Pepper (Capsicum annuum L.) carrying the gds (corresponding to bs5) gene can prevent the development of bacterial leaf spot disease without HR. However, little is known regarding the development of the resistance mechanism encoded by gds, especially its influence on the bacterium. Here, the effect of gds was compared with pattern-triggered immunity (PTI), another form of asymptomatic resistance, to reveal the interactions and differences between these two defense mechanisms. The level of resistance was examined by its effect on the bacterial growth and in planta expression of the stress and pathogenicity genes of Xanthomonas euvesicatoria. PTI, which was activated with a Pseudomonas syringae hrcC mutant pretreatment, inhibited the growth of Xanthomonas euvesicatoria to a greater extent than gds, and the effect was additive when PTI was activated in gds plants. The stronger influence of PTI was further supported by the expression pattern of the dpsA bacterial stress gene, which reached its highest expression level in PTI-induced plants. PTI inhibited the hrp/hrc expression, but unexpectedly, in gds plant leaves, the hrp/hrc genes were generally expressed at a higher level than in the susceptible one. Abstract: The DNA-binding protein from stationary phase (Dps) protein family plays an important role in protecting microorganisms from oxidative and nutritional stresses. In silico analysis of the promoter region of alr3808, a dpsA homologue from the cyanobacterium Nostoc sp. PCC7120 shows putative iron-boxes with high homology with those recognized by FurA (ferric uptake regulator). Evidence for the modulation of dpsA by FurA was obtained using in vitro and in vivo approaches. SELEX linked to PCR was used to identify P dpsA as a FurA target. Concurrently, EMSA assays showed high affinity of FurA for the dpsA promoter region. DpsA expression analysis in an insertional mutant of the alr1690-αfurA message (that exhibited an increased expression of FurA) showed a reduced synthesis of DpsA. These studies suggest that FurA plays a significant role in the regulation of the DpsA. [Copyright &y& Elsevier]

The sigD and sigE genes, which encode two alternative σ-factors from the unicellular marine cyanobacterium Synechococcus sp. PCC 7002, were cloned and characterized. Strains in which the sigD and sigE genes were insertionally inactivated were viable under standard laboratory conditions, indicating that SigD and SigE are group 2 σ-factors. When stationary-phase cells were diluted into fresh growth medium, it was observed that the sigE mutant strain required longer times to re-establish exponential growth than the wild-type strain. By monitoring the growth rates in such dilution experiments, it was observed that the lag times for the mutant strain became progressively longer as the original cultures progressed towards stationary phase. Transcripts for the sigE gene initially increased and subsequently decreased as cells grew further into stationary phase. It was determined that a functional SigE protein is required for the expression of the starvation-induced protein DpsA/PexB. The results suggest that SigE is involved in the transcription of genes specifically expressed in the post-exponential phase. [ABSTRACT FROM AUTHOR]

Operating staffs of a nuclear power plant (NPP) are responsible for returning the NPP to a stable state and alert authorities if there is the potential for offsite radiological consequences following an accident. An operator support tool (OST) using deep learning techniques and trained by data from dynamic probabilistic safety/risk assessment (DPSA/DPRA) is proposed to assist the NPP personnel in decision-making. The DPSA/DPRA methodology employs time-dependent branching conditions based on the evolving state of the NPP and accounts for complex hardware/process/software/human interactions to predict possible outcomes of the initiating event. A large number of scenarios generated from the DPSA/DPRA performed for a pressurized water reactor station blackout as a function of time were used to train the OST to predict possible offsite dose outcomes at 2-mile and 10-mile site boundaries for emergency response planning.
