Ridomil Gold SL (45.3% a.i. mefenoxam) is a widely used chemical fungicide for the control of oomycetes. However, its impact on fungal communities remains unexplored. Therefore, the goal of this study was to examine the effects of mefenoxam on the temporal dynamics of fungal taxonomic and functional diversities during carrot cultivation under four treatment groups: mefenoxam application with and without Pythium inoculation, and untreated control groups with and without Pythium inoculation. Our in vitro sensitivity assay showed that the maximum recommended concentration of mefenoxam, 0.24 ppm, did not suppress the mycelial growth of P. irregulare. At 100 ppm, mycelial growth was only reduced by 11.4%, indicating that the isolate was resistant to mefenoxam. MiSeq sequencing data revealed transient taxonomic variations among treatments 2 weeks post-treatment. Mortierella dominated the fungal community in the mefenoxam-Pythium combination treatment, as confirmed through PCR using our newly designed Mortierella-specific primers. Conversely, mefenoxam-Pythium combination had adverse effects on Penicillium, Trichoderma, and Fusarium, and decrease the overall alpha diversity. However, these compositional changes gradually reverted to those observed in the control by the 12th week. The predicted ecological functions of fungal communities in all Pythium and mefenoxam treatments shifted, leading to a decrease in symbiotrophs and plant pathogen functional groups. Moreover, the community-level physiological profiling approach, utilizing 96-well Biolog FF microplates, showed discernible variations in the utilization of 95 diverse carbon sources among the treatments. Notably, arbutin, L-arabinose, Tween 80, and succinamic acid demonstrated a strong positive association with Mortierella. Our findings demonstrate that a single application of mefenoxam at its recommended rate triggers substantial taxonomic and functional shifts in the soil fungal community. Considering this impact, the conventional agricultural practice of repeated mefenoxam application is likely to exert considerable shifts on the soil ecosystem that may affect agricultural sustainability.

Carrot (Daucus carota L.) is an important vegetable crop that is rich in carotenoids and is widely cultivated throughout the world. Alternaria leaf blight (ALB), caused by infection with Alternaria dauci (A. dauci), is the most serious fungal disease in carrot production. Although several quantitative trait loci associated with ALB resistance have been identified, the genetic mechanisms underlying this resistance remain largely unelucidated. The aim of the present study was to clarify the infection mode of A. dauci and examine the molecular mechanisms underlying carrot cultivar adaptation to ALB by RNA sequencing. Microscopic observation revealed that A. dauci invades leaf tissues by entering through stomata, and resistant germplasms may significantly inhibit the infection and colonization of A. dauci. In addition, transcriptomic analyses were performed to detect the key pathways and genes associated with the differential responses between ALB-resistant (HB55) and ALB-susceptible (14088) carrot cultivars. These results suggest that the secondary metabolic process, phenylpropanoid biosynthesis, and tyrosine metabolism might play important roles in the resistance of carrots to A. dauci. Three candidate genes (LOC108208301, LOC108215577, and LOC108224339) that were specifically upregulated in the resistant carrot cultivar 'HB55' after A. dauci infection were identified as the key resistance response genes. These findings provide insights into the resistance mechanism of carrots to ALB, as well as key candidate genes and information on expression regulation for the molecular breeding of carrot disease resistance. SUMMARY: Carrot (Daucus carota) is one of the most popular and nutritious vegetable crops worldwide. However, significant yield losses occur every year due to leaf blight, a disease caused by a fungal pathogen (Alternaria dauci). Past research on resistance to leaf blight disease in carrots has been slow because of the low‐quality genome assemblies of both carrot and the pathogen. Here, we report the greatly improved assemblies and annotations of telomere‐to‐telomere (T2T) reference genomes of carrot DH13M14 (451.04 Mb) and A. dauci A2016 (34.91 Mb). Compared with the previous carrot genome versions, our assembly featured notable improvements in genome size, continuity, and completeness of centromeres and telomeres. In addition, we generated a time course transcriptomic atlas during the infection of carrots by A. dauci and captured their dynamic gene expression reprogramming during the interaction process. During infection, A. dauci genes encoding effectors and enzymes responsible for the degradation of plant cell wall components, e.g., cellulose and pectin, were identified, which appeared to increase pathogenic ability through upregulation. In carrot, the coordinated gene expression of components of pattern‐ and effector‐triggered immunity (PTI and ETI) in response to A. dauci attack was characterized. The biosynthesis or signal transduction of plant hormones, including JA, SA, and ethylene, was also involved in the carrot response to A. dauci. This work provides a foundation for understanding A. dauci pathogenic progression and carrot defense mechanisms to improve carrot resistance to leaf blight disease. The Carrot Database (CDB) developed also provides a useful resource for the carrot community. Carrots, a globally cultivated root vegetable crop, are renowned for their nutritional and functional properties. However, the deep utilization and development of carrots and their derived products are limited in numerous countries, with a particular deficiency in advanced deep‐processing and transformation technologies. Consequently, the value of carrot products is diminished and resources are wasted. This review explores the nutritional value and characteristics of different carrot varieties, providing a comprehensive overview of the current state of carrot product processing. These products include primary processed products such as minimally processed carrots, dried carrots, pickles, preserves, fruit purees, yoghurt, juice, and essential oils, as well as processed by‐products including functional phytochemical extractions such as carotenoids, polyphenols, dietary fibers, active peptides, and glycoproteins. This review also covers other by‐products, including feeds, fast food boxes, and bioethanol production. Furthermore, cutting‐edge technologies related to carrot processing are discussed. Gibberellins (GAs) are a class of plant hormones that can affect plant growth and development. GA-oxidases are rate-limiting enzymes, which play a direct role in GA accumulation in plants. However, the roles of GA-oxidase on carrot (Daucus carota L.) taproot development are still unclear. In this study, two GA-oxidase genes, DcGA20ox2 and DcGA2ox1, were identified in carrot. Transgenic carrot plants were obtained by using Agrobacterium-mediated genetic transformation method. The results showed that overexpression of DcGA20ox2 significantly promoted the accumulation of active GAs in carrot, increased plant height, generated more branches, and enhanced xylem development. Overexpression of DcGA2ox1 significantly reduced the total contents of active GAs compared with the control group, resulting in a dwarf phenotype and markedly increased lignin content of the transgenic carrot. The expression profiling showed that the genes of GA metabolic pathway responded to the negative feedback regulation mechanism. At the same time, the expression of most genes in lignin biosynthesis and polymerization process was up-regulated, corresponding to the massive accumulation of lignin. These findings indicated that DcGA20ox2 and DcGA2ox1 affected carrot growth and development by regulating the levels of endogenous GAs. The present study reviews the knowledge to date on the carrot cyst nematode Heterodera carotae, which has become the primary threat to the carrot sector following the withdrawal of the last chemical soil fumigants in Europe. A keyword co-occurrence network was used to structure this review into five research areas: i) biology and epidemiology of H. carotae; ii) molecular identification and phylogeny; iii) population genetics and genomics; iv) control methods and strategies; and v) root exudates and hatching stimulation. Our findings indicate that H. carotae is an under-studied plant-parasitic nematode species, with several identified alternative control methods that require further investigation. Carrot (Daucus carota L.) is a globally significant root vegetable, rich in bioactive and nutritional compounds. Carrot is a crop with a wide range of phenotypic variability including colour, shape, size, and other characteristics. The aim of this work was to determine and compare selected morphological traits and chemical composition of 24 carrot varieties grown under open field conditions in Slovakia. Altogether, 11 qualitative characteristics and 6 quantitative parameters were evaluated. The carrot roots were also analysed for essential chemical constituents including reducing sugars (fructose, glucose, saccharose), carotenoids, dietary fibre, dry matter, ash, and mineral compounds. A wide range of diversity in morphological characteristics and chemical composition indicates a high variability of studied varieties. The root length ranged from 59 to 205 mm, root diameter varied from 17.7 to 39.0 mm and root weight ranged from 21.0 to 74.8 g. Based on the measurement results, saccharose was the dominant sugar ranging from 1.89 to 4.67 g.100 g-1 of fresh weight, carotenoid content varied from 85.94 to 374.24 µg.g-1 of fresh weight, the amount of dietary fibre ranged from 25.67 to 36.02% of dry matter. Considerable differences were also observed in the content of macro and microelements [mg.100 g-1] ranging as follows: P (17.5 - 52.0), K (110.0 - 337.0), Na (65.9 - 186.0), Fe (0.324 - 3.340), Cu (0.042 - 0.126), Zn (0.152 - 0.945) and Mn (0.062 - 0.189). The presented results can provide useful information for carrot growers and consumers in terms of morphological characteristics as well as the chemical composition of individual carrot varieties. The study aims to compare and analyze the levels of heavy metal contaminants in Solanum tuberosum (Irish Potato) and Daucus carota (Carrot) in Bokkos Local Government Area Plateau State, Nigeria, assessing potential health risks associated with consuming these crops. The research was focused to five heavy metals (Cd, Cr, Cu, Ni, and Pb) in Irish potato and carrot sample from five different farms. The concentrations of these metals were gotten in decreasing order as follows: (DIM) Irish potato (A) Cd > Pb > Cr > Ni; (EDI) Irish potatoes (C) Cu > P B > Cr B > Ni B > Cd; (DI) carrot (A), Cu > B > Cu B > P Ni; and (DIMA) carrot(A) Pb Cd Cu Pb. The health risk assessment indices for the populations through the consumption of contaminated fish were estimated based on the daily intake of metals relative to the reference oral dose (RfD) for each metal. The mean concentration of Pb, Cd, Ni and Cd in tubers was less than the recommended values of FAO/WHO. The risk analysis factors considered were transfer factor (TF), EDI, DIM, bioaccumulation factor (BF), target hazard quotient (THQ), and Health risk index (HRI) evaluated for this study showed moderate to less high risk levels with respect to human exposure through consumption of the vegetables and not the soil as may be indicate in transfer factor. The current studies were carried out to explore antifungal potential of bacteria isolated from rhizospheric region of Daucus carota against Rhizoctonia. Of the total 21 bacterial isolates isolated, only 3 isolates namely CVO-3, CVO-4 and CRY-O-2 were shown antifungal potential against Rhizoctonia. Besides having antifungal potential these isolates also have some plant growth promoting attributes that will be an extra benefit of using these isolates as a biofungicides against R. solani. Thus, these isolates could be used as biofungicides against Rhizoctonia to prevent a variety of plant diseases like sheath blight in Oryza sativa, stem canker and black scurf in Solanum tuberosum and seedling diseases in Brassica napus. The isolates CVO-3, CVO-4 and CRY-O-2 were also characterized biochemical level by using methods based on Determinative Bacteriology given in Bergey’s manual and at molecular level using 16S rRNA sequencing and sequences of 16S rDNA segment were submitted to NCBI in GenBank database with the accession numbers OR775574 (Lysinibacillus boronitolerans ss3cvo3), OR775575 (Lysinibacillus boronitolerans ss4cvo4) and OR708523 (Shouchella rhizosphaerae ss1cryo2). The present investigation is an approach towards the use of these bacterial isolates from carrot rhizosphere to control the activities in the rhizosphere of other cropping plants.

Carrot (Daucus carota L. subsp. sativus) cultivation is important in Argentina, with the Garay department in Santa Fe province being a key production area. In recent years, dry rot symptoms have afflicted various carrot cultivars, resulting in substantial field losses and postharvest discards. Several soilborne pathogens, including Fusarium species, have been implicated in similar carrot diseases globally. This study aimed to identify the specific causal agent responsible for dry rot in carrots in central Argentina to enable targeted disease management strategies. Samples of symptomatic carrots were collected from fields in the Garay department. Fungal isolates were obtained from the infected tissue and subjected to morphological and molecular analyses. Pathogenicity tests were conducted to confirm the ability of the isolates to cause disease symptoms in carrots. Based on morphological and molecular identification, isolates were identified as Fusarium nirenbergiae, Fusarium triseptatum and Fusarium solani. Pathogenicity tests demonstrated that the three Fusarium species were capable of causing dry rot symptoms in carrots. This study represents the first report of Fusarium nirenbergiae, Fusarium triseptatum and Fusarium solani causing dry rot in carrots in Argentina. [ABSTRACT FROM AUTHOR]

The lipoxygenase cascade in plants is a source of oxylipins (oxidized fatty acid derivatives), which play an important role in regulatory processes and formation of plant response to stress factors. Some of the most common enzymes of the lipoxygenase cascade are 13-specific hydroperoxide lyases (HPLs, also called hemiacetal synthases) of the CYP74B subfamily. In this work, we identified and cloned the CYP74B34 gene from carrot (Daucus carota L.) and described the biochemical properties of the corresponding recombinant enzyme. The CYP74B34 enzyme was active towards 9- and 13-hydroperoxides of linoleic (9-HPOD and 13-HPOD, respectively) and α-linolenic (9-HPOT and 13-HPOT, respectively) acids. CYP74B34 specifically converted 9-HPOT and 13-HPOT into aldo acids (HPL products). The transformation of 13-HPOD led to the formation of aldo acids and epoxyalcohols [products of epoxyalcohol synthase (EAS) activity] as major and minor products, respectively. At the same time, conversion of 9-HPOD resulted in the formation of epoxyalcohols as the main products and aldo acids as the minor ones. Therefore, CYP74B34 is the first enzyme with a double HPL/EAS activity described in carrot. The presence of these catalytic activities was confirmed by analysis of the oxylipin profiles for the roots from young seedlings and mature plants. In addition, we substituted amino acid residues in one of the catalytically essential sites of the CYP74B34 and CYP74B33 proteins and investigated the properties of the obtained mutant enzymes. Liposomes coated with chitosan by ionic gelation with tripolyphosphate (TPP-chitosomes) are interesting particles for stabilizing active compounds. However, the encapsulation condition must be optimized. The aim of this study was to optimize the encapsulation of phenolics and carotenoids of carrot pomace in TPP-chitosomes by using a Central Composite Design 23 and response surface methodology. The independent variables were the phospholipid (0.8–4.2 mg/mL), chitosan (2.6–9.4 mg/mL), and carrot pomace (4–14 g/100 mL of ethanol) concentrations; the responses were the encapsulation efficiency in TPP-chitosomes (EE) of phenolics, a-carotene, and b-carotene and the particle size and zeta potential of the particles. The zeta potential ranged from +17 to +37 mV, indicating that the liposomes were coated with chitosan and that the particle sizes were in the nanometric to submicrometric scale. The optimized condition for encapsulating carotenoids was 2.5 mg/mL phospholipids, 6.0 mg/mL chitosan, and 12 g of carrot pomace/100 mL of ethanol. In this condition, the EE of phenolics and α- and β-carotene was 95%, 98%, and 99%, respectively. Therefore, TPP-chitosomes containing encapsulated phenolics and carotenoids, which can be obtained from agro-industrial by-products, have potential application as natural pigments in food or cosmetics. The purple carrot cultivar 'Purple Sun' (Daucus carota L.) is characterized by a relevant content of phenolic compounds and anthocyanins, which may play an important role in reducing the risk of chronic diseases and in the treatment of metabolic syndrome. In the present study, the genetic diversity, phytochemical composition, and bioactivities of this outstanding variety were studied for the first time. Genetic analysis by molecular markers estimated the level of genetic purity of this carrot cultivar, whose purple-pigmented roots were used for obtaining the purple carrot ethanol extract (PCE). With the aim to identify specialized metabolites potentially responsible for the bioactivities, the analysis of the metabolite profile of PCE by LC-ESI/LTQ Orbitrap/MS/MS was carried out. LC-ESI/HRMS analysis allowed the assignment of twenty-eight compounds, putatively identified as isocitric acid (1), phenolic acid derivatives (2 and 6), hydroxycinnamic acid derivatives (9, 10, 12–14, 16, 17, 19, 22, and 23), anthocyanins (3–5, 7, 8, 11, and 18), flavanonols (15 and 21), flavonols (20 and 24), oxylipins (25, 26, and 28), and the sesquiterpene 11-acetyloxytorilolone (27); compound 26, corresponding to the primary metabolite trihydroxyoctanoic acid (TriHOME), was the most abundant compound in the LC-ESI/HRMS analysis of the PCE, and hydroxycinnamic acid derivatives followed by anthocyanins were the two most represented groups. The antioxidant activity of PCE, expressed in terms of reactive oxygen species (ROS) level and antioxidant enzymes activity, and its pro-metabolic effect were evaluated. Moreover, the antibacterial activity on Gram (−) and (+) bacterial strains was investigated. An increase in the activity of antioxidant enzymes (SOD, CAT, and GPx), reaching a maximum at 0.5 mg/mL of PCE with a plateau at higher PCE concentrations (1.25, 2.5, and 5.0 mg/mL), was observed. PCE induced an initial decrease in ROS levels at 0.1 and 0.25 mg/mL concentrations, reaching the ROS levels of control at 0.5 mg/mL of PCE with a plateau at higher PCE concentrations (1.25, 2.5, and 5.0 mg/mL). Moreover, significant antioxidant and pro-metabolic effects of PCE on myoblasts were shown by a reduction in ROS content and an increase in ATP production linked to the promotion of mitochondrial respiration. Finally, the bacteriostatic activity of PCE was shown on the different bacterial strains tested, while the bactericidal action of PCE was exclusively observed against the Gram (+) Staphylococcus aureus. The bioactivities of PCE were also investigated from cellular and molecular points of view in colon and hematological cancer cells. The results showed that PCE induces proliferative arrest and modulates the expression of important cell-cycle regulators. For all these health-promoting effects, also supported by initial computational predictions, 'Purple Sun' is a promising functional food and an optimal candidate for pharmaceutical and/or nutraceutical preparations. Post‐harvest diseases cause significant economic losses in the carrot production chain. In this study, storage losses and fungal pathogens causing them were analysed in the carrot yield from 52 different field plots in four areas in Finland in 3 years (2016–2018). Over 30,000 carrots were sampled and analysed at three time points during cold storage at 0–1°C. In March, after 5–6 months' storage, the average loss due to diseases was 20%–21% every year. Decay of the root tip was the most common disease symptom, followed by pits on the side and black rot in the crown, detected in 69.2%, 15.0% and 9.0% of the symptomatic samples, respectively. Both intensive carrot cultivation practice and early timing of harvest increased storage losses. Pathogens in 3057 symptomatic carrot tissue samples were isolated by culturing, and fungal species were identified. The most common fungal species detected were Mycocentrospora acerina, Botrytis cinerea and Fusarium spp., especially F. avenaceum. However, the frequency of different pathogens varied between the different years and time points during storage. Species‐specific PCR tests revealed that M. acerina and F. avenaceum were present in many early time‐point samples where they could not yet be detected by the culturing method. In Finland, this study on carrot post‐harvest diseases is the first large‐scale survey in which the fungal pathogens were isolated and identified by laboratory tests. The article investigates the antifungal potential of bacterial isolates from the rhizosphere of Daucus carota against the plant pathogens Fusarium udum and Fusarium oxysporum. Topics include the impact of Fusarium infections on crops and the production of mycotoxins, the challenges in managing Fusarium due to its persistence in soil and host tissue, and the potential of using bacterial isolates as biocontrol agents instead of chemical fungicides.

Abstract Background Carrot is an important vegetable crop grown worldwide. The major economic problem in carrot cultivation is yellow disease caused by Bactericera trigonica, which induces biotic stress and has the greatest impact on crop productivity. Comprehensive studies on the mechanism of carrot defense response to biotic stress caused by B. trigonica infestation have yet to be conducted. Methods The changes in photosynthetic pigments, proline, TPC, H2O2 and MDA content, DPPH radical scavenging ability, and antioxidant enzyme activity of SOD, CAT, and POX in carrot leaves in response to insect sex (female and male), rapid response (during the first six hours), and long-term response to B. trigonica infestation were evaluated. Results The results of our study strongly suggest that B. trigonica infestation causes significant changes in primary and secondary metabolism and oxidative status of carrot leaves. Photosynthetic pigment content, TPC, and DPPH and CAT activities were significantly reduced in carrot leaves in response to insect infestation. On the other hand, proline, H2O2 content, and the activity of the antioxidant enzymes superoxide dismutase and peroxidase were increased in carrot leaves after B. trigonica infestation. The results indicate that B. trigonica attenuates and delays the oxidative stress responses of carrot, allowing long-term feeding without visible changes in the plant. Carrot responded to long-term B. trigonica infestation with an increase in SOD and POX activity, suggesting that these enzymes may play a key role in plant defense mechanisms. Conclusions This is the first comprehensive study strongly suggesting that B. trigonica infestation causes significant changes in primary and secondary metabolism and an attenuated ROS defense response in carrot leaves that enables long-term insect feeding. The information provides new insights into the mechanisms of carrot protection against B. trigonica infestation.

Infection with the bacterium 'Candidatus Liberibacter solanacearum' (Lso) is suspected to cause severe damage in carrot leading to high carrot weight loss. This study investigates three main aspects: (i) whether there is a reduction of carrot root weight under field conditions due to Lso infection; (ii) the correlation between Lso infection rate in carrot plants and occurrence of the psyllid Trioza apicalis as the known vector for Lso in carrot, and (iii) the comparison between symptoms described in literature and observed symptoms associated with Lso infection. Therefore, field surveys were conducted from 2018 to 2021 on organically managed carrot fields in Lower Saxony, Germany. Two Rebell orange sticky traps were placed per field replaced on a weekly basis. Captured T. apicalis were morphologically identified and counted. Carrot plant samples were collected from T. apicalis infested fields. Discolouration of foliage and further symptoms on carrots were investigated. For detection of Lso in carrot plants and psyllids, the samples were analysed using PCR. The infection rate of carrot plants varied between 2.5% and 80% per field with low abundance of T. apicalis. Of the asymptomatic plants, 39.4% were infected with Lso, while 80.9% of the symptomatic plants were infected with Lso. The weight of Lso-positive carrot samples was not significantly reduced compared to Lso-negative samples. No economically relevant losses of carrot yield were reported by farmers participating in this study. This indicates that there was no effect of Lso infection on carrot root weight during this study. Of the symptoms associated with a Lso infection in carrots, only leaf discolouration could be confirmed. Background: Carrot is an important vegetable crop grown worldwide. The major economic problem in carrot cultivation is yellow disease caused by Bactericera trigonica, which induces biotic stress and has the greatest impact on crop productivity. Comprehensive studies on the mechanism of carrot defense response to biotic stress caused by B. trigonica infestation have yet to be conducted. Methods: The changes in photosynthetic pigments, proline, TPC, H2O2 and MDA content, DPPH radical scavenging ability, and antioxidant enzyme activity of SOD, CAT, and POX in carrot leaves in response to insect sex (female and male), rapid response (during the first six hours), and long-term response to B. trigonica infestation were evaluated. Results: The results of our study strongly suggest that B. trigonica infestation causes significant changes in primary and secondary metabolism and oxidative status of carrot leaves. Photosynthetic pigment content, TPC, and DPPH and CAT activities were significantly reduced in carrot leaves in response to insect infestation. On the other hand, proline, H2O2 content, and the activity of the antioxidant enzymes superoxide dismutase and peroxidase were increased in carrot leaves after B. trigonica infestation. The results indicate that B. trigonica attenuates and delays the oxidative stress responses of carrot, allowing long-term feeding without visible changes in the plant. Carrot responded to long-term B. trigonica infestation with an increase in SOD and POX activity, suggesting that these enzymes may play a key role in plant defense mechanisms. Conclusions: This is the first comprehensive study strongly suggesting that B. trigonica infestation causes significant changes in primary and secondary metabolism and an attenuated ROS defense response in carrot leaves that enables long-term insect feeding. This study conducted in Gasabo District, Nyacyonga marshland aimed to investigate the effect of water hyacinth (Eichhornia crassipes) organic fertilizer on the vegetative growth of the Royal Chantenay variety of carrot (Daucus carota). The experiment was conducted using a randomized complete block design (RCBD) with three replications. Four treatments were applied: T1 (control, no fertilizer), T2 (NPK 17-17-17), T3 (25% water hyacinth organic fertilizer + 75% NPK 17-17-17), and T4 (50% water hyacinth organic fertilizer + 50% NPK 17-17-17). The growth parameters assessed included plant height, leaf number, and root diameter. Results showed that the application of water hyacinth organic fertilizer significantly influenced the vegetative growth of Royal Chantenay carrot plants. T3 and T4 treatments enhanced plant height, leaf number, and root diameter compared to the control (T1) and NPK 17-17-17 (T2). The highest vegetative growth parameters were observed in T4, indicating that a higher concentration of water hyacinth organic fertilizer positively affected the growth of carrot plants. The findings suggest that water hyacinth organic fertilizer can be a viable alternative to chemical fertilizers in promoting the vegetative growth of Royal Chantenay carrot plants. The utilization of water hyacinth as an organic fertilizer can contribute to sustainable agricultural practices by reducing dependence on synthetic inputs.

This study evaluated the effect of carrot flour (CF; Daucus carota L.) as a partial substitute for the basal diet of Cobb 500 broiler chicken on skin pigmentation, production parameters and profitability. A total of 64 chickens were separated according to sex and assigned to four diets, with four replicates of two birds each. The control groups were fed a standard diet, and the experimental groups were fed the same conventional diet with replacement levels of 10, 15, and 20% CF. No significant differences were observed in feed consumption, weight gain or feed conversion ratio. However, diets with carrot flour improved feed intake, promoted weight gain and optimized feed efficiency with 10% in males and 15% in females. No mortality was recorded in the experimental groups. The intensity of pigmentation in the chickens’ skin increased as the level of substitution increased, while sex influenced color expression. Diets with CF improved profitability especially in males. In conclusion, the replacement of the basal diet with CF in the diet of Cobb 500 broilers had positive effects on the evaluated parameters, therefore, it can be used as an alternative source of natural pigment in the production of yellow-skinned broilers. Daucus carota L., a member of the Apiaceae family, comprises 13 subspecies, with one being cultivated (D. carota L. ssp. sativus (Hoffm.) Arcang.) and the remaining being wild. Traditionally, the wild carrot has been recognized for its antilithic, diuretic, carminative, antiseptic, and anti-inflammatory properties and has been employed in the treatment of urinary calculus, cystitis, gout, prostatitis, and cancer. While extensive literature is available on the phytochemical, pharmacological, and therapeutic evaluations of the cultivated carrot, limited information has been published on the wild carrot. A thorough search was conducted on the phytochemical composition, folk-medicine uses, and pharmacological properties of wild carrot subspecies (Daucus carota L. ssp. carota). Various electronic databases were consulted, and the literature spanning from 1927 to early 2023 was reviewed. Thirteen wild Daucus carota subspecies were analyzed, revealing over 310 compounds, including terpenoids, phenylpropenoids, flavonoids, and phenolic acids, with 40 constituting more than 3% of the composition. This review also highlights the antioxidant, anticancer, antipyretic, analgesic, antibacterial, antifungal, hypolipidemic, and hepato- and gastroprotective properties of wild carrot subspecies. Existing in vitro and in vivo studies support their traditional uses in treating infections, inflammation, and cancer. However, further research on other subspecies is required to confirm additional applications. Carrots as a representative type of delicious and healthy vegetables has become increasingly popular due to its unique flavors and nutritional value. Vast amount of research has been completed on chemical composition and aroma profile of fresh carrot. However, the knowledge of fresh carrot is only scattered. Therefore, a more comprehensive review to fresh carrot is requisite to be reported. This review first describes the botany of carrots; then, the chemical ingredients and their various functions are discussed. The volatile constituents and the effects of terpenoids on aroma profiles of fresh carrot are also discussed. Finally, sources of variation in the formation and development of carrot aroma through heat processing and dehydration are analyzed. This information will further help researchers to solve the problem of insufficient aroma of carrot products and standardize for each type of carrot through improving the production process.

Liposomes coated with chitosan by ionic gelation with tripolyphosphate (TPP-chitosomes) are interesting particles for stabilizing active compounds. However, the encapsulation condition must be optimized. The aim of this study was to optimize the encapsulation of phenolics and carotenoids of carrot pomace in TPP-chitosomes by using a Central Composite Design 23 and response surface methodology. The independent variables were the phospholipid (0.8–4.2 mg/mL), chitosan (2.6–9.4 mg/mL), and carrot pomace (4–14 g/100 mL of ethanol) concentrations; the responses were the encapsulation efficiency in TPP-chitosomes (EE) of phenolics, a-carotene, and b-carotene and the particle size and zeta potential of the particles. The zeta potential ranged from +17 to +37 mV, indicating that the liposomes were coated with chitosan and that the particle sizes were in the nanometric to submicrometric scale. The optimized condition for encapsulating carotenoids was 2.5 mg/mL phospholipids, 6.0 mg/mL chitosan, and 12 g of carrot pomace/100 mL of ethanol. In this condition, the EE of phenolics and α- and β-carotene was 95%, 98%, and 99%, respectively. Therefore, TPP-chitosomes containing encapsulated phenolics and carotenoids, which can be obtained from agro-industrial by-products, have potential application as natural pigments in food or cosmetics. TPP-chitosomes can also be used to encapsulate other types of natural pigments.

Introduction: Medicinal plants, including spontaneous or cultured herbaceous and forest products, represent an inexhaustible source of traditional and effective remedies thanks to their active major compounds. The present work consists of an ethnobotanical study of three species namely, Daucus carota, Dittrichia viscosa, and Salvia officinalis, commonly used in Taounate region (Northern Morocco) to treat various diseases. Methods: An ethnopharmacological survey was conducted in Taounate region during a period of three months from January to March 2022, using semi-structured individual interviews. Then, the collected data were analyzed statistically using Microsoft Office software "Excel 2013" and System Package for Social Sciences (SPSS). Results: Leaves, flowers, and stems were the most common parts used to prepare traditional remedies. Decoction, infusion, and cataplasm were the most used preparation methods, and the oral route was the most common method of administration for the studied plants. Moreover, the plants intervened in the treatment of digestive, genito-urinary, dermatological, neurological, and metabolic diseases. Conclusion: Information collected during this study shed light on the interesting know-how in traditional herbal medicine in the study area, and on the frequent use of medicinal plants as an alternative to synthetic drugs by the population of Taounate, to treat different diseases. Thereafter, the study’s results can constitute an important database for pharmacologists, phytochemists, toxicologists, and clinical researchers for the development of new drugs based on natural substances.

This study is based on an investigation of the transport phenomenon, specifically the quantification of arsenic diffusion in carrots within a temperature range of 89 °C–99 °C using a thin plate model. Studying the diffusion of arsenic in carrots is important due to its toxicity, as it can concentrate during cooking. The World Health Organization considers arsenic as one of the ten chemical substances of public health concern. In this study, biennial hybrid carrots of the Nantesa variety were cooked whole with their epidermis in an aqueous solution containing diarsenic trioxide with an As concentration of 5 mgL−1 at 89 °C, 94 °C, and 99 °C. The cooking times of the carrots at different temperatures were based on a specific degree of tenderness, with a value of ≤3 kg m−2. The evaluated data showed consistency with increasing temperature. The calculated effective diffusion coefficients at temperatures of 89 °C, 94 °C, and 99 °C were 5.84E-09 m2s-1, 1.08E-08 m2s-1, and 2.51E-08 m2s-1 for the flesh (DL), and 1.601E-11 m2s-1, 2.15E-11 m2s-1, and 4.39E-11 m2s-1 for the epidermis (DE), respectively. The activation energy for diffusion was determined to be 159.54 kJmol−1 for the and 110.68 kJmol−1 for the epidermis. Similar behaviours were observed in different radial positions of the carrot, where the arsenic content decreased from the periphery to the centre, consistent with studies on diffusion phenomena with other solutes in food. The novelty was the detailed quantification of arsenic diffusion in the Nantes-type hybrid carrot matrix. This study is limited to a specific concentration of 5 mgL−1 of arsenic solution. The findings of this study may have significant implications for public health and food safety.

Sensory parameters as well as the volatile and non-volatile compound profiles of sixteen carrot cultivars were recorded to obtain insight into consumer preference decisions. The sensory test was carried out with a consumer panel of 88 untrained testers allowing a clear acceptance-based differentiation of the cultivars. Five individual sensory characters (sweetness, overall aroma, bitterness, astringency and off-flavor) supported this discrimination. Chemical analyses of volatile organic compounds, polyacetylenes, phenylpropanoids and sugars enabled us to correlate the influence of these ingredients on sensory perception. Higher concentrations of α-pinene, hexanal, styrene and acetophenone correlated with a better acceptance, as well as sweetness and overall aroma perception. In contrast, a low acceptance as well as a stronger perception of bitterness, astringency and off-flavor correlated with enhanced concentrations of camphene, bornylacetate, borneol, myristicine, falcarindiol, falcarindiol-3-acetate, laserin and epilaserin. Premise: Cultivated species and their wild relatives often hybridize in the wild, and the hybrids can survive and reproduce in some environments. However, it is unclear whether cultivar alleles are permanently incorporated into the wild genomes or whether they are purged by natural selection. This question is key to accurately assessing the risk of escape and spread of cultivar genes into wild populations. Methods: We used genomic data and population genomic methods to study hybridization and introgression between cultivated and wild carrot (Daucus carota) in the United States. We used single nucleotide polymorphisms (SNPs) obtained via genotyping by sequencing for 450 wild individuals from 29 wild georeferenced populations in seven states and 144 cultivars from the United States, Europe, and Asia. Results: Cultivated and wild carrot formed two genetically differentiated groups, and evidence of crop–wild admixture was detected in several but not all wild carrot populations in the United States. Two regions were identified where cultivar alleles were present in wild carrots: California and Nantucket Island (Massachusetts). Surprisingly, there was no evidence of introgression in some populations with a long‐known history of sympatry with the crop, suggesting that post‐hybridization barriers might prevent introgression in some areas. Conclusions: Our results provide support for the introgression and long‐term persistence of cultivar alleles in wild carrots populations. We thus anticipate that the release of genetically engineered (GE) cultivars would lead to the introduction and spread of GE alleles in wild carrot populations. [ABSTRACT FROM AUTHOR]

Bioethanol is a liquid biofuel produced from the digestion of biomass and usable waste of organic origin. The objective of this research was to obtain bioethanol from carrot (Daucus carota) residues of the Peruvian Chantenay variety, with a high content of lignocellulosic substances. The in-batch process method of enzymatic hydrolysis, with Aspergillus niger amyloglucosidase, and fermentation, with Saccharomyces cerevisiae yeast, was applied. The ferment was steam distilled and chemically characterized. The process was evaluated by controlling pH and enzyme/yeast mass ratio through the response surface optimization. The optimum conditions for the best values of TSS and % ethanol content for the distilled product were a time of 300 min, yeast/enzyme mass ratio of 24.0, and pH of 4.98. The results showed a significant decrease in sugars in the hydrolysis and fermentation stages, optimum alcohol content in the distilled product of 92.48% (v/v), lower organic compound content, and net calorific value of 23.82 MJ/kg, which is higher than those reported in the literature. Beta-carotene and lycopene are important carotenoids present in carrots (Daucus carota). In this study, a fast and efficient HPLC method was developed for the determination of beta-carotene and lycopene from Daucus carota. The method involved extraction of the carotenoids from the vegetable with acetone, followed by separation on a C30 column using a mobile phase of acetonitrile-methanolwater. The detection was carried out at 470 nm for beta-carotene and lycopene. The method was validated in terms of linearity, precision, accuracy, and sensitivity. The developed method was found to be sensitive, specific, and precise, with a limit of detection of 0.05 µg/mL for beta-carotene and lycopene. The highest concentration of beta-carotene content is found in the Atomic Red variety (8.5 mg/100g) and the lowest in the Purple Haze F1 hybrid (2.2 mg/100g). On the other hand, the content of lycopene in the tested samples ranged from 0.05 to 0.1 mg/100 g, with the highest content found in the Atomic Red variety and the lowest in the Danvers variety. The method was successfully applied to the determination of beta-carotene and lycopene in Daucus carota samples.

Many unit operations in the food industry are diffusional driven. These processes are usually very slow and difficult to handle for specific groups of raw materials. Vacuum impregnation (VI) is one example. Impregnating low-porous or densely-structured materials is problematic and often requires low pressure, which can negatively affect product quality and be expensive in energy consumption. This research aimed to evaluate ultrasound (US) as a factor in intensifying mass transfer and enhancing its effectiveness in the VI process. Experiments on impregnation enhanced with ultrasound applied at different stages of the process were carried out. Carrot, a difficult-to-process raw material, was impregnated with ascorbic acid as a mass transfer marker. The process’s effectiveness and selected quality parameters were then analyzed. Ultrasound was found to have a positive influence on mass transfer during VI. The effects of ultrasound enhancement were different for particular processes, and depended on the stage of the application and duration of US exposure. The greatest increase in the tissue’s ascorbic acid content (60% compared to the non-ultrasound-assisted process) was observed when ultrasound was applied continuously throughout the process. Applying ultrasound only during the relaxation (at atmospheric pressure) or aeration periods resulted in a similar effect – c.a. 20% increase in the marker’s content. The smallest increase (10%) was observed when ultrasound was applied only during the vacuum period. Applying US did not result in any unfavorable color change. In most cases, pH decreased, which is favorable for the semi-product’s stability. The carotenoid and phenolic compounds’ content did not decrease. The results unequivocally indicate that ultrasound has great potential for use as a mass transfer accelerator in the VI process for low porosity materials. The effectiveness of the US is influenced not only by pressure but also by exposure duration. The synergistic effect observed using ultrasound-enhanced impregnation throughout the process confirmed this hypothesis.

Introduction: Sustainable agricultural practices for controlling crop pests are urgently needed to reduce the reliance on chemical pesticides, which have long-term detrimental effects on ecosystems. In this study, we assessed the effectiveness of arbuscular mycorrhizal fungi (AMF) and vermicompost (Vc) supplementation, alone and in combination, in mitigating the negative impacts of Meloidogyne incognita infestation on carrot (Daucus carota L.) growth, development, and physiology. Methods: We measured different plant growth parameters such as plant height and biomass accumulation, several plant physiological parameters such as the levels of photosynthetic pigments, phenolics, and the activity of defense enzymes such as peroxidases and polyphenol oxidases, and evaluated the severity of Meloidogyne incognita nematode infestation on plants treated or not treated with vermicompost (Vc) and/or arbuscular mycorrhizal fungi (AMF). Results: Our findings show that M. incognita significantly affects plant growth, biomass accumulation, and photosynthetic pigment and carotenoid content. The incorporation of Vc and AMF into the soil, either individually or in combination, significantly alleviates the negative effects of nematode infestation on carrot plants. This was accompanied by the induction of phenolic compounds and defense enzymes such as peroxidases (+15.65%) and polyphenol oxidases (29.78%), and by a reduction in the severity of nematode infestation on Vc and AMF-treated plants compared to nematode-infested plants. Principal component analysis (PCA) shows significant correlations between various of the studied parameters. In particular, we observed negative correlations between the application of AMF and Vc alone and in combination and disease severity, and positive correlations between plant growth, photosynthetic pigments phenol content, and activity of defense enzymes. Discussion: Our study highlights the relevance of cultural practices and beneficial microorganisms for the sustainable and environmentally friendly management of agricultural pests. Carrot (Daucus carota L.) seed quality is affected by umbel position due to uneven maturation of carrot seeds produced in different umbel orders. However, keeping this in view, we tested whether seed quality could be improved with the suppression of tertiary umbels under exogenous auxin application. Using auxin-producing bacterial isolates, i.e., Bacillus sp. MN54, Enterobacter sp. MN17, Pantoea sp. MN34, and Burkholderia phytofirmans PsJN, the arrangements of carrot umbel order were evaluated in terms of quality carrot seed production. The results revealed that auxin production by plant-growth-promoting rhizobacteria showed significant differences among measured growth indices, yield, and seed quality attributes. The selected endophytic strains co-applied with auxin via foliar application improved all growth- and yield-related traits, as well as the enzymatic activities of carrots. Noticeably, MN17+L-tryptophan and MN34+L-tryptophan effectively minimized the number of tertiary umbels by increasing the number of secondary umbels. Furthermore, treating with PsJN+L-tryptophan and MN34+L-tryptophan resulted in reduced conductivity of seed leachates and malondialdehyde levels in primary, secondary, and tertiary umbel seeds. These findings collectively indicate the potential of the foliar application of PsJN+L-tryptophan and MN34+L-tryptophan to effectively alter umbel arrangement, leading to improved yield and seed quality. This study implies that carrot seed producers can consider employing specific PGPB strains, particularly MN34+L-tryptophan, to suppress tertiary umbels and achieve higher yields of high-quality carrot seeds. Main conclusion: DcMYB11, an R2R3 MYB gene associated with petiole anthocyanin pigmentation in carrot, was functionally characterized. A putative enhancer sequence is able to increase DcMYB11 activity. The accumulation of anthocyanin pigments can exhibit different patterns across plant tissues and crop varieties. This variability allowed the investigation of the molecular mechanisms behind the biosynthesis of these pigments in several plant species. Among crops, carrots have a well-defined anthocyanin pigmentation pattern depending on the genic background. In this work, we report on the discovery of DNA structural differences affecting the activity of an R2R3 MYB (encoded by DcMYB11) involved in anthocyanin regulation in carrot petiole. To this end, we first verified the function of DcMYB11 using heterologous systems and identified three different alleles which may explain differences in petiole pigmentation. Characterization of the DcMYB11 alleles at the 5' upstream sequence unveiled a sequence that functions as a putative enhancer. In conclusion, this study provides novel insight into the molecular mechanisms controlling anthocyanin accumulation in carrot. Three garden vegetables—radish, carrot and lettuce—were cultivated in a pot experiment using two soils from the Příbram area polluted mainly by cadmium (Cd), zinc (Zn), lead (Pb) and chromium (Cr). The soils of the Příbram district, Czech Republic, are heavily polluted as a result of the atmospheric deposition of toxic elements originating from historic lead–silver mining and smelting activities. The results showed that lettuce absorbed the highest amounts of toxic elements (Cd 28 and 30, Cr 12 and 13, Zn 92 and 205 mg·kg−1 DW), except Pb, which was higher in radish (30 and 49 mg·kg−1 DW). Changes in macronutrient contents in edible parts were not found, except for sulfur. A higher total free amino acids (fAAs) accumulation was shown in all vegetables in more contaminated soil, with the highest fAA content being in radish. A group of essential fAAs reached 7–24% of total fAAs in vegetables. The risk to human health was characterized using the target hazard quotient and total hazard index (HI). The cumulative effect of the consumption of vegetables with HI > 1 showed possible non-carcinogenic health effects for lettuce and carrot. HI decreased in the order Cd > Pb > Cr > Zn. The carcinogenic risk of toxic elements decreased in the order Cd > Cr > Pb (0.00054, 0.00026, 0.00003). These values showed a carcinogenic risk from the consumption of lettuce and carrot and confirmed that the adult population of the studied area is at high risk if lettuce and carrot cultivated in this area are consumed daily. A great challenge of scientific research with carrot in semi-arid regions is obtaining the best productive characteristics and economic conditions by optimizing the green manure amounts using spontaneous species from the Caatinga biome, such as hairy woodrose (Merremia aegyptia L.) and roostertree (Calotropis procera Ait.). Therefore, this work aimed to agronomically and economically optimize the production of carrot roots in monocropping and of their components when fertilized with equitable biomass amounts of these spontaneous species in two cropping seasons in a semi-arid environment. The experimental design was a randomized complete block design, with five treatments and five replications. The treatments consisted of equitable amounts of hairy woodrose and roostertree biomass in amounts of 16, 29, 42, 55, and 68 t ha1 on a dry basis. In each experiment, an additional carrot treatment without fertilizer (absolute control) was planted for comparison with the other treatments to determine maximum physical and economic efficiency. Carrot fertilization was performed with the incorporation of 48.87 t ha1 of M. aegyptia and C. procera dry biomass into the soil to obtain the maximum optimized productive efficiency (commercial productivity). The maximum optimized agroeconomic efficiency (net income) of carrot cultivation was obtained when 49.64 t ha1 of M. aegyptia and C. procera dry biomass was added to the soil. The use of biomass from these spontaneous species from the Caatinga biome as green manure is a viable technology for tuberose producers in monocropping in semiarid environments. Haploid technologies are used to create homozygous lines for accelerated breeding. We aimed to optimize the technology for using the isolated microspore culture in vitro to obtain doubled haploids of the carrot (Daucus carota L.). We studied two carrot varieties with different responsiveness to embryogenesis, Altajskaya lakomka and Breeding line 17. Carrot microspores were isolated from buds and cultivated in liquid nutrient media supplemented with an antibiotic and activated carbon in vitro. They were exposed to different thermal treatments. The experiment showed the benefits of combining cold pre-treatment of buds (5°C for 1 day) with heat shock of isolated microspores in vitro (32°C for 2 days). The induction of embryogenesis on the NLN-13 medium was twice as high as on the MSm-13 medium. The use of 1% activated carbon in 0.5% agarose increased the yield of embryoids by more than 1.5 times. 100 mg/L of ampicillin was found to be the most efficient concentration. After 30 days of cultivation under optimized conditions, the yield was 161.3 and 44.0 embryoids per Petri dish for the cultivar Altajskaya lakomka and Breeding line 17, respectively. The induction of carrot embryogenesis is determined by the type and duration of thermal stress, the composition of the nutrient medium, the use of activated carbon as a sorbent, the addition of β-lactam antibiotics, and the type of explant exposed to thermal treatment. Our technology enabled us to obtain homozygous doubled haploid lines of carrots during a year, and these lines were included in the breeding process to create F1 hybrids.

بیماری پوسیدگی نرم هویج از بیماری­های مهم این محصول در مزرعه و در دوران بعد از برداشت است. از آن جایی که هویج مصرف تازه­خوری دارد استفاده از سموم شیمیایی برای کنترل بیماری پوسیدگی­نرم سلامت بهداشتی محصول را تهدید می­کند؛ لذا در تحقیق حاضر کارایی روش گرمادرمانی به عنوان یک روش ایمن در کنترل بیماری پوسیدگی نرم مورد بررسی قرار گرفت. بدین منظور دو روش هوای گرم و آب گرم در دما­های 40، 45 و50 درجه سلسیوس در مدت زمان­های مختلف استفاده شد. مایه­زنی نمونه­های هویج با ایجاد زخم در دو محل روی هویج و غوطه­ورسازی نمونه در سوسپانسیون باکتری (OD600 = 0.3) انجام شد. بیست­وچهار ساعت بعد از مایه­زنی، تیمارهای مختلف گرمایی روی آن­ها اعمال شد و یک هفته بعد از مایه­زنی نتایج با اندازه­گیری وزن بافت له شده هر نمونه مورد ارزیابی قرار گرفت. نمونه­های تیمارشده با هیپوکلریت سدیم و تتراسیکلین و نمونه­های بدون تیمار شیمیایی و حرارتی به­عنوان شاهد استفاده شدند. آزمایش به صورت طرح کاملا تصادفی در سه تکرار اجرا شد. مقایسه میانگین با استفاده از آزمون دانکن در سطح پنج درصد انجام شد. تاثیر تیمارها در کاهش وزن هویج نیز ارزیایی شد. نتایج بیانگر تاثیر مثبت گرمادرمانی در کاهش معنی­دار لهیدگی بود. از بین تیمارهای مورد بررسی موثرترین تیمار هوای گرم 50 درجه سلسیوس در مدت زمان 10 دقیقه بود. کاهش وزن هویج در این تیمار با شاهد تفاوت معنی­دار نداشت.

Drought is one of the major environmental challenges constraining the production of agricultural crops, including carrot. Seed germination is the initial and most critical stage of crop establishment, and it is very sensitive to drought stress because water scarcity affects the enzymatic solubilization of stored metabolites in seeds that provide energy for the growth of germinating embryo. Few studies evaluating the effect of drought stress on carrot seed germination of more than a few cultivars grown under stress have been published. Therefore, the present study was designed to define the appropriate osmotic potential for evaluating drought tolerance of carrot, evaluate the response of diverse carrot germplasm to drought stress during seed germination to identify drought-tolerant accessions that may be used by plant breeders, and evaluate the relation between amylase activity and germination rate of drought-tolerant and drought-sensitive accessions. To identify an appropriate screening osmotic potential, two commercial cultivars and two United States Department of Agriculture inbreeds were evaluated at six osmotic potentials (00, −0.30, −0.51, −0.58, −0.80, and −1.05 MPa); −0.58 MPa was identified as the optimal osmotic potential for screening the drought tolerance of carrot seed. Cultivated and wild carrot plant introductions (PIs) (n = 200 and n = 50, respectively) from the National Plant Germplasm System were evaluated for drought tolerance under nonstress and simulated drought stress conditions (00 MPa and −0.58 MPa, respectively) by calculating the absolute decrease (AD) in percent germination, inhibition index (II), relative drought tolerance (RDT), and drought tolerance index (DTI). All measurements of drought tolerance identified significant differences among accessions; the AD in seed germination ranged from 0.0% to 69.3%, II ranged from 0.0% to 80.2%, RDT ranged from 0.2 to 1.0, and DTI ranged from 0.13 to 1.47. All wild carrot accessions displayed low levels of drought tolerance, but PI 652387 and PI 177381 (both from Turkey) and PI 274297 (Pakistan) were most drought-sensitive, whereas cultivated accessions PI 643114 (United States), PI 652208 (China), and PI 502347 (Uzbekistan) were most drought-tolerant. Tolerant accessions displayed much higher α-amylase activity under nonstress conditions than sensitive accessions, and α-amylase activity of tolerant accessions was also reduced less with seed germination under increasing osmotic potential (range, 0.0 to −1.05 MPa) than sensitive accessions over 24, 48, and 72 hours of seed germination. This is the first evaluation of drought stress tolerance during seed germination and the enzymatic response of diverse carrot germplasms under simulated drought stress.

In traditional oriental medicine, carrots (Daucus carota L.) are considered effective medicinal herbs; however, the use of D. carota leaves (DCL) as therapeutic agents has not been explored in depth. Therefore, we aimed to demonstrate the value of DCL, generally treated as waste while developing plants for wide industrial availability. Six flavone glycosides were isolated and identified from DCL, and their constituents were identified and quantitated using an NMR and HPLC/UV method, which was optimized and validated. The structure of chrysoeriol-7-rutinoside from DCL was elucidated for the first time. The method exhibited adequate relative standard deviation (<1.89%) and recovery (94.89–105.97%). The deglycosylation of DCL flavone glycosides by Viscozyme L and Pectinex was assessed. Upon converting the reaction contents to percentages, the luteolin, apigenin, and chrysoeriol groups showed values of 85.8, 33.1, and 88.7%, respectively. The enzyme-treated DCL had a higher inhibitory effect on TNF-α and IL-2 expression than that of the carrot roots or carrot leaves without enzyme treatments. The present study characterized a genetically and phenotypically diverse collection of 27 purple and two non-purple (one orange and one yellow) carrot accessions for concentration of root anthocyanins, phenolics, and carotenoids, and antioxidant capacity estimated by four different methods (ORAC, DPPH, ABTS, FRAP), in a partially replicated experimental design comprising data from two growing seasons (2018 and 2019). Broad and significant (p < 0.0001) variation was found among the accessions for all the traits. Acylated anthocyanins (AA) predominated over non-acylated anthocyanins (NAA) in all the accessions and years analyzed, with AA accounting for 55.5–100% of the total anthocyanin content (TAC). Anthocyanins acylated with ferulic acid and coumaric acid were the most abundant carrot anthocyanins. In general, black or solid purple carrots had the greatest TAC and total phenolic content (TPC), and the strongest antioxidant capacities, measured by all methods. Antioxidant capacity, estimated by all methods, was significantly, positively, and moderately-to-strongly correlated with the content of all individual anthocyanins pigments, TAC, and TPC, in both years (r = 0.59–0.90, p < 0.0001), but not with the carotenoid pigments lutein and β-carotene; suggesting that anthocyanins and other phenolics, but not carotenoids, are major contributors of the antioxidant capacity in purple carrots. We identified accessions with high concentration of chemically stable AA, with potential value for the production of food dyes, and accessions with relatively high content of bioavailable NAA that can be selected for increased nutraceutical value (e.g., for fresh consumption).
