Quinoa (Chenopodium quinoa) cultivation has become increasingly popular in NW Europe but little is known about the performance of contract-free varieties in this region. In this study, we phenotyped 25 quinoa varieties on a single-plant basis in a field trial in Belgium. In addition, we optimized breeding tools such as NIRS (near-infrared reflectance spectroscopy) to estimate the seed crude protein content and a multiplex PCR set to identify true F1 progeny from pair crosses. We identified 14 varieties with sufficiently early maturity, 17 varieties with plant height below 150 cm, 21 large-seeded varieties, four varieties with a crude protein content exceeding 15%, and two low-saponin varieties. A variety of seed colors and plant morphological traits was observed. Seed yield was not correlated with maturity, plant height or saponin content, but was negatively correlated with seed crude protein content. NIRS could accurately predict seed crude protein content with a determination coefficient of 0.94. Our multiplex SSR set could correctly identify the paternity in 77% to 97% of progeny, depending on the pair cross. In conclusion, our study identified various contract-free varieties that may be suitable for cultivation in NW Europe. Transgenic lines engineered through wild type Rhizobium rhizogenes display an altered phenotype known as the Ri phenotype. This phenotype includes a more compact plant habit, which has proved useful to obtain more compact varieties that require less chemical growth regulation. Here, we develop a method for the molecular and cytogenetic characterization of Cape daisy (Osteospermum fruticosum Norl.) Ri lines in order to predict segregation of pRi T-DNA genes. Analysis of copy number variation (CNV) by means of digital PCR indicated large variation in the copy number of the inserted root oncogenic loci (rol) genes, ranging from 1 to more than 15 copies. In addition, up to 9 copies of the auxin biosynthesis genes (aux) were present in a single Ri line. Visualization of pRiA4 and pRi1724 rol and aux insertion in 4 Ri lines was performed through Fluorescence In Situ Hybridization. The number of rol integrated loci varied from 1 to 3 loci. In contrast, the different TR-gene copies were confined to a single locus which consistently co-localized with a TL locus, this was demonstrated for the first time. Based on CNV and FISH a single Ri line, harboring 7 pRi1724 rol gene copies dispersed over 3 integration loci, was selected for breeding. Copy number segregation in R1 progeny of 2, 3, 4 and 5 pRi1724 copies was confirmed, indicating that the evaluation of the breeding value of first generation Ri lines is possible through CNV and FISH. Hardy geranium is a popular ornamental plant with a high market demand. As a result, there is a strong need for continuous innovation to improve its ornamental qualities. Interspecific hybridization is a widely used technique for introducing desirable traits and creating new cultivars with added value. To explore the possibilities of creating novelty within the Geranium genus, we conducted 2438 interspecific crosses over three flowering seasons, resulting in the successful harvest of 445 seeds and the final production of 82 seedlings. Using embryo rescue increased the germination rate of a seedling from 0.17% to 1.01%. To validate the paternal genetic contribution in the new seedlings, we used amplified fragment length polymorphism (AFLP) to assess the proportion of uniquely inherited paternal markers in all seedlings. AFLP analysis confirmed that 54 of the 82 seedlings were indeed true hybrids (65.8% hybridization success rate). A morphological evaluation of the flowers and leaves, including flower diameter (mm), style length (mm), number of flowers, flowering period (weeks), and color of both flowers and leaves, provided further confirmation of the F1 hybrid status of the seedlings. The results of our interspecific breeding demonstrated the efficiency of interspecific hybridization in hardy geraniums and its potential to create distinctive and novel cultivars for the ornamental market. Chitin soil amendment is known to improve soil quality, plant growth and stress resilience, but the underlying mechanisms are not well understood. In this study, we monitored chitin's effect on lettuce physiology every two weeks through an eight-week growth period, analyzed the early transcriptional reprogramming and related metabolomic changes of lettuce, in response to crab chitin treatment in peat-based potting soil. In commercial growth conditions, chitin amendment still promoted lettuce growth, increased chlorophyll content, the number of leaves and crop head weight from week six. The flavonoid content in lettuce leaves was altered as well, showing an increase at week two but a decrease from week six. Transcriptomic analysis showed that over 300 genes in lettuce root were significantly differentially expressed after chitin soil treatment. Gene Ontologyterm (GO) enrichment analysis revealed statistical overrepresentation of GO terms linked to photosynthesis, pigment metabolic process and phenylpropanoid metabolic process. Further analysis of the differentially expressed genes (DEGs) showed that the flavonoid pathway was mostly upregulated whereas the bifurcation of upstream phenylpropanoid pathway towards lignin biosynthesis was mostly downregulated. Metabolomic analysis revealed the upregulation of salicylic acid, chlorogenic acid, ferulic acid, and p-coumaric acid in chitin-treated lettuce seedlings. These phenolic compounds (PCs) mainly influence the phenylpropanoid biosynthesis pathway and may play important roles in plant defense reactions. Rose is the most economically important ornamental plant. However, cold stress seriously affects the survival and regrowth of garden roses in northern regions. Cold acclimation was studied using two genotypes (Rosa wichurana and R. hybrida ‘Yesterday’) selected from a rose breeding program. During the winter season (November to April), the cold hardiness of stems, soluble sugar content, and expression of dehydrins and the related key genes in the soluble sugar metabolism were analyzed. ‘Yesterday’ is more cold-hardy and acclimated faster, reaching its maximum cold hardiness in December. R. wichurana is relatively less cold-hardy, only reaching its maximum cold hardiness in January after prolonged exposure to freezing temperatures. Dehydrin transcripts accumulated significantly during November–January in both genotypes. Soluble sugars are highly involved in cold acclimation, with sucrose and oligosaccharides significantly correlated with cold hardiness. Sucrose occupied the highest proportion of total soluble sugars in both genotypes. During November– January, downregulation of RhSUS was found in both genotypes, while upregulation of RhSPS was observed in ‘Yesterday’ and upregulation of RhINV2 was found in R. wichurana. Oligosaccharides accumulated from November to February and decreased to a significantly low level in April. RhRS6 had a significant upregulation in December in R. wichurana.
