit urine and goat manure and their interaction with the growth and yield. The study used a Factorial Randomized Block Design (RBD). The first factor was rabbit urine liquid organic fertilizer (LOF) with a concentration of 0, 2, and 4%. The second factor was goat manure with doses of 0, 37, and 75 g/polybag. The results showed that applying rabbit urine (LOF) and goat manure increased the growth and yield of gotu kola. Application of 2% rabbit urine LOF significantly increased stalk length compared to controls. Gotu kola plants given 37 g/polybag of goat manure produced stolons that were significantly longer than those treated with goat manure 75 g/polybag and had significantly heavier roots than other treatments. Common symptoms of depressive disorders include anhedonia, sleep problems, and reduced physical activity. Drugs used to treat depression mostly aim to increase serotonin signaling but these can have unwanted side effects. Depression has also been treated by traditional medicine using plants like Centella asiatica (CA) and this has been found to be well tolerated. However, very few controlled studies have addressed CA's protective role in depression, nor have the active compounds or mechanisms that mediate this function been identified. To address this issue, we used Drosophila melanogaster to investigate whether CA can improve depression-associated symptoms like anhedonia and decreased climbing activity. We found that a water extract of CA provides resilience to stress induced phenotypes and that this effect is primarily due to mono-caffeoylquinic acids found in CA. Furthermore, we describe that the protective function of CA is due to a synergy between chlorogenic acid and one of its isomers also present in CA. However, increasing the concentration of chlorogenic acid can overcome the requirement for the second isomer. Lastly, we found that chlorogenic acid acts via calcineurin, a multifunctional phosphatase that can regulate synaptic transmission and plasticity and is also involved in neuronal maintenance. Plant-based nootropics are a diverse group of natural drugs that can improve cognitive abilities through various physiological mechanisms, especially in cases where these functions are weakened or impaired. In many cases, the nootropics enhance erythrocyte plasticity and inhibit aggregation, which improves the blood's rheological properties and increases its flow to the brain. Many of these formulations possess antioxidant activity that protects brain tissue from neurotoxicity and improves the brain's oxygen supply. They can induce the synthesis of neuronal proteins, nucleic acids, and phospholipids for constructing and repairing neurohormonal membranes. These natural compounds can potentially be present in a great variety of herbs, shrubs, and even some trees and vines. The plant species reviewed here were selected based on the availability of verifiable experimental data and clinical trials investigating potential nootropic effects. Original research articles, relevant animal studies, meta-analyses, systematic reviews, and clinical trials were included in this review. Selected representatives of this heterogeneous group included Bacopa monnieri (L.) Wettst., Centella asiatica (L.) Urban, Eleutherococcus senticosus (Rupr. & Maxim.) Maxim., Ginkgo biloba L., Lepidium meyenii Walp., Panax ginseng C.A. Meyer, Paullinia cupana Kunth, Rhodiola rosea L., Schisandra chinensis (Turcz.) Baill., and Withania somnifera (L.) Dunal. The species are depicted and described, together with their active components and nootropic effects, and evidence of their efficacy is presented. The study provides brief descriptions of the representative species, their occurrence, history, and the chemical composition of the principle medicinal compounds, with uses, indications, experimental treatments, dosages, possible side effects, and contraindications. Most plant nootropics must be taken at optimal doses for extended periods before measurable improvement occurs, but they are generally very well tolerated. Their psychoactive properties are not produced by a single molecule but by a synergistic combination of several compounds. The use of gotu kola in food and beverages has increased over the years due to its beneficial and nutritional properties. The intent of this study is for the public utilization of the gotu kola and its improvement. It further investigates the sensory evaluation of dried fettuccine pasta enriched with gotu kola Centella asiatica among the three treatments in terms of aroma, color, taste and texture and to determine if there was a significant difference among the three treatments. This study used the experimental-descriptive research design employing a self-made modified questionnaire as a main tool in gathering the data through 4-point modified Hedonic scale for the level of liking. The data gathered were tabulated using the weighted mean and ANOVA to determine the significant difference in the respondent's liking among the three treatments. There was a total of 85 respondents composed of 55 selected food technology experts (25 instructors handling food technology and food related subjects and 30 students taking Master of Arts in Teaching Vocational Education major in Food Technology) and 30 consumers. The findings revealed that treatment 3 was "like very much" while, treatments 1 and 2 were "like moderately". The result showed there was a significant difference in the level of liking of the dried fettucine pasta in three treatments. Thus, the researcher recommended to utilize gotu kola as a potential ingredient to enrich the pasta for possible income generating project for extension activities. [ABSTRACT FROM AUTHOR]

Insufficient sleep is a common lifestyle which induces stress and has been shown to enhance the generation of free radicals, thus resulting in cellular injury. Gotu kola is a medicinal plant with established antioxidant profile. This study aims to evaluate the probable ameliorative effect of Nigerian-processed Gotu kola supplement known as Centella lujica on liver function in mice subjected to stress initiated by sleep disruption. Thirty mice (22+2g) were randomly assigned into five groups. Treatment lasted for seven days. Group 1 served as control (not sleep deprived), Group 2 was sleep deprived, mice in groups 3-5 in addition to being sleep deprived for the final 72 h of treatment, received Centella lujica (50 mg/kg), (100 mg/kg), astaxanthin (50mg/kg) respectively. Liver enzyme activity and hepatic oxidative stress parameters were assessed thereafter. Hepatocellular changes were also recorded using histological techniques. Pre-treatment of sleep-deprived mice with Centella lujica supplement caused a reduction in prooxidants with a concurrent increase in antioxidant levels. Meanwhile, the opposite effect was the case in the sleep-deprived group. Also, liver enzyme activity and liver cell degradation were significantly (p <0.05) reduced in the centella groups when compared to the sleep deprived group. In conclusion, Centella lujica supplement caused a reduction in oxidative stress and enhanced liver function in mice exposed to 72 h sleep restriction. [ABSTRACT FROM AUTHOR]

Chronic D-galactose (D-gal) administration causes cognitive impairment and is used widely in animal models for anti-aging studies. Centella asiatica (CA), a traditional herbal medicine, has been used as a brain tonic to enhance memory. This study evaluates the neuroprotective role of an ethanolic extract of Centella asiatica (CAE) against D-gal-induced aging in rats. Healthy male rats were divided into three groups: Control, D-gal, and D-gal + CAE. The Control group received normal saline (i.p.), whereas the D-gal group received D-gal (120 mg/kg b.w., i.p.), and the D-gal + CAE group received D-gal (120 mg/kg b.w., i.p.) and CAE (300 mg/kg b.w., orally) daily for 42 days. Behavioral and brain biochemical and histopathological changes were assessed after treatment. The results of the behavioral study depicted that D-gal significantly reduces the spontaneous alternation and locomotor activity indicating behavioral and cognitive impairment. Biochemical studies showed that D-gal significantly increases the oxidative stress and acetylcholinesterase activity (AChE) in rat brain. Histopathological study showed that D-gal disturbs the normal architecture of hippocampal and cortical cells, indicating degeneration in these brain areas. D-gal and CAE co-treatment for 42 days attenuated the behavioral, biochemical, and neuroanatomical impairments caused by the D-gal; it markedly suppresses the D-gal-induced oxidative stress and AChE activity in the brain, and maintains the normal cellular architecture in hippocampal and cortical areas. Thus, this study shows that CAE can protect the brain from the adverse effects of D-gal (e.g., memory loss and cognitive impairment) by modulating AChE activity and oxidative stress. Low water potential related stresses are regulated by modifying water uptake and loss to avoid low water potential, accumulating solutes which in turn enhance active principles and its gene expressions. Present study examined effect of in vitro induced absorption of mannitol and PEG (poly ethelene glycol) 6000 in Indian pennywort, Centella asiatica (L.) Urb., neutraceutical plant, evidenced by phenotypic, molecular and phytochemical analyses. Both mannitol and PEG 6000 induce water deficit conditions in plants and retarded normal plant biomass in terms of fresh and dry weights. These effects were significantly less severe in plants subjected to mannitol, compared to PEG. PEG and mannitol imposed water deficit, resulted in decline in major active compound, asiaticoside evidenced by HPTLC of asiaticoside content. Differential expression of some selected key genes in the asiaticoside pathway including squalene synthase and β amyrin synthase by qPCR, confirmed decrease in transcript level expression of asiaticoside, whereas upregulated transcript level expression was observed in cycloartenol synthase for synthesis of phytosterols. Estimation of total flavonoids and phenolics under different water deficit conditions were found declined. In conclusion, water deficit by mannitol and PEG 6000 can significantly affects processes associated with biomass growth and ability to synthesize secondary metabolites in C. asiatica. Background: Fitness is the state of the body does not experience significant fatigue after doing routine activities. While in Traditional Chinese Medicine (TCM), fitness is defined as the good circulation of Qi-energy and Xue-blood that can cause the function of the organs of Zhang Fu to run well. Physical predisposition is influenced by various factors and assessed in various ways, one of which is by measuring VO2 max calculated by doing cooper test that is running for 12 minutes on the track 400 meters. A person’s VO2 max level indicates the level of physical fitness. Based on the examination that has been done, the patient is exposed to excessive heat pathogens in the Stomach, there are humid pathogens in the body, and Qi and Xue are not sufficient. Purpose: To find out the effect of acupuncture therapy and herbal administration in complementary traditional medicine on improving fitness. Methods: To improve fitness is done acupuncture therapy and herbal therapy. Acupuncture therapy is performed at the Zusanli point (ST 36) and Sanyinjiao point (SP 6). Herbal therapy given during treatment is infusion of leaves Gotu Kola (Centella asiatica (L). Urb) and Liquorice (Glycyrrhiza glabra L.). To improve fitness, acupuncture therapy was performed 12 times in 4 stages of therapy. For herbal therapy infusa leaves Gotu Kola fresh as much as 2.5 grams and 2 grams of Liquorice, taken daily 3 times a day. Results: Acupuncture and herbal therapy that has been done for 26 days showed good results. This is evidenced by the results of copper test VO2 max increased 44.3% and no more complaints felt by patient. Conclution: Acupuncture therapy at the point of Zusanli (ST36), Sanyinjiao (SP6) accompanied by gotu kola herbs (Centella asiatica (L.) Urb) and liquorice (Glycyrrhiza glabra L.) can improve fitness.

Centella asiatica is a medicinal herb commonly known as Asiatic pennywort or gotu kola. The species is valued for its medicinal and nutritional properties. It is a perennial plant with leaves and stems that can be consumed as a green leafy vegetable. It is used as a folk remedy to cure various mild and chronic diseases due to its anti-rheumatic, antipyretic, antibacterial, antiviral, and anti-inflammatory effects, and as a mental rejuvenator. Although the species is widely distributed throughout the tropics and subtropics, its recognition was limited. The morphological description of C. asiatica is not well-documented in Myanmar, in Burmese or other local languages. Plant assessment via morphological markers is one of the ultimate methods for the primary description and characterization of their phenotypic traits. The objectives of this study were focused on the description of Myanmarese C. asiatica grown in Japan through morphological markers and a brief overview of its ethnobotanical use in Asia. Morphological characterization revealed the quantitative and qualitative differences regarding several traits among assessed C. asiatica samples. Furthermore, the study can provide information on the primary C. asiatica cultivation system in Japan, as it can be a potentially new and economically important crop for the Japanese market.

Stretch marks result from mechanical factors, namely stretching of the skin due to the development of subcutaneous structures (interstitial adipose tissue) or biochemical factors (the presence of excessive glucocorticoids that inhibit fibroblast activity and proliferation). There is no truly effective treatment for removing striae once they have formed. This study evaluates the effectiveness of administering Gotu Kola Extract (Centella asiatica) on the stretch mark. This paper was a Quasi-Experimental study with a pretest-posttest design. This study was conducted at the Independent Practice Midwife (IMP) Zummatul Atika in October-December 2020. The population was postpartum mothers (days 0-6) with stretch marks. Meanwhile, there were 12 respondents by purposive sampling technique. The independent variable was Gotu kola extract lotion, while the dependent variable was stretch marks with ratio data. Collecting data with a structured interview to gather the characteristics of respondents and participant observation to evaluate changes (number of stretch marks, color, moisture, and skin condition) in stretch marks before and after intervention during one month. The Gotu kola extract lotion was made in the pharmacy laboratory of PGRI Adi Buana University Surabaya within several stages. The data analysis used the paired T-test, and the result showed p= 0.000, indicating a significant difference before and after the intervention. This study concludes that administering Gotu Kola Extract (Centella asiatica) lotion effectively reduces skin pigmentation and lines in stretch marks, increases skin moisture, and refines skin texture.

Cholesterol in quail meat is one of the factors causing consumers to control consumption of quail meat. On the other hand, the antioxidant content in quail meat could attracts the consumers. Cinnamon (Cinnamomum sp.) bark and gotu kola (Centella asiatica) leaves are medicinal plants that contain many polyphenol compounds. The objective of this study was to obtain the most optimum formula made from cinnamon and gotu kola as a feed supplement to produce the quail meat which has low cholesterol but rich in antioxidants. This study used a completely randomized design which consisting of six groups with three replications. The six groups included one control (P0), 5 treatments consisted of 5% cinnamon (P1), 5% gotu kola (P2), combination of cinnamon and gotu kola powder with ratio 5%:5% (P3), 5%:10% (P4) and 10%:5% (P5). The results showed that food supplements made from cinnamon bark and gotu kola leaves could increase the body weight, antioxidant levels, and reduce cholesterol levels in quail meat. Cinnamon as feed supplement with a concentration of 5% or a combination of cinnamon and gotu kola with a ratio of 5%: 10% provided the best effect on antioxidants and cholesterol level in meat. The use of cinnamon and gotu kola supplementation to produce high antioxidant and low cholesterol of quail met have not studied yet. The result of the study would be beneficial for developing the healthy, safe and good quality of quail meat.

Due to an increase in the aging population, age-related diseases and age-related changes, such as diminished cognition and sleep disturbances, are an increasing health threat. It has been suggested that an increase in oxidative stress underlies many of these changes. Current treatments for these diseases and changes either have low efficacy or have deleterious side effects preventing long-time use. Therefore, alternative treatments that promote healthy aging and provide resilience against these health threats are needed. The herbs Withania somnifera and Centella asiatica may be two such alternatives because both have been connected with reducing oxidative stress and could therefore ameliorate age-related impairments. To test the effects of these herbs on behavioral phenotypes induced by oxidative stress, we used the Drosophila melanogaster sniffer mutant which has high levels of oxidative stress due to reduced carbonyl reductase activity. Effects on cognition and mobility were assessed using phototaxis assays and both, W. somnifera and C. asiatica water extracts improved phototaxis in sniffer mutants. In addition, W. somnifera improved nighttime sleep in male and female sniffer flies and promoted a less fragmented sleep pattern in male sniffer flies. This suggests that W. somnifera and C. asiatica can ameliorate oxidative stress-related changes in behavior and that by doing so they might promote healthy aging in humans.

The in vivo effects of Centella asiatica L. Urban (Family: Apiaceae; CA) on diabetes‐induced testicular fatty acid misdistribution and oxidative injury were investigated. Diabetic rats were treated with vehicle, CA or metformin daily for 14 days by oral gavage. Fatty acid (FA) content in testis was analysed using gas chromatography‐flame ionisation detection while redox indices were measured as peroxide value (PV), malondialdehyde (MDA), oxygen radical antioxidant capacity (ORAC), reduced glutathione (GSH), glutathione S‐transferase (GST) and glutathione peroxidase (GPx) activities. Diabetes increased omega‐6 (61%), and decreased omega‐3 (23%) and monounsaturated fatty acids (MUFA; 18%) compared to non‐diabetic controls. Oxidative injury in diabetic rats was confirmed by increases in PV (112%) and MDA (77%) in addition to decreases in GSH (41%) and activities of GST (19%) & GPx (24%) compared to non‐diabetic controls. CA treatment led to 17% reduction in omega‐6 and 33% rise in MUFA compared to diabetic controls. Additionally, CA ameliorated the oxidative injury and improved antioxidant capacity by increasing GSH (49%), GST (16%) and GPx (23%) when compared to diabetic controls. Data suggest CA potential in alleviating the alterations caused by diabetes in testes through effects on omega‐6 and MUFA; and via increased GSH level and dependent enzyme activities. Background and objective: Topical aminophylline, caffeine, yohimbe, l‐carnitine, and gotu kola (Centella asiatica) may aid in reducing body fat. Lipoxyderm™ contains these ingredients and was used to test if fat loss of the thigh, in conjunction with a low intensity exercise program and restricted calorie intake, was enhanced via the topical application of this lotion. Methods: This was a double‐blind, placebo‐controlled, within‐group study that investigated the effects of Lipoxyderm™ on thigh fat mass, circumference, and skinfold thickness. Seven participants underwent pre/post‐exercise testing for weight, bilateral thigh circumference/skinfold thickness, and body composition/thigh fat mass assessment via dual‐energy X‐ray absorptiometry. Participants followed a hypocaloric diet, walked 150 minutes/wk, and were randomly assigned to apply a placebo to one leg and Lipoxyderm™ to their other leg for 28 days. Separate two‐way mixed factorial repeated measures ANOVAs were used to compare the effects of Lipoxyderm™ to the placebo on thigh circumference, skinfold thickness, and fat mass. Results: A significant time x group interaction was found for thigh circumference (F1,6 = 18.2, P = 0.005), skinfold thickness (F1,6 = 14.6, P = 0.009), and fat mass (F1,6 = 37.1, P = 0.001). Conclusions: A twice‐daily topical application of Lipoxyderm™ for 28 days compared to a placebo combined with a walking program and a restricted caloric intake is more effective at reducing thigh circumference (1.2 vs 0.8 cm), thigh skinfold thickness (3.7 vs 2.0 mm), and thigh fat mass (100.0 g vs 57.3 g). This review describes in detail the phytochemistry and neurological effects of the medicinal herb Centella asiatica (L.) Urban. C. asiatica is a small perennial plant that grows in moist, tropical and sub-tropical regions throughout the world. Phytochemicals identified from C. asiatica to date include isoprenoids (sesquiterpenes, plant sterols, pentacyclic triterpenoids and saponins) and phenylpropanoid derivatives (eugenol derivatives, caffeoylquinic acids, and flavonoids). Contemporary methods for fingerprinting and characterization of compounds in C. asiatica extracts include liquid chromatography and/or ion mobility spectrometry in conjunction with high-resolution mass spectrometry. Multiple studies in rodent models, and a limited number of human studies, support C. asiatica’s traditional reputation as a cognitive enhancer, as well as its anxiolytic and anticonvulsant effects. Neuroprotective effects of C. asiatica are seen in several in vitro models, for example against beta amyloid toxicity, and appear to be associated with increased mitochondrial activity, improved antioxidant status, and/or inhibition of the pro-inflammatory enzyme, phospholipase A2. Neurotropic effects of C. asiatica include increased dendritic arborization and synaptogenesis, and may be due to modulations of signal transduction pathways such as ERK1/2 and Akt. Many of these neurotropic and neuroprotective properties of C. asiatica have been associated with the triterpene compounds asiatic acid, asiaticoside and madecassoside. More recently, caffeoylquinic acids are emerging as a second important group of active compounds in C. asiatica, with the potential of enhancing the Nrf2-antioxidant response pathway. The absorption, distribution, metabolism and excretion of the triterpenes, caffeoylquinic acids and flavonoids found in C. asiatica have been studied in humans and animal models, and the compounds or their metabolites found in the brain. This review highlights the remarkable potential for C. asiatica extracts and derivatives to be used in the treatment of neurological conditions, and considers the further research needed to actualize this possibility. Centella asiatica (L.) Urb. (CA; Family: Apiaceae) remains a major herb in the indigenous medicines of many Asian and African countries. Scientists have been delving deeper to fully understand the rationale for the various anecdotal therapeutic claims ascribed to its usage. Here, we present a comprehensive analysis of the published research on CA within the past three decades. The original articles and conference papers on CA research between 1991 and 2021 indexed on the Scopus® database were retrieved and analyzed by VOSviewer® software. The ∼2000 published research articles have been cited > 38,000 times in the period under study. The dominance of India, Malaysia, China, Thailand, and Indonesia out of the 81 countries that contributed to CA research is obvious. These Asian countries altogether contributed more than two-thirds of the publication count. However, the Italian scientist, Belcaro GV affiliated with the Università Degli Studi "G. d'Annunzio" Chieti – Pescara, Italy led the individual pack with 26 publications on CA while Kim OT of the Rural Development Administration, South Korea is the most cited author with 532 citations. The Thai universities – Chulalongkorn, Mahidol, Khon Kaen, Prince of Songkla, and Chiang Mai are actively involved in the research on CA having published ∼7 % of the total outputs and citations within the last 31 years. The primary focus of the research on CA has been its efficacy in the management of wound healing and other chronic conditions. As Americans use botanical medicines in ever-increasing numbers, they are moving beyond capsules and tinctures, and turning to assorted preparations that will complete their herbal medicine chest. According to The Arthritis Foundation, almost 45% of patients apply ointments or rubs for oseteoarthritis. Topical preparations have a long history in traditional herbal medicine systems, and makeup a significant proportion of the armamentarium of indigenous practitioners. Although contemporary patients are often resistant to the inconvenience of topical preparations, they often will become compliant after a notable success. The nomenclature of these topical preparations, as one would expect, is murky. The use of solid-phase extraction (SPE) for solving interference problems observed in simple HPLC assays involving nutraceuticals is demonstrated. Anionexchange SPE is used to remove an interfering compound coeluting with the triterpene glycoside madecassoside in gotu kola (Centella asiatica). C18 SPE is used to remove an interfering compound coeluting with parthenolide in feverfew (Tanacetum parthenium). and as a general cleanup for valerenic acids in valerian (Voleriana officinalis). Due to an increase in the aging population, age-related diseases and age-related changes, such as diminished cognition and sleep disturbances, are an increasing health threat. It has been suggested that an increase in oxidative stress underlies many of these changes. Current treatments for these diseases and changes either have low efficacy or have deleterious side effects preventing long-time use. Therefore, alternative treatments that promote healthy aging and provide resilience against these health threats are needed. The herbs Withania somnifera and Centella asiatica may be two such alternatives because both have been connected with reducing oxidative stress and could therefore ameliorate age-related impairments. To test the effects of these herbs on behavioral phenotypes induced by oxidative stress, we used the Drosophila melanogaster sniffer mutant which has high levels of oxidative stress due to reduced carbonyl reductase activity. Effects on cognition and mobility were assessed using phototaxis assays and both, W. somnifera and C. asiatica water extracts improved phototaxis in sniffer mutants. In addition, W. somnifera improved nighttime sleep in male and female sniffer flies and promoted a less fragmented sleep pattern in male sniffer flies. This suggests that W. somnifera and C. asiatica can ameliorate oxidative stress-related changes in behavior and that by doing so they might promote healthy aging in humans.

Background—Alzheimer's disease (AD) is a multifactorial, progressive, neurodegenerative disease that is characterized by memory loss, personality changes, and a decline in cognitive function. While the exact cause of AD is still unclear, recent studies point to lifestyle, diet, environmental, and genetic factors as contributors to disease progression. The pharmaceutical approaches developed to date do not alter disease progression. More than two hundred promising drug candidates have failed clinical trials in the past decade, suggesting that the disease and its causes may be highly complex. Medicinal plants and herbal remedies are now gaining more interest as complementary and alternative interventions and are a valuable source for developing drug candidates for AD. Indeed, several scientific studies have described the use of various medicinal plants and their principal phytochemicals for the treatment of AD. This article reviews a subset of herbs for their anti-inflammatory, antioxidant, and cognitive-enhancing effects. Methods—This article systematically reviews recent studies that have investigated the role of neuroprotective herbs and their bioactive compounds for dementia associated with Alzheimer's disease and pre-Alzheimer's disease. PubMed Central, Scopus, and Google Scholar databases of articles were collected, and abstracts were reviewed for relevance to the subject matter. Conclusions—Medicinal plants have great potential as part of an overall program in the prevention and treatment of cognitive decline associated with AD. Nerve growth factor (NGF), a typical neurotrophin, has been characterized by the regulation of neuronal cell differentiation and survival involved in learning and memory functions. NGF has a main role in neurite extension and synapse formation by activating the cyclic adenosine monophosphate-response-element-binding protein (CREB) in the hippocampus. The purpose of this study was to determine whether a mixture of Gotu Kola, Cnidium fruit, and Goji berry (KYJ) enhances memory function by inducing NGF-mediated actions both in vitro and in vivo. The KYJ combination increased NGF concentration and neurite length in C6 glioma and N2a neuronal cells, respectively. Additionally, we discovered memory-enhancing effects of KYJ through increased NGF-mediated synapse maturation, CREB phosphorylation, and cell differentiation in the mouse hippocampus. These findings suggest that this combination may be a potential nootropic cognitive enhancer via the induction of NGF and NGF-dependent activities. [ABSTRACT FROM AUTHOR]

Background: Dysregulation of glucose and glycogen metabolism are crucial mechanisms implicated in type 2 diabetes mellitus (T2DM). Centella asiatica (L.) Urban (Apiaceae) has been utilized as a traditional medicine in Africa and Asia for centuries and is commercially available as a dietary supplement., Aim: We explored for the first time, the possible efficacy of Centella asiatica (CA) extract in ameliorating T2DM-induced changes in key enzymes involved in glucose and glycogen metabolism in the rat skeletal muscle., Methods: Diabetic rats were orally treated with vehicle, CA (500 and 1000 mg/kg) or metformin (300 mg/kg) daily for 14 days. Skeletal muscle activities of hexokinase (HK), phosphofructokinase (PFK) and fructose 1,6-bisphosphatase (FBPase) were determined by spectrophotometric assays while those of glycogen synthase (GS) and glycogen phosphorylase (GP) were assayed radio-chemically. Histological examination of skeletal muscle was also performed., Results: Rats with induced T2DM had reduced activities of HK (25%), PFK (88%), and GS (38%) when compared to non-diabetic rats. Treatment of diabetic rats with CA500 increased the activities of PFK (7-fold), and FBPase (23%). Further, treatment of diabetic rats with CA1000 also increased the activities of GS (27%) and GP (50%) with little change in these parameters for diabetic rats treated with CA500. These effects probably led to the reduced blood glucose level and elevated skeletal muscle glycogen content observed in CA-treated rats relative to diabetic controls. Furthermore, CA treated rats had reduced the morphological damage of skeletal muscle fibres compared to the non-treated diabetic control rats., Conclusion: Our findings strongly suggest that the anti-diabetic effects of CA in part target muscle glucose and glycogen metabolism and hence supporting its folkloric medical use as an anti-diabetic remedy., (Copyright © 2019. Centella asiatica L. (Gotu Kola) is a nutritionally important plant and a valued traditional medicine in South East Asia. In this review, the chemical composition, nutritional values, and health benefits of C. asiatica have been discussed in detail to emphasize its usage as traditional food and medicine. C. asiatica is one of the most commonly used green leafy vegetables (GLVs) in some countries including Sri Lanka due to its high amounts of medicinally important triterpenoids and beneficial carotenoids. It is consumed in the form of GLVs and in the preparation of juice, drink, and other food products. It is also known to contain vitamins B and C, proteins, important minerals, and some other phytonutrients such as flavonoids, volatile oils, tannins, and polyphenol. In vitro and in vivo studies have shown important health benefits like antidiabetic, wound-healing, antimicrobial, memory-enhancing, antioxidant, and neuroprotecting activities. However, detailed scientific approaches on clinical trials regarding health benefits and nutritional values of C. asiatica are limited, hindering the perception of its benefits, mechanisms, and toxicity in order to develop new drug prototypes. In vitro studies have shown that the method of processing C. asiatica has an impact on its nutritional values and health-related beneficial compounds.
