194 9.-38 fig.-31 tab.-8 anexos., [EN], RISKS AND BENEFITS OF THE INSECT-RESISTANT Bt MAIZE (MON810) CULTIVATED IN SPAIN. The use of insect-resistant genetically modified (GM) crops which express insecticidal Cry proteins from Bacillus thuringiensis is an effective method to control pests of major economic importance. Nowadays, the only insect-resistant GM plants commercially cultivated in the EU are those maize varieties derived from the event MON810, engineered to express the Cry1Ab insecticidal protein (Bt maize). This toxin efficiently control two key pests: the Mediterranean corn borer, Sesamia nonagrioides (Lefèbvre) (Lepidoptera: Noctuidae) and the European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae). Immature stages of both species have endophytic behaviour, being the main damage caused by the stalk tunneling of larger larvae. As a consequence, Bt maize has shown high efficacy to control these pests, especially in areas where they are in high densities. Among the EU, Spain is the member state with the highest adoption rate of Bt maize, presenting more than 130.000 hectares in 2014 (representing the 30% of the total maize cultivated in the country). The extended use of Bt maize into agriculture has raised controversy in Europe, mainly due to the potential negative effects that the extended spread of these GM plants could cause to the environment. Despite Bt maize is highly specific to control these two corn borers, other arthropods associated with the GM plant that are not targeted (non-target arthropods) could be exposed and negatively affected by the toxin. So far, several laboratory and field studies have assessed the toxicity of the Cry1Ab protein against different functional guilds of non-target arthropods and their exposure to the toxin. Nevertheless, there are still certain groups and aspects associated with the cultivation of Bt maize that require more research, as the effects of Bt maize on non-target arthropods inhabiting the aerial parts of the maize and on the soil microarthropod communities, as well as their exposure to the insecticidal toxin in the field. Since not all the non-target arthropods can be tested, it has been proposed the selection of a representative subset of them (referred as surrogate species) for consideration in the risk assessment of GM plants. The species selection should be performed according to different selection criteria, such as: ecological function, geographical distribution, potential exposure to the toxin, economic relevance and feasibility to be taxonomically identified in the field and to perform bioassays in the laboratory., In Southern Europe, maize is a crop highly prone to suffer infections by moulds of the genera Fusarium and Aspergillus. During plant colonization they can produce root, stem and cob rot, and in addition they can synthesize secondary metabolites, including mycotoxins, which are highly toxic for mammals. The accumulation of mycotoxins in food represents a serious risk for human and animal health and causes important economical losses in the agrofood industry, due to reductions in the quality of the agricultural raw materials. To avoid the concern related with the presence of mycotoxins in the food chain, the EU has developed a restrictive and specific normative that provide the highest levels of these secondary metabolites that can be present in feed and food. Due to the complexity of removing these mycotoxins from the food at a low cost, without affecting the properties of the food, the EU have developed strategies to prevent the contamination by the moulds that produce them. The different species of moulds infecting a specific crop could have different toxigenic profiles. Thus, their taxonomical identification in a particular matrix could be useful to predict the mycotoxins that might occur. It has been described that the presence of insects, such as the corn borers, in the plants could favor their contamination with mycotoxigenic moulds, acting the larvae as disease vectors or penetrating the moulds into the plant through the wounds produced by them. Besides, mould proliferation could be magnified by the stress produced in the plants after they have been damaged by insects. In this context, a reduction in the concentration of mycotoxins could be possible, at least in part, if the damage by these pests is controlled, being the use of Bt maize an important control method. This relationship has been observed in different studied carried out in the past decade. Nevertheless, the relation between the attack of corn borers and the presence of fungal infections is not very obvious. Moreover, in Spain the epidemiological data referring to the mycotoxigenic moulds that contaminate maize are scarce. Based on the above, the general objectives of this Doctoral Thesis were: 1) To evaluate the effects of continuous cultivation of maize MON810 on the non-target soil microarthropods and on the community of arthropods present in the aerial part of the plant, assessing their exposure levels to the toxin, and identifying those taxa that fulfill the requirements to be employed as indicators in environmental risk assessments of Bt maize under Mediterranean agroecological conditions; and 2) to assess differences in levels of contamination with mycotoxigenic moulds and in the presence of mycotoxins, between cobs from Bt maize and insect damaged cobs from non-Bt maize, to determine whether the presence of the mycotoxigenic moulds is a good indicator of the contamination with mycotoxins and to analyze if the corn borers, O. nubilalis and S. nonagrioides act as vehicles of these moulds., Arthropods present in the aerial parts of the plants and the soil microarthropods were studied in an experimental farm-scale field in Madrid during three consecutive years (2009-2011), using yellow sticky traps and soil samples, respectively. Among the former, the most abundant insects were cicadellids (Hemiptera: Cicadellidae), delphacids (Hemiptera: Delphacidae), mymarids (Hymenoptera: Mymaridae), trichogrammatids (Hymenoptera: Trichogrammatidae), thrips (Thysanoptera) and chloropids (Diptera: Chloropidae). More than 64 % of the total captures were represented by the cicadellid Zyginidia scutellaris (herbivore) and the mymarid Anagrus spp. (parasitoid), closely related between them and with the maize plant. No negative effects of Bt maize were found on the abundance of these taxa, though differences among years were commonly present. We report for the first time the exposure to the protein Cry1Ab expressed in MON810-maize, in two cicadellids collected in the field, Z. scutellaris and Empoasca spp. Mites and collembolans were the most abundant edaphic microarthropods collected from soil samples, representing together the 88% of the total specimens collected. They were identified to suborder and species level, respectively. Mites and collembolans abundances and the frequency of occurrence of collembolan species did not rely on the type of maize, except for Parisotoma notabilis (Schäffer), which was more abundant and frequent in Bt maize. However, significant differences in abundance among years were common in both groups. Noticeably, we found higher values of species richness and diversity of collembolans in Cry1Ab-expressing Bt maize than in non-Bt plots. Moreover, the toxin was detected for the first time in collembolans from the genus Entomobrya collected in the field, evidencing that these microarthopods are exposed to the toxin expressed in MON810-maize. At the same time, we have detected the presence of Cry1Ab toxin in a fraction of the decaying soil organic matter (OM) from the rhizosphere soil samples from Bt maize plots. The toxin was detected up to three months after maize harvest, with values ranging between 0.10 and 0.18 ng Cry1Ab/mg OM. When the insecticidal activity of the OM purified toxin was evaluated, low toxicity against O. nubilalis was found, indicating low concentration in an active form. From the results presented herein, we propose the herbivorous Z. scutellaris, the parasitoid Anagrus spp. and the communities of soil mites and collembolans as appropriate taxonomic groups to be employed as indicators of the possible effects of Bt maize. This selection is based on their exposure to the Cry1Ab toxin in the field, regular presence in maize fields, feasibility to be sampled and taxonomically identified and efficacy to detect differences between non-Bt and Bt maize after the analysis of their abundances. Also, the species richness and diversity indices have proven to be useful in the detection of differences between both types of maize., During two consecutive seasons we have collected corncobs from non-Bt maize fields damaged by of corn borers and from Bt maize fields, in different fields located in three areas of Spain were maize is normally cultivated (Extremadura, Albacete y Valle del Ebro). The presence of 16 species of the genus Fusarium and Aspergillus was analyzed in the collected corncobs, by specific PCR protocols. Samples contaminated by F. verticillioides, F. proliferatum, F. subglutinans, F. graminearum, F. sporotrichioides, A. flavus and the aggregate A. niger were detected. The species belonging to the section Liseola, F. verticillioides, F. proliferatum y F. subglutinans, were the only that showed a higher frequency of occurrence in the samples from non- Bt maize. This is the first time that mycotoxigenic species, mainly from these three species of Fusarium, are isolated from larvae of the corn borers O. nubilalis and S. nonagrioides collected from commercial maize fields. Fumonisins were the mycotoxins more frequently found infecting the corncobs, being the damaged non-Bt ones significantly higher contaminated than corncobs from Bt maize. High levels of fumonisins have been related with the occurrence of corn borers in the corncob and with the presence of fungal species that produce these mycotoxins (F. verticillioides and F. proliferatum). The high frequency of occurrence presented by F. subglutinans in non-Bt corncobs rise alert about contamination of maize with moniliformin and other emerging mycotoxins. The frequency of occurrence of the rest of the moulds and mycotoxins (DON, T-2, HT-2, ZEA, OTA y aflatoxins) showed no relation with the type of maize cultivated. In summary, we can conclude that the continuous cultivation of MON810 maize did not negatively affect the arthropods present in the aerial parts of the plants and the soil microarthropods studied herein, despite the exposure to the Cry1Ab toxin was proved in field specimens belonging to both groups. The selection of key groups, to be used as indicators to detect negative effects of Bt maize proposed here (the herbivorous Z. scutellaris, the parasitoid Anagrus spp., and the detritivore communities of soil mites and collembolans), represents a practical tool to be implemented in risk assessment evaluation of GM plants, demanded by Europe for new events and for the post-market monitoring of Bt-maize (MON810). Moreover, due to the efficient control that Bt maize exert on corn borers, it represents an effective method to reduced the levels of mycotoxins in comparison with non-Bt maize. The isogenic maize damaged by corn borers was principally contaminated with fumonisins, which is related with the elevated presence of moulds of the section Liseola, main producers of these mycotoxins. The presence of the mycotoxigenic moulds F. verticillioides and F. proliferatum in plants damaged by the corn borers, O. nubilalis and S. nonagrioides has demonstrated to be a good indicator of contamination with fumonisins., [ES], RIESGOS Y BENEFICIOS DEL CULTIVO EN ESPAÑA DE MAÍZ Bt (MON810) RESISTENTE A INSECTOS. El uso de plantas genéticamente modificadas (GM) resistentes a insectos que expresan oxinas Cry de Bacillus thuringiensis ofrece un control altamente efectivo sobre plagas de gran importancia económica. Actualmente, la única planta GM resistente a insectos que se cultiva comercialmente en Europa es el maíz MON810 que expresa la toxina Cry1Ab (maíz Bt), activa frente a los taladros del maíz Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae) y Sesamia nonagrioides (Lefèbvre) (Lepidoptera: Noctuidae). Los daños de ambas especies son producidos por las larvas, que tienen un comportamiento endófito, es decir, penetran en el interior de la caña de la planta y se alimentan de su médula. Por este motivo, el maíz Bt es el método de control que se ha mostrado más eficaz en zonas con alta incidencia de dichas plagas. España es el único país de la UE que ha adoptado de forma significativa este cultivo, habiéndose sembrado en 2014 más de 130.000 ha, lo que representó el 30% del total de maíz. La incorporación del maíz Bt a la agricultura ha suscitado una gran controversia en Europa debido, entre otros motivos, a los posibles efectos que la liberación a gran escala de estas plantas puede producir sobre el medio ambiente. A pesar que el maíz Bt controla de manera específica a los taladros, los artrópodos asociados temporal y/o espacialmente al agroecosistema hacia los que no va dirigido este control (artrópodos nodiana), pueden estar expuestos a la toxina, y por tanto verse afectados por la misma. Hasta el momento se han realizado numerosos estudios de campo y de laboratorio con artrópodos de distintos grupos funcionales presentes en el cultivo del maíz para evaluar la toxicidad de la proteína Cry1Ab sobre ellos, así como el nivel de exposición al que pueden estar sometidos. Sin embargo, a pesar de toda la información recabada, aún quedan aspectos relevantes por conocer acerca de los efectos del cultivo del maíz Bt sobre algunos artrópodos no-diana que se encuentran en las partes aéreas de la planta y sobre los microartrópodos que habitan en el suelo, así como de su exposición a la toxina insecticida en el campo. Como no es posible llevar a cabo evaluaciones de riesgo ambiental de plantas GM para todas las especies presentes en un agroecosistema, se ha propuesto la utilización de especies indicadoras, sensibles a cambios en el medio ambiente o importantes por su papel ecológico, que actuarían como representantes de una comunidad de artrópodos determinada. En general, la selección de estas especies se basa en diversos criterios como su función ecológica, distribución geográfica, la posible exposición a la toxina, la facilidad para la identificación taxonómica en campo, el manejo en el laboratorio o la importancia económica., El maíz es un cultivo particularmente propenso en el sur de Europa a sufrir infecciones por hongos de los géneros Fusarium y Aspergillus. Al colonizar la planta algunos de estos hongos pueden causar podredumbre de raíces, tallos y mazorcas, además de sintetizar metabolitos secundarios, entre los que se encuentran las micotoxinas, que son altamente tóxicas para los mamíferos. La acumulación de toxinas en los alimentos representa un riesgo para la salud de los seres humanos y los animales, además de producir pérdidas económicas en la industria agroalimentaria al disminuir la calidad de la materia prima. Debido al problema que supone la entrada de micotoxinas a la cadena alimentaria, la UE ha desarrollado normativas específicas, cada vez más restrictivas, que marcan los niveles máximos permitidos de estos metabolitos en diferentes productos alimenticios. La dificultad de eliminar las micotoxinas de forma eficaz sin afectar a las propiedades de la materia prima con un bajo coste, hace que una de las estrategias para controlar su aparición sea la prevención de la contaminación con hongos productores de las mismas. Las distintas especies fúngicas que aparecen en un cultivo pueden presentar perfiles toxígenos diferentes, por lo que la identificación de los hongos productores en un alimento puede ser útil para predecir qué toxinas podrían aparecer. Se ha descrito que la entrada de insectos en la planta, como los taladros del maíz, puede favorecer la contaminación con hongos micotoxígenos, al actuar como vectores de la enfermedad o al penetrar los hongos a través de las heridas producidas. Además, la proliferación de los hongos se ve favorecida por el estrés producido en la planta por el daño de los insectos. Por ello, la reducción de la concentración de micotoxinas en el maíz sería, en parte, posible con la toma de medidas para controlar las plagas de taladros, siendo una de ellas el cultivo de maíz Bt. En la última década se ha relacionado el menor ataque de taladros en el maíz Bt con una disminución en los niveles de micotoxinas. Sin embargo, la asociación entre el ataque de taladros y la aparición de infecciones fúngicas no siempre está tan clara. Además, en España, los datos epidemiológicos sobre los hongos micotoxígenos que contaminan el maíz son escasos. Por todo lo expuesto, los objetivos generales de esta Tesis fueron: 1) evaluar los efectos del cultivo continuado de maíz MON810 sobre la comunidad de artrópodos de la parte aérea de la planta y los microartrópodos edáficos, analizando su nivel de exposición a la toxina e identificando los grupos más adecuados para ser utilizados en la evaluación de riesgo ambiental en condiciones agroclimáticas mediterráneas; y 2) estudiar las diferencias en la contaminación con hongos micotoxígenos y en la producción de micotoxinas entre mazorcas de maíz no-Bt con daño de taladro y maíz Bt, determinar si la presencia de los hongos micotoxígenos es un buen indicador de la contaminación con micotoxinas de las muestras y analizar el papel de O. nubilalis y S. nonagrioides como portadores de hongos productores de micotoxinas., Durante tres años consecutivos (2009-2011) se estudiaron los artrópodos de la parte aérea de la planta y los microartrópodos del suelo, recogidos mediante trampas adhesivas amarillas y muestras de suelo, respectivamente, de parcelas de maíz Bt y no- Bt en un campo experimental situado en San Fernando de Henares (Madrid). De los primeros, los más abundantes fueron los cicadélidos (Hemiptera: Cicadellidae), delfácidos (Hemiptera: Delphacidae), mimáridos (Hymenoptera: Mymaridae), tricogramátidos (Hymenoptera: Trichogrammatidae), tisanópteros (Thysanoptera) y clorópidos (Diptera: Chloropidae). Más del 64% del total de las capturas estuvieron compuestas por el cicadélido Zyginidia scutellaris (Herrich-Schäffer) (herbívoro) y el mimárido Anagrus spp. (parasitoide), que presentan una estrecha relación entre ellos y con la planta de maíz. No se encontraron efectos negativos del cultivo del maíz Bt en la abundancia de estos grupos, aunque en la mayoría de los casos hubo diferencias entre los años. Se demostró por primera vez la exposición, en condiciones de campo, de dos cicadélidos, Z. scutellaris y Empoasca spp., a la proteína Cry1Ab que expresa el maíz MON810. De los microartrópodos edáficos capturados a partir de las muestras de suelo recogidas a lo largo de todo el año, ácaros y colémbolos representaron el 88% de todos los individuos capturados, siendo identificados hasta nivel de suborden y especie, respectivamente. La abundancia de ácaros y colémbolos y la frecuencia de aparición de las principales especies de colémbolos no dependió del tipo de maíz, excepto para la especie Parisotoma notabilis (Schäffer), más abundante y frecuente en las parcelas de maíz Bt. Sin embargo, diferencias significativas entre años fueron comunes en ambos grupos. Además, se encontraron valores más altos de riqueza y diversidad de especies de colémbolos en las parcelas de maíz Bt que en las no-Bt. La toxina Cry1Ab fue detectada por primera vez en colémbolos del género Entomobrya recogidos en campo, lo que demuestra su exposición a la toxina que expresa el maíz MON810. Asimismo, se evaluaron los niveles de Cry1Ab en una fracción de la materia orgánica (MO) de la rizosfera procedente de parcelas de maíz Bt, detectándose la toxina hasta tres meses después de la cosecha con valores que oscilaron entre 0,10 y 0,18 ng Cry1Ab/mg MO., Sin embargo, presentó una reducida toxicidad frente a la especie susceptible O. nubilalis, lo que indica que su concentración en forma activa en el suelo fue baja. A partir de losresultados obtenidos, se propone al herbívoro Z. scutellaris, al parasitoide Anagrus spp., y a las comunidades de detritívoros de ácaros y colémbolos del suelo como taxones idóneos para ser utilizados como indicadores de los posibles efectos del maíz Bt en función de su exposición a la toxina Cry1Ab en condiciones de campo, su presencia habitual en cultivos de maíz, la asequibilidad que presentan para ser muestreados e identificados y su utilidad para detectar diferencias entre el maíz Bt y no-Bt analizando sus abundancias. Además, los parámetros riqueza y diversidad de especies de colémbolos fueron adecuados para detectar diferencias entre ambos tipos de maíz., Durante dos campañas consecutivas se recogieron mazorcas de maíz no-Bt que tenían daño de taladro y de maíz Bt de diferentes campos comerciales situados en tres áreas maiceras de España (Extremadura, Albacete y Valle del Ebro). Se analizó la presencia en as mazorcas de 16 especies de los géneros Fusarium y Aspergillus mediante protocolos específicos de PCR, detectándose muestras contaminadas con F. verticillioides, F. proliferatum, F. subglutinans, F. graminearum, F. sporotrichioides, el agregado A. niger y A. flavus. Las especies de la sección Liseola, F. verticillioides, F. proliferatum y F. subglutinans, fueron las únicas que mostraron una frecuencia mayor en las muestras de maíz no-Bt que en las de Bt. Por primera vez se han aislado hongos micotoxígenos, principalmente de estas tres especies de Fusarium, a partir de larvas de O. nubilalis y S. nonagrioides recogidas en campos comerciales de maíz. Las fumonisinas fueron las micotoxinas que con mayor frecuencia contaminaron las mazorcas, siendo la concentración de las mismas significativamente más alta en las mazorcas de maíz no-Bt que en las de maíz Bt. Esta mayor contaminación se ha relacionado con la presencia de los hongos productores de estas micotoxinas (F. verticillioides y F. proliferatum). La alta presencia de F. subglutinans en las mazorcas no-Bt alerta de la posibilidad de contaminación del maíz con moniliformina y otras micotoxinas emergentes, recomendándose su vigilancia. La frecuencia de aparición del resto de hongos y toxinas estudiadas (DON, T-2, HT-2, ZEA, OTA y aflatoxinas) no pareció estar relacionada con el tipo de maíz, Bt o no-Bt. Como conclusión, nuestros resultados revelan que el cultivo continuado de maíz Bt MON810 no afecta negativamente a los artrópodos de la parte aérea de la planta ni a los microartrópodos del suelo, a pesar de que se ha demostrado la exposición de especies pertenecientes a ambas comunidades a la toxina Cry1Ab en condiciones de campo. La selección de grupos indicadores de efectos de maíz Bt llevada a cabo (el herbívoro Z. scutellaris, el parasitoide Anagrus spp., y las comunidades de detritívoros de ácaros y colémbolos del suelo) puede tener una aplicación en las evaluaciones de riesgo exigidas en Europa para las plantas GM que incorporen nuevos eventos y en los programas de seguimiento de los efectos del maíz Bt (MON810). Por otro lado, el maíz Bt, al no presentar ataque de taladros, ha demostrado ser muy eficaz en la reducción de micotoxinas en comparación con el maíz no-Bt. Éste estuvo fundamentalmente contaminado con fumonisinas, la toxina más frecuente en maizales españoles, debido a la mayor presencia de hongos de la sección Liseola, que incluye a los principales productores de las mismas. La presencia de F. verticillioides y F. proliferatum en plantas dañadas por los taladros O. nubilalis y S. nonagrioides ha demostrado ser buen indicador de una probable contaminación con fumonisinas.
