The Earth Explorer Atmospheric Dynamics Mission (ADM-Aeolus) of ESA will be the first-ever satellite to provide global observations of wind profiles from space. Its single payload, namely the Atmospheric Laser Doppler Instrument (ALADIN) is a directdetection high spectral resolution Doppler Wind Lidar (DWL), operating at 355 nm, with a fringe-imaging receiver (analysing aerosol and cloud backscatter) and a double-edge receiver (analysing molecular backscatter). In order to meet the stringent mission requirements on wind retrieval, ESA is conducting various science support activities for the consolidation of the on-ground data processing, calibration and sampling strategies. Results from a recent laboratory experiment to study Rayleigh-Brillouin scattering and improve the characterisation of the molecular lidar backscatter signal detected by the ALADIN double-edge Fabry- Perot receiver will be presented in this paper. The experiment produced the most accurate ever-measured Rayleigh-Brillouin scattering profiles for a range of temperature, pressure and gases, representative of Earth’s atmosphere. The measurements were used to validate the Tenti S6 model, which is implemented in the ADM-Aeolus ground processor. First results from the on-going Vertical Aeolus Measurement Positioning (VAMP) study will be also reported. This second study aims at the optimisation of the ADM-Aeolus vertical sampling in order to maximise the information content of the retrieved winds, taking into account the atmospheric dynamical and optical heterogeneity. The impact of the Aeolus wind profiles on Numerical Weather Prediction (NWP) and stratospheric circulation modelling for the different vertical sampling strategies is also being estimated.

The AVR9 elicitor from the fungal pathogen Cladosporium fulvum induces defense-related responses, including cell death, specifically in tomato (Lycopersicon esculentum Mill.) plants that carry the Cf-9 resistance gene. To study biochemical mechanisms of resistance in detail, suspension cultures of tomato cells that carry the Cf-9 resistance gene were initiated. Treatment of cells with various elicitors, except AVR9, induced an oxidative burst, ion fluxes, and expression of defense-related genes. Agrobacterium tumefaciens-mediated transformation of Cf9 tomato leaf discs with Avr9-containing constructs resulted efficiently in transgenic callus formation. Although transgenic callus tissue showed normal regeneration capacity, transgenic plants expressing both the Cf-9 and the Avr9 genes were never obtained. Transgenic F1 seedlings that were generated from crosses between tomato plants expressing the Avr9 gene and wild-type Cf9 plants died within a few weeks. However, callus cultures that were initiated on cotyledons from these seedlings could be maintained for at least 3 months and developed similarly to callus cultures that contained only the Cf-9 or the Avr9 gene. It is concluded, therefore, that induction of defense responses in Cf9 tomato cells by the AVR9 elicitor is developmentally regulated and is absent in callus tissue and cell-suspension cultures, which consists of undifferentiated cells. These results are significant for the use of suspension-cultured cells to investigate signal transduction cascades.

The RTP tokamak (R = 0.72 m, a = 0.164 m, B-phi < 2 5.T, I-p = < 150 kA) is equipped with three gyrotrons (2 x 60 GHz, 180 kW, 100 ms each; 1 x 110 GHz, 500 kW, 200 ms) for electron cyclotron heating (ECH) and current drive (ECCD). The power from one of the 60 GHz gyrotrons is launched via an adjustable mirror from the high field side (HFS) in the 1X-mode. The power of both other gyrotrons is sent in perpendicularly to the toroidal magnetic field from the low field side (LFS). A comprehensive set of high-resolution multichannel plasma diagnostics is available to study the detailed behaviour of various plasma phenomena. First, recent diagnostic innovations are briefly discussed. Then, new physics results are presented for ohmic and EC heated plasmas. ECCD, slide-away discharges, discharges with a hollow temperature profile and MHD phenomena, including sawteeth and disruptions. are treated.

A 'non-local' response of the plasma core triggered by peripheral plasma perturbations other than laser ablation is found in the RTP tokamak. Oblique pellet injection (OPI) has been used to induce fast cooling of the peripheral plasma. In response, an inward cold pulse (T-e drop) and a slightly delayed core T-e rise are observed. A somewhat similar 'non-local' response is observed when the peripheral plasma is heated by modulated electron cyclotron heating or by fast current ramps, i.e. the core temperature drops in response to the peripheral heating. The plasma conditions for the occurrence of the 'non-local' response have been investigated. The core T-e rise following OPI is associated with the formation of a large temperature gradient in the region 1 < q < 2. The T-e rise is largest at low electron density and for large pellet deposition radii. Above a critical density the T-e rise disappears and only the (weaker) drop in core T-e is observed. Time dependent transport simulations show that the propagation of the inward cold pulse is consistent with local transport, while the core T-e rise is a slower phenomenon requiring a large transient drop of chi(e) in the region 1 < q < 2. In the Rijnhuizen Tokamak Project plasmas, a transient rise of the core electron temperature is observed when hydrogen pellets are injected tangentially to induce fast cooling of the peripheral region. High-resolution Thomson scattering measurements show that the T-e rise is associated with large temperature gradients in the region 1 < q < 2. This region acts as a layer of transiently increased thermal resistivity (transport barrier) when probed by fast heat pulses from modulated electron cyclotron heating.
