Thus, knowing the preferences of the consumers will enhance pedagogical skills.Objective: To characterize pregnant women’s preferences regarding the fitness instructor’s quality characteristics in specific group exercise sessions for this population.Methods: A qualitative study was conducted in 2021. The Fitness Instructor Quality Questionnaire was applied to 29 pregnant women engaged in prenatal group fitness classes, aged between 24 and 48 years. This questionnaire includes 25 items, grouped in two dimensions, scored with a Likert scale with 7 levels of importance (1 = not at all important; 7 = extremely important). Descriptive statistics were used to characterize pregnant women’s opinion (mean, standard deviation, minimum and maximum).Results: The global average of items was 6.41, showing that pregnant women give importance to the Quality of the fitness instructor. Relational Quality has the highest average (6.48), close to Pedagogical-Technical Quality dimension (6.34). The most important items for pregnant women were Ethics (6.76), Communication (6.72), and Availability, Motivation, and Punctuality (6.69). The less important items were Musical Domain (5.24), Image (5.69), and Physical Fitness (5.86).Conclusion: Pregnant women give great importance to the Quality of the fitness instructor, although they value some indicators more than others, as identified in other studies in different populations. Considering the results of this study, it can be recommended that fitness instructors delivering prenatal exercise programs should address these factors related to Quality. It can be emphasized that the fitness instructor should have an intervention that shows willingness to listen to pregnant women, allows them time to perform the exercises, encourages them to practice, without overestimating aspects related to image or fitness level.Keywords: exercise, pregnant, fitness, instructor, quality, group classes, Fitness Instructor Quality Questionnaire

22. Extemporaneously compounded oral medicines in European hospital pharmacies

Pharmaceutical compounding corresponds to the preparation of customised medicines in order to meet the specific needs of patients, which cannot be met by the proprietary medicines provided by the pharmaceutical industry. Historically, pharmaceutical compounding dates back to the very origins of pharmacy and, ever since, it has been an integral part of pharmacy practice. Nevertheless, little is known regarding current compounding practices in Europe and, therefore, the aim of this project was to identify and characterise the oral compounded medicines most frequently dispensed in European hospital pharmacies. The research method adopted was a large-scale, international (European) survey and the research instrument was a self-completion (country-specific) questionnaire. A total of 11 European countries were included in the research: Portugal, UK, Switzerland, Poland, Netherlands, Denmark, Slovenia, Finland, Spain, France and Germany. For most countries, a purposive sample of hospitals was contacted and invited to contribute data regarding the oral compounded medicines most frequently dispensed in their pharmacies. The pilot-study was launched in Portugal but fieldwork was undertaken in most countries. Information regarding legislation, professional organisations and information sources relevant to pharmaceutical compounding was also collected. The oral compounded medicines most frequently dispensed in hospital pharmacy varied considerably throughout Europe, from traditional cachets in Poland to complex tablets in the Netherlands and Denmark. A wide range of active substances, including NTI drugs, and dosage strengths were dispensed. Compounded medicines were prepared individually and also in batches of variable sizes. There is little consistency of compounding practices in Europe and there is a need for common legislation, professional organisations and information sources. This project corresponds to the largest and most complex research in pharmaceutical compounding across Europe and aims to contribute to the harmonisation of quality and safety of compounded medicines in Europe.

The objective of this thesis is to investigate the use of computers with artificial intelligence methods for the teaching of basic literacy skills to be applied eventually to the teaching of illiterate adults in Brazil. The main question addressed in this research, consequently is whether these two apparently conflicting approaches could be combined to create a method that would be technically PA oriented but at the same time could concentrate on meaning by using thematic vocabularies as stimuli for teaching. Would it be possible to find words to illustrate all the phonological features with which a PA method deals using a thematic vocabulary? To answer this question diverse concepts, languages and tools have been developed in order to allow the selection of thematic vocabularies, the description of PA curricula, the distribution of thematic words across PA curricula, the description of teaching activities and the definition of the teaching strategy rules to orient the teaching sequence. The resultant vocabularies have been evaluated and the outcomes of the research have been assessed by literacy experts. A prototype that can be started through the Internet for delivering experimental teaching activities has also been developed and demonstrated.

- Abstract
Objective: To assess the repeatability and suitability for multicentre studies of MScanFit motor unit number estimation (MUNE), which involves modelling compound muscle action potential (CMAP) scans. Methods: Fifteen groups in 9 countries recorded CMAP scans twice, 1–2 weeks apart in healthy subjects from abductor pollicis brevis (APB), abductor digiti minimi (ADM) and tibialis anterior (TA) muscles. The original MScanFit program (MScanFit-1) was compared with a revised version (MScanFit-2), designed to accommodate different muscles and recording conditions by setting the minimal motor unit size as a function of maximum CMAP. Results: Complete sets of 6 recordings were obtained from 148 subjects. CMAP amplitudes differed significantly between centres for all muscles, and the same was true for MScanFit-1 MUNE. With MScanFit-2, MUNE differed less between centres but remained significantly different for APB. Coefficients of variation between repeats were 18.0% for ADM, 16.8% for APB, and 12.1% for TA. Conclusions: It is recommended for multicentre studies to use MScanFit-2 for analysis. TA provided the least variable MUNE values between subjects and the most repeatable within subjects. Significance: MScanFit was primarily devised to model the discontinuities in CMAP scans in patients and is less suitable for healthy subjects with smooth scans.

- Abstract
Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with a lifetime risk of one in 350 people and an unmet need for disease-modifying therapies. We conducted a cross-ancestry genome-wide association study (GWAS) including 29,612 patients with ALS and 122,656 controls, which identified 15 risk loci. When combined with 8,953 individuals with whole-genome sequencing (6,538 patients, 2,415 controls) and a large cortex-derived expression quantitative trait locus (eQTL) dataset (MetaBrain), analyses revealed locus-specific genetic architectures in which we prioritized genes either through rare variants, short tandem repeats or regulatory effects. ALS-associated risk loci were shared with multiple traits within the neurodegenerative spectrum but with distinct enrichment patterns across brain regions and cell types. Of the environmental and lifestyle risk factors obtained from the literature, Mendelian randomization analyses indicated a causal role for high cholesterol levels. The combination of all ALS-associated signals reveals a role for perturbations in vesicle-mediated transport and autophagy and provides evidence for cell-autonomous disease initiation in glutamatergic neurons.

- Abstract
Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease with an estimated heritability between 40 and 50%. DNA methylation patterns can serve as proxies of (past) exposures and disease progression, as well as providing a potential mechanism that mediates genetic or environmental risk. Here, we present a blood-based epigenome-wide association study meta-analysis in 9706 samples passing stringent quality control (6763 patients, 2943 controls). We identified a total of 45 differentially methylated positions (DMPs) annotated to 42 genes, which are enriched for pathways and traits related to metabolism, cholesterol biosynthesis, and immunity. We then tested 39 DNA methylation–based proxies of putative ALS risk factors and found that high-density lipoprotein cholesterol, body mass index, white blood cell proportions, and alcohol intake were independently associated with ALS. Integration of these results with our latest genome-wide association study showed that cholesterol biosynthesis was potentially causally related to ALS. Last, DNA methylation at several DMPs and blood cell proportion estimates derived from DNA methylation data were associated with survival rate in patients, suggesting that they might represent indicators of underlying disease processes potentially amenable to therapeutic interventions.
