Underground telecommunication cables are highly susceptible to damage from excavation activities. Preventing accidental damage to underground telecommunication cables is critical and necessary. In this study, we present field trial results of monitoring excavation activities near underground fiber cables using an intensity-based phase-sensitive optical time-domain reflectometer (φ-OTDR). The reasons for choosing intensity-based φ-OTDR for excavation monitoring are presented and analyzed. The vibration signals generated by four typical individual excavation events, i.e., cutting, hammering, digging, and tamping at five different field trial sites, as well as five different mixed events in the fifth field trial site were investigated. The findings indicate that various types of events can generate vibration signals with different features. Typically, fundamental peak frequencies of cutting, hammering and tamping events ranged from 30 to 40 Hz, 11 to 15 Hz, and 30 to 40 Hz, respectively. Digging events, on the other hand, presented a broadband frequency spectrum without a distinct peak frequency. Moreover, due to differences in environmental conditions, even identical excavation events conducted with the same machine may also generate vibration signals with different characteristics. The diverse field trial results presented offer valuable insights for both research and the practical implementation of excavation monitoring techniques for underground cables. The aim of the present study was to monitor the dynamics and to measure the safety and efficacy of a live, attenuated, thermosensitive Mycoplasma anserisalpingitidis vaccine candidate, namely MA271, in geese breeder flocks under field conditions. Two rearing flocks were vaccinated with MA271 at 4 weeks of age and boosted at 24 weeks of age by cloaca inoculation (1 ml) and eye-dropping (60 µl). The geese then were transported to multi-aged breeding farms. Two breeding flocks served as controls. Colonization of the cloaca by MA271 showed 75% maximum prevalence between 4 and 6 weeks after the first vaccination. Then the prevalence decreased to 25% until the cooler, humid fall months which coincided with the booster vaccination. Boosting raised cloacal colonization to 100%. No clinical signs were observed in the vaccinated birds. After transportation to five multi-aged breeding farms, the wild-type strain appeared as well as MA271 in three flocks. In one flock, the wild-type strain completely displaced MA271, while in one flock only MA271 was detected. Only wild-type strains were detected in the control flocks; however, due to an HPAI outbreak, both flocks were exterminated before the end of the study. Based on the available data, the median percentage of infertile eggs was 3.7-5.1% in the MA271 vaccinated flocks, and 7.7% in the non-vaccinated flock. In conclusion, MA271 can colonize the cloaca of geese under field conditions. MA271 proved to be safe and presumably protects against M. anserisalpingitidis-induced reproduction losses. Land reclamations often demand large quantities of fill materials. However, many countries commonly engaged in land reclamation are facing an escalating scarcity of natural fill sources. To address this issue, this study proposes a sustainable land reclamation approach that involves dredged marine sediments as fill materials. The dredged marine sediments are improved by a combined method that incorporates the use of horizontal drains with vacuum preloading for preliminary soil treatment and vertical drains with vacuum preloading for further improvement. A field trial has been designed and conducted to verify the performance of the proposed preliminary treatment of marine sediment slurry using prefabricated horizontal drains (PHDs) and vacuum preloading under a "membrane-free" condition. Field measurements, including settlement, pore pressure, effective earth pressure, vacuum pressure, water content, and undrained shear strength, were systematically recorded during the field trial period. After a four-month treatment, the average water content of the filled soil was slightly below 50%, which is one-third of the initial water content. Owing to the combined effect of the vacuum preloading and drying process, a crust layer with the undrained shear strength of 40 kPa was formed on the top of the treated soil. Below the crust layer, the average undrained shear strength of the treated marine deposits was approximately 32 kPa. Overall, the notable effect of using PHDs and the "membrane-free" vacuum preloading method to rapidly consolidate marine deposits slurry has been successfully demonstrated with field monitoring under the site trial condition. There has always been a specific focus on nitrogen fertilization in sugar beet production due to its important effect on sugar beet root yield and quality. For stable sugar beet growth and satisfactory root yield and quality, balanced N fertilization is crucial. Thus, this study aimed to investigate spring N fertilization in two seasons as the following treatments: N0—control, N1—only pre-sowing fertilization, and N2—pre-sowing with topdressing. Four different genotypes were included in the study (Serenada, Colonia, Fred, and Danton). The experiment was set up in a plain area, belonging to the temperate climate zone in Eastern Croatia (Županja and Vrbanja), with the long-term mean (LTM) (March–October) air temperature around 16 °C and the total precipitation of 515 mm. Pre-sowing N fertilization had a smaller impact on root yield in the year with higher precipitation (31% higher than LTM). Therefore, the average yields with pre-sowing fertilization (N1) and pre-sowing fertilization with top dressing (N2) were very similar and were only 7% higher than those of the control. In a season with less rainfall (29% less than LTM), pre-sowing fertilization with top dressing (N2) had a more pronounced effect on the increase in sugar beet root yield, which was 17% higher compared to that of the control treatment. The sugar beet sucrose content and quality parameters (brei impurities, loss of sugar in molasses, extractable sugar) differed when N fertilization was applied among locations in both seasons. The white sugar yield was the highest at N2 treatment with pre-sowing and topdressing N fertilization. In general, according to the average of all locations and years of research, the Serenada hybrid achieved the highest average root yield (81.1 t ha−1), while Colonia exhibited the highest root sugar content (14.5%) and white sugar yield (9.7 t ha−1).

BACKGROUND: Ultra‐low volume (ULV) space sprays aerosolize insecticide formulated products (FP) to contact flying mosquitoes, while barrier sprays expose mosquitoes to FP residue on vegetation and other surfaces. Centers for Disease Control and Prevention bottle bioassays used to assess insecticide resistance are based on residual active ingredient (AI) exposure and do not directly relate to FP efficacy. The current pilot study developed a novel compact wind tunnel for mosquito exposure to FP. Caged Aedes albopictus and Culex pipiens/quinquefasciatus were exposed to undiluted Biomist®3 + 15 FP (permethrin AI) or air (control) within the wind tunnel, transferred to new cages, and held in a 28 °C incubator. Separate mosquitoes were exposed to residual permethrin AI (8 μg mL−1) in bottle bioassays. Mortality was monitored 15, 30, 60, and 120 min post‐exposure. RESULTS: Chi‐square tests (P < 0.05) showed significantly higher mortality in Aedes compared to Culex populations for most time points in both bioassay and wind tunnel exposure groups. As expected, mosquitoes exposed to Biomist®3 + 15 showed higher mortality rates than bottle bioassay exposure to permethrin. Two Culex colonies resistant to permethrin in bottle bioassays were susceptible to Biomist®3 + 15 in the wind tunnel. CONCLUSION: The novel compact wind tunnel developed here may be an alternative to field trials for testing FP efficacy, avoiding factors such as weather, logistical planning, and extended personnel hours. The wind tunnel could allow programs to conveniently test efficacy of multiple FP. During the previous field study with a robot and its interaction with mall visitors, we observed a surprising event during which a leaflet-distributing robot was abused, although it was subsequently helped by one of its previous abusers. After analyzing 72.25 hours of video data, we identified 47 cases where a robot dropped a leaflet and classified them according to following three criteria: 1) interaction between the potential helper or others with the robot before it dropped the leaflet, 2) the nature of the interaction (abused or not), and 3) whether it was helped. Using the Trajectory Equifinality Model (TEM), we analyzed 19 cases where the robot was helped. We identified the following interaction process that started with individuals who paid attention to the robot, whether they had abusive or non-abusive interactions with it, whether they noticed its failure, and finally whether they helped it. The presence of others encouraged the person to focus on the robot, and the interactions with it led to helping, regardless whether the interaction was abusive. The absence of others when the robot dropped the leaflet encouraged helping. The findings of this study will motivate interaction designs for social robots that can leverage human help. An outbreak of camelpox occurred in the Mangistau region of Kazakhstan in 2019. To control the outbreak of camelpox and to prevent its further spread to other regions, camels were vaccinated using live and inactivated camelpox vaccines produced in Kazakhstan. To evaluate the efficacy of these camelpox vaccines in the field, vaccine trials used 172 camels on camel farms in the Beineu district. Of these, 132 camels were vaccinated using a live attenuated camelpox vaccine and 40 camels were vaccinated using an inactivated vaccine to observe immunogenicity and safety. The live vaccine was inoculated into camels by scarification at a dose of 5 × 104 EID50, and the inactivated vaccine was injected intramuscularly at 5 mL twice, with an interval of 35 days. During the safety evaluation, camels administered either vaccine displayed no clinical signs of illness or any adverse effects. Post-vaccination seroconversion demonstrated that the live attenuated vaccine started to elicit antibody responses in some animals as early as day seven, while, by day 28, 99% of vaccinated camels responded. For camels immunized with the inactivated vaccine, seroconversion began on day 21 at low titers ranging from 1:2 to 1:4. Ninety days post vaccination, 77% of the camels demonstrated an immune response that was up to a titer of 1:16. The antibody response waned six months post vaccination in camels vaccinated with two types of vaccine. Nonetheless, both vaccines were 100% effective at preventing clinical disease in vaccinated camels during the camelpox outbreak. All unvaccinated camels became ill, with manifestations of clinical signs characteristic of camelpox. Following these successful field trials in Kazakhstan, a vaccination program for camels, to control camelpox using the domestically produced live attenuated camelpox vaccine, has started. Red seabream iridovirus (RSIV) is a major cause of marine fish mortality in Korea, with no effective vaccine available since its first occurrence in the 1990s. This study evaluated the efficacy of a formalin-killed vaccine against RSIV in rock bream under laboratory and field conditions. For the field trial, a total of 103,200 rock bream from two commercial marine cage-cultured farms in Southern Korea were vaccinated. Farm A vaccinated 31,100 fish in July 2020 and monitored them for 18 weeks, while farm B vaccinated 30,700 fish in August 2020 and monitored them for 12 weeks. At farm A, where there was no RSIV infection, the vaccine efficacy was assessed in the lab, showing a relative percentage of survival (RPS) ranging from 40% to 80%. At farm B, where natural RSIV infections occurred, cumulative mortality rates were 36.43% in the vaccinated group and 80.32% in the control group, resulting in an RPS of 54.67%. The RSIV-infectious status and neutralizing antibody titers in serum mirrored the cumulative mortality results. This study demonstrates that the formalin-killed vaccine effectively prevents RSIV in cage-cultured rock bream under both laboratory and field conditions. [ABSTRACT FROM AUTHOR]

Introgression of resistance genes from wild or related species is a common strategy to improve disease resistance of wheat cultivars. Pm17 is a gene that confers powdery mildew resistance in wheat. It encodes an NLR type of immune receptor and was introgressed from rye to wheat as part of the 1RS chromosome arm translocation several decades ago. So far it has not been possible to separate Pm17 from its co-introgressed rye genes due to suppressed recombination. Here we tested in the field transgenic Bobwhite wheat overexpressing Pm17 without any other rye genes. Four transgenic events showed high levels of PM17 protein accumulation, strong powdery mildew resistance, and no pleiotropic effects during three field seasons. We used a combined approach of transgene insertion and cross-breeding to generate lines co-expressing Pm17 and Pm3 , or Pm17 and Pm8. Blumeria graminis f. sp. tritici infection tests confirmed additive, race-specific resistance of the two pyramided transgenes in lines Pm17+Pm3b and Pm17+Pm8. Furthermore, pyramided lines showed strong powdery mildew resistance during three field seasons. We conclude that the combination of overexpressed NLR genes from the extended gene pool broadens and diversifies wheat disease resistance. [ABSTRACT FROM AUTHOR]

Naturally occurring variability within a study region harbors valuable information on relationships between biological variables. Yet, spatial patterns within these study areas, e.g., in field trials, violate the assumption of independence of observations, setting particular challenges in terms of hypothesis testing, parameter estimation, feature selection, and model evaluation. We evaluate a number of spatial regression methods in a simulation study, including more realistic spatial effects than employed so far. Based on our results, we recommend generalized least squares (GLS) estimation for experimental as well as for observational setups and demonstrate how it can be incorporated into popular regression models for high-dimensional data such as regularized least squares. This new method is available in the BioConductor R-package pengls. Inclusion of a spatial error structure improves parameter estimation and predictive model performance in low-dimensional settings and also improves feature selection in high-dimensional settings by reducing “red-shift”: the preferential selection of features with spatial structure. In addition, we argue that the absence of spatial autocorrelation (SAC) in the model residuals should not be taken as a sign of a good fit, since it may result from overfitting the spatial trend. Faba bean (Vicia faba L.) is a legume crop grown in diverse climates worldwide. It has a high potential for increased cultivation to meet the need for more plantbased proteins in human diets, a prerequisite for a more sustainable food production system. Characterization of diversity panels of crops can identify variation in and genetic markers for target traits of interest for plant breeding. In this work, we collected a diversity panel of 220 accessions of faba bean from around the world consisting of gene bank material and commercially available cultivars. The aims of this study were to quantify the phenotypic diversity in target traits to analyze the impact of breeding on these traits, and to identify genetic markers associated with traits through a genome-wide association study (GWAS). Characterization under field conditions at Nordic latitude across two years revealed a large genotypic variation and high broad-sense heritability for eleven agronomic and seed quality traits. Pairwise correlations showed that seed yield was positively correlated to plant height, number of seeds per plant, and days to maturity. Further, susceptibility to bean weevil damage was significantly higher for early flowering accessions and accessions with larger seeds. In this study, no yield penalty was found for higher seed protein content, but protein content was negatively correlated to starch content. Our results showed that while breeding advances in faba bean germplasm have resulted in increased yields and number of seeds per plant, they have also led to a selection pressure towards delayed onset of flowering and maturity. DArTseq genotyping identified 6,606 single nucleotide polymorphisms (SNPs) by alignment to the faba bean reference genome. These SNPs were used in a GWAS, revealing 51 novel SNP markers significantly associated with ten of the assessed traits. Three markers for days to flowering were found in predicted genes encoding proteins for which homologs in other plant species regulate flowering. Altogether, this work enriches the growing pool of phenotypic and genotypic data on faba bean as a valuable resource for developing efficient breeding strategies to expand crop cultivation. Service robots are used in a range of service situations, including those where they are required to recommend products. However, when robots use subjective statements for product recommendations, users may not place sufficient trust in the robot's recommendations which may degrade its recommendation capability. This study proposes a method to improve the perceived trustworthiness of the robot's subjective statements and thus promote the user's food purchase behavior. The proposed recommendation method involves explicitly sharing the robot's experience with the user and improving the perceived trustworthiness of the robot's subjective statements by performing a pseudo-eating behavior. The results of two online surveys show that the robot's statements and recommendations accompanied by the pseudo-eating behavior improved the trust in the robot's subjective statement and consequently, the user's willingness to purchase products. Further, the results of a field experiment in a real bakery store showed that the proposed method is comparable to recommendation methods proposed in previous works. The results obtained through the online surveys and the field experiment indicate that robots are trusted even when they express subjective statements for recommendations by explicitly sharing the experience with the user. Abstract. Plants function in symbiosis with numerous microorganisms, which might contribute to their adaptation and performance. In this study, we tested whether fungal strains in symbiotic interaction with roots of Celtica gigantea , a wild grass adapted to nutrient-poor soils in semiarid habitats, could improve the field performance of the agricultural cereal tritordeum (Triticum durum  ×  Hordeum chilense). Seedlings of tritordeum were inoculated with 12 different fungal strains isolated from roots of Celtica gigantea that were first proved to promote the growth of tritordeum plants under greenhouse conditions. The inoculated seedlings were transplanted to field plots at two locations belonging to different climatic zones in terms of mean temperatures and precipitation in the Iberian Peninsula. Only one strain, Diaporthe iberica T6, had a significant effect on plant height, number of tillers and grain yield in one location. This result showed a substantial divergence between the results of greenhouse and field tests. In terms of grain nutritional quality, several parameters were differentially affected at both locations: Diaporthe T6, Pleosporales T7, Zygomycota T29 and Zygomycota T80 increased the content of total carotenoids, mainly lutein, in the colder location; whereas gluten proteins increased with several treatments in the warmer location. In conclusion, early inoculation of tritordeum plants with fungal symbionts had substantial beneficial effects on subsequent plant growth and development in the field. Drosophila suzukii Matsumura (Diptera: Drosophilidae) is a polyphagous invasive vinegar fly. Females can penetrate certain soft-skinned grapevine varieties with their strong serrated ovipositor to lay their eggs within berries, which eventually leads to the decay and collapse of grapes. Alternative pest control options were tested in practically oriented field trials set up in two vineyards in Styria. The effect of the mineral particle films diatomaceous earth, kaolin and muscovite mica was compared to an untreated control. All products were complemented with wetting agents to increase the efficacy and/or the rain resistance of the film. Due to the long waiting times after application in organic production, only some of the trials also comprised a comparable insecticide variant (Spinosad). Treatments were applied from the beginning of infestations until harvest, and berries were sampled once to twice a week, depending on pest pressure. Evaluation of treatment effects was based on a microscopic analysis of berries for oviposition and a visual evaluation of the clusters. Both kaolin and diatomaceous earth led to a statistically significant reduction in oviposition and to a significantly improved macroscopic appearance of clusters compared to the control, whereas the effect of muscovite mica was negligible. A direct comparison of particle film and spinosad treatment was only expedient in one experiment, and in this case, the effect of kaolin on oviposition was significantly superior. Kaolin and diatomaceous earth might complement established integrated pest management practices against Drosophila suzukii and contribute to the reduction of chemical plant protection agents in the future.

Abstract Metabolic engineering for hyperaccumulation of lipids in vegetative tissues of high biomass crops promises a step change in oil yields for the production of advanced biofuels. Energycane is the ideal feedstock for this approach due to its exceptional biomass production and persistence under marginal conditions. Here, we evaluated metabolically engineered energycane with constitutive expression of the lipogenic factors WRINKLED1 (WRI1), DIACYLGLYCEROL ACYLTRANSFERASE1 (DGAT1), and OLEOSIN1 (OLE1) for the accumulation of triacylglycerol (TAG), total fatty acid (TFA), and biomass under field conditions at the University of Florida‐IFAS experiment station near Citra, Florida. TAG and TFA accumulation were highest in leaves (up to 9.9% and 12.9% of DW, respectively), followed by juice from crushed stems, stems, and roots. TAG and TFA accumulation increased up to harvest time and correlated highest with OLE1 and DGAT1 expression. Biomass dry weight, TAG, and TFA content differed greatly depending on DGAT1 and OLE1 expression in transgenic lines with similar WRI1 expression. Biomass did not significantly differ between WT and line L2 with DAGT1 and OLE1 expressed at low levels and TAG and TFA accumulating to 12‐ and 1.6‐fold that of WT leaves, respectively. In contrast, line L13, with intron‐mediated enhancement of DGAT1 expression, displayed a 245‐ to 330‐fold increase in TAG and a 4.75‐ to 6.45‐fold increase in TFA content compared with WT leaves and a biomass reduction of 52%. These results provide the basis for developing novel feedstocks for expanding plant lipid production and point to new prospects for advanced biofuels.

The capsule expansion technique (CET) is a novel active measure to control the deformation of subsurface structures induced by underground engineering construction. CET has been applied to tunnel deformation control, but its application in pile foundations is rarely reported. In this paper, a field trial and three-dimensional coupled consolidation finite-element analysis were performed to study the interaction of capsule–soil–pile and control efficiency of CET. Field trial results verify the feasibility of CET in controlling pile horizontal deformation, and the control efficiency is 60% higher than that of Tube-a-Manchette grouting with 40%. Numerical back analyses indicate that the pile maximum displacement increases almost linearly with the expansion diameter, while the control efficiency of CET hardly changes. The total stress of the soil in front of the pile is significantly reduced by 20.3% due to the dissipation of excess pore-water pressure, resulting in the reverse displacement of the pile. High excess pore pressure induced by CET would cause the soil to bear additional stress after dissipating, leading to the reverse displacement of the soil and further diminishing the control efficiency. Moreover, control efficiency can be improved by reducing the surrounding excess pore-water pressure induced by expansion and increasing the excess pore-water pressure at the back side of the pile. Management of the raccoon rabies virus variant in North America is conducted primarily using oral rabies vaccination (ORV). When a sufficient proportion of the population is vaccinated ((60%), rabies transmission can be eliminated. To date, ORV programs have successfully controlled and eliminated raccoon rabies in rural areas, but there has been less success in urban areas. We studied the proportions of rabies virus neutralizing antibodies (RVNA) in a raccoon (Procyon lotor) population during a 3-yr ORV trial in developed areas of Burlington, Vermont, US. We used a modified N-mixture model to estimate raccoon abundance, RVNA seroprevalence, and capture rates jointly to examine factors that relate to ORV success to better inform management. We found that raccoon abundance was lower in less-developed areas compared to urban centers. Raccoon RVNA seroprevalence decreased as population abundance increased; it increased as the average age of the population increased. Nontarget opossum (Didelphis virginiana) captures correlated with a decrease in raccoon RVNA seroprevalence in low-development areas, suggesting that they may be competing for baits. The target bait density across the entire study area was 150 baits/km2, but a hand baiting strategy was heavily concentrated on roads, resulting in uneven bait densities within sampling sites (0–484 baits/km2 ). Uneven bait distribution across the study area may explain low RVNA seroprevalence in some locations. • Gardening has been shown to have a positive effect on the perceived quality of life of elderly persons. • Though modern Information Technology (IT) solutions can strengthen different care services, these are usually not combined with gardening trials. • The paper presents the summary results of a recent quantitative, pre- and post-test field trial on IT-supported leisure gardening in a European setting. • We found significant improvements in the perceived quality of life of the participants. The study presented in this paper aimed to assess the effect of an Information Technology enabled community gardening program for older adults, developed by an international consortium. We have executed a quantitative, pre- and post-test field trial with older adult volunteers to test the proposed programme in two European countries, Italy and Belgium (n =98). We used standardized and ad hoc questionnaires to measure changes in the volunteers' mental and psychological state during the trial. The statistical data analysis sought for differences in the pre- and post-test values of the key scores related to the perceived quality of life and benefits of gardening via paired-samples t-tests, and also tried to identify the important factors of significant changes via logistic regression. We found significant improvements in the perceived benefits of gardening and also in the scores computed from the WHO Quality of Life instruments, especially in the social sub-domains. The improvements were associated with the country, age, marital state and education of the volunteers. Higher age or being widow, divorced or single increased the odds of a significant improvement in the scores in more than one sub-domains. Though the two trial settings were different in some aspects, the observed significant improvements generally confirmed the positive effects of gardening concerning the perceived quality of life and benefits of gardening. Metabolic engineering for hyperaccumulation of lipids in vegetative tissues of high biomass crops promises a step change in oil yields for the production of advanced biofuels. Energycane is the ideal feedstock for this approach due to its exceptional biomass production and persistence under marginal conditions. Here, we evaluated metabolically engineered energycane with constitutive expression of the lipogenic factors WRINKLED1 (WRI1), DIACYLGLYCEROL ACYLTRANSFERASE1 (DGAT1), and OLEOSIN1 (OLE1) for the accumulation of triacylglycerol (TAG), total fatty acid (TFA), and biomass under field conditions at the University of Florida‐IFAS experiment station near Citra, Florida. TAG and TFA accumulation were highest in leaves (up to 9.9% and 12.9% of DW, respectively), followed by juice from crushed stems, stems, and roots. TAG and TFA accumulation increased up to harvest time and correlated highest with OLE1 and DGAT1 expression. Biomass dry weight, TAG, and TFA content differed greatly depending on DGAT1 and OLE1 expression in transgenic lines with similar WRI1 expression. Biomass did not significantly differ between WT and line L2 with DAGT1 and OLE1 expressed at low levels and TAG and TFA accumulating to 12‐ and 1.6‐fold that of WT leaves, respectively. In contrast, line L13, with intron‐mediated enhancement of DGAT1 expression, displayed a 245‐ to 330‐fold increase in TAG and a 4.75‐ to 6.45‐fold increase in TFA content compared with WT leaves and a biomass reduction of 52%. Countries in arid climates, such as Qatar, require efficient water-saving strategies and nitrogen treatment for vegetable production. Vegetable importation constituted approximately USD 352 million of Qatar's 2019 GDP; hence, enhancing local production is essential. This study investigated the effect of varying nitrogen and water levels on cucumber (Cucumis sativus L.) fruit yield. Various water management strategies were also evaluated. A split plot design was employed with two water levels (W1: 50% deficit irrigation, W2: 100% full irrigation) and three nitrogen levels (N1: 50 kg N ha−1, N2: 70 kg N ha−1, N3: 100 kg N ha−1) to examine cucumber yield and physiological response. Our findings revealed that using minimal drip irrigation and reducing nitrogen levels significantly enhanced the growth, SPAD index, fruit characteristics, and yield components of cucumber. Drip irrigation had a greater influence on cucumber production than nitrogen levels. Shoot height increased by 4% from W2N1 (T1) to W1N3 (T6) and 4.93% from W2N2 (T2) to W1N2 (T5). Fruit length and width increased by 10.63% and 13.41% from T2 and T1 to T5, respectively. The highest total yield occurred at T5, followed by T6, T2, W2N3 (T3), W1N1 (T4), and T1 at 34.5, 29.1, 27.6, 25.8, 25.2, and 20.4 t/ha, respectively. The optimal combination comprised 50% deficit irrigation (W1) and 70 kg N ha−1 (N2) nitrogen. IntroductionWe previously demonstrated efficacy of an 8-antigen recombinant subunit vaccine against a single species homologous Teladorsagia circumcincta challenge in lambs and in lambing ewes in pen trials. We subsequently demonstrated efficacy of a simplified, 2-antigen, version of this vaccine in lambs in pen trials. Here, we test both vaccines in lambing ewes in a field setting.MethodsIn the work presented here, 12 adjacent plots were seeded with a mixed infection of several common species of parasitic nematodes of sheep in temperate regions, including T. circumcincta. Ewes (n = 144), in groups of 12, grazed for 2 years on these plots and, in the first year, six of these groups of ewes were vaccinated with a 2-antigen prototype vaccine against T. circumcincta prior to mating and then again prior to lambing. In the following year these ewes were immunised again, this time with the 8-antigen prototype vaccine against T. circumcincta prior to mating and then prior to lambing. Throughout both seasons antigen-specific serum antibody levels in ewes and faecal worm egg counts (FEC) in ewes and their lambs were monitored, along with nematode species diversity at lambing.ResultsImmunised ewes produced elevated serum antibody levels to each of the vaccine antigens following immunisation but their FEC levels were not statistically significantly impacted by vaccination with either vaccine. FEC levels were also not impacted in lambs co-grazing the pastures with these immunised ewes. Nematode species diversity was not significantly impacted by vaccination in either year.DiscussionThe immunosuppressive effects of co-infecting gastrointestinal nematodes, the absence of vaccine cross-protection against co-infecting species and the influence of the periparturient relaxation in immunity probably all contributed to the inability of either vaccine to protect against T. circumcincta infection in field trials in the work presented here.

Faba bean (Vicia faba L.) is a legume crop grown in diverse climates worldwide. It has a high potential for increased cultivation to meet the need for more plant-based proteins in human diets, a prerequisite for a more sustainable food production system. Characterization of diversity panels of crops can identify variation in and genetic markers for target traits of interest for plant breeding. In this work, we collected a diversity panel of 220 accessions of faba bean from around the world consisting of gene bank material and commercially available cultivars. The aims of this study were to quantify the phenotypic diversity in target traits to analyze the impact of breeding on these traits, and to identify genetic markers associated with traits through a genome-wide association study (GWAS). Characterization under field conditions at Nordic latitude across two years revealed a large genotypic variation and high broad-sense heritability for eleven agronomic and seed quality traits. Pairwise correlations showed that seed yield was positively correlated to plant height, number of seeds per plant, and days to maturity. Further, susceptibility to bean weevil damage was significantly higher for early flowering accessions and accessions with larger seeds. In this study, no yield penalty was found for higher seed protein content, but protein content was negatively correlated to starch content. Our results showed that while breeding advances in faba bean germplasm have resulted in increased yields and number of seeds per plant, they have also led to a selection pressure towards delayed onset of flowering and maturity. DArTseq genotyping identified 6,606 single nucleotide polymorphisms (SNPs) by alignment to the faba bean reference genome. These SNPs were used in a GWAS, revealing 51 novel SNP markers significantly associated with ten of the assessed traits. Three markers for days to flowering were found in predicted genes encoding proteins for which homologs in other plant species regulate flowering. Altogether, this work enriches the growing pool of phenotypic and genotypic data on faba bean as a valuable resource for developing efficient breeding strategies to expand crop cultivation.

Abstract Background Probiotics are used in the broiler industry to increase production performance. Most often a probiotic is applied by mixing it in the feed, but studies have shown that earlier application may be advantageous. Therefore, in ovo application where the probiotic is administrated into the egg before hatch has been investigated as an alternative application method. However, in ovo application may impact hatchability negatively and may not be feasible at all hatcheries. The purpose of this study was to investigate the effect of a novel non-invasive method for mass application before hatch. The probiotic (E. faecium 669) was applied as a single dose by spray on the unhatched eggs and production performance and development of the cecal microbiota until slaughter was compared with a control flock. Through 16S rRNA sequencing of cecal samples from 25 broilers at day 7, 21 and 37 we compared the microbiota composition and richness for each group. The study was repeated for additional recording of production performance and re-isolation of the probiotic E. faecium from the intestine. Results In both trials the probiotic E. faecium could be re-isolated from the yolk sac and intestine at hatch and at day 7. Broilers in the probiotic treated groups had a higher performance in terms of bodyweight at day 34 and European production efficiency factor. Finally, a significant reduction of first-week and overall mortality was observed in the probiotic group in the first trial. Based on 16S rRNA profiling, significant differences in alpha diversity were found exclusively at day 37. Estimation of beta diversities, however, identified significant differences in microbiota composition between the control and probiotic group at day 7, 21 and 37. Conclusion The probiotic E. faecium strain successfully colonized broilers before/during hatch after a single spray application at day 18 of incubation. Positive effects of the probiotic were observed in multiple production parameters, including reduced mortality in trial 1, and microbiota analyses indicate significantly different microbiota compositions throughout the experimental phase. Taken together, the novel low-tech mass administration of E. faecium (669) may be considered a feasible strategy for improvements of production parameters in broiler production.

Roof rats and other rodents are a common pest in agriculture, causing extensive amounts of damage and losses. Poultry farms provide an abundance of resources that attract rodents but our ability to control them in these locations is limited. Bait stations can become sources of nesting or go unused if the target species only travels in aerial locations. Any uncontrolled rats will quickly overpopulate agricultural buildings due to their high reproductive rates. We tested alternative baiting devices at a large poultry farm to develop a station that was easily utilized in aerial locations and well accepted by roof rats. We deployed ContraPest®, a contraceptive liquid bait, in the devices and tracked consumption monthly. We monitored the rat populations for 16 months with remote cameras to measure changes in activity before and during ContraPest baiting using a general index approach. Linear regression showed a significant relationship between ContraPest consumption and the general index. As rats continuously consumed ContraPest from the new baiting devices, activity steadily declined. Within one year of using ContraPest, rat activity reduced by 94%, showing a significant difference from starting activity levels. These results demonstrate the impact an antifertility agent can have on rat populations when used successfully within an integrated pest management program.

Red seabream iridovirus (RSIV) is a major cause of marine fish mortality in Korea, with no effective vaccine available since its first occurrence in the 1990s. This study evaluated the efficacy of a formalin-killed vaccine against RSIV in rock bream under laboratory and field conditions. For the field trial, a total of 103,200 rock bream from two commercial marine cage-cultured farms in Southern Korea were vaccinated. Farm A vaccinated 31,100 fish in July 2020 and monitored them for 18 weeks, while farm B vaccinated 30,700 fish in August 2020 and monitored them for 12 weeks. At farm A, where there was no RSIV infection, the vaccine efficacy was assessed in the lab, showing a relative percentage of survival (RPS) ranging from 40% to 80%. At farm B, where natural RSIV infections occurred, cumulative mortality rates were 36.43% in the vaccinated group and 80.32% in the control group, resulting in an RPS of 54.67%. The RSIV-infectious status and neutralizing antibody titers in serum mirrored the cumulative mortality results. This study demonstrates that the formalin-killed vaccine effectively prevents RSIV in cage-cultured rock bream under both laboratory and field conditions.

An outbreak of camelpox occurred in the Mangistau region of Kazakhstan in 2019. To control the outbreak of camelpox and to prevent its further spread to other regions, camels were vaccinated using live and inactivated camelpox vaccines produced in Kazakhstan. To evaluate the efficacy of these camelpox vaccines in the field, vaccine trials used 172 camels on camel farms in the Beineu district. Of these, 132 camels were vaccinated using a live attenuated camelpox vaccine and 40 camels were vaccinated using an inactivated vaccine to observe immunogenicity and safety. The live vaccine was inoculated into camels by scarification at a dose of 5 × 104 EID50, and the inactivated vaccine was injected intramuscularly at 5 mL twice, with an interval of 35 days. During the safety evaluation, camels administered either vaccine displayed no clinical signs of illness or any adverse effects. Post-vaccination seroconversion demonstrated that the live attenuated vaccine started to elicit antibody responses in some animals as early as day seven, while, by day 28, 99% of vaccinated camels responded. For camels immunized with the inactivated vaccine, seroconversion began on day 21 at low titers ranging from 1:2 to 1:4. Ninety days post vaccination, 77% of the camels demonstrated an immune response that was up to a titer of 1:16. The antibody response waned six months post vaccination in camels vaccinated with two types of vaccine. Nonetheless, both vaccines were 100% effective at preventing clinical disease in vaccinated camels during the camelpox outbreak. All unvaccinated camels became ill, with manifestations of clinical signs characteristic of camelpox. Following these successful field trials in Kazakhstan, a vaccination program for camels, to control camelpox using the domestically produced live attenuated camelpox vaccine, has started.

The need for an agricultural system in the Mediterranean countries consuming fewer chemicals and respecting the environment becomes a pressing element. The use of natural beneficial microorganisms that enhance soil fertility could be a promising solution. Lebanese Beneficial Microorganisms (LBM) were extracted and evaluated for their capacity of promoting plant growth. Two combinations of LBM were compared to chemical fertilizers: alone (treatment B) or with an organic fertilizer (treatment A). Under field conditions, significant positive effects on height, leaf biomass, and fruit production were obtained starting from day 15 for both tested crops. Treatment A was the most efficient in enhancement of different growth parameters. Hence, Capsicum annuum shoots were 44.4% longer with 99.5% and 51.2% increase in the number of leaves and flowers per plant, respectively. Fruit yield increased over the control by 31.7% in C. annuum and 37.5% in Solanum lycopersicum . However, treatment B exhibited the highest significant values of root length and weight in S. lycopersicum . This study highlights the efficiency of both LBM combinations in the total absence of chemical fertilizers and the increase in their outcome by the addition of organic products. The use of native microbial consortia represents a novel strategy for the development of biofertilizers.

Abstract Background Gray blight is among the most destructive diseases that affect tea plants worldwide. In this study, microorganisms from deep-sea sediment samples for those with antagonistic effects were screened against gray blight caused by Pestalotiopsis theae. Results Thirty-two and twenty-eight morphologically different deep-sea bacteria and fungi were isolated, respectively. Isolates B5 and A65 clearly inhibited the pathogens in vitro and were prepared as wettable agent powders for evaluation in micro-plot field trials. Foliar application of the 48-h culture of B5 (1 × 108, 2 × 107, 1 × 107 colony-forming units (CFU)/ml) significantly reduced the incidence of gray blight disease. Compared to the untreated control, spraying with B5 inhibited gray blight disease by 78.57%. Isolate B5 was identified as Bacillus subtilis B5 in morphological and 16S rDNA sequence analyses. The foliar application of 7-day cultures of A65 (1 × 108, 2 × 107, 1 × 107 CFU/ml) significantly reduced the incidence of gray blight disease. A65 (108 CFU/ml) inhibited gray blight disease by 75.46% and was identified as Paecilomyces lilacinus A65 in morphologically and internally transcribed spacer sequence analyses. Conclusions These candidate microbial pesticides may inhibit gray wilt in tea, replace chemical pesticides’ use without causing environmental pollution, and promote the development of green agriculture.

Salmonella enterica can be carried by pigs and can reach the final product and the consumer. Thus, Salmonella reduction strategies along the swine production chain should be studied. In this sense, the objective of the study was to evaluate a subunit vaccine based on secondary antigens administered orally against natural infection in swine farms in Brazil. A field trial study was conducted to estimate the effect of the vaccination on the Salmonella spp. seroprevalence, presence in lymph nodes, and fecal content in commercial pig herds in a vertical integration system belonging to an agroindustry. In this work, we present the results of the inoculation of canola seeds (Brassica napus L.) with Trichoderma viride strains that promote the growth of plants. Seven morphologically different strains of T. viride (TvI-VII) were shown to be capable of synthesizing auxins and exhibited cellulolytic and pectinolytic activities. To gain a deeper insight into the molecular mechanisms underlying canola–T. viride interactions, we analyzed the canola stress genes metallothioneins (BnMT1-3) and stringent response genes (BnRSH1-3 and BnCRSH). We demonstrated the presence of cis-regulatory elements responsive to fungal elicitors in the promoter regions of B. napus MT and RSH genes and observed changes in the levels of the transcripts of the above-mentioned genes in response to root colonization by the tested fungal strains. Of the seven tested strains, under laboratory conditions, T. viride VII stimulated the formation of roots and the growth of canola seedlings to the greatest extent. An experiment conducted under field conditions during drought showed that the inoculation of canola seeds with a suspension of T. viride VII spores increased yield by 16.7%. There was also a positive effect of the fungus on the height and branching of the plants, the number of siliques, and the mass of a thousand seeds. Flavonoids are antioxidant phytochemicals that confer a beneficial effect on human health. We have previously developed and characterized eight lettuce (Latuca sativa L.) lines that accumulated high levels of diverse flavonoids and their precursors in controlled environment conditions. Three Rutgers Scarlet lettuce (RSL) lines selected in tissue culture for deep-red color (RSL-NAR, RSL-NBR, RSL-NFR) accumulate anthocyanins and quercetin, three lines identified in a chemically mutagenized red lettuce population accumulate kaempferol (KfoA and KfoB) or naringenin chalcone (Nco), and two lines that were spontaneous green mutants derived from the red line RSL-NAR (GSL, GSL-DG) accumulate quercetin. These eight lines were field-grown in the Salinas Valley of California for four years together with seven control accessions of varying colors (light green, dark green, red, and dark red). At market maturity, a substantial variation in plant composition was observed, but the three RSL lines consistently accumulated high levels of cyanidin, GSL and GSL-DG accumulated the highest levels of quercetin, KfoA and KfoB accumulated kaempferol, and Nco amassed naringenin chalcone, confirming that these mutant lines produce high levels of beneficial phytochemicals under field conditions. Mutant lines and control accessions were also assessed for their biomass production (plant weight, height, and width), overall content of pigments (leaf chlorophyll and anthocyanins), resistance to diseases (downy mildew, lettuce drop, and Impatiens necrotic spot virus), postharvest quality of processed tissue (deterioration and enzymatic discoloration), and composition of 23 mineral elements. All but one mutant line had a fresh plant weight at harvest comparable to commercial leaf cultivars; only Nco plants were significantly (p < 0.05) smaller. Rhizosphere bacteria can provide multiple benefits to plants, including increased nutrient supply, pathogen/disease control, and abiotic stress tolerance, but results from pot trials do not always translate to field conditions. This study tested whether rhizosphere biocontrol bacteria can also provide plant growth promotion and how benefits can be provided at a commercial farm. Commercial lettuce seeds and plants were treated with rhizosphere biocontrol bacteria Bacillus velezensis UQ9000N, B. amyloliquefaciens 33YE, Brevibacillus laterosporus 4YE, and Pseudomonas azotoformans UQ4510An. 33YE increased the head diameter, plant height, and fresh weight of the Green Moon cultivar, while 33YE, UQ4510An, and UQ9000N increased the fresh and dry weight of Liston, a more heat-tolerant cultivar, via a single seed treatment or repeat root treatments under nursery and field conditions across different inoculation schedules and growth stages. Significant growth promotion was also demonstrated when inoculating field plants after transplanting (in particular for 33YE). Applications of these microbial biostimulants to lettuce seeds or plantlets potentially enable earlier transplanting and earlier harvests. Repeat inoculations using irrigation water and long-lasting formulations may further advance the benefits of these biostimulants as microbial biofertilisers for plant growth promotions in the field. The use of plant biostimulants, also known as bioeffectors (BEs), has attracted increasing attention as an environmentally friendly strategy for more sustainable crop production. BEs are substances or microorganisms that are applied to plants or the surrounding soil to stimulate natural processes to enhance nutrient uptake, stress tolerance, and plant growth. Here, we tested the effectiveness of five BEs to enhance maize growth and phosphorus (P) uptake from various recycled P fertilizers in a series of pot and field experiments. First, the impact of two bacterial BEs and one soil-specific plant-based BE on crop performance was assessed in a 4-week screening experiment conducted in two arable, P-deficient soils of differing soil pH (a silty clay loam of pH 7.1 and a silty loam of pH 7.8) amended with recycled P-fertilizers (rock phosphate, biogas digestate, green waste compost, composted dairy manure, and chicken manure pellets). Then, for each soil type, the plant growth-promoting effect of the most promising BE-fertilizer combinations was re-assessed in an 8-week experiment. In addition, over a period of up to 3 years, three field experiments were conducted with maize in which up to two bacterial BEs were used either alone or in combination with a plant-based BE. Our experiments show that while BEs in combination with specific P-fertilizers can promote maize growth within the first weeks of growth under controlled conditions, the observed effects vanished in the long term, both in pots and under field conditions. In a tracing experiment, in which we tested the persistence of one bacterial BE over a period of 5 weeks, we observed a drastic decrease in colony-forming units already 2 weeks after inoculation. As previously shown in other studies, our data indicate that the plant growth-promoting effects of BEs found under controlled conditions are not directly transferable to field conditions. We developed an admonishing service for a shopworker robot and conducted a field trial to investigate the impressions of a shop's staff and customers. Applying the admonishing service in a real-world robot is difficult due to the high risk of rejection by society. We wanted to achieve an acceptable admonishing service while simultaneously avoiding the impression of a forceful request from a machine. We proposed a harmonized design that provided friendly and admonishing services. First, we interviewed a shop's staff to learn their strategies for both friendly and admonishing services. From our evaluation of the interview results, we derived three design principles: friendly impressions, zero erroneous admonishments, and polite requests. Based on the design principles, we implemented our harmonized design on a social robot that guides customers to product locations and admonishes those who are not wearing face masks. We conducted a 13-day field trial in a retail shop and interviewed the customers and shopworkers to learn their impressions of our robot. The results of the field trial imply that our harmonized design approach is successful. Both the customers and the shop staff had overall positive impressions of the robot, its admonishing and friendly services, and expressed an intention to use it in the future. This study aimed to evaluate the efficacy of a virus-like particle (VLP) vaccine containing the open reading frame 2 of porcine circovirus type 2d (PCV2d) in a farm environment where natural infections associated with porcine circovirus-associated disease are endemic. The vaccine trial was conducted on three farms (H, M, and Y) with a history of infections including porcine reproductive and respiratory syndrome virus (PRRSV), PCV, Mycoplasma, and E. coli. Farm H, as well as farms M and Y, experienced natural PCV2 infection between 4 and 8 weeks post-vaccination (wpv), and 8 and 12 wpv, respectively. Viremia levels of all farms were significantly (p < 0.05) lower in vaccinated piglets than the control group after natural infection. In all farms, serum immunoglobulin G levels peaked at 8 wpv in the vaccinated groups, surpassing those in the control groups. Furthermore, neutralizing antibody titers were significantly (p < 0.05) higher in the vaccinated groups than the control groups in farms H and Y (0–8 wpv). However, there were no significant differences between the vaccinated and control group in neutralizing antibody titers of farm M (0–20 wpv). In terms of body weight, vaccinated piglets from all three farms showed significantly increased average weights at 12 wpv compared to the control groups. In conclusion, our study revealed noteworthy differences in viremia and body weight gain between vaccinated and control animals on three farms. BACKGROUND: Although co‐formulants constitute a substantial portion of the total plant protection product (PPP) mass applied to crops, data on residue formation and the behaviour of these substances on plants are scarce. In an earlier study we demonstrated that co‐formulants commonly used in PPPs can form considerable residues, i.e., in the low to medium mg/kg range, but normally decline rapidly within few days. In the field trial reported here, we aimed to identify the major decline processes of co‐formulants. Residues of co‐formulants were therefore monitored in parsley and lettuce grown in an open field as well as under foil tunnels equipped with either an overhead or a drip irrigation system. RESULTS: Dissipation of three anionic surfactants was markedly faster when crops (parsley and lettuce) were exposed to natural rainfall or irrigation from above compared to drip irrigation. In contrast, the decline of three volatile organic solvents was not affected by rain or irrigation, but was dependent on the crop, with much shorter half‐lives in lettuce than in parsley. Furthermore, dilution through plant growth contributed significantly to the reduction of residues over time. CONCLUSION: In this work we substantiate earlier findings on the magnitude and dissipation of residues of anionic surfactants and solvents representing the most important co‐formulant classes. The chosen experimental setup allowed differentiation between decline processes and we confirm that foliar wash‐off is a major dissipation process for anionic surfactants. Methane leakage from point sources in the oil and gas industry is a major contributor to global greenhouse gas emissions. The majority of such emissions come from a small fraction of “super-emitting” sources. We evaluate the emission detection and quantification capabilities of Kairos Aerospace’s airplane-based hyperspectral imaging methane emission detection system for methane fluxes of 18–1,025 kg per hour of methane (kgh(CH4 )). In blinded controlled releases of methane conducted over 4 days in San Joaquin County, CA, Kairos detected 182 of 200 valid nonzero releases, including all 173 over 15 kgh(CH4 ) per meter per second (mps) of wind and none of the 12 nonzero releases below 8.3 kgh(CH4 )/mps. Nine of the 26 releases in the partial detection range of 5–15 kgh(CH4 )/mps were detected. There were no false positives: Kairos did not detect methane during any of the 21 negative controls. Plume quantification accuracy depends on the wind measurement technique, with a parity slope of 1.15 (s ¼ 0.037, R2 ¼ 0.84, N ¼ 185) using a cup-based wind meter and 1.45 (s ¼ 0.059, R2 ¼ 0.80, N ¼ 157) using an ultrasonic anemometer. Performance is comparable even with only modeled wind data. For emissions above 15 kgh/mps, quantification error scales as roughly 30%–40% of emission size, even when using wind reanalysis data instead of ground-based measurements. This reflects both uncertainty in wind measurements and in Kairos’ estimates. These findings suggest that at 2 mps winds under favorable environmental conditions in the United States, Kairos could detect and quantify over 50% of total emissions by identifying super-emitting sources.

This study presents the possible consequences of maintaining the current regulatory regime of the experimental release of genetically modified higher plants in the EU for the products of new genomic techniques (NGTs). Currently, the experimental release is a crucial stage before the authorization of a product for the market. By analyzing the data on the performance of field trials in the EU (numbers, sizes, dominating countries) and comparing the present regulatory provisions with those of selected third countries (including new provisions adopted in the UK), this study shows that the current framework of GMO (genetically modified organisms) field trials is ill-fitted for breeding activities. Due to strict limitations placed on the operator of a field trial in the EU, easing the regulatory burdens on the authorization of certain NGT products for the market may not provide researchers (especially, plant breeders) the competitive position they need if the present legal conditions for carrying out GMO field trials with certain NGT products (especially, those that are considered GMOs covered by the EU GMO legislation) are not going to change as well.

The use of plant biostimulants, also known as bioeffectors (BEs), has attracted increasing attention as an environmentally friendly strategy for more sustainable crop production. BEs are substances or microorganisms that are applied to plants or the surrounding soil to stimulate natural processes to enhance nutrient uptake, stress tolerance, and plant growth. Here, we tested the effectiveness of five BEs to enhance maize growth and phosphorus (P) uptake from various recycled P fertilizers in a series of pot and field experiments. First, the impact of two bacterial BEs and one soil-specific plant-based BE on crop performance was assessed in a 4-week screening experiment conducted in two arable, P-deficient soils of differing soil pH (a silty clay loam of pH 7.1 and a silty loam of pH 7.8) amended with recycled P-fertilizers (rock phosphate, biogas digestate, green waste compost, composted dairy manure, and chicken manure pellets). Then, for each soil type, the plant growth-promoting effect of the most promising BE–fertilizer combinations was re-assessed in an 8-week experiment. In addition, over a period of up to 3 years, three field experiments were conducted with maize in which up to two bacterial BEs were used either alone or in combination with a plant-based BE. Our experiments show that while BEs in combination with specific P-fertilizers can promote maize growth within the first weeks of growth under controlled conditions, the observed effects vanished in the long term, both in pots and under field conditions. In a tracing experiment, in which we tested the persistence of one bacterial BE over a period of 5 weeks, we observed a drastic decrease in colony-forming units already 2 weeks after inoculation. As previously shown in other studies, our data indicate that the plant growth-promoting effects of BEs found under controlled conditions are not directly transferable to field conditions. It is suggested that the drastic decline in inoculated bacterial strains in the tracing experiment is the reason for the decline in plant growth effect.

Xylan that comprises roughly 25% of hardwood biomass is undesirable in biorefinery applications involving saccharification and fermentation. Efforts to reduce xylan levels have therefore been made in many species, usually resulting in improved saccharification. However, such modified plants have not yet been tested under field conditions. Here we evaluate the field performance of transgenic hybrid aspen lines with reduced xylan levels and assess their usefulness as short-rotation feedstocks for biorefineries. Three types of transgenic lines were tested in four-year field tests with RNAi constructs targeting either Populus GT43 clades B and C (GT43BC) corresponding to Arabidopsis clades IRX9 and IRX14, respectively, involved in xylan backbone biosynthesis, GATL1.1 corresponding to AtGALT1 involved in xylan reducing end sequence biosynthesis, or ASPR1 encoding an atypical aspartate protease. Their productivity, wood quality traits, and saccharification efficiency were analyzed. The only lines differing significantly from the wild type with respect to growth and biotic stress resistance were the ASPR1 lines, whose stems were roughly 10% shorter and narrower and leaves showed increased arthropod damage. GT43BC lines exhibited no growth advantage in the field despite their superior growth in greenhouse experiments. Wood from the ASPR1 and GT43BC lines had slightly reduced density due to thinner cell walls and, in the case of ASPR1, larger cell diameters. The xylan was less extractable by alkali but more hydrolysable by acid, had increased glucuronosylation, and its content was reduced in all three types of transgenic lines. The hemicellulose size distribution in the GALT1.1 and ASPR1 lines was skewed towards higher molecular mass compared to the wild type. These results provide experimental evidence that GATL1.1 functions in xylan biosynthesis and suggest that ASPR1 may regulate this process. In saccharification without pretreatment, lines of all three constructs provided 8-11% higher average glucose yields than wild-type plants. In saccharification with acid pretreatment, the GT43BC construct provided a 10% yield increase on average. The best transgenic lines of each construct are thus predicted to modestly outperform the wild type in terms of glucose yields per hectare. A field trial examined the impact of nitrogen sources and application stages on yield components. The research was conducted at the Agronomy Research Farm, The University of Agriculture Peshawar, during the winter of 2022. The experiment was arranged in a Randomized Complete Block Design (RCBD) with three replications. The experiment consisted of three factors: the first factor involved Nitrogen Levels (NL) (100, 120, and 140 kg. ha-1), the second factor pertained to application sources (S), urea and Farm Yard Manure (FYM) (urea + FYM), and the third factor addressed application stages (A) (sowing, tillering, and stem elongation). Nitrogen levels significantly affected spikes m-2 (255.0), net grains weight spike-1 (2.6 g), grains spike-1 (52.2) and thousand grains weight (48.1 g), respectively. Application stages significantly affected net gain weight (2.5 g) and thousand grains weight (46.9 g). Applications sources also significantly affected grains spike-1 (52.7). The interaction NL × S significantly affected grains spike-1. Similarly, the interaction S × A also suggestively affected grains spike-1. The interaction NL × A affected the thousand-grain weight significantly. The research concluded that applying integrated nitrogen sources (Urea + FYM) at a rate of 120 kg. ha-1 at an application stage of stem elongation gives a higher yield and is recommended to the end users.
