BACKGROUND: The parasitic mite, Varroa destructor has posed a threat to the health and survival of European honey bees, Apis mellifera worldwide. There is a prevailing belief that small comb cells could provide a management tool against Varroa mites. However, the hypothesis that smaller cells can impede Varroa reproduction has not been fully tested. Here, we tested this hypothesis under laboratory conditions by using two distinct Varroa in vitro rearing systems: one involved gelatin capsules of different sizes, specifically size 00 (0.95 mL) versus size 1 (0.48 mL), and the second consisted of brood comb cells drawn on 3D printed foundations with varying cell sizes, ranging from 5.0 mm to 7.0 mm at 0.5 mm intervals. RESULTS: The results showed that mother mites in size 00 cells had significantly lower fecundity and fertility compared to those in size 1 cells. Interestingly, the reproductive suppression in larger cells could be reversed by adding an extra worker larva. Similarly, gonopore size of mother mites was smaller in size 00 cells, but restored with another host larva. Furthermore, both the fecundity and fertility of mother mites decreased linearly with the size of brood comb cells. CONCLUSIONS: Our results suggest that the reproduction of V. destructor is hindered by larger cells, possibly because larger brood cells disperse or weaken host volatile chemical cues that are crucial for Varroa reproduction. Honey bee population inhabiting among different geographic regions often exhibit distinctive traits and phenotypes as a means of enhancing their adaptation to local environmental conditions. Morphological difference of worker was used to reflect the honey bee population locality adaptation and as one of the important markers of species evolution. Here, four comb cell and seven morphology characters of honey bee Apis cerana from two adjacent populations of Yunnan Province were measured to explore the effect of local adaptation on worker comb cell and morphology. Results showed that the characters of worker comb cell and morphology was significant difference between samples from these two local populations of A. cerana. It was separated into two groups which corresponding into the general A. cerana geographic population of Yungui Plateau and South Yunnan base on worker comb cell and morphology character. The worker comb cell size and morphology of A. cerana from Qujing were found to be significantly larger than those from Cangyuan. The indices that showed the most significantly different between populations were cell depth and right forewing length. Moreover, a strong positive correlation was observed between the cell diagonal diameter and the right forewing length, with a correlation coefficient 0.575. In conclusion, honey bee A. cerana population appear high variation in comb cell and morphology to adapt the locality environment. The forewing length of worker was the most significant variation indices between geographic populations, and could be as an indicator of worker cell and morphology among different A. cerana geographic populations. Temperatures of worker- and drone-brood rearing in various hive locations were compared in both colonies kept on small-cell combs (4.90 mm) (SMC) and standard-cell combs (5.50 mm) (STC) in two seasons. Temperatures close to the worker-brood comb placed near the rightmost storage-comb were lower than those near the worker brood in the nest centre but equal to those near the outskirt drone-brood comb (34.37-35.24°C) regardless of the month and the comb-cell size. Temperatures of the brood rearing in the SMC did not differ from those in the STC, independently on the location (center-periphery) and the brood type (drone-worker). Occasionally, they were even higher in the STC near the peripheral drone-brood comb and in the nest centre. We concluded that the drones which are involved in colony reproduction could affect its thermoregulation. The peripheral drone brood can be heated just as well as the worker brood, if the colony is strong enough and has the proper drone-worker ratio. Therefore, it is doubtful whether a higher temperature near the worker brood in the SMC limit the development of the V. destructor population. A lower temperature may not be a factor in encouraging V. destructor females to prefer trap-drone-combs for reproduction in the SMC. Strong field colonies may be especially prone to such behaviour. Therefore, temperature cannot be considered a mechanism of effective Varroa control in SMC.
