Urbanization has reshaped the distribution of biodiversity on Earth, but we are only beginning to understand its effects on ecological communities. While urbanization may have homogenization effects strong enough to blur the large-scale patterns in interaction networks, urban community patterns may still be associated with climate gradients reflecting large-scale biogeographical processes. Using 103 hummingbird-plant mutualistic networks across continental Americas, including 176 hummingbird and 1,180 plant species, we asked how urbanization affects species interactions over large climate gradients. Urban networks were more generalized, exhibiting greater interaction overlap. Higher generalization was also associated with lower precipitation in both urban and natural areas, indicating that climate affects networks irrespective of habitat type. Urban habitats also showed lower hummingbird functional trait diversity and over/underrepresentation of specific clades. From the plant side, urban communities had a higher prevalence of nonnative nectar plants, which were more frequently visited by the hummingbird species occurring in both urban and natural areas. Therefore, urbanization affected hummingbird-plant interactions through both the composition of species and traits, as well as floral resource availability. Taken together, we show that urbanization consistently modifies ecological communities and their interactions, but climate still plays a role in affecting the structure of these novel communities over the scale of continents., Competing Interests: Competing interests statement:The authors declare no competing interest.

Our study identified strains of the A/H5N1 virus in analyzed samples of subsistence poultry, wild birds, and mammals, belonging to clade 2.3.4.4b, genotype B3.2, with very high genetic similarity to strains from Chile, Uruguay, and Argentina. This suggests a migratory route for wild birds across the Pacific, explaining the phylogenetic relatedness. The Brazilian samples displayed similarity to strains that had already been previously detected in South America. Phylogeographic analysis suggests transmission of US viruses from Europe and Asia, co-circulating with other lineages in the American continent. As mutations can influence virulence and host specificity, genomic surveillance is essential to detect those changes, especially in critical regions, such as hot spots in the HA, NA, and PB2 sequences. Mutations in the PB2 gene (D701N and Q591K) associated with adaptation and transmission in mammals were detected suggesting a potential zoonotic risk. Nonetheless, resistance to neuraminidase inhibitors (NAIs) was not identified, however, continued surveillance is crucial to detect potential resistance. Seneca Valley virus (SVV) is a non-enveloped RNA virus and the only member of the Senecavirus A (SVA) species, in the Senecavirus genus, Picornaviridae family. SVV infection causes vesicular lesions in the oral cavity, snout and hooves of pigs. This infection is clinically indistinguishable from trade-restrictions-related diseases such as foot-and-mouth disease. Other clinical manifestations include diarrhoea, anorexia, lethargy, neurological signs and mortality in piglets during their first week of age. Before this study, Chile was considered free of vesicular diseases of swine, including SVV. In April 2022, a suspected case of vesicular disease in a swine farm was reported in Chile. The SVV was confirmed and other vesicular diseases were ruled out. An epidemiological investigation and phylogenetic analyses were performed to identify the origin and extent of the outbreak. Three hundred ninety-five samples from 44 swine farms were collected, including faeces (208), oral fluid (28), processing fluid (14), fresh semen (61), environmental samples (80) and tissue from lesions (4) for real-time RT-PCR detection. Until June 2022, the SVV has been detected in 16 out of 44 farms, all epidemiologically related to the index farm. The closest phylogenetic relationship of the Chilean SVV strain is with viruses collected from swine in California in 2017. The direct cause of the SVV introduction has not yet been identified; however, the phylogenetic analyses suggest the USA as the most likely source. Since the virus remains active in the environment, transmission by fomites such as contaminated feed cannot be discarded. Holoprosencephaly is the most common brain malformation in humans and it is a complex genetic disorder. We report on a patient with holoprosencephaly caused by a rare ZIC2 mutation presenting a bifid nose associated with a nasal fistula and an epidermal cyst, besides hypernatremia. The patient was a 1 year and 4 months old girl that developed an important neuropsychomotor delay. Currently, she uses a wheelchair to move around and only emits sounds. Computed tomography (CT) scan revealed a semilobar holoprosencephaly and a Dandy-Walker variant. Head magnetic resonance imaging also disclosed corpus callosum agenesis and prefrontal subarachnoid space enlargement. On physical examination at 1 year and 4 months of age, we verified growth retardation, microcephaly, bilateral epicantic fold, upslanting palpebral fissures, bifid nose, and limbs spasticity secondary to hypertonia. Later, she began to present hypernatremia; however, its precise cause was not identified. At 6 years and 10 months of age, a nasal fistula was suspected. Facial CT scan showed an epidermal cyst at cartilaginous portion of the nasal septum. High resolution GTG-Banding karyotype was normal. However, molecular analysis through direct sequencing technique showed a mutation at regulatory region of the ZIC2 gene: c.1599*954T > A, a genetic variation previously described only in a Brazilian patient. Our patient presented findings still not reported in literature among patients with holoprosencephaly, including those with ZIC2 mutations. Objective: This study aimed to evaluate the marginal fit of zirconia and lithium disilicate ceramic restorations processed by computer-aided design (CAD)/computer-aided manufacturing (CAM) system., Materials and Methods: Ten models were prepared from a metal die, on which scanning was carried out to obtain the virtual model for the processing of zirconia and lithium disilicate ceramics using the CAD/CAM technique. Gypsum models were fabricated and used for scanning the restorations. The infrastructures were designed using software, and CAD/CAM was used to fabricate the restorations (n = 10). The analysis of the marginal discrepancy was measured at 12 points of the margin using the original metallic die. For each crown, an arithmetic average of the discrepancy values was obtained., Results: The marginal fit of the copings of lithium disilicate was 133.10 ± 26.87 μm and zirconia was 127.34 ± 47.97 μm. There was no statistically significant difference (P = 0.7709) between the materials., Conclusion: The marginal fit is similar between lithium disilicate and zirconia ceramics milled in the CAD/CAM system used., Competing Interests: None

The aim of this study was standardization and application of polymerase chain reaction (PCR) for the detection of contaminants in cell cultures, sera and trypsin. Five PCR protocols were standardized to assess the presence of genetic material from mycoplasma, porcine circovirus 1 (PCV1), bovine leukemia virus (BLV) or bovine viral diarrhea virus (BVDV) in cell culture samples. PCR reactions for the genes GAPDH and beta-actin were used to evaluate the efficiency of nucleic acid extraction. The PCR protocols were applied to 88 cell culture samples from eight laboratories. The tests were also used to assess potential contamination in 10 trypsin samples and 13 fetal calf serum samples from different lots from five of the laboratories. The results showed the occurrence of the following as DNA cell culture contaminants: 34.1% for mycoplasma, 35.2% for PCV1, 23.9% for BVDV RNA and 2.3% for BLV. In fetal calf sera and trypsin samples BVDV RNA and PCV1 DNA was detected. The results demonstrated that cell culture, sera and trypsin used by different laboratories show a high rate of contaminants. Abstract: Generally, natural environments have been transformed into small forest remnants, with the consequent habitat loss and species extinction. The North Paraná State is not an exception, since only 2 to 4% of the original ecosystem occurs in small fragments of Stational Semidecidual Forest. We studied the species richness and abundance of bats in two forest fragments from the Fazenda Congonhas, in Rancho Alegre city, Parana State, Brazil. Four samplings were undertaken in a legally protected native area (107.8 ha) and in a reforested area (11.8 ha) between April 2007 and March 2008. Samplings began at nightfall and lasted six hours,during two consecutive nights in each location. The individuals were captured using eight mist nets, with the same capture effort in both environments. A total of 397 individuals, 14 species and 10 genera were captured in the native area; while in the reforested area, 105 individuals, six species and four genera. Artibeus lituratus was the most common species in both fragments (n = 328, 65.3%), followed by Artibeus fimbriatus (n = 44, 8.8%) and Artibeus jamaicensis (n = 30, 6.0%). Other species including Platyrrhinus lineatus, Carollia perspicillata, Sturnira lilium, Chrotopterus aurintus, Desmodus rotundus, Michronycteris megalotis, Phyllostomus hastatus, Phyllostomus discolor, Myoti levis, Myotis nigricans and Lasiurus blossevillii, accounted for 19.9% of the captures. The native area presented higher values of species richness (S = 14) and diversity (H' = 1.4802) in comparison to the reforested area (S = 6, H '= 0.57015). The t-test evidenced a significant difference between diversity among the sites (t = 7.1075). Chao 1 index indicated that the sampling effort recorded approximately 78% from the total species richness for the native area and 75% for the reforested area. Therefore, the preservation of the forest fragment is essential since it provides habitat for a diverse community of bats. Forest management and reforestation actions may prevent drastic changes in the microclimate of neighboring areas within the forest fragment, and could allow the occupation of available niches in the area, by opportunistic and generalist species.
