To investigate if the molecular control of DV patterning is conserved inside an insect order, the emergent model hemiptera species Rhodnius prolixus was studied. We found that R. prolixus BMP pathway controls the entire DV axis, with a broader effect respective to Toll, as shown for the hemiptera Oncopeltus fasciatus. Different from O. fasciatus, the unique R. prolixus short gastrulation (sog) and the twisted gastrulation (tsg) orthologues do not antagonize, but rather favour embryonic BMP signalling. Our results reinforce the hypothesis that hemiptera rely preferentially on BMPs for DV patterning but that, surprisingly, in R. prolixus Sog and Tsg proteins exert only a positive role to establish a dorsal-to-ventral BMP gradient. Since sog has been reported to be lost from orthopteran and hymenopteran genomes, our results indicate that Sog's role to modify BMP activity varies greatly in different insect species.

11. Permeability, roughness and topography of enamel after bleaching: tracking channels of penetration with silver nitrate

Abstract

Aim: This study evaluated the surface roughness, topography and permeability of bovine enamel by profilometry and scanning electron microscopy (SEM) with and without silver nitrate solution, after exposure to different bleaching agents. Methods: Fifty-two enamel samples were randomly divided into four groups (n=13): CP16% –16% carbamide peroxide - Whiteness Perfect; HP6% - 6% hydrogen peroxide - White Class; HP35% - 35% hydrogen peroxide Whiteness HP Maxx; and Control - not bleached and kept in artificial saliva. For roughness analysis, average surface roughness (Ra) and flatness coefficient (Rku) parameters were used. The topography and permeability were examined by SEM. For permeability evaluation, the samples were immersed in a 50% silver nitrate solution and analyzed using a backscattered electron and secondary electron mode. Results: For the roughness (Ra) evaluation, Kruskal-Wallis and Wilcoxon Signed Ranks Test were used, showing an increase on the surface roughness in all bleached groups. The Rku parameter suggested changes on enamel integrity. The SEM micrographs indicated changes on enamel topography and different levels of silver nitrate penetration in the samples of the bleached groups. In the overall analysis, the bleaching agents promoted surface changes and higher silver nitrate penetration when compared to the control group. Conclusions: It may be concluded that different bleaching agents might alter the topography and roughness of enamel surface. Moreover, the higher infiltration of silver nitrate suggests an easier penetration path for the oxygen molecules into the dentin substrate.

Abstract

Since historical times, the inherent human fascination with pearls turned the freshwater pearl mussel Margaritifera margaritifera (Linnaeus, 1758) into a highly valuable cultural and economic resource. Although pearl harvesting in M. margaritifera is nowadays residual, other human threats have aggravated the species conservation status, especially in Europe. This mussel presents a myriad of rare biological features, e.g. high longevity coupled with low senescence and Doubly Uniparental Inheritance of mitochondrial DNA, for which the underlying molecular mechanisms are poorly known. Here, the first draft genome assembly of M. margaritifera was produced using a combination of Illumina Paired-end and Mate-pair approaches. The genome assembly was 2.4 Gb long, possessing 105,185 scaffolds and a scaffold N50 length of 288,726 bp. The ab initio gene prediction allowed the identification of 35,119 protein-coding genes. This genome represents an essential resource for studying this species' unique biological and evolutionary features and ultimately will help to develop new tools to promote its conservation.

Abstract

Coleoid cephalopod molluscs comprise squid, cuttlefish and octopuses, and represent nearly the entire diversity of modern cephalopods. Sophisticated adaptations such as the use of colour for camouflage and communication, jet propulsion and the ink sac highlight the unique nature of the group. Despite these striking adaptations, there are clear parallels in ecology between coleoids and bony fishes. The coleoid fossil record is limited, however, hindering confident analysis of the tempo and pattern of their evolution. Here we use a molecular dataset (180 genes, approx. 36 000 amino acids) of 26 cephalopod species to explore the phylogeny and timing of cephalopod evolution. We show that crown cephalopods diverged in the Silurian–Devonian, while crown coleoids had origins in the latest Palaeozoic.While the deep-sea vampire squid and dumbo octopuses have ancient origins extending to the Early Mesozoic Era, 242+38 Ma, incirrate octopuses and the decabrachian coleoids (10-armed squid) diversified in the Jurassic Period. These divergence estimates highlight the modern diversity of coleoid cephalopods emerging in the Mesozoic Marine Revolution, a period that also witnessed the radiation of most ray-finned fish groups in addition to several other marine vertebrates. This suggests that that the origin of modern cephalopod biodiversity was contingent on ecological competition with marine vertebrates.

Abstract

Birds are the most species-rich class of tetrapod vertebrates and have wide relevance across many research fields. We explored bird macroevolution using full genomes from 48 avian species representing all major extant clades. The avian genome is principally characterized by its constrained size, which predominantly arose because of lineage-specific erosion of repetitive elements, large segmental deletions, and gene loss. Avian genomes furthermore show a remarkably high degree of evolutionary stasis at the levels of nucleotide sequence, gene synteny, and chromosomal structure. Despite this pattern of conservation, we detected many non-neutral evolutionary changes in protein-coding genes and noncoding regions. These analyses reveal that pan-avian genomic diversity covaries with adaptations to different lifestyles and convergent evolution of traits.

Abstract

To better determine the history of modern birds, we performed a genome-scale phylogenetic analysis of 48 species representing all orders of Neoaves using phylogenomic methods created to handle genome-scale data. We recovered a highly resolved tree that confirms previously controversial sister or close relationships. We identified the first divergence in Neoaves, two groups we named Passerea and Columbea, representing independent lineages of diverse and convergently evolved land and water bird species. Among Passerea, we infer the common ancestor of core landbirds to have been an apex predator and confirm independent gains of vocal learning. Among Columbea, we identify pigeons and flamingoes as belonging to sister clades. Even with whole genomes, some of the earliest branches in Neoaves proved challenging to resolve, which was best explained by massive protein-coding sequence convergence and high levels of incomplete lineage sorting that occurred during a rapid radiation after the Cretaceous-Paleogene mass extinction event about 66 million years ago.

Abstract

Single crystals of 0.5% Cr3+-doped Cs2NaGaF6 were studied by means of x-ray diffraction and polarized Raman scattering. The crystal exhibits a unique stacking interaction, with a hexagonal structure with R3̄m symmetry. Polarized Raman spectra recorded at 16 and 300 K reveal sidebands that are ascribed to the local vibrational of the [CrF6] coordination unit, which differ markedly from those of the Cs2NaGaF6 host lattice. The results are compared to findings for other elpasolite lattices, and the room temperature luminescence quantum yields are discussed in terms of a delicate balance between electron-phonon strength and lattice distortion.

Abstract

Ceramic materials doped with magnetic ions that present emission in the visible and near-infrared spectral regions are very attractive due to their inherent tunability. Possible applications include their utilization as optoelectronic and display devices, as spintronic material, in signal transmission and information storage, in the fabrication of special papers, as dosimetric materials and room temperature solid state lasers. Materials doped with tetrahedrally coordinated Co2+ present wide bands originated from electronic transitions in the ionic unfilled 3d electronic shell. The Co2+ 3d electrons are outside of the ion core and therefore their optical properties are directly affected by static and dynamic properties of ligand anions. The magnesium gallate MgGa2O4 is a partially inverted spinel described as a AB2O4 material with two possible positions for A2+ and B3+ cations. Polycrystalline MgGa2O4 : Co2+ samples were produced by solid-state reactions between ultra-pure raw oxides MgO, β-Ga2O3 and the desired CoCO3 quantities. Photoluminescence data at room temperature and 77 K of MgGa2O4 polycrystalline samples doped with 0.1 and 1.0% of Co2+ are presented. The visible emission observed is attributed to the 4T1(4P) →4A2(4F) spin-allowed transition of Co2+ ions tetrahedrally coordinated by O2- ions. The photoluminescence intensity decreases with temperature, but 90% of the 77 K emission integrated intensity remains at room temperature. Moreover, from lifetime results we estimate that Co2+ emission quantum efficiency is about 1.0 at room temperature. We also observe that between 0.1 and 1.0% of Co2+ the luminescence intensity decreases. For 1.0% of Co2+ the luminescence intensity is 37% of the obtained for 0.1%. This fact is attributed to non-radiative transfer processes of impurity ion relaxation that become competitive at 1.0% of Co2+ and show that there is a strongly impurity-concentration luminescence dependence. We also observed that for the higher concentration the band is slightly shifted to the infrared region. Due to low impurity ion concentration it would not be expected an energy transfer between Co2+ ions, therefore non-radiative decay processes are responsible by temperature luminescence quenching. 39. Permeability, roughness and topography of enamel after bleaching: tracking channels of penetration with silver nitrate.

Abstract

Aim: This study evaluated the surface roughness, topography and permeability of bovine enamel by profilometry and scanning electron microscopy (SEM) with and without silver nitrate solution, after exposure to different bleaching agents. Methods: Fifty-two enamel samples were randomly divided into four groups (n=13): CP16% -16% carbamide peroxide - Whiteness Perfect; HP6% - 6% hydrogen peroxide - White Class; HP35% - 35% hydrogen peroxide Whiteness HP Maxx; and Control - not bleached and kept in artificial saliva. For roughness analysis, average surface roughness (Ra) and flatness coefficient (Rku) parameters were used. The topography and permeability were examined by SEM. For permeability evaluation, the samples were immersed in a 50% silver nitrate solution and analyzed using a backscattered electron and secondary electron mode. Results: For the roughness (Ra) evaluation, Kruskal-Wallis and Wilcoxon Signed Ranks Test were used, showing an increase on the surface roughness in all bleached groups. The Rku parameter suggested changes on enamel integrity. The SEM micrographs indicated changes on enamel topography and different levels of silver nitrate penetration in the samples of the bleached groups. In the overall analysis, the bleaching agents promoted surface changes and higher silver nitrate penetration when compared to the control group. Conclusions: It may be concluded that different bleaching agents might alter the topography and roughness of enamel surface. Moreover, the higher infiltration of silver nitrate suggests an easier penetration path for the oxygen molecules into the dentin substrate. [ABSTRACT FROM AUTHOR]

Abstract

In the last 100 years major depression has increased worldwide. In this study we provided coconut fat (CF, rich in saturated fatty acids) or fish oil (FO, rich in n-3 polyunsaturated fatty acids) to female rats throughout pregnancy and lactation and then to their offspring post-weaning and examined lipid brain profile and the possible effect of FO as antidepressant agent in the offspring in adulthood (F1). Rats were submitted to forced swimming test, elevated plus maze, Morris water maze and open field. Peroxidation rate in the cerebral cortex and hippocampus were measured. Docosahexaenoic acid (DHA) concentration in dam's milk, eicosapentaenoic acid (EPA) and DHA concentration in hippocampus and cerebral cortex from F1 rats FO supplemented increased significantly when compared to control (C) and CF rats. Arachidonic acid/EPA ratio in the cerebral cortex and hippocampus decreased in rats submitted to forced swimming test. Peroxidation rate were not different between the groups. Immobility time in the forced swimming test in FO group was reduced ( p <0.01) when compared to C and CF rats. We conclude that lifelong intake of FO was able to induce an antidepressant effect with EPA and DHA concentration increased in the cerebral cortex and hippocampus. [ABSTRACT FROM AUTHOR]
