Gas monitoring is a prerequisite to understanding the exchange, diffusion, and migration processes of natural gases within underground environments, which are involved in several applications such as geological sequestration of CO2. In this study, three different techniques (micro-GC, infrared, and Raman spectroscopies) were deployed on an experimental flooded borehole for monitoring purposes after CO2 injection. The aim was to develop a real-time chemical monitoring device to follow dissolved gas concentrations by measurements in water inside the borehole but also at the surface through a gas collection system in equilibrium with the borehole water. However, all three techniques must be calibrated to provide the most accurate quantitative data. For this, a first step of calibration in the laboratory was carried out. A new calibrations were required to determine partial pressure and/or concentrations of gases in water or in the gas collection system. For gas phase analysis, micro-GC, FTIR spectroscopy, and Raman spectroscopy were compared. New calibration of the micro-GC was done for CO2, CH4, and N2 with uncertainty from ±100 ppm to 1.5 mol% depending on the bulk concentration and the type of gas. The FTIR and Raman spectrometers were previously calibrated for CO2, and CO2, N2, O2, CH4, and H2O, respectively with an accuracy of 1–6% depending on concentration scale, gas and spectrometer. Dissolved CO2 in water was measured using a Raman spectrometer equipped with an immersion probe. The uncertainty on the predicted dissolved CO2 concentration and partial pressure was ±0.003 mol·kg−1 and ±0.05 bar, respectively.

Abstract Mutations in mitochondrial energy-producing genes lead to a heterogeneous group of untreatable disorders known as primary mitochondrial diseases (MD). Leigh syndrome (LS) is the most common pediatric MD and is characterized by progressive neuromuscular affectation and premature death. Here, we show that daily cannabidiol (CBD) administration significantly extends lifespan and ameliorates pathology in two LS mouse models, and improves cellular function in fibroblasts from LS patients. CBD delays motor decline and neurodegenerative signs, improves social deficits and breathing abnormalities, decreases thermally induced seizures, and improves neuropathology in affected brain regions. Mechanistically, we identify peroxisome proliferator-activated receptor gamma (PPARγ) as a key nuclear receptor mediating CBD’s beneficial effects, while also providing proof of dysregulated PPARγ expression and activity as a common feature in both mouse neurons and fibroblasts from LS patients. Taken together, our results provide the first evidence for CBD as a potential treatment for LS.

This is the first viral metagenomic analysis of grapevine conducted in Mexico. During the summer of 2021, 48 plants displaying virus-like symptoms were sampled in Queretaro, an important grapevine-producing area of Mexico, and analyzed for the presence of viruses via high-throughput sequencing (HTS). The results of HTS were verified by real-time RT-PCR following a standardized testing scheme (Protocol 2010). Fourteen different viruses were identified, including grapevine asteroid mosaic-associated virus (GAMaV), grapevine Cabernet Sauvignon reovirus (GCSV), grapevine fanleaf virus (GFLV), grapevine fleck virus (GFkV), grapevine Pinot gris virus (GPGV), grapevine red globe virus (GRGV), grapevine rupestris stem pitting-associated virus (GRSPaV), grapevine rupestris vein feathering virus (GRVFV), grapevine Syrah virus 1 (GSyV-1), grapevine virus B (GVB), and grapevine leafroll-associated viruses 1, 2, 3, 4 (GLRaV1, 2, 3, 4). Additionally, divergent variants of GLRaV4 and GFkV, and a novel Enamovirus-like virus were discovered. This is the first report of GAMaV, GCSV, GLRaV4, GPGV, GRGV, GRVFV, and GSyV-1 infecting grapevines in Mexico; the impact of these pathogens on production is unknown.

Summary: Background: High temperatures have been associated with increased mortality, with evidence reported predominately in large cities and for total cardiovascular or respiratory deaths. This case-crossover study examined heat-related cause-specific cardiopulmonary mortality and vulnerability factors using small-area data from Germany. Methods: We analyzed daily counts of cause-specific cardiopulmonary deaths from 380 German districts (2000–2016) and daily mean temperatures estimated by spatial–temporal models. We applied conditional quasi-Poisson regression using distributed lag nonlinear models to examine heat effects during May–September in each district and random-effects meta-analysis to pool the district-specific estimates. Potential individual- and district-level vulnerability factors were examined by subgroup analyses and meta-regressions, respectively. Findings: Heat was associated with increased mortality risks for all cardiopulmonary sub-causes. The relative risk (RR) of total cardiovascular and respiratory mortality for a temperature increment from the 75th to the 99th percentile was 1.24 (95% confidence interval: 1.23, 1.26) and 1.34 (1.30, 1.38), respectively. The RRs of cardiovascular sub-causes ranged from 1.16 (1.13, 1.19) for myocardial infarction to 1.32 (1.29, 1.36) for heart failure. For respiratory sub-causes, the RR was 1.27 (1.22, 1.31) for COPD and 1.49 (1.42, 1.57) for pneumonia. We observed greater susceptibility related to several individual- and district-level characteristics, e.g., among females or in highly urbanized districts. Heat vulnerability factors remained consistent between urban and rural areas. Interpretation: Our study highlights heat-related increases in cause-specific cardiopulmonary mortality across Germany and identifies key vulnerability factors, offering insights for improving public health practices to mitigate heat-related health impacts. One of the options considered to mitigate greenhouse gas concentrations in the atmosphere is underground storage of CO2. There is a strong need for enhancing and developing methods that would help throughout the duration life of such underground storage, to ensure the safety and able to monitor the evolution of the injected CO2 plume. Among these, geochemical methods can play an important role. Here, we describe results acquired under the research programme “Géocarbone-Monitoring”, partially funded by the French National Research Agency, on the Montmiral natural analogue in South-Eastern France. Other results obtained under the same research programme in the French Massif Central are reported elsewhere in this volume. Spot sampling methods allowing a great geographical coverage and continuous measurements on selected points were undertaken in 2006 and 2007, in order to determine soil gas concentrations and fluxes as well as carbon isotope ratio determinations. One important result is that without any evidence of deep CO2 leakage, both CO2 concentrations and fluxes appear to be higher than can be explained only by biological activities. Further investigations are thus needed to understand the gas evolution better throughout the year. Une des options envisagées pour réguler les concentrations de gaz à effet de serre dans l’atmosphère est le stockage souterrain du CO2. Dans ce domaine existe un fort besoin de renforcer et de développer des méthodes susceptibles d’être utilisées tout au long de la durée de vie de ces stockages souterrains, afin de s’assurer de leur sécurité et de pouvoir suivre l’évolution du panache de CO2 injecté. Parmi elles, les méthodes géochimiques peuvent jouer un rôle important. Nous décrivons ici les résultats acquis dans le cadre du programme de recherche « Géocarbone-Monitoring » financé en partie par l’Agence Nationale de la Recherche sur l’analogue naturel de Montmiral dans le Sud-Est de la France. D’autres résultats obtenus dans le cadre de ce même programme de recherche sont rapportés dans un autre article présent dans ce volume. Des méthodes d’échantillonnage ponctuelles permettant une grande couverture géographique et des mesures en continu sur certains points sélectionnés ont été entreprises en 2006 et 2007, afin de permettre la détermination des concentrations en gaz du sol et des flux, ainsi que des déterminations des rapports isotopiques du carbone. Sans aucune preuve d’une fuite de CO2 profond, à la fois les concentrations et les flux de CO2 semblent être plus élevés que ne pouvant s’expliquer par l’effet des seules activités biologiques. Des études plus approfondies sont donc nécessaires pour mieux comprendre l’évolution des gaz tout au long de l’année.

This study presents an overview of some results obtained within the French ANR (National Agency of Research) supported Géocarbone-Monitoring research program. The measurements were performed in Sainte-Marguerite, located in the French Massif Central. This site represents a natural laboratory for CO2/fluid/rock interactions studies, as well as CO2 migration mechanisms towards the surface. The CO2 leaking character of the studied area also allows to test and validate measurements methods and verifications for the future CO2 geological storage sites. During these surveys, we analyzed soil CO2 fluxes and concentrations. We sampled and analyzed soil gases, and gas from carbo-gaseous bubbling springs. A one-month continuous monitoring was also tested, to record the concentration of CO2 both in atmosphere and in the soil at a single point. We also developed a new methodology to collect soil gas samples for noble gas abundances and isotopic analyses, as well as carbon isotopic ratios. Our geochemical results, combined with structural geology, show that the leaking CO2 has a very deep origin, partially mantle derived. The gas rises rapidly along normal and strike-slip active faults. CO2 soil concentrations (also showing a mantle derived component) and CO2 fluxes are spatially variable, and reach high values. The recorded atmospheric CO2 is not very high, despite the important CO2 degassing throughout the whole area. Cette étude présente les principaux résultats de campagnes de monitoring géochimique menées en 2006 et 2007 dans le cadre du projet Géocarbone-Monitoring, sur le site de Sainte-Marguerite, situé dans le Massif Central. Ce site constitue un « laboratoire naturel » pour l’étude des interactions CO2/fluides/roches et des mécanismes de migration du CO2 vers la surface, à l’échelle des temps géologiques. Le caractère particulièrement émissif de cet « analogue » permet également de tester et valider des méthodes de mesure et de surveillance des futurs sites de stockage de CO2. Au cours des campagnes de terrain, nous avons analysé des flux de CO2 entre le sol et l’atmosphère, et nous avons prélevé et analysé à la fois des gaz des sols, et du gaz provenant de sources carbo-gazeuses, présentes dans toute la région. Un dispositif de « monitoring continu » dans le temps a également été testé, afin d’enregistrer conjointement les teneurs en CO2 de l’atmosphère et dans le sol en un point précis. Nous avons pu mettre au point un suivi géochimique basé sur la composition isotopique des gaz rares prélevés dans les sols. L’ensemble de nos résultats, confronté à la géologie de terrain, nous a permis de mettre en évidence l’origine mantellique du CO2. Ce CO2 remonte rapidement à la surface à l’état gazeux, le long de failles normales et/ou décrochantes, actives actuellement. Les teneurs et flux de CO2 dans le sol sont spatialement variables et élevés, et montrent également une origine mantellique. Les teneurs atmosphériques semblent faiblement augmenter par rapport à l’important dégazage observé dans la région.

Metabolic adaptations to negative energy balance, as well as lipomobilization, influence inflammatory responses, immune function, and oxidative stress in animals. This study aimed to evaluate the biochemical profile of Mediterranean buffaloes with different levels of lipomobilization from the prepartum to the postpartum period. A total of 76 Mediterranean buffaloes were enrolled, and a weekly blood sample was taken from 7 weeks before to 6 weeks after calving. The concentration of non-esterified fatty acids (NEFAs) was determined in serum and was used to categorize buffaloes into three lipomobilization groups 7 weeks before calving: mild (NEFA-I; NEFA ≤ 0.29 mEq/L; n = 18), medium (NEFA-II; 0.29 0.29 mEq/L; NEFA-II and NEFA-III) at 7 weeks before calving should be monitored more closely to reduce the risk of metabolic diseases. Furthermore, the medium (NEFA-II) and severe (NEFA-III) lipomobilization groups could be associated with differences in the animals’ ability to manage their metabolic status. Specifically, the severe mobilization group was most associated with a greater energy deficit during both the prepartum and postpartum periods without oxidative stress. On the contrary, the medium mobilization group was associated with a less severe energy deficit but was also associated with an inflammatory status and oxidative stress during the prepartum period. These distinctions highlight the need for tailored management strategies to address varying levels of metabolic stress in dairy buffaloes.

This article deals with the investigation of iron losses in toroidally wound laminated magnetic cores excited with wide-bandgap-device-based power electronic converters. The study aims to analyze the impact of selected pulsewidth modulation voltages on the iron losses, through an extensive experimental measurement campaign. In particular, four toroidal specimens made of different magnetic materials are supplied by pulsewidth-modulated voltage waveforms with switching frequencies ranging from 1 to 350 kHz and different deadtimes. Test campaigns have been conducted with the dual objectives of critically reviewing an engineering method proposed in the prior literature for predicting iron losses under distorted voltage waveforms. Additionally, the aim is to extend this estimation model to accommodate the highest frequencies currently employed in high-speed ac motor drives equipped with wide-bandgap semiconductor power converters.

The increase in the frequency and intensity of drought events expected in the coming decades in Western Europe may disturb forest biogeochemical cycles and create nutrient deficiencies in trees. One possible origin of nutrient deficiency is the disturbance of the partitioning of the green leaf pool during the leaf senescence period between resorption, foliar leaching and senesced leaves. However, the effects of drought events on this partitioning and the consequences for the maintenance of tree nutrition are poorly documented. An experiment in a beech forest in Meuse (France) was conducted to assess the effect of drought events on nutrient canopy exchanges and on the partitioning of the green leaf pool during the leaf senescence period. The aim was to identify potential nutritional consequences of droughts for trees. Monitoring nutrient dynamics, including resorption, chemistry of green and senesced leaves, foliar absorption and leaching in mature beech stands from 2012 to 2019 allowed us to compare the nutrient exchanges for three nondry and three dry years (i.e., with an intense drought event during the growing season). During dry years, we observed a decrease by almost a third of the potassium (K) partitioning to resorption (i.e. resorption efficiency), thus reducing the K reserve in trees for the next growing season. This result suggests that with the increased drought frequency and intensity expected for the coming decades, there will be a risk of potassium deficiency in trees, as already observed in a rainfall exclusion experiment on the same study site. Reduced foliar leaching and higher parititioning to the senesced leaves for K and phosphorus (P) were also observed. In addition, a slight increase in nitrogen (N) resorption efficiency occurred during dry years which is more likely to improve tree nutrition. The calcium (Ca) negative resorption decreased, with no apparent consequence in our study site. Our results show that nutrient exchanges in the canopy and the partitioning of the green leaf pool can be modified by drought events, and may have consequences on tree nutrition.

The aim of this study was to evaluate the relation of inflammatory biochemical markers as neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR) and systemic inflammation index (SII) with POD occurrence.Methods: This was a single-center, retrospective, observational study. We analyzed the perioperative data of patients who had undergone elective vascular surgery procedures. The occurrence of delirium after procedure was used to divide the population in two groups: POD-pos and POD-neg group. ROC curves were performed to find the appropriate cut-off values of NLR, PLR and SII. Multivariate analysis was used to identify the independent predictors for POD.Results: A total of 646 patients were enrolled. Mean age was 76.2± 9.8 years, 68.4% were male. Seventy-three patients (11.3%) developed POD. Mean hospital stay was significantly increased in the POD-pos group (6.1± 5.4 vs 3.2± 2.8 days, p=0.0001). In-hospital reinterventions were more frequent in the POD-pos group (8.2% vs 3.8%). Blood values analysis reported significant differences: Hb, NLR, PLR, SII, creatinine and RCP were strongly increased (p< 0.05) in the POD-pos group. ROC curves identified cut-off values for NLR> 3.57, PLR> 139.2 and SII> 676.4. Multivariate analysis revealed that age, Renal Failure, peripheral revascularization procedures, major amputation, general anesthesia, hospitalization in the previous month, NLR> 3.57 and SII> 676.4 were independent risk factors for POD.Conclusion: POD represents a common complication of vascular surgery patients. Our study demonstrated that NLR, PLR and SII are reliable and readily available laboratory predictors of POD in vascular surgery that could help in POD risk-stratification.Keywords: delirium, vascular disease, risk assessment, vascular surgery, elderly

The intricate behavior of particle acceleration and transport mechanisms complicates the overall efforts in formulating a comprehensive understanding of solar energetic particle (SEP) events; these efforts include observations of low-energy particles (from tens of keV to hundreds of MeV) by space-borne instruments and measurements by the ground-based neutron monitors of the secondary particles generated in the Earth atmosphere by SEPs in the GeV range. Numerous space-borne missions provided good data on the nature/characteristics of these solar particles in past solar cycles, but more recently—concurrently with the rise toward the maximum of solar cycle 25—the High-Energy Particle Detector (HEPD-01) proved to be well suited for the study of solar physics and space weather. Its nominal 30–300 MeV energy range for protons can enlarge the detection capabilities of solar particles at low Earth orbit, closer to the injection limit of many SEP events. In this work, we characterize three SEP events within the first six months of 2022 through spectral and velocity dispersion analysis, assessing the response of HEPD-01 to >M1 events.

Abstract Omega 3 polyunsaturated fatty acids (n-3 PUFA) are known to have beneficial effects on cardiovascular and metabolic health. However, whether different sources of n-3 PUFA, for instance fatty fish vs vegetable oils, could elicit different effects on glucose and lipid metabolism, remains to be determined. Herein we examine recent findings showing that while a plant-based n-3 PUFA supplementation for six months can reduce fasting blood glucose, marine-based n-3 PUFA can instead reduce serum levels of triglycerides. We also discuss the potential molecular mechanisms that could underlie these different effects on the regulation of glycolipid metabolism.

Salmonella isolated from dairy farms has a significant effect on animal health and productivity. Different serogroups of Salmonella affect both human and bovine cattle causing illness in both reservoirs. Dairy cows and calves can be silent Salmonella shedders, increasing the possibility of dispensing Salmonella within the farm. The aim of this study was to determine the genomic characteristics of Salmonella isolates from dairy farms and to detect the presence of virulence and antimicrobial resistance genes. A total of 377 samples were collected in a cross-sectional study from calves, periparturient cow feces, and maternity beds in 55 dairy farms from the states of Aguascalientes, Baja California, Chihuahua, Coahuila, Durango, Mexico, Guanajuato, Hidalgo, Jalisco, Queretaro, San Luis Potosi, Tlaxcala, and Zacatecas. Twenty Salmonella isolates were selected as representative strains for whole genome sequencing. The serological classification of the strains was able to assign groups to only 12 isolates, but with only 5 of those being consistent with the genomic serotyping. The most prevalent serovar was Salmonella Montevideo followed by Salmonella Meleagridis. All isolates presented the chromosomal aac(6′)-Iaa gene that confers resistance to aminoglycosides. The antibiotic resistance genes qnrB19, qnrA1, sul2, aph(6)-Id, aph(3)-ld, dfrA1, tetA, tetC, flor2, sul1_15, mph(A), aadA2, blaCARB, and qacE were identified. Ten pathogenicity islands were identified, and the most prevalent plasmid was Col(pHAD28). The main source of Salmonella enterica is the maternity areas, where periparturient shedders are contaminants and perpetuate the pathogen within the dairy in manure, sand, and concrete surfaces. This study demonstrated the necessity of implementing One Health control actions to diminish the prevalence of antimicrobial resistant and virulent pathogens including Salmonella.

As central nervous system (CNS)-related disorders present an increasing cause of global morbidity, mortality, and high pressure on our healthcare system, there is an urgent need for new insights and treatment options. The endocannabinoid system (ECS) is a critical network of endogenous compounds, receptors, and enzymes that contribute to CNS development and regulation. Given its multifaceted involvement in neurobiology and its significance in various CNS disorders, the ECS as a whole is considered a promising therapeutic target. Despite significant advances in our understanding of the ECS’s role in the CNS, its complex architecture and extensive crosstalk with other biological systems present challenges for research and clinical advancements. To bridge these knowledge gaps and unlock the full therapeutic potential of ECS interventions in CNS-related disorders, a plethora of molecular–genetic tools have been developed in recent years. Here, we review some of the most impactful tools for investigating the neurological aspects of the ECS. We first provide a brief introduction to the ECS components, including cannabinoid receptors, endocannabinoids, and metabolic enzymes, emphasizing their complexity. This is followed by an exploration of cutting-edge imaging tools and genetic models aimed at elucidating the roles of these principal ECS components. Special emphasis is placed on their relevance in the context of CNS and its associated disorders.

Introduction: Since 2010, the Ovation Abdominal Stent Graft System has offered a new sealing concept, achieved by a sealing ring filled with polymer 13 mm from the renal arteries. In the latest version, called Ovation Alto, the sealing ring is relocated 6 mm closer to the top of the fabric. This study describes the early clinical outcomes, after CE Mark approval in August 2020, of endovascular aneurysm repair with the Alto endograft. In the last decades, climate change and the rapid urbanization due to the development of the coastal economy have led to biodiversity loss and the fragmentation of habitat in many coastal zones. The presence of protected areas cannot prevent the progress of land degradation. However, these areas are very important because they provide significant ecosystem services and affect local tourism. With regard to increasing adaptation strategies to human pressures and climate change, the present study proposes a detailed monitoring activity and an ecological restoration plan which could improve the resilience of a protected coastal zone in the Pantano forest of Policoro, located on the Ionian coast (southern Italy). In this area, continuous phenomena of intensive deforestation, hydraulic reclamation actions, and fires have reduced the native species of particular naturalistic value, favouring the advancement of desertification, coastal erosion, and saltwater intrusion. The proposed actions are derived from a preliminary analysis on maps, UAV-images, climate data and from meetings with the local community. The operative process detailed in this article could be applied to other protected areas which are subjected to the same phenomena and problems.

Emerging evidence has shown that microRNAs (miRNAs) play critical role in the pathogenesis of several disorders. In the present minireview, we focus our attention on the functional role of a specific miRNA, namely miR-34a, in the pathophysiology of frailty and diabetes mellitus. Based on the current literature, we speculate that this miRNA may serve as a potential biomarker of frailty in diabetic older adults. Additionally, its actions on oxidative stress might represent a druggable target to obtain new potentials treatments.
