Abstract

Spherical structures built from uni- and multilamellar lipid bilayers (LUV and MLV) are nowadays considered not just as nanocarriers of various kinds of therapeutics, but also as the vehicles that, when coupled with gold (Au) nanoparticles (NPs), can also serve as a tool for imaging and discriminating healthy and diseased tissues. Since the presence of Au NPs or their aggregates may affect the properties of the drug delivery vehicle, we investigated how the shape and position of Au NP aggregates adsorbed on the surface of MLV affect the arrangement and conformation of lipid molecules. By preparing MLVs constituted from 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) in the presence of uncoated Au NP aggregates found i) both within liposome core and on the surface of the outer lipid bilayer, or ii) adsorbed on the outer lipid bilayer surface only, we demonstrated the maintenance of lipid bilayer integrity by microscopic techniques (cryo-TEM, and AFM). The employment of SERS and FTIR-ATR techniques enabled us not only to elucidate the lipid interaction pattern and their orientation in regards to Au NP aggregates but also unequivocally confirmed the impact of Au NP aggregates on the persistence/breaking of van der Waals interactions between hydrocarbon chains of DPPC. Abstract

Deoxyribonucleic acid (DNA) detection is essential for the accurate and early diagnosis of a disease. In this study, a femtomolar DNA detection method based on the exploitation of the localized surface plasmon (LSP) resonance of gold nanoparticles (AuNPs) was developed. We prepared Poly Ethylen Glycol (PEG) functionalized AuNPs with a specific DNA capture probe (CP) directly modified on the gold surface. Two strategies are proposed using different kinds of CP to detect the target DNA (tDNA). In the first strategy, CP is the complementary of the complete sequence of the DNA (CCP method). For the second strategy, we used two CPs, which were half complementary to tDNA, and these were hybridized with tDNA to form sandwich structures (MIX method). The results showed that our detection methods are highly sensitive and that the limits of detection of 124 am and 2.54 fm tDNA can be reached when using the CCP and MIX methods, respectively. In addition, the specificity of our two strategies is also demonstrated with mismatched DNAs. The proposed method provides a simple, fast, sensitive and specific DNA biosensor, which has the potential to be used for point-of-care tests (POCT).

Abstract

Raman spectroscopy has become a popular tool for analyzing biological samples such as DNA. It allows us to access to the vibrational levels of molecules and thus to identify the chemical composition and to observe the structure of molecular systems.[1] In this work, we recorded the SERS spectra of DNA strands with different sequences and grafted at the surface of gold nanoparticles. Our objective is to study the correlation between the DNA sequences and its base composition and the intensity of Raman bands observed in SERS. To reach a high density of DNA on the surface of the gold nanoparticles, the DNA strands include a thiol group (SH) at their 5' end and are conjugated to gold nanoparticles using the freeze-thaw cycle method. SERS measurements are performed on dried drops deposited on a glass slide. We recorded 5 spectra for each sample using a 785nm excitation wavelength. We observed several bands that can be assigned to the different bases and to the phosphate backbone. For instance, the bands of ring breathing mode of adenine and the carbonic skeleton of the DNA are observable at 733cm-1 and 1029cm-1 respectively.[2] We then compare the relative intensity of the different bands such as the 733cm-1 or 1029cm-1 ones. We found that the relative intensity of the 733cm-1 band is correlated with the amount of adenine in the DNA sequence. This study provides a new approach for reliable quantification and analysis of genetic information associated with DNA molecule sequencing.

Abstract

Among various cavitand molecules, cyclodextrins are extensively studied due to their ability to form host-guest complexes with small hydrophobic molecules. Aiming to explore cyclodextrin implementation on the scopes related to the environmental pollution monitoring or remediation, extensive studies for understanding the cyclodextrin-based host-guest complex formation with selected targeted substances are conducted. In this context, two polycyclic aromatic hydrocarbons, naphthalene and fluoranthene as well as toluene as a member of volatile organic compounds, were studied regarding their ability to encapsulate into cyclodextrin cavities. Synthesised complexes were examined by thermogravimetric analysis combined with Raman spectroscopy. The obtained results demonstrated that the size between targeted molecules and the cyclodextrin cavities strongly correlates with its ability to engage in complexation. Thus, this latter parameter plays an important role in the inclusion complex formation as well as in the strength of the interaction between the molecules.

International audience; Optimum amplification in surface enhanced Raman scattering (SERS) from individual nanoantennas is expected when the excitation is slightly blue-shifted with respect to the localized surface plasmon resonance (LSPR), so that the LSPR peak falls in the middle between the laser and the Stokes Raman emission. Recent experiments have shown when moving the excitation from the visible to the near-infrared that this rule of thumb is no more valid. The excitation has to be red-shifted with respect to the LSPR peak, up to 80 nm, to obtain highest SERS. Such discrepancy is usually attributed to a near-field (NF) to far-field (FF) spectral shift. Here we critically discuss this hypothesis for the case of gold nanocylinders. By combining multiwavelength-excitation SERS experiments with numerical calculations, we show that the red-shift of the excitation energy does not originate from a spectral shift between the extinction (FF) and the near-field distribution (NF), which is found to be not larger than 10 nm. Rather, it can be accounted for by looking at the peculiar spectral dependence of the near-field intensity on the cylinders diameter, characterized by an initial increase, up to 180 nm diameter, followed by a decrease and a pronounced skewness.
