Tardigrades are microscopic animals that survive desiccation by inducing biostasis. To survive drying tardigrades rely on intrinsically disordered CAHS proteins, which also function to prevent perturbations induced by drying in vitro and in heterologous systems. CAHS proteins have been shown to form gels both in vitro and in vivo, which has been speculated to be linked to their protective capacity. However, the sequence features and mechanisms underlying gel formation and the necessity of gelation for protection have not been demonstrated. Here we report a mechanism of fibrillization and gelation for CAHS D similar to that of intermediate filament assembly. We show that in vitro, gelation restricts molecular motion, immobilizing and protecting labile material from the harmful effects of drying. In vivo, we observe that CAHS D forms fibrillar networks during osmotic stress. Fibrillar networking of CAHS D improves survival of osmotically shocked cells. We observe two emergent properties associated with fibrillization; (i) prevention of cell volume change and (ii) reduction of metabolic activity during osmotic shock. We find that there is no significant correlation between maintenance of cell volume and survival, while there is a significant correlation between reduced metabolism and survival. Importantly, CAHS Ds fibrillar network formation is reversible and metabolic rates return to control levels after CAHS fibers are resolved. This work provides insights into how tardigrades induce reversible biostasis through the self-assembly of labile CAHS gels.

The moss Physcomitrium patens is a model system for the evolutionary study of land plants, and as such, it may contain as yet unannotated genes with functions related to the adaptation to water deficiency that was required during the water-to-land transition. In this study, we identified a novel gene, Bryophyte Co-retained Gene 1 (BCG1), in P. patens that is responsive to dehydration and rehydration. Under de- and rehydration treatments, BCG1 was significantly co-expressed with DHNA , which encodes a dehydrin (DHN). Examination of previous microarray data revealed that BCG1 is highly expressed in spores, archegonia (female reproductive organ), and mature sporophytes. In addition, the bcg1 mutant showed reduced dehydration tolerance, and this was accompanied by a relatively low level of chlorophyll content during recovery. Comprehensive transcriptomics uncovered a detailed set of regulatory processes that were affected by the disruption to BCG1. Experimental evidence showed that BCG1 might function in antioxidant activity, the abscisic acid pathway, and in intracellular Ca2+ homeostasis to resist desiccation. LEA_1 domain-containing proteins constitute a class of late-embryogenesis-abundant proteins that are highly hydrophilic and predominantly accumulate in mature seeds. Though LEA_1 proteins have been proven to be essential for seed desiccation tolerance and longevity, little information is available on their roles in non-seed storage organs. In this study, a first genome-wide characterization of the LEA_1 gene family was conducted in tigernut (Cyperus esculentus L., Cyperaceae), whose underground tubers are desiccation tolerant with a moisture content of less than 6%. Five family members identified in tigernut are comparative to four to six found in seven other Cyperaceae plants, but relatively more than three reported in Arabidopsis. Further comparison of 125 members from 29 plant species supports early divergence of the LEA_1 family into two phylogenetic groups before angiosperm radiation, and gene expansion in tigernut was contributed by whole-genome duplications occurring after the split with the eudicot clade. These two phylogenetic groups could be further divided into six orthogroups in the momocot clade, five of which are present in tigernut and the remaining one is Poaceae specific. Frequent structural variation and expression divergence of paralogs were also observed. Significantly, in contrast to seed-preferential expression of LEA_1 genes in Arabidopsis, rice, and maize, transcriptional profiling and qRT-PCR analysis revealed that CeLEA1 genes have evolved to predominantly express in tubers, exhibiting a seed desiccation-like accumulation during tuber development. Moreover, CeLEA1 transcripts in tubers were shown to be considerably more than that of their orthologs in purple nutsedge, another Cyperaceae plant producing desiccation-sensitive tubers. These results imply species-specific activation and key roles of CeLEA1 genes in the acquisition of desiccation tolerance of tigernut tubers as observed in orthodox seeds. Our findings not only improve the understanding of lineage-specific evolution of the LEA_1 family, but also provide valuable information for further functional analysis and genetic improvement in tigernut. Cissus quadrangularis is a succulent vine that degrades forests where it is not native by growing over trees and causing them to break or by impeding regeneration. Methods for its control have been tried but no satisfactory approach has been found yet. We carried out an experiment to analyze how much desiccation Cissus can endure before losing its ability to grow when rehydrated, using fragments of 0.5, 1, 2, and 3 internodes to test if desiccation tolerance was affected by fragment length. We found that Cissus remains viable after losing up to 80% of its weight, with shorter fragments losing viability (capacity to grow) at 70% weight loss. No fragments sustained viability at 90% water loss, establishing a critical threshold for Cissus desiccation tolerance. Our study also showed that shorter fragments (0.5 internodes) were less viable compared to longer ones (1, 2 or 3 internodes). Cissus has a remarkable tolerance to desiccation. Therefore, management strategies should ensure complete dehydration of Cissus fragments to prevent its regrowth. Reducing fragments to smaller sizes could amplify the effectiveness of control measures by reducing their viability, but risks of increasing propagule numbers should be considered. Discontinuous hydration and dehydration (HD) cycles refer to controlled imbibition followed by dehydration before seed germination. Here, we investigated whether the level of imbibition before HD cycles affects the physiology of Tabebuia heterophylla seeds and seedlings. Seeds were imbibed for 10 h (T1; phase I of imbibition) or 35 h (T2; phase II), dehydrated, and progressively rehydrated one to four times (HD cycles). Germination and biochemical parameters (membrane integrity; total soluble, reducing, and nonreducing (NRS) sugars; proteins, amino acids, proline, H2O2, catalase, ascorbate peroxidase, and glutathione reductase activity) were quantified at the last rehydration step of each cycle. Biometric and biochemical parameters (including pigments) were analysed in seedlings 60 days after germination. HD cycles at T1 led to reduced seed germination and greater plasma membrane damage, higher enzyme activity (catalase and glutathione reductase) and accumulation of NRS, total amino acids, and proline compared to the controls and T2 treatment. Cellular damage became more severe with more HD cycles. HD cycles at T2 synchronized germination regardless of the number of cycles and also had a priming effect. T2 seeds had less NRS, total amino acids, and proline content than T1. HD cycles at T1 produced seedlings with higher carotenoid and total chlorophyll content than controls and T2, while seedlings from HD cycles at T2 had higher amounts of osmoprotectants. HD cycles at T2 benefited seeds and seedlings more than at T1. This suggests that the physiological and biochemical effects of HD cycles in seeds modulate seedling plasticity, depending on water availability, potentially promoting increased tolerance to recurrent droughts that will be intensified with ongoing climate changes. Summary: Water scarcity, resulting from climate change, poses a significant threat to ecosystems. Syntrichia ruralis, a dryland desiccation‐tolerant moss, provides valuable insights into survival of water‐limited conditions.We sequenced the genome of S. ruralis, conducted transcriptomic analyses, and performed comparative genomic and transcriptomic analyses with existing genomes and transcriptomes, including with the close relative S. caninervis. We took a genetic approach to characterize the role of an S. ruralis transcription factor, identified in transcriptomic analyses, in Arabidopsis thaliana.The genome was assembled into 12 chromosomes encompassing 21 169 protein‐coding genes. Comparative analysis revealed copy number and transcript abundance differences in known desiccation‐associated gene families, and highlighted genome‐level variation among species that may reflect adaptation to different habitats. A significant number of abscisic acid (ABA)‐responsive genes were found to be negatively regulated by a MYB transcription factor (MYB55) that was upstream of the S. ruralis ortholog of ABA‐insensitive 3 (ABI3). We determined that this conserved MYB transcription factor, uncharacterized in Arabidopsis, acts as a negative regulator of an ABA‐dependent stress response in Arabidopsis.The new genomic resources from this emerging model moss offer novel insights into how plants regulate their responses to water deprivation. Water is essential to all life on earth. It is a major component that makes up living organisms and plays a vital role in multiple biological processes. It provides a medium for chemical and enzymatic reactions in the cell and is a major player in osmoregulation and the maintenance of cell turgidity. Despite this, many organisms, called anhydrobiotes, are capable of surviving under extremely dehydrated conditions. Less is known about how anhydrobiotes adapt and survive under desiccation stress. Studies have shown that morphological and physiological changes occur in anhydrobiotes in response to desiccation stress. Certain disaccharides and proteins, including heat shock proteins, intrinsically disordered proteins, and hydrophilins, play important roles in the desiccation tolerance of anhydrobiotes. In this review, we summarize the recent findings of desiccation tolerance in the budding yeast Saccharomyces cerevisiae. Aims and background: The resurrection plant Myrothamnus flabellifolia tolerates complete desiccation and is a great model for studying how plants cope with extreme drought. Root-associated microbes play a major role in stress tolerance and are an attractive target for enhancing drought tolerance in staple crops. However, how these dynamics play out under the most extreme water limitation remains underexplored. This study aimed to identify bacterial and fungal communities that tolerate extreme drought stress in the bulk soil, rhizosphere, and endosphere of M. flabellifolia. Methods: High-throughput amplicon sequencing was used to characterise the microbial communities associated with M. flabellifolia. Results: The bacterial phyla that were most abundant across all compartments were Acidobacteriota, Actinobacteriota, Chloroflexota, Planctomycetota, and Pseudomonadota, while the most abundant fungal phyla were Ascomycota and Basidiomycota. Although the bulk soil hosted multiple beneficial root-associated microbes, the rhizosphere compartment showed the highest functional diversity of bacteria and fungi. In contrast, the endosphere exhibited a low abundance and diversity of microbes. These findings share consistent with the theory that M. flabellifolia recruits soil microbes from the bulk to the rhizosphere and finally to the endosphere. It is possible that these microbes could promote drought tolerance in associated plant tissues. Conclusion: We find that compartments act as the major driver of microbial diversity, but the soil physicochemical factors also influence microbial composition. Main conclusion: The resurrection plant Boea hygrometrica selectively recruits and assembles drought-specific microbial communities across the plant–soil compartments, which may benefit plant growth and fitness under extreme drought conditions. Plant-associated microbes are essential for facilitating plant growth and fitness under drought stress. The resurrection plant Boea hygrometrica in natural habitats with seasonal rainfall can survive rapid desiccation, yet their interaction with microbiomes under drought conditions remains unexplored. This study examined the bacterial and fungal microbiome structure and drought response across plant–soil compartments of B. hygrometrica by high-throughput amplicon sequencing of 16S rRNA gene and internal transcribed spacer. Our results demonstrated that the diversity, composition, and functional profile of the microbial community varied considerably across the plant–soil compartments and were strongly affected by drought stress. Bacterial and fungal diversity was significantly reduced from soil to endosphere and belowground to aboveground compartments. The compartment-specific enrichment of the dominant bacteria phylum Cyanobacteriota and genus Methylorubrum in leaf endosphere, genera Pseudonocardia in rhizosphere soil and Actinoplanes in root endosphere, and fungal phylum Ascomycota in the aboveground compartments and genera Knufia in root endosphere and Cladosporium in leaf endosphere composed part of the core microbiota with corresponding enrichment of beneficial functions for plant growth and fitness. Moreover, the recruitment of dominant microbial genera Sphingosinicella and Plectosphaerella, Ceratobasidiaceae mycorrhizal fungi, and numerous plant growth-promoting bacteria involving nutrient supply and auxin regulation was observed in desiccated B. hygrometrica plants. Our results suggest that the stable assembled drought-specific microbial community of B. hygrometrica may contribute to plant survival under extreme environments and provide valuable microbial resources for the microbe-mediated drought tolerance enhancement in crops. Desiccation tolerance evolved recurrently across diverse plant lineages to enable survival in water-limited conditions. Many resurrection plants are polyploid, and several groups have hypothesized that polyploidy contributed to the evolution of desiccation tolerance. However, due to the vast phylogenetic distance between resurrection plant lineages, the rarity of desiccation tolerance, and the prevalence of polyploidy in plants, this hypothesis has been difficult to test. Here, we surveyed natural variation in morphological, reproductive, and desiccation tolerance traits across several cytotypes of a single species to test for links between polyploidy and increased resilience. We sampled multiple natural populations of the resurrection grass Microchloa caffra across an environmental gradient ranging from mesic to xeric in South Africa. We describe two distinct ecotypes of M. caffra that occupy different extremes of the environmental gradient and exhibit consistent differences in ploidy, morphological, reproductive, and desiccation tolerance traits in both field and common growth conditions. Interestingly, plants with more polyploid genomes exhibited consistently higher recovery from desiccation, were less reproductive, and were larger than plants with smaller genomes and lower ploidy. These data indicate that selective pressures in increasingly xeric sites may play a role in maintaining and increasing desiccation tolerance and are mediated by changes in ploidy. The Valdivian region has a temperate rainy climate with differences in rainfall throughout the year. This heterogeneity results in periods of summer drought that expose the poikilohydric epiphytes to desiccation. With this research, we aim to answer different research questions related to phorophyte preference, response to desiccation, and response to radiation. How does the diversity of macrolichens vary at a local and microclimate scale in three tree species within an evergreen forest? What is the tolerance limit of macrolichens against prolonged desiccation, according to evaluation of the maximum efficiency of PSII (Fv/Fm) and pigment concentration? What is the tolerance limit against a potential increase in radiation? We found that macrolichen communities are determined by tree species, which regulate the suitability of the substrate by modifying the temperature and humidity conditions. In addition, our results show a rapid photosynthetic alteration in temporal exposure to desiccation, measured through Fv/Fm and pigment concentration. Our results showed that the most sensitive lichens to radiation and desiccation are not coincident. We confirm the low tolerance of macrolichen species to high radiation, reflected in the saturation profile obtained for the set studied. ABSTRACT: Desiccation sensitivity is a condition present in seeds of several tree species of economic and ecological importance, which makes such seeds unable to tolerate drying and storage. Thus, studies that seek strategies to maintain the viability of such seeds after being subjected to drying are necessary. An alternative for this purpose is the use of priming in solutions of known osmotic potential. Solutions of polyethylene glycol (PEG), glycerol, as well as phytohormones such as abscisic acid (ABA) and salicylic acid (SA) are related to stress tolerance in seeds. The objective of this work was to study desiccation sensitive seeds of Inga vera, Eugenia uniflora and Tapirira guianensis, using solutions of PEG, SA, sucrose, glycerol and combinations of SA+PEG in an attempt of reducing seed desiccation sensitivity. The responses varied according to the species, but in general, successful results in maintaining the viability of the seeds after drying were obtained from the treatmen of seed with sucrose, glycerol and combinations of PEG and SA. Treatment of seeds with these compounds is promising in reducing the desiccation sensitivity of seeds of tree species.

To increase the resilience of forests to drought and other hazards, foresters are increasingly planting mixed stands. This requires knowledge about the drought response of tree species in pure and mixed-culture neighborhoods. In addition, drought frequently interacts with continued atmospheric nitrogen (N) deposition. To disentangle these factors for European beech, Norway spruce and Douglas fir, we conducted a replicated 3-factorial sapling growth experiment with three moisture levels, (high, medium, and low), two N levels (high and ambient), and pure and mixed-culture neighborhoods. We measured biomass, stomatal conductance (GS), shoot water potential (at predawn: ΨPD, midday, and turgor loss point: ΨTLP), branch xylem embolism resistance (Ψ50) and minimum epidermal conductance (G min). The three species differed most with respect to G min (10-fold higher in beech than in the conifers), hydroscape area (larger in beech), and the time elapsed to reach stomatal closure (TΨGS90) and ΨTLP (TTLP; shorter in beech), while Ψ50 and ΨTLP were remarkably similar. Neighborhood (pure vs mixed-culture) influenced biomass production, water status and hydraulic traits, notably G S (higher in Douglas fir, but lower in spruce and beech, in mixtures than pure culture), hydraulic safety margin (smaller for beech in mixtures), and TΨGS90 and TTLP (shorter for spruce in mixture). High N generally increased GS , but no consistent N effects on leaf water status and hydraulic traits were detected, suggesting that neighbor identity had a larger effect on plant water relations than N availability. We conclude that both tree neighborhood and N availability modulate the drought response of beech, spruce, and Douglas fir. Species mixing can alleviate the drought stress of some species, but often by disadvantaging other species. Thus, our study suggests that stabilizing and building resilience of production forests against a drier and warmer climate may depend primarily on the right species choice; species mixing can support the agenda. The extreme low humidity and temperatures in Antarctica make it one of the harsher areas for life on our planet. In a global change context, environmental barriers that prevented the arrival of alien species in Antarctica are weakening. Deception Island, one of the four active volcanoes of Antarctica, is especially vulnerable to the impacts of alien species. Geothermal areas (GA) in this Island offer unique microclimatic conditions that could differentially affect native and alien soil arthropods. Here we explore the desiccation tolerance of a native (Cryptopygus antarcticus) and an alien (Proisotoma minuta) springtail (Collembola) species to these extreme environmental conditions. GA and non‐geothermal areas (NGA) were selected to evaluate intra‐ and interspecific variation in desiccation tolerance. Populations of P. minuta from GA had greater desiccation tolerance than populations from NGA. However, desiccation tolerance of C. antarcticus did not differ between GA and NGA. This native species had greater desiccation tolerance than the alien P. minuta, but also greater body size. Our findings show that the alien P. minuta responds differently to environmental conditions than the native C. antarcticus. Furthermore, body size may influence desiccation tolerance in these two springtail species. Physiological maturity impacts seed quality through various mechanisms including vigor, desiccation tolerance, dormancy induction, synthesis of raw materials (including seed storage proteins), and the reorganization of metabolisms. Peanut seed development can be classified into seven classes with four incremental stages per class. Based on the mesocarp color, the final three stages are commonly referred to as "orange", "brown", and "black". In 2017, freshly harvested pods from one genotype of runner market-type peanuts grown under conventional practices were obtained from the University of Georgia research facility. The pods were removed from the plant material and 'pod blasted' to reveal the mesocarp. After separation, the remainder of the pod outer layer was removed, and the seeds were segregated for proteomic analysis. The raw peanuts were analyzed by bottom-up LC-MS/MS proteomics, which was conducted by the Proteomics Resource Center at the Rockefeller University, to identify the significant protein composition differences in each maturity class. The proteomic data revealed differentially expressed proteins as a function of maturity class with multiple functions including plant defense, metabolism, cell signaling, nutrient accumulation, and packaging. Understanding the processes needed for seed maturation will enable peanut scientists to evaluate the traits needed for robust germination, hardiness of the seed in response to disease, and nutrient quality. SUMMARY: The effects of four stressors, temperature, irradiance, desiccation and salinity, on the photochemical efficiency (ΔF/Fm′) of an edible green alga, Caulerpa chemnitzia var. laetevirens from Kagoshima, Japan, were determined for optimizing cultivation and shipping environments using a pulse amplitude modulation‐chlorophyll fluorometer. The ΔF/Fm′ remained stable at 24–34°C during the 3‐day temperature exposures ranging from 8 to 36°C; however, it dropped at higher and lower temperatures. During continuous exposure for 6 h to irradiance levels of 400 (low) and 1000 (high) μmol photons m−2 s−1 at 16, 24 and 32°C, the decrease in ΔF/Fm′ was pronounced at high irradiance. Moreover, ΔF/Fm′ also dropped at 16°C even under low irradiance, suggesting the occurrence of low temperature–light stress. Desiccation experiments under 50% humidity and up to 5 h of aerial exposure at 24°C and dim light (20 μmol photons m−2 s−1) indicated that ΔF/Fm′ was well tolerated within 1 h of desiccation; however, it dropped quickly as the desiccation period prolonged, suggesting that, if more than 20% of the interior water was lost, ΔF/Fm′ dropped progressively. However, during a similar experiment up to 14 days of aerial exposure under saturated humidity (99%) and dim light (12:12 h light/dark photocycle), as the interior water of the frond was maintained because of the saturated humidity, ΔF/Fm′ appeared to be well maintained for up to 5 days. Similar to desiccation, this alga exhibited stenohaline characteristics (30–40 psu) under the salinity gradient of 0–80 psu for up to 7 days of culture at 24°C and under dim light (12:12 h light/dark). In conclusion, this alga can be cultivated by modifying the techniques used for cultivating Japanese Caulerpa lentillifera and adjusting its environment accordingly. The incorporation of biological control agents (BCAs) such as Trichoderma spp. in agricultural systems favors the transition towards sustainable practices of plant nutrition and diseases control. Novel bioproducts for crop management are called to guarantee sustainable antagonism activity of BCAs and increase the acceptance of the farmers. The encapsulation in polymeric matrices play a prominent role for providing an effective carrier/protector and long-lasting bioproduct. This research aimed to study the influence of biopolymer in hydrogel capsules on survival and shelf-life of T. koningiopsis. Thus, two hydrogel capsules prototypes based on alginate (P1) and amidated pectin (P2), containing conidia of T. koningiopsis Th003 were formulated. Capsules were prepared by the ionic gelation method and calcium gluconate as crosslinker. Conidia releasing under different pH values of the medium, survival of conidia in drying capsules, storage stability, and biocontrol activity against rice sheath blight (Rhizoctonia solani) were studied. P2 prototype provided up to 98% survival to Th003 in fluid bed drying, faster conidia releasing at pH 5.8, storage stability greater than 6 months at 18 °C, and up to 67% of disease reduction. However, both biopolymers facilitate the antagonistic activity against R. solani, and therefore can be incorporated in novel hydrogel capsules-based biopreparations. This work incites to develop novel biopesticides-based formulations with potential to improve the delivery process in the target site and the protection of the active ingredient from the environmental factors. Desiccation tolerance has emerged as a potential strategy for crop improvement. However, the application of desiccation tolerance in crops, particularly rice, has been challenging and limited. Studies in angiosperms suggest that desiccation in vegetative tissues shares a common set of genes, developed convergently by modifying shared pathways associated with seed desiccation. The drought-tolerant Oryza sativa ssp. indica cv MR303 has demonstrated the capability to withstand desiccation state for a short period of time. Using this variety, we aim to investigate the factors that limit rice plants from achieving desiccation tolerance. The irreversibility of the cell wall emerges as a primary factor hindering rice plants from acquiring desiccation tolerance. Analysis of cell wall composition and transcriptomic data reveals an accumulation of secondary cell wall components and the expression of cell wall-related genes involved in secondary wall formation and modification, suggesting that these factors could be underlying causes for the irreversibility of the cell wall. Such factors contribute to the rigidity and stiffness of the cell wall, leading to its irreversibility during the rehydration process. This, in turn, impedes the recovery of rice after dehydration.

Abstract Background Drought is one of the main consequences of global climate change and this problem is expected to intensify in the future. Resurrection plants evolved the ability to withstand the negative impact of long periods of almost complete desiccation and to recover at rewatering. In this respect, many physiological, transcriptomic, proteomic and genomic investigations have been performed in recent years, however, few epigenetic control studies have been performed on these valuable desiccation-tolerant plants so far. Results In the present study, for the first time for resurrection plants we provide evidences about the differential chromatin accessibility of Haberlea rhodopensis during desiccation stress by ATAC-seq (Assay for Transposase Accessible Chromatin with high-throughput sequencing). Based on gene similarity between species, we used the available genome of the closely related resurrection plant Dorcoceras hygrometricum to identify approximately nine hundred transposase hypersensitive sites (THSs) in H. rhodopensis. The majority of them corresponds to proximal and distal regulatory elements of different genes involved in photosynthesis, carbon metabolism, synthesis of secondary metabolites, cell signalling and transcriptional regulation, cell growth, cell wall, stomata conditioning, chaperons, oxidative stress, autophagy and others. Various types of binding motifs recognized by several families of transcription factors have been enriched from the THSs found in different stages of drought. Further, we used the previously published RNA-seq data from H. rhodopensis to evaluate the expression of transcription factors putatively interacting with the enriched motifs, and the potential correlation between the identified THS and the expression of their corresponding genes. Conclusions These results provide a blueprint for investigating the epigenetic regulation of desiccation tolerance in resurrection plant H. rhodopensis and comparative genomics between resurrection and non-resurrection species with available genome information.

LEA_1 domain-containing proteins constitute a class of late-embryogenesis-abundant proteins that are highly hydrophilic and predominantly accumulate in mature seeds. Though LEA_1 proteins have been proven to be essential for seed desiccation tolerance and longevity, little information is available on their roles in non-seed storage organs. In this study, a first genome-wide characterization of the LEA_1 gene family was conducted in tigernut (Cyperus esculentus L., Cyperaceae), whose underground tubers are desiccation tolerant with a moisture content of less than 6%. Five family members identified in tigernut are comparative to four to six found in seven other Cyperaceae plants, but relatively more than three reported in Arabidopsis. Further comparison of 125 members from 29 plant species supports early divergence of the LEA_1 family into two phylogenetic groups before angiosperm radiation, and gene expansion in tigernut was contributed by whole-genome duplications occurring after the split with the eudicot clade. These two phylogenetic groups could be further divided into six orthogroups in the momocot clade, five of which are present in tigernut and the remaining one is Poaceae specific. Frequent structural variation and expression divergence of paralogs were also observed. Significantly, in contrast to seed-preferential expression of LEA_1 genes in Arabidopsis, rice, and maize, transcriptional profiling and qRT-PCR analysis revealed that CeLEA1 genes have evolved to predominantly express in tubers, exhibiting a seed desiccation-like accumulation during tuber development. Moreover, CeLEA1 transcripts in tubers were shown to be considerably more than that of their orthologs in purple nutsedge, another Cyperaceae plant producing desiccation-sensitive tubers. These results imply species-specific activation and key roles of CeLEA1 genes in the acquisition of desiccation tolerance of tigernut tubers as observed in orthodox seeds. Our findings not only improve the understanding of lineage-specific evolution of the LEA_1 family, but also provide valuable information for further functional analysis and genetic improvement in tigernut.

Observed climate change in northern high latitudes is strongest in winter, but still relatively little is known about the effects of winter climate change on tundra ecosystems. Ongoing changes in winter climate and snow cover will change the intensity, duration and frequency of frost events. Bryophytes form a major component of northern ecosystems but their responses to winter climate changes are largely unknown.Here, we studied how changes in overall winter climate and snow regime affect frost damage in three common bryophyte taxa that differ in desiccation tolerance in a subarctic tundra ecosystem. We used a snow manipulation experiment where bryophyte cores were transplanted from just above the tree line to similar elevation (i.e. current cold climate) and lower elevation (i.e. near‐future warmer climate scenario) in Abisko, Sweden. Here, we measured frost damage in shoots of Ptilidium ciliare, Hylocomium splendens and Sphagnum fuscum with the relative electrolyte leakage (REL) method, during late winter and spring in two consecutive years. We hypothesized that frost damage would be lower in a milder climate (low site) and higher under reduced snow cover and that taxa from moister habitats with assumed low desiccation tolerance would be more sensitive to lower temperature and thinner snow cover than those from drier and more exposed habitats.Contrary to our expectations, frost damage was highest at low elevation, while the effect of snow treatment differed across sites and taxa. At the high site, frost damage was reduced under snow addition in the taxon with the assumed lowest desiccation tolerance, S. fuscum. Surprisingly, frost damage increased with mean temperature in the bryophyte core of the preceding 14 days leading up to REL measurements and decreased with higher frost degree sums, that is, was highest in the milder climate at the low site.Synthesis Our results imply that climate warming in late winter and spring increases frost damage in bryophytes. Given the high abundance of bryophytes in tundra ecosystems, higher frost damage could alter the appearance and functioning of the tundra landscape, although the short and long‐term effects on bryophyte fitness remain to be studied. The outstanding desiccation tolerance of Cronobacter sakazakii (C. sakazakii) enables long-term persistence in food products with low-water activity to increase the infection risk, especially in low-birth-weight, immuno-compromised neonates, and infants less than 4 weeks of age. In our previous study, the disruption of glutathione transport-related gene gsiD by transposon was found to significantly increase its inactivation rate under drying stress challenges. However, the mechanism underlying the association between glutathione transport and desiccation tolerance of C. sakazakii remains to be clarified. In this study, the mechanism underlying their association was investigated in detail by constructing the gsiD gene deletion mutant. gsiD gene deletion was found to cause the dysfunction of the glutathione transport system GsiABCD and the limitation of glutathione import. The resulting decrease in intracellular glutathione caused the decreased potassium ions uptake and increased potassium ions efflux, inhibited the proline synthesis process, limited extracellular glutathione utilization, increased oxidant stress, reduced biofilm formation, and increased outer membrane permeability, which may be the main reasons for the significant reduction of the desiccation tolerance of C. sakazakii. To survive extreme drying (anhydrobiosis), many organisms, spanning every kingdom of life, accumulate intrinsically disordered proteins (IDPs). For decades, the ability of anhydrobiosis‐related IDPs to form transient amphipathic helices has been suggested to be important for promoting desiccation tolerance. However, evidence empirically supporting the necessity and/or sufficiency of helicity in mediating anhydrobiosis is lacking. Here, we demonstrate that the linker region of CAHS D, a desiccation‐related IDP from the tardigrade Hypsibius exemplaris, that contains significant helical structure, is the protective portion of this protein. Perturbing the sequence composition and grammar of the linker region of CAHS D, through the insertion of helix‐breaking prolines, modulating the identity of charged residues, or replacement of hydrophobic amino acids with serine or glycine residues results in variants with different degrees of helical structure. Importantly, correlation of protective capacity and helical content in variants generated through different helix perturbing modalities does not show as strong a trend, suggesting that while helicity is important, it is not the only property that makes a protein protective during desiccation. Tardigrada (water bears) are a group of small invertebrates known for their ability to survive extreme desiccation by entering an anhydrobiotic state. This state is characterized by the loss of ≤ 98% of body water, resulting in body shrinkage and almost complete cessation of metabolism. In anhydrobiosis, multiple mechanical and biochemical stress effects, such as protein denaturation and aggregation, DNA damage, formation of reactive oxygen species, membrane disintegration and degeneration of cellular compartments, are counteracted by various proteins. Although multiple studies of individual proteins and tardigrade proteomes have been conducted, our knowledge of tardigrade anhydrobiosis-related proteins has not yet been reviewed. To fill this gap, based on available papers, we present a review of all tardigrade proteins known to contribute to desiccation survival, including intrinsically disordered proteins, heat shock proteins and reactive oxygen species-scavenging enzymes. Water is essential to all life on earth. It is a major component that makes up living organisms and plays a vital role in multiple biological processes. It provides a medium for chemical and enzymatic reactions in the cell and is a major player in osmoregulation and the maintenance of cell turgidity. Despite this, many organisms, called anhydrobiotes, are capable of surviving under extremely dehydrated conditions. Less is known about how anhydrobiotes adapt and survive under desiccation stress. Studies have shown that morphological and physiological changes occur in anhydrobiotes in response to desiccation stress. Certain disaccharides and proteins, including heat shock proteins, intrinsically disordered proteins, and hydrophilins, play important roles in the desiccation tolerance of anhydrobiotes. In this review, we summarize the recent findings of desiccation tolerance in the budding yeast Saccharomyces cerevisiae. We also propose that the yeast under desiccation could be used as a model to study neurodegenerative disorders.

Water is essential for metabolism and all life processes. Despite this, many organisms distributed across the kingdoms of life survive near-complete desiccation or anhydrobiosis. Increased intracellular viscosity, leading to the formation of a vitrified state is necessary, but not sufficient, for survival while dry. What properties of a vitrified system make it desiccation-tolerant or -sensitive are unknown. We have analyzed 18 different in vitro vitrified systems, composed of one of three protective disaccharides (trehalose, sucrose, or maltose) and glycerol, quantifying their enzyme-protective capacity and their material properties in a dry state. Protection conferred by mixtures containing maltose correlates strongly with increased water content, increased glass-transition temperature, and reduced glass former fragility, while the protection of glasses formed with sucrose correlates with increased glass transition temperature and the protection conferred by trehalose glasses correlates with reduced glass former fragility. Thus, in vitro different vitrified sugars confer protection through distinct material properties. Next, we examined the material properties of a dry desiccation tolerant and intolerant life stage from three different organisms. The dried desiccation tolerant life stage of all organisms had an increased glass transition temperature and reduced glass former fragility relative to its dried desiccation intolerant life stage. These results suggest in nature organismal desiccation tolerance relies on a combination of various material properties. This study advances our understanding of how protective and non-protective glasses differ in terms of material properties that promote anhydrobiosis. This knowledge presents avenues to develop novel stabilization technologies for pharmaceuticals that currently rely on the cold-chain. Statement of significance: For the past three decades the anhydrobiosis field has lived with a paradox, while vitrification is necessary for survival in the dry state, it is not sufficient. Understanding what property(s) distinguishes a desiccation tolerant from an intolerant vitrified system and how anhydrobiotic organisms survive drying is one of the enduring mysteries of organismal physiology. Here we show in vitro the enzyme-protective capacity of different vitrifying sugars can be correlated with distinct material properties. However, in vivo, diverse desiccation tolerant organisms appear to combine these material properties to promote their survival in a dry state.

Desiccation is a severe survival problem for organisms. We have been studying the desiccation tolerance mechanisms in the true slime mold Physarum polycephalum. We measured the trehalose content of P. polycephalum vegetative cells (plasmodia) and drought cells (sclerotia). Surprisingly, we found that the content in sclerotia was about 473-fold greater than in the plasmodia. We then examined trehalose metabolism-related genes via RNAseq, and consequently found that trehalose 6-phosphate phosphorylase (T6pp) expression levels increased following desiccation. Next, we cloned and expressed the genes for T6pp, trehalose 6-phosphate synthase/phosphatase (Tps/Tpp), maltooligosyltrehalose trehalohydrolase (TreZ), and maltooligosyltrehalose synthase (TreY) in E. coli. Incidentally, TreY and TreZ clones have been reported in several prokaryotes, but not in eukaryotes. This report in P. polycephalum is the first evidence of their presence in a eukaryote species. Recombinant T6pp, TreY, and TreZ were purified and confirmed to be active. Our results showed that these enzymes catalyze reactions related to trehalose production, and their reaction kinetics follow the Michaelis-Menten equation. The t6pp mRNA levels of the sclerotia were about 15-fold higher than in the plasmodia. In contrast, the expression levels of TreZ and TreY showed no significant change between the sclerotia and plasmodia. Thus, T6pp is probably related to desiccation tolerance, whereas the contribution of TreY and TreZ is insufficient to account for the considerable accumulation of trehalose in sclerotia. • Study for the desiccation tolerance mechanisms in a true slime mold Physarum polycephalum. • A large amount of trehalose was accumulated in the drought cells (sclerotia). • The genes (t6pp , tps/tpp , treZ , treY) related to trehalose metabolism were cloned. • T6pp is probably related to desiccation tolerance. • This is the first report of treY and treZ genes from eukaryotic species. Antarctic macroalgal communities of the upper intertidal zone are particularly poor compared with other coastal regions. Exposure to desiccation and freezing combined with the abrasive effect of ice threatens the life of sessile organisms. One of the few species able to colonize this environment is the rhodophyte Pyropia endiviifolia. It belongs to the Bangiales, one of the oldest extant clades of photosynthetic eukaryotes, which has occurred for more than one billion years with basically the same morphological structure. Considering that the extent of Antarctic glaciation is a geologically recent event, we hypothesized that pre-adaptations to desiccation in bangiophytes may have contributed to the success of P. endiviifolia in Antarctica. To test this, we compared its photosynthetic performance and tolerance to desiccation and freezing with those from a related intertidal species, the temperate Atlantic species Porphyra linearis. As evidenced by gas exchange and chlorophyll fluorescence, P. endiviifolia is more adapted to high irradiances than P. linearis. The former species was also more desiccation-tolerant, and showed a higher glass transition temperature. Both species did not differ in chlorophyll content per dry weight, and tolerance to −20°C, even though the ice-nucleation temperature was much higher in P. endiviifolia. Membrane integrity may depend on fatty acid composition in P. endiviifolia, while on enhanced tocopherol in P. linearis. Overall, both species show different strategies to deal with freezing temperatures: supercooling in P. linearis vs. freezing-tolerance in P. endiviifolia. This matches with the probability of being subjected to sub-zero temperatures in their natural environments (lower in the case of P. linearis). Surprisingly, the higher risk of dehydration in the natural habitat of P. linearis is not matched by a higher desiccation tolerance. This observation does not support the initial hypothesis of the study but suggests the opposite: the acclimation to a cold environment results in higher desiccation tolerance. For the present study, we collected the Ulvophyceae species Trentepohlia aurea from limestone rock near Berchtesgaden, Germany, and the closely related taxa T. umbrina from Tilia cordata tree bark and T. jolithus from concrete wall both in Rostock, Germany. Freshly sampled material stained with Auramine O, DIOC6, and FM 1–43 showed an intact physiological status. Cell walls were depicted with calcofluor white and Carbotrace. When subjected to three repeated and controlled cycles of desiccation over silica gel (~ 10% relative humidity) followed by rehydration, T. aurea recovered about 50% of the initial photosynthetic yield of photosystem II (YII). In contrast, T. umbrina and T. jolithus recovered to 100% of the initial YII. HPLC and GC analysis of compatible solutes found highest proportions of erythritol in T. umbrina and mannitol/arabitol in T. jolithus. The lowest total compatible solute concentrations were detected in T. aurea, while the C/N ratio was highest in this species, indicative of nitrogen limitation. The prominent orange to red coloration of all Trentepohlia was due to extremely high carotenoid to Chl a ratio (15.9 in T. jolithus, 7.8 in T. aurea, and 6.6. in T. umbrina). Photosynthetic oxygen production was positive up to ~ 1500 µmol photons m−2 s−1 with the highest Pmax and alpha values in T. aurea. All strains showed a broad temperature tolerance with optima for gross photosynthesis between 20 and 35 °C. The presented data suggest that all investigated Trentepohlia species are well adapted to their terrestrial lifestyle on exposed to sunlight on a vertical substrate with little water holding capacity. Nevertheless, the three Trentepohlia species differed concerning their desiccation tolerance and compatible solute concentrations. Several plant growth–promoting bacteria (PGPB) are gram-negative, and their cell viability is affected during the bio-inoculant production. Hence, formulation-drying processes provide challenges that limit the adoption of these beneficial microorganisms in sustainable agricultural production. Among delivery system strategies for gram-negative PGPB, the encapsulating cells in biopolymeric materials are emerging as a promising alternative. This research aims to evaluate the effect of additives and crosslinking agents on the survival of the consortium of Herbaspirillum frisingense AP21, Azospirillum brasilense D7, and Rhizobium leguminosarum T88 in hydrogel capsules. Three crosslinkers and diverse potential drying protectors were tested. Calcium gluconate provides notable consortium survival advantages regarding colony-forming units (CFUs) (losses of up to 4 log CFU) compared to calcium lactate and calcium chloride (up to 6 log CFU). Additives such as skimmed milk, whey protein, and Gelita® EC improve the recovery of viable cells after the drying process, demonstrating an increase in cell survival of the three bacteria by up to 4 log CFU. The combination of these substances into a capsule prototype extends the storage stability of bacterial consortium up to 3 months at 18 ± 2 °C. This study expands the knowledge for formulating gram-negative PGPB consortium, regarding the crosslinker and drying protector relationship on encapsulation processes with drying survival and further storage stability performance. The Valdivian region has a temperate rainy climate with differences in rainfall throughout the year. This heterogeneity results in periods of summer drought that expose the poikilohydric epiphytes to desiccation. With this research, we aim to answer different research questions related to phorophyte preference, response to desiccation, and response to radiation. How does the diversity of macrolichens vary at a local and microclimate scale in three tree species within an evergreen forest? What is the tolerance limit of macrolichens against prolonged desiccation, according to evaluation of the maximum efficiency of PSII (Fv/Fm) and pigment concentration? What is the tolerance limit against a potential increase in radiation? We found that macrolichen communities are determined by tree species, which regulate the suitability of the substrate by modifying the temperature and humidity conditions. In addition, our results show a rapid photosynthetic alteration in temporal exposure to desiccation, measured through Fv/Fm and pigment concentration. Our results showed that the most sensitive lichens to radiation and desiccation are not coincident. We confirm the low tolerance of macrolichen species to high radiation, reflected in the saturation profile obtained for the set studied. The lichen community in the evergreen forest showed high complexity and vulnerability, pointing to the importance of more research.

Physiological maturity impacts seed quality through various mechanisms including vigor, desiccation tolerance, dormancy induction, synthesis of raw materials (including seed storage proteins), and the reorganization of metabolisms. Peanut seed development can be classified into seven classes with four incremental stages per class. Based on the mesocarp color, the final three stages are commonly referred to as “orange”, “brown”, and “black”. In 2017, freshly harvested pods from one genotype of runner market-type peanuts grown under conventional practices were obtained from the University of Georgia research facility. The pods were removed from the plant material and ‘pod blasted’ to reveal the mesocarp. After separation, the remainder of the pod outer layer was removed, and the seeds were segregated for proteomic analysis. The raw peanuts were analyzed by bottom-up LC-MS/MS proteomics, which was conducted by the Proteomics Resource Center at the Rockefeller University, to identify the significant protein composition differences in each maturity class. The proteomic data revealed differentially expressed proteins as a function of maturity class with multiple functions including plant defense, metabolism, cell signaling, nutrient accumulation, and packaging. Understanding the processes needed for seed maturation will enable peanut scientists to evaluate the traits needed for robust germination, hardiness of the seed in response to disease, and nutrient quality.

Bryophytes play a crucial role in the ecosystem's water compartment due to their unique ability to retain water. However, their role within temperate native ecosystems is mostly unknown. To address this knowledge gap, a study was conducted on Terceira Island (Azores), focusing on 14 bryophyte species found at different altitudes (40 m, 683 m, and 1012 m); five samples were collected monthly, per species and location, and their fresh, saturated, and dry weights were examined in the laboratory; four species were collected from more than one site. Generalized linear models (GLM) were used to assert the influence of climate factors (temperature, precipitation, and relative humidity) and environmental variables on two water indicators: field water content (FWC) and relative water content (RWC). None of the examined factors, per se, were able to explain all cases. Species appear to respond to climate according to a limiting factor effect: at lower elevations, precipitation was determinant, while at medium elevations, FWC was influenced by a combination of precipitation and relative humidity. At higher elevations, temperature was retained for seven of the nine studied species. The RWC values indicated that the 14 bryophyte species remained hydrated throughout the year but rarely reached their maximum water-holding capacity, even at the highest altitude. Understanding the mechanisms by which native bryophytes acquire, store, and release water is crucial for comprehending the resilience of native vegetation in the face of climate change. Beauveria bassiana is a cosmopolitan entomopathogenic fungus that can infect over 1000 insect species. During growth inside the host, B. bassiana transitions from hyphal to yeast-like unicellular growth as blastospores. Blastospores are well suited as an active ingredient in biopesticides due to their ease of production by liquid fermentation. Herein, we investigated the impact of hyperosmotic growth environments mediated by ionic and non-ionic osmolytes on two strains of B. bassiana (ESALQ1432 and GHA) relevant to growth morphology, blastospore production, desiccation tolerance, and insecticidal activity. Polyethylene glycol (PEG200) increased osmotic pressure in submerged cultures leading to decreased blastospore size but higher blastospore yields for one strain. Morphologically, decreased blastospore size was linked to increased osmotic pressure. However, smaller blastospores from PEG200 supplemented cultures after air-drying exhibited delayed germination. Ionic osmolytes (NaCl and KCl) generated the same osmotic pressure (2.5–2.7 MPa) as 20% glucose and boosted blastospore yields (> 2.0 × 109 blastospores mL−1). Fermentation performed in a bench-scale bioreactor consistently promoted high blastospore yields when using NaCl (2.5 MPa) amended media within 3 days. Mealworm larvae (Tenebrio molitor) were similarly susceptible to NaCl-grown blastospores and aerial conidia in a dose-time-dependent manner. Collectively, these results demonstrate the use of hyperosmotic liquid culture media in triggering enhanced yeast-like growth by B. bassiana. Understanding the role of osmotic pressure on blastospore formation and fitness will hasten the development of viable commercial fungal biopesticides. The family Termitidae is renowned for its diverse nesting behaviors, with the evolution of epigeal and arboreal nests hypothesized to increase desiccation stress due to greater exposure to air. However, these nests may also alleviate desiccation stress through humidity regulation. To explore the implications of acquiring epigeal and arboreal nests, we investigated desiccation tolerance traits in 16 Termitidae termite species with varying nest types and analyzed trait correlations. Principal component analysis revealed that termites constructing epigeal and arboreal nests exhibited reduced water loss rates and enhanced survival under desiccated conditions. Furthermore, termites building arboreal nests displayed a notably higher water content. Redundancy analysis demonstrated that nest types accounted for a substantial portion (57.2%) of the observed variation in desiccation tolerance. These findings support the hypothesis that epigeal and arboreal nests in termites are associated with increased desiccation stress and increased desiccation tolerance. The cosolvent 2,2,2‐trifluoroethanol (TFE) is often used to mimic protein desiccation. We assessed the effects of TFE on cytosolic abundant heat soluble protein D (CAHS D) from tardigrades. CAHS D is a member of a unique protein class that is necessary and sufficient for tardigrades to survive desiccation. We find that the response of CAHS D to TFE depends on the concentration of both species. Dilute CAHS D remains soluble and, like most proteins exposed to TFE, gains α‐helix. More concentrated solutions of CAHS D in TFE accumulate β‐sheet, driving both gel formation and aggregation. At even higher TFE and CAHS D concentrations, samples phase separate without aggregation or increases in helix. Trees of forest importance are a group of special interest for conservation actions due to their use value. To determine whether they can be conserved in seed banks, we studied the tolerance to desiccation and storage behaviour of four species from north-eastern Argentina: Handroanthus impetiginosus, Jacaranda mimosifolia, Neltuma alba y Pterogyne nitens. Desiccation tolerance was evaluated by decreasing the moisture content of the seeds to 4-5%. Storage behaviour was determined by storing dried seeds at -20, 5 and 20 °C for three, six and twelve months. The potential longevity of the seeds was estimated with an accelerated aging experiment. Response variable in all cases was the percentage of germinated seeds. The results indicated that the species have orthodox behaviour and can therefore be stored in seed banks. Throughout twelve months of storage the N. alba y P. nitens seeds remained dormant and had to be scarified to germinate. The potential longevity of the seeds allowed the species to be classified as short-lived (H. impetiginosus), intermediate-lived (J. mimosifolia y P. nitens), and long-lived (N. alba). The results will be useful to establish ex situ conservation strategies for these species, which should be complemented with in situ conservation actions. [ABSTRACT FROM AUTHOR]

Summary: The entomopathogenic nematode (EPN), Heterorhabditis bacteriophora , is a biocontrol agent with almost world-wide distribution, thus providing a huge source of biodiversity available for exploitation in breeding programmes to improve beneficial traits. For management of economically important insect pests, the nematode is mass-produced in liquid culture, formulated with inert carriers, and transported under controlled temperature conditions to maintain its quality. The biocontrol potential of nematode materials is defined by their genetic background. Previous reports have assessed the phenotypic plasticity of H. bacteriophora. Major beneficial traits are field efficacy influenced by their virulence, longevity and field persistence and tolerance to abiotic stresses like oxidative stress, desiccation and extreme low and high temperatures. For several of these traits high heritability values (h 2) have been reported, providing a platform for trait improvement by selection. Modern breeding uses molecular markers to track beneficial traits. Recent studies reported hundreds of single nucleotide polymorphisms (SNPs) in H. bacteriophora. This investigation revived a large set of cryopreserved wild types, hybrids and phenotyped materials of H. bacteriophora. Additional phenotypic data on the virulence (LC50) of H. bacteriophora strains against mealworm (Tenebrio molitor) were produced ranging from 1.4 ± 0.3 to 8.0 ± 3.6 nematodes per insect. Markers linked to beneficial traits were used for a comprehensive genotype analysis transferring reported SNP sequence information into the multiplex platform SeqSNP. Specific alleles associated to beneficial traits were identified and 65 materials were classified for the presence of relevant SNPs associated to beneficial traits. Traits in species that influence invasion success may vary in populations across its invaded range. The aquatic New Zealand mud snail, Potamopyrgus antipodarum, reproduces parthenogenetically in its invaded range, and a few distinct clonal genotypes have been identified in North America. Much of the spread of the snail in North America has been the result of unintentional overland transport by recreational water users. Thus, desiccation tolerance may play an important role in the invasion success of this species. The primary goal of these experiments is to determine if variation in desiccation tolerance exists between populations of this species. Here we compared multiple multi-locus genotypes (MLGs) and populations within those genotypes with regard to their desiccation tolerance. We conducted three experiments. The first compared the survival rate over time of snails from three populations and two MLGs with regard to their ability to survive being completely removed from water. The second experiment examined different size classes of snails from the same population, and the third experiment compared four different populations and two MLGs genotypes with respect to their survival rate over time when removed from water but being kept in moist conditions. We found larger snails tolerate desiccation longer than smaller snails with snails between 4 and 4.6 mm surviving at a rate of more than 50% after 12 h while smaller snails survived at a less than 5% rate after 12 h. We also found significant variation both between and within MLGs in their survival rate when completely removed from water and dried with the MLG from the western US having a more than 50% greater survival probability than the eastern MLGs at both 18 and 24 h out of water. When removed from water and kept moist all MLGs had a near 100% survival rate at 60 days at 7 °C, and most survived at a greater than 90% rate at 60 days at 17 °C, while no MLG's survived past 30 days at 27 °C. The biological risks of the deep space environment must be elucidated to enable a new era of human exploration and scientific discovery beyond low earth orbit (LEO). There is a paucity of deep space biological missions that will inform us of the deleterious biological effects of prolonged exposure to the deep space environment. To safely undertake long-term missions to Mars and space habitation beyond LEO, we must first prove and optimize autonomous biosensors to query the deep space radiation environment. Such biosensors must contain organisms that can survive for extended periods with minimal life support technology and must function reliably with intermittent communication with Earth. NASA's BioSentinel mission, a nanosatellite containing the budding yeast Saccharomyces cerevisiae, is such a biosensor and one of the first biological missions beyond LEO in nearly half a century. It will help fill critical gaps in knowledge about the effects of uniquely composed, chronic, low-flux deep space radiation on biological systems and in particular will provide valuable insight into the DNA damage response to highly ionizing particles. Due to yeast's robustness and desiccation tolerance, it can survive for periods analogous to that of a human Mars mission. In this study, we discuss our optimization of conditions for long-term reagent storage and yeast survival under desiccation in preparation for the BioSentinel mission. We show that long-term yeast cell viability is maximized when cells are air-dried in trehalose solution and stored in a low-relative humidity and low-temperature environment and that dried yeast is sensitive to low doses of deep space-relevant ionizing radiation under these conditions. Water is critical for the survival of most cells and organisms. Remarkably, a small number of multicellular animals are able to survive nearly complete drying. The phenomenon of anhydrobiosis, or life without water, has been of interest to researchers for over 300 years. In this review we discuss advances in our understanding of protectants and mechanisms of desiccation tolerance that have emerged from research in three anhydrobiotic invertebrates: brine shrimp (Artemia), roundworms (nematodes), and tardigrades (water bears). Discovery of molecular protectants that allow each of these three animals to survive drying diversifies our understanding of desiccation tolerance, and convergent themes suggest mechanisms that may offer a general model for engineering desiccation tolerance in other contexts.

Haematoccocus lacustris (previously H. pluvialis) is a desiccation-tolerant unicellular freshwater green alga. During acclimation to desiccation, astaxanthin-rich lipid bodies and low-molecular-weight antioxidants (α-tocopherol, glutathione) accumulate, while the chloroplast area and chlorophyll contents decrease, which may facilitate desiccation tolerance by preventing damage mediated by reactive oxygen species (ROS). Here, we investigated the influence of moisture, light, oxygen, and temperature on redox homeostasis and cell longevity. Respiration and unbound freezable water were detectable in cells equilibrated to ≥90% relative humidity (RH), a threshold above which viability considerably shortened. At 92.5% RH and 21 °C, antioxidants depleted over days as cells lost viability, especially in an oxygen-rich atmosphere, supporting the role of ROS production in uncoupled respiration in viability loss. At 80% RH and 21 °C, redox homeostasis was maintained over weeks, and longevity was less influenced by oxygen. Light and oxygen was a lethal combination at 92.5% RH, under which pigments bleached, while in the dark only astaxanthin bleached. Viability positively correlated with glutathione concentrations across all treatments, while correlation with α-tocopherol was weaker, indicating limited viability loss from lipid peroxidation at 80% RH. In cells equilibrated to 50% RH, longevity and redox homeostasis showed strong temperature dependency, and viability was maintained at sub-zero temperatures for up to three years, revealing cryogenic storage to be an optimal strategy to store H. lacustris germplasm.
