Accurately assessing the balance between acoustic wave energy fluxes and radiative losses is critical for understanding how the solar chromosphere is thermally regulated. We investigate the energy balance in the chromosphere by comparing deposited acoustic flux and radiative losses under quiet and active solar conditions using non–local thermodynamic equilibrium inversions with the Stockholm Inversion Code. To achieve this, we utilize spectroscopic observations from the Interferometric BIdimensional Spectrometer in the Na i 5896 Å and Ca ii 8542 Å lines and from the Interface Region Imaging Spectrograph in the Mg ii h and k lines to self-consistently derive spatially resolved velocity power spectra and cooling rates across different heights in the atmosphere. Additionally, we use snapshots of a three-dimensional radiative magnetohydrodynamics simulation to investigate the systematic effects of the inversion approach, particularly the effect of attenuation on the velocity power spectra and the determination of the cooling rates. The results indicate that inversions potentially underestimate acoustic fluxes at all chromospheric heights while slightly overestimating the radiative losses when fitting these spectral lines. However, even after accounting for these biases, the ratio of acoustic flux to radiative losses remains below unity in most observed regions, particularly in the higher layers of the chromosphere. We also observe a correlation between the magnetic field inclination in the photosphere and radiative losses in the low chromosphere in plage, which is evidence that the field topology plays a role in the chromospheric losses. Pro-inflammatory cytokines, chemokines and ROS (reactive oxygen species) are excessively produced in endotoxaemia. However, attempting to inhibit all of these inflammatory signalling pathways at the same time in order to prevent endotoxaemia is difficult. In a previous study we observed that activation of P2X7 receptors elicited the release of IL (interleukin)-1ß from LPS (lipopolysaccharide)-incubated vessels. In the present study, we hypothesize that P2X7 receptor activation is the initial event leading to vascular dysfunction following LPS treatment. LPS-induced decreases in MAP (mean arterial pressure) and pressor responses to NE (noradrenaline) were attenuated in P2X7KO (P2X7-knockout) mice. Hyporeactivity in response to PE (phenylephrine) in isolated mesenteric arteries by LPS treatment was also observed in C57BL/6 [WT (wild-type)] mice, which was prevented by IL1ra (IL-1 receptor antagonist), L-NAME (NG-nitro-L-arginine methyl ester) and indomethacin and in P2X7KO mice. In addition, treatment with IL1ra plus L-NAME produced an additive inhibition of LPS-induced vascular hyporeactivity, suggesting different signalling pathways between IL-1ß and NOS (NO synthase). LPS-induced plasma levels of IL-1ß, TNFa (tumour necrosis factor a), IL-10, vascular eNOS (endothelial NOS) and COX2 (cyclo-oxygenase 2) protein expression, as determined by ELISA and Western blot, observed in WT mice were inhibited by IL1ra and in P2X7KO mice. These results suggest that P2X7 receptor activation involves an initial upstream mechanism of LPS-induced vascular dysfunction, which is associated with IL-1ß-mediated eNOS, COX2 activation and TNFa release. Excessive nitric oxide (NO) production by inducible NO synthase has been implicated in the hyporesponsiveness to vasoconstrictors present in septic shock. Here we show that a brief incubation (30 min) of rat aorta rings with NO donors renders the vessels hyporesponsive to phenylephrine for several hours. Contraction of rings without endothelium by phenylephrine (0.1 nM to 100 µM) was decreased by 50–60% after incubation (30 min) with sodium nitroprusside (3–300 µM) or S-nitroso-acetyl-d,l-penicillamine (SNAP; 70–200 µM). This decrease was characterized by reductions in maximal response and rightwards shifts of phenylephrine concentration/response curves, present even 130 min after NO donor removal. Soluble guanylate cyclase inhibitors methylene blue (10 µM) and 1H-(1,2,4)-oxadiazol-(4,3-a)-quinoxalin-1-one (ODQ, 1 µM) or the potassium channel blockers TEA (tetraethylammonium; 10 mM) and charybdotoxin (100 nM) inhibited the hyporesponsiveness to phenylephrine induced by the NO donors. In contrast, 4-aminopyridine (1 mM) and glibenclamide (10 µM) had no effect. Our results show that incubation with NO donors reproduces the hyporesponsiveness to phenylephrine and that NO alone accounts for most, if not all, the refractoriness to vasoconstrictors present in septic shock. In addition, soluble guanylate cyclase activation and opening of potassium channels, more specifically the calcium-activated subtype, play a predominant role in this NO-induced hyporesponsiveness to phenylephrine in the rat aorta. Antigenic drift, the gradual accumulation of amino acid substitutions in the influenza virus hemagglutinin (HA) receptor protein, enables viral immune evasion. Antibodies (Abs) specific for the drift-resistant HA stem region are a promising universal influenza vaccine target. Although anti-stem Abs are not believed to block viral attachment, here we show that complement component 1q (C1q), a 460-kilodalton protein with six Ab Fc-binding domains, confers attachment inhibition to anti-stem Abs and enhances their fusion and neuraminidase inhibition. As a result, virus neutralization activity in vitro is boosted up to 30-fold, and in vivo protection from influenza PR8 infection in mice is enhanced. These effects reflect increased steric hindrance and not increased Ab avidity. C1q greatly expands the anti-stem Ab viral escape repertoire to include residues throughout the HA, some of which cause antigenic alterations in the globular region or modulate HA receptor avidity. We also show that C1q enhances the neutralization activity of non-receptor binding domain anti-SARS-CoV-2 spike Abs, an effect dependent on spike density on the virion surface. These findings demonstrate that C1q can greatly expand Ab function and thereby contribute to viral evolution and immune escape.

Hypertrophic cardiomyopathy (HCM) is the most common inherited cardiac disease. Recently, a connection has been observed between the presence of first-degree atrioventricular block (FDAVB) and cardiovascular outcomes, although the pathophysiology of this association remains poorly understood. Considering the period 2000-2023, we retrospectively included HCM patients at sinus rhythm at the first appointment and sought possible interactions of FDAVB (defined as PR interval >200 ms) with different clinical and imaging variables and with the occurrence of cardiovascular events, including atrial fibrillation (AF). A total of 97 patients were included, of whom 57 (58.8%) were men, with a mean age of 51±19 years, and 14 (14.4%) had FDAVB. During a median of 4.29 (P25 1.92, P75 7.67) years of follow-up, 35 cardiovascular events occurred, including 13 de novo diagnoses of AF, 8 hospitalizations due to heart failure, 8 new-onset strokes, 4 myocardial infarctions, and 2 implantations of cardio defibrillators in secondary prevention; no HCM-related death occurred. We did not find any association between outcomes and the presence of FDAVB. The role of FDAVB as a prognostic marker in HCM patients requires further investigation. We found that FDAVB patients were older, more frequently reported dyspnea, had a larger QRS duration, a higher E/e' ratio, and lower maximal left ventricle wall thickness by magnetic resonance (p<0.05). After multivariable analysis, FDAVB was independently associated with a higher echocardiographic E/e' ratio (p=0.039) (odds ratio=1.588). This is the first paper to document an independent association between FGAVB and a higher E/e' ratio in HCM patients.

Extreme environmental conditions eventually limit plant growth [J. R. Dinneny, Annu. Rev. Cell Dev. Biol. 35 , 1-19 (2019), N. Gigli-Bisceglia, C. Testerink, Curr. Opin. Plant Biol. 64 , 102120 (2021)]. Here, we reveal a mechanism that enables multiple external cues to get integrated into auxin-dependent growth programs in Arabidopsis thaliana . Our forward genetics approach on dark-grown hypocotyls uncovered that an imbalance in membrane lipids enhances the protein abundance of PIN-LIKES (PILS) [E. Barbez et al. , Nature 485 , 119 (2012)] auxin transport facilitators at the endoplasmic reticulum (ER), which thereby limits nuclear auxin signaling and growth rates. We show that this subcellular response relates to ER stress signaling, which directly impacts PILS protein turnover in a tissue-dependent manner. This mechanism allows PILS proteins to integrate environmental input with phytohormone auxin signaling, contributing to stress-induced growth adaptation in plants.

Ethnopharmacological Relevance: Cochlospermum regium is well-known as "Algodãozinho do cerrado" in folk Brazilian medicine, and is used to fight infections, inflammation and skin disorders., Aim of the Study: To identify the phytochemical constituents and the effects of the ethanolic extract of C. regium leaves (EECR) on inflammation and pain, and the effects of C. regium gel (GEECR) on wound healing., Materials and Methods: Animals were treated with EECR (30-300 mg/kg) or GEECR (1.25 and 2.5%) and studies were conducted using carrageenan-induced pleurisy and paw edema tests, formalin-induced pain model, and excision wound model., Results: In total, 25 compounds, including quercitrin, methyl gallate, and 1,2,3,4,6-pentagalloylhexose, with highest detectability were identified. Background: 1,3-Thiazolidin-4-ones are heterocycles whose importance in the pharmaceutical market has already been established. Many of these synthetic derivatives, which contain a thiazolidinone nucleus, are currently used in various commercial formulations or are already in clinical trials for the treatment of disease for their anticonvulsant, antihypertensive and antidiabetic activities in addition to their activity against Streptomyces. 1,3-Thiazolidin-4-ones are produced mainly by cyclo condensation between an imine (generated in situ by the reaction of an aldehyde with an amine) and α-mercaptoacetic acid, known as the MALI (Mercaptoacetic Acid Looking Imine) reaction., Objective: A solvent-free methodology was developed to synthesize a 1,3-thiazolidin-4-one family by the MALI reaction. An apparatus was developed to grind a solid-liquid mixture of anilines, benzaldehydes and thioglycolic acid to activate the reaction. This apparatus was named MGRE (mechanical grinding reaction equipment)., Methods: Substituted aniline 2 (4 mmol), substituted benzaldehyde 1 (4 mmol) and thioglycolic acid 3 (12 mmol) were placed in a mortar. The reagents were macerated using the MGRE at room temperature for a specified time period. At the end of the reaction, the product was poured into ice, the precipitate formed was neutralized (with 5% NaHCO 3 ), and the solution was extracted in ethyl acetate and dried in MgSO 4 . The solid was recrystallized from MeOH/H 2 O., Results: The developed MGRE is a modification of a rod used in a mechanical stirrer. This adaptation is inexpensive and simple in construction, and it enables reactions to occur over long periods of time that would be exhaustive for manual grinding. Fifteen (1,3) thiazolidin-4-ones were produced. The products were synthesized using the solvent-free MALI-MGRE procedure., Conclusions: The MALI-MGRE methodology developed to produce 1,3-thiazolidin-4-ones showed a good reaction scope, has an easy work-up and is solvent-free. Consequently, MALI-MGRE is classified as a green methodology. An innovation of this study is the construction of the MGRE, which involves modifying the rod in a mechanical stirrer. Introduction: Leprosy is a neglected, infectious, granulomatous and chronic disease caused by the pathological agent Mycobacterium leprae . The course of the disease is more aggressive in patients under 15 years of age, but the current treatment offered worldwide consists of solid forms, by the combination of antibiotics such as rifampicin, clofazimine and dapsone. This represents results in lack of adherence in pediatric patients and drug therapy failure, although numerous formulations and technologies have already been developed., Area Covered: This study aims to analyze the technological evolution of the pharmaceutical treatment of leprosy, aimed at children. A review of patents around the world was conducted to look for technical and clinical aspects of formulations and devices., Expert Opinion: Innovative formulations for pediatric patients were classified according to the routes of administration as oral, inhalable, injectable and transdermal. The formulations were organized as alternatives for pediatric therapy, taking into account the physicochemical aspects of drugs and the physiological aspects of pediatric patients. Among the difficulties for the patented formulations to reach the market, of special note is the low stability of the physicochemical characteristics of the drugs. Optimization of formulations would favor the pediatric treatment of leprosy, aiming at therapeutic success.

Multiple internal and external signals modulate the metabolism, intercellular transport and signaling of the phytohormone auxin. Considering this complexity, it remains largely unknown how plant cells monitor and ensure the homeostasis of auxin responses. PIN-LIKES (PILS) intracellular auxin transport facilitators at the endoplasmic reticulum are suitable candidates to buffer cellular auxin responses because they limit nuclear abundance and signaling of auxin. We used forward genetics to identify gloomy and shiny pils (gasp) mutants that define the PILS6 protein abundance in a post-translational manner. Here, we show that GASP1 encodes an uncharacterized RING/U-box superfamily protein that impacts on auxin signaling output. The low auxin signaling in gasp1 mutants correlates with reduced abundance of PILS5 and PILS6 proteins. Mechanistically, we show that high and low auxin conditions increase and reduce PILS6 protein levels, respectively. Accordingly, non-optimum auxin concentrations are buffered by alterations in PILS6 abundance, consequently leading to homeostatic auxin output regulation. Sewage sludge (SS), a solid residue of effluent treatment, is rich in organic matter and nutrients, while also containing heavy metals and other potential contaminants. The feasibility of employing SS as a substrate component for seedling production depends on its composition and dose, as well as on the tolerance limit of individual plant species. To expand the knowledge base on the use of SS in the production of plants native to the Brazilian Cerrado biome, we evaluated the physiological responses and quality of Alibertia edulis seedlings grown under distinct SS concentrations. Chlorophyll a fluorescence (ChlF), stomatal conductance (gs), leaf temperature (Lt), biomass, growth, and seedling quality were investigated. At 25%, SS improved growth, biomass, and seedling quality, while substrates containing 50% or more SS affected gs, Lt, and ChlF. Seedling quality was strongly worsened by SS at 75% and 100%, as shown by lower biomass and impaired growth, including leaf symmetry loss and leaf deformities possibly related to contaminants, particularly heavy metals. Accordingly, we conclude that SS exhibited potential as a fertilizer at concentrations below 50%, but exerted a toxic effect on seedlings at higher concentrations.

Perillyl alcohol (POH) is a monocyclic terpene that has strong antitumor activity. Brain tumors are particularly difficult to treat with therapeutic agents, and clinical trials have shown their low tolerance through oral administration. We proposed the entrapment of POH into an oil-in-water chitosan nanoemulsion aiming its intranasal administration for brain targeting. An ultra-performance liquid chromatography/tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for the quantitation of total metabolite perillic acid (PA) in plasma and brain of rats. The rat samples containing the metabolite were treated by liquid-liquid extraction with acetonitrile. The mobile phase was 0.1% formic acid in water (solvent A) and 0.1% formic acid in methanol (solvent B), at a flow rate of 0.3 mL min -1 in gradient elution. The chromatography was run for 10 min, and analytical curves were built in acetonitrile, plasma, and brain. The PA was detected in positive ion mode with multiple reaction monitoring. The method has shown high selectivity, sensitivity, and throughput. The low quantification limits of 162, 178, and 121 ng mL -1 for acetonitrile, brain, and plasma, respectively, indicate a good detectability of the method. The repeatability and precision observed were within the limits recommended in the literature. The accuracy of the method was verified through high recovery rates (106-118%). The validated method was successfully applied to the pharmacokinetic study of the metabolite PA after the intranasal administration of free or POH-loaded nanoemulsion in rats. Ursolic acid (UA) is a naturally occurring triterpene that has been investigated for its antitumor activity. However, its lipophilic character hinders its oral bioavailability, and therapeutic application. To overcome these limitations, chitosan (CS) modified poly (lactic acid) (PLA) nanoparticles containing UA were developed, characterized, and had their oral bioavailability assessed. The nanoparticles were prepared by emulsion-solvent evaporation technique and presented a mean diameter of 330 nm, zeta potential of +28 mV, spherical shape and 90% encapsulation efficiency. The analysis of XRD and DSC demonstrated that the nanoencapsulation process induced to UA amorphization. The in vitro release assay demonstrated that 53% of UA was released by diffusion after 144 h, following a second-order release kinetics. In simulated gastrointestinal fluids and mucin interaction tests, CS played an important role in stability and mucoadhesiveness improvement of PLA nanoparticles, respectively. In the presence of erythrocytes, nanoparticles proved their hemocompatibility. In tumor cells, nanoparticles presented lower cytotoxicity than free UA, due to slow UA release. After a single oral dose in rats, CS modified PLA nanoparticles increased the UA absorption, reduced its clearance and elimination, resulting in increased bioavailability. Glyphosate (Gly) is the most widely used herbicide in the world and has broad-spectrum and non-selective activity. Its indiscriminate use hence risks contamination of water bodies and can affect living organisms, especially sensitive or resistant non-target plants. Despite this, studies on physiological mechanisms and Gly remediation in Neotropical aquatic plants remain limited. This study aims to evaluate the physiological mechanisms of the aquatic macrophyte Salvinia biloba on exposure to different concentrations of a Gly commercial formulation (Gly-CF) and a Gly analytical standard (Gly-AS). Furthermore, using square-wave voltammetry (SWV), we determined whether the studied plant could remove Gly from water. Our data suggest that Gly-AS and Gly-CF induce similar physiological responses in S. biloba. However, Gly-CF was more phytotoxic. Depending on the concentration, the two forms of Gly affected the plants, decreasing the chlorophyll a and b contents and the photosystem II (PSII) photochemical activity. The data also revealed that Gly promoted oxidative stress and increased the shikimic acid concentration. At the same time, the plants removed Gly from water, with 100% removal for 1 mg L -1 Gly and above 60% removal for the other concentrations studied. Therefore, our results suggest that S. biloba may be a potential phytoremediation agent for low Gly concentrations, since 1 mg L -1 Gly was completely removed and exhibited low phytotoxicity. Background: We found in the literature, an excellent review of the Biginelli reaction that addresses the methodologies for obtaining enantiopure dihydropyrimidinones (DHPMs). In 1992, optically pure DHPMs were obtained by fractional crystallization of the diastereomeric ammonium salt derivative with (S)-(-) and (R)- (+)-α-methyl benzylamine and by other chiral resolution techniques, such as chiral high-performance liquid chromatography (HPLC). Asymmetric syntheses of these compounds are also explained in the literature. The main strategy is to use acid catalysts such as organophosphates, organometallic complexes, amines and diamines, nanocomposites, and chiral ionic liquids, e.g., L-prolinium sulfate (Pro2SO4)., Objective: The objective was to study the Biginelli reaction with a chiral aldehyde., Methods: A mixture of ethyl acetoacetate (0.26 g, 3 mmol), urea (0.18 g, 3 mmol) and ethyl lactate (EL) (1 mL) was left under heating at 70°C and stirring for 1 h. Next, (-)-(1R)-myrtenal (0.45 g, 3 mmol) was added, and the medium was heated for 5 h more until the formation of a white solid. Ten millilitres of distilled water was added, and the product was extracted with CH2Cl2 (3 x 4 mL). The solvent was evaporated, and the product was recrystallized from ethanol-water., Results and Discussion: (+)-Myrtenal was used as a chiral substrate for a study that led to ethyl (R)-4-((1R,5S)- 6,6-dimethylbicyclo [3.1.1]hept-2-en-2-yl)-6-methyl-2-oxo-1,2,3,4-tetrahydropyrimidine-5-carboxylate by the Biginelli synthesis using EL as a green solvent. The result is the first example of the enantiospecific Biginelli reaction. The product was exhaustively characterized by several physical analysis methods, i.e., 1H, 13C and 2D nuclear magnetic resonance (NMR) spectroscopies, infrared (IR) spectroscopy, mass spectrometry (MS), and high-resolution MS (HRMS), and its structure was unequivocally elucidated by X-ray crystallography., Conclusion: Compound (4R)-4-(6,6-dimethylbicyclo[3.1.1]hept-2-en-2-yl)-6-methyl-2-oxo-1,2,3,4- tetrahydropyrimidine- 5-ethyl carboxylate is the first example of the enantiospecific Biginelli reaction. In addition, the process has the advantage of using EL as a green solvent. Many flavonoids have been shown to present good results for the treatment of gastric ulcers. Baicalein, a bioactive flavonoid derived from the Scutellaria baicalensis Georgi root, possesses several biological effects, such as anti-inflammatory and antioxidant. This study was conducted to assess the gastroprotective properties of baicalein. Anti-ulcerogenic assay was performed using the protocol of ulcer induced by ethanol/HCl in mice; then, the role of presynaptic α 2 -receptors, sulfhydryl (SH) compounds, nitric oxide (NO), prostaglandin (PG) and ATP-sensitive K + (K ATP ) channels in gastroprotection of baicalein was investigated. The levels of reduced glutathione (GSH) and the myeloperoxidase (MPO) activity were measured in the gastric mucosa. Parameters of gastric secretion (volume, [H + ] and pH) were determined with or without the presence of the secretagogue agent histamine, as well as mucus in gastric contents, by the pylorus ligation model. In vitro H + ,K + -ATPase activity was also determined. Baicalein (10, 30 and 100 mg/kg) exhibited a dose related gastroprotective effect (P < 0.001) against acidified ethanol-induced lesions. The intraperitoneal treatment of mice with a α 2 -adrenoreceptor antagonist (yohimbine; 2 mg/kg), a SH compounds blocker (N-ethylmaleimide, NEM; 10 mg/kg), a non-selective inhibitor of NO synthase (N w -nitro-L-arginine methyl ester hydrochloride, L-NAME; 10 mg/kg), a non-selective inhibitor of cyclo-oxygenase (indomethacin; 10 mg/kg) or a K ATP channel blocker (glibenclamide; 10 mg/kg) was able to reverse (P < 0.001) the gastroprotective response caused by baicalein (30 mg/kg). Baicalein (30 mg/kg; P < 0.05) was able to increase GSH levels and decreasing MPO activity. The intraduodenal treatment with baicalein (30 and 100 mg/kg) significantly increased (P < 0.05) the gastric mucus secretion. Additionally, the treatment with baicalein reduced (30 and 100 mg/kg; P < 0.05) the secretion volume and total acid secretion, and also increased (10, 30 and 100 mg/kg; P < 0.001) the pH value, after pylorus ligature. Baicalein (30 mg/kg) was also effective in inhibiting the effects of histamine on gastric secretion (volume, [H + ] and pH; P < 0.001). Baicalein at 10 and 30 μg/mL showed anti-H + ,K + -ATPase activity. In conclusion, the present results provide convincing evidence that baicalein could be used as a cytoprotective (preventive effect) and anti-ulcerogenic (anti-secretory effect) agent in the gastric ulcers., (Copyright © 2016 Elsevier Ireland Ltd. Taekwondo has evolved into a modern-day Olympic combat sport. The physical and physiological demands of modern-day taekwondo competition require athletes to be competent in several aspects of fitness. This review critically explores the physical and physiological characteristics of taekwondo athletes and presents implications for training and research. International taekwondo athletes possess low levels of body fat and a somatotype that characterises a blend of moderate musculoskeletal tissue and relative body linearity. While there is some variation in the maximum oxygen uptake of taekwondo athletes, moderate to high levels of cardio-respiratory fitness are necessary to support the metabolic demands of fighting and to facilitate recovery between consecutive matches. Taekwondo athletes demonstrate high peak anaerobic power characteristics of the lower limbs and this attribute appears to be conducive to achieving success in international competition. The ability to generate and sustain power output using both concentric and 'stretch-shortening cycle' muscle actions of the lower limbs may be important to support the technical and tactical actions in combat. Taekwondo competitors also display moderate to high maximum dynamic strength characteristics of the lower and upper extremities, and moderate endurance properties of the trunk and hip flexor musculature. The dynamic nature of the technical and tactical actions in the sport demand high flexibility of the lower limbs. More extensive research is required into the physical and physiological characteristics of taekwondo athletes to extend existing knowledge and to permit specialised conditioning for different populations within the sport.

Objective and Design: To investigate the effect of nitric oxide (NO) donors on inflammatory mouse paw edema (MPE)., Materials and Methods: Mice previously treated with sodium nitroprusside (SNP; 1.5, 5 and 10 micromol/kg) or S-nitroso-N-acetyl-DL-penicillamine (SNAP; 7, 14 and 28 micromol/kg) were injected with inflammatory mediators in the paw. Paw edema, myeloperoxidase activity and vascular dye leakage were measured., Results: Pre-treatment with SNP and SNAP (4 h or 12 h) reduced (approximately 50%) MPE induced by carrageenan, dextran sulfate, bradykinin and histamine but not by serotonin. Pre-treatment with SNP also inhibited carrageenan-induced increases in myeloperoxidase activity and vascular dye leakage. Methylene blue blocked the SNP-induced reduction in MPE when injected 30 min before or 2 h after SNP, but not 4 or 6 h after the NO donor. Tetraethylammonium blocked the SNP-induced reduction in MPE if injected 30 min before or 2, 4 or 6 h after SNP., Conclusions: NO donors have a long-lasting anti-inflammatory effect in MPE, which involves guanylate cyclase and tetraethylammonium-sensitive potassium channels.

We investigated the effects of the exposure of the rat vascular system to nitric oxide (NO), using infusion of either NO donor sodium nitroprusside (SNP) or S-nitroso-acetyl-DL-penicillamine (SNAP) on mean arterial pressure (MAP) responses to vasoconstrictors (phenylephrine, angiotensins I and II) and to vasodilators (bradykinin, acetylcholine, SNP, and iloprost). SNP (250 nmol/kg/ min) or SNAP (85 nmol/kg/min) infused for 30 min decreased MAP by 40 to 60 mm Hg. MAP returned to normal levels 5 to 10 min after the end of infusion. After infusion of SNP or SNAP the effects of phenylephrine, angiotensin I, and angiotensin II were reduced by 40 to 80%, whereas the responses to bradykinin or acetylcholine were enhanced by 50 to 80%. These changes in vascular responsiveness persisted for at least 24 h after the SNP infusion. Pretreatment with either tetraethylammonium (360 micromol/kg) or 4-aminopyridine (4-AP; 1 micromol/kg) did not alter the effects of phenylephrine or bradykinin in control animals, but prevented SNP-induced changes in responsiveness to phenylephrine or bradykinin. On the other hand, administration of tetraethylammonium, even 24 h after SNP infusion, reversed hyporesponsiveness to phenylephrine, whereas 4-AP was ineffective. Tetraethylammonium and 4-AP did not alter the increased responses to bradykinin. Glibenclamide was without effect in any situation. These results indicate that NO-induced changes on vascular responsiveness to vasoconstrictors and vasodilators are much more profound and long-lasting than described previously and that the effects of NO appear to be, at least in part, mediated by persistent activation of a tetraethylammonium-sensitive population of K+ channels.
