Documentation of exposure of threatened and endangered sea turtles to petroleum and dispersant released into the northern Gulf of Mexico (GoM) during the Deepwater Horizon (DWH) oil spill was a critical component of the Natural Resource Damage Assessment (NRDA) process. Substances collected from the skin of oiled and suspected oiled turtles were analyzed for petroleum hydrocarbons to determine oiling status and oil sources. Tissue, gastroenteric and bile samples from a subset of visibly oiled and unoiled turtles that died during the spill in 2010 and in 2011 were analyzed for evidence of internal exposure and absorption of polycyclic aromatic hydrocarbons (PAHs) and the dispersant component dioctyl sodium sulfosuccinate (DOSS). The volume of external oil collected from sea turtles was sufficient to confirm the presence of petroleum on 61% of turtles, and oil from the DWH spill was identified as the source in 97% of those turtles in which conclusive comparison was possible. Visibly oiled turtles had higher concentrations of tissue PAH or biliary fluorescent PAH metabolites compared to those determined in unoiled animals. Findings in most of the unoiled turtles were suggestive of low-level PAH exposure from various sources that may represent background values for sea turtles from the northern GoM. DOSS levels were below the limit of quantitation in all samples analyzed except in an esophagus sample of a heavily oiled sea turtle. Overall, the results for petroleum or petroleum-derived compounds of both external and internal samples of sea turtles supported visual observations of oiling.

Polycyclic aromatic hydrocarbons (PAHs) have frequently been suspected of governing crude oil toxicity because of similar morphological defects in fish. However, PAH concentrations are often not high enough to explain the observed crude oil toxicity. We hypothesize that one PAH can enhance the metabolism and toxicity of another PAH when administered as a mixture. Early life stage Atlantic haddock ( Melanogrammus aeglefinus ) were in this study exposed to phenanthrene in the presence and absence of 3-methylchrysene that is known to induce the metabolic enzyme cytochrome P450 1A via cyp1a gene expression. Uptake, metabolism, and multiple toxicity endpoints were then measured in a time-course study up to 3 days post-hatching. Passive dosing provided aqueous concentrations ≈180 μg/L for phenanthrene and ≈0.6 μg/L for 3-methylchrysene, which resulted in tissue concentrations ≈60 μg/g ww for phenanthrene and ≈0.15 μg/g ww for 3-methylchrysene. The low concentration of 3-methylchrysene led to the elevated expression of cyp1a but no toxicity. Levels of phenanthrene metabolites were 5-fold higher, and morphological defects and cardiotoxicity were consistently greater when co-exposed to both compounds relative to phenanthrene alone. This work highlights the metabolic activation of PAH toxicity by a co-occurring PAH, which can lead to excess toxicity, synergistic effects, and the overproportional contribution of PAHs to crude oil toxicity.

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous environmental contaminants that are widely monitored in marine biota from urbanized areas, due to their toxicity to aquatic organisms. Teleost fish can quickly metabolize PAHs into hydroxylated forms (OHPAHs) that, in some cases, are more toxic than the parent (unmetabolized) PAHs. But due to this fast metabolism, monitoring traditional parent PAHs in fish can cause underestimation on assessing PAH exposure. In addition, environmental levels of individual OHPAH metabolites are lacking in the literature worldwide. Therefore, we developed a rapid and accurate analytical method in which a number of individual OHPAHs metabolites are measured simultaneously in fish bile, via liquid chromatography coupled with tandem mass spectrometry, including low and high molecular weight mono- and diol-OHPAHs. We analyzed bile samples of 119 English sole (Parophrys vetulus) collected from 14 Puget Sound, WA, USA, sites, which has multiple sources of PAHs, including urban stormwater runoff, wastewater effluents, as well as an inactive creosote facility. The mean (± SD) biliary summed OHPAH (∑OHPAH) concentrations determined in English sole from urban, near-urban, and non-urban sites were 790 ± 1400 (n = 46), 310 ± 330 (n = 44) and 130 ± 200 (n = 29) ng/mL, respectively, with a maximum reaching 9400 ng/mL in a sample from an urban site. We compared these novel biliary OHPAH metabolite data with parent PAHs measured in stomach content of the same individual sole. Biliary ∑OHPAH concentrations were significantly correlated with the levels of ∑PAH in stomach content, however, with major differences in their distribution. Persistent organic pollutants were investigated in the blood of living seabirds (Pterodroma arminjoniana, Onychoprion fuscatus, Sula dactylatra, Anous minutus, Anous stolidus and Sula leucogaster) from two Brazilian Conservation Units - Trindade Island and São Pedro and São Paulo Archipelago. ∑PCBs (0.55 to 55.09 ng/g wet weight (ww), ∑DDTs (0.01 to 17.36 ng/g ww) and Mirex (0.01 to 5.53 ng/g ww) were predominant in all samples. Trindade petrel Pterodroma arminjoniana presented higher values than other seabirds, which is potentially related to diet and migratory behavior. Fluctuations in the trophic ecology data, through carbon and nitrogen stable isotope ratios, warn of temporal variation in diet and foraging and highlight the importance of using a nondestructive matrix to allow long-term monitoring of POP contamination in seabirds from Brazilian Conservation Units. The purpose of the present study was to evaluate the efficacy of the treatment with a recombinant cysteine proteinase from Leishmania, rldccys1, associated with allopurinol or miltefosine on Leishmania (Leishmania) infantum chagasi-infected hamsters. Golden Syrian hamsters infected with L. (L.) infantum chagasi were treated with either miltefosine (46 mg/kg) or allopurinol (460 mg/kg) alone by oral route or associated with rldccys1 (150 µg/hamster) by subcutaneous route for 30 days. Infected hamsters were also treated with miltefosine (46 mg/kg) plus rldccys1 (150 µg/hamster) for 30 days (phase 1) followed by two additional doses of rldccys1 (250 µg/hamster) (phase 2). After the end of treatment, the animals were analyzed for parasite load, body weight, serum levels of immunoglobulins, cytokine expression, and drug toxicity. The data showed a significant decrease of parasite load in infected hamsters treated with allopurinol or miltefosine alone or associated with rldccys1, as well as in those treated with rldccys1 alone. Significantly lower levels of serum IgG were detected in hamsters treated with allopurinol plus rldccys1. The treatment with miltefosine associated with rldccys1 prevented relapse observed in animals treated with miltefosine alone. A significant loss of body weight was detected only in some hamsters treated with miltefosine for 1 month and deprived of this treatment for 15 days. There were no significant differences in transcript expression of IFN-γ and IL-10 in any of treated groups. Neither hepatotoxicity nor nephrotoxicity was observed among controls and treated groups. These findings open perspectives to further explore this immunochemotherapeutic schedule as an alternative for treatment of visceral leishmaniasis., (© 2022. The use of rosemary essential oil (RO) and its combination with nisin (RO+N) in preventing the multiplication of Alicyclobacillus acidoterrestris in orange juice was evaluated. The minimum inhibitory and bactericidal concentrations (MIC and MBC) for RO were both 125 μg ml -1 while RO+N displayed a synergistic effect. The use of RO and RO+N at concentrations of 1, 4 and 8× MIC in orange juice for 96 h was evaluated in terms of their sporicidal effectiveness. With regard to the action against A. acidoterrestris spores, RO at 8× MIC was sporostatic, whereas RO+N at 1× MIC was sporicidal. Morphological changes in the structure of the micro-organism after treatment were also observed by microscopy. Furthermore, flow cytometric analysis showed that most cells were damaged or killed after treatment. In general, the antioxidant activity after addition of RO+N decreased with time. Aluminum smelter-derived polycyclic aromatic hydrocarbons (PAHs) in outmigrant juvenile Chinook salmon were evaluated in Kitimat Arm, British Columbia, Canada from 2000 to 2004, and in 2015. Decades of continual smelter operations by Rio Tinto resulted in PAH contamination of marine sediments at levels associated with adverse effects in juvenile salmon. Recently, smelter operations have undergone process changes to reduce PAH input to the environment. The PAH concentrations in juvenile Chinook salmon observed in 2000 to 2004, at sites nearest the smelter were comparable to salmon in other urban areas where reduced disease resistance was observed; the levels were lower in 2015 than 2000-2004 suggesting that the recent process changes were effective. The palladacycle complex DPPE 1.2 was previously shown to inhibit Leishmania (Leishmania) amazonensis infection in vitro and in vivo . The present study aimed to evaluate the effect of DPPE 1.2 associated with a recombinant cysteine proteinase, rLdccys1, and the adjuvant Propionibacterium acnes on L. (L.) amazonensis infection in two mouse strains, BALB/c, and C57BL/6. Treatment with this association potentiated the leishmanicidal effect of DPPE 1.2 resulting in a reduction of parasite load in both strains of mice which was higher compared to that found in groups treated with either DPPE 1.2 alone or associated with P. acnes or rLdccys1. The reduction of parasite load in both mice strains was followed by immunomodulation mediated by an increase of memory CD4 + and CD8 + T lymphocytes, IFN-γ levels and reduction of active TGF-β in treated animals. No infection relapse was observed 1 month after the end of treatment in mice which received DPPE 1.2 associated with rLdccys1 or rLdccys1 plus P. acnes in comparison to that exhibited by animals treated with DPPE 1.2 alone. Evaluation of serum levels of AST, ALT, urea, and creatinine showed no alterations among treated groups, indicating that this treatment schedule did not induce hepato or nephrotoxicity. These data indicate the potential use of this association as a therapeutic alternative for cutaneous leishmaniasis caused by L. The present study focused on the activity of the palladacycle complex DPPE 1.1 on Leishmania (Leishmania) amazonensis . Promastigotes of L. (L.) amazonensis were destroyed in vitro by nanomolar concentrations of DPPE 1.1, whereas intracellular amastigotes were killed at drug concentrations fivefold less toxic than those harmful to macrophages. L. (L.) amazonensis -infected BALB/c mice were treated by intralesional injection of DPPE 1.1. Animals treated with 3.5 and 7.0 mg/kg of DPPE 1.1 showed a significant decrease of foot lesion sizes and a parasite load reduction of 93 and 99%, respectively, when compared to untreated controls. Furthermore, DPPE 1.1 was non-toxic to treated animals. The cathepsin B activity of L. (L.) amazonensis amastigotes was inhibited by DPPE 1.1 as demonstrated spectrofluorometrically by use of a specific fluorogenic substrate. Analysis of T-cells populations in mice treated with DPPE 1.1 and untreated controls was performed by fluorescence-activated cell sorter (FACS). IFN-γ was measured in supernatants of lymphocytes from popliteal and inguinal lymph nodes isolated from treated and untreated mice and stimulated with L. (L.) amazonensis amastigotes extract and active TGF-β was evaluated in supernatants of foot lesions; both dosages were carried out by means of a double-sandwich ELISA assay. A significant increase of TCD4 + and TCD8 + lymphocytes and IFN-γ secretion was displayed in mice treated with DPPE 1.1 compared to untreated animals, whereas a significant reduction of active TGF-β was observed in treated mice. These findings open perspectives for further investment in DPPE 1.1 as an alternative option for the chemotherapy of cutaneous leishmaniasis.

Pituitary-hormone signaling plays critical roles in the onset and progression of gametogenesis in vertebrates. This study characterized expression patterns of pituitary gonadotropin beta-subunits (fshb and lhb), brain-type aromatase (cyp19a1b), androgen (ar1, ar2) and estrogen receptors (esr1, esr2a, esr2b), and changes in plasma steroid levels by liquid chromatography/tandem mass spectrometry in wild sablefish (Anoplopoma fimbria, order Scorpaeniformes) during a complete reproductive cycle. Transcripts for fshb increased during early gametogenesis and peaked in late vitellogenic females and late recrudescent males, while expression of lhb reached maximum levels in periovulatory and spermiating fish. Pituitary levels of cyp19a1b and ar1 were strongly correlated with those of lhb in females and males, increasing during gametogenesis and reaching maximum levels prior to spawning. By contrast, expression of ar2, and the three estrogen receptors differed between female and male sablefish. 17β-estradiol (E2) was the dominant steroid in females during vitellogenesis, while a range of at least 6 steroids (11β-hydroxyandrostenedione, testosterone [T], E2, 11-ketotestosterone [11KT], 11-deoxycortisol, and 17α,20β,21-trihydroxyprogesterone) were detected at similar levels in males during testicular development. Prior to spawning, a marked increase in 4-androstenedione, T, 11KT and E2 was found in both periovulatory females and spermiating males. In conclusion, the concomitant changes in plasma androgen levels and pituitary ar1 expression during gametogenesis suggest a specific role for androgens in pituitary hormone regulation of reproduction in sablefish. This study evaluated the PAH bioavailability from Santos Bay (Brazil) in 4 species of fish, using PAH biliary metabolites. The collection was done monthly, between July and December, in three different regions of Santos Bay. The metabolites were analyzed through a high performance liquid chromatograph with fluorescence detectors. Total metabolites concentrations ranged from 65.5 to 589μgg -1 of bile, evidencing PAH bioavailability on Santos Bay. Levels of phenanthrene and benzo[a]pyrene metabolites were in the classification range of areas moderate contaminated. Those concentrations were lower in Nebris microps and higher in Sphoeroides testudineus (p<0.05). Naphthalene metabolites concentrations did not differ significantly among fish species and were in the classification range of low contaminated areas. There were no significant spatial and temporal differences in levels among sampled areas.
