The gut is an intricate and diverse organ system for investigating visceral pattern generation. The gut made an early evolutionary breakthrough. There is evidence that the molecular mechanisms governing the development of gastrointestinal patterns are species-specific. Some have suggested using marine drugs to treat gastrointestinal disorders. The study, which used an integrated network pharmacology approach, aimed to find out how well fucoidan worked and what Fucus vesiculosus might be doing to help prevent digestive problems. Researchers gathered information on how potential bioactive components of brown seaweed and their associated targets interact with the disease gene targets of gastrointestinal disorders. We studied the signaling pathways linked to F. vesiculosus in the context of gut health treatments using a KEGG (Kyoto Encyclopedia of Genes and Genomes pathway) and GO (Gene ontology) enrichment analysis. We built and studied the protein–protein interaction (PPI) and compound-target networks using Cytoscape. PPI network analysis yielded a total of 44 important targets, including BCL2, ESR1, and STAT3. The KEGG enrichment analysis revealed a strong correlation between the signaling pathways used in the treatment and the prevention of gastrointestinal disorders. We docked significant genes with TNF, STAT3, BCL2, and ALB. Fucoxanthin and fucoidan possess beneficial properties for improving gut health by treating gastric-intestinal issues. This study showed that by controlling several targets and many pathways with multiple components, F. vesiculosus exhibited preventive effects against gastrointestinal disorders. Geographic isolation is a factor that is known to affect species composition and diversity in marine systems. In this study, we tested the effects of geographic isolation on invertebrate fauna living among the belt-forming foundation species Fucus vesiculosus in the northern Baltic Sea. Sampling was carried out on reefs with varying levels of isolation (0.5, 1 and 2 km) in July 2021 in the Åland islands (between 59.96°N 20.24°E and 59.75°N 20.84°E). The total number of taxa, abundance, Shannon-Wiener index, Pielou's evenness and biomass were calculated and the effects of isolation were tested using generalised linear models. Multivariate methods were used to analyse differences in the invertebrate community composition. Although the number of taxa and Shannon´s diversity index did not vary along the isolation gradient, the total abundance of fauna decreased with increasing isolation. In addition, some taxa-specific patterns were observed, e.g. the abundance of insect larvae decreased with isolation while isopods were more numerous in the most isolated locations. As diversity and the number of taxa were similar regardless of isolation, shallow Fucus vesiculosus belts likely act as important biodiversity refuges and potential stepping-stones for invertebrates in remote outer archipelago areas. In the 19th century, a lower intertidal macrophyte, Fucus serratus (Linnaeus), from western Europe was introduced to Nova Scotia, Canada, where it successfully established, co-existing with native macrophytes Fucus vesiculosus (Linnaeus) and Ascophyllum nodosum (Linnaeus). We first examined whether a common gastropod in Nova Scotia, Littorina obtusata (Linnaeus, 1758), which grazes on the native macrophytes, has exploited the invasive and on finding this, we secondly examined whether there has been any phenotypic differentiation on the invasive. Among 98 sites surveyed around Nova Scotia and Newfoundland in May and June 1994, 11 had the invasive macrophyte, all of which showed colonization by L. obtusata including egg masses, juveniles, and adults. Among 2135 shells photographed for digital image analyses, those on the invasive differed from those on the native macrophytes with respect to (1) RGB (red/green/blue) channels, (2) HSV (hue, saturation, brightness) phenotypes, (3) protoconch pattern, and (4) adult shell size. Nitrogen and carbon stable isotope signatures on muscle tissues from a subset of L. obtusata indicate foraging fidelity on the invasive rather than temporary occupation. We suggest that these cumulative phenotypic responses to the invasive macrophyte that vary in extent and direction within and among localities reflect localized adaptation and offer a unique opportunity for quantifying early stages of phenotypic and genomic differentiation in a novel ecological niche. BACKGROUND: The drive towards ensuring the sustainability of bioresources has been linked with better valorising primary materials and developing biorefinery pipelines. Seaweeds constitute valuable coastal resources with applications in the bioenergy, biofertiliser, nutrition, pharmaceutical and cosmetic sectors. Owing to the various sought‐after metabolites they possess, several seaweed species are commercially exploited throughout Western Europe, including Ireland. Here, four commercially relevant brown (Fucus serratus and Fucus vesiculosus) and red (Chondrus crispus and Mastocarpus stellatus) seaweed species were sampled during a spring tide in July 2021 on moderately exposed shores across three coastal regions in the west of Ireland. RESULTS: Significant regional differences were identified when specimens were analysed for carbohydrates (max. 80.3 μg glucose eq mg−1 DW), proteins (max. 431.3 μg BSA eq. mg−1 DW), lipids (max. 158.6 mg g−1 DW), pigment signature and antioxidant potential. Protein content for F. serratus recorded a twofold difference between northern and southern specimens. The antioxidant potential of F. vesiculosus and M. stellatus returned greater activity compared to F. serratus and C. crispus, respectively. Multivariate analysis showed a clear latitudinal pattern across the three western coastal regions (north, west and south) for both F. vesiculosus and F. serratus. CONCLUSION: F. vesiculosus thalli from the northwest were richer in pigment content while the F. serratus thalli from the northwest were richer in antioxidants. Such biogeographic patterns in the biochemical make‐up of seaweeds need consideration for the development of regional integrated aquaculture systems and the optimisation of the biomass content for targeted downstream applications. © 2024 Society of Chemical Industry. The presence of heavy metals in wastewater is an environmental concern and the current treatment procedures are very expensive so it is necessary to find effective and inexpensive biosorbents. In this study, Fucus vesiculosus was used as a biosorbent for the biosorption of Cr(III) ions from the aqueous solutions. Biosorption parameters, such as pH, adsorbent dose, contact time, and initial concentrations of Cr(III) had the most impact on the sorption process. The required pH value for sorption was 5, the biosorbent dose was 4.0 g/L, the contact time was seen to occur after 90 min, and the Cr(III) removal decreased from 98.9 to 92%. The maximum biosorption capacity of chromium was 14.12 mg/g. FTIR analysis of Fucus vesiculosus biomass before the sorption process contains carboxyl, amino, hydroxyl, alkyne, and carbonyl groups, and according to the analysis after the sorption process, it was found that Cr(III) metal ions were incorporated within the sorbent during the interaction with (=C–H) active functional groups. The biosorption data were found to be perfectly suited by Langmuir equilibrium isotherm model. Summary: Algae and field crops can innovate and improve pasta; however, incorporation of high‐fibre materials can weaken the protein‐starch matrix. This study's aim was to verify how ratios of xanthan gum (XG) and linseed flour (LF) would influence physical, sensory attributes and bioactivity of pasta supplemented with algae. Fucus vesiculosus and Chlorella vulgaris were incorporated into durum wheat along with LF or XG. Samples were evaluated for optimal cooking time (OCT), cooking loss, colour variation, texture profile analysis (TPA), sensory evaluation, total phenolic content and antioxidant activity. XG increased cooking time and cooking loss. The phenolic content and antioxidant activity were highest in samples with LF. TPA showed texture is influenced by incorporation of algae. Sensory analysis showed that LF‐enriched pasta had better acceptance. Observations in biochemical composition and physical parameters revealed the potential to formulate dry or fresh pasta with a better nutritional content. [ABSTRACT FROM AUTHOR]

Recent agricultural research has prioritized the development of environmentally friendly management strategies to ensure food security, among which the application of biostimulants such as brown algae extracts, arbuscular mycorrhizal fungi (AMF), and their combination are included. The experimental protocol was based on the factorial combination of two planting times (4 May and 1 June) and seven biostimulant treatments (three brown algae species, Cystoseria tamariscifolia—C.t.; Fucus vesiculosus—F.v.; Padina pavonica—P.p.; arbuscular mycorrhizal fungi—AMF; C.t. + AMF; F.v. + AMF; P.p. + AMF) plus an untreated control. The earlier transplant resulted in a higher yield, due to the higher number of fruits per plant, and a higher plant fresh and dry biomass. The treatments with P.p. and F.v. extracts and the combination P.p. + AMF led to the highest yields (56.7 t ha−1), mainly due to the highest fruit number per plant. The earlier planting time led to higher values of dry residue, soluble solids, firmness, and colour component 'a'. The highest values of fruit dry residue were recorded under the F.v. and P.p. extracts, and the combinations F.v. + AMF and P.p. + AMF, the highest soluble solid content with P.p. treatment, and firmness under P.p. + AMF. The highest levels of 'L' and 'a' fruit colour components were obtained under the P.p. extract treatment, of 'b' upon the application of P.p. and F.v. extract, and AMF + P.p. and AMF + F.v. The later planting time led to significantly higher values of the antioxidant parameters, as did the application of the P.p. extract and P.p. + AMF. CAT activity was more intense corresponding to the later tomato crop cycle, P.p. extract, and AMF + P.p. Overall, our study highlights the potential of biostimulants, particularly brown algae extracts and their combination with AMF, to improve tomato yield, antioxidant properties, and biochemical activities. The extracellular release of dissolved organic carbon (DOC) from marine macroalgae supports coastal ecosystem function by supplying photosynthetically fixed carbon to higher trophic levels via the microbial loop. Despite its widely acknowledged biogeochemical importance, DOC is not typically included in primary production estimates of coastal systems. Additionally, little is known about how changes in species composition and coverage will affect the supply of DOC to coastal systems. Within the context of the Baltic Sea, anthropogenic forces are driving a decline in habitat forming kelp species (Fucus vesiculosus) which is superseded by filamentous/turfing algal species, a pattern of change observed globally in numerous other aquatic systems. To evaluate how the drivers of this change may impact the flow of carbon within the Baltic Sea coastal system, the production of DOC by the filamentous algae (Ectocarpus siliculosus) was examined and its rate of release determined (0.095 mg C · g DW-1 · h-1 in light and 0.070 mg C · g DW-1 · h-1 in dark). In addition, bioassays were used to assess the short-term DOC use by bacteria as a proxy of the lability of the released products, with the majority (28.7% released in light and 18.6% released in dark) of the products remaining after 120 hours. This data was linked with long term macroalgae biomass and coverage surveys in order to assess changes in macroalgae community structure through time and to produce standing stock estimates of F. vesiculosus. DOC production as a metric of algal coverage was used to make a preliminary estimation of how changes in community structure may impact the flow of carbon within the system. Our results suggest that decreased levels of DOC released by filamentous algae relative to F. vesiculosus will act to reduce microbial activity. Our model estimates the presence of approximately 150,000 tonnes of F. vesiculosus (dry weight) within the Estonian coastal system translating to an annual release of 7,391 tonnes of DOC. Our study indicates that filamentous-dominated systems have likely altered carbon flow, impacting the broader ecology of the Baltic Sea. Consequently, the loss of kelp species and the expansion of filamentous algae may alter carbon dynamics, with important ecological consequences for other coastal systems globally. Abstract Enzymatic degradation of algae cell wall carbohydrates by microorganisms is under increasing investigation as marine organic matter gains more value as a sustainable resource. The fate of carbon in the marine ecosystem is in part driven by these degradation processes. In this study, we observe the microbiome dynamics of the macroalga Fucus vesiculosus in 25-day-enrichment cultures resulting in partial degradation of the brown algae. Microbial community analyses revealed the phylum Pseudomonadota as the main bacterial fraction dominated by the genera Marinomonas and Vibrio. More importantly, a metagenome-based Hidden Markov model for specific glycosyl hydrolyses and sulphatases identified Bacteroidota as the phylum with the highest potential for cell wall degradation, contrary to their low abundance. For experimental verification, we cloned, expressed, and biochemically characterised two α-L-fucosidases, FUJM18 and FUJM20. While protein structure predictions suggest the highest similarity to a Bacillota origin, protein–protein blasts solely showed weak similarities to defined Bacteroidota proteins. Both enzymes were remarkably active at elevated temperatures and are the basis for a potential synthetic enzyme cocktail for large-scale algal destruction.

The global food industry faces a critical challenge in ensuring sustainable practices to meet the demands of a growing population while minimizing environmental impact. At the same time, consumer awareness and the demand for quality products drive innovation and inspire positive changes in the food supply chain. Aiming to create a more sustainable and nutrient-rich alternative, this study is summarized by characterizing the physical and chemical characteristics of algae-enriched chickpea hummus: an innovative approach to popular food products. The algae-enriched hummuses were developed with an incorporation (6% w/w) of Gelidium corneum and Fucus vesiculosus seaweeds and Chlorella vulgaris (hetero and autotrophic) microalgae to reveal their technological potential and evaluate the nutritional and rheological characteristics relative to a control hummus (without algae). From a nutritional perspective, the main results indicated that hummus enriched with microalgae showed an increase in protein content and an improved mineral profile. This was particularly notable for the seaweed F. vesiculosus and the autotrophic microalga C. vulgaris, leading to claims of being a "source of" and "rich in" various minerals. Additionally, the antioxidant activity of hummus containing F. vesiculosus and C. vulgaris increased significantly compared to the control. From a rheological perspective, incorporating algae into the humus strengthened its structure. The use of beach-cast macroalgae as a fertilizer (F) or soil amendment (SA) is coming back into focus, due to its highly efficient transformation of CO2, nutrients, salts and minerals from its aqueous surroundings into biomass. This research studied the hydrothermal carbonization (HTC) of Fucus vesiculosus macroalgae to hydrochar and evaluated its feasibility for use in soil applications. F. vesiculosus was submitted to HTC following a full factorial design of experiments with three HTC process parameters varied to assess their impact on the hydrochars: temperature (T: 160, 190, 220 °C), solid content (%So: 20, 35%), and process water recirculation (PWrec: yes and no). In general, F. vesiculosus and its hydrochars were rich in nutrients, but also contained regulated heavy metals. Investigation of the partitioning behavior of inorganic elements between the hydrochars and process water showed that heavy metals like Cr, Pb, Co and Cu tended to accumulate in the hydrochar, unaffected by HTC conditions. Nutrients such as P, N, B, and Mn were primarily found in the hydrochar and could be partially influenced to transfer to process water by changing %So and T. The correlation between the mass fractions of 22 elements in the hydrochar and HTC process parameters was studied. T was the most influential parameter, showing a significant positive correlation for eleven elements. %So and PWrec showed inconsistent effects on different elements. When process water was recirculated, some elements decreased (Ca, Cd, Fe) while others increased (K, Na, B, N) in the hydrochar. Assessment against various regulations and standards for F and SA revealed that F. vesiculosus complied with Cd limit values for most rules including the EURF and B, and was regulated only in the RAL for SA, over the limit value. In contrast, the limit value of Cd for both F and SA applications was surpassed in the 13 hydrochars. The contents of N, P, K, S, and Na in the feedstock and hydrochars complied with European F and SA rules, while they were too high for German rules on SA. The other limits for F rules were achieved (under certain HTC process parameters) except for P (lower than the requirements in F for F. vesiculosus and its hydrochars). Highlights: The brown algae complied with most limit values for Cd content to be applied as soil amendment or as fertilizer. Hydrochars from brown algae did not comply with any Cd limit values. No HTC process condition reduced the Cd content to comply with the requirements. Inorganic mass fractions were mostly influenced by temperature (positively correlated with 11/22 elements), followed by water recirculation (7) and solid content (6). Seaweeds are foundation species on rocky shores that are exposed to a series of stressors. These include wave exposure, desiccation, grazing, and human activities, with habitat fragmentation as a common result. As part of climate and biodiversity changes, we are expecting increased winds and more frequent storm events, warmer climate, and trophic downgrading of ecosystems, causing overgrazing events. Here, we present the results from a two-factorial mesocosm experiment on the relationship between wave exposure, habitat fragmentation and growth (branching and tip elongation) of Fucus vesiculosus in the intertidal and upper subtidal. We found that intertidal seaweeds branched significantly more at low than high wave exposure. There was no effect of habitat fragmentation on branching, and no effects on tip growth from either wave exposure or habitat fragmentation. The experiment suffered greatly from uncontrolled grazing. Gastropod coverage was higher inside seaweed patches than in the seaweed free areas between and outside of the patches. In basins with highly fragmented seaweed patches, we found more gastropods at low than high wave exposure, whereas in basins with low fragmentation we found more gastropods at higher wave exposure. Phragmites australis is a common helophyte, covering much of the sheltered and shallow soft bottoms along the coasts of the Baltic Sea. Despite the expansion of P. australis over the past decades, there is little information on aquatic macroinvertebrates within P. australis beds. In this study, we examined the effect of large-scale (wave exposure, nutrients) and small-scale (distance from the seaward edge, live and dead stalk density, epiphyte and rhizome biomass) drivers on the density, taxa richness, diversity, and community structure of epifauna and infauna in monospecific P. australis beds around the Åland Islands and the Archipelago Sea. We found that higher wave exposure and nutrient levels generally supported higher epi- and infauna abundance and taxa richness. The effects on Shannon–Wiener diversity were less evident apart from an increase of the infauna diversity in the Archipelago Sea with increasing nutrient levels. On a local scale, the distance from the seaward edge, live and dead stalk density, and epiphyte biomass had varying effects on both epi- and infauna communities in the different regions. Rhizome biomass had no effect on either the epi- or infauna abundance, taxa richness, or diversity. Furthermore, according to existing studies, other habitats, e.g., Zostera marina meadows, Fucus vesiculosus belts, and vegetated soft-bottomed shallow bays, are generally characterized by more abundant fauna, except for the infauna, which had a higher density in P. australis beds than in vegetated soft-bottomed shallow bays. Background: Brown seaweed is promising for the treatment of type 2 diabetes mellitus (T2DM). Its bioactive constituents can positively affect plasma glucose homeostasis in healthy humans. We investigated the effect of the brown seaweeds Sargassum (S.) fusiforme and Fucus (F.) vesiculosus in their natural form on glucose regulation in patients with T2DM. Methods: We conducted a randomized, double-blind, placebo-controlled pilot trial. Thirty-six participants with T2DM received, on a daily basis, either 5 g of dried S. fusiforme, 5 g of dried F. vesiculosus, or 0.5 g of dried Porphyra (control) for 5 weeks, alongside regular treatment. The primary outcome was the between-group difference in the change in weekly average blood glucose levels (continuous glucose monitoring). The secondary outcomes were the changes in anthropometrics, plasma lipid levels, and dietary intake. The data were analyzed using a linear mixed-effects model. Results: The change in weekly average glucose levels was 8.2 ± 2.1 to 9.0 ± 0.7 mmol/L (p = 0.2) in the S. fusiforme group (n = 12) and 10.1 ± 3.3 to 9.2 ± 0.7 mmol/L (p = 0.9) in the F. vesiculosus group (n = 10). The between-group difference was non-significant. Similarly, no between-group differences were observed for the changes in the secondary outcomes. Discussion: A daily intake of 5 g of fresh, dried S. fusiforme or F. vesiculosus alongside regular treatment had no differential effect on weekly average blood glucose levels in T2DM. Sulfated marine polysaccharides, so-called fucoidans, have been shown to exhibit anti-inflammatory and immunomodulatory activities in retinal pigment epithelium (RPE). In this study, we tested the effects of different fucoidans (and of fucoidan-treated RPE cells) on retinal microglia to investigate whether its anti-inflammatory effect can be extrapolated to the innate immune cells of the retina. In addition, we tested whether fucoidan treatment influenced the anti-inflammatory effect of RPE cells on retinal microglia. Three fucoidans were tested (FVs from Fucus vesiculosus, Fuc1 and FucBB04 from Laminaria hyperborea) as well as the supernatant of primary porcine RPE treated with fucoidans for their effects on inflammatory activated (using lipopolysaccharide, LPS) microglia cell line SIM-A9 and primary porcine retinal microglia. Cell viability was detected with a tetrazolium assay (MTT), and morphology by Coomassie staining. Secretion of tumor necrosis factor alpha (TNFα), interleukin 1 beta (IL1β) and interleukin 8 (IL8) was detected with ELISA, gene expression (NOS2 (Nitric oxide synthase 2), and CXCL8 (IL8)) with qPCR. Phagocytosis was detected with a fluorescence assay. FucBB04 and FVs slightly reduced the viability of SIM-A9 and primary microglia, respectively. Treatment with RPE supernatants increased the viability of LPS-treated primary microglia. FVs and FucBB04 reduced the size of LPS-activated primary microglia, indicating an anti-inflammatory phenotype. RPE supernatant reduced the size of LPS-activated SIM-A9 cells. Proinflammatory cytokine secretion and gene expression in SIM-A9, as well as primary microglia, were not significantly affected by fucoidans, but RPE supernatants reduced the secretion of LPS-induced proinflammatory cytokine secretion in SIM-A9 and primary microglia. The phagocytosis ability of primary microglia was reduced by FucBB04. In conclusion, fucoidans exhibited only modest effects on inflammatorily activated microglia by maintaining their cell size under stimulation, while the anti-inflammatory effect of RPE cells on microglia irrespective of fucoidan treatment could be confirmed, stressing the role of RPE in regulating innate immunity in the retina. Polyphenols (PP) found in brown algae are known for their wide range of biological activities including noteworthy antitumor properties. This article presents a method for obtaining an active polyphenolic extract from the Arctic alga Fucus vesiculosus with 98% purity and radical scavenging activity equivalent to 862 mg of ascorbic acid per gram of extract. Immunostimulant effects of polyphenols were assessed in vitro using venous blood from two groups of people: healthy people (HP) and people with chronic undifferentiated lymphocytic leukemia (LP). Polyphenols activated the surface properties of immunocompetent cells. Specifically, polyphenols dose-dependently increased the percentage of cells' spreading and adhesion by 2–3 times. Additionally, polyphenols increased the number of activated lymphocytes in the LP blood to levels characteristic of HP. Given their natural origin, high activity, non-toxicity, and straightforward production process, these studied polyphenols exhibit immense potential for use as new pharmaceuticals or as active components with immunostimulatory effects. Проведен анализ научной литературы, посвященной морским водорослям как дополнительному источнику питательных веществ и биологически активных соединений. Результаты исследований доказывают, что бурые водоросли являются перспективным сырьем для использования в питании человека. Водоросли содержат 7 аминокислот, незаменимых для взрослого человека. Присутствие монойодтирозина и дийодтирозина в составе данного биообъекта обуславливает гормоноподобное воздействие белков, содержащихся в нем, на организм человека. Жиры бурых водорослей представлены как насыщенными, так и ненасыщенными жирными кислотами, поэтому авторы статьи предполагают, что водоросли могут стать одним из альтернативных источников эссенциальных длинноцепочечных полиненасыщенных жирных кислот, оказывающих положительное влияние на функционирование сердечно-сосудистой и нервной систем, а также на когнитивное развитие детей. Особо следует подчеркнуть, что бурые водоросли являются источником фукостерина, который характеризуется гипохолестеринемическим, гипотензивным, антиоксидантным, противовоспалительным, противоопухолевым и антидиабетическим действием. Углеводы водорослей представлены уникальными по строению и биологическому действию полисахаридами: альгинатами, обладающими противовоспалительными, антиоксидантными и иммуномодулирующими свойствами; фукоиданами, имеющими антибактериальное, антиоксидантное и противоопухолевое действие; ламинаринами, оказывающими иммуномодулирующее, антиоксидантное, противоопухолевое и антибактериальное влияние. Полифенолы, содержащиеся в водорослях, не встречаются в наземных растениях. Водоросли являются источником йода и других минеральных веществ, а также витаминов. В настоящее время блюда и продукты из водорослей уже введены в рацион населения некоторых стран. Background: Snakebite envenomation (SBE) causes diverse toxic effects in humans, including disability and death. Current antivenom therapies effectively prevent death but fail to block local tissue damage, leading to an increase in the severity of envenomation; thus, seeking alternative treatments is crucial. Methods: This study analyzed the potential of two fucoidan sulfated polysaccharides extracted from brown seaweeds Fucus vesiculosus (FVF) and Undaria pinnatifida (UPF) against the fibrinogen or plasma coagulation, proteolytic, and phospholipase A2 (PLA2) activities of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom. The toxicity of FVF and UPF was assessed by the hemocompatibility test. Results: FVF and UPF did not lyse human red blood cells. FVF and UPF inhibited the proteolytic activity of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom by approximately 25%, 50%, and 75%, respectively, while all venoms led to a 20% inhibition of PLA2 activity. UPF and FVF delayed plasma coagulation caused by the venoms of B. jararaca and B. neuwiedi but did not affect the activity of B. jararacussu venom. FVF and UPF blocked the coagulation of fibrinogen induced by all these Bothropic venoms. Conclusion: FVF and UPF may be of importance as adjuvants for SBE caused by species of Bothrops, which are the most medically relevant snakebite incidents in South America, especially Brazil. High blood cholesterol levels are a major risk factor for cardiovascular diseases. A purified aqueous extract of Fucus vesiculosus, rich in phlorotannins and peptides, has been described for its potential to inhibit cholesterol biosynthesis and intestinal absorption. In this work, the effect of this extract on intestinal cells' metabolites and proteins was analysed to gain a deeper understanding of its mode of action on lipids' metabolism, particularly concerning the absorption and transport of exogenous cholesterol. Caco-2 cells, differentiated into enterocytes, were exposed to the extract, and analysed by untargeted metabolomics and proteomics. The results of the metabolomic analysis showed statistically significant differences in glutathione content of cells exposed to the extract compared to control cells, along with an increased expression of fatty acid amides in exposed cells. A proteomic analysis showed an increased expression in cells exposed to the extract compared to control cells of FAB1 and NPC1, proteins known to be involved in lipid metabolism and transport. To the extent of our knowledge, this study is the first use of untargeted metabolomics and a proteomic analysis to investigate the effects of F. vesiculosus on differentiated Caco-2 cells, offering insights into the molecular mechanism of the extract's compounds on intestinal cells. Newly synthesized cerium oxide was successfully obtained by the hard templating route. The optimal As(III) and As(V) adsorption onto the studied adsorbent was reached for the initial pH of 4.0 and a contact time of 10 h. The highest static adsorption capacities for As(III) and As(V) were 92 mg g−1 and 66 mg g−1, respectively. The pseudo-second-order model was well fitted to the As(III) and As(V) experimental kinetics data. The Langmuir model described the As(III) and As(V) adsorption isotherms on synthesized material. The adsorption mechanism of the studied ions onto the synthesized cerium oxide was complex and should be further investigated. The optimal solid–liquid ratio during the proposed aqueous extraction of inorganic As from the Fucus vesiculosus algae was 1:50. Fucoidan, a sulfate polysaccharide obtained from brown seaweed, has various bioactive properties, including anti-inflammatory, anti-cancer, anti-viral, anti-oxidant, anti-coagulant, anti-thrombotic, anti-angiogenic, and anti-Helicobacter pylori properties. However, the effects of low-molecular-weight fucoidan (LMW-F) on melanoma cell lines and three dimensional (3D) cell culture models are not well understood. This study aimed to investigate the effects of LMW-F on A375 human melanoma cells and cryopreserved biospecimens derived from patients with advanced melanoma. Ultrasonic wave was used to fragment fucoidan derived from Fucus vesiculosus into smaller LMW-F. MTT and live/dead assays showed that LMW-F inhibited cell proliferation in both A375 cells and patientderived melanoma explants in a 3D-printed collagen scaffold. The PTEN/AKT pathway was found to be involved in the anti-melanoma effects of fucoidan. Western blot analysis revealed that LMW-F reduced the phosphorylation of Bcl-2 at Thr 56, which was associated with the prevention of anti-apoptotic activity of cancer cells. The worldwide prevalence of obesity impacts more than 600 million adults. Successfully managing weight is effective in reducing the risk of chronic diseases, but sustaining long-term weight loss remains a challenge. Although there are supplements based on algae that claim to aid in weight loss, there is a notable scarcity of scientific evidence supporting their effectiveness, and their regular consumption safety remains inadequately addressed. In this work, commercially available Arthrospira (Spirulina) platensis Gomont and/or Fucus vesiculosus L. supplements showed moderate capacity to inhibit the activity of carbohydrate-metabolizing enzymes, and to scavenge biologically relevant reactive species. IC25 values varying between 4.54 ± 0.81 and 66.73 ± 5.91 µg of dry extract/mL and between 53.74 ± 8.42 and 1737.96 ± 98.26 µg of dry extract/mL were obtained for α-glucosidase and aldose reductase, respectively. A weaker effect towards α-amylase activity was observed, with a maximum activity of the extracts not going beyond 33%, at the highest concentrations tested. Spirulina extracts showed generally better effects than those from F. vesiculosus. Similar results were observed concerning the antiradical capacity. In a general way, the extracts were able to intercept the in vitro-generated reactive species nitric oxide (•NO) and superoxide anion (O2•−) radicals, with better results for O2•−scavenging with the spirulina samples (IC25 values of 67.16 and 122.84 µg of dry extract/mL). Chemically, similar pigment profiles were observed between spirulina supplements and the authenticated counterpart. However, fucoxanthin, the chemotaxonomic marker of brown seaweeds, was not found in F. vesiculosus samples, pointing to the occurrence of a degradation phenomenon before, during, or after raw material processing. Osteosarcoma is a bone cancer primarily affecting teenagers. It has a poor prognosis and diminished quality of life after treatment due to chemotherapy side effects, surgical complications and post-surgical osteoporosis risks. The sulphated polysaccharide fucoidan, derived from brown algae, has been a subject of interest for its potential anti-cancer properties and its impact on bone regeneration. This study explores the influence of crude, low-molecular-weight (LMW, 10–50 kDa), medium-molecular-weight (MMW, 50–100 kDa) and high-molecular-weight (HMW, >100 kDa) fractions from Sargassum filipendula, harvested from the Colombian sea coast, as well as crude fucoidan from Fucus vesiculosus, on a specific human osteoprogenitor cell type, human embryonic-derived mesenchymal stem cells. Fourier transform infrared spectroscopy coupled with attenuated total reflection (FTIR-ATR) results showed the highest sulphation levels and lowest uronic acid content in crude extract from F. vesiculosus. There was a dose-dependent drop in focal adhesion formation, proliferation and osteogenic differentiation of cells for all fucoidan types, but the least toxicity was observed for LMW and MMW. Transmission electron microscopy (TEM), JC-1 (5,50,6,60-tetrachloro-1,10,3,30-tetraethylbenzimi-dazolylcarbocyanine iodide) staining and cytochrome c analyses confirmed mitochondrial damage, swollen ER and upregulated autophagy due to fucoidans, with the highest severity in the case of F. vesiculosus fucoidan. Stress-induced apoptosis-like cell death by F. vesiculosus fucoidan and stress-induced necrosis-like cell death by S. filipendula fucoidans were also confirmed. LMW and MMW doses of <200 ng/mL were the least toxic and showed potential osteoinductivity. This research underscores the multifaceted impact of fucoidans on osteoprogenitor cells and highlights the delicate balance between potential therapeutic benefits and the challenges involved in using fucoidans for post-surgery treatments in patients with osteosarcoma. Featured Application: This article reports the bioactivity activity of macroalgae pigments against fungal agents of human skin diseases. Since this topic has not been thoroughly explored, this article presents a compilation of techniques to obtain the highest pigment yield, optimizing the methodology for both the extraction and separation of pigments, together with the anti-dermatophytic activity. Seaweeds have been explored as a natural resource of compounds of interest due to their bioactivities. Although many studies report the interest and the application of seaweeds in various areas, from food or human health to the economy, these data mostly focus on raw extracts and not on specific compounds, such as seaweed pigments. Fungal infections of the skin, nails, and hair caused by dermatophytes are the most common fungal infections worldwide. These pathologies require long periods of topical and/or systemic treatment associated with adverse effects and increased antifungal resistance. So, this study had two objectives: the first was to isolate and characterize the pigments of the seaweeds Calliblepharis jubata and Fucus vesiculosus; the second was to assess their antifungal activity. The extraction of pigments was performed using a method of extraction by exhaustion, and the purification was achieved via column chromatography. Three techniques were used to characterize the pigments: thin-layer chromatography (TLC), UV–visible spectrophotometry, and Fourier-transform infrared spectroscopy (FTIR). The antifungal activity against the three most common dermatophytes, Trichophyton rubrum, Trichophyton mentagrophytes, and Microsporum canis, was evaluated using a microdilution methodology, following the EUCAST international standards. It was possible to observe that the extracts obtained from the seaweed C. jubata, corresponding to the purified pigment phycobiliprotein, and the crude extract (an enriched extract) showed antifungal activity against the three fungal agents of human skin infection. Brown macroalgae are rich sources of nutrients and health-promoting compounds. Nevertheless, their consumption is still limited by their strong organoleptic characteristics, thus requiring the development of extraction strategies to profit from their nutritional value. To fulfil this, two sequential extraction approaches were developed, differing in the solvent used in the first extraction step, water in approach 1 or food-grade ethanol in approach 2, to obtain economic and affordable extracts rich in specific compounds from Fucus vesiculosus. The use of water in the first step of extraction allowed us to recover water-soluble phlorotannins, laminarans and mannuronic-rich alginates, making the subsequent 70% ethanol extract richest in fucoxanthin (0.07% algae DW), and the hot water fractions purest in fucoidans and alginates with a lower mannuronic-to-guluronic (M/G) ratio (2.91). Conversely, when beginning extraction procedures with 96% ethanol, the recovered yields of phlorotannins increased (0.43 g PGE/100 g algae DW), but there was a concomitant seven-fold decrease in the recovery of fucoxanthin in the subsequent 70% ethanol extract. This approach also led to less pure hot water fractions containing fucoidans, laminarans and alginates with a higher M/G ratio (5.50). Overall, this work unveiled the potential of the first extraction steps in sustainable and holistic cascade strategies to modulate the composition of food-grade extracts, creating prospects of their application as tailored functional ingredients in food products. Whether more species are required to maintain ecosystem functioning as spatial scale increases or across environmental gradients has frequently been studied by examining whether different species drive ecosystem functioning in different sites. However, this approach does rule out the counterfactual scenario where a few species could potentially maintain ecosystem functioning across sites as this requires examining which species can (or cannot) compensate for the loss of others.Here, I used an observational study and a field‐based transplant experiment to examine the effects of species loss on biomass productivity in an intertidal marine macroalgal system. I calculated the number of species required to maintain biomass productivity across four depth zones reflecting a water emersion gradient using two commonly used observational approaches. Then, I combined hypothetical simulated extinction scenarios with field‐based transplant data of relative growth rates of all species across the four depth zones to explore how the number of species required to maintain biomass productivity across depth zones changed under counterfactual scenarios where species compensated for species loss.The observational analyses suggested that between three and four species were required to maintain productivity across the depth zones. The simulated extinction scenarios did not. Rather, decreases in biomass productivity due to the loss of some species (e.g. Fucus spiralis, Ascophyllum nodosum) were easily compensated by other species (e.g. Fucus vesiculosus). The gut is an intricate and diverse organ system for investigating visceral pattern generation. The gut made an early evolutionary breakthrough. There is evidence that the molecular mechanisms governing the development of gastrointestinal patterns are species-specific. Some have suggested using marine drugs to treat gastrointestinal disorders. The study, which used an integrated network pharmacology approach, aimed to find out how well fucoidan worked and what Fucus vesiculosus might be doing to help prevent digestive problems. Researchers gathered information on how potential bioactive components of brown seaweed and their associated targets interact with the disease gene targets of gastrointestinal disorders. We studied the signaling pathways linked to F. vesiculosus in the context of gut health treatments using a KEGG (Kyoto Encyclopedia of Genes and Genomes pathway) and GO (Gene ontology) enrichment analysis. We built and studied the protein–protein interaction (PPI) and compound-target networks using Cytoscape. PPI network analysis yielded a total of 44 important targets, including BCL2, ESR1, and STAT3. The KEGG enrichment analysis revealed a strong correlation between the signaling pathways used in the treatment and the prevention of gastrointestinal disorders. We docked significant genes with TNF, STAT3, BCL2, and ALB. Fucoxanthin and fucoidan possess beneficial properties for improving gut health by treating gastric-intestinal issues. This study showed that by controlling several targets and many pathways with multiple components, F. vesiculosus exhibited preventive effects against gastrointestinal disorders.

Brown seaweeds are a rich source of carotenoids, particularly fucoxanthin, which has a wide range of potential health applications. Fucoxanthin fluctuates within and among seaweeds over time, frustrating efforts to utilise this resource. Thus, we require comprehensive analyses of long- and short-term concentrations across species in field conditions. Here, we used High Performance Liquid Chromatography to compare fucoxanthin content in four brown macroalgae, Ascophyllum nodosum, Fucus serratus, Fucus vesiculosus and Saccharina latissima, monthly for 1 year. F. serratus and F. vesiculosus had significantly higher fucoxanthin content (mg/g), which was highest in Spring (0.39 ± 0.04) and Autumn (0.45 ± 0.04) [mean (± SE)]. Two species, A. nodosum and F. serratus, were collected monthly at the same location for a further two non-consecutive years. For both A. nodosum and F. serratus, a significant interaction effect of seasons and years was identified, highlighting that there is variation in fucoxanthin content among and within species over time. We also show that fucoxanthin content differs significantly among months even within seasons. Therefore, it is not sufficient to assess fucoxanthin in single months to represent seasonality. We discuss how weather, nutrients and reproduction may have driven the seasonal variation, and reveal patterns of fucoxanthin concentration that can provide information concerning its availability for many important medical functions. Arctic brown algae are considered a promising industrial-scale source of bioactive sub-stances as polysaccharides, polyphenols, and low-molecular secondary metabolites. Conventional technologies for their processing are focused mainly on the isolation of polysaccharides and involve the use of hazardous solvents. In the present study a "green" approach to the fractionation of brown algae biomass based on the dissolution in ionic liquids (ILs) with 1-butil-3-methylimidazolium (bmim) cation with further sequential precipitation of polysaccharides and polyphenols with acetone and water, respectively, is proposed. The effects of IL cation nature, temperature, and treatment duration on the dissolution of bladderwrack (Fucus vesiculosus), yields of the fractions, and their chemical composition were studied involving FTIR and NMR spectroscopy, as well as size-exclusion chromatography and monosaccharide analysis. It was shown that the use of bmim acetate ensures almost complete dissolution of plant material after 24 h treatment at 150 °C and separate isolation of the polysaccharide mixture (alginates, cellulose, and fucoidan) and polyphenols (phlorotannins) with the yields of ~40 and ~10%, respectively. The near-quantitative extraction of polyphenolic fraction with the weight-average molecular mass of 10–20 kDa can be achieved even under mild conditions (80–100 °C). Background: Brown seaweed is promising for the treatment of type 2 diabetes mellitus (T2DM). Its bioactive constituents can positively affect plasma glucose homeostasis in healthy humans. We investigated the effect of the brown seaweeds Sargassum (S.) fusiforme and Fucus (F.) vesiculosus in their natural form on glucose regulation in patients with T2DM. Methods: We conducted a randomized, double-blind, placebo-controlled pilot trial. Thirty-six participants with T2DM received, on a daily basis, either 5 g of dried S. fusiforme, 5 g of dried F. vesiculosus, or 0.5 g of dried Porphyra (control) for 5 weeks, alongside regular treatment. The primary outcome was the between-group difference in the change in weekly average blood glucose levels (continuous glucose monitoring). The secondary outcomes were the changes in anthropometrics, plasma lipid levels, and dietary intake. The data were analyzed using a linear mixed-effects model. Results: The change in weekly average glucose levels was 8.2 ± 2.1 to 9.0 ± 0.7 mmol/L (p = 0.2) in the S. fusiforme group (n = 12) and 10.1 ± 3.3 to 9.2 ± 0.7 mmol/L (p = 0.9) in the F. vesiculosus group (n = 10). The between-group difference was non-significant. Similarly, no between-group differences were observed for the changes in the secondary outcomes. Discussion: A daily intake of 5 g of fresh, dried S. fusiforme or F. vesiculosus alongside regular treatment had no differential effect on weekly average blood glucose levels in T2DM.

Sulfated marine polysaccharides, so-called fucoidans, have been shown to exhibit anti-inflammatory and immunomodulatory activities in retinal pigment epithelium (RPE). In this study, we tested the effects of different fucoidans (and of fucoidan-treated RPE cells) on retinal microglia to investigate whether its anti-inflammatory effect can be extrapolated to the innate immune cells of the retina. In addition, we tested whether fucoidan treatment influenced the anti-inflammatory effect of RPE cells on retinal microglia. Three fucoidans were tested (FVs from Fucus vesiculosus, Fuc1 and FucBB04 from Laminaria hyperborea) as well as the supernatant of primary porcine RPE treated with fucoidans for their effects on inflammatory activated (using lipopolysaccharide, LPS) microglia cell line SIM-A9 and primary porcine retinal microglia. Cell viability was detected with a tetrazolium assay (MTT), and morphology by Coomassie staining. Secretion of tumor necrosis factor alpha (TNFα), interleukin 1 beta (IL1β) and interleukin 8 (IL8) was detected with ELISA, gene expression (NOS2 (Nitric oxide synthase 2), and CXCL8 (IL8)) with qPCR. Phagocytosis was detected with a fluorescence assay. FucBB04 and FVs slightly reduced the viability of SIM-A9 and primary microglia, respectively. Treatment with RPE supernatants increased the viability of LPS-treated primary microglia. FVs and FucBB04 reduced the size of LPS-activated primary microglia, indicating an anti-inflammatory phenotype. RPE supernatant reduced the size of LPS-activated SIM-A9 cells. Proinflammatory cytokine secretion and gene expression in SIM-A9, as well as primary microglia, were not significantly affected by fucoidans, but RPE supernatants reduced the secretion of LPS-induced proinflammatory cytokine secretion in SIM-A9 and primary microglia. The phagocytosis ability of primary microglia was reduced by FucBB04. In conclusion, fucoidans exhibited only modest effects on inflammatorily activated microglia by maintaining their cell size under stimulation, while the anti-inflammatory effect of RPE cells on microglia irrespective of fucoidan treatment could be confirmed, stressing the role of RPE in regulating innate immunity in the retina.

It is estimated that more than one-third of people aged over 16 reported living with inflammation-related chronic illness. Fucoidan, sulfated polysaccharide, has been proven to modulate immune response and inhibit inflammation in both in vitro and in vivo research. In the present study the effect of commercially available fucoidan from Fucus vesiculosus (with purity 97%) on immune response was investigated. Two cell lines, human fibroblasts (HDFa) and mouse macrophages (J774A.1) were used in the study. Cells were activated with pro-inflammatory stimulus (LPS/IFN-γ or PMA) and treated with fucoidan. Results of NO inhibition assay revealed that fucoidan has anti-inflammatory activity and significantly reduces the level of NO produced by the cells. A significant decrease in secretion of IL-6 from both cell lines was observed. Moreover, fucoidan inhibits IL-12 production by mouse macrophages. The obtained results indicate that fucoidan not only acts as anti-inflammatory compound, but also has immunomodulatory activity. Enhanced secretion of TNF-α and IL-8 from macrophages and fibroblasts, respectively, was also noted. The antioxidant activity of fucoidan has also been studied. Fucoidan demonstrates dose-dependent scavenging properties by DPPH assay. Fucoidan's simultaneous ability to reduce inflammation, activate macrophages, and remove free radicals may be used as a tool to fight chronic inflammation disorders. These all contribute to the effective immunoprevention of cancer or other diseases related to the impaired function of the immune cells.

Simple Summary: Searching for new feed products suitable for poultry nutrition is an important tool for improving productive and immunity status of poultry. Thus, our present study aimed to evaluate the impact of using the thermally modified mineral adsorbent shungite and the dried seaweed meal Fucus vesiculosus in different doses in the diets of Brown Nick cross laying hens on their productivity indicators, digestibility of nutrients, nitrogen utilization, morphological blood profile, immunity status indicators, and egg quality. Our results showed that the inclusion of the abovementioned feed additives improves both productive performance and immunity status of Brown Nick cross laying hens as well as decreases the incidence of fatty liver occurrence. The main purpose of this study was to assess the impact of using the thermally modified mineral adsorbent shungite (MAS) and the dried seaweed meal Fucus vesiculosus (DSM) with different doses in Brown Nick cross laying hens' diet on their productivity, nutrient digestibility, morphological and blood profile, immunity status, and egg quality. A total of 261,720 hens were used in this experiment at the age of 63 weeks, and they were randomly divided into 5 groups (feeding program) with six repetitions of 8724 chickens in each. The first served (control) as a control group where laying hens were fed the basal diet that was used on the farm only; the second and the third groups represented MAS+ and MAS++, where they received the basal diet supplemented by 0.1% and 0.25% (or 1.0 kg/t and 2.5 kg/t of feed) of the mineral adsorbent shungite (MAS) which was provided in the feed in powder form (5 microns) and was added to the feed at the feed mill; the fourth and fifth groups represented DSM+ and DSM++, which received the basal diet provided with 0.1% and 0.25% (or 1.0 kg/t and 2.5 kg/t of feed) of dried seaweed meal of F. vesiculosus algae (DSM). The average egg weight over the entire period of the experiment revealed significant differences between the experimental groups and represented in the control group 65.20 vs. 66.88, 66.87 and 68.10 and 68.13 g in the MAS+ and MAS++, and DSM+ and DSM++ groups, respectively. Once the dried seaweed meal F. vesiculosus (DSM) was used, the crude protein increased significantly (p < 0.05) in egg yolk by 2.64 and 2.67%, carotenoids by 1.13 and 1.20 mg/g DM. The inclusion of both MAS and DSM feed additives revealed a significant decrease in the level of crude fat (lipids) in their liver when compared with the control group. The level of erythrocytes (RBCs) increased (p < 0.05) in the MAS+ and MAS++ and DSM+ and DSM++ groups when compared to the control group. Similarly, a significant increase was noted in hemoglobin when DSM was supplemented when compared to the control one. Moreover, the number of heterophils increased (p < 0.05) in groups of MAS and DSM when compared to the control group. The percentage of phagocytic activity increased significantly by 5.39, 6.90, and 7.18% in MAS++, DSM+, and DSM++, respectively, relative to the control group. On the other hand, the phagocytic number decreased (p < 0.05) by 1.15 and 1.12 conditional units in MAS+ and MAS++ and by 1.03 and 0.83 conditional units in DSM+ and DSM++ when compared to the control group, respectively. The urgent need to remediate ocean acidification has brought attention to the ability of marine macrophytes (seagrasses and seaweeds) to take up carbon dioxide (CO2) and locally raise seawater pH via primary production. This physiological process may represent a powerful ocean acidification mitigation tool in coastal areas. However, highly variable nearshore environmental conditions pose uncertainty in the extent of the amelioration effect. We developed experiments in aquaria to address two interconnected goals. First, we explored the individual capacities of four species of marine macrophytes (Ulva lactuca, Zostera marina, Fucus vesiculosus and Saccharina latissima) to ameliorate seawater acidity in experimentally elevated pCO2. Second, we used the most responsive species (i.e., S. latissima) to assess the effects of high and low water residence time on the amelioration of seawater acidity in ambient and simulated future scenarios of climate change across a gradient of irradiance. We measured changes in dissolved oxygen, pH, and total alkalinity, and derived resultant changes to dissolved inorganic carbon (DIC) and calcium carbonate saturation state (Ω). While all species increased productivity under elevated CO2, S. latissima was able to remove DIC and alter pH and Ω more substantially as CO2 increased. Additionally, the amelioration of seawater acidity by S. latissima was optimized under high irradiance and high residence time. However, the influence of water residence time was insignificant under future scenarios. Finally, we applied predictive models as a function of macrophyte biomass, irradiance, and residence time conditions in ambient and future climatic scenarios to allow projections at the ecosystem level. Algae are a rich but unexplored source of fibers with the potential to contribute to the next generation of prebiotics. The sulfated brown algae polysaccharide, fucoidan, is mainly composed of the deoxy-hexose L-fucose, which can be metabolized to 1,2-propanediol (1,2-PD) or lactate by gut microbes as precursors of propionate and butyrate. It was the aim of this study to investigate the impact of fucoidan on the fermentation capacity of the fecal microbiota and to compare to fucose. In batch fermentations of fecal microbiota collected from 17 donor samples, fucose promoted the production of propionate while no consistent effect was observed for commercial fucoidan and Fucus vesiculosus extract prepared in this study containing laminarin and fucoidan. H2S production was detected under all tested conditions, and levels were significantly lower in the presence of fucose in a dose-dependent manner. The addition of high fucose levels led to higher relative abundance of microbial 1,2-PD and lactate cross-feeders. Our results highlight that fucose and not fucoidan addition impacted fermentation capacity and increased the proportions of propionate and butyrate, which allows for precise modulation of intestinal microbiota activity. In the Archipelago Sea as in most other parts of the Baltic Sea, the bladder wrack (Fucus vesiculosus) is a foundation species of the littoral communities of the rocky shores. It sustains a community of epiphytic algae, herbivorous crustaceans and molluscs and various fish. Recently we have noticed a steep decline in the occurrence of the herbivorous crustaceans and molluscs in many sites in the Archipelago Sea. We hypothesise that a key factor contributing to this decline is the recent introduction of the Harris mud crab (Rhithropanopeus harrisii), which was first sighted in 2009 in this region. Importantly, because there are no native crabs in the northern parts of the Baltic Sea, the mud crab is a completely novel kind of predator in the ecosystem and the herbivorous crustaceans and molluscs may be particularly susceptible to it. Here, we document a dramatic decline of the typical herbivores occurring on the bladder wrack, possibly indicating an ongoing regime shift, by comparing our recent samples from across the Archipelago Sea with data collected a decade before the sighting of the mud crab. Moreover, we demonstrate a spatio-temporal association between the decline, particularly of the key herbivore species, the isopod Idotea balthica, and the establishment of the mud crab. We also present experimental evidence for a strong predator-prey -link between the mud crab and the isopod I. balthica. Marine algae are sources of bioactive components with defensive properties of great value against microbial infections. This study investigated the bioactivity of extracts from brown algae Fucus vesiculosus against rotavirus, the worldwide leading cause of acute gastroenteritis in infants and young children. Moreover, one of the extracts was tested against four foodborne bacteria: Campylobacter jejuni, Escherichia coli, Salmonella Typhimurium, and Listeria monocytogenes, and the non-pathogenic: E. coli K12. In vitro tests using MA104 cells revealed that both whole algae extracts and crude fucoidan precipitates neutralized rotavirus in a dose-responsive manner. The maximum neutralization activity was observed when the rotavirus was incubated with 100 μg mL−1 of the hydrochloric acid-obtained crude fucoidan (91.8%), although crude fucoidan extracted using citric acid also demonstrated high values (89.5%) at the same concentration. Furthermore, molecular weight fractionation of extracts decreased their antirotaviral activity and high molecular weight fractions exhibited higher activity compared to those of lower molecular weight. A seaweed extract with high antirotaviral activity was also found to inhibit the growth of C. jejuni, S. Typhimurium, and L. monocytogenes at a concentration of 0.2 mg mL−1. The brown macroalga Fucus vesiculosus is a foundation species in temperate rocky shores, subjected to seasonally fluctuating environmental conditions. To obtain a more complete picture of the seasonality of F. vesiculosus ecophysiology in the northern Baltic Sea, in situ photochemistry, elemental ratios and chlorophyll a and c content of the alga were investigated in field campaigns conducted in different months throughout the year during 2017. Carbon, nitrogen, carbon to nitrogen ratio and chlorophyll a and c content of the alga varied substantially throughout the year, with highest carbon content observed in summer, and highest nitrogen content in winter. C:N ratio in F. vesiculosus apical tissue ranged from 8.6 in February to 48.3 in July. Chlorophyll a and c content followed inversely the seasonal patterns of ambient irradiance. High chlorophyll a and c content in winter was associated with higher maximum photosynthetic efficiency of energy conversion (Fv/Fm), but not with efficiency of photosynthetic energy conversion under light limitation (α). Electron transport rate correlated strongly with seawater temperature, and the highest electron transport rates were observed in summer and correlated with highest internal carbon content of the alga. Redundancy analysis conducted on measured environmental variables against physiological responses identified day of year, temperature and macronutrients in seawater as the most important variables driving the observed seasonal patterns in F. vesiculosus ecophysiology. The results suggest elevated temperatures may increase Fucus growth and photosynthesis rates in the study area. Highlights Two distinct physiological states of Fucus vesiculosus were identified. Background: Snakebite envenomation (SBE) causes diverse toxic effects in humans, including disability and death. Current antivenom therapies effectively prevent death but fail to block local tissue damage, leading to an increase in the severity of envenomation; thus, seeking alternative treatments is crucial. Methods: This study analyzed the potential of two fucoidan sulfated polysaccharides extracted from brown seaweeds Fucus vesiculosus (FVF) and Undaria pinnatifida (UPF) against the fibrinogen or plasma coagulation, proteolytic, and phospholipase A2 (PLA2) activities of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom. The toxicity of FVF and UPF was assessed by the hemocompatibility test. Results: FVF and UPF did not lyse human red blood cells. FVF and UPF inhibited the proteolytic activity of Bothrops jararaca, B. jararacussu, and B. neuwiedi venom by approximately 25%, 50%, and 75%, respectively, while all venoms led to a 20% inhibition of PLA2 activity. UPF and FVF delayed plasma coagulation caused by the venoms of B. jararaca and B. neuwiedi but did not affect the activity of B. jararacussu venom. FVF and UPF blocked the coagulation of fibrinogen induced by all these Bothropic venoms. Conclusion: FVF and UPF may be of importance as adjuvants for SBE caused by species of Bothrops, which are the most medically relevant snakebite incidents in South America, especially Brazil.

High blood cholesterol levels are a major risk factor for cardiovascular diseases. A purified aqueous extract of Fucus vesiculosus, rich in phlorotannins and peptides, has been described for its potential to inhibit cholesterol biosynthesis and intestinal absorption. In this work, the effect of this extract on intestinal cells’ metabolites and proteins was analysed to gain a deeper understanding of its mode of action on lipids’ metabolism, particularly concerning the absorption and transport of exogenous cholesterol. Caco-2 cells, differentiated into enterocytes, were exposed to the extract, and analysed by untargeted metabolomics and proteomics. The results of the metabolomic analysis showed statistically significant differences in glutathione content of cells exposed to the extract compared to control cells, along with an increased expression of fatty acid amides in exposed cells. A proteomic analysis showed an increased expression in cells exposed to the extract compared to control cells of FAB1 and NPC1, proteins known to be involved in lipid metabolism and transport. To the extent of our knowledge, this study is the first use of untargeted metabolomics and a proteomic analysis to investigate the effects of F. vesiculosus on differentiated Caco-2 cells, offering insights into the molecular mechanism of the extract’s compounds on intestinal cells.

The worldwide prevalence of obesity impacts more than 600 million adults. Successfully managing weight is effective in reducing the risk of chronic diseases, but sustaining long-term weight loss remains a challenge. Although there are supplements based on algae that claim to aid in weight loss, there is a notable scarcity of scientific evidence supporting their effectiveness, and their regular consumption safety remains inadequately addressed. In this work, commercially available Arthrospira (Spirulina) platensis Gomont and/or Fucus vesiculosus L. supplements showed moderate capacity to inhibit the activity of carbohydrate-metabolizing enzymes, and to scavenge biologically relevant reactive species. IC25 values varying between 4.54 ± 0.81 and 66.73 ± 5.91 µg of dry extract/mL and between 53.74 ± 8.42 and 1737.96 ± 98.26 µg of dry extract/mL were obtained for α-glucosidase and aldose reductase, respectively. A weaker effect towards α-amylase activity was observed, with a maximum activity of the extracts not going beyond 33%, at the highest concentrations tested. Spirulina extracts showed generally better effects than those from F. vesiculosus. Similar results were observed concerning the antiradical capacity. In a general way, the extracts were able to intercept the in vitro-generated reactive species nitric oxide (•NO) and superoxide anion (O2•−) radicals, with better results for O2•−scavenging with the spirulina samples (IC25 values of 67.16 and 122.84 µg of dry extract/mL). Chemically, similar pigment profiles were observed between spirulina supplements and the authenticated counterpart. However, fucoxanthin, the chemotaxonomic marker of brown seaweeds, was not found in F. vesiculosus samples, pointing to the occurrence of a degradation phenomenon before, during, or after raw material processing. Our findings can contribute to providing data to allow regulatory entities (e.g., EFSA and FDA) to better rule these products in a way that can benefit society.

Brown macroalgae are rich sources of nutrients and health-promoting compounds. Nevertheless, their consumption is still limited by their strong organoleptic characteristics, thus requiring the development of extraction strategies to profit from their nutritional value. To fulfil this, two sequential extraction approaches were developed, differing in the solvent used in the first extraction step, water in approach 1 or food-grade ethanol in approach 2, to obtain economic and affordable extracts rich in specific compounds from Fucus vesiculosus. The use of water in the first step of extraction allowed us to recover water-soluble phlorotannins, laminarans and mannuronic-rich alginates, making the subsequent 70% ethanol extract richest in fucoxanthin (0.07% algae DW), and the hot water fractions purest in fucoidans and alginates with a lower mannuronic-to-guluronic (M/G) ratio (2.91). Conversely, when beginning extraction procedures with 96% ethanol, the recovered yields of phlorotannins increased (0.43 g PGE/100 g algae DW), but there was a concomitant seven-fold decrease in the recovery of fucoxanthin in the subsequent 70% ethanol extract. This approach also led to less pure hot water fractions containing fucoidans, laminarans and alginates with a higher M/G ratio (5.50). Overall, this work unveiled the potential of the first extraction steps in sustainable and holistic cascade strategies to modulate the composition of food-grade extracts, creating prospects of their application as tailored functional ingredients in food products.

Why species that in their core areas mainly reproduce sexually become enriched with clones in marginal populations ("geographic parthenogenesis") remains unclear. Earlier hypotheses have emphasized that selection might promote clonality because it protects locally adapted genotypes. On the other hand, it also hampers recombination and adaptation to changing conditions. The aim of the present study was to investigate the early stages of range expansion in a partially clonal species and what drives an increase in cloning during such expansion. We used genome‐wide sequencing to investigate the origin and evolution of large clones formed in a macroalgal species (Fucus vesiculosus) during a recent expansion into the postglacial Baltic Sea. We found low but persistent clonality in core populations, while at range margins, large dominant clonal lineages had evolved repeatedly from different sexual populations. A range expansion model showed that even when asexual recruitment is less favourable than sexual recruitment in core populations, repeated bottlenecks at the expansion front can establish a genetically eroded clonal wave that spreads ahead of a sexual wave into the new area. Genetic variation decreases by drift following repeated bottlenecks at the expansion front. This results in the emerging clones having low expected heterozygosity, which corroborated our empirical observations. We conclude that Baker's Law (clones being favoured by uniparental reproductive assurance in new areas) can play an important role during range expansion in partially clonal species, resulting in a complex spatiotemporal mosaic of clonal and sexual lineages that might persist during thousands of generations. A brown seaweed consumed worldwide, Fucus vesiculosus, has been used to prevent atherosclerosis and hypercholesterolemia, among other uses. However, the mechanisms of action that lead to these effects are not yet fully understood. This work aims to study the in vitro effect of an aqueous extract of F. vesiculosus, previously characterized as rich in phlorotannins and peptides, on the expression of different proteins involved in the synthesis and transport of cholesterol. A proteomic analysis, Western blot, and qRT-PCR analysis were performed to identify protein changes in HepG2 cells exposed to 0.25 mg/mL of the F. vesiculosus extract for 24 h. The proteomic results demonstrated that, in liver cells, the extract decreases the expression of four proteins involved in the cholesterol biosynthesis process (CYP51A1, DHCR24, HMGCS1 and HSD17B7). Additionally, a 12.76% and 18.40% decrease in the expression of two important transporters proteins of cholesterol, NPC1L1 and ABCG5, respectively, was also observed, as well as a 30% decrease in NPC1L1 mRNA levels in the cells exposed to the extract compared to control cells. Our study reveals some of the mechanisms underlying the actions of bioactive compounds from F. vesiculosus that may explain its previously reported hypocholesterolemic effect, future prospecting its use as a functional food. Fucoidans are discussed as antiviral agents, and fucoidan from Undaria pinnatifida (UpF), in particular has gained interest as potential food additive in antinoroviral strategies. As the competitive blocking activity of antinoroviral agents increases with the valency of terminal nonreducing fucose on the competitor, an effective processing of fucoidans to inhibitory oligosaccharides will depend on basic structural features of the polysaccharide. We demonstrate increased antiviral binding activity of processed low-mass UpF generated by hydrothermal degradation contrasting with decreased efficacy of low-mass fucoidan from Fucus vesiculosus. As this finding is in conflict with current structural models of UpF, we undertook a re-investigation of the glycan backbone in UpF. Applying solvolytical desulfation combined with enzymatic cleavage of low-mass fucoidan by endo-β6-galactanase and terminal labeling of oligosaccharides by deutero-reduction and bis-5-phenyl-3-methyl-1-pyrazolone (PMP) substitution, evidence from mass spectrometry and methylation linkage analysis of the oligosaccharides indicates that fucoses and galactoses in the glycan backbone are organized in homomeric blocks, where oligo-fucoses branch off from a galactane-type core: Fuc(1-3Fuc)n1-3[Gal(1-6Gal)n1-6]Gal(1-6Gal)n. Dietary variability and the degradation and incorporation of macroalgae in key macroinvertebrate consumers were examined (1) in a monitoring field study including a natural attached canopy habitat and an adjacent habitat receiving natural accumulations of detritus, and (2) in a manipulative in situ experiment of macroalgal detritus at two different depths (3 and 6 m) in the archipelago of SW Finland. The monitoring field study, examining species-specific dietary responses across three sampling dates in natural macroalgal stands, showed that a pulse of drifting filamentous macroalgae shaped the dietary compositions of the abundant benthic macroinvertebrate consumers and that accumulations of drifting filamentous macroalgae were rapidly incorporated into the food web through epigrazers. The in situ field experiment simulating a natural accumulation event and the degradation process of Fucus vesiculosus during 60 days showed that algal decomposition progressed relatively slowly at both depths. Detectable increasing incorporation of Fucus-derived matter to epigrazers and detritivorous bivalves occurred after 2−3 weeks, while simultaneously the incorporation of filamentous algae decreased over time. Hence, the ecological role of decomposing F. vesiculosus might be more important in areas where the algal matter can accumulate for several months. The effect of depth influenced the food incorporation of typical epigrazers. The increasing depth from 3 to 6 m lowered the median proportion of Fucus-derived matter incorporated into the macrofauna community approximately by 10% points compared to the shallower depth of 3 m. A fucoidan discovered in the plant Fucus vesiculosus, which lowered the molecular weight of fucoidan, was ideal for its application in the pharmaceutical and food sectors. The aim was to study the impact of ultrasound process parameters on the molecular weight, structure, and antioxidant activity of fucoidan. For optimization of sonochemical process parameters such as temperature, sonication time, and power (intensity), Box–Behnken design (BBD) through the response surface method (RSM) at fixed fucoidan concentrations was compared with a normal process. The outcomes demonstrated that sonochemical treatment significantly decreased molecular weight (Mw) to 318 kDa compared to the control process (815 kDa). Antioxidant activity tests revealed that the sonication treatment significantly increased antioxidant activity (88.9% compared to 65.3% with the control process). Through use of the BBD model, we found that the ideal conditions for degradation of fucoidan were a temperature of 33 °C, sonication time of 40 min, and sonication power of 102.5 W/cm2. Under these conditions, the quadratic model was fitted and the experimental values for Mw and antioxidant activity (318 kDa and 87.4%) were close to the predicated values (316 kDa and 87.9%). According to the findings, sonication treatment is a useful method for lowering fucoidan levels with no observable changes in the monosaccharide units of fucoidan through scanning electron microscope, X-Ray diffraction, and Fourier-transform infrared (FTIR) analysis. A comprehensive approach to the management of brown seaweed—Fucus vesiculosus was presented. An algal extract, which served as a biostimulant of plant growth was produced using ultrasound-assisted extraction (UAE). The concentration of the extract (20, 40, 60, 80, 100%), which had the greatest influence on biometric parameters of radish, was determined in germination tests. The seaweed itself as well as the produced post-extraction residue were used in doses of 2 and 4 g/kg as soil additives, stimulating plant growth in the initial phase. Pot tests for sorghum carried out under optimal conditions (20% extract and 2 g/kg of soil additive) had a positive effect on the plant weight, length and the content of chlorophyll in comparison with the control group treated with distilled water. Additionally, preliminary studies on the bioremediation of soil contaminated with Zn(II) ions with the use of both soil additives were performed. It was shown that the immobilization of Zn(II) ions in the soil by the applied additives reduced the bioaccumulation of zinc in the aerial part of plants as compared with the group cultivated in the contaminated soil but without additive. Accordingly, by producing plant biostimulants by UAE it was also possible to successfully manage the post-extraction residue following the concept of a bio-based economy. [ABSTRACT FROM AUTHOR]
