O molho madeira se destaca pelo paladar e elegância na culinária. Como é bastante servido em eventos de grande porte como casamentos e aniversários, necessita ser preparado com certa antecedência e congelado. Porém, alguns possíveis problemas podem ser observados no descongelamento, como a segregação de fases e modificação dos aspectos originais, levando à perda das características sensoriais do produto. Assim, visando uma melhor condição dos aspectos de qualidade, é necessário o congelamento e descongelamento correto do mesmo após o preparo. O objetivo deste trabalho foi avaliar os diferentes tipos de congelamento e descongelamento do molho madeira, determinando o tipo de congelamento que garantisse que os aspectos do molho descongelado fossem os mesmos do molho recém-preparado, além de determinar o tempo de congelamento do mesmo. A composição físico-química do molho madeira foi determinada a fim de determinar suas propriedades termofísicas como massa específica, calor específico, condutividade e difusividade térmica que obtiveram como resultado 787,92 kg.m^-3, 3,85 kJ.kg^-1.K^-1, 0,53 W.m^-1.K^-1 e 1,3 x 10^-5 m 2.s^-1, respectivamente. Esses valores foram utilizados no cálculo do tempo de congelamento segundo os modelos matemáticos de Nagoaka, Pham, Plank, os quais foram comparados ao tempo de congelamento determinado experimentalmente por meio da curva de congelamento. O tempo de congelamento foi de 4,5 horas, mais próximo ao valor calculado pelo modelo de Plank. O congelamento rápido apresentou influência mais positiva, sendo o mais adequado para o produto. The madeira sauce is distinguished by the palate and elegance in the cuisine. As it is well- attended in large events such as weddings and birthdays, it needs to be prepared in advance and frozen. However, some possible problems can be observed in thawing, such as phases segregation and modification of the original aspects, leading to the loss of the product sensorial characteristics. Thus, aiming a better condition of quality aspects, it is necessary to freeze and thaw it correctly after preparation. The objective of this work was to evaluate the different types of freezing and thawing of the madeira sauce, determining the type of freezing that would ensure that the aspects of the thawed sauce were the same as the freshly prepared sauce, besides determining its freezing time. The physico-chemical composition of the madeira sauce was obtained in order to determine its thermophysical properties as specific mass, specific heat, thermal conductivity and diffusivity that resulted in 787,92 kg.m^-3, 3,85 kJ.kg^-1.K^-1, 0,53 W.m^-1.K^-1 and 1,3 x 10^-5 m 2.s^-1, respectively. These values ​ ​ were used to calculate the freezing time according to the Nagoaka, Pham and Plank mathematical models, which were compared to the freezing time determined experimentally by means of the freezing curve. The freezing time was 4.5 hours, closer to the value calculated by the Plank model. Quick freezing had a more positive influence, being the most suitable for the product.

A delipidação química tem sido utilizada como alternativa para a melhoria da criotolerância em embriões produzidos in vitro (PIV). Este estudo foi realizado objetivando avaliar o efeito da Lcarnitina (LC) sobre o desenvolvimento e a sobrevivência de embriões bovinos PIV vitrificados pelo método Cryotop no primeiro ensaio, e no segundo ensaio o efeito comparado da LC e Forskolin em embriões criopreservados por Cryotop (Experimento 1), ou por congelamento lento modificado (Experimento 2). So that, the product can be prepared in this period, to improve the condition of the quality and organoleptic aspects, it is necessary its freezing after preparation. However, due to the freezing, potential problems can be observed, such as segregation of phases and difference to its original aspects. The purpose of this study was to evaluate the production system as well as the process of freezing and thawing of the madeira sauce, aiming to guarantee aspects of freshly prepared sauce. Therefore, physicochemical analysis in order to determine the properties of the fresh sauce were made, obtaining value as 787,92 kg.m-3, 3,85 kJ.kg-1.K-1, 0,53 W.m-1.K-1 e 1,3 x 10-5 m2.s-1 to specific weight, specific heat, thermal conductivity and thermal diffusivity, respectively, as well as the construction of its quick and slow freezing curves for indication of freezing time, these being 4 hours and 30 minutes and -1,8 ºC, respectively. Thus, it was compared to the values ??obtained by mathematical models of Nagoaka, Pham and Plank. The quick freezing had more positive influence than the slow freezing, so, the appearance of madeira sauce after this type of freezing was closest to the freshly prepared sauce, then this was the most appropriate for the product. Objective To assess the viability of bovine ovarian tissue after cryopreservation through either slow freezing or vitrification, and to compare it to that of control tissue by performing morphological analyses. Methods The study included 20 bovine ovarian cortex fragments that were divided into control, vitrification, and slow freezing groups. Each group consisted of four fragments of the same ovary, two fixed without cultivation, and two fixed with cultivation. Tissues were evaluated based on follicular morphology immediately after heating and after 7 days of culture, and compared with the control group. Results A total of 240 fragments were analyzed, generating a sample of 1,344 follicles without cultivation and 552 with cultivation. When the non-cultivated samples were classified as non-atretic follicles, 572 were found in the control group, 289 in the vitrification group, and 373 in the slow freezing group, showing no significant differences. When classified as atretic, 46 follicles were found in the control group, 23 in the vitrification group, and 41 in the slow freezing group, also showing no statistical difference. In the post-culture sample, an evolution of the follicular stages could be observed. This finding was important to support that the follicles considered non-atretic in the non-cultivated group were actually viable in the morphological evaluation. Conclusion With no differences between the protocols, vitrification was shown to be an advanced and alternative method for patients who will undergo treatments that Resumo Objetivo avaliar a viabilidade do tecido ovariano bovino após a criopreservação, utilizando congelamento lento e vitrificação, e comparando com o tecido controle por meio de análises morfológicas. Métodos o estudo incluiufragmentos de córtex de vinte ovários bovinos divididos em grupos controle, vitrificação e congelamento lento. Cada grupo foi composto por quatro fragmentos do mesmo ovário, sendo dois fragmentos fixados sem cultivo e dois fragmentos fixados pós-cultivo. Os tecidos foram avaliados pela morfologia folicular logo após o aquecimento e após sete dias de cultivo, e comparados com o grupo controle. Resultados um total de 240 fragmentos foi analisado, gerando uma amostra de 1.344 folículos sem cultivo e 552 pós-cultivo. Quando a amostra sem cultivo teve seus folículos agrupados em não atrésicos, obtivemos 572 no grupo controle, 289 no vitrificação, e 373 no congelamento lento, não apresentando diferença estatística. Quando agrupados em atrésicos, o grupo controle apresentou 46 folículos, o vitrificação, 23, e o congelamento lento, 41, não apresentando também diferença estatística. Na amostra pós-cultivo, podemos observar uma evolução dos estágios foliculares: esse achado foi importante para sustentar que os folículos considerados não atrésicos na avaliação morfológica sem cultivo estavam realmente viáveis. Conclusão não havendo diferenças entre os protocolos, a vitrificação se mostra um avanço e um método alternativo para pacientes que irão se submeter a tratamentos que podem levar a uma falência ovariana, uma vez que a metodologia é mais barata, mais rápida e mais bem adaptável a uma rotina de um laboratório.

Summary Background: an optimal formulation for vitrifying in vitro-produced (IVP) bovine embryos is currently unavailable. Objective: to estimate whether differences in composition of vitrification solutions may affect the viability of IVP embryos as compared to that of embryos cryopreserved by a conventional slow-freezing method. Methods: bovine IVP embryos were cryopreserved by two methods: 1) a slow controlled-rate (1.5 M ethylene glycol (EG)), or 2) vitrification by using two different vitrification and thawing/warming solutions: (1) Protocol V1: commercial vitrification Kit, and (2) Protocol V2: defined vitrification (20% EG; 20% dimethyl sulfoxide (DMSO); 20% fetal bovine serum (FBS)) and warming (20% FBS; 0.2 M sucrose) solutions. Embryo viability was recorded at 24, 48, and 72 h after thawing/warming by evaluating the number of embryos that re-expanded and developed to the hatching blastocyst stage.
