This work presents an investigation of the degradation of paracetamol via heterogeneous photocatalysis, aiming to magnetically immobilize the catalyst in a continuous process. Catalyst immobilization was conducted on aggregated flower-like structures. The CoFe2O4@Nb5O2 catalyst was characterized using a Vibrating Sample Magnetometer (VSM). The effects of the magnetic immobilization of the catalyst, flow, residence time, adsorption, and photolysis were evaluated. Additionally, catalyst reuse cycles were analyzed. The results indicated that a longer residence time favors the degradation of paracetamol due to the increase in the contact time of the effluent catalyst. At a flow rate of 20 mL·min−1, a degradation of 27% was obtained. Photolysis and adsorption tests indicated that residence time was not an important factor for paracetamol degradation. For the photolysis test, in the first cycle, the values obtained were in the range of 6.0–8.5%. The adsorption results indicated ~10% removal.

Abstract How prostate cancer cells and their precursors mediate changes in the tumor microenvironment (TME) to drive prostate cancer progression is unclear, in part due to the inability to longitudinally study the disease evolution in human tissues. To overcome this limitation, we perform extensive single-cell RNA-sequencing (scRNA-seq) and molecular pathology of the comparative biology between human prostate cancer and key stages in the disease evolution of a genetically engineered mouse model (GEMM) of prostate cancer. Our studies of human tissues reveal that cancer cell-intrinsic activation of MYC signaling is a common denominator across the well-known molecular and pathological heterogeneity of human prostate cancer. Cell communication network and pathway analyses in GEMMs show that MYC oncogene-expressing neoplastic cells, directly and indirectly, reprogram the TME during carcinogenesis, leading to a convergence of cell state alterations in neighboring epithelial, immune, and fibroblast cell types that parallel key findings in human prostate cancer.

The present study deals with the reuse of agro-industrial waste with a specific focus on biochar (processed plant biomass or biochar) consisting of organic and inorganic waste biomass subjected to thermochemical processes. The objective of this work is to carry out a systematic review of the literature according to the Methodi Ordinatio methodology and select a bibliographic portfolio of high relevance to this study that makes it possible to present the concepts, applications and interest on the part of companies in including biochar in their processes, as well as addressing the environmental impacts linked to incorrect waste disposal. In this sense, biochar presents an interesting potential solution from both a waste management and environmental point of view. The current challenge is studies that prove economic viability.

ObjectivesAdverse childhood experiences (ACEs) are associated with mental health issues and substance use. Having a substance use disorder increases the risk of overdose (OD). Research on ACEs and risk of OD is limited. This study examined the relationship between ACE scores and a self-reported history of OD among patients in an addiction and mental health outpatient setting.MethodsThis single-center, cross-sectional design included adults in a dual-diagnosis addiction and mental health outpatient recovery and treatment program from November 2017 to August 2020. Patients (N = 115) were assessed with self-report questionnaires, which included ACEs and history of OD. Bivariate and multivariable logistic regression was used to determine factors associated with self-reported OD history. We assessed the reliability and validity of the ACEs scale.ResultsOf the 115 participants, 26 (22.6%) reported a past OD at intake. The mean ACE score for participants with an OD history, as compared with those with no history of OD, was 4.0 (standard deviation, 2.7) vs 2.3 (standard deviation, 2.2). In the multivariable regression, a higher ACE score was associated with history of OD (adjusted odds ratio, 1.23; 95% confidence interval, 1.00-1.50; P = 0.0456).ConclusionsGiven the observed association between OD and higher ACE scores, patients presenting for treatment in outpatient dual-diagnosis clinics should be screened for ACEs and OD history, providing the opportunity for treatment with trauma-informed care and/or referral to appropriate services.

Universal access to electricity is beneficial for the socio-economic development of a country and the development of smart communities. Unfortunately, the electrification of remote off-grid areas, especially in developing countries, is rather slow due to geographic and economic barriers. In the Philippines, specifically, many electrified off-grid areas are underserved, with access to electricity being limited to only a few hours a day. This is mainly due to the high dependence on diesel power plants (DPPs) for electrifying these areas. To address these problems, hybrid renewable energy systems (HRESs) have been considered good electrification alternatives and have been extensively studied for their techno-economic and financial feasibility for Philippine off-grid islands. In this work, articles published from 2012 to 2023 focusing on off-grid Philippine rural electrification were reviewed and classified based on their topic. The taxonomical analysis of collected studies shows that there is a saturation of works focusing on the technical and economic aspects of off-grid electrification. Meanwhile, studies focusing on environmental and socio-political factors affecting HRES off-grid electrification are lagging. A bibliographic analysis of the reviewed articles also showed that there is still a lack of a holistic approach in studying off-grid electrification in the Philippines. There are only a few works that extend beyond the typical techno-economic study. Research works focusing on environmental and socio-political factors are also mainly isolated and do not cross over with technical papers. The gap between topic clusters should be addressed in future works on off-grid electrification.

Solidity is an important parameter in the design, manufacture, and operation concerning energy yield and performance efficiency of VAWTs. This paper examined detailed aerodynamic analysis of solidity effects at constant Re and at low Reynolds numbers, the flow field and forces (lift, drag, and Torque coefficients) aerodynamics at various azimuth positions, and the changes in the performance and the flow field of VAWTs at a wide range of TSR. The simulation of two three-bladed VAWT configurations, σ = 0.26 (C = 0.03 m) and σ = 0.34 C = 0.04 m) over 1.5 ≤ λ ≤ 5.5 tip speed ratios on the CL, CD, CM, and speed were monitored and extracted, and the blades surrounding flow field is plotted from the CFD simulations. Performance (Cp -λ) curves, force hysteresis, and plots of the flow field for the turbines are matched at low λ = 2 5, high λ = 4, and Reynolds number to explain solidity effects on the VAWTs performance and aerodynamics thus providing good insight into the underlying aerodynamics for the differences in power coefficient between the two rotors in detail. This allowed the linking of the performance to the detailed force and flow field aerodynamics at various azimuth angles. It has been shown that the initiation of blade stall and stall vortices shedding started earlier on the σ = 0.26 VAWT than the σ =0.34 VAWT, thus affecting the force attained and ultimately limiting the rotor efficiency of σ = 0.26 compared with the σ =0.34 rotor and provided an improved understanding of solidity phenomenon that will help in the design, production and operation of VAWTs.

This paper documents the recent changes in the structure and estimation procedures of the SAMBA model, providing a complete description of the decision problems that each economic agent faces, the first order conditions that solve those problems, and the new techniques employed to estimate the model. This updated version of the model incorporates new features, such as involuntary unemployment, imported goods in the consumption bundle and a new identified vector auto-regressive process for the rest of the world. Reflecting these changes, the set of observables was expanded to include, for instance, participation rates in the labor market and an exogenous measure of output gap. In face of increased complexity and the large number of observables, the model was estimated using Sequential Monte Carlo (SMC) methods, allowing for a smaller sensitivity to the choice of priors.

Abstract Cyclic high-dose testosterone administration, known as bipolar androgen therapy (BAT), is a treatment strategy for patients with metastatic castration-resistant prostate cancer (mCRPC). Here, we report the results of a multicenter, single arm Phase 2 study (NCT03554317) enrolling 45 patients with heavily pretreated mCRPC who received BAT (testosterone cypionate, 400 mg intramuscularly every 28 days) with the addition of nivolumab (480 mg intravenously every 28 days) following three cycles of BAT monotherapy. The primary endpoint of a confirmed PSA50 response rate was met and estimated at 40% (N = 18/45, 95% CI: 25.7–55.7%, P = 0.02 one-sided against the 25% null hypothesis). Sixteen of the PSA50 responses were achieved before the addition of nivolumab. Secondary endpoints included objective response rate (ORR), median PSA progression-free survival, radiographic progression-free survival (rPFS), overall survival (OS), and safety/tolerability. The ORR was 24% (N = 10/42). Three of the objective responses occurred following the addition of nivolumab. After a median follow-up of 17.9 months, the median rPFS was 5.6 (95% CI: 5.4–6.8) months, and median OS was 24.4 (95% CI: 17.6–31.1) months. BAT/nivolumab was well tolerated, resulting in only five (11%) drug related, grade-3 adverse events. In a predefined exploratory analysis, clinical response rates correlated with increased baseline levels of intratumoral PD-1 + T cells. In paired metastatic tumor biopsies, BAT induced pro-inflammatory gene expression changes that were restricted to patients achieving a clinical response. These data suggest that BAT may augment antitumor immune responses that are further potentiated by immune checkpoint blockade.

Abstract Patients with prostate cancer (PC) generally do not respond favorably to immune checkpoint inhibitors, which may be due to a low abundance of tumor-infiltrating lymphocytes even when mutational load is high. Here, we identified a patient who presented with high-grade primary prostate cancer with two adjacent tumor nodules. While both nodules were mismatch repair-deficient (MMRd), exhibited pathogenic MSH2 and MSH6 alterations, had a high tumor mutational burden (TMB), and demonstrated high microsatellite instability (MSI), they had markedly distinct immune phenotypes. The first displayed a dense infiltrate of lymphocytes (“hot nodule”), while the second displayed significantly fewer infiltrating lymphocytes (“cold nodule”). Whole-exome DNA analysis found that both nodules shared many identical mutations, indicating that they were derived from a single clone. However, the cold nodule appeared to be sub-clonal relative to the hot nodule, suggesting divergent evolution of the cold nodule from the hot nodule. Whole-transcriptome RNA analysis found that the cold nodule demonstrated lower expression of genes related to antigen presentation (HLA) and, paradoxically, classical tumor immune tolerance markers such as PD-L1 (CD274) and CTLA-4. Immune cell deconvolution suggested that the hot nodule was enriched not only in CD8+ and CD4 + T lymphocytes, but also in M1 macrophages, activated NK cells, and γδ T cells compared to the cold nodule. This case highlights that MMRd/TMB-high PC can evolve to minimize an anti-tumor immune response, and nominates downregulation of antigen presentation machinery (HLA loss) as a potential mechanism of adaptive immune evasion in PC.

Recent studies have shown that chlorophyll sensitization can improve the performance of semiconductors like TiO2 in photocatalytic reactions and light-harvesting technologies, such as solar cells. Faced with the search for renewable energy sources and sustainable technologies, the application of this natural pigment has been gaining prominence. The present work addresses some of the main possibilities of chlorophyll-TiO2 combination, presenting the most relevant aspects affecting chlorophyll extraction and TiO2 sensitization.

Objective: The present study examined if identification with mainstream American culture (acculturation) and heritage culture (enculturation) are differentially associated with blackouts and other drinking consequences among male and female college students of color. Participants: Participants were college students (N = 150) who self-identified as a racial/ethnic minority and endorsed blackouts in the past year. Methods: Regression models were used to examine gender-by-acculturation/enculturation interaction effects on alcohol-induced blackout and other alcohol-related consequences. Results: While acculturation was not significantly associated with either drinking outcome, enculturation showed a significant relationship with blackout frequency. Gender moderated this relationship; greater enculturation was associated with increased blackout frequency among male but not female students. Conclusions: The present findings suggest the importance of considering the interplay between enculturation and gender in understanding alcohol use among college students of color. Men who endorse high levels of enculturation may be at an increased risk of experiencing negative drinking-related consequences.

BackgroundTelomeres are located at chromosomal termini and function to maintain genomic integrity. Telomere dysfunction is a well-recognized contributor to aging and age-related diseases, such as prostate cancer. Since telomere length is highly heritable, we postulate that stromal cell telomere length in the tissue of a particular solid organ may generally reflect constitutive stromal cell telomere length in other solid organs throughout the body. Even with telomere loss occurring with each round of cell replication, in general, telomere length in prostate stromal cells in mid-life would still be correlated with the telomere length in stromal cells in other organs. Thus, we hypothesize that prostate stromal cell telomere length and/or telomere length variability is a potential indicator of the likelihood of developing future solid cancers, beyond prostate cancer, and especially lethal cancer.MethodsTo explore this hypothesis, we conducted a cohort study analysis of 1,175 men who were surgically treated for prostate cancer and were followed for death, including from causes other than their prostate cancer.ResultsIn this cohort study with a median follow-up of 19 years, we observed that longer prostate stromal cell telomere length measured in tissue microarray spots containing prostate cancer was associated with an increased risk of death from other solid cancers. Variability in telomere length among these prostate stromal cells was possibly positively associated with risk of death from other solid cancers.ConclusionStudying the link between stromal cell telomere length and cancer mortality may be important for guiding the development of cancer interception and prevention strategies.

The evolution of the filamentation instability produced by two counter-streaming pair plasmas is studied with particle-in-cell (PIC) simulations in both one (1D) and two (2D) spatial dimensions. Radiation friction effects on particles are taken into account. During the nonlinear stage of the instability, a strong broadening of the particle energy spectrum occurs accompanied by the formation of a peak at twice their initial energy. A simple theory of the peak formation is presented. The presence of radiative losses does not change the dynamics of the instability but affects the structure of the particle spectra.
