Abstract

In prostate cancer, androgen receptor (AR)-targeting agents are very effective in various disease stages. However, therapy resistance inevitably occurs, and little is known about how tumor cells adapt to bypass AR suppression. Here, we performed integrative multiomics analyses on tissues isolated before and after 3 months of AR-targeting enzalutamide monotherapy from patients with high-risk prostate cancer enrolled in a neoadjuvant clinical trial. Transcriptomic analyses demonstrated that AR inhibition drove tumors toward a neuroendocrine-like disease state. Additionally, epigenomic profiling revealed massive enzalutamide-induced reprogramming of pioneer factor FOXA1 from inactive chromatin sites toward active cis-regulatory elements that dictate prosurvival signals. Notably, treatment-induced FOXA1 sites were enriched for the circadian clock component ARNTL. Posttreatment ARNTL levels were associated with patients' clinical outcomes, and ARNTL knockout strongly decreased prostate cancer cell growth. Our data highlight a remarkable cistromic plasticity of FOXA1 following AR-targeted therapy and revealed an acquired dependency on the circadian regulator ARNTL, a novel candidate therapeutic target.SignificanceUnderstanding how prostate cancers adapt to AR-targeted interventions is critical for identifying novel drug targets to improve the clinical management of treatment-resistant disease. Our study revealed an enzalutamide-induced epigenomic plasticity toward prosurvival signaling and uncovered the circadian regulator ARNTL as an acquired vulnerability after AR inhibition, presenting a novel lead for therapeutic development. 4. The Role of Teaching Staff in Fostering Perceived Employability of University Students

Abstract

For university students, perceived employability is gaining growing importance in dealing with occupational uncertainty. However, how perceived employability is shaped in the university setting is still under-researched. This study examines how support from university teaching staff influences perceived employability in a sample of Italian final-year university students. We draw on Conservation on Resources Theory to hypothesise a positive impact of support from teaching staff on students' perceived employability. In addition, we contend that perceived employability predicts students' psychological well-being. To test our hypotheses, one hundred twenty-seven university students completed a survey three times over a 10-month period. The results confirm that support from teaching staff enhances students' perceived employability. Moreover, a positive effect of perceived employability on students' psychological well-being emerges. Perceived employability also appears to mediate the relationship between support from teaching staff and psychological well-being. Using a time-lagged approach, this study enhances the empirical knowledge about antecedents and outcomes of students' perceived employability. This scholarly work underlines universities' responsibility in refining teachers' roles and didactical practices to equip their students with career resources.

11. The Curvilinear Relation between Work Predictability and Creativity

Abstract

In this study, we investigate how the level of work control predictability affects employee creativity. Specifically, we examine whether supervisor and coworker support moderate the predictability-creativity relationship. We use survey data from 128 employee--supervisor dyads from a governmental organization in Belgium. Multilevel analyses demonstrated a significant inverted U-curve relationship between predictability and leader-rated creativity, indicating that too little or too much work control predictability contributes less to employee creativity compared to moderate levels of work control predictability. Moreover, supervisor and coworker support moderate this relationship in such a way that for employees who receive low support the inverted U-curve relationship is significantly stronger compared to employees that receive high support. This is the first study that focuses on work control predictability in relation to creative performance.

16. Self-Perceived Employability and Psychological Well-Being Among Italian Students and Graduates: A Three-Wave Cross-Lagged Study.

Abstract

This study investigated the reciprocal relationship between self-perceived employability (SPE) and psychological well-being (PWB), a connection previously suggested by theory but not yet explored empirically in early career research. Drawing from the Conservation of Resources theory, we analysed the reciprocal effects between SPE and PWB. Using data collected from 376 Italian university students and graduates, we tested the hypotheses with the random intercept cross-lagged panel model (RI-CLPM) to analyse the SPE-PWB reciprocal linkage at both the between-person and within-person levels. While we observed a reciprocal SPE-PWB association across three waves at the between-person level, we obtained mixed findings at the within-person level. This study is the first to test the reciprocal associations between SPE and PWB among labour market entrants adopting a within-person approach. The findings offer new empirical insights into SPE during early career stages, with implications for both future research and practical applications. [ABSTRACT FROM AUTHOR]

Abstract

In industrial product development with customers, it is well‐known that lead userness is associated with novel and commercially attractive products. We take a next step by analyzing if lead userness is related to product development that potentially affects industry‐level practices. Our main hypothesis is that lead userness results in the pursuit of Schumpeterian product opportunities: disequilibrating, radical, rare, and based on new knowledge and creative activities. In contrast, at low lead userness, more Kirznerian product opportunities are expected—which maintain industry standards. The hypothesis is supported by data from 139 high‐tech small firms. Next, we anticipate that factors related to the selection of lead users, and the effective processing of their inputs, moderate the connection between lead userness and Schumpeterian opportunity. As for selection, we find a stronger connection when firms collaborate with "new" customers. Conversely, when involved customers have been business partners already for a long time, the pursuit of Schumpeterian opportunities is mitigated. Also, tentative evidence is found that the connection between lead userness and Schumpeterian opportunity amplifies when involved customers are less dominant market players. Finally, when it comes to processing lead users' input, we report tentative evidence that firms' R&D intensity should be higher (indicating ability to process lead users' inputs and make continued development efforts). Our findings help to explain why lead users have been associated with industry dynamics, and provide guidance to identify "relevant" lead users when firms seek product opportunities that potentially disequilibrate their industry.
