Fargesin is an important bioactive furofuran lignan isolated from different plant species. Despite presenting potent biological activities, its stereochemical characterization has relied mostly on empirical correlations of optical rotation, an approach considered risky that commonly leads to misassignments and error propagation. Additionally, the enantiomeric purity of fargesin isolates used for biological assays has not been previously investigated. Herein, we report the enantioresolution of a scalemic mixture of fargesin isolated from Aristolochia warmingii along with the first unambiguous determination of the absolute configuration of each enantiomer by means of optical rotatory dispersion, as well as electronic and vibrational circular dichroism aided by quantum‐chemical calculations. Natural ectoine, (+)-(4S)-2-methyl-1,4,5,6-tetrahydropyrimidine-4-carboxylic acid, is an extremely important small biomolecule belonging to the class of osmolytic/osmoprotective compounds. It stabilizes biomacromolecules such as DNA and proteins and protects them from denaturation by heat, dehydration, and UV radiation. The rapidly growing interest in this compound resulted in currently exclusive biotechnological production, while a chemical process along with enantioseparation as an alternative has not yet been established. An improved chemical synthesis of racemic ectoine starting from γ-butyrolactone in very good yield is described. Regioselective monoacetylation is achieved by the complexation of a copper(II)-ion with two molecules of 2,4-diamonobutyric acid in the key synthetic step. The racemic ectoine was synthesized with the aim of being successfully enantioseparated for the first time by high-performance liquid chromatography (HPLC) using a teicoplanin-based Chiral-T column in different solvent systems. The presence of (+)-ectoine was determined and quantified using an HPLC protocol on the Synergy Polar-RP column in fermentation broths inoculated with different strains of Streptomyces sp. bacteria isolated from the Adriatic Sea and grown on different NaCl concentrations. The adsorption dynamics of the LL- and DD-enantiomers of Leu-Leu and Gly-Gly on a Chirobiotic V column packed with a CSP bearing glycopeptide antibiotic vancomycin was studied and compared with the adsorption dynamics of the same dipeptides on a Chirobiotic R column packed with a CSP with bonded antibiotic ristocetin A. The column efficiencies were essentially different. The heights equivalent to a theoretical plate (HETP) on Chirobiotic R were 2-4 times larger than on Chirobiotic V, and the shape of van Deemter plots, strongly convex-upwards on the former column, demonstrated minor (Leu-Leu) or lacking (Gly-Gly) convexity on the latter column. This difference is attributed to differences in the binding kinetics of dipeptides to the antibiotics, fast for vancomycin and slow for ristocetin A. Since the kinetic C term of the van Deemter equation cannot explain such a large discrepancy in HETP for the same analyte on two columns, and since the A and B terms for the Chirobiotic R column were essentially larger than for the Chirobiotic V column, it is suggested that binding kinetics influences eddy and longitudinal diffusion processes associated with those terms. It is also hypothesized that unusually high values of van Deemter B coefficients occasionally found in chiral chromatography are not experimental artifacts but the manifestation of coupling the effects of slow adsorption kinetics and longitudinal diffusion.

The existence of slow adsorption-desorption kinetics in chiral liquid chromatography is common knowledge. This may significantly contribute to worsening the efficiency and kinetic performance of a chromatographic run, especially when high flow rates are employed. Many attempts and protocols have been proposed to access this term, the so-called c ads , but they are based on different (theoretical) assumptions. As a consequence, no official method is available for the estimation of the adsorption-desorption kinetics term. In this work, a novel approach to access c ads is presented. This procedure combines experimental results obtained with kinetic and thermodynamic measurements. The investigations have been performed on two zwitterionic teicoplanin chiral stationary phases (CSPs) based on 1.9 μ m fully porous and 2.0 μ m superficially porous particles (FPPs and SPPs), using Z-D,L-Methionine as probe molecule. Kinetic studies have been performed through the combination of both stop-flow and dynamic measurements, while adsorption isotherms have been calculated through Inverse Method. This study has confirmed that, on both particle formats, analyte diffusion on the surface of the particle is negligible, meaning that adsorption is localized, and it has been demonstrated that adsorption-desorption kinetics is strongly dependent on particle geometry and, in particular, on the loading of chiral selector. These findings are fundamental not only to unravel novel aspects of the complex enantiorecognition mechanism but also to optimize the employment of CSPs for ultra-fast and preparative applications. Synthetic therapeutic peptides are a complex and popular class of pharmaceuticals. In recent years, peptides with proven therapeutic activity have gained significant interest in the market. The determination of synthetic peptide enantiomeric purity plays a critical role in the evaluation of the quality of the medicine. Since racemization is one of the most common side reactions occurring in AAs or peptides, enantiomeric impurities such as D‐isomers can form during the peptide synthesis or can be introduced from the starting materials (e.g., AAs). The therapeutic effect of a synthetic or semi‐synthetic bioactive peptide molecule depends on its AA enantiomeric purity and secondary/tertiary structure. Therefore, the enantiomeric purity determination for synthetic peptides is supportive for interpreting unwanted therapeutic effects and determining the quality of synthetic peptide therapeutics. However, enantiomeric purity analysis encounters formidable analytical challenges during chromatographic separation, as D/L isomers have identical physical–chemical properties except stereochemical configuration. To ensure peptides AA stereochemical configuration whether in the free or bound state, sensitive and reproducible quantitative analytical method is mandatory. In this regard, numerous analytical techniques were emerged for the quantification of D‐isomeric impurities in synthetic peptides, but still, very few reports are available in the literature. Thus, the purpose of this paper is to provide an overview of the importance, regulatory requirements, and various analytical methods used for peptide enantiomeric purity determination. A series of nine racemic trans-β-lactam ureas were analyzed for enantiomer separation by high-performance liquid chromatography (HPLC) and supercritical fluid chromatography (SFC). The separations were performed on three immobilized polysaccharide-based chiral analytical columns (CHIRAL ART Amylose-SA, CHIRAL ART Cellulose-SB and CHIRAL ART Cellulose-SC). In HPLC mode, a normal-phase consisting of n-hexane/2-PrOH (90/10, v/v), a polar organic mobile phase consisting of 100% MeOH or 100% EtOH, and a non-standard mobile phase consisting of 100% dimethyl carbonate (DMC) were investigated. In SFC mode, the mobile phases CO2/alcohol (80/20, v/v) and CO2/DMC/alcohol (MeOH or EtOH; 70/24/6, v/v/v or 60/32/8, v/v/v) were investigated. The best achieved enantioseparation of trans-β-lactam ureas was obtained with an Amylose-SA column. We have shown that the green solvent dimethyl carbonate (DMC) can be efficiently used as a mobile phase in HPLC mode as well as in SFC mode along with the addition of polar organic modifiers (MeOH or EtOH). The importance of developing chiral chromatographic methods to monitor and ensure the purity of enantiomers is underscored by the distinct biological properties exhibited by the different forms of ketoprofen. We employed a thin-layer chromatographic approach to separate ketoprofen from commercially available tablet formulations. Our method utilized frovatriptan as the chiral selector, which was incorporated into the stationary phase through a silica gel slurry. The mobile phase consisted of a combination of acetonitrile, methanol, and triethylamine in a ratio of 6:2:2, with a pH of 9. Frovatriptan also served as the chiral selector in the stationary phase, maintaining a pH of 5. The detection and quantitation limits for the ketoprofen enantiomers were determined to be 8.4 μg spot-1 and 25.2 μg spot-1, respectively. Notably, this study marks the first-ever application of frovatriptan as a chiral selector in thinlayer chromatography to resolve ketoprofen enantiomers. [ABSTRACT FROM AUTHOR]

High molecular weight polycyclic aromatic hydrocarbons (HMW-PAHs) are an example of a family of compounds that exist with isomeric mixtures. While multi-instrument techniques may isolate many isomeric PAHs at qualitative and quantitative levels, complete separation of this class of environmental pollutant has not yet been attained, leaving ample room for method improvement. In this research, the separation of 21 commercially available isomers of MM 302 Da was studied using polysaccharide-based chiral immobilized materials as stationary phase. Isocratic, normal phase runs were performed using mixed concentrations and combinations of 302 Da PAH isomers. Retention times and calibration data were determined using a chiral chromatographic setup. Selectivity values are given for all the evaluated HMW-PAH isomers, retention and intensity factors were also calculated based on the directions outlined in Environmental Protection Agency Method 550.1, along with chromatographic peak resolution values. With the use of chiral separation, a total of 16 HMW-PAH peaks are visible on the chromatogram of the 21 MM 302 Da PAH synthetic mixture. This group of molecules does not possess chiral centers in the classical sense; however, larger molecules such as these may possess pseudochiral characteristics, providing a little-researched opportunity for separation strategy of formerly considered achiral molecules to be separated using chiral stationary phases. The resolution of this method makes it suitable for the identification and quantitation of MM 302 Da PAH isomers fractionated from a larger complex sample. We established a validated high‐performance liquid chromatography method utilizing Frovatriptan as a chiral derivatizing agent to separate the enantiomers of ketoprofen from commercially available tablet formulations. The chiral derivatizing agent facilitated the synthesis of diastereomeric derivatives of ketoprofen, which were separated on a reversed‐phase C18 column using a mobile phase composed of acetonitrile and 0.1% trifluoroacetic acid under gradient elution from 30% to 70% of acetonitrile. The method demonstrated a limit of detection and quantitation of 2.52 and 8.31 ng/ml for the derivatives, respectively. Validation of the method was performed following the guidelines set by the International Council for Harmonisation to ensure accuracy, precision, and reliability, making it suitable for pharmaceutical industry applications. Notably, this study introduces the novel application of Frovatriptan as a chiral derivatizing agent in high‐performance liquid chromatography for the separation of ketoprofen enantiomers. The separation on the enantiomers of thirteen newly synthesised liquid crystalline racemic mixtures obtained on the basis of (R,S)-2-hexanol was studied in HPLC on a chiral column prepared by covalent immobilisation of the chiral selector, namely amylose tris(3-chloro-5-methylphenylcarbamate) on silica. The mobile phases (MPs) consisting of n-heptane/isopropanol (IPA) or acetonitrile (ACN)/water (H2O) at different volume ratios were used. All measurements were made at room temperature. The order of the elution was also determined. The chromatographic parameters such as: resolution - (R), selectivity - (α) and number of theoretical plates - (N) were determined. Separation and quantification of amphetamine enantiomers are commonly used to distinguish between consumption of prescription amphetamine (mostly S‐amphetamine) and illicit forms of the drug (racemate). In this study, electromembrane extraction with prototype conductive vials was combined with ultra‐high performance supercritical fluid chromatography (UHPSFC‐MS/MS) to quantify R‐ and S‐amphetamine in urine. Amphetamine was extracted from 100 μL urine, diluted with 25 μL internal standard solution and 175 μL 130 mM formic acid, across a supported liquid membrane (SLM) consisting of 9 μL of a 1:1(w/w) mixture of 2‐nitrophenyloctyl ether (NPOE) and bis(2‐ethylhexyl)phosphite (DEHPi) into an acceptor phase containing 300 μL 130 mM formic acid. The extraction was facilitated by the application of 30 V for 15 min. Enantiomeric separation was achieved using UHPSFC‐MS/MS with a chiral stationary phase. The calibration range was 50–10,000 ng/mL for each enantiomer. The between‐assay CV was ≤5%, within‐assay CV ≤ 1.5%, and bias within ±2%. Recoveries were 83%–90% (CV ≤ 6%), and internal standard corrected matrix effects were 99–105 (CV ≤ 2%). The matrix effects ranged from 96% to 98% (CV ≤ 8%) when not corrected by the internal standard. The EME method was compared with a chiral routine method that employed liquid–liquid extraction (LLE) for sample preparation. Assay results were in agreement with the routine method, and the mean deviation between methods was 3%, ranging from −21% to 31%. Finally, sample preparation greenness was assessed using the AGREEprep tool, which resulted in a greenness score of 0.54 for conductive vial EME, opposed to 0.47 for semi‐automated 96‐well LLE. The interaction between human serum albumin (HSA) and apremilast (APR), a novel antipsoriatic drug, was characterized by multimodal analytical techniques including high-performance liquid chromatography (HPLC), fluorescence spectroscopy and molecular docking for the first time. Using an HSA chiral stationary phase, the APR enantiomers were well separated, indicating enantioselective binding between the protein and the analytes. The influence of chromatographic parameters—type and concentration of the organic modifier, buffer type, pH, ionic strength of the mobile phase, flow rate and column temperature—on the chromatographic responses (retention factor and selectivity) was analyzed in detail. The results revealed that the eutomer S-APR bound to the protein to a greater extent than the antipode. The classical van 't Hoff method was applied for thermodynamic analysis, which indicated that the enantioseparation was enthalpy-controlled. The stability constants of the protein–enantiomer complexes, determined by fluorescence spectroscopy, were in accordance with the elution order observed in HPLC (KR-APR-HSA = 6.45 × 103 M−1, KS-APR-HSA = 1.04 × 104 M−1), showing that, indeed, the later-eluting S-APR displayed a stronger binding with HSA. Molecular docking was applied to study and analyze the interactions between HSA and the APR enantiomers at the atomic level. It was revealed that the most favored APR binding occurred at the border between domains I and II of HSA, and secondary interactions were responsible for the different binding strengths of the enantiomers. The mechanisms of retention and separation of the enantiomers of chiral oxazolopyrroloquinolones on a stationary phase with graftеd macrocyclic antibiotic ristocetin A under conditions of high-performance liquid chromatography with aqueous methanol mobile phases have been studied. The retention factor was found to decrease monotonically when the CH3OH concentration in the mobile phase increased to 90 vol %; then it slightly increased as the composition of the mobile phase approached pure methanol. The thermodynamics of adsorption of oxazolopyrroloquinolones in different regions of this dependence was studied. A mathematical model has been proposed; it describes well the experimental data and allows for analyte solvation in the mobile phase and competitive adsorption (with mobile phase components) on a chiral selector. Phosphatidylglycerol (1,2-diacyl- sn -glycero-3-phospho-glycerol) (PG) is one of the most abundant lipids in biological membranes. However, the chirality of the carbon atom in glycerol phosphate differs among the three kingdoms: bacteria, archaea, and eukaryotes. It is commonly assumed that archaea, as well as bacteria and eukaryotes, produce only one isomer of PG. Archaeal membranes consist of phospholipids with glycerol-1-phosphate in the S configuration, while the phospholipids of the other two kingdoms contain glycerol-3-phosphate with (R) stereochemistry. Another chiral atom is found in glycerol with non-esterified hydroxy groups. Considering the high temperatures that accompanied the origin of life on Earth, it becomes obvious that it is necessary to clarify the importance of membrane lipids in early evolutionary times. To reconstruct the effect of high temperatures on membrane lipids, it is ideal to use microorganisms originating from a thermophilic environment analogous to the early Earth, such as the thermal groundwater of the famous spa town of Karlovy Vary. Here, we prepared all four isomers of PG, i.e., (R,S, R,R, S,R) , and (S,S) , by organic synthesis and analyzed the representation of individual molecular species in seven bacteria isolated from the Karlovy Vary thermal springs using chiral chromatography - mass spectrometry. Our results provide evidence that five of these strains produce all four isomers of PG and that this production is highly dependent on the cultivation temperature. Subsequent analysis by chiral chromatography revealed that the ratio of isomers, enantiomers, and diastereoisomers depends on the cultivation temperature of individual strains. A new synthesis strategy for the preparation of cellulose derivative-based chiral selectors and the subsequent mild immobilization onto pre-functionalized silica gel are introduced, utilizing Cu(I)-catalyzed alkyne-azide Huisgen cycloaddition ("click") chemistry. A cellulose 3,5-dimethylphenyl carbamate derivative carrying propynyl carbamate groups was prepared by a combination of carbonate aminolysis and isocyanate chemistry. For immobilization, 3-azidopropyl-functionalized silica gel as an inert carrier was used, synthesized via a 3-chloropropyl intermediate. The chiral selector, as well as the inorganic/organic hybrid materials (silica gel/chiral selector), were comprehensively characterized by ATR-FTIR, solid-state 13C and 29Si NMR, liquid-state NMR, GPC, TGA, and elemental analysis. The enantioseparation performance of the immobilized-type chiral stationary phase was evaluated by HPLC with a set of representative chiral test analytes and different eluents and compared to a respective coated-type (=non-covalently bound) chiral stationary phase carrying the same selector quality and quantity on the same silica gel matrix. The immobilization did not adversely affect the chiral separation performance; on the contrary, in some chromatographic separations the immobilized-type chiral stationary phase surprisingly even surpassed the coated reference material. The β-blocker (S)-bisoprolol hemifumarate has been synthesised in 96% enantiomeric excess with 19% total yield in a six-step synthesis. A transesterification reaction of the racemic chlorohydrin 1-chloro-3-(4-((2-isopropoxyethoxy)methyl)phenoxy)propan-2-ol catalysed by lipase B from Candida antarctica resulted in the R-chlorohydrin in high enantiomeric purity. Reaction of this building block with isopropylamine in methanol gave (S)-bisoprolol, and further reaction with fumaric acid gave (S)-bisoprolol fumarate in 96% ee. Chiral carbon nanoparticles (CNPs) represent a rapidly evolving area of research for optical and biomedical technologies. Similar to small molecules, applications of CNPs as well as fundamental relationships between their optical activity and structural asymmetry would greatly benefit from their enantioselective separations by chromatography. However, this technique remains in its infancy for chiral carbon and other nanoparticles. The possibility of effective separations using high performance liquid chromatography (HPLC) with chiral stationary phases remains an open question whose answer can also shed light on the components of multiscale chirality of the nanoparticles. Herein, we report a detailed methodology of HPLC for successful separation of chiral CNPs and establish a path for its future optimization. A mobile phase of water/acetonitrile was able to achieve chiral separation of CNPs derived from L‐ and D‐cysteine denoted as L‐CNPs and D‐CNPs. Molecular dynamics simulations show that the teicoplanin‐based stationary phase has a higher affinity for L‐CNPs than for D‐CNPs, in agreement with experiments. The experimental and computational findings jointly indicate that chiral centers of chiral CNPs are present at their surface, which is essential for the multiple applications of these chiral nanostructures and equally essential for interactions with biomolecules and circularly polarized photons. The β-blocker (S)-betaxolol has been synthesized in 99% enantiomeric excess (ee) from the commercially available precursor 4-(2-hydroxyethyl)phenol. The racemic chlorohydrin 1-chloro-3-(4-(2-(cyclopropylmethoxy)ethyl)phenoxy)propan-2-ol was esterified with vinyl acetate catalyzed by lipase B from Candida antarctica, which gave the R-chlorhydrin (R)-1-chloro-3-(4-(2-(cyclopropylmethoxy)ethyl)phenoxy)propan-2-ol in 99% ee with 38% yield. The enantiomeric excess of the R-chlorohydrin was retained in an amination reaction with isopropylamine in methanol to yield (S)-betaxolol in 99% ee and with 9% overall yield. When separating enantiomers on chiral crystals of achiral substances, it was found that a relatively high enantioselectivity is accompanied by a low efficiency of columns. Therefore, work was carried out to evaluate the influence of the coefficients of the van Deemter equation on the broadening of the peaks of camphor enantiomers on a chiral stationary phase based on enantimorphic α-NiSO4⋅6H2O crystals (retgersite). From an experimental van Deemter curve, the contributions of vortex and longitudinal diffusion, as well as the resistance to mass transfer, were calculated in the value of an introduced magnitude equivalent to the theoretical plate (h). It is shown that the right branch of the curve for enantiomers of camphor differs. It is established that this difference is caused by different values of resistance to mass transfer in the stationary phase Cs caused by the difference in diffusion coefficients in the pore space. The β-blocker (S)-esmolol, has been synthesized in 97% enantiomeric excess and 26% total yield in a four-step synthesis, with a transesterification step of the racemic chlorohydrin methyl 3-(4-(3-chloro-2-hydroxypropoxy)phenyl)propanoate, catalysed by lipase B from Candida antarctica from Syncozymes, Shanghai, China. The β-blocker (S)-penbutolol, has been synthesized in 99% enantiomeric excess and in 22% total yield. The transesterification step of the racemic chlorohydrin 1-chloro-3-(2-cyclopentylphenoxy)propan-2-ol was catalyzed by the same lipase as used for the esmolol building block. We have used different bases for the deprotonation step of the starting phenols, and vinyl butanoate as the acyl donor in the transesterification reactions. The reaction times for the kinetic resolution steps catalysed by the lipase varied from 23 to 48 h, and were run at 30–38 °C. Specific rotation values confirmed the absolute configuration of the enantiopure drugs, however, an earlier report of the specific rotation value of (S)-esmolol is not consistent with our measured specific rotation values, and we here claim that our data are correct. Compared to the previously reported syntheses of these two enantiopure drugs, we have replaced toluene or dichloromethane with acetonitrile, and replaced the flammable acetyl chloride with lithium chloride. A new total synthesis of the pharmacologically active β-carboline alkaloid brevicolline is described. The new synthetic approach is based on a commercially available and inexpensive starting material and reagents leading to a practical synthesis of the racemic target molecule, the natural (S)-enantiomer, and its antipode. Initially, the construction of the β-carboline skeleton and functionalization at the C(4) position have been accomplished. The formation of dihydropyrrole structural unit was obtained as the result of an Au-catalyzed hydroamination reaction, which was followed by a reduction that led to the chiral intermediate. The synthetic route described here is developed to ensure the sustainable access of the racemic brevicolline in 11 steps with improved 48% overall yield compared to the previously reported methods. Enantioseparation of the newly synthesized series of novel quinoline-2(1H)-one epoxide structures rac-6a–c and rac-8a–c, named marinoepoxides, is described. Marinoepoxide rac-6a, the key intermediate in the total synthesis of natural products marinoaziridines A and B, as well as their structural analogues, was synthesized by addition of the achiral ylide generated in situ from the sulfonium salt 5 or 7, to the carbon-oxygen double bond of the corresponding quinoline-2(1H)-one-4-carbaldehyde 4a–c in good yield. Separation of enantiomers of (±)-2,3,3-trisubstituted marinoepoxides rac-6a–c and (±)-trans-2,3-disubstituted marinoepoxides rac-8a–c was studied using two immobilized polysaccharide type chiral stationary phases (CSPs); tris-(3,5-dichlorophenylcarbamoyl)cellulose stationary phase (CHIRAL ART Cellulose-SC) and tris-(3,5-dimethylphenylcarbamoyl)amylose stationary phase (CHIRAL ART Amylose-SA). Separating pure enantiomers from racemates has long been a challenging and significant task across various scientific disciplines of biology, medicine, and chemistry. Over the past two decades, metal–organic frameworks (MOFs) have emerged as one of the most sought-after materials, showcasing unparalleled advantages in various applications. Chiral MOFs, in particular, have garnered increasing attention in enantiomeric separation. Unlike conventional materials, MOFs offer greater potential for achieving enantiomeric separation in chromatographic techniques due to their customizable chiral structures and adjustable pore environments. In this review, we aim to outline the remarkable progress and achievements of MOFs as stationary phases in the field of chromatographic enantiomeric separation including gas chromatography, high-performance liquid chromatography, and capillary electrochromatography. The discussions about MOF synthesis, column packing strategy, and MOF-based column performance, will improve our understanding about the design, immobilization, and separation mechanism for the MOF stationary phase. we developed an effective protein precipitation method for determination of levamlodipine in human plasma using LC–MS/MS. Sample extraction was carried out by using liquid–liquid extraction in 96-well plate format. (S)-Amlodipine-d4 was used as internal standard (IS). The chromatographic separation was achieved using Philomen Chiral MX (2) column (3 μm, 2.1 × 100 mm). Mobile phase A was comprised of Acetonitrile (ACN), Mono ethanol amine (MEA) and Iso-Propyl alcohol (IPA) (1000:1:10, v/v/v), Mobile phase B was IPA-ACN (2:1, v/v). The flow rate was 0.4 mL/min. The total run time of each sample was 4.0 min with gradient elution. LC-MS/MS spectra were generated in positive ion mode, and multiple reaction monitoring (MRM) was used to detect the following transitions: m/z 409.20 → 238.15 for levamlodipine and 415.25 → 240.20 for (S)-Amlodipine-d4 (the IS). The method was linear from 50 to 10000 pg/mL（R2＝0.9988489）,and the lower limit of quantification (LLOQ) was 50 pg/mL. This method was applied to a bioequivalence study of levamlodipine. Two minor phospholipids, i.e., mono- and/or dimethylphosphatidylethanolamines, are widespread in many organisms, from bacteria to higher plants and animals. A molecular mixture of methyl-PE and dimethyl-PE was obtained from total lipids by liquid chromatography and further identified by mass spectrometry. Total methyl-PE and dimethyl-PE were cleaved by phospholipase C, and the resulting diacylglycerols, in the form of acetyl derivatives, were separated into alkyl-acyl, alkenyl-acyl, and diacylglycerols. Reversed-phase LC/MS allowed dozens of molecular species to be identified and further analyzed. This was performed on a chiral column, and identification by tandem positive ESI revealed that diacyl derivatives from all four bacteria were mixtures of both R and S enantiomers. The same applied to alkenyl-acyl derivatives of anaerobic bacteria. Analysis thus confirmed that some bacteria biosynthesize phospholipids having both sn-glycerol-3-phosphate and sn-glycerol-1-phosphate as precursors. These findings were further supported by data already published in GenBank. The use of chiral chromatography made it possible to prove that both enantiomers of glycerol phosphate of some molecular species of mono- and dimethylphosphatidylethanolamines are present. The result of the analysis can be interpreted that the cultured bacteria do not have homochiral membranes but, on the contrary, have an asymmetric, i.e., heterochiral membranes. Amphetamine (speed), methamphetamine (crystal meth), and 3,4‐methylenedioxy‐N‐methylamphetamine (MDMA, ecstasy) represent the most frequently abused amphetamine‐type stimulants (ATS). Differences in pharmacological potency and metabolism have been shown for the enantiomers of all three stimulants. Legal consequences in cases of drug possession may also differ according to the German law depending on the enantiomeric composition of the seized drug. Therefore, enantioselective monitoring of seized specimens is crucial for legal and forensic casework. Various kinds of samples of amphetamine (n = 143), MDMA (n = 94), and methamphetamine (n = 528) that were seized in southern Germany in 2019 and 2020 were analyzed for their chiral composition using different chromatographic methods. Whereas all samples of amphetamine and MDMA were racemic mixtures, the chiral composition of the methamphetamine specimens was diverse. Although the vast majority (n = 502) was present as (S)‐methamphetamine, also specimens containing pure (R)‐methamphetamine (n = 7) were found. Furthermore, few samples (n = 8) were of racemic nature or contained non‐racemic mixtures of both enantiomers (n = 10). Because methamphetamine appears in varying enantiomeric compositions, any seizure should be analyzed using an enantioselective method. Amphetamine and MDMA, on the other hand, currently appear to be synthesized exclusively via racemic pathways and are not chirally purified. Nafronyl is a pharmaceutically active compound used as a drug ingredient. It occurs in form of four stereoisomers, with however different biological activities. A procedure for preparation of the most active stereoisomer of nafronyl was developed. The stereoselective synthesis was used to produce a pair of diastereoisomers, which included the target stereoisomer. The mixture obtained was then separated by chiral chromatography in a batch mode. The efficiency of the separation in a continuous mode in a classical simulated moving‐bed system was assessed. To improve the yield of the whole operation, the less active diastereoisomer was subjected to diastereoisomeric conversion, which delivered back the pair of diastereoisomers that could be recycled to the chromatographic separation. Retention and separation factors of enantiomers of several dipeptides were measured on chiral stationary phases with grafted macrocyclic antibiotic ristocetin A from different manufacturers, Chirobiotic R and Nautilus-R, using buffered and unbuffered methanol–water mixtures with 0 to 90% (v/v) of methanol as mobile phases. A remarkable difference in enantioselectivity toward both hydrophilic and hydrophobic dipeptides was observed between these two stationary phases that were explained by different configurations of the binding sites of the selectors resulted from different anchoring reactions applied to graft the antibiotic moiety to the silica support and also by different number of ionizable amino groups of the grafted selectors. A study is performed of mechanisms of the retention and separation of enantiomers of a series of chiral oxazolopyrroloquinolones on a stationary phase with grafted macrocyclic antibiotic ristocetin A under the conditions of high-performance liquid chromatography using water–acetonitrile mobile phases. It is found that the dependence of the retention factor on the concentration of CH3CN in the mobile phase has a U-shape. An explanation of this phenomenon is given. The thermodynamics of the oxazolopyrroloquinolone adsorption in different parts of this dependence is studied. It is assumed that the adsorption complex forms through inclusion of the oxazole moiety of the adsorbate molecule in the chiral cavity of the grafted selector. It has become increasingly popular in the field of chiral separation, and many applications have been introduced during the last years. This review mainly focuses on the current topics, applications, and trends in chiral separation by counter‐current chromatography. It contains the development of modern counter‐current chromatography apparatus, theory of counter‐current chromatography, overview of applications of chiral counter‐current chromatography enantioseparation, its current situation, and challenges. This manuscript presents the development, validation and application of a multi-residue supercritical fluid chromatography coupled with tandem mass spectrometry method for the analysis of 140 chiral and non-chiral chemicals of emerging concern in environmental samples, with 81 compounds being fully quantitative, 14 semi-quantitative and 45 qualitative, validated according to European Medicine Agency (EMA) guidelines (European Medicines Agency 2019). One unified LC-MS method was used to analyse all analytes, which were split into three injection methods to ensure sufficient peak resolution. The unified method provided an average of 113% accuracy and 4.5% precision across the analyte range. Limits of detection were in the range of 35 pg L−1–0.7 μg L−1, in both river water and wastewater, with an average LOD of 33 ng L−1. The method was combined with solid-phase extraction and applied in environmental samples, showing very good accuracy and precision, as well as excellent chromatographic resolution of a range of chiral enantiomers including beta-blockers, benzodiazepines and antidepressants. The method resulted in quantification of 75% of analytes in at least two matrices, and 56% in the trio of environmental matrices of river water, effluent wastewater and influent wastewater, enabling its use in monitoring compounds of environmental concern, from their sources of origin through to their discharge into the environment. Professor William H. Pirkle (1934–2018) made a profound impact on modern chemistry by inventing and popularizing widely used techniques for the analysis and purification of enantiomers, contributions that paved the way for the subsequent advances in the discovery, development, and manufacture of enantiopure pharmaceuticals, agrochemicals, and specialty chemicals. Pirkle's pioneering 1966 demonstration of the use of chiral solvating agents for the nuclear magnetic resonance determination of enantiopurity led to a lifelong interest in understanding the supramolecular interactions responsible for enantiodifferentiation. Ongoing research into the chromatographic resolution of stereoisomers throughout the 1970s led in 1981 to the very first commercialization of a chiral stationary phase for the high‐performance liquid chromatography (HPLC) separation of enantiomers. The availability of this and subsequent "Pirkle columns" had a deep and lasting impact, becoming widely embraced by the chemical sciences research community worldwide and spearheading the wholesale changeover to HPLC as the preferred technique for measuring enantiopurity. Doc Pirkle was a highly creative, independent, and fun‐loving collaborator whose circle of friends extends around the globe. His research group at the University of Illinois, often referred to as The Pirkle Zoo, became a refuge for an interesting assembly of characters who flourished under his mentorship and guidance. A chiral stationary phase is proposed on the basis of 3,4,9,10-perylenetetracarboxylic acid (3,4,9,10-perylentetracarbonacid, PTCA). The PTCA molecule itself is not chiral; however, it can form chiral supramolecular structures. Symmetry breaking during the self-assembly of PTCA supramolecular structures was carried out similarly to the Viedma ripening process. The obtained stationary phases with PTCA multilayers on an inert solid support and C18 silica were used to separate racemates in gas and liquid chromatography, respectively. It was found that the proposed stationary phases exhibit enantioselectivity with respect to pairs of enantiomers bearing a hydroxyl group at an asymmetric carbon atom. Enantiomers of butanol-2, pentanol-2, and 1-methoxy-propanol-2 were separated by gas chromatography. The separation of menthols was achieved in the normal-phase HPLC mode. In the present study, an in silico methodology able to define the binding modes adopted by carnosine enantiomers in the setting of the chiral recognition process is described. The inter‐ and intramolecular forces involved in the enantioseparation process with the Teicoplanin A2‐2 chiral selector and carnosine as model compound are successfully identified. This approach fully rationalizes, at a molecular level, the (S) < (R) enantiomeric elution order obtained under reversed‐phase conditions. Consistent explanations were achieved by managing molecular dynamics results with advanced techniques of data analysis. As a result, the time‐dependent identification of all the interactions simultaneously occurring in the chiral selector‐enantiomeric analyte binding process was obtained. Accordingly, it was found that only (R)‐carnosine is able to engage a stabilizing charge–charge interaction through its ionized imidazole ring with the carboxylate counter‐part on the chiral selector. Instead, (S)‐carnosine establishes intramolecular contacts between its ionized functional groups, that limit its conformational freedom and impair the association with the chiral selector unit. 17β-Hydroxysteroid dehydrogenase type 3 (17β-HSD3) is expressed at high levels in testes and seminal vesicles; it is also present in prostate tissue and involved in gonadal and non-gonadal testosterone biosynthesis. The enzyme is membrane-bound, and a crystal structure is not yet available. Selective aryl benzylamine-based inhibitors were designed and synthesised as potential agents for prostate cancer therapeutics through structure-based design, using a previously built homology model with docking studies. Potent, selective, low nanomolar IC50 17β-HSD3 inhibitors were discovered using N-(2-([2-(4-chlorophenoxy)phenylamino]methyl)phenyl)acetamide (1). The most potent compounds have IC50 values of approximately 75 nM. Compound 29, N-[2-(1-Acetylpiperidin-4-ylamino)benzyl]-N-[2-(4-chlorophenoxy)phenyl]acetamide, has an IC50 of 76 nM, while compound 30, N-(2-(1-[2-(4-chlorophenoxy)-phenylamino]ethyl)phenyl)acetamide, has an IC50 of 74 nM. Racemic C-allyl derivative 26 (IC50 of 520 nM) was easily formed from 1 in good yield and, to determine binding directionality, its enantiomers were separated by chiral chromatography. Absolute configuration was determined using single crystal X-ray crystallography. Only the S-(+)-enantiomer (32) was active with an IC50 of 370 nM. The increased use and applicability of Cannabis and Cannabis-derived products has skyrocketed over the last 5 years. With more and more governing bodies moving toward medical and recreational legalization, the need for robust and reliable analytical testing methods is also growing. While many stationary phases and methods have been developed for this sort of analysis, chiral stationary phases (CSPs) are unique in this area; not only can they serve their traditional chiral separation role, but they can also be used to perform achiral separations. Given that mixtures of cannabinoids routinely contain enantiomers, diastereomers, and structural isomers, this offers an advantage over the strictly achiral-only analyses. This work presents the separation of a 10-cannabinoid mixture on several polysaccharide-based sub-2 µm CSPs with both normal-phase and reversed-phase ultra-high-performance liquid chromatography (UHPLC) conditions. Along with the separation of the mixture, appropriate single-peak identification was performed to determine the elution order and reported where applicable.

In forensic toxicology, amphetamine intoxications represent one of the most common case groups and present difficult questions for toxicologists. Estimating the time of consumption and the current influence of the stimulant is particularly difficult when only total amphetamine concentrations are considered. Stereoselective analysis and the consideration of metabolites can provide valuable information to facilitate interpretation. An enantioselective liquid chromatography−tandem mass spectrometry (LC-MS/MS) method for detection of amphetamine, norephedrine and 4-hydroxyamphetamine was developed. Validation showed satisfactory selectivity, sensitivity, linearity (0.5–250 ng/mL), precision and accuracy for all enantiomers. The method was applied to a collective of 425 forensic serum samples and 30 serum samples from psychiatric inpatients stating their last time of amphetamine consumption. Norephedrine and 4-hydroxyamphetamine were detected more frequently at higher amphetamine concentrations and at lower amphetamine (R)/(S) concentration ratios, possibly indicating recent consumption. Mean (R)/(S) ratio of amphetamine was 1.14, whereas higher ratios (mean 1.36) were found for amphetamine concentrations below 100 ng/mL. The (R)/(S) ratios of psychiatric inpatients significantly correlated with the reported time intervals to last consumption. The use of amphetamine (R)/(S) ratios and the simultaneous detection of metabolites are promising factors that can facilitate estimation of consumption time and current impairment.

This review draws attention to the use of chiral monolithic silica HPLC columns for the enantiomeric separation and determination of chiral compounds. Properties and advantages of monolithic silica HPLC columns are also highlighted in comparison to conventional particle-packed, fused-core, and sub-2-µm HPLC columns. Nano-LC capillary monolithic silica columns as well as polymeric-based and hybrid-based monolithic columns are also demonstrated to show good enantioresolution abilities. Methods for introducing the chiral selector into the monolithic silica column in the form of mobile phase additive, by encapsulation and surface coating, or by covalent functionalization are described. The application of molecular modeling methods to elucidate the selector–selectand interaction is discussed. An application for enantiomeric impurity determination is also considered.

Culinary sage, Salvia officinalis L., is a popular spice plant commonly used throughout the world. In this study, 35 odorants were identified in dried sage via solvent-assisted flavor evaporation (SAFE) and aroma extract dilution analysis (AEDA), including 9 that were identified in sage for the first time. Fifteen odorants were quantitated by stable isotope dilution analysis (SIDA), and their odor activity values (OAVs) were determined. Odorants with high OAVs included (2 E,6Z )-nona-2,6-dienal, 1,8-cineole, and β-myrcene. A formulated aroma simulation model closely matched the aroma profile of an aqueous infusion of dried sage. Enantiomeric proportions of selected odorants were determined by chiral gas chromatography. Furthermore, 6 different sage cultivars were grown in the greenhouse, dried under the same conditions, and analyzed. Sensory analysis determined that all cultivars were dominated by an herbaceous sensory attribute and had varying intensities of eucalyptus, mint, clove, pine, green, earthy, floral, and citrus notes. Cultivars with varying intensities of herbaceous, eucalyptus, pine, and green sensory notes correlated with the OAVs of α-thujone/β-thujone, 1,8-cineole, α-pinene, and (2 E ,6 Z )-nona-2,6-dienal, respectively. This study identified the odorants driving the sensory profiles of different sage cultivars and serves as a foundation for future studies on the aroma chemistry of culinary sage.

The chromatographic behavior and adsorption of enantiomers of glycylaspartic acid on the Nautilus-E chiral stationary phase with grafted eremomycin antibiotic were studied. Acetate buf er mixtures with variable pH prepared in a water—methanol mixed solvent (volume ratio 60 : 40) were used as acetate buf ers. The retention of Gly—Asp was found to be enantioselective and depending on the ratio of various ionic forms of dipeptide in the mobile phase. The ratio is determined by the pH. A model of adsorption equilibrium assuming independent binding of diverse ionic forms with the stationary phase, which is characteristic of energy uniformity, was proposed. This model is well consistent with the experimental data. Features of application of the equilibrium dispersion and transport dispersion dynamic elution models for the determination of the adsorption isotherms by the inverse method are discussed. Evaluating lumefantrine enantioselectivity and properly quantifying its enantiomers is essential for a deeper understanding of the impact of the enantioselectivity in the pharmacodynamic and pharmacokinetic properties of the drug. For this, a chiral chromatographic method was developed and validated for the separation of lumefantrine enantiomers. The method developed employed a Chiralpack AD-H column (150 × 4.6 mm, 5 μm), with amylose tris-3,5-dimethylphenylcarbamate stationary phase, at 25 °C, mobile phase composed of hexane and isopropanol (97:3), flow rate of 1.0 mL/min and detection at 335 nm. The method showed adequate resolution for lumefantrine enantiomers and was validated for linearity (48–72 µg/ml for each enantiomer), precision, accuracy, selectivity and robustness. The proposed method was successfully applied for determination of lumefantrine enantiomers in tablet formulations and may be useful to future researchers for chiral evaluation of lumefantrine.
