Bone tissue engineering (BTE) is a biomedical area that develops scaffolds capable of mimicking and repairing damage on bone tissue. For this, a popular and nonexpensive 3D printing technique named fused deposition modeling (FDM) has been used. The potential use of 3D scaffolds of poly(butylene adipate-co-terephthalate (PBAT) has been investigated in BTE since this polymer presents good biocompatibility and degradability, as well as mechanical properties like those offered by the natural bone. A single material does not have the features to promote cell adhesion, proliferation, and differentiation; the incorporation of bioactive substances can overcome this issue. This work aimed to develop 3D printed PBAT scaffolds by FDM technique and coated them with hydroxyapatite (H), bioglass (B), and gelatin (G) by solution immersion technique to obtain a functional biomaterial to be applied on the BTE. Structural characteristics and morphological and mechanical properties of the 3D scaffolds were evaluated. The cell proliferation and mineralization of pre-osteoblastic cells (MC3T3-E1) were accessed by methylthiazolyldiphenyl-tetrazolium bromide, sulphorodhamine B, and Alizarin red assay, respectively. 3D printed PBAT scaffolds were successfully obtained by FDM, and the surface modification of 3D PBAT was proven through the changes observed in structural and morphological characteristics. In addition, the mechanical properties were improved in the modified scaffolds. Also, the coated 3D PBAT scaffolds with any bioactive substance increased and promoted cell proliferation and pre-osteoblastic differentiation. Therefore, the combination of 3D PBAT scaffolds and H, G, and/or B (doped with niobium) is an alternative to produce functional biomaterials for BTE. [ABSTRACT FROM AUTHOR]

This study aimed to investigate if apical periodontitis in different periods changes systemic levels of the antioxidant and pro-oxidant parameters in Wistar rats. Twenty-four rats were randomly allocated into healthy animals, apical periodontitis at 14 days (AP14) and apical periodontitis at 28 days (AP28). The first mandibular molars were accessed in the AP groups, and the pulp chamber was exposed to the oral environment, inducing the apical lesion. After 14 and 28 days, the animals were anesthetized, euthanized, and hemimandibles were collected formicro-computed tomography (micro-CT) analysis tomeasure lesion volume, bone volume (BV), percent of bone to total tissue volume (BV/TV), trabecular thickness (Tb.Th), trabecular number (Tb.N), and trabecular space (Tb.Sp). A histological examination of the remaining bone was also performed. Finally, blood samples were collected for oxidative biochemistry analysis, investigating glutathione (GSH), Trolox equivalent antioxidant capacity (TEAC), and lipid peroxidation (TBARS). The lesion volume was greater at 28 than at 14 days, as shown by micro-CT. AP14 and AP28 had decreased BV and Tb.Th, but only AP28 showed a reduction in BV/TV. Tb.N and Tb. Sp were increased in apical periodontitis at 28 days. In the histopathological analysis, AP14 had focal regions of moderate mononuclear inflammatory infiltrate, and AP28 had an intense inflammatory infiltrate with bacterial colonies. In the biochemical evaluation, GSH, TEAC, and TBARS were increased after 14 days. However, GSH returned to control levels, TEAC was similar to AP14, and TBARS increased significantly after 28 days. Therefore, the oxidative biochemistry response was modulated according to the progression of periapical damage. After 14 days, the organism could still react to the injury. Aim: This study analyzed public procurements for different endodontic materials used in the Brazilian public health system and evaluated the variables related to their cost. Methods: A time-series study was performed by screening materials for endodontic application in the public Brazilian Databank of Healthcare Prices from 2010 to 2019. Data were categorized according to material composition and clinical application. The collated variables were used in a multiple linear regression model to predict the impact of unit price in procurement processes. Results: A total of 5,973 procurement processes (1,524,693 items) were evaluated. Calcium hydroxides were found in 79% of the observations (4,669 processes). Prices drop each year by US$1.87 while MTAs and epoxy resins are increasingly purchased at higher prices (US$50.87; US$67.69, respectively). The microregion, the procurement modality, and the type of institution had no influence on unit prices in the adjusted model (p > 0.05). Conclusions: Calcium hydroxide-based materials were the cheapest and most frequently purchased endodontic materials in the public health care system. Novel formulations are being implemented into clinical practice over time and their cost may be a barrier to the broad application of materials such as MTAs, despite their effectiveness. Purpose: To synthesize niobium silicate particles (SiNb) and incorporate into resin-based luting agents. Methods: SiNb particles were synthesized and characterized by x-ray diffraction, FTIR spectroscopy, particle size, and specific surface area. Luting agents were formulated with 50 wt% BisGMA, 30 wt% TEGDMA and 20 wt% UDMA. BAPO was used as a photoinitiator in 1mol%. The SiNb particles were incorporated into the agents at concentrations of 50 wt% (SiNb50%) or 65% wt% (SiNb65%). Barium glass particles at the same concentrations were used as controls (SiBa50% and SiBa65%). Refractive index, degree of conversion (DC), polymerization kinetics, softening in solvent, radiopacity, film thickness, color stability, flexural strength (FS) and micro shear bond strength (µSBS) were evaluated. Results: SiNb particles were successfully synthesized with an adequate structure to be applied as inorganic fillers. SiNb groups had higher DC, lower %ΔKHN, greater film thickness and greater radiopacity than the SiBa groups. Color stability was greater for SiNb50% and SiNb65% after six months of storage, as demonstrated by ΔE00. ΔWID values were lower for the SiBa groups. FS results decreased over time, and lower values were found for SiNb. In µSBS, the values of SiNb65% were higher at 24 h (45.22 MPa) and at six months (36.83 MPa), with statistically differences from values for the SiBa groups. Conclusions: SiNb particles were successfully synthesized using the sol-gel method, and their incorporation into luting agents at a concentration of up to 65% improved the physicomechanical characteristics and color stability of these agents. The aim of this study was to evaluate the influence of different calcium phosphates (CaPs) on the physical, biological, and remineralizing properties of experimental resin-based sealants (RBSs). Triethylene-glycol dimethacrylate (90wt%) and bisphenol A-glycidyl methacrylate (10wt%) were used to produce resin-based sealants. Hydroxyapatite (SHAp), α-tricalcium phosphate (Sα-TCP) and octacalcium phosphate (SOCP) were added to the sealants in a 10wt% concentration. One group without CaPs was used as the control group (SCG). The degree of conversion (DC) was assessed with Fourier-transformed infrared spectroscopy, whereas cytotoxicity was tested with the HaCaT keratinocyte cell line. The ultimate tensile strength (UTS) was used to assess the mechanical strength of the experimental RBSs. Sealed enamel was used for colorimetric assay. Mineral deposition was assessed with Raman spectroscopy after 7, 14, and 28 days of sample immersion in artificial saliva. Scanning electron microscopy was used to analyze the surface morphology after 28 days of immersion. The addition of 10wt% of fillers significantly reduced the DC of sealants. SOCP groups showed reduced cell viability. Higher UTS was found for Sα-TCP a nd SHAp. The color analysis showed that SGC and demineralized teeth presented higher mismatches with the sound tissue. Mineral deposition was observed for SHAp a nd S α-TCP after 7 days, with increased phosphate content and mineral deposits for SHAp after 28 days. RBS with the addition of 10% HAp promoted increased mineralization in vitro after 28 days, and did not affect cell viability, DC, mechanical properties, or RBS color in the enamel. Orthodontic adhesives with antimicrobial and remineralizing properties may be an alternative to control white spot lesions around brackets. The aim of this study is to develop an experimental orthodontic adhesive containing boron nitride nanotubes (BNNT) and alkyl trimethyl ammonium bromide (ATAB). Methacrylate (BisGMA and TEGDMA) monomers were used to formulate the adhesives. Four experimental groups were produced with the addition of 0.1 wt.% BNNT (GBNNT); 0.1 wt.% ATAB (GATAB); and 0.2 wt.% BNNT with ATAB (GBNNT/ATAB); in the control group, no fillers were added (GCtrl). The degree of conversion, cytotoxicity, softening in solvent, contact angle and free surface energy, antibacterial activity, shear bond strength, and mineral deposition were evaluated. Adhesives achieved degree of conversion higher than 50% and cell viability higher than 90%. GBNNT and GATAB adhesives exhibited reduced softening in solvent. Mean free surface energy was decreased in the GBNNT adhesive. Significant reduction in bacterial growth was observed in the GBNNT/ATAB. No statistical difference was found for shear bond strength. Mineral deposition was found in GBNNT, GATAB, and GBNNT/ATAB groups after 14 and 28 days. The addition of 0.2% BNNT/ATAB to an experimental orthodontic adhesive inhibited bacterial growth and induced mineral deposition without affecting the properties of the material.
