The pervasiveness of deep space radiation remains a confounding factor for the transit of humans through our solar system. Spacecraft shielding both protects astronauts but also contributes to absorbed dose through galactic cosmic ray interactions that produce secondary particles. The resultant biological effects drop to a minimum for aluminum shielding around 20 g/cm2 but increase with additional shielding. The present work evaluates for the first time, the impact of secondary pions on central nervous system functionality. The fractional pion dose emanating from thicker shielded spacecraft regions could contribute up to 10% of the total absorbed radiation dose. New results from the Paul Scherrer Institute have revealed that low dose exposures to 150 MeV positive and negative pions, akin to a Mars mission, result in significant, long-lasting cognitive impairments. These surprising findings emphasize the need to carefully evaluate shielding configurations to optimize safe exposure limits for astronauts during deep space travel.

In this work, the properties of LiF crystals grown using Li of different isotopic compositions are described from the standpoint of their application as fluorescent nuclear track detectors used in measurements in the neutron radiation fields. The crystals were grown using two techniques: the Czochralski method and the micro-pulling-down method. Three isotopic compositions of Li were studied: natural, highly enriched in 6Li, and highly enriched in 7Li. It was found that 6LiF detectors are about six times more sensitive to thermal (low energy) neutrons than natural LiF, which significantly decreases the lower detection limit. 7LiF detectors are insensitive to thermal neutrons, which makes it easier to detect tracks due to other radiation modalities, such as energetic ions or nuclei recoiled in collisions with high-energy neutrons. Besides the response to neutron radiation, no other significant differences in the crystal properties were identified, irrespective of the isotopic composition and crystal growth method employed. Background: Flight attendants face various risk factors in their working environments, particularly occupational exposure to cosmic radiation. This study aimed to assess cancer risk among air transportation industry workers, including flight attendants, in Korea by constructing a cohort using national health registry-based data and analyzing cancer incidence risk. Methods: We used the Korea National Health Insurance Service database from 2002 to 2021 to construct a cohort of 37,011 workers in the air transportation industry. Cancer incidence was defined using the tenth version of the International Classification of Diseases. We calculated the age- and sex-specific standardized incidence ratios (SIRs) and 95% confidence intervals (CIs) by applying the cancer incidence rate of the general population between 2002 and 2019. Results: Approximately 5% of the cohort developed cancer. Overall, the cancer incidence in the cohort was similar to or lower than that of the general population, with the SIRs for all cancers being lower. However, significantly higher SIRs were observed for nasopharyngeal cancer (SIR, 3.21; 95% CI, 1.71–5.48) and non-Hodgkin lymphoma (SIR, 1.57; 95% CI, 1.02–2.32) in male workers and breast and genital cancer (SIR, 1.51; 95% CI, 1.34–1.70) and thyroid cancer (SIR, 1.25; 95% CI, 1.05–1.47) in female workers. Conclusions: The lower overall cancer incidence among air transportation industry workers observed in this study could indicate the "healthy worker effect"; however, the incidences of certain cancers were higher than those in the general population. Given that these workers are exposed to multiple occupational and lifestyle-related risk factors, including cosmic radiation, further studies are necessary to determine radiation-induced cancer risk while considering potential confounding factors. Purpose: Space exploration poses unique challenges to astronauts, especially the effects of space radiation and microgravity (μG). Understanding molecular responses to these factors is crucial for ensuring astronaut health, and this study aimed to identify transcriptomic changes in mouse hypothalamic cell line N38 (mHypoE-N38) caused by simulated space environments. Method: Four experimental groups were established, namely, a ground condition group (GC; the control group), a proton irradiated group (the space radiation group), a simulated μG group, and a proton irradiated × simulated μG group (the combination group). RNA sequencing and quantitative real-time polymerase chain reaction were performed to investigate key altered genes and to validate them. Results: Three hundred and fifty-five differentially expressed genes were identified. Notably, the expressions of UCN2 and UGT1A5 genes were upregulated in all three experimental groups, suggesting a shared regulatory mechanism with potential consequences for brain function during space missions. Moreover, the study revealed significant alterations in genes belonging to the USP17 and ZSCAN4 families, indicating active response to DNA damage and telomere maintenance. PCR results validated that UGT1A5, USP17 family, and ZSCAN4 families (ZSCAN4C, ZSCAN4D, and ZSCAN4F) were significantly upregulated at the mRNA level in the combination group, while UCN2, ZSCAN4A, and ZSCAN4B were not reproduced. Conclusion: The present study on mHypoE-N38 cells exposed to space environments revealed a complex molecular narrative with disease-oriented implications. The knowledge gained might serve as a cornerstone for developing strategies to mitigate potential health risks associated with extended exposure to space-related stressors.

For missions beyond low Earth orbit to the moon or Mars, space explorers will encounter a complex radiation field composed of various ion species with a broad range of energies. Such missions pose significant radiation protection challenges that need to be solved in order to minimize exposures and associated health risks. An innovative galactic cosmic ray simulator (GCRsim) was recently developed at the NASA Space Radiation Laboratory (NSRL) at Brookhaven National Laboratory (BNL). The GCRsim technology is intended to represent major components of the space radiation environment in a ground analog laboratory setting where it can be used to improve understanding of biological risks and serve as a testbed for countermeasure development and validation. The current GCRsim consists of 33 energetic ion beams that collectively simulate the primary and secondary GCR field encountered by humans in space over the broad range of particle types, energies, and linear energy transfer (LET) of interest to health effects. A virtual workshop was held in December 2020 to assess the status of the NASA baseline GCRsim. Workshop attendees examined various aspects of simulator design, with a particular emphasis on beam selection strategies. Experimental results, modeling approaches, areas of consensus, and questions of concern were also discussed in detail. This report includes a summary of the GCRsim workshop and a description of the current status of the GCRsim. This information is important for future advancements and applications in space radiobiology.

This article summarizes a Symposium on Radiation risks of the central nervous system held virtually at the 67th Annual Meeting of the Radiation Research Society, 3-6 October 2021. Repeated low-dose radiation exposure over a certain period could lead to reduced neuronal proliferation, altered neurogenesis, neuroinflammation and various neurological complications, including psychological consequences, necessitating further research in these areas. Four speakers from radiation biology, genetics and epidemiology presented the latest data from their studies seeking insights into this important topic. This symposium highlighted new and important directions for further research on mental health disorders, neurodegenerative conditions and cognitive impairment. Future studies will examine risks of mental and behavioral disorders and neurodegenerative diseases following protracted radiation exposures to better understand risks of occupational exposures as well as provide insights into risks from exposures to galactic cosmic rays.

Astronauts will encounter extended exposure to galactic cosmic radiation (GCR) during deep space exploration, which could impair brain function. Here, we report that in male mice, acute or chronic GCR exposure did not modify reward sensitivity but did adversely affect attentional processes and increased reaction times. Potassium (K+)-stimulation in the prefrontal cortex (PFC) elevated dopamine (DA) but abolished temporal DA responsiveness after acute and chronic GCR exposure. Unlike acute GCR, chronic GCR increased levels of all other neurotransmitters, with differences evident between groups after higher K+-stimulation. Correlational and machine learning analysis showed that acute and chronic GCR exposure differentially reorganized the connection strength and causation of DA and other PFC neurotransmitter networks compared to controls which may explain space radiation-induced neurocognitive deficits.

Long-duration space exploratory missions to the Earth's moon and the planet Mars are actively being planned. Such missions will require humans to live for prolonged periods beyond low earth orbit where astronauts will be continuously exposed to high energy galactic cosmic rays (GCRs). A major unknown is the potential impact of GCRs on the risks of developing degenerative cardiovascular disease, which is a concern to NASA. A ground-based rat model has been used to provide a detailed characterization of the risk of long-term cardiovascular disease from components of GCRs at radiation doses relevant to future human missions beyond low earth orbit. Six month old male WAG/RijCmcr rats were irradiated at a ground-based charged particle accelerator facility with high energy ion beams broadly representative of GCRs: protons, silicon and iron. Irradiation was given either as a single ion beam or as a combination of three ion beams. For the doses used, the single ion beam studies did not show any significant changes in the known cardiac risk factors and no evidence of cardiovascular disease could be demonstrated. In the three ion beam study, the total cholesterol levels in the circulation increased modestly over the 270 day follow up period, and inflammatory cytokines were also increased, transiently, 30 days after irradiation. Perivascular cardiac collagen content, systolic blood pressure and the number of macrophages found in the kidney and in the heart were each increased 270 days after irradiation with 1.5 Gy of the three ion beam grouping. These findings provide evidence for a cardiac vascular pathology and indicate a possible threshold dose for perivascular cardiac fibrosis and increased systemic systolic blood pressure for complex radiation fields during the 9 month follow up period. The development of perivascular cardiac fibrosis and increased systemic systolic blood pressure occurred at a physical dose of the three ion beam grouping (1.5 Gy) that was much lower than that required to show similar outcomes in earlier studies with the same rat strain exposed to photons. Further studies with longer follow up periods may help determine whether humans exposed to lower, mission-relevant doses of GCRs will develop radiation-induced heart disease.

Interplanetary space travel poses many hazards to the human body. To protect astronaut health and performance on critical missions, there is first a need to understand the effects of deep space hazards, including ionizing radiation, confinement, and altered gravity. Previous studies of rodents exposed to a single such stressor document significant deficits, but our study is the first to investigate possible cumulative and synergistic impacts of simultaneous ionizing radiation, confinement, and altered gravity on behavior and cognition. Our cohort was divided between 6-month-old female and male mice in group, social isolation, or hindlimb unloading housing, exposed to 0 or 50 cGy of 5 ion simplified simulated galactic cosmic radiation (GCRsim). We report interactions and independent effects of GCRsim exposure and housing conditions on behavioral and cognitive performance. Exposure to GCRsim drove changes in immune cell populations in peripheral blood collected early after irradiation, while housing conditions drove changes in blood collected at a later point. Female mice were largely resilient to deficits observed in male mice. Finally, we used principal component analysis to represent total deficits as principal component scores, which were predicted by general linear models using GCR exposure, housing condition, and early blood biomarkers.

This paper describes events of anomalously high energy transfer to a micro-object by fragments of nuclei generated in nuclear interactions in the environment on board a spacecraft in flight in low-Earth orbit. An algorithm has been developed that allows for the calculation of the absorbed energy from one or more fragments - products of nuclear interaction. With this algorithm the energy distributions for a spherical micro-volume in an aqueous medium were calculated. And the resulting absorbed energy spectra from nuclear fragments and from primary cosmic rays were compared. The role of nuclear interactions in events of large energy transfers in micro-objects in the field of primary cosmic radiation has been evaluated. The calculations performed in this study showed that the energy in a micro-volume from nuclear events can be several times higher compared to the energy imparted by primary space radiation. [Display omitted] The scientific paper presents studies of the radiation-protective features of a polymer composite material for protection against space radiation based on poly-tetra-fluoro-ethylene, Bi 2 O 3 bismuth oxide, WC tungsten carbide, B 4 C boron carbide, TiH 1.7 titanium hydride shot. The composite provides for a high degree of protection against ionizing radiation, including protection against exposure to secondary neutrons: macroscopic fast neutrons removal cross-section (E > 2 MeV) 0.1331–0.1414 cm−1, thermal-velocity neutrons removal cross-section (E < 0.4 eV), cm−1 0.1183–0.1257 cm−1. A shield made of the tailored composition of the proposed 6 cm thick polymer composite attenuates the intensity of neutron radiation by more than 50 % at energies from 0.1 to 5 MeV, in particular, the greatest attenuation of the neutron radiation intensity occurs at an energy of 0.1 MeV with a sharp increase in attenuation when passing through a 2 cm thick shield and complete absorption occurs passing through a 6 cm thick shield. This material can be used to protect astronauts and radio-electronic equipment from the effects of adverse or harmful space factors, including exposure to secondary neutrons, and this material can also be used to protect crews of ships with mobile nuclear reactors such as atomic submarines and nuclear-powered icebreakers. The climatic conditions on Mars are significantly different from those on Earth, which is the main factor affecting the potential architectural solutions on this planet. Above all, high levels of cosmic radiation of solar and extragalactic origin require specific architectural solutions, such as a safe location and sufficient protection, which are determined by a complex set of factors. These factors include the accessibility of building materials on site, the range and predictability of changes in climatic conditions, and human needs. In this article, the significant influence of radiation on architecture, which should be addressed when planning temporary or permanent resilient settlements on Mars, is examined in the case of manned missions to the planet. For this purpose, a comprehensive literature review is conducted, focused on the latest radiation-related discoveries. As the main outcome, two main schematic architectural solutions are proposed as a step toward more advanced projects. In addition, the gaps that need to be filled in the current approach to the design process, related to the insufficiently strong focus on multidisciplinary site analysis with radiation as its main factor (as is necessary for erecting an inhabitable base) are addressed.

Many years have passed since the first spaceflight, and in addition to the enormous technological progress, the level of understanding of human physiology in space is also increasing. The presented paper aims to summarize the recent research findings on the influence of the space environment (microgravity, pressure differences, cosmic radiation, etc.) on the human body systems during short-term and long-term space missions. The review also presents the biggest challenges and problems that must be solved in order to extend safely the time of human stay in space. In the era of increasing engineering capabilities, plans to colonize other planets, and the growing interest in commercial space flights, the most topical issues of modern medicine seems to be understanding the effects of long-term stay in space, and finding solutions to minimize the harmful effects of the space environment on the human body. In recent decades, the exposure of aviation crews and passengers to cosmic radiation has been progressively increasing due to the growing need of flights. Therefore, there is a need for radiation assessment software tools to act as a complement to other radiation protection techniques and countermeasures. In this work, the exposure to cosmic radiation is estimated for flights from Athens International Airport to various international destinations, by performing Monte Carlo simulations with the validated tool DYASTIMA/DYASTIMA-R. The results of the estimated total ambient dose equivalent, as well as the ambient dose equivalent rate for different flights, applying a typical flying level and constant atmospheric conditions, are presented for the first time. This study is carried out for different phases of solar activity for the time period from 1996 to 2019, which includes the two recent Solar Cycles 23 and 24. [ABSTRACT FROM AUTHOR]

The nucleosynthetic Cr isotope anomalies provides useful information to trace the source and origin of extraterrestrial samples, but it is usually influenced by high‐energy cosmic rays, and evaluating such effect of cosmic rays in lunar samples is especially important. Those cosmic radiation particles (primary particles) can react with lunar materials, creating many secondary particles. Both primary and secondary particles can produce cosmogenic nuclides on the Moon. Radiation Environment and Dose at the Moon (REDMoon) is a novel GEANT4 Monte‐Carlo model built to simulate the interactions of space particles with the lunar surface and subsurface content. Using this model, we simulate the production of cosmogenic Cr isotopes (50Cr, 52Cr, 53Cr, 54Cr) at different depths of lunar surface, and compare the contribution of different reactions generating these nuclides. The results suggest that spallation reactions are the most important process producing cosmogenic Cr isotopes. We also analyze the relationship between 53Cr/52Cr and 54Cr/52Cr predicted by our model and compare it with different Apollo samples. As previously studied, we also find an approximate linear relationship between ɛ53Cr and ɛ54Cr, where ɛ53Cr (or ɛ54Cr) is the relative deviation from the standard 53Cr/52Cr ratio (or 54Cr/52Cr ratio), normalized to 1/10,000. Furthermore, we reveal a change of this linear relationship in different depths of lunar surface. Besides, we investigate how the slopes can be influenced by exposure age and the Fe/Cr ratio. With these additional factors carefully considered, the comparison between our modeled results and the measurements is better than previous studies. Plain Language Summary: Cosmic rays arriving at the Moon can change the isotopic compositions on the lunar surface including chromium isotopes which can be used to constrain the evolution history of the Moon. Here, we use the particle transport model to simulate the high‐energy particles and the chromium isotopes generated in the lunar material by different cosmic ray sources. We compare different processes producing cosmogenic Cr isotopes and suggest that the spallation reactions play the main role. After taking the effect of solar energetic particles and the decay of cosmogenic 53Mn into consideration, we reach a better agreement between our model prediction and the Apollo lunar samples about the relationship between ɛ53Cr and ɛ54Cr. Such an improved analysis of the lunar cosmogenic nuclides contributes to a better understanding of the origin of the Moon. Recent spaceflights involving nonprofessional people have opened the doors to the suborbital space tourism business. However, they have also drawn public attention to the safety and hazards associated with space travel. Unfortunately, space travel involves a myriad of health risks for people, ranging from DNA damage caused by radiation exposure to the hemodynamic changes that occur when living in microgravity. In fact, the primary pathogenetic role is attributed to cosmic radiation, since deep space lacks the protective benefit of Earth's magnetic shielding. The second risk factor for space-induced pathologies is microgravity, which may affect organ function and cause a different distribution of fluid inside the human body. Both cosmic radiation and microgravity may lead to the alteration of cellular homeostasis and molecular changes in cell function. These, in turn, might have a direct impact on heart function and structure. The aim of this review is to draw attention to the fact that spaceflights constitute a novel frontier in biomedical research. We summarize the most important clinical and experimental evidence regarding the cardiovascular effects of cosmic radiation and microgravity. Finally, we highlight that unraveling the mechanisms underlying how space radiation and microgravity affect the cardiovascular system is crucial for identifying potential countermeasures and developing effective therapeutic strategies. During future space missions, astronauts will be exposed to cosmic radiation and microgravity (μG), which are known to be health risk factors. To examine the differentially expressed genes (DEG) and their prevalent biological processes and pathways as a response to these two risk factors simultaneously, 1BR-hTERT human fibroblast cells were cultured under 1 gravity (1G) or simulated μG for 48 h in total and collected at 0 (sham irradiated), 3 or 24 h after 1 Gy of X-ray or Carbon-ion (C-ion) irradiation. A three-dimensional clinostat was used for the simulation of μG and the simultaneous radiation exposure of the samples. The RNA-seq method was used to produce lists of differentially expressed genes between different environmental conditions. Over-representation analyses were performed and the enriched biological pathways and targeting transcription factors were identified. Comparing sham-irradiated cells under simulated μG and 1G conditions, terms related to response to oxygen levels and muscle contraction were identified. After irradiation with X-rays or C-ions under 1G, identified DEGs were found to be involved in DNA damage repair, signal transduction by p53 class mediator, cell cycle arrest and apoptosis pathways. The same enriched pathways emerged when cells were irradiated under simulated μG condition. Nevertheless, the combined effect attenuated the transcriptional response to irradiation which may pose a subtle risk in space flights. [ABSTRACT FROM AUTHOR]

There has been growing interest within the space industry for long‐duration manned expeditions to the Moon and Mars. During deep space missions, astronauts are exposed to high levels of galactic cosmic radiation (GCR) and microgravity which are associated with increased risk of oxidative stress and endothelial dysfunction. Oxidative stress and endothelial dysfunction are causative factors in the pathogenesis of erectile dysfunction, although the effects of spaceflight on erectile function have been unexplored. Therefore, the purpose of this study was to investigate the effects of simulated spaceflight and long‐term recovery on tissues critical for erectile function, the distal internal pudendal artery (dIPA), and the corpus cavernosum (CC). Eighty‐six adult male Fisher‐344 rats were randomized into six groups and exposed to 4‐weeks of hindlimb unloading (HLU) or weight‐bearing control, and sham (0Gy), 0.75 Gy, or 1.5 Gy of simulated GCR at the ground‐based GCR simulator at the NASA Space Radiation Laboratory. Following a 12–13‐month recovery, ex vivo physiological analysis of the dIPA and CC tissue segments revealed differential impacts of HLU and GCR on endothelium‐dependent and ‐independent relaxation that was tissue type specific. GCR impaired non‐adrenergic non‐cholinergic (NANC) nerve‐mediated relaxation in the dIPA and CC, while follow‐up experiments of the CC showed restoration of NANC‐mediated relaxation of GCR tissues following acute incubation with the antioxidants mito‐TEMPO and TEMPOL, as well as inhibitors of xanthine oxidase and arginase. These findings indicate that simulated spaceflight exerts a long‐term impairment of neurovascular erectile function, which exposes a new health risk to consider with deep space exploration.During deep space missions, astronauts are exposed to high levels of galactic cosmic radiation and microgravity which are associated with increased risk of oxidative stress and endothelial dysfunction. In this work, rats faced simulated spaceflight conditions. Ex vivo physiological analysis of the dIPA and CC revealed impaired vasoreactivity due to increased oxidative stress and arginase, resulting in diminished nitric oxide action. The GRACE – Follow On (GRACE-FO) satellite mission (2018-now) hosts the novel Laser Ranging Interferometer (LRI), a technology demonstrator for proving the feasibility of laser interferometry for inter-satellite ranging measurements. The GRACE-FO mission extends the valuable climate data record of changing mass distribution in the system Earth, which was started by the original GRACE mission (2002–2017). The mass distribution can be deduced from observing changes in the distance of two low-earth orbiters employing interferometry of electromagnetic waves in the K-Band for the conventional K-Band Ranging (KBR) and in near-infrared for the novel LRI. This paper identifies possible radiation-induced Single Event Upset (SEU) events in the LRI phase measurement. We simulate the phase data processing within the Laser Ranging Processor (LRP) and use a template-based fitting approach to determine the parameters of the SEU and subtract the events from the ranging data. Galactic cosmic radiation (GCR), composed of highly energetic and fully ionized atomic nuclei, produces diverse deleterious effects on the body. In researching the neurological risks of GCR exposures, including during human spaceflight, various ground-based single-ion GCR irradiation paradigms induce differential disruptions of cellular activity and overall behavior. However, it remains less clear how irradiation comprising a mix of multiple ions, more accurately recapitulating the space GCR environment, impacts the central nervous system. We therefore examined how mixed-ion GCR irradiation (two similar 5-6 beam combinations of protons, helium, oxygen, silicon and iron ions) influenced neuronal connectivity, functional generation of activity within neural circuits and cognitive behavior in mice. In electrophysiological recordings we find that space-relevant doses of mixed-ion GCR preferentially alter hippocampal inhibitory neurotransmission and produce related disruptions in the local field potentials of hippocampal oscillations. Such underlying perturbation in hippocampal network activity correspond with perturbed learning, memory and anxiety behavior.

The airborne radiation monitoring has been used in geophysics for more than forty years and now it also has its important role in emergency monitoring. The aircraft background and the cosmic gamma-rays contribute to the measured gamma spectrum on the aircraft board. This adverse effect should be eliminated before the data processing. The paper describes two semiparametric methods to estimate the full spectrum aircraft background and cosmic gamma-ray contribution from spectra measured at altitudes where terrestrial contribution is negligible. The methods only assume to know possible peak positions in spectra and their full width at half maximum, that can be easily obtained e.g. from terrestrial measurement. The methods were applied to real experimental data acquired on Mi-17 and Bell 412 helicopter boards. The IRIS airborne gamma-ray spectrometer, with 4 × 4 L NaI(Tl) crystals, produced by Pico Envirotec Inc., Canada, was used on helicopters’ boards. To obtain valid estimate of the aircraft background and the cosmic contribution, the measurements over sea and large water areas were carried out. However, the satisfactory results over inland were also achieved comparing with those acquired over large water areas.

Deep space travel presents a number of measurable risks including exposure to a spectrum of radiations of varying qualities, termed galactic cosmic radiation (GCR) that are capable of penetrating the spacecraft, traversing through the body and impacting brain function. Using rodents, studies have reported that exposure to simulated GCR leads to cognitive impairments associated with changes in hippocampus function that can persist as long as one-year post exposure with no sign of recovery. Whether memory can be updated to incorporate new information in mice exposed to GCR is unknown. Further, mechanisms underlying long lasting impairments in cognitive function as a result of GCR exposure have yet to be defined. Here, we examined whether whole body exposure to simulated GCR using 6 ions and doses of 5 or 30 cGy interfered with the ability to update an existing memory or impact hippocampal synaptic plasticity, a cellular mechanism believed to underlie memory processes, by examining long term potentiation (LTP) in acute hippocampal slices from middle aged male mice 3.5-5 months after radiation exposure. Using a modified version of the hippocampus-dependent object location memory task developed by our lab termed "Objects in Updated Locations" (OUL) task we find that GCR exposure impaired hippocampus-dependent memory updating and hippocampal LTP 3.5-5 months after exposure. Further, we find that impairments in LTP are reversed through one-time systemic subcutaneous injection of the histone deacetylase 3 inhibitor RGFP 966 (10 mg/kg), suggesting that long lasting impairments in cognitive function may be mediated at least in part, through epigenetic mechanisms.

Ionizing radiations encountered by astronauts on deep space missions produce biological damage by two main mechanisms: (1) Targeted effects (TE) due to direct traversals of cells by ionizing tracks. (2) Non-targeted effects (NTE) caused by release of signals from directly hit cells. The combination of these mechanisms generates non-linear dose response shapes, which need to be modeled quantitatively to predict health risks from space exploration. Here we used a TE + NTE model to analyze data on APC(1638N/+) mouse tumorigenesis induced by space-relevant doses of protons, 4He, 12C, 16O, 28Si or 56Fe ions, or γ rays. A customized weighted Negative Binomial distribution was used to describe the radiation type- and dose-dependent data variability. This approach allowed detailed quantification of dose-response shapes, NTE- and TE-related model parameters, and radiation quality metrics (relative biological effectiveness, RBE, and radiation effects ratio, RER, relative to γ rays) for each radiation type. Based on the modeled responses for each radiation type, we predicted the tumor yield for a Mars-mission-relevant mixture of these radiations, using the recently-developed incremental effect additivity (IEA) synergy theory. The proposed modeling approach can enhance current knowledge about quantification of space radiation quality effects, dose response shapes, and ultimately the health risks for astronauts.

The proposed deep space exploration to the moon and later to Mars will result in astronauts receiving significant chronic exposures to space radiation (SR). SR exposure results in multiple neurocognitive impairments. Recently, our cross-species (mouse/rat) studies reported impaired associative memory formation in both species following a chronic 6-month low dose exposure to a mixed field of neutrons (1 mGy/day for a total dose pf 18 cGy). In the present study, we report neutron exposure induced synaptic plasticity in the medial prefrontal cortex, accompanied by microglial activation and significant synaptic loss in the hippocampus. In a parallel study, neutron exposure was also found to alter fluorescence assisted single synaptosome LTP (FASS-LTP) in the hippocampus of rats, that may be related to a reduced ability to insert AMPAR into the post-synaptic membrane, which may arise from increased phosphorylation of the serine 845 residue of the GluA1 subunit. Thus, we demonstrate for the first time, that low dose chronic neutron irradiation impacts homeostatic synaptic plasticity in the hippocampal-cortical circuit in two rodent species, and that the ability to successfully encode associative recognition memory is a dynamic, multicircuit process, possibly involving compensatory changes in AMPAR density on the synaptic surface.

A recognized risk of long-duration space travel arises from the elevated exposure astronauts face from galactic cosmic radiation (GCR), which is composed of a diverse array of energetic particles. There is now abundant evidence that exposures to many different charged particle GCR components within acute time frames are sufficient to induce central nervous system deficits that span from the molecular to the whole animal behavioral scale. Enhanced spacecraft shielding can lessen exposures to charged particle GCR components, but may conversely elevate neutron radiation levels. We previously observed that space-relevant neutron radiation doses, chronically delivered at dose-rates expected during planned human exploratory missions, can disrupt hippocampal neuronal excitability, perturb network long-term potentiation and negatively impact cognitive behavior. We have now determined that acute exposures to similar low doses (18 cGy) of neutron radiation can also lead to suppressed hippocampal synaptic signaling, as well as decreased learning and memory performance in male mice. Our results demonstrate that similar nervous system hazards arise from neutron irradiation regardless of the exposure time course. While not always in an identical manner, neutron irradiation disrupts many of the same central nervous system elements as acute charged particle GCR exposures. The risks arising from neutron irradiation are therefore important to consider when determining the overall hazards astronauts will face from the space radiation environment.

BackgroundCosmic radiation exposures have been found to elicit cognitive impairments involving a wide-range of underlying neuropathology including elevated oxidative stress, neural stem cell loss, and compromised neuronal architecture. Cognitive impairments have also been associated with sustained microglia activation following low dose exposure to helium ions. Space-relevant charged particles elicit neuroinflammation that persists long-term post-irradiation. Here, we investigated the potential neurocognitive benefits of microglia depletion following low dose whole body exposure to helium ions.MethodsAdult mice were administered a dietary inhibitor (PLX5622) of colony stimulating factor-1 receptor (CSF1R) to deplete microglia 2 weeks after whole body helium irradiation (4He, 30 cGy, 400 MeV/n). Cohorts of mice maintained on a normal and PLX5622 diet were tested for cognitive function using seven independent behavioral tasks, microglial activation, hippocampal neuronal morphology, spine density, and electrophysiology properties 4-6 weeks later.ResultsPLX5622 treatment caused a rapid and near complete elimination of microglia in the brain within 3 days of treatment. Irradiated animals on normal diet exhibited a range of behavioral deficits involving the medial pre-frontal cortex and hippocampus and increased microglial activation. Animals on PLX5622 diet exhibited no radiation-induced cognitive deficits, and expression of resting and activated microglia were almost completely abolished, without any effects on the oligodendrocyte progenitors, throughout the brain. While PLX5622 treatment was found to attenuate radiation-induced increases in post-synaptic density protein 95 (PSD-95) puncta and to preserve mushroom type spine densities, other morphologic features of neurons and electrophysiologic measures of intrinsic excitability were relatively unaffected.ConclusionsOur data suggest that microglia play a critical role in cosmic radiation-induced cognitive deficits in mice and, that approaches targeting microglial function are poised to provide considerable benefit to the brain exposed to charged particles.

In this study, we investigated the levels of background radiation in different locations across the Eastern part of Koshi province, specifically in Taplejung, Panchthar, Ilam, and Jhapa. We used a portable Geiger Muller counter to collect data from twenty different locations, with five sites taken from each district. The average absorbed dose rate was found to be 0.243±0.035 mSv/y. The highest measured value of absorbed dose was 0.335±0.041 mSv/y at Pathivara temple in Taplejung, and the lowest was found to be 0.197±0.039 mSv/y at Kakarvita, Jhapa. The results suggest that these four districts do not pose any radiation risk because it was below the threshold of risk (1mSv/y). We also measured the variation of absorption dose with altitude which is positively correlated with altitude with a correlation coefficient of +0.57. This might be because of the surge in cosmic radiation with an increase in altitude.

Melanins are complex pigments with various biological functions and potential applications in space exploration and biomedicine due to their radioprotective properties. Aspergillus niger, a fungus known for its high radiation resistance, is widely used in biotechnology and a candidate for melanin production. In this study, we investigated the production of fungal pyomelanin (PyoFun) in A. niger by inducing overproduction of the pigment using L-tyrosine in a recombinant 1hmgA mutant strain (OS4.3). The PyoFun pigment was characterized using three spectroscopic methods, and its antioxidant properties were assessed using a DPPH-assay. Additionally, we evaluated the protective effect of PyoFun against non-ionizing radiation (monochromatic UV-C) and compared its efficacy to a synthetically produced control pyomelanin (PyoSyn). The results confirmed successful production of PyoFun in A. niger through inducible overproduction. Characterization using spectroscopicmethods confirmed the presence of PyoFun, and the DPPH-assay demonstrated its strong antioxidant properties. Moreover, PyoFun exhibited a highly protective effect against radiation-induced stress, surpassing the protection provided by PyoSyn. The findings of this study suggest that PyoFun has significant potential as a biological shield against harmful radiation. Notably, PyoFun is synthesized extracellularly, differing it from other fungal melanins (such as L-DOPA- or DHN-melanin) that require cell lysis for pigment purification. This characteristic makes PyoFun a valuable resource for biotechnology, biomedicine, and the space industry. The field of cosmic radiation at low-Earth orbit (LEO) has a complex composition. It always contains a component of secondary charged particles, formed by the products of nuclear interactions of the primary high-energy radiation with the nuclei of spacecraft's shielding material, electronic components and biological matter on board. Generation of this secondary radiation can be observed in some track detectors in the form of "stars" formed by tracks—fragments with a common vertex. The energy absorbed by the medium in the region adjacent to the interaction vertex can reach abnormally high values because of its intersection by several particle fragments. In the present paper, a methodology is considered to calculate the energy imparted by such fragments to a spherical sensitive volume in an aqueous medium. The energy distributions for three fragment events were calculated for different positions of the vertex relative to the spherical volume. The obtained data were analyzed and were compared with the distribution for a uniform fluence of secondary particles. It was shown that as the fragmentation vertex approaches the boundary of the sensitive micro-volume, the probability of events with anomalously high energy transfers, higher than the energies from single fragments, increases. The method can be applied to calculate absorbed energy distributions from secondary radiation in media of different elemental composition than that used in the present work. In the future, it is of interest to apply the method for example to study the energy imparted from secondary fragments to a silicon medium, to quantify the number of single event upsets in electronic components. Health risks from galactic cosmic rays (GCR) in space travel above low earth orbit remain a concern. For many years accelerator experiments investigating space radiation induced prevalence of murine Harderian gland (HG) tumorigenesis have been performed to help estimate GCR risks. Most studies used acute, relatively low fluence, exposures. Results on a broad spectrum of individual ions and linear energy transfers (LETs) have become available. However, in space, the crew are exposed simultaneously to many different GCR. Recent upgrades at the Brookhaven NASA Space Radiation Laboratory (NSRL) now allow mixtures in the form of different one-ion beams delivered in rapid sequence. This paper uses the results of three two-ion mixture experiments to illustrate conceptual, mathematical, computational, and statistical aspects of synergy analyses and also acts as an interim report on the mixture experiments' results. The results were interpreted using the following: (a) accumulated data from HG one-ion accelerator experiments; (b) incremental effect additivity synergy theory rather than simple effect additivity synergy theory; (c) parsimonious models for one-ion dose-effect relations; and (d), computer-implemented numerical methods encapsulated in freely available open source customized software. The main conclusions are the following. As yet, the murine HG tumorigenesis experimental studies show synergy in only one case out of three. Moreover, some theoretical arguments suggest GCR-simulating mixed beams are not likely to be synergistic. However, more studies relevant to possible synergy are needed by various groups that are studying various endpoints. Especially important is the possibility of synergy among high-LET radiations, since individual high-LET ions have large relative biological effectiveness for many endpoints. Airborne gamma-ray surveys are useful for many applications, ranging from geology and mining to public health and nuclear security. In all these contexts, the ability to decompose a measured spectrum into a linear combination of background source terms can provide useful insights into the data and lead to improvements in the techniques that use spectral energy windows. Multiple methods for the linear decomposition of spectra exist but are subject to various drawbacks, such as allowing negative photon fluxes or requiring detailed Monte Carlo modeling. We propose using non-negative matrix factorization (NMF) as a data-driven approach to spectral decomposition. Using aerial surveys that include flights over water, we demonstrate that the mathematical approach of NMF finds physically relevant structure in the aerial gamma-ray background, namely, that measured spectra can be expressed as the sum of nearby terrestrial emission, distant terrestrial emission, and radon and cosmic emission. These NMF background components are compared with the background components obtained by noise-adjusted singular value decomposition (NASVD), which contain negative photon fluxes and, thus, do not represent the emission spectra in as straightforward a way. Finally, we comment on the potential areas of research that are enabled by NMF decompositions, such as new approaches to spectral anomaly detection and data fusion.

Extended travel in deep space poses potential hazards to the reproductive function of female and male astronauts, including exposure to cosmic radiation, microgravity, increased gravity (hypergravity), psychological stress, physical stress and circadian rhythm disruptions. This Review focuses on the effects of microgravity, hypergravity and cosmic radiation. Cosmic radiation contains protons, helium nuclei and high charge and energy (HZE) particles. Studies performed on Earth in which rodents were exposed to experimentally generated HZE particles have demonstrated a high sensitivity of ovarian follicles and spermatogenic cells to HZE particles. Exposure to microgravity during space flight and to simulated microgravity on Earth disrupts spermatogenesis and testicular testosterone synthesis in rodents, whereas the male reproductive system seems to adapt to exposure to moderate hypergravity. A few studies have investigated the effects of microgravity on female reproduction, with findings of disrupted oestrous cycling and in vitro follicle development being cause for concern. Many remaining data gaps need to be addressed, including the effects of microgravity, hypergravity and space radiation on the male and female reproductive tracts, hypothalamic-pituitary regulation of reproduction and prenatal development of the reproductive system as well as the combined effects of the multiple reproductive hazards encountered in space.

Space weather can impede normal aviation operations through communication blackouts, GNSS‐based navigation and surveillance failures, and elevated cosmic radiation, consequently resulting in necessary flight plan adjustments and considerable economic costs. Although space weather effects have been heavily emphasized, the literature on the economic effects on aviation is limited. In this study, we estimate the economic impacts from the perspective of air traffic management, assuming an extremely strong space weather event like the 2003 Halloween solar storm would occur in 2019 with a booming air transport industry in recent years. We find that (a) as the high‐frequency communication blackouts may lead to polar flight rerouting and cancellations, possible daily economic costs could range from €0.21 million to €2.20 million per day; (b) during the satellite navigation failure period in the continental United States, as aircraft utilizes ground navigation aids as a backup, the increased flying time and disrupted descent approach operations may lead to additional cost of €2.43 million; (c) a surveillance failure can reduce airspace capacity and increase the workload of air traffic controllers, resulting in fatigue and perhaps risking flight safety; (d) to prevent massive cosmic radiation exposure, the economic costs of flight cancellations can be from €2.77 million to €48.97 million, depending on the cosmic radiation dose limits for a given plan. Our study indicates that severe space weather events may briefly disrupt normal aviation operations and cause substantial economic losses if future aviation equipment and technology are fragile to its effects. Plain Language Summary: Space weather events have the potential to have a significant economic impact on many vital national infrastructure systems, including electricity grids, the oil and gas industry, and Global Navigation Satellite Systems (GNSS). Particularly, space weather can impair normal aviation operations from high‐frequency communication blackouts, satellite navigation failure, GNSS‐based surveillance failure, and enhanced aviation radiation exposure. Consequently, air traffic management methods are required, and the economic effects of space weather on aviation are inevitable. Due to the lack of actual flight operation data during past space weather events, it is still unclear what the economic losses will be. Therefore, this study tries to estimate the economic costs based on the assumption that a space weather event like the Halloween solar storm of 2003 would occur in 2019. We can conclude that the cost caused by massive cosmic radiation dose is the highest due to flight cancellations. In contrast, the cost caused by satellite navigation failure is the least due to the backup of ground navigation systems. We believe this study can serve as a baseline for future space weather economic implications. In recent decades, the exposure of aviation crews and passengers to cosmic radiation has been progressively increasing due to the growing need of flights. Therefore, there is a need for radiation assessment software tools to act as a complement to other radiation protection techniques and countermeasures. In this work, the exposure to cosmic radiation is estimated for flights from Athens International Airport to various international destinations, by performing Monte Carlo simulations with the validated tool DYASTIMA/DYASTIMA-R. The results of the estimated total ambient dose equivalent, as well as the ambient dose equivalent rate for different flights, applying a typical flying level and constant atmospheric conditions, are presented for the first time. This study is carried out for different phases of solar activity for the time period from 1996 to 2019, which includes the two recent Solar Cycles 23 and 24.

During future space missions, astronauts will be exposed to cosmic radiation and microgravity (μG), which are known to be health risk factors. To examine the differentially expressed genes (DEG) and their prevalent biological processes and pathways as a response to these two risk factors simultaneously, 1BR-hTERT human fibroblast cells were cultured under 1 gravity (1G) or simulated μG for 48 h in total and collected at 0 (sham irradiated), 3 or 24 h after 1 Gy of X-ray or Carbon-ion (C-ion) irradiation. A three-dimensional clinostat was used for the simulation of μG and the simultaneous radiation exposure of the samples. The RNA-seq method was used to produce lists of differentially expressed genes between different environmental conditions. Over-representation analyses were performed and the enriched biological pathways and targeting transcription factors were identified. Comparing sham-irradiated cells under simulated μG and 1G conditions, terms related to response to oxygen levels and muscle contraction were identified. After irradiation with X-rays or C-ions under 1G, identified DEGs were found to be involved in DNA damage repair, signal transduction by p53 class mediator, cell cycle arrest and apoptosis pathways. The same enriched pathways emerged when cells were irradiated under simulated μG condition. Nevertheless, the combined effect attenuated the transcriptional response to irradiation which may pose a subtle risk in space flights.

As NASA prepares for a mission to Mars, concerns regarding the health risks associated with deep space radiation exposure have emerged. Until now, the impacts of such exposures have only been studied in animals after acute exposures, using dose rates ∼1.5×105 higher than those actually encountered in space. Using a new, low dose-rate neutron irradiation facility, we have uncovered that realistic, low dose-rate exposures produce serious neurocognitive complications associated with impaired neurotransmission. Chronic (6 month) low-dose (18 cGy) and dose rate (1 mGy/d) exposures of mice to a mixed field of neutrons and photons result in diminished hippocampal neuronal excitability and disrupted hippocampal and cortical long-term potentiation. Furthermore, mice displayed severe impairments in learning and memory, and the emergence of distress behaviors. Behavioral analyses showed an alarming increase in risk associated with these realistic simulations, revealing for the first time, some unexpected potential problems associated with deep space travel on all levels of neurological function.

There exists a wide degree of genetic variation within the normal human population which includes disease free individuals with heterozygote defects in major DNA repair genes. A lack of understanding of how this genetic variation impacts cellular phenotypes that inform cancer risk post heavy ion exposure poses a major limitation in developing personalized cancer risk assessment astronauts. We initiated a pilot study with Human Mammary Epithelial Cell strains (HMEC) derived from wild type, a p16 silenced derivative of wild type, and various genetic variants that were heterozygote for DNA repair genes; BRCA1, BRCA2 and ATM. Cells strains were exposed to different high and low LET radiation qualities to generate both simple and complex lesions and centrosome aberrations were examined as a surrogate marker of genomic instability and cancer susceptibility post different exposures. Our results indicate that centrosome aberration frequency is higher in the genetic variants under study. The aberration frequency increases with dose, complexity of the lesion generated by different radiation qualities and age of the individual. This increase in genomic instability correlates with elevated check-point activation post radiation exposure. These studies suggest that the influence of individual genetics on cell cycle regulation could modify the degree of early genomic instability in response to complex lesions and potentially define cancer predisposition in response to HZE exposure. These results will have significant implications in estimating cancer susceptibility in genetically variant individuals exposed to HZE particles.

ADA, short for Astroparticle Detectors Array, is an educational project aiming to detect cosmic radiation and possibly high-energy particles known as ultra-high-energy cosmic rays (UHECRs) or even to spot a supernova event. Its working process is the same as that used in professional cosmic ray observatories: it consists of simple detectors spread over the entire Italian territory and beyond. The detectors are hosted among high schools, associations, and private astronomical observatories. ADA has been operating since 2013 and was brought about with the intention of promoting astroparticle physics to any given level of outreach. Furthermore, ADA is becoming an interesting tool not only for teachers but also for independent and keen scientists. Over the years, we have discovered the importance of having a long series of data for studying the relation between and among cosmic rays, weather, and space weather and to investigate the main cause of oscillations in cosmic ray data. In this paper, we show what we find to be the most compelling results, such as the beautiful symmetry of the behavior between muons and the atmospheric temperature and, likewise, the evident anti-correlation between the intensity of the muons at ground level compared with solar activity. NUSES is a new project aiming to explore and test innovative technological and observational approaches for satellite-borne particle detectors dedicated to the study of cosmic radiation, astrophysical neutrinos, Sun-Earth environment, Space weather and Magnetospheric-Ionospheric-Lithospheric Coupling (MILC). The satellite will host two payloads, named TERZINA and ZIRÈ. TERZINA will test innovative observational techniques devoted to the hunt for astrophysical neutrino signals, looking at the Cherenkov light produced by extensive air showers generated in the atmosphere. The ZIRÈ experiment will exploit new technological approaches devoted to flux measurements of electrons, protons and light nuclei with energies spanning from few to hundreds of MeVs, but also to the detection of cosmic MeV photons and to the monitoring of eventual MILC signals. During extraordinary space weather, cosmic radiation can be significant enough to pose a threat to aircrew health. Traditional methods of reducing massive cosmic radiation exposure include flight cancellation, lowering flying altitudes, and flight rerouting. However, flight cancellation can result in additional financial expenditures, while lowering flight altitudes and rerouting can consequently cause more fuel consumption or even violation of airspace rights. As a result, we use a multi‐objective optimization model to assign optimal flight altitude and speed to reduce the overall weighted sum of cosmic radiation and fuel consumption. The simulation scenario is based on a space weather event with dramatically increased cosmic radiation that occurs during a routine international flight from Tokyo to London. Our results show that the proposed model can reduce fuel consumption while satisfying cosmic radiation limits recommended by the Council of the European Union if the forecasts of cosmic radiation are sufficiently accurate. In addition, a Pareto frontier is provided as a tactical air traffic management guideline. Our study provides insight into future policymaking for air transportation during harsh space weather conditions. Plain Language Summary: Cosmic radiation is made up of high‐energy protons and atomic nuclei from the Sun and distant galaxies. The radiation intensity at airplane cruising altitudes and high latitudes is higher, due to reduced protection from Earth's atmosphere and magnetic field, than that at low altitude and low latitude regions. Therefore, aircrews receive more radiation considering their long‐term exposure at high altitudes. The radiation level may increase significantly during severe space weather events. Traditionally airlines may cancel flights, reroute flights, or lower flight altitudes to mitigate radiation. However, these traditional methods have drawbacks like greater financial expenditures and fuel consumption and are sometimes even infeasible from air traffic management standpoint. To control the exposure to cosmic radiation and reduce fuel consumption while keeping normal aircraft operation, we propose a multi‐objective optimization model to assign flight altitudes and speeds along a multi‐segment route. The modeling is applied to an international flight from Tokyo to London assuming a space weather event similar to the one on 20 January 2005 occurs. We demonstrate that the proposed flight routes can successfully minimize fuel consumption while satisfying cosmic radiation limit standards. This can be a valuable example for air traffic management when dealing with space weather effects.
