Blood cells in Drosophila serve primarily innate immune responses. Various stressors influence blood cell homeostasis regarding both numbers and the proportion of blood cell types. The principle molecular mechanisms governing hematopoiesis are conserved amongst species and involve major signaling pathways like Notch, Toll, JNK, JAK/Stat or RTK. Albeit signaling pathways generally rely on the activity of protein kinases, their specific contribution to hematopoiesis remains understudied. Here, we assess the role of Serine/Threonine kinases with the potential to phosphorylate the transcription factor Su(H) in crystal cell homeostasis. Su(H) is central to Notch signal transduction, and its inhibition by phosphorylation impedes crystal cell formation. Overall, nearly twenty percent of all Drosophila Serine/Threonine kinases were studied in two assays, global and hemocyte-specific overexpression and downregulation, respectively. Unexpectedly, the majority of kinases influenced crystal cell numbers, albeit only a few were related to hematopoiesis so far. Four kinases appeared essential for crystal cell formation, whereas most kinases restrained crystal cell development. This group comprises all kinase classes, indicative of the complex regulatory network underlying blood cell homeostasis. The rather indiscriminative response we observed opens the possibility that blood cells measure their overall phospho-status as a proxy for stress-signals, and activate an adaptive immune response accordingly. In this FlyBook chapter, we present a survey of the current literature on the development of the hematopoietic system in Drosophila The Drosophila blood system consists entirely of cells that function in innate immunity, tissue integrity, wound healing, and various forms of stress response, and are therefore functionally similar to myeloid cells in mammals. The primary cell types are specialized for phagocytic, melanization, and encapsulation functions. As in mammalian systems, multiple sites of hematopoiesis are evident in Drosophila and the mechanisms involved in this process employ many of the same molecular strategies that exemplify blood development in humans. Drosophila blood progenitors respond to internal and external stress by coopting developmental pathways that involve both local and systemic signals. An important goal of these Drosophila studies is to develop the tools and mechanisms critical to further our understanding of human hematopoiesis during homeostasis and dysfunction.

Sea urchins are the best-known models in echinoderm immunology and only four coelomocytes – i.e. phagocytes, vibratile cells, and red and colorless spherulocytes – have traditionally been observed in these animals. However, recent studies have demonstrated that this number could be underestimated, which consequently may hinder a better understanding of sea urchin physiology. Here, we investigated the cells of one of the most known sea urchin genera in the world: the genus Paracentrotus. This genus comprises two species with distinct geographic distributions, thus providing an excellent scenario for comparative analyses. In this context, through an integrative approach consisting of living cells, cytological preparations, and scanning electron microscopy (SEM), we analyzed morphological, morphometric, and cytochemical characteristics of the coelomocytes of Paracentrotus lividus (Lamarck, 1816) and Paracentrotus gaimardi (Blainville, 1825). Seven different coelomic subpopulations were found in these species, including the traditional types and the granular spherulocyte, crystal cell, and progenitor cell. For all spherulocyte subpopulations, a set of morphologically similar cells were observed in both cytological and SEM preparations. Based on morphology, morphometry, and cytochemistry, we observed that these morphotypes could be organized in a sequence, which we interpreted as a maturation process. Then, we raise one hypothesis that explains how spherulocytes mature. Lastly, taking into account the all results achieved by this study, we discuss (1) how the morphological, morphometric, and cytochemical characteristics observed in Paracentrotus coelomocytes may be used to identify the coelomocytes of other sea urchins; (2) the physiological implications of our findings. Considering that P. lividus is one of the most studied sea urchins in the world, the new findings obtained here may shed new light on traditional aspects of sea urchin immunology (e.g. coelomocytes function), as well as stimulate research on new aspects of Echinoidea and even in Echinodermata immunology. 1 – The coelomocytes of the only two species belonging to the genus Paracentrotus were studied. 2 – The integrative approach used here allowed the identification of three new cell types. 3 – Morphological, morphometric, and cytochemical data revealed a putative maturation process for spherulocytes. 4 – The data obtained for Paracentrotus sea urchins will be useful to identify coelomocytes in other echinoids. With the growing importance of the black soldier fly (Hermetia illucens) for both sustainable food production and waste management as well as for science, a great demand of understanding its immune system arises. Here, we present the first description of the circulating larval hemocytes with special emphasis on uptake of microorganisms and distinguishing hemocyte types. With histological, zymographic, and cytometric methods and with a set of hemocyte binding lectins and antibodies, the hemocytes of H. illucens are identified as plasmatocytes, crystal cells, and putative prohemocytes. Total hemocyte counts (THC) are determined, and methods for THC determination are compared. Approximately 1100 hemocytes per microliter hemolymph are present in naive animals, while hemocyte density decreases dramatically shortly after wounding, indicating a role of hemocytes in response to wounding (and immune response in general). The determination of the relative abundance of each hemocyte type (differential hemocyte count, DHC) revealed that plasmatocytes are highly abundant, whereas prohemocytes and crystal cells make up only a small percentage of the circulating cells. Plasmatocytes are not only the most abundant but also the professional phagocytes in H. illucens. They rapidly engulf and take up bacteria both in vivo and in vitro, indicating a very potent cellular defense against invading pathogens. Larger bioparticles such as yeasts are also removed from circulation by phagocytosis, but slower than bacteria. This is the first analysis of the potent cellular immune response in the black soldier fly, and a first toolbox that helps to identify hemocyte (types) is presented. [ABSTRACT FROM AUTHOR]

Maintenance and differentiation of progenitor cells is essential for proper organ development and adaptation to environmental stress and injury. In Drosophila melanogaster, the haematopietic system serves as an ideal model for interrogating the function of signaling pathways required for progenitor maintenance and cell fate determination. Here we focus on the role of the Hippo pathway effectors Yorkie and Scalloped in mediating and facilitating Notch signaling-mediated lineage specification in the lymph gland, the primary center for haematopoiesis within the developing larva. We discuss the regulatory mechanisms which promote Notch activity during normal haematopoiesis and its modulation during immune challenge conditions. We provide additional evidence establishing the hierarchy of signaling events during crystal cell formation, highlighting the relationship between Yorkie, Scalloped and Lozenge, while expanding on the role of Yorkie in promoting hemocyte survival and the developmental regulation of Notch and its ligand, Serrate, within the lymph gland. Finally, we propose additional areas of exploration that may provide mechanistic insight into the environmental and non-cell autonomous regulation of cell fate in the blood system.

Identifying high‐performance non‐precious metal‐based catalysts at the cathode is a major challenge for future practical applications. Herein, a soft‐template route through a self‐assembly arrangement of sulfur sources was successfully developed, facilitating the anion exchange. In addition, compared with pristine cobalt disulfide synthesized without templates, the cobalt disulfide prepared using the new method presented a lattice shrinking phenomenon due to the hindrance of cobalt hydroxide crystal cell. Based on X‐ray absorption spectroscopy (XAS) and density functional theory (DFT) calculation, increased occupancy of eg orbitals was verified for the cobalt disulfide after shrinkage, which was the main factor for enhancing the intrinsic activity of the catalyst. Besides the microscopic morphologic structure, elementary composition, and the valence state of the elements, the possible growth process of the cobalt disulfide was also discussed in detail. As catalyst for the oxygen reduction reaction, CoS2 showed a similar half‐wave potential (0.81 vs. 0.84 V for Pt/C) and higher diffusion‐limiting current density (reaching 5.33 vs. 5.19 mA cm−2 for Pt/C) than a commercial Pt/C catalyst. Trichoplax adhaerens are simple animals with no nervous system, muscles or body axis. Nevertheless, Trichoplax demonstrate complex behaviors, including responses to the direction of the gravity vector. They have only six somatic cell types, and one of them, crystal cells, has been implicated in gravity reception. Multiple crystal cells are scattered near the rim of the pancake-shaped animal; each contains a cup-shaped nucleus and an intracellular crystal, which aligns its position according to the gravity force. Little is known about the development of any cell type in Trichoplax, which, in the laboratory, propagate exclusively by binary fission. Electron and light microscopy were used to investigate the stages by which crystal cells develop their mature phenotypes and distributions. Nascent crystal cells, identified by their possession of a small crystal, were located farther from the rim than mature crystal cells, indicating that crystal cells undergo displacement during maturation. They were elongated in shape and their nucleus was rounded. The crystal develops inside a vacuole flanked by multiple mitochondria, which, perhaps, supply molecules needed for the biomineralization process underlying crystal formation. This research sheds light on the development of unique cells with internal biomineralization and poses questions for further research.

Haematopoiesis is a complex process in which the regulatory mechanisms of several implicated transcription factors remain uncertain. Drosophila melanogaster is an excellent model to resolve the unanswered questions about the blood cell development. This study describes the role of Beadex, a Drosophila homologue of LIM domain only 2 (LMO2), in haematopoiesis. Mutants of Beadex were analysed for blood cell abnormalities. Crystal cells, a subset of haemocytes, were significantly more in Beadex hypermorphic flies. Similarly, Beadex misexpression in prohemocytes altered the crystal cell numbers. Stage-specific misexpression analyses demonstrated that Beadex functions after the prohemocytes enter the crystal cell lineage. We also discovered that Pannier–U-shaped complex is a negative regulator of the crystal cell differentiation and is possibly negatively regulated by Beadex through its interaction with Pannier. We, therefore, suggest the mechanism of two novel regulators of crystal cell specification—Beadex and Pannier—during Drosophila haematopoiesis. [ABSTRACT FROM AUTHOR]

Abstract The production of reactive oxygen species (ROS) is a prominent response to infection among innate immune cells such as macrophages and neutrophils. To better understand the relationship between antimicrobial and regulatory functions of blood cell ROS, we have characterized the ROS response to infection in Drosophila hemocytes. Using fluorescent probes, we find a biphasic hemocyte ROS response to bacterial infection. In the first hour, virtually all hemocytes generate a transient ROS signal, with nonphagocytic cells including prohemocytes and crystal cells displaying exceptionally strong responses. A distinct, and more delayed ROS response starting at 90 min is primarily within cells that have engulfed bacteria, and is sustained for several hours. The early response has a clear regulatory function, as dampening or intensifying the intracellular ROS level has profound effects on plasmatocyte activation. In addition, ROS are necessary and sufficient to activate JNK signalling in crystal cells, and to promote JNK-dependent crystal cell rupture. These findings indicate that Drosophila will be a promising model in which to dissect the mechanisms of ROS stimulation of immune activation. Symbiotic bacteria assist in maintaining homeostasis of the animal immune system. However, the molecular mechanisms that underlie symbiont-mediated host immunity are largely unknown. Tsetse flies (Glossina spp.) house maternally transmitted symbionts that regulate the development and function of their host’s immune system. Herein we demonstrate that the obligate mutualist, Wigglesworthia, up-regulates expression of odorant binding protein six in the gut of intrauterine tsetse larvae. This process is necessary and sufficient to induce systemic expression of the hematopoietic RUNX transcription factor lozenge and the subsequent production of crystal cells, which actuate the melanotic immune response in adult tsetse. Larval Drosophila’s indigenous microbiota, which is acquired from the environment, regulates an orthologous hematopoietic pathway in their host. These findings provide insight into the molecular mechanisms that underlie enteric symbiont-stimulated systemic immune system development, and indicate that these processes are evolutionarily conserved despite the divergent nature of host-symbiont interactions in these model systems.

In a niche under the skin in Drosophila larvae, blood cells called plasmatocytes can transform into other classes of blood cell.

Virtually all species of coelomate animals contain blood cells that display a division of labor necessary for homeostasis. This functional partition depends upon the balance between proliferation and differentiation mostly accomplished in the hematopoietic organs. In Drosophila melanogaster, the lymph gland produces plasmatocytes and crystal cells that are not released until pupariation. Yet, throughout larval development, both hemocyte types increase in numbers. Mature plasmatocytes can proliferate but it is not known if crystal cell numbers increase by self-renewal or by de novo differentiation. We show that new crystal cells in third instar larvae originate through a Notch-dependent process of plasmatocyte transdifferentiation. This process occurs in the sessile clusters and is contingent upon the integrity of these structures. The existence of this hematopoietic tissue, relying on structure-dependent signaling events to promote blood homeostasis, creates a new paradigm for addressing outstanding questions in Drosophila hematopoiesis and establishing further parallels with vertebrate systems.

Hematopoietic homeostasis requires the maintenance of a reservoir of undifferentiated blood cell progenitors and the ability to replace or expand differentiated blood cell lineages when necessary. Multiple signaling pathways function in these processes, but how their spatiotemporal control is established and their activity is coordinated in the context of the entire hematopoietic network are still poorly understood. We report here that loss of the gene Rabex-5 in Drosophila causes several hematopoietic abnormalities, including blood cell (hemocyte) overproliferation, increased size of the hematopoietic organ (the lymph gland), lamellocyte differentiation and melanotic mass formation. Hemocyte-specific Rabex-5 knockdown was sufficient to increase hemocyte populations, increase lymph gland size and induce melanotic masses. Rabex-5 negatively regulates Ras, and we show that Ras activity is responsible for specific Rabex-5 hematopoietic phenotypes. Surprisingly, Ras-independent Notch protein accumulation and transcriptional activity in the lymph gland underlie multiple distinct hematopoietic phenotypes of Rabex-5 loss. Thus, Rabex-5 plays an important role in Drosophila hematopoiesis and might serve as an axis coordinating Ras and Notch signaling in the lymph gland. [ABSTRACT FROM AUTHOR]

The invertebrate Drosophila melanogaster has been a powerful model for understanding blood cell development and immunity. Drosophila is a holometabolous insect, which transitions through a series of life stages from embryo, larva and pupa to adulthood. In spite of this, remarkable parallels exist between Drosophila and vertebrate macrophages, both in terms of development and function. More than 90% of Drosophila blood cells (hemocytes) are macrophages (plasmatocytes), making this highly tractable genetic system attractive for studying a variety of questions in macrophage biology. In vertebrates, recent findings revealed that macrophages have two independent origins: self-renewing macrophages, which reside and proliferate in local microenvironments in a variety of tissues, and macrophages of the monocyte lineage, which derive from hematopoietic stem or progenitor cells. Like vertebrates, Drosophila possesses two macrophage lineages with a conserved dual ontogeny. These parallels allow us to take advantage of the Drosophila model when investigating macrophage lineage specification, maintenance and amplification, and the induction of macrophages and their progenitors by local microenvironments and systemic cues. Beyond macrophage development, Drosophila further serves as a paradigm for understanding the mechanisms underlying macrophage function and cellular immunity in infection, tissue homeostasis and cancer, throughout development and adult life. Melanization contributes to arthropod-specific innate immunity through deposition of melanin at wound sites or around parasites, with concomitant release of microbicidal reactive oxygen species. Melanization requires sequential activation of proteolytic enzymes in the hemolymph, including the final enzyme pro-phenoloxidase. Black cells ( Bc ) is a mutation causing spontaneous melanization of Drosophila crystal cells, a hemocyte cell type producing phenoloxidases. Bc individuals exhibit circulating black spots but fail to melanize upon injury. Although Bc is widely used as a loss-of-function mutant of phenoloxidases, the mutation causing Bc remained unknown. Here, we identified a single point mutation in the pro-phenoloxidase 1 ( PPO1 ) gene of Bc flies causing an Alanine to Valine change in the C-terminal domain of PPO1, predicted to affect the conformation of the N-terminal pro-domain cleavage site at a distance and causing uncontrolled catalytic activity. Genomic insertion of a PPO1 A480V transgene phenocopies Black cells , proving that A480V is indeed the causal mutation of the historical Bc phenotype. [ABSTRACT FROM AUTHOR]

Single crystals of ZnP2 with a developed natural faceting characteristic of tetragonal symmetry were grown for the first time using a resublimation method. The atomic composition and crystal cell constants of t-ZnP2 tetragonal crystals were determined. Structures that feature the straightening and photovoltaic effect covered the natural faces of the crystals. Studies of the photosensitivity of the obtained structures in natural and linearly polarized radiation made it possible to elucidate the character of interband transitions and to estimate the forbidden-band gap for the t-ZnP2 crystals. Natural photopleochroism has been found and studied in structures based on single crystals with the natural faceting.
