High-throughput antibody sequencing allows in-depth insights into human antibody repertoires. To investigate the characteristics of antibody repertoires in patients with chronic HBV infection, we performed Illumina sequencing and IMGT/HighV-QUEST analysis of B lymphocytes from healthy adults and the HBV carriers with high or low level of viral replication. The comparative study revealed high levels of similarity between the IgM and IgG repertoires of the HBV carriers and the healthy adults, including the somatic mutations in V regions, the average CDR3 length, and the occurrence of junctional modifications. Nevertheless, the diversity of the unique clones decreased and some clusters of unique clones expanded in the IgM repertoire of chronic HBV carriers (CHB) compared with healthy adults (HH) and inactive HBV carriers (IHB). Such difference in clone diversity and expansion was not observed in the IgG repertoires of the three populations. More shared antibody clones were found between the IgM repertoires of IHB and HH than that found between CHB and HH (7079 clones vs. 2304 clones). Besides, the biased used IGHD genes were IGHD2-2 and IGHD3-3 in CHB library but were IGHD3-10 and IGHD3-22 in IHB and HH library. In contrast, for IgG repertories, the preferred used VDJ genes were similar in all the three populations. These results indicated that low level of serum HBV might not induce significant changes in BCR repertoires, and high level of HBV replication could have more impacts on IgM repertories than IgG repertoires. Taken together, our findings provide a better understanding of the antibody repertoires of HBV chronically infected individuals.

One of the threats in the diversity loss of the primary gene pool of Vanilla planifolia is the lack of information on existing level of polymorphism in cultivated germplasm, and the different expressions of this polymorphism. For this reason, it is proposed to study the chemical polymorphism of the four phytochemicals that define the vanilla aroma quality in fruits (vanillin, vanillic acid, p-hydroxybenzaldehyde, p-hydroxybenzoic acid) by HPLC analysis (High Performance Liquid Chromatography) of 25 collections of unknown genotype, grown in the region Totonacapan Puebla-Veracruz, Mexico. The results identified a selection process, domestication in fruit aroma of vanilla, during which increased the participation of vanillin and reduced the presence of three minor compounds (vanillic acid, p-hydroxybenzaldehyde and p-hydroxybenzoic acid) in the global aroma. We distinguished a total of six chemotypes of V. planifolia in the Totonacapan region, some chemotypes with wild aromatic characteristics (low participation of vanillin) related to the material less cultivated in the region and domesticated chemotypes with high participation of vanillin, for the most cultivated material. The results show that the diversification of the chemotypes of V. planifolia is not related to environmental variation. The data indicate that in the possible center of origin of vanilla, there is phytochemical polymorphism, which indirectly suggests the existence of genetic polymorphism, essential for the design of a breeding program for optimizing the use and conservation of diversity of the primary gene pool of Vanilla planifolia. Background and Aims Grevillea rhizomatosa is a spreading shrub which exhibits multiple breeding strategies within a narrow area in the fire-prone heathlands of eastern Australia. Reproductive strategies include self-compatibility, self-incompatibility and clonality (with and without sterility). The close proximity of contrasting breeding systems provides an opportunity to explore the evolution of sterility and to compare and contrast the origins of genotypic diversity (recombinant or somatic) against degrees of sexual expression. Methods ISSR markers for 120 band positions (putative loci) were used to compare genetic diversity among five populations at a macro-scale of 5 m between samples (n = 244 shrubs), and at a micro-scale of nearest neighbours for all plants in five 25-m2 quadrats with contrasting fertilities (n = 162 shrubs). Nearest-neighbour sampling included several clusters of connected ramets. Matrix incompatibility (MIC) analyses were used to evaluate the relative contribution of recombination and somatic mutation to genotype diversity. Key Results High levels of genotypic diversity were found in all populations regardless of fertilities (fertile populations, G/N ≥ 0·94; sterile populations, G/N ≥ 0·97) and most sterile populations had a unique genetic profile. Somatic mutations were detected along connected ramets in ten out of 42 ramet clusters. MIC analyses showed that somatic mutations have contributed to diversity in all populations and particularly so in sterile populations. Conclusions Somatic mutations contribute significantly to gene diversity in sterile populations of Grevillea rhizomatosa, the accumulation of which is the likely cause of male and female sterility. High levels of genetic diversity therefore may not always be synonymous with sexual fitness and genetic health. We hypothesize that frequent fires drive selection for clonal reproduction, at the cost of flowering such that sexual functions are not maintained through selection, and the build-up of somatic mutations in meristems results in high genotype diversity at the cost of pollen and ovule fertilities.
