Changes in dietary composition affect the rumen microbiota in ruminants. However, information on the effects of dietary concentrate-to-forage ratio changes on yak rumen bacteria and metabolites is limited. This study characterized the effect of three different dietary concentrate-to-forage ratios (50:50, C50 group; 65:35, C65 group; 80:20, C80 group) on yak rumen fluid microbiota and metabolites using 16S rRNA gene sequencing and liquid chromatography-mass spectrometry (LC-MS) analyses. Rumen fermentation parameters and the abundance of rumen bacteria were affected by changes in the dietary concentrate-to-forage ratio, and there was a strong correlation between them. At the genus level, higher relative abundances of norank_f__F082, NK4A214_group, Lachnospiraceae_NK3A20_group, Acetitomaculum, and norank_f__norank_o__Clostridia_UCG-014 were observed with a high dietary concentrate-to-forage ratio (P < 0.05). Combined metabolomic and enrichment analyses showed that changes in the dietary concentrate-to-forage ratio significantly affected rumen metabolites related to amino acid metabolism, protein digestion and absorption, carbohydrate metabolism, lipid metabolism, and purine metabolism. Compared with the C50 group, 3-methylindole, pantothenic acid, D-pantothenic acid, and 20-hydroxy-leukotriene E4 were downregulated in the C65 group, while spermine and ribose 1-phosphate were upregulated. Compared to the C50 group, Xanthurenic acid, tyramine, ascorbic acid, D-glucuronic acid, 6-keto-prostaglandin F1a, lipoxin B4, and deoxyadenosine monophosphate were upregulated in the C80 group, while 3-methylindole and 20-hydroxy-leukotriene E4 were downregulated. All metabolites (Xanthurenic acid, L-Valine, N-Acetyl-L-glutamate 5-semialdehyde, N-Acetyl-L-glutamic acid, Tyramine, 6-Keto-prostaglandin F1a, Lipoxin B4, Xanthosine, Thymine, Deoxyinosine, and Uric acid) were upregulated in the C80 group compared with the C65 group. Correlation analysis of microorganisms and metabolites provided new insights into the function of rumen bacteria, as well as a theoretical basis for formulating more scientifically appropriate feeding strategies for yak.

Summary: This study evaluated the effects of dietary concentrate levels and 2‐methylbutyrate (2MB) supplementation on performance, ruminal fermentation, bacteria abundance, microbial enzyme activity and urinary excretion of purine derivatives (PD) in steers. Eight ruminally cannulated Simmental steers (12 months of age; 389 ± 3.7 kg of body weight) were used in a replicated 4 × 4 Latin square design with a 2 × 2 factorial arrangement. Moderate‐concentrate (400 g/kg diet [MC]) or high‐concentrate (600 g/kg diet [HC]) diets were fed with or without 2MB (0 g/day [2MB−] or 15.0 g/day [2MB+]). Dry matter intake and average daily gain increased, but feed conversion ratio decreased with the HC diet or 2MB supplementation. Ruminal pH decreased, but total volatile fatty acid increased with the HC diet or 2MB supplementation. Molar proportion of acetate and acetate‐to‐propionate ratio decreased with the HC diet, but increased with 2MB supplementation. Propionate molar proportion and ruminal NH3‐N content increased with the HC diet, but decreased with 2MB supplementation. Neutral detergent fibre degradability decreased with the HC diet, but increased with 2MB supplementation. Crude protein degradability increased with the HC diet or 2MB supplementation. Abundance of Ruminococcus albus, Ruminococcus flavefaciens, Fibrobacter succinogenes and Bufyrivibrio fibrisolvens as well as activities of carboxymethyl cellulase, cellobiase, xylanase and pectinase decreased with the HC diet, but increased with 2MB supplementation. However, abundance of Prevotella ruminicola and Ruminobacter amylophilus as well as activities of α‐amylase and protease increased with the HC diet or 2MB supplementation. Total PD excretion also increased with the HC diet or 2MB supplementation. The results suggested that growth performance, ruminal fermentation, CP degradability and total PD excretion increased with increasing dietary concentrate level from 40% to 60% or 2MB supplementation. The observed diet × 2MB interaction indicated that supplementation of 2MB was more efficacious for improving growth performance, ruminal fermentation and total PD excretion with promoted ruminal bacteria abundance and enzyme activity in the MC diet than in the HC diet. The ruminal microbiota plays major roles in feed digestion. The composition and fermentation of the bacterial communities in 3 important ruminant species have been studied previously. Here, we extended this research to the effect of concentrate-to-forage ratios on ruminal bacterial communities in Tarim red deer ( Cervus elaphus yarkandensis). Different concentrate-to-forage ratios (2:8, 3:7, 4:6, and 5:5) were fed to Tarim red deer for 20 days. Ruminal bacterial communities were elucidated by 16S ribosomal RNA gene sequencing on an Illumina HiSeq 2500 platform. The microbial composition and biodiversity at the different concentrate-to-forage ratio levels were analyzed using clustering of operational taxonomic units based on 97% sequence identity, taxonomic classification at the phylum and genus levels, α diversity, and β diversity. Rumen microorganisms of deer fed a diet with a concentrate-to-forage ratio of 2:8 had the highest species diversity, followed by ratios of 3:7, 4:6, and 5:5. The community structure of the A1 and A2 samples and the A3 and A4 samples was similar. The bacterial composition appeared to be affected by diet, with a lower dietary concentrate level tending to increase the richness and diversity of ruminal bacteria in the rumen of Tarim red deer. This study aimed to evaluate the effect of concentrate-to-forage ratio (C:F) on the performance, blood profile, and milk physicochemical characteristics of mid-lactation cows fed a corn silage-based diet. Twenty four Holstein cows, with BW 575 ± 70 kg, body condition score (BCS) 3.1 ± 0.2, milk yield 18.4 ± 3.0 kg, and days in milk (DIM) 121 ± 21 were randomly allocated into three treatments with C:F ratios of 35:65, 45:55, and 55:45 on a dry matter (DM) basis. Data was submitted to analyses of variance and regression. Increasing C:F from 35 to 55 % linearly enhanced milk production (22 to 23.6 kg day) and serum urea nitrogen (16.8 to 19.6 mg/dL), while it linearly reduced lactose and fat in milk (4.8 to 4.6 %; 3.9 to 3.6 %, respectively). Body weight, BCS, milk acidity, ethanol stability, coagulation time, and milk and blood mineral contents did not differ among treatments. During the last period of measurements, increased C:F reduced urinary pH and milk urea nitrogen. Changes of concentrate-to-forage proportion from 35 to 55 % increased milk yield, altered chemical composition without changing BW, BCS, acidity, stability, and mineral content of milk and blood attributes. This study was conducted to evaluate the effects of dietary forage type and concentrate-to-forage ratio (CTFR) on CH4 emissions from dairy cows in China using the SF6 tracer method Rumen fluid concentrations of NH4-N and VFA, rumen fluid pH, and daily milk yield were measured as well The dietary regimens included: corn stalks (dry corn stalks after corn harvest) as the forage source at CTFR of 40:60 (ration A), corn silage as the forage source at CTFR of 40:60 (ration B), and corn silage as the forage source at CTFR of 60:40 (ration C). Twelve dairy cows (body weight = 525 ±12 kg, mean ± SE) were divided into three groups of four animals each, balanced in age and body weight, and randomly allocated to the dietary treatments. The study was run for 25 days, with the first 15 days used for adjustment and the subsequent 10 days used for response measurement. The dietary treatments ranked ration A > ration B > ration C for CH4 emissions quantity (L d-1 head-1 ), rumen fluid pH, acetic acid concentration, and ratio of acetic acid and propionic acid concentration. For example, CH4 emissions (mean ±SE) for rations A, B, and C were 353±13.4, 283±7.48, and 263 ±9.04 L d-1 head -1, respectively, or 335 ±12.75, 270 ± 7.12, and 250 ± 8.6 L d-1 AU-1 (AU = 500 kg live weight). On the other hand, the treatments ranked ration A < ration B < ration C for concentrations of NH3-N, propionic acid, and butyric acid in the tureen fluid This study indicates that CH4 emissions intensity from dairy cows (g kg-1 milk output) can be significantly reduced by using corn silage as a forage source and by increasing the content of concentrates in the ration. Compared with rations B and C, ration A increased CH4 emissions by 25% and 34% (p < 0.01), respectively.
