Sepsis is an inevitable stage of bacterial invasion characterized by an uncontrolled inflammatory response resulting in a syndrome of multiorgan dysfunction. Most conventional antibiotics used to treat sepsis are efficacious, but they have undesirable side effects. The green synthesised Ag NPs were synthesized by 5 g of the earthworm extract dissolved in a volume of 500mL of distilled water and then added to 2,500 mL aqueous solution of 1mM silver nitrate at 40 °C. After 4 h, the mixture was then allowed to dry overnight at 60 °C. Later, Ag NPs were washed and collected. They were characterized by X-ray diffraction, ultraviolet-visible spectroscopy, and transmission electron microscopy. Sepsis model as induced by feces-intraperitoneal injection method. Eighteen male mice were assigned into three main groups: the control group, the sepsis-model group, and the Ag NPs-treated group. The control group received a single oral dose of distilled water and, after two days, intraperitoneally injected with 30% glycerol in phosphate buffer saline. The Sepsis-model group received a single oral dose of distilled water. Ag NPs - The treated group received a single oral dose of 5.5 mg/kg of Ag NPs. After two days, the sepsis-model group and Ag NPs-treated group were intraperitoneally injected with 200 µL of faecal slurry. Ag NPs treatment in septic mice significantly decreased liver enzyme activities, total protein, and serum albumin. Moreover, Ag NPs significantly enhanced kidney function, as indicated by a significant decrease in the levels of creatinine, urea, and uric acid. In addition, Ag NPs showed a powerful antioxidant effect via the considerable reduction of malondialdehyde and nitric oxide levels and the increase in antioxidant content. The histopathological investigation showed clear improvement in hepatic and kidney architecture. The gut microbiota is a key and primary stimulus for the development of a host's immune system. The early establishment of the gut microbiota is affected by several perinatal factors but little is known about their influence on shaping normal immune development and, consequently, on the programming of future health. The analysis of different immune compounds is well-documented in serum samples; however, their presence in faecal samples has not been studied, and this information could be valuable in early life. In this context, the authors of this study aimed to both describe the immunological faecal profile of a cohort of one-month-old infants and describe the impact of different perinatal factors, exploring possible associations between immune compounds and gut microbiota in faecal samples. Clear differences in immune profile were observed between full-term and premature infants. Breastfeeding increases IgG2, IgG4, and IgA; in addition, male babies showed some increased Igs, among other observations. Overall, the findings of this study reinforce the hypothesis that microorganisms and immune compounds interact with each other in the early neonatal gut and that understanding these interactions in depth will help us comprehend the influence of the gut microbiota on short- and long-term infant health outcomes. Simple Summary: Transportation is one of the most stressful situations in pig production. The maximum duration of a journey should be determined considering the pigs' coping capacity. The effect of a typical journey (200 km in Argentina) on pig physiological stress response was assessed using animal-friendly techniques. Fattening pigs (n = 18) were transported for three hours; 10 received a dexamethasone injection 7 h after the end of the journey. A control group (n = 18) was not transported, and 10 individuals received dexamethasone. The levels of faecal glucocorticoid metabolites and hair cortisol and cortisone were measured. The transported pigs had higher levels of faecal glucocorticoid metabolites after transportation than the control group. The level of faecal glucocorticoid metabolites returned to normal values faster in transported pigs treated with dexamethasone than in the control group. The amount of glucocorticoids in their hair was not related to the stressful stimuli; in contrast, the cortisone/cortisol ratio yielded better results. Since glucocorticoid measurements in hair are still a questionable stress biomarker in pigs, further studies are needed to develop animal-based transport protocols. The results showed that pigs were able to cope with a 3 h road trip. In the near future, a non-invasive glucocorticoid analysis could be used to characterize pigs' coping capacity on a farm. Pig homeostasis is challenged by stressful production practices, like road transportation. Glucocorticoids (GCs) are mediators of reactive homeostasis, and their concentrations are frequently used as a stress indicator. The adrenocortical activity of fattening female and castrated male pigs was monitored over a 5-day longitudinal study. A bi-factorial experimental design was applied on day 2; 18 pigs in pen 1 were transported for 3 h (T; 1.2 m2/pig), and 18 pigs were kept in pen 2 (NT). Ten pigs from each pen were treated with dexamethasone (T-D or NT-D), and eight with saline solution (T-SS or NT-SS). Adrenocortical activity was assessed by measuring the levels of faecal glucocorticoid metabolites (FGMs) and hair cortisol and cortisone. In T-SS pigs, the level of FGMs was higher after transportation than in NT-SS pigs. The level of FGMs of T-D pigs initially increased but then reached similar levels to those of NT-SS sooner than T-SS. In contrast, hair cortisol and cortisone did not respond to the treatments. Nevertheless, the hair cortisone/cortisol ratio increased due to transport and decreased after dexamethasone administration. Daily faecal sampling proved still more reliable than 60-day hair sampling for assessing adrenocortical activity. Transported pigs recovered their adrenocortical baseline levels within 24 h. Geohelminths (soil-transmitted helminths) tend to thrive in moist soil conditions, and the poor hygiene systems of underdeveloped countries have enhanced their transmission at an alarming rate. A cross-sectional study was conducted to determine the effects of soil type, pH, and temperature on geohelminths found in soil samples from Mkpat Enin LGA, Akwa Ibom State. The soil samples were examined using the centrifugal floatation method for geohelminth eggs and larvae. Among the 200 soil samples examined, an overall prevalence of 67.0% was recorded. Geohelminths recovered were Ascaris lumbricoides, which contaminated 38 (28.36%), Trichuris trichiura 27(20.15 %), hookworms 26(19.40 %), Strongyloides stercoralis 24(17.91 %), and Toxocara canis 19(14.18 %) soil samples. Soil samples obtained from farmlands and toilets had a higher contamination rate (82 %) than those from schools and roadsides (68.0% and 36.0%, respectively). Loamy soil recorded 77(66.38%) contaminated soil samples at a mean temperature of 28.7oC ± 0.1, and pH of 6.9 ± 0.03, compared to sandy soil with 32(69.57 %) at a mean temperature of (27.5oC ± 0.3) and pH of 5.7 ± 0.04, then clayey soil with 25(65.79 %) at a mean temperature of 27.0oC ± 0.1 and pH of 5.3 ± 0.04. There was no significant correlation between the prevalence of each geohelminth and soil type, temperature, or pH. The type of soil, soil temperature within the measured range, and soil alkalinity as measured do not appear to be major factors influencing the overall prevalence of geohelminths. The analysis also showed that these factors do not have a strong influence on the prevalence of individual geohelminth species. The eggs of these geohelminths were more abundant than their larvae. Little is known regarding how the microbiota reacts to repeated acute stress events in dogs, and whether it is a predictor of stress response. In this study, we explored the impact on the gut microbiota and digestive health with two common events many pet dogs find stressful. Twenty healthy adult dogs, living within a colony, were exposed to either car travel or separation three times across eight-week intervals. Faecal samples were collected 24 h before, within 24 h, and 24–48 h after. Faecal quality and pH, and microbiota diversity and composition were analysed in context with wider published work on physiological stress measures. No significant changes were observed in faecal quality or pH with either stress event at any timepoint, indicating all pets remained in good digestive health. Microbiota analysis demonstrated no significant impact on alpha or beta diversity with either stressor. Microbial signatures previously linked to stress were not identified in these dogs and no changes were observed in the functional gut composition. Irrespective of whether the pet was considered "stressed" (i.e., exhibited an increase in serum cortisol), there was no effect on the microbiota and no taxa were predictive of stress response. Collectively, this work demonstrates, for this population, certain acute stress events have no meaningful impact on the canine gut microbiota, and it has no impact on the associated stress response. The study aimed to explore the connection between horses’ total tract apparent dry matter digestibility (ttaDMD) of various roughages and faecal particle size (FPS), to develop a tool to rank roughage digestibility. Six horses, varying in size and breed, were fed six distinct roughages (four cuts of fescue, perennial ryegrass/clover mix, and straw). Each feeding period lasted 16 days, with a 14-day adaptation phase. Faecal samples underwent wet sieving analysis. Roughage chemical composition was evaluated using NIRs. Faecal and roughage acid insoluble ash content were used to estimate ttaDMD and FPS was determined by wet sieving using a set of 6 sieves of different mesh sizes (S1 to S6; 6.50, 4.50, 2.36, 1.18, 0.6 and 0.15 mm respectively). The effects of roughage ttaDMD on FPS across sieves were analysed using GLM. Linear regressions and correlations were performed between faecal residues on each sieve and mesh sizes of the sieves to create a mathematical tool to rank ttaDMD based on FPS distribution. Increased ttaDMD was associated with larger FPS, with perennial ryegrass/clover mix (ttaDMD 74.44%) and straw (ttaDMD 44.57%) resulting in smaller and larger faecal particles respectively. Correlations were observed between ttaDMD, S1 and S2 (r = 0.639 and 0.668; p ≤ 0.001). The angular coefficient of the linear function between faecal residues on each sieve support the idea to estimate roughages digestibility from FPS (R2 = 0.4153). Consequently, determining faecal residues across sieves of varying mesh sizes could serve as an indirect method for ranking roughages digestibility.

The Shiga toxin producing Escherichia coli (STEC) are considered to be one of the most important groups of emerging public health pathogens with cattle being the main reservoir. The objective of this study was to isolate and characterise Escherichia coli Shiga toxins in dairy cattle farms. A total of 252 faeces samples were collected from healthy cows belonging to 37 farms. PCR screening of samples for the common sequences of stx1/stx2 genes and stx1 and stx2 genes resulted in a STEC faecal excretion prevalence of 59.5% at the farm level and 26.6% at the individual level. Among positive animals, 85.1% carried STEC with a single stx1 gene and 14.9% with the stx1 and stx2 genes. Immunomagnetic separation was performed on 40 PCR-positive samples (10/10 positive for the stx1 and stx2 genes and 30/57 positive only for stx1). Biochemical identification revealed the presence of 66 E. coli strains (27.5%). The search for virulence genes on these strains by PCR showed that only twenty-two (33.33%) were STEC. The presence of the stx1, stx2, ehx and eae genes was characterised in 30.3%, 4.54%, 13.63% and 1.51% of the strains, respectively, indicating that the virulotype with stx alone was dominant. Serological identification showed the absence of O157 sero-groups and the presence of O1(2), O2, O18(2), O128 sero-groups. The susceptibility testing of STEC showed 68.18% resistance to chloramphenicol, 63.64% to neomycin, 59.1% to ampicillin, 22.73% to trimethoprim + sulfamethoxazole and 9.1% to amoxicillin + clavulanic acid and nalidixic acid. Four strains showed multi-resistance. Bovine carriage of STEC constitutes a public health risk by contamination of milk and meat. To protect human health, it is necessary to limit the bovine STEC shedding.

The role of infectious agents in gastrointestinal diseases is becoming greater, because normally the human intestines have many bacteria with an increased concentration in the lower parts of the gastrointestinal tract. The most common among the colonizing microorganisms are the so-called problematic gram-negative bacteria. In recent years, the role of one microbe in the progression of gastrointestinal diseases - Klebsiella pneumoniae - has been increasing. This bacterium is often isolated from patients with autoimmune and oncological problems such as Crohn's disease, ulcerative colitis and colorectal carcinoma. Context: DNA recovered from predator faeces (scats) can be used to determine the presence of fauna and shed light on their life histories and inter-species interactions. DNA metabarcoding, which involves concurrent amplification and sequencing of DNA from multiple taxa, represents an important advance by enabling the simultaneous detection of multiple species from such samples. Although an attractive proposition, metabarcoding requires 'universally' applicable genetic markers that can discriminate among a broad range of taxa, while also targeting sequences that are sufficiently short to be amplified from degraded DNA. Aims: To identify, evaluate, and test metabarcoding DNA markers suitable for the detection of marsupials and other Australian fauna from terrestrial predator scats found in nature. Methods: We apply a bioinformatic approach using publicly available DNA databases and a locally derived and marker-specific reference-DNA database to evaluate the diagnostic ability and likelihood of amplification of candidate metabarcoding markers for marsupials and other taxa that may be consumed by predators. Key results: We identify two markers (12SV5 and 16SMam) that are suitable for use and successfully identify marsupial sequences at a high level of resolution. These markers work best in combination because they bring complementary levels of primer specificity and diagnostic ability in detecting multiple prey species as well as the predator. We also show that these samples work well in predator scats sampled from the wild in Tasmania. Conclusions: These markers provide a useful tool for surveying mammalian predators and their prey and could also be applied to eDNA analyses from other sample types. Improvements to the reference database and further development of markers targeting different taxonomic groups will improve the resolution and usefulness of this approach. Implications: Metabarcoding of predator scats provides a potent approach to non-invasive wildlife survey that offers the opportunity for the detection of multiple species across all vertebrates. Introduction: To consider the growing health issues caused by antibiotic resistance from a "one health" perspective, the contribution of meat production needs to be addressed. While antibiotic resistance is naturally present in microbial communities, the treatment of farm animals with antibiotics causes an increase in antibiotic resistance genes (ARG) in the gut microbiome. Pigs are among the most prevalent animals in agriculture; therefore, reducing the prevalence of antibiotic-resistant bacteria in the pig gut microbiome could reduce the spread of antibiotic resistance. Probiotics are often studied as a way to modulate the microbiome and are, therefore, an interesting way to potentially decrease antibiotic resistance. Methods: To assess the efficacy of a probiotic to reduce the prevalence of ARGs in the pig microbiome, six pigs received either treatment with antibiotics (tylvalosin), probiotics (Pediococcus acidilactici MA18/5M; Biopower® PA), or a combination of both. Their faeces and ileal digesta were collected and DNA was extracted for whole genome shotgun sequencing. The reads were compared with taxonomy and ARG databases to identify the taxa and resistance genes in the samples. Results: The results showed that the ARG profiles in the faeces of the antibiotic and combination treatments were similar, and both were different from the profiles of the probiotic treatment (p < 0.05). The effects of the treatments were different in the digesta and faeces. Many macrolide resistance genes were detected in a higher proportion in the microbiome of the pigs treated with antibiotics or the combination of probiotics and antibiotics. Resistance-carrying conjugative plasmids and horizontal transfer genes were also amplified in faeces samples for the antibiotic and combined treatments. There was no effect of treatment on the short chain fatty acid content in the digesta or the faeces. Conclusion: There is no positive effect of adding probiotics to an antibiotic treatment when these treatments are administered simultaneously. [ABSTRACT FROM AUTHOR]

The transmission of pathogens from reservoir to recipient host species, termed pathogen spillover, can profoundly impact plant, animal, and public health. However, why some pathogens lead to disease emergence in a novel species while others fail to establish or do not elicit disease is often poorly understood. There is strong evidence that deformed wing virus (DWV), an (+)ssRNA virus, spills over from its reservoir host, the honeybee Apis mellifera, into the bumblebee Bombus terrestris. However, the low impact of DWV on B. terrestris in laboratory experiments suggests host barriers to virus spread in this recipient host. To investigate potential host barriers, we followed the spread of DWV genotype B (DWV-B) through a host's body using RT-PCR after experimental transmission to bumblebees in comparison to honeybees. Inoculation was per os, mimicking food-borne transmission, or by injection into the bee's haemocoel, mimicking vector-based transmission. In honeybees, DWV-B was present in both honeybee faeces and haemolymph within 3 days of inoculation per os or by injection. In contrast, DWV-B was not detected in B. terrestris haemolymph after inoculation per os, suggesting a gut barrier that hinders DWV-B's spread through the body of a B. terrestris. DWV-B was, however, detected in B. terrestris faeces after injection and feeding, albeit at a lower abundance than that observed for A. mellifera, suggesting that B. terrestris sheds less DWV-B than A. mellifera in faeces when infected. Barriers to viral spread in B. terrestris following oral infection may limit DWV's impact on this spillover host and reduce its contribution to the community epidemiology of DWV. AbstractRepeated use of rodenticides for rodent pest management leads to the development of bait shyness and resistance in rodents. Therefore, using pheromones for rodent management could be a significant step forward. The present study was conducted to determine the response of house rats towards odour of faecal extracts of conspecifics of different age groups and sexes. Rats of different maturity groups and sexes were exposed to 20% faecal extract of mature and immature male and female rats. Results indicated that faecal extract of estrous stage mature females has a significant attractant/repellent effect on mature rats of opposite/same sex, respectively. Gas chromatography-linked mass spectrometry of the faecal extract of estrous female rats identified 62 compounds, of which 16 were already reported in other animals as pheromonal compounds and might be used in communication by house rats. In the future, these identified compounds can be used in a fixed proportion after testing their efficacy to develop a stable lureto increase the trappability and acceptability of antifertility agents and rodenticide-based baits for the management of house rats. Cryptosporidium, an enteric protozoan parasite, poses economic losses in animal production and has zoonotic potential. This study aimed to determine the prevalence of cryptosporidium and associated risk factors in local pig breeds within Donga Local Government Area of Taraba State, Nigeria. A cross-sectional study was conducted, and a total of 384 faecal samples were collected from pigs in eight sampling sites. The samples were examined using the modified Ziehl-Neelsen staining technique. The results showed an overall prevalence of 27.9% (107/384) for cryptosporidium oocysts. The prevalence was significantly higher in pre-weaned piglets (64%) compared to post-weaned (26.9%) and adult pigs (24.14%). Female pigs had a significantly higher prevalence (49.21%) than males (17.4%), and diarrhoeic pigs had a significantly higher prevalence (55.2%) than non-diarrhoeic (22.9%). The prevalence was higher in the rainy season (42.4%) than in the dry season (10.3%). The detection of Cryptosporidium spp. oocysts in the faeces of pigs suggests that these animals may serve as potential reservoirs and shedders of this protozoan parasite, contaminating the environment and serving as a source of infection to other animals and humans. Abstract Background Sarcocystis species are obligatorily heteroxenous protozoan parasites with predator–prey life cycles. Global Knowledge about the epidemiology and the distribution pattern of different Sarcocystis species in dog feces are very scarce. Therefore, the current investigation was conducted to declare the occurrence of Sarcocystis in the fecal specimens of the most common canids in Egypt, the domestic dogs, and to identify the species present using various parasitological and molecular approaches. Methods A total of 100 dog fecal samples were collected and screened using fecal sugar flotation test for the presence of Sarcocystis oocysts/sporocysts. Additionally, thirty samples were used for genomic DNA extraction. The 18S rRNA gene fragment was the target of primers for a PCR, followed by purification and sequencing of the amplicons. Results Currently, the results obtained reviewed that 4% of fecal samples were positive for Sarcocystis spp. using LM. Additionally, Sarcocystis spp. were verified in sixteen dogs (53.3%, 16/30) using PCR and subsequent sequencing protocols. Statistically, insignificant difference in prevalence of sarcocystosis relative to age and gender was noticed. Morphologically, the detected sporocysts measured 13.2–16.0 × 9.4–11 μm. Based on the 18S rRNA gene, sequencing analysis of amplicons from sporocysts DNA revealed 99.82% nucleotide homology with published S. tenella partial nucleotide sequences from sheep in Iraq and Iran. Conclusions This is the first molecular evidence in support of the final host role of domestic dogs in the life cycle of S. tenella in Egypt, which provides a precious diagnostic tool for further epidemiological studies and for the assessment of the effectiveness of control measures for this disease.

The aim of this research was to investigate the influence of dietary humic substances on growth efficiency, faeces indicators, and the bacteriological composition of faeces in piglets. Twenty-four crossbred young pigs (Slovakian White x Landrace) in total were allotted into two groups: the experimental and the control groups. They were fed a diet with humic substances supplemented at 0.5% in the experimental group. In this study, there were no significant differences in the monitored production indicators of piglets. The addition of humic substances increased the dry matter of faeces in the experimental group (P < 0.001). Other indicators in the faeces samples were not significantly affected by dietary treatment. We observed a significant decrease in the counts of coliform bacteria in the faeces (P < 0.05) of the experimental group. The results also showed a tendency to increase the counts of the lactic acid bacteria and a decrease in the counts of the Enterobacteriaceae in the piglets’ experimental group compared to the control group. A decrease in coliform bacteria counts in the faeces of experimental animals showed a positive effect on the microbiota in the intestine. It was concluded that the addition of humic substances to the diet of piglets could have a positive impact on gut flora without having a negative effect on production performance or other fecal characteristics.

Abstract Background Reproducible and reliable studies of cat and dog faecal microbiomes are dependent on many methodology-based variables including how the faecal stools are sampled and stored prior to processing. The current study aimed to establish an appropriate method for sampling and storing faecal stools from cats and dogs which may also be applied to privately-owned pets. The approach investigated the effects of storing faeces for up to 12 h at room temperature and sampling from various locations within the stool in terms of microbial diversity, relative taxa abundances and DNA yield. Faeces were collected from 10 healthy cats and 10 healthy dogs and stored at room temperature (20 °C). Samples were taken from various locations within the stool (the first emitted part (i), the middle (ii) and the last emitted end (iii), at either surface or core) at 0, 0.5, 1, 2, 3, 6 and 12 h, stabilised and stored at -80 °C. DNA was extracted from all samples, using Illumina NovaSeq. Results Faecal bacterial composition of dogs and cats shown no statistically significant differences in alpha diversity. Bacteroidetes, Firmicutes, Proteobacteria and Actinobacteria were the most prevalent phyla. Cat and dog samples were characterized by a dominance of Prevotella, and a lack of Fusobacterium in feline stools. Room temperature storage of cat and dog faecal samples generally had no significant effect on alpha diversity, relative taxa abundance or DNA yield for up to 12 h. Sampling from regions i, ii or iii of the stool at the surface or core did not significantly influence the outcome. However, surface cat faecal samples stored at room temperature for 12 h showed a significant increase in two measures of alpha diversity and there was a tendency for a similar effect in dogs. When comparing samples with beta diversity measures, it appeared that for dog and cat samples, individual effect has the strongest impact on the observed microbial diversity (R2 0.64 and 0.88), whereas sampling time, depth and horizontal locations significantly affected the microbial diversity but with less impact. Conclusion Cat and dog faeces were stable at room temperature for up to 12 h, with no significant changes in alpha diversity, relative taxa abundance and DNA concentration. Beta diversity analysis demonstrated that despite an impact of the sampling storing time and the surface of the sampling, we preserved the identity of the microbial structure linked to the individual. Finally, the data suggest that faecal stools stored for > 6 h at room temperature should be sampled at the core, not the surface.

ABSTRACTChronic wasting disease (CWD) is a fatal transmissible spongiform encephalopathy that affects both free-ranging and farmed cervid species, including mule deer, white-tailed deer, and elk (Odocoileus hemionus, Odocoileus virginianus, and Cervus canadensis). Due to the long incubation period and variability of clinical signs, CWD can expand and spread to new areas before they reach diagnostically detectable levels. Antemortem testing methods currently available can be difficult to obtain and to be applied to the large numbers required for adequate surveillance. However, key volatile biomarkers could be harnessed for non-invasive antemortem surveillance. Detection dogs are the most effective tool currently available for volatile detection; dogs can effectively complete wildlife surveys at rates surpassing that of humans. This study is the first to demonstrate that trained detection dogs can be used as an antemortem test for CWD. First, we trained three dogs to differentiate between CWD-positive and CWD-negative white-tailed deer faeces in a laboratory setting. Dogs spent significantly more time at the positive sample than the negative samples, suggesting that they differentiated between the positive and negative volatile signatures. We then trained the same dogs to search for CWD-positive faecal samples in a more naturalistic field setting. In the field, dogs found 8/11 CWD-positive samples and had an average false detection rate of 13%. These results suggest that dogs can be trained to differentiate CWD-positive faeces from CWD-negative faeces in both laboratory and field settings. Future studies will compare canine accuracy to other antemortem methods, as well as improved canine training methods.

The gut microbiota is a key and primary stimulus for the development of a host’s immune system. The early establishment of the gut microbiota is affected by several perinatal factors but little is known about their influence on shaping normal immune development and, consequently, on the programming of future health. The analysis of different immune compounds is well-documented in serum samples; however, their presence in faecal samples has not been studied, and this information could be valuable in early life. In this context, the authors of this study aimed to both describe the immunological faecal profile of a cohort of one-month-old infants and describe the impact of different perinatal factors, exploring possible associations between immune compounds and gut microbiota in faecal samples. Clear differences in immune profile were observed between full-term and premature infants. Breastfeeding increases IgG2, IgG4, and IgA; in addition, male babies showed some increased Igs, among other observations. Overall, the findings of this study reinforce the hypothesis that microorganisms and immune compounds interact with each other in the early neonatal gut and that understanding these interactions in depth will help us comprehend the influence of the gut microbiota on short- and long-term infant health outcomes.

Pig homeostasis is challenged by stressful production practices, like road transportation. Glucocorticoids (GCs) are mediators of reactive homeostasis, and their concentrations are frequently used as a stress indicator. The adrenocortical activity of fattening female and castrated male pigs was monitored over a 5-day longitudinal study. A bi-factorial experimental design was applied on day 2; 18 pigs in pen 1 were transported for 3 h (T; 1.2 m2/pig), and 18 pigs were kept in pen 2 (NT). Ten pigs from each pen were treated with dexamethasone (T-D or NT-D), and eight with saline solution (T-SS or NT-SS). Adrenocortical activity was assessed by measuring the levels of faecal glucocorticoid metabolites (FGMs) and hair cortisol and cortisone. In T-SS pigs, the level of FGMs was higher after transportation than in NT-SS pigs. The level of FGMs of T-D pigs initially increased but then reached similar levels to those of NT-SS sooner than T-SS. In contrast, hair cortisol and cortisone did not respond to the treatments. Nevertheless, the hair cortisone/cortisol ratio increased due to transport and decreased after dexamethasone administration. Daily faecal sampling proved still more reliable than 60-day hair sampling for assessing adrenocortical activity. Transported pigs recovered their adrenocortical baseline levels within 24 h. Dexamethasone attenuated the response to transport.

Cereulide is an emetic toxin produced by some strains of Bacillus cereus. This bacterial toxin, a cyclic 1.2 kDa dodecadepsipeptide, is stable to heat and acids and causes nausea and vomiting when ingested via contaminated food. This work aimed to develop and validate a targeted analytical method applying liquid chromatography-tandem mass spectrometry (LC–MS/MS) to quantify this toxin in food and human faeces. Samples were extracted with acetonitrile in the presence of 13C6-cereulide, a labelled internal standard, and purified by centrifugation and filtration. The limits of quantification were 0.5 and 0.3 µg kg−1 for food and faeces, respectively. The linearity of the method was very good, with calculated R2 values above 0.995. The mean recovery of the method was within the acceptable range of 70.0%–120.0%, the repeatability was not higher than 7.3%, and the highest intra-laboratory reproducibility was 8.9%. The estimated range for the expanded measurement uncertainty was between 5.1% and 18.0%. The LC–MS/MS method was used to analyse one food sample (rice) from a Belgian foodborne outbreak and five faecal samples from patients with clinical symptoms after consumption of the contaminated rice. Supplemental feed for free‐ranging deer is often advocated by managers to compensate for winter survival. However, providing extra food has great costs so it is important to know how essential supplemental feed is for deer populations. In our study, supplemental feed is supplied year round for free‐ranging Père David's deer Elaphurus davidianus since early 2019 to help them survive food shortages in the Jiangsu Dafeng Milu National Nature Reserve. To understand use of supplemental feed, we quantified the biomass contributions of C3 plants, C4 plants and supplemental feed consumed by free‐ranging deer by assessing their faecal carbon and nitrogen stable isotope values in May, July, October and December. The diet–faeces trophic shift of deer was assessed using semi‐captive animals. Faeces in both areas were collected and faecal δ13C values revealed more variable diets of free‐ranging deer than semi‐captive deer in different months. MixSIAR model showed that C4 plants contributed greatly to deer diet biomass (75.1, 82, 64.3, 79.4%, respectively in May, July, October and December) compared with C3 plants and supplemental feed. We suggest that managers should consider the efficiency of long‐term supplemental feed and consider the selection of C3 and C4 plants during restoration. [ABSTRACT FROM AUTHOR]

Seed dispersal influences the survival and distribution of plant species and is an important mechanism for maintaining floristic diversity. Bushpigs (Potamochoerus larvatus) are large mammals of indigenous forests and well-wooded areas of eastern and southern Africa and may play an important role as seed dispersers. However, the diversity of fruits consumed and the effect of digestion on seeds remains poorly understood. This study was conducted from October 2018 to January 2019 and gives first insights into seed dispersal by bushpigs in the Soutpansberg Mountain range, South Africa. Here, we collected bushpig faeces and assessed the number of seeds and seed species found. Furthermore, germination experiments with digested and nondigested seeds and fruits were conducted. Our results show that bushpigs are omnivorous, consuming fruits and seeds of 126 different plant species. The proportion of seeds per plant species found in faeces was unhomogeneously distributed with a few hyperabundant species, in particular Searsia chirindensis (55%). The germination experiments showed that gut passage did, overall, not influence germination or seedling growth rates. However, seeds remaining within fruits showed reduced growth, pointing to a high importance of fruit consumption for seed fate. Our results suggest that bushpigs are major seed dispersers of a large variety of fruiting plant species in wooded areas of Africa. [ABSTRACT FROM AUTHOR]

Background: Sarcocystis species are obligatorily heteroxenous protozoan parasites with predator–prey life cycles. Global Knowledge about the epidemiology and the distribution pattern of different Sarcocystis species in dog feces are very scarce. Therefore, the current investigation was conducted to declare the occurrence of Sarcocystis in the fecal specimens of the most common canids in Egypt, the domestic dogs, and to identify the species present using various parasitological and molecular approaches. Methods: A total of 100 dog fecal samples were collected and screened using fecal sugar flotation test for the presence of Sarcocystis oocysts/sporocysts. Additionally, thirty samples were used for genomic DNA extraction. The 18S rRNA gene fragment was the target of primers for a PCR, followed by purification and sequencing of the amplicons. Results: Currently, the results obtained reviewed that 4% of fecal samples were positive for Sarcocystis spp. using LM. Additionally, Sarcocystis spp. were verified in sixteen dogs (53.3%, 16/30) using PCR and subsequent sequencing protocols. Statistically, insignificant difference in prevalence of sarcocystosis relative to age and gender was noticed. Morphologically, the detected sporocysts measured 13.2–16.0 × 9.4–11 μm. Dung beetles feed mainly on scarce and ephemeral mammalian faeces making them vulnerable to declines of mammal populations. Therefore, studying how diverse groups like the Scarabaeinae subfamily utilize alternative food sources, such as decomposing invertebrates, enhances our understanding of their distribution and resilience in defaunated areas. One poorly studied aspect of the feeding habits of dung beetles is the potential differences between the sexes and their predatory behaviour. We evaluated how the sex (male and female) of individuals of Chalconotus convexus affected individual resource attraction to different bait types. Since C. convexus is known to feed on faeces, carrion and dead millipedes, we also test whether it can predate live individuals of millipedes. We used a combination of two field samplings and one laboratory experiment, using pitfall traps baited with faeces, carrion, millipedes and millipede's defensive compounds. While the preference for millipede was not associated with sex, we found a significant relationship between faeces and carrion, where females of C. convexus are significantly less attracted to faeces when compared to males and also males are significantly less attracted to carrion when compared to females. Furthermore, we also show that while actively consuming dead millipedes, C. convexus is unable to predate on live individuals. Our study illustrates that the versatility of food sources may help to explain the high abundance and wide distribution of C. convexus in Africa. [ABSTRACT FROM AUTHOR]

Especially in the temperate zone, female bats are exposed to increased energy demands during gestation. In addition, abiotic factors, such as ambient temperature and pathogen load, can significantly affect reproduction success. Bats are considered natural reservoirs for a wide variety of pathogenic agents, and have developed strong immune systems to cope. Nevertheless, very little is known about the influence of these pathogenic microorganisms on bat health as most infections are asymptomatic. In this study, we monitored presence of non-tuberculous mycobacteria (NTM) in female common noctule bats Nyctalus noctula during gestation. Eight NTM species, subspecies or complexes (Risk Group 1: Mycobacterium hiberniae and M. terrae complex; Risk Group 2: M. arupense, M. avium ssp. hominissuis, M. fortuitum, M. interjectum, M. peregrinum, M. septicum) were confirmed as part of the female gut microbiome during gestation, with representation changing during foetal development. Following hibernation, most females were infected, though infection load and severity were relatively low. Just prior to parturition, however, a high proportion of females were infected with a high, often pathogenic, NTM load. Females who gave birth to twins had a higher (though non-significant) proportion of positive samples after hibernation and before parturition. Negative correlations between two consecutive measurements suggest that female N. noctula can cope with NTM loading, even during energy-demanding pregnancies. Bat faeces appear to be a source of numerous NTM species and, as such, bats can be considered as vectors for NTM spread in the environment. [ABSTRACT FROM AUTHOR]

Introduction: To consider the growing health issues caused by antibiotic resistance from a “one health” perspective, the contribution of meat production needs to be addressed. While antibiotic resistance is naturally present in microbial communities, the treatment of farm animals with antibiotics causes an increase in antibiotic resistance genes (ARG) in the gut microbiome. Pigs are among the most prevalent animals in agriculture; therefore, reducing the prevalence of antibiotic-resistant bacteria in the pig gut microbiome could reduce the spread of antibiotic resistance. Probiotics are often studied as a way to modulate the microbiome and are, therefore, an interesting way to potentially decrease antibiotic resistance.Methods: To assess the efficacy of a probiotic to reduce the prevalence of ARGs in the pig microbiome, six pigs received either treatment with antibiotics (tylvalosin), probiotics (Pediococcus acidilactici MA18/5M; Biopower® PA), or a combination of both. Their faeces and ileal digesta were collected and DNA was extracted for whole genome shotgun sequencing. The reads were compared with taxonomy and ARG databases to identify the taxa and resistance genes in the samples.Results: The results showed that the ARG profiles in the faeces of the antibiotic and combination treatments were similar, and both were different from the profiles of the probiotic treatment (p < 0.05). The effects of the treatments were different in the digesta and faeces. Many macrolide resistance genes were detected in a higher proportion in the microbiome of the pigs treated with antibiotics or the combination of probiotics and antibiotics. Resistance-carrying conjugative plasmids and horizontal transfer genes were also amplified in faeces samples for the antibiotic and combined treatments. There was no effect of treatment on the short chain fatty acid content in the digesta or the faeces.Conclusion: There is no positive effect of adding probiotics to an antibiotic treatment when these treatments are administered simultaneously.

Alterations in faecal lactobacilli in autistic children have been reported, but little is known related to age and disorder severity. We used a culture-based method and partial 16S rRNA gene sequencing to isolate and identify lactobacilli strains from faeces of Tunisian autistic children (ASD group) and compared them with strains isolated from siblings (SIB) and children from the general population (GP). The ASD group displayed an increased number of different species compared to SIB and GP. Differences in species abundance with age accounted for a significant decrease in the abundance of Lactiplantibacillus plantarum/Lactiplantibacillus pentosus isolates in the GP at the age of 8–10 years compared to the age of 4–7 years, and to a significantly lower abundance of Lacticaseibacillus rhamnosus in the ASD group with respect to SIB and the GP at the age of 8–10 years. Simpson’s and Shannon–Wiener indices showed a more pronounced species diversity increase with age in the GP group compared to the ASD and SIB groups. Minor differences were found in lactobacilli prevalence and in species diversity between children with severe and mild-to-moderate ASD. Overall, we found substantial differences in the profile of faecal lactobacilli species in the ASD and GP groups at the age of 8–10 years.

AbstractNumerous studies have analyzed the distribution and concentration of microplastic particles (MPs) in various aquatic habitats, and estimated how they affect residual biota. Decidedly less research concerns how organisms (e.g. ichthyofauna) affect the quantitative and qualitative characteristics of MPs in the water. Therefore, the aim of our study was to test, if fish can increase the vertical flux of MPs to sediments. This would be the result of the combined effect of increased sedimentation rate and the active transport resulting from vertical migration of the fish in the simultaneous presence of a thermal gradient (and in turn water density gradient). We conducted eight 5-day-long experiments in thermally stratified twin vertical columns filled with a suspension of polystyrene beads of 25 and 250 µm in size, with initial density of 1000 and 200 × L−1, respectively. In each of the experiments, we placed sediment traps at three different depths in each of the columns, and four planktivorous fish in one of them. During the experiments, we monitored the distribution of fish. At the end of the experiment, we determined the number of MPs and the amount of detritus in both sediment traps and water. We found that the presence of planktivorous fish significantly increased the vertical flux of MPs to sediments and decreased the density of MPs in the water column. These effects seem to be more likely due to the increased sedimentation rate of MPs due to the fish activity, rather than to their active transport, as MPs in the experimental variant were covered with detritus to a greater extent, than in the control, and the fish remained in the subsurface layer in the water column. Our study seems to be the first one showcasing the effect of fish on the concentration and distribution of MPs in aquatic ecosystems.

Escherichia coli is quite a common and sometimes pathogenic bacterium that can cause acute to severe infection in poultry birds affecting their performance. E. coli can also cause a few extra-intestinal infections too in poultry birds. Nowadays antimicrobial resistance is quite a menace which is reported from several gram-negative bacteria including E. coli. These traits in E. coli are governed by a few β-lactamases producing genes which may also be associated with plasmids in the pathogen. This study aimed to detect the presence of extended-spectrum (ESBL) and AmpC (ACBL) beta-lactamases producing E. Coli from fecal samples of diseased and apparently healthy poultry birds from different districts of West Bengal, India. A total of 177 (62.32%) E. coli isolates were detected and confirmed from the poultry fecal samples/ cloacal swabs (n=284) collected in this study. All isolates were typical in morphology. One hundred thirty-two (74.58%) bacterial isolates were positive for phenotypical beta-lactamases production, of which 83 (46.89%) were positive for phenotypical ACBL production. A total of 41 and 17 isolates were found to be positive for PCR detection of the blaCTXM and blaSHV genes respectively. No blaTEM gene was detected. Again 95 (53.67%) isolates possessed the blaAmpC gene as detected in this study. Antibiogram of the ESBLproducing E. coli isolates (49) revealed a significantly high level of resistance against most of the commonly used antimicrobials like ceftriaxone, ampicillin (both 100%), cefotaxime (97.96%), ceftazidime (91.84%) amoxicillin/clavulanic acid (85.71%), azithromycin, tetracycline, norfloxacin (@70-75%). These isolates were found to be sensitive (@ 96-81%) against a few antimicrobials, viz. amikacin, imipenem, gentamicin, and ampicillin/sulbactam which is a major point of concern.

Breastfeeding represents a strong selective factor for shaping the infant gut microbiota. Besides providing nutritional requirements for the infant, human milk is a key source of oligosaccharides, human milk oligosaccharides (HMOs), and diverse microbes in early life. This study aimed to evaluate the influence of human milk microbiota and oligosaccharides on the composition of infant faecal microbiota at one, three, and nine months postpartum. We profiled milk microbiota, HMOs, and infant faecal microbiota from 23 mother–infant pairs at these time points. The predominant genera in milk samples were Streptococcus, Staphylococcus, and an unclassified Enterobacteriaceae genus-level taxon (Enterobacteriaceae uncl.), whereas the infant faecal microbiota was predominated by Bifidobacterium, Bacteroides, and Enterobacteriaceae uncl. Mother–infant dyads frequently shared bacterial amplicon sequence variants (ASVs) belonging to the genera Bifidobacterium, Streptococcus, Enterobacteriaceae uncl., Veillonella, Bacteroides, and Haemophilus. The individual HMO concentrations in the milk showed either no change or decreased over the lactation period, except for 3-fucosyllactose (3-FL), which increased. Neither maternal secretor status nor HMO concentrations were significantly associated with microbiota composition at the different ages or the bacterial ASVs of maternal milk and infant faeces. This study suggests an age-dependent role of milk microbes in shaping the gut microbiota, while variations in HMO concentrations show limited influence.

Differences in species abundance with age accounted for a significant decrease in the abundance of Lactiplantibacillus plantarum/Lactiplantibacillus pentosus isolates in the GP at the age of 8–10 years compared to the age of 4–7 years, and to a significantly lower abundance of Lacticaseibacillus rhamnosus in the ASD group with respect to SIB and the GP at the age of 8–10 years. Simpson's and Shannon–Wiener indices showed a more pronounced species diversity increase with age in the GP group compared to the ASD and SIB groups. Minor differences were found in lactobacilli prevalence and in species diversity between children with severe and mild-to-moderate ASD. This study was carried out to identify T. solium eggs in feacal droppings of one hundred (100) unrestrained pigs in different sampling areas of WapanNghaku and Mission Quarters in Wukari, Southern Taraba State, Nigeria, using the formol-ether-sedimentation and direct wet mount techniques. Taenia solium (T. solium), also known as pork tapeworm, is a segmented intestinal parasite of humans and pigs that is endemic in many developing countries. The prevalence of eggs in both diagnostic methods was compared using percentages. The prevalence of T. solium eggs using the formol-ether-sedimentation and direct wet mount technique was 48% and 32% respectively. In both diagnostic isolation methods, the rate was highest in the WapanNghaku (formol-ethersedimentation: 56%; direct wet mount: 40%) than in the Mission Quarters areas (formol-ethersedimentation: 40%; direct wet mount: 20%). This study which describes for the first time, the study on T. solium in nomadic pigs and techniques to identify their eggs in faeces and determine their prevalences in Wukari, has improved data on the epidemiology of T. solium by showing that nomadic pig farming in Wukari predisposes transmission of taeniasis in the study population of pigs that serve as reservoirs of T. solium eggs. Furthermore, these eggs can be isolated with more accuracy using the formol-ether-sedimentation technique which is simple. Whilst a robust surveillance data is advocated, there is need to adopt a quality intensive system of pig management to maintain good hygienic environment and disease free pork for consumption in the study areas. Mortality is prevalent in koala joeys being hand reared/rehabilitated, with little knowledge about the causes of such high rates of mortality. Wildlife hospitals/rehabilitation expose koala joeys to novel husbandry environments that create stressful stimulus for these animals. Furthermore, loss of the mother and/or being raised by a human can be threatening and/or stressful. The hypothalamic–pituitary–adrenal axis plays a vital role in mediating the stress endocrine response of vertebrate species including marsupials (by adrenocortical release of glucocorticoids such as cortisol), however, there are no studies that quantify cortisol metabolites in koala joeys. To contribute to this dearth of knowledge, the primary aim of this study was to quantify the levels of faecal cortisol metabolites (as an index of physiological stress) in koala joeys undergoing rehabilitation. A total of 39 faecal samples were collected from four koala joeys admitted at a wildlife hospital in New South Wales, Australia. The samples were processed and analysed for faecal cortisol metabolites (FCM) using a polyclonal R4866 cortisol enzyme-immunoassay which has been previously biologically validated in adult koalas. The results indicated that there was significant inter-individual variation in FCM concentrations among the joeys, however, these differences were not based on the sex of the joeys. The average FCM concentration among joeys varied from 18.34 to 44.18 ng/g. Our study provides the first record of physiological stress in male and female rescued koala joeys by indexing FCM concentrations during rehabilitation. Koala joeys undergoing rehabilitation often die from unknown causes. We indexed physiological stress in rehabilitating koala joeys by monitoring faecal cortisol metabolites. We found significant inter-individual differences in faecal cortisol metabolite concentrations between koala joeys, not related to the sex of the joeys. These results provide the first record of concentrations of faecal cortisol metabolites in rehabilitating koala joeys, which will prove valuable for future assessments of stress in koala joeys. Animal constructions are the outcomes of complex evolutionary, behavioural, and ecological forces. A brief review of diverse animal builders, the materials used, and the functions they provide their builders is provided to develop approaches to studying faecal-based constructions and faecal-carrying in leaf beetles (Coleoptera: Chrysomelidae). Field studies, rearing, dissections, photography, and films document shields constructed by larvae in two species in two tribes of the subfamily Cassidinae, Calyptocephala attenuata (Spaeth, 1919) (Spilophorini), and Cassida sphaerula Boheman, 1853 (Cassidini). Natural history notes on an undetermined Cassidini species and Stolas cucullata (Boheman, 1862) (Tribe Mesomphaliini) outline the life cycle of tortoise beetles and explain terms. Commonly, the cassidine shield comprises exuviae onto which faeces are daubed, producing a pyramidal-shaped shield that can cover most of the body (up to the pronotum). In Cal. attenuata the larval shield comprises only exuviae, while in Cass. sphaerula, instar 1 initiates the shield by extending its telescopic anus to apply its own faeces onto its paired caudal processes; at each moult the exuvia is pushed to the caudal process base but remains attached, then more faeces are applied over it. The larva’s telescopic anus is the only tool used to build and repair the shield, not mouthparts or legs, and it also applies chemicals to the shield. Pupae in Cal. attenuata retain part of the exuviae-only shield of instar VI, while pupae in Cass. sphaerula retain either the entire 5th instar larval shield (faeces + all exuviae) or only the 5th larval exuvia. The caudal processes are crucial to shield construction, shield retention on the body, and as materials of the central scaffold of the structure. They also move the shield, though the muscular mechanism is not known. Altogether the faecal + exuviae shields may represent a unique morpho-behavioural synapomorphy for the crown-clade Cassidinae (10 tribes, ~ 2669 species) and may have been a key innovation in subsequent radiation. Abstract Background Calves undergo nutritional, metabolic, and behavioural changes from birth to the entire weaning period. An appropriate selection of weaning age is essential to reduce the negative effects caused by weaning-related dietary transitions. This study monitored the faecal microbiome and plasma metabolome of 59 female Holstein calves during different developmental stages and weaning times (early vs. late) and identified the potential associations of the measured parameters over an experimental period of 140 days. Results A progressive development of the microbiome and metabolome was observed with significant differences according to the weaning groups (weaned at 7 or 17 weeks of age). Faecal samples of young calves were dominated by bifidobacterial and lactobacilli species, while their respective plasma samples showed high concentrations of amino acids (AAs) and biogenic amines (BAs). However, as the calves matured, the abundances of potential fiber-degrading bacteria and the plasma concentrations of sphingomyelins (SMs), few BAs and acylcarnitines (ACs) were increased. Early-weaning at 7 weeks significantly restructured the microbiome towards potential fiber-degrading bacteria and decreased plasma concentrations of most of the AAs and SMs, few BAs and ACs compared to the late-weaning event. Strong associations between faecal microbes, plasma metabolites and calf growth parameters were observed during days 42–98, where the abundances of Bacteroides, Parabacteroides, and Blautia were positively correlated with the plasma concentrations of AAs, BAs and SMs as well as the live weight gain or average daily gain in calves. Conclusion The present study reported that weaning at 17 weeks of age was beneficial due to higher growth rate of late-weaned calves during days 42–98 and a quick adaptability of microbiota to weaning-related dietary changes during day 112, suggesting an age-dependent maturation of the gastrointestinal tract. However, the respective plasma samples of late-weaned calves contained several metabolites with differential concentrations to the early-weaned group, suggesting a less abrupt but more-persistent effect of dietary changes on host metabolome compared to the microbiome.

The transmission of pathogens from reservoir to recipient host species, termed pathogen spillover, can profoundly impact plant, animal, and public health. However, why some pathogens lead to disease emergence in a novel species while others fail to establish or do not elicit disease is often poorly understood. There is strong evidence that deformed wing virus (DWV), an (+)ssRNA virus, spills over from its reservoir host, the honeybee Apis mellifera, into the bumblebee Bombus terrestris. However, the low impact of DWV on B. terrestris in laboratory experiments suggests host barriers to virus spread in this recipient host. To investigate potential host barriers, we followed the spread of DWV genotype B (DWV-B) through a host’s body using RT-PCR after experimental transmission to bumblebees in comparison to honeybees. Inoculation was per os, mimicking food-borne transmission, or by injection into the bee’s haemocoel, mimicking vector-based transmission. In honeybees, DWV-B was present in both honeybee faeces and haemolymph within 3 days of inoculation per os or by injection. In contrast, DWV-B was not detected in B. terrestris haemolymph after inoculation per os, suggesting a gut barrier that hinders DWV-B’s spread through the body of a B. terrestris. DWV-B was, however, detected in B. terrestris faeces after injection and feeding, albeit at a lower abundance than that observed for A. mellifera, suggesting that B. terrestris sheds less DWV-B than A. mellifera in faeces when infected. Barriers to viral spread in B. terrestris following oral infection may limit DWV’s impact on this spillover host and reduce its contribution to the community epidemiology of DWV.

ABSTRACT: The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.

Objectives: Inflammatory bowel disease (IBD) and irritable bowel syndrome (IBS) are increasingly prevalent disorders. Faecal calprotectin is useful in the differential diagnosis of IBD from IBS and monitoring IBD activity. We verified the Bühlmann fCAL turbo faecal calprotectin assay on the Binding Site, Optilite benchtop analyser. Design: Accuracy, precision, lower limit of quantitation (LLoQ), and linearity of the Bühlmann fCAL turbo faecal calprotectin assay on the Binding Site, Optilite benchtop analyser were ascertained. Comparison with the Bühlmann Quantum Blue fCAL extended and DiaSorin, Liaison calprotectin assays were also undertaken. Difference between assays was evaluated using the Wilcoxon signed-rank test and method comparison was undertaken using Spearman’s rank correlation (rs), difference plots and Passing-Bablok regression analyses. Simple Summary: Stable isotope analysis is a technique increasingly used for studying many aspects of wildlife behaviour, such as diet, migrations, and trophic relationships. However, scarce information is available on how isotopes are assimilated into the body under conditions such as nutritional stress situations or intrasexual competition for mates. In this study, we evaluated how intrasexual competition between male red deer from free-ranging populations in Andalusia (Spain) is reflected in the ratio of stable isotopes of carbon and nitrogen (δ13C and δ15N) in faeces. Our results showed that in populations with high intrasexual competition δ15N values in faeces were significantly lower than in low intrasexual competition populations. No differences were found for δ13C. This study provides a basis for interpreting δ15N values related to different protein usage under various nutritional stress situations in male red deer and novel applications of the use of nitrogen isotopes. Isotopic analysis of carbon and nitrogen in faeces is a reliable methodology for studying ecology in wildlife. Here, we tested this technique to detect variations in carbon and nitrogen isotopic ratios (δ13C and δ15N) in two different intrasexual competition scenarios of male Iberian red deer (Cervus elaphus hispanicus) using faeces of individuals collected during hunting actions in South-eastern Spain. The carbon isotopic ratio (δ13C) was not found to be significant, likely due to similar diet composition in all individuals. However, the nitrogen isotopic ratio (δ15N) was found to be lower in populations where sexual competition between males during the rut was higher compared to low-competition populations. Therefore, this study suggests a different use of proteins by an individual male red deer depending on the sexually competitive context in which he lives. Although further research is needed, these results show the potential of isotopic analysis as a tool for studying individual and populational variations in the level of intrasexual competition, with implications in evolutionary ecology and population management. Estuarine and coastal waters are areas of potential concern for antimicrobial resistance because of the discharge of wastewater from sewage treatment plants and the run-off from urban and agricultural lands. Herein, we evaluate the antimicrobial resistance profiles in bacteria from dugongs (Dugong dugon), mammals that inhabit and feed in shallow coastal regions and, thus, are vulnerable to encountering water and sediment contaminated by human activities. Bacterial isolates were cultured from fresh faeces of four wild dugongs, as well as from one sediment sample from a dugong foraging ground in Queensland, Australia. Ten bacterial isolates underwent phenotypic antimicrobial susceptibility testing using disc diffusion and minimum inhibitory concentration testing, and genotypic resistance and virulence gene identification through whole genome sequencing. Four Staphylococcus warneri isolates and one Bacillus cereus isolate from dugong faeces were resistant to penicillin, with two S. warneri isolates also displaying resistance to trimethoprim. Four Escherichia coli isolates were all resistant to ampicillin. Resistance genes, including fosB, BcII, dfrC, blaZ, and mdfA, were identified in the isolates cultured from dugong faeces with two virulence genes (gad and lpfA) identified in all E. coli isolates. Lysinibacillus sphaericus cultured from the marine sediment and B. cereus from dugong faeces displayed phenotypic multidrug resistance (across categories of nonextended spectrum cephalosporins, penicillins and beta-lactamase inhibitors, and clindamycin; and for L. sphaericus, phosphonic acids). These results demonstrate the role that dugongs can play as a sentinel species for antimicrobial resistance in the coastal waters across their range, which includes both disturbed urban and rural regions. Hedgehogs, which are kept as pets worldwide, are prohibited from being sold or kept as pets in Türkiye. The aim of the study was to investigate Salmonella and endo-parasite carriage in hedgehogs and to determine the antibacterial resistance profiles of isolated Salmonella agents. Free-range hedgehogs living in parks and gardens of Istanbul that were brought to veterinary clinics by animal lovers for check-up and treatment were used for sampling. Thirty faecal samples were taken from the animals. For bacteriological examinations, samples were seeded on selective media, and then the isolates were identified by conventional methods as Salmonella spp. Identification and antibiotic susceptibilities of the isolates were determined by the BD Phoenix Automated Microbiology System. All faecal samples were also investigated by PCR for Salmonella spp. specific gene regions. The aminoglycoside and quinolone resistance profiles of the strains were analysed by PCR. On parasitological examination, samples were analysed for parasites’ eggs, oocysts, and larvae. Two Salmonella isolates were determined as Salmonella enterica serovar Typhimurium (S. Typhimurium), whose antigenic formula was 4,5,12:i:1,2. The isolates were phenotypically resistant to ciprofloxacin but did not harbour any of the genes examined. Out of 30 faecal samples, 19 (63.3%) were positive for one or more genera of parasites. The hedgehogs were infected with the species of Capillaria spp. (60%), Crenosoma spp. (13.3%), Eimeria spp. (10%) and Acanthocephala spp. (6.6%). Knowledge of endogenous nutrient losses is important when estimating the nutrient requirements of animals. It has been suggested that faecal endogenous phosphorus (P) losses differ between growing and adult horses, but studies on foals are scarce. In addition, studies on foals on forage‐only diets with different P contents are lacking. Thus this study: (1) assessed faecal endogenous P losses in foals fed a grass haylage‐only diet close to or below estimated P requirements; (2) evaluated use of serum cross‐linked carboxyterminal telopeptides of type‐I collagen (CTx) as a marker of bone resorption secondary to low‐P intake; and (3) examined whether analysis of faecal P concentration on a dry matter (DM) basis could be used as an indicator of P intake. Six foals were fed three grass haylages (fertilised to contain different amounts of P: 1.9, 2.1, 3.0 g/kg DM) for 17‐day periods in a Latin square design. Total collection of feaces was performed by the end of each period. Faecal endogenous P losses were estimated using linear regression analysis. There was no difference in the concentration of CTx in plasma between diets in samples collected on the last day of each period. A correlation was found (y = 0.64x − 1.51; r2 = 0.75, p < 0.0001) between P intake and faecal P content, but regression analysis indicated that underestimation as well as overestimation of intake is likely if faecal P content is used to assess intake. It was concluded that faecal endogenous P losses in foals are low, probably no higher than in adult horses. It was also concluded that plasma CTx cannot be used to assess short‐term low‐P intake in foals and that faecal P content cannot be used to assess differences in P intake, at least not when P intake is close to or below estimated P requirements.
