Background: Deletions in the β-globin cluster are uncommon and cause thalassemia (thal) with hereditary persistence of fetal hemoglobin. They constitute a heterogenous group of disorders characterized by absent or reduced synthesis of adult hemoglobin (Hb A) and increased synthesis of fetal hemoglobin (Hb F). Although the clinical severity of these disorders are asymptomatic owing to the increased Hb F levels, the molecular basis is very heterogenous due to the large deletions in the β-globin cluster spanning both HBD and HBB genes. Here, we describe a Tunisian family carrying a novel deletion mutation causing (δβ)°-thalassemia. Methods: The amounts of hemoglobin fractions were measured by capillary electrophoresis of hemoglobin. Amplification and sequencing of different regions on the β-gene cluster were performed by Sanger method. Results: Family study and genetic analysis revealed a large deletion mutation in the β-globin cluster of 14.5 kb (NG_000,007.3:g. 58,253 to g.72837del14584) at the homozygous state in the patient and at heterozygous state at the other members of the family. This deletion removes the HBD and HBB genes. Conclusions: In our knowledge, this new large deletion is described for the first time in the Tunisian population and in the world, designed Tunisian(δβ)0 in Ithanet database (IthaID: 3971). Therefore, it is important to identify the deletion leading to δβ-thalassemia carriers at the molecular level, to highlight the importance of recognizing the clinical features and implementing appropriate testing to clarify the diagnosis and manage the condition. Background: Several mutations including the single nucleotide polymorphisms (SNPs) in the GSTP1 and deletional mutations of GSTM1 and GSTT1 isoforms of the glutathione S transferase gene are of significant importance in the etiology and severity of prostate cancer (Pca) with varying degrees of association in different population groups. We investigated the association of these mutations with the prevalence and severity of the Pca in an urban population of Eastern India. Materials and Methods: In the present case–control study, deletional mutations of the GSTM1 and GSTT1 were detected by their presence or absence in the gel electrophoresis after the polymerase chain reaction (PCR). The A to G SNP of the GSTP1 gene resulting in Ile105Val was detected using Restriction Fragment Length Polymorphism after PCR. Severity of Pca was assessed by International Society of Urological Pathology (ISUP) grading. Polymorphic variations obtained were correlated to the prevalence and severity of Pca. Results: The Chi-square tests showed a significantly greater prevalence of all three mutations in the case group. However, the post hoc ANOVA and multiple logistic regression analysis revealed that only the A to G SNP of the GSP1 gene among the three mutations showed significant association with the severity of the Pca as indicated by the ISUP gradings. Conclusion: The results indicated that although the A to G transitional SNP of the GSTP1 and the deletional SNPs of GSTM1 and GSTT1 acted all as important factors for increasing the prevalence of Pca, the A to G SNP of the GSTP1 gene resulting in Ile105Val only contributed to the severity and spread of the disease in our study population. AbstractWe report the haematological parameters and molecular characterization of beta zero (β°) South East Asia (SEA) deletion in the HBB gene cluster with unusually high levels of Hb F compared to a classical heterozygous beta zero (β°)-thalassaemia.MethodsRetrospective study on 17 cases of (β°) South East Asia (SEA) deletion from 2016 to 2019 referred to Institute for Medical Research were conducted. The clinical information and haematological profiles were evaluated. The mutation was analyzed, and the results were compared with other β°-thalassaemia groups. For HBB gene genotyping, all the cases were subjected for multiplex gap-PCR, 5 cases were subjected for HBB gene sequencing for exclusion of compound heterozygous with other beta variants. Co-inheritance of α-thalassaemia were determined using multiplex gap-PCR and multiplex ARMS-PCR.ResultsSeventeen cases were positive for β°-thal SEA deletion. The medians for the MCV and MCH of β°-thal SEA deletion were significantly higher than for all heterozygote β°-thalassaemia traits (Mann Whitney test, p  < 0.05). Patients with β°-thal SEA deletion had elevated levels of Hb A2 consistent with β-thalassaemia traits, with Hb F levels consistent with HPFH or δβ-thalassaemia carriers. The median for Hb A2 was 4.00 + 1.00%, similar to that observed in other β°-thalassaemia groups except for IVS 1-1 mutation (median 5.30 + 0.45%) and β°-Filipino (∼45 kb deletion) deletion (median 6.00 + 0.58). Interestingly, we found that Hb F levels for β°-thal SEA deletion were statistically higher than other β°-thalassaemia mutations (median 19.00 + 5.50%, p  < 0.05), except for the β°-thal 3.5 kb deletion group., Conclusion: We conclude that β°-thal SEA deletion has a unique haematological parameters of beta zero thalassaemia trait.
