This study summarized and evaluated the cost-benefit analysis methods for typical water environment pollution control measures in China. We conducted a comprehensive comparison of these measures, whose advantages and disadvantages were examined, so the application models and characteristics can be delved into. Through integrating methods and analyzing application cases, we refined a combination of measures with high efficiency and low cost in controlling China's water environment. In our analysis of cost-benefit, we emphasized the importance of implementing scientifically designed control measures and employing rigorous methods to addressing water environment pollution. In order to achieve rapid response and efficient treatment of China's environmental problems, we advocated for establishing a cross regional pollution control mechanism. That is, the collaborative platform and scientific suggestions for all participants in this field should be provided, through interdisciplinary integration, development of domestic models, and expansion of databases. Agricultural nutrients nitrogen and phosphorus can subsequently be transported to waterways and are often managed through the adoption of best management practices (BMPs). However, we have a poor understanding of how the use of BMPs varies spatially and how BMP adoption might be related to nutrient yields in surface waters. To address this, we performed a survey of agricultural landowners across New York State and compared this with estimates of annual incremental nitrogen and phosphorus yields of agricultural origin from the Spatially Referenced Regressions On Watershed attributes (SPARROW) model. Using these socio-behavioral data and SPARROW predictions, we perform colocation analysis to identify areas where watersheds with high nutrient yield from agriculture are collocated with non-use of agricultural BMPs. This colocation analysis offers a novel methodology for identifying areas where monitoring of waterways and promotion of best management practices could be targeted to achieve the greatest benefits. Several differentiation protocols have enabled the generation of intermediate mesoderm (IM)-derived cells from human pluripotent stem cells (hPSC). However, the substantial variability between existing protocols for generating IM cells compromises their efficiency, reproducibility, and overall success, potentially hindering the utility of urogenital system organoids. Here, we examined the role of high levels of Nodal signaling and BMP activity, as well as WNT signaling in the specification of IM cells derived from a UCSD167i-99-1 human induced pluripotent stem cells (hiPSC) line. We demonstrate that precise modulation of WNT and BMP signaling significantly enhances IM differentiation efficiency. Treatment of hPSC with 3 μM CHIR99021 induced TBXT+/MIXL1+ mesoderm progenitor (MP) cells after 48 h of differentiation. Further treatment with a combination of 3 μM CHIR99021 and 4ng/mL BMP4 resulted in the generation of OSR1+/GATA3+/PAX2+ IM cells within a subsequent 48 h period. Molecular characterization of differentiated cells was confirmed through immunofluorescence staining and RT-qPCR. Hence, this study establishes a consistent and reproducible protocol for differentiating hiPSC into IM cells that faithfully recapitulates the molecular signatures of IM development. This protocol holds promise for improving the success of protocols designed to generate urogenital system organoids in vitro, with potential applications in regenerative medicine, drug discovery, and disease modeling. Cartilage, a flexible and smooth connective tissue that envelops the surfaces of synovial joints, relies on chondrocytes for extracellular matrix (ECM) production and the maintenance of its structural and functional integrity. Melatonin (MT), renowned for its anti-inflammatory and antioxidant properties, holds the potential to modulate cartilage regeneration and degradation. Therefore, the present study was devoted to elucidating the mechanism of MT on chondrocytes. The in vivo experiment consisted of three groups: Sham (only the skin tissue was incised), Model (using the anterior cruciate ligament transection (ACLT) method), and MT (30 mg/kg), with sample extraction following 12 weeks of administration. Pathological alterations in articular cartilage, synovium, and subchondral bone were evaluated using Safranin O-fast green staining. Immunohistochemistry (ICH) analysis was employed to assess the expression of matrix degradation-related markers. The levels of serum cytokines were quantified via Enzyme-linked immunosorbent assay (ELISA) assays. In in vitro experiments, primary chondrocytes were divided into Control, Model, MT, negative control, and inhibitor groups. Western blotting (WB) and Quantitative RT-PCR (q-PCR) were used to detect Silent information regulator transcript-1 (SIRT1)/Nuclear factor kappa-B (NF-κB)/Nuclear factor erythroid-2-related factor 2 (Nrf2)/Transforming growth factor-beta (TGF-β)/Bone morphogenetic proteins (BMPs)-related indicators. Immunofluorescence (IF) analysis was employed to examine the status of type II collagen (COL2A1), SIRT1, phosphorylated NF-κB p65 (p-p65), and phosphorylated mothers against decapentaplegic homolog 2 (p-Smad2). In vivo results revealed that the MT group exhibited a relatively smooth cartilage surface, modest chondrocyte loss, mild synovial hyperplasia, and increased subchondral bone thickness. ICH results showed that MT downregulated the expression of components related to matrix degradation. ELISA results showed that MT reduced serum inflammatory cytokine levels. In vitro experiments confirmed that MT upregulated the expression of SIRT1/Nrf2/TGF-β/BMPs while inhibiting the NF-κB pathway and matrix degradation-related components. The introduction of the SIRT1 inhibitor Selisistat (EX527) reversed the effects of MT. Together, these findings suggest that MT has the potential to ameliorate inflammation, inhibit the release of matrix-degrading enzymes, and improve the cartilage condition. This study provides a new theoretical basis for understanding the role of MT in decelerating cartilage degradation and promoting chondrocyte repair in in vivo and in vitro cultured chondrocytes. Nonylphenol (NP) is a known endocrine-disrupting chemical (EDC) that has been shown to affect bone development in mammals. However, the detrimental impacts of NP on the skeletal growth and development of aquatic species, especially bony fish, remain poorly understood. Bone morphogenic proteins (BMPs), essential for bone formation and osteoblast differentiation, act through the BMP-Smad signaling pathway. In this study, two BMP genes, BMP2 and BMP4, were cloned and characterized in the red crucian carp (Carassius auratus red var.). The full-length cDNAs of BMP2 and BMP4 were 2029 bp and 2095 bp, respectively, encoding polypeptides of 411 and 433 amino acids, and share a typical TGF-β domain with other BMPs. The tissue expression patterns of both genes were identified, showing ubiquitous expression across all studied tissues. Additionally, the exposure of embryos or adult fish to NP stress resulted in a downregulation of BMP2, BMP4, and other genes associated with the BMP-Smad signaling pathway. Moreover, the combined treatment of adult fish with NP and the specific BMP receptor inhibitor significantly reduced these genes' expression. These findings elucidate the mechanism of NP stress on BMP2 and BMP4, suggesting a role for the BMP-Smad signaling pathway in the response to endocrine-disrupting chemicals in fish. Assessment and modelling of hydro-sedimentological flows of a high-altitude river system is a critical step for developing and managing sustainable water resource projects and best management practices (BMPs) in the downslope regions of the Indian Himalayan Region (IHR). A field study was carried out to measure the hydraulic parameters such as water pressure, water flow rate, and stage of the 6th order glacier-fed river to quantify hydro-sedimentological flows using area-velocity and vacuum filtration method for 3 successive years during 2018–2020. Further, a process-based hydrological model: Soil and Water Assessment Tool (SWAT), is used to simulate the hydro-sedimentological flows. The statistical indices such as coefficient of determination (R2), Nash–Sutcliffe efficiency (NSE), and percentage bias (PBIAS) attain higher values during both calibration and validation periods. The snowmelt and rainfall contributions to the total streamflow range from 17–35 % and 27–34 %, respectively. The measured and modelled hydro-sedimentological flows show high variability with a high coefficient of variation (COV > 1). However, the mean suspended sediment load (SSL) carried by the river was low compared to the other glacier-fed rivers. The physical weathering rate (PWR) dominates the chemical weathering rate (CWR) for the study years. This might be due to higher crushing of the region and weathering of base rock materials. The PWR and CWR of the basin are less than that of the western Himalayan regions. This study also underscores the necessity of basin management plans in the Himalaya, emphasizing erosion identification, snowmelt and glacier melt in streamflow, and customized groundwater recharge strategies through GIS mapping, providing essential insights for sustainable land and water resource management in changing climatic conditions. Identifying vulnerable areas to erosion within the watershed and implementing best management practices (BMPs) are crucial steps in mitigating watershed degradation by minimizing sediment yields. The present study evaluates and identifies the BMPs in the Seybouse basin, northeastern Algeria, using the Soil and Water Assessment Tool (SWAT) model. After successful calibration and validation, the model demonstrated a satisfactory ability to simulate monthly discharge and sediment. Then, the calibrated model was employed to evaluate the efficacy of diverse management practices in sediment control. In the SWAT, three soil and conservation practices, as well as vegetated filter strips (VFSs), grade stabilization structures (GSSs), and terracing were evaluated. The average annual sediment yield in the Seybouse watershed is determined to be 14.43 t/ha year, constituting 71% of the total soil loss. VFS demonstrated a sediment reduction of 37.30%, GSS 20.40%, and terracing 42.30%. Among these strategies, terracing results in the greatest reduction, followed by VFS. The results of this study area can be useful for informed decision-making regarding optimal watershed management strategies. To investigate whether the Anti-Müllerian Hormone (AMH), an ovarian hormone belonging to the Transforming Growth Factor β superfamily, may represent a possible candidate for use as a bone anabolic factor. We performed in vitro studies on Human Osteoblasts (HOb) to evaluate the expression and the functionality of AMHRII, the AMH receptor type-2, and investigate the effects of exogenous AMH exposure on osteogenic gene expression and osteoblast functions. We reported the first evidence for the expression and functionality of AMHRII in HOb cells, thus suggesting that osteoblasts may represent a specific target for exogenous AMH treatment. Furthermore, the exposure to AMH exerted a stimulatory effect on HOb cells leading to the activation of osteogenic genes, including the upregulation of osteoblastic transcription factors such as RUNX and OSX, along with increased deposition of mineralized nodules. Our findings proved interesting clues on the stimulatory effects of AMH on mature osteoblasts expressing its specific receptor, AMHRII. This study may therefore have translation value in opening the perspective that AMH may be an effective candidate to counteract the bone loss in osteoporotic patients by selectively targeting osteoblast with minimal off-target effect. Diffuse pollution control through the implementation of best management practices (BMPs) is essential for integrated water management. This study presents a comprehensive approach that integrates monitoring and deterministic water quality modeling to assess the effectiveness of various BMPs and point pollution control scenarios for improving water quality. The pilot study area (PSA) is the Acısu Creek watershed in the south of Turkiye. The hydrological model MapShed was employed to estimate sediment and nutrient load reductions by BMPs and these estimates were used as inputs to the Water Quality Analysis Simulation Program (WASP8) for water quality simulations. The MapShed model was calibrated in the Lower Aksu Stream basin from 2013 to 2019, achieving satisfactory results. Calibration and verification of the WASP8 model were performed using one year of monthly water quality monitoring results, showing good agreement between measurements and predictions. Diffuse pollution control scenarios were effective in decreasing pollutant concentrations. The BMP scenario for management of pastureland and livestock wastes led to total nitrogen (TN) and total phosphorus (TP) load reductions of 43% and 34% from pastureland, and 61.5% and 57.9% from farm animals, respectively, but the watershed annual loads decreased only by 4%. The combined BMP scenario targeting agricultural practices, pasture management and animal waste control provided the maximum reduction in total annual diffuse pollution loads by 35% for TN and 29% for TP. The holistic nature of this study contributes to the advancement of water quality modeling and provides valuable insights for sustainable water resources planning and decision-making. Non-point source pollution poses a continuous threat to the quality of Great Lakes waters. To abate this problem, the Great Lakes Agricultural Stewardship Initiative (GLASI) was initiated in Ontario, Canada, with the primary aim of reducing phosphorus pollution. Therefore, a case-study analysis of the Wigle Creek watershed, one of the six priority watersheds under the GLASI program, was undertaken to evaluate the effectiveness of various existing and potential future Best Management Practices (BMPs) and to identify BMPs that might aid in mitigating the watershed's contribution to phosphorus loads reaching Lake Erie. Given the watershed's very flat topography, hydrological/nutrient modeling proved an extremely challenging exercise. The Soil and Water Assessment Tool (SWAT) model was used in this evaluation. Several digital elevation model (DEM) options were considered to accurately describe the watershed and represent flow conditions. A 30 m resolution DEM, implementing a modified burning in of streams based on ground truthing, was finally employed to develop the SWAT model's drainage framework. The model was first calibrated for flow, sediment, and phosphorus loads. The calibrated model was used to evaluate the ability of potential BMPs (minimum tillage, no-till, retiring croplands into pasture, retiring croplands into forest, winter wheat cover crop, and vegetative filter strips) to reduce phosphorus loads compared to implemented practice. Converting all croplands into pasture or forest significantly decreased P loads reaching Lake Erie. Comparatively, a winter wheat cover crop had minimal effect on reducing phosphorus loading. This study conducted a spatio-temporal analysis of runoff, total suspended sediment, suspended particulate carbon, nitrogen, and phosphorus loadings within the 2.06 km2 Steppler subwatershed in southern Manitoba of Canada based on 11 years of field monitoring data collected at nine stations. Results showed that the nutrient losses were very small because of the implementation of multiple BMPs in the study area. However, a high spatio-temporal variation of runoff and water quality parameters was found for the nine fields within the subwatershed. The average runoff coefficient was 0.19 at the subwatershed outlet with sediment, suspended particulate carbon, total nitrogen, and total phosphorus losses of 73.8, 6.10, 4.54, and 0.76 kg/ha respectively. Spring snowmelt runoff was about 74.5% of the annual runoff at the subwatershed outlet, while for sediment, suspended particulate carbon, total nitrogen, and total phosphorus, the proportions were 61.1%, 63.6%, 74.9%, and 81.2% respectively during the monitoring period, which suggests that BMPs designed for reducing nutrient loadings from snowmelt runoff would be more effective than BMPs designed for reducing pollutant loading from rainfall storms in the study area. Research findings from this study will benefit the enhancement of current BMPs and the development of new BMPs in the region to minimize soil and nutrient losses from agricultural fields and improve water quality in receiving water bodies. The aim of this study is to evaluate the current water quality status of one of the urban rivers in Malaysia, called Sungai Air Hitam. The river's water supply is not only unsuitable for the inhabitants but also hazardous to the aquatic species that depend on it. In order to simulate the water quality formulation of the river, the Model for Urban Stormwater Improvement Conceptualization (MUSIC) was used. The effects of various best management practices (BMPs) components have been examined to improve the river's water quality. This study also investigated different scenarios of the expected future changes in the land cover and the quality of the river. As the proportion of impervious surfaces increases, the urban hydrology cycle can be significantly altered, resulting in an increase in volumes and peak flows, and a decrease in storage, infiltration, and interception. The MUSIC results have shown significant reductions in biochemical oxygen demand (BOD), total suspended solids (TSS), total phosphorus (TP), and total nitrogen (TN) after introducing BMPs. It was also noticed that the prediction of pollutants falls within the acceptable range set by the Urban Stormwater Management Manual for Malaysia (MSMA) 2nd edition. For the land cover, it was found that the total reduction of BOD, TSS, TP, and TN for existing land use is 92.5 %, 94.5 %, 90.7 % and 91.9 %. Meanwhile, the total reduction in future land use is 81.6 % for BOD, 86.2 % for TSS, 80.9 % for TP and 80.8 % for TN. From the simulation results, it was observed that the application of BMPs has successfully reduced the observed mean BOD concentration from 92.38 mg/L (Class V) to 6.93 mg/L (Class IV) of the national water quality standards, NWQS, water quality index. As a result, the water quality index of the overall catchment has improved from Class IV to Class III (WQ1, WQ3, and WQ4) and from Class V to IV (WQ2) with the application of the BMPs. This assessment aims to raise awareness within the Sungai Air Hitam community regarding the importance of preserving river cleanliness and understanding the long-term environmental impact of water quality. These findings underscore the importance of an integrated system in managing urban water systems, which can offer valuable insight to the decision-makers.

The calcium dependent Calpain proteases are modulatory enzymes with important roles in cell cycle control, development and immunity. In the fly model Drosophila melanogaster Calpain A cleaves Cactus/IkappaB and consequently modifies Toll signals during embryonic dorsal-ventral (DV) patterning. Here we explore the role of Calpains in the hemiptera Rhodnius prolixus, an intermediate germband insect where the Bone Morphogenetic Protein (BMP) instead of the Toll pathway plays a major role in DV patterning. Phylogenetic analysis of Calpains in species ranging from Isoptera to Diptera indicates an increase of Calpain sequences in the R. prolixus genome and other hemimetabolous species. One locus encoding each of the CalpC, CalpD and Calp7 families, and seven Calpain A/B loci are present in the R. prolixus genome. Several predicted R. prolixus Calpains display a unique architecture, such as loss of Calcium-binding EF-hand domains and loss of catalytic residues in the active site CysPc domain, yielding catalytically dead Calpains A/B. Knockdown for one of these inactive Calpains results in embryonic DV patterning defects, with expansion of ventral and lateral gene expression domains and consequent failure of germ band elongation. In conclusion, our results reveal that Calpains may exert a conserved function in insect DV patterning, despite the changing role of the Toll and BMP pathways in defining gene expression territories along the insect DV axis.

In this study, an attempt has been made to evaluate the best management practices (BMPs) in a poorly degraded and economically fragile watershed area in Shivalik hills, Northern India. A scenario-based approach was followed to evaluate the existing BMPs and a few hypothetically implemented BMPs based on SWAT modeling. A regionalization approach was adopted for calibrating the SWAT model for ungauged micro-watersheds within the study area. After successful calibration, soil and water assessment tool (SWAT) model was simulated for several BMPs that are in practice such as Check dams, vegetation and fencing so as to learn about their effectiveness in controlling sediment and nutrient yield. Other hypothetically installed BMPs, such as contouring, terracing, grassed waterways and filter strips were also evaluated in untreated micro and sub-watersheds. The cost–benefit analysis of these hypothetical BMPs revealed that the average reduction in nutrients was maximum for grassed waterways and minimum for terracing. Overall, the scenario-based analysis revealed that conservation practices, in the otherwise degraded watershed, can prove to be beneficial for sustainability of its natural resources. INTRODUCTION: Osteoporosis is an important health problem and there is no effective treatment yet. phosphodiesterase 5 inhibitors are promising agents for the treatment of osteoporosis. In this study, we aimed to determine the effects of phosphodiesterase 5 inhibitors (vardenafil, tadalafil, and udenafil) on bone morphogenic protein-2 and 9 (BMP-2 and 9), vascular endothelial growth factor (VEGF), and oxidative stress markers (malondialdehyde and CoQ10) in liver tissue of rats with ovariectomy-induced osteoporosis. METHODS: 50 Albino wistar female rats were randomly divided into 5 groups of 10 rats per group. Groups were the sham-operated, ovariectomise (OVEX), OVEX + Tadalafil, OVEX + udenafil and OVEX + vardenafil, respectively. VEGF, BMP-2 and 9 levels were measured by ELISA kits. To detect levels of MDA and CoQ10, we used high pressure liquid chromatography method. RESULTS: The levels of VEGF, BMP-2 and 9 levels in the groups that applied PDE-5 inhibitors were significantly higher than in the sham and OVEX groups. There was no significant difference between the OVEX+vardenafil and OVEX+udenafil groups in terms of VEGF, BMP-2 and 9 levels. The levels of MDA and CoQ10 were significantly lower in the groups that applied PDE5 inhibitors than in the OVEX group. When the histological and immunohistochemical results were examined, it was seen that angiogenesis was significantly higher in PDE-5 inhibitor groups. DISCUSSION AND CONCLUSION: In conclusion, we can say that these inhibitors may have positi ve effects on bone mineralization and remodelling by inducing the expression of VEGF, BMP-2 and 9 in liver tissue.

Together, these findings suggest that MT has the potential to ameliorate inflammation, inhibit the release of matrix-degrading enzymes, and improve the cartilage condition. This study provides a new theoretical basis for understanding the role of MT in decelerating cartilage degradation and promoting chondrocyte repair in in vivo and in vitro cultured chondrocytes.

Nonylphenol (NP) is a known endocrine-disrupting chemical (EDC) that has been shown to affect bone development in mammals. However, the detrimental impacts of NP on the skeletal growth and development of aquatic species, especially bony fish, remain poorly understood. Bone morphogenic proteins (BMPs), essential for bone formation and osteoblast differentiation, act through the BMP-Smad signaling pathway. In this study, two BMP genes, BMP2 and BMP4, were cloned and characterized in the red crucian carp (Carassius auratus red var.). The full-length cDNAs of BMP2 and BMP4 were 2029 bp and 2095 bp, respectively, encoding polypeptides of 411 and 433 amino acids, and share a typical TGF-β domain with other BMPs. The tissue expression patterns of both genes were identified, showing ubiquitous expression across all studied tissues. Additionally, the exposure of embryos or adult fish to NP stress resulted in a downregulation of BMP2, BMP4, and other genes associated with the BMP-Smad signaling pathway. Moreover, the combined treatment of adult fish with NP and the specific BMP receptor inhibitor significantly reduced these genes’ expression. These findings elucidate the mechanism of NP stress on BMP2 and BMP4, suggesting a role for the BMP-Smad signaling pathway in the response to endocrine-disrupting chemicals in fish.

Objective To investigate whether the Anti-Müllerian Hormone (AMH), an ovarian hormone belonging to the Transforming Growth Factor β superfamily, may represent a possible candidate for use as a bone anabolic factor.Methods We performed in vitro studies on Human Osteoblasts (HOb) to evaluate the expression and the functionality of AMHRII, the AMH receptor type-2, and investigate the effects of exogenous AMH exposure on osteogenic gene expression and osteoblast functions.Results We reported the first evidence for the expression and functionality of AMHRII in HOb cells, thus suggesting that osteoblasts may represent a specific target for exogenous AMH treatment. Furthermore, the exposure to AMH exerted a stimulatory effect on HOb cells leading to the activation of osteogenic genes, including the upregulation of osteoblastic transcription factors such as RUNX and OSX, along with increased deposition of mineralized nodules.Conclusion Our findings proved interesting clues on the stimulatory effects of AMH on mature osteoblasts expressing its specific receptor, AMHRII. This study may therefore have translation value in opening the perspective that AMH may be an effective candidate to counteract the bone loss in osteoporotic patients by selectively targeting osteoblast with minimal off-target effect.

Osteoinductive BMPs require a suitable delivery system for treating various pathological conditions of the spine and segmental bone defects. INFUSE, the only commercially available BMP-based osteoinductive device, consisting of rhBMP2 on bovine absorbable collagen sponge (ACS) showed major disadvantages due to serious side effects. A novel osteoinductive device, OSTEOGROW, comprised of rhBMP6 dispersed within autologous blood coagulum (ABC) is a promising therapy for bone regeneration, subjected to several clinical trials for diaphysial bone repair and spinal fusion. In the present study, we have examined the release dynamics showing that the ABC carrier provided a slower, more steady BMP release in comparison to the ACS. Rat subcutaneous assay was employed to evaluate cellular events and the time course of ectopic osteogenesis. The host cellular response to osteoinductive implants was evaluated by flow cytometry, while dynamics of bone formation and maintenance in time were evaluated by histology, immunohistochemistry and micro CT analyses. Flow cytometry revealed that the recruitment of lymphoid cell populations was significantly higher in rhBMP6/ABC implants, while rhBMP2/ACS implants recruited more myeloid populations. Furthermore, rhBMP6/ABC implants more efficiently attracted early and committed progenitor cells. Dynamics of bone formation induced by rhBMP2/ACS was characterized by a delayed endochondral ossification process in comparison to rhBMP6/ABC implants. Besides, rhBMP6/ABC implants induced more ectopic bone volume in all observed time points in comparison to rhBMP2/ACS implants. These results indicate that OSTEOGROW was superior to INFUSE due to ABC's advantages as a carrier and rhBMP6 superior efficacy in inducing bone.

This study was conducted in the Atwood and Tappan Lakes watersheds of the Tuscarawas basin of Ohio. The flow, total nitrogen (TN), and total phosphorus (TP) loadings were monitored with the help of local stakeholders for a few years at various locations of the watershed to develop the Soil and Water Assessment Tool (SWAT). The multi-site SWAT model calibration and validation were accomplished with a reasonable model performance. In the next step, the scenario analysis was conducted in the SWAT model using various BMPs, including vegetative filter strips, grass waterways, fertilizer reduction, crop rotation, and cover crops to evaluate their performance in reducing TN and TP from the watershed. While BMPS in many studies are decided based on researchers' intuition, these BMPs were selected based on active consultation with the local stakeholders, who were engaged in the reduction of TN and TP loadings from the watersheds. Since the SWAT model calibration for TN and TP was not as good as the hydrologic model calibration, various scenarios of TN and TP reduction using BMPs were investigated for several years using both calibrated and uncalibrated SWAT models. We examined all the BMPs in 12 sub-watersheds of the Atwood and 10 sub-watersheds of the Tappan Lake watershed. The analysis indicated that the management practices of cover crops (rye) in combination with grass waterways with a 10% fertilizer reduction could minimize the TN and TP loading by as much as 88%, without significantly compromising the agricultural yield. However, a 10% fertilizer reduction without any BMPs could reduce TN and TP by just 9%. The cover crop (rye) including 10% fertilizer reduction with grass waterways seemed to be the most effective in reducing TN and TP, whereas the implementation of a filter strip led to a 70% reduction and was the next effective BMPs in reducing TN and TP loadings. In general, TN losses were reduced by 8% to 53%, while TP losses were reduced by 7% to 88%, depending on the BMPs used. By and large, the TN and TP reduction achieved through the calibrated model was not significantly different from the uncalibrated model, even though the reduction using the calibrated model was slightly higher for all scenarios than that of the uncalibrated model. The TN and TP loadings were highly sensitive to cattle grazing. When just 50% of the cattle were permitted to graze, the model predicted that there would be a 40% increase in total nitrogen and a 70% increase in total phosphorus in both watersheds. In the present studies, we are reporting synthesis, structural, microstructural, optical dielectric, and magnetic properties of pristine and Ni-doped CdO (Cd1-xNixO, 0% ≤ x ≤ 10%) nanoparticles. Co-precipitation method was used to synthesize these nanoparticles. X-ray diffraction patterns show the cubic phase of polycrystalline Cd1–xNixO nanoparticles along with the formation of secondary NiO phase at higher Ni-content. The XPS spectra indicate the presence of oxygen vacancies along with mixed oxidation states of nickel (Ni2+ and Ni3+) and cadmium (Cd2+, Cd4+). FESEM micrographs show non uniform distribution of grains in these nanoparticles. The observed Raman peaks show that Ni2+ ions are successfully substituted at the Cd2+ lattice sites in the host lattice of CdO. The decrease in ac-conductivity of CdO with Ni-doping indicates the segregation of defects (Cd interstitials and oxygen vacancies) at grain boundaries. The conduction phenomena are explained by correlated barrier hopping mechanism in these nanoparticles. The optical band-gap broadening of CdO with Ni-doping show the influence of carrier densities. Subsurface (or tile) drainage improves land productivity by enhancing soil aeration and preventing water-logged conditions. However, the continuous expansion of drained agricultural lands and reliance on synthetic fertilizer in the Midwestern United States have increasingly facilitated nitrate transport from agricultural fields to surface water bodies. Hence, there is a need to implement various agricultural best management practices (BMPs) in order to reduce the adverse water quality impacts resulting from excess nitrate, such as eutrophication and the formation of hypoxic zones. In this study, we used a SWAT+ model to assess the overall impacts on the riverine nitrate load and crop yield in the corn–soybean cropping system based on a combination of different management practices. The corn and soybean yields simulated with the model were found to be in good agreement with the observed yields for both the calibration and validation periods. The long-term simulation over a period of 30 years showed a reduction in the nitrate load of up to 32% without impacting the crop yield. The model results suggest that by reducing the current N application rate by 20% and using a 40:60 split between spring pre-plant and side-dressing N applications combined with cereal rye as a cover crop in corn–soybean rotation, one can potentially reduce nitrate losses without impacting crop yields. This study will help researchers, stakeholders, and farmers to explore and adopt alternative management practices beneficial for offsetting the environmental impacts of agricultural productions on the watershed scale. IntroductionTimber harvests influence coarse woody debris (CWD) dynamics both initially and long-term—contributing a significant amount of CWD as slash immediately after harvest, but also removing some or all of the mature trees necessary to produce CWD over time. Whereas shelterwood and other similar timber harvest systems retain varying amounts of the overstory, preserving CWD production after harvest, commercial clearcutting essentially eliminates sources of fresh CWD until regenerating trees are large enough to contribute CWD through fallen limbs or trunks, often decades after harvest. Forestry best management practices (BMPs) are critical for reducing the water quality impacts of timber harvest, but their effects on riparian and stream CWD are not well understood.MethodsThe current project explores CWD dynamics (surveyed in 2001, 2004, 2009, and 2020) in three eastern Kentucky watersheds receiving differing timber harvest treatments in 1983: unharvested control, BMPs (clearcut with a protected streamside management zone), and no BMPs (clearcut).ResultsAnalysis of hydrology data over the period of record demonstrated significant flooding in 2004 that likely “reset” CWD in the study watersheds. Coarse woody debris volume was higher in control (3.33 m3/ha) than no BMP (1.03 m3/ha) in 2020, with CWD accumulation rates ranging from 0.039 m3/ha in the no BMP watershed to 0.19 m3/ha in the control.DiscussionWhile not significantly different, CWD volume was nominally higher in the BMP watershed than the No BMP watershed, suggesting that, in addition to their many other benefits, streamside management zones help facilitate CWD provisioning during stand initiation after a commercial clearcut harvest. Furthermore, this study suggests that provisioning of CWD may not recover after clearcut harvesting for 100 years or more.

This investigation explores the impact of various management practices on nitrate leaching and crop yield in two specific areas in Slovenia. The issue of nitrate leaching from agricultural land is a worldwide threat to drinking water, and despite years of research efforts, universal solutions are still unknown. The two chosen study sites are significant because of their importance for agricultural production and because groundwater aquifers beneath are main sources of drinking water, which makes imposing mitigation measures challenging. One of the areas was reported to be of "bad" status according to the Water Framework Directive criteria, while the other is at risk of reaching this status if nitrate concentrations in groundwater continue to rise. This research used the SWAT model to simulate nitrate leaching and crop yield changes under different agricultural scenarios on different soil types. It aimed to accomplish two objectives: first, to identify parts of the case study areas where the existing combination of soil conditions and agricultural practices enables a high potential for nitrate leaching; second, to identify agricultural practices that decrease nitrate leaching from various soil types while maintaining crop yields in each area. By identifying the most vulnerable locations and the most promising practices, we generated a chart of best management practices for specific soil types as a guide that extension services can use to advise farmers on potential management improvements. The main findings demonstrate that reducing fertilizer application, both organic and inorganic, in areas where the rates exceed crop requirements may not have a significant impact on crop production. However, these reductions often resulted in a noticeable decrease in nitrate exports. The results also showed that soil type is crucially important and should always be considered when evaluating the effects of agricultural management on crop yields and nitrate leaching. Abstract Pollution of a watershed by different land uses and agricultural practices is becoming a major challenging factor that results in deterioration of water quality affecting human health and ecosystems. Sustainable use of available water resources warrants reduction of Non-Point Source (NPS) pollutants from receiving water bodies through best management practices (BMPs). A hydrologic model such as the Soil and Water Assessment Tool (SWAT) can be used for analyzing the impacts of various BMPs and implementing of different management plans for water quality improvement, which will help decision makers to determine the best combination of BMPs to maximize benefits. The objective of this study is to assess the potential reductions of sediments and nutrient loads by utilizing different BMPs on the Yarra River watershed using the SWAT model. The watershed is subdivided into 51 sub-watersheds where seven different BMPs were implemented. A SWAT model was developed and calibrated against a baseline period of 1998–2008. For calibration and validation of the model simulations for both the monthly and annual nutrients and sediments were assessed by using the Nash–Sutcliffe efficiency (NSE) statistical index. The values of the NSE were found more than 0.50 which indicates satisfactory model predictions. By utilizing different BMPs, the highest pollution reduction with minimal costs can be done by 32% targeted mixed-crop area. Furthermore, the combined effect of five BMPs imparts most sediments and nutrient reductions in the watershed. Overall, the selection of a BMP or combinations of BMPs should be set based on the goals set in a BMP application project.

Abstract Background This study aimed to get a deeper insight into new osteosarcoma (OS) signature based on bone morphogenetic proteins (BMPs)-related genes and to confirm the prognostic pattern to speculate on the overall survival among OS patients. Methods Firstly, pathway analyses using Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) were managed to search for possible prognostic mechanisms attached to the OS-specific differentially expressed BMPs-related genes (DEBRGs). Secondly, univariate and multivariate Cox analysis was executed to filter the prognostic DEBRGs and establish the polygenic model for risk prediction in OS patients with the least absolute shrinkage and selection operator (LASSO) regression analysis. The receiver operating characteristic (ROC) curve weighed the model’s accuracy. Thirdly, the GEO database (GSE21257) was operated for independent validation. The nomogram was initiated using multivariable Cox regression. Immune infiltration of the OS sample was calculated. Finally, the three discovered hallmark genes’ mRNA and protein expressions were verified. Results A total of 46 DEBRGs were found in the OS and control samples, and three prognostic DEBRGs (DLX2, TERT, and EVX1) were screened under the LASSO regression analyses. Multivariate and univariate Cox regression analysis were devised to forge the OS risk model. Both the TARGET training and validation sets indicated that the prognostic biomarker-based risk score model performed well based on ROC curves. In high- and low-risk groups, immune cells, including memory B, activated mast, resting mast, plasma, and activated memory CD4 + T cells, and the immune, stromal, and ESTIMATE scores showed significant differences. The nomogram that predicts survival was established with good performance according to clinical features of OS patients and risk scores. Finally, the expression of three crucial BMP-related genes in OS cell lines was investigated using quantitative real-time polymerase chain reaction (qRT-PCR) and western blotting (WB). Abstract The NF‐κB family of transcription factors plays an important role in skeletal development and bone homeostasis. In osteoblast cells, NF‐κB signaling has been shown to suppress survival, proliferation, and differentiation. Furthermore, pharmacological suppression of NF‐κB enhances osteoblast differentiation and bone formation. Thus, NF‐κB antagonists are promising candidates as anabolic agents for enhancing bone mass. In this study, we describe the mechanism by which nobiletin, an inhibitor of NF‐κB activity, regulates osteoblast differentiation and mineralization. We found that in MC3T3‐E1 osteoblast cells, nobiletin inhibited a TNF‐α responsive NF‐κB luciferase reporter and also decreased the induction of classical NF‐κB target genes by TNF‐α. Consistent with this, nobiletin prevented TNF‐α ‐mediated suppression of osteogenesis and potently enhanced the differentiation and mineralization of MC3T3‐E1 cells. Likewise, in an in vivo BMP2‐induced ectopic bone formation assay, nobiletin markedly enhanced ossicle bone volume. Western blotting and SMAD‐responsive luciferase assays also demonstrated that NF‐κB suppression of BMP signaling could be inhibited by nobiletin. Thus, our data suggest that mechanistically, nobiletin prevents the endogenous repression of BMP signaling by TNF‐α, thereby enhancing osteoblast activity. In conclusion, nobiletin is a novel NF‐κB antagonist that may be a useful anabolic agent for bone formation.

Bone fragility is the product of defects in bone mass and bone quality, both of which show sex-specific differences. Despite this, the cellular and molecular mechanisms underpinning the sexually dimorphic control of bone quality remain unclear, limiting our ability to effectively prevent fractures, especially in postmenopausal osteoporosis. Recently, using male mice, we found that systemic or osteocyte-intrinsic inhibition of TGFβ signaling, achieved using the 9.6-kb DMP1 promoter-driven Cre recombinase (TβRIIocy-/- mice), suppresses osteocyte perilacunar/canalicular remodeling (PLR) and compromises bone quality. Because systemic TGFβ inhibition more robustly increases bone mass in female than male mice, we postulated that sex-specific differences in bone quality could likewise result, in part, from dimorphic regulation of PLR by TGFβ. Moreover, because lactation induces PLR, we examined the effect of TGFβ inhibition on the female skeleton during lactation. In contrast to males, female mice that possess an osteocyte-intrinsic defect in TGFβ signaling were protected from TGFβ-dependent defects in PLR and bone quality. The expression of requisite PLR enzymes, the lacunocanalicular network (LCN), and the flexural strength of female TβRIIocy-/- bone was intact. With lactation, however, bone loss and induction in PLR and osteocytic parathyroid hormone type I receptor (PTHR1) expression, were suppressed in TβRIIocy-/- bone, relative to the control littermates. Indeed, differential control of PTHR1 expression, by TGFβ and other factors, may contribute to dimorphism in PLR regulation in male and female TβRIIocy-/- mice. These findings provide key insights into the sex-based differences in osteocyte PLR that underlie bone quality and highlight TGFβ signaling as a crucial regulator of lactation-induced PLR. © 2020 American Society for Bone and Mineral Research.

MicroRNAs (miRNAs) are a family of small, single-stranded, and non-protein coding RNAs about 19 to 22 nucleotides in length, that have been reported to have important roles in the control of bone development. MiRNAs have a strong influence on osteoblast differentiation through stages of lineage commitment and maturation, as well as via controlling the activities of osteogenic signal transduction pathways. Generally, miRNAs may modulate cell stemness, proliferation, differentiation, and apoptosis by binding the 3′-untranslated regions (3′-UTRs) of the target genes, which then can subsequently undergo messenger RNA (mRNA) degradation or protein translational repression. MiRNAs manage the gene expression in osteogenic differentiation by regulating multiple signalling cascades and essential transcription factors, including the transforming growth factor-beta (TGF-β)/bone morphogenic protein (BMP), Wingless/Int-1(Wnt)/β-catenin, Notch, and Hedgehog signalling pathways; the Runt-related transcription factor 2 (RUNX2); and osterix (Osx). This shows that miRNAs are essential in regulating diverse osteoblast cell functions. TGF-βs and BMPs transduce signals and exert diverse functions in osteoblastogenesis, skeletal development and bone formation, bone homeostasis, and diseases. Herein, we highlighted the current state of in vitro and in vivo research describing miRNA regulation on the canonical TGF-β/BMP signalling, their effects on osteoblast linage, and understand their mechanism of action for the development of possible therapeutics. In this review, particular attention and comprehensive database searches are focused on related works published between the years 2000 to 2022, using the resources from PubMed, Google Scholar, Scopus, and Web of Science. Polycrystalline nanoparticles of Al-doped and Ce-co-doped ZnO were processed through sol)gel co-precipitation. Crystallite sizes determined from XRD were in the range of 10.90 to 48.77 nm. FTIR spectra indicated a stretching mode of Zn-O bond. The insertion of Al and Ce at Zn-O site created Al-O and Ce-O bonds. UV-vis spectra favoured the formation of impurity levels by doping and co-doping. Their overlapping with the conduction band edge led to the reduction of band-gap. Blue-green emission which arises from radiative recombination of a photogenerated hole with an electron occupying oxygen vacancy was observed in photo)luminescence spectra. FESEM suggested granular growth in undoped ZnO which changed to diverse structures such as nano-bar and cluster of elongated grains. Significant improvement in room temperature ferromagnetism (RTFM) was noticed by co-doping of Ce in Al-doped ZnO. Non-point source pollution poses a continuous threat to the quality of Great Lakes waters. To abate this problem, the Great Lakes Agricultural Stewardship Initiative (GLASI) was initiated in Ontario, Canada, with the primary aim of reducing phosphorus pollution. Therefore, a case-study analysis of the Wigle Creek watershed, one of the six priority watersheds under the GLASI program, was undertaken to evaluate the effectiveness of various existing and potential future Best Management Practices (BMPs) and to identify BMPs that might aid in mitigating the watershed’s contribution to phosphorus loads reaching Lake Erie. Given the watershed’s very flat topography, hydrological/nutrient modeling proved an extremely challenging exercise. The Soil and Water Assessment Tool (SWAT) model was used in this evaluation. Several digital elevation model (DEM) options were considered to accurately describe the watershed and represent flow conditions. A 30 m resolution DEM, implementing a modified burning in of streams based on ground truthing, was finally employed to develop the SWAT model’s drainage framework. The model was first calibrated for flow, sediment, and phosphorus loads. The calibrated model was used to evaluate the ability of potential BMPs (minimum tillage, no-till, retiring croplands into pasture, retiring croplands into forest, winter wheat cover crop, and vegetative filter strips) to reduce phosphorus loads compared to implemented practice. Converting all croplands into pasture or forest significantly decreased P loads reaching Lake Erie. Comparatively, a winter wheat cover crop had minimal effect on reducing phosphorus loading.

The bone morphogenic protein (BMP) family is a member of the TGF-beta superfamily and plays a crucial role during the onset of gut inflammation and arthritis diseases. Recent studies have reported a connection with the gut–joint axis; however, the genetic players are still less explored. Meanwhile, BDMC33 is a newly synthesized anti-inflammatory drug candidate. Therefore, in our present study, we analysed the genome-wide features of the BMP family as well as the role of BMP members in gut-associated arthritis in an inflammatory state and the ability of BDMC33 to attenuate this inflammation. Firstly, genome-wide analyses were performed on the BMP family in the zebrafish genome, employing several in silico techniques. Afterwards, the effects of curcumin analogues on BMP gene expression in zebrafish larvae induced with TNBS (0.78 mg/mL) were determined using real time-qPCR. A total of 38 identified BMP proteins were revealed to be clustered in five major clades and contain TGF beta and TGF beta pro peptide domains. Furthermore, BDMC33 suppressed the expression of four selected BMP genes in the TNBS-induced larvae, where the highest gene suppression was in the BMP2a gene (an eight-fold decrement), followed by BMP7b (four-fold decrement), BMP4 (four-fold decrement), and BMP6 (three-fold decrement). Therefore, this study reveals the role of BMPs in gut-associated arthritis and proves the ability of BDMC33 to act as a potential anti-inflammatory drug for suppressing TNBS-induced BMP genes in zebrafish larvae. Forest roads often increase runoff and sediment loss, thus greatly impacting hydrological processes in mountainous watersheds. While there has been previous investigation on best management practices (BMPs) to reduce soil erosion from forest roads, few studies have attempted to optimize BMPs based on how much they can decrease sediment connectivity between forest roads and streams. To close this gap in knowledge, we analyzed the spatial relationship between forest roads and streams, presented the spatial distribution of sediment connectivity by integrating the forest roads into the calculation of the index of connectivity (IC), determined how sediment connectivity would respond to additional BMPs through simulating scenarios, and used these data to optimize the BMPs so they would intercept the greatest sediment loads. We found that forest roads and streams in the Xiangchagou watershed in the Dabie Mountain area of China tend to occur within 180 m of each other; however, within the same buffer zones, streams are more often accompanied by forest roads. IC was greatest near road–stream crossings but smaller near streams and forest roads, and it tended to decrease as the buffer distance increased. Furthermore, we found that sediment connectivity was decreased through running a variety of scenarios that used sediment basin and riparian buffers as BMPs between forest roads and streams. Specifically, within this watershed, riparian buffers should be 64 m wide, and there should be 30 sediment basins with a minimum upslope drainage area of 2 ha. At these quantities, the BMPs in this watershed would significantly affect sediment connectivity. By contrast, beyond these thresholds, increasing the width of riparian buffers or the number of sediment basins does not lead to meaningful sediment reductions. In this way, we were able to use the mean change point method to determine the optimal sediment basin quantity (30 with corresponding minimum upslope drainage area of 2 ha) and the optimal riparian buffer width (64 m) for the Xiangchagou watershed. While these results are a first approximation in a novel research area, they can guide forest managers and stakeholders to design and optimize BMPs that control the delivery of eroded sediments associated with forest roads. The origin of ferromagnetism in nanoparticles of polycrystalline Cd1−xMnxO (x = 0, 2%, 10%) prepared using co-precipitation method is presented. The XPS studies indicate the presence of oxygen vacancies along with mixed oxidation states of manganese (Mn4+ and Mn2+) and cadmium (Cd2+, Cd4+). The room temperature isothermal hysteresis measurements show that upon Mn doping, there is enhancement in ferromagnetic ordering and in coercivity of CdO nanoparticles. The zero-field-cooled and field-cooled magnetization as function of temperature indicates the presence of disordered magnetic structure, which may be due to the formation of bound magnetic polarons by mixed valance states of Mn cations and oxygen vacancies. The enhancement in dielectric constant with Mn substitution also points to the enhancement of space charge polarization and the frequency-dependent ac-conductivity indicates the small polarons hoping mechanism of conductivity. As demand for wood energy rises, it's vital to understand the environmental impact of various harvest types, considering different best management practices (BMPs) and site conditions, for sustainable forest management. This study provides a comparative analysis of postharvest site characteristics between biomass and conventional harvest sites located in the Coastal Plain region of the southeastern US. A total of 30 recently harvest sites (15 each for biomass and conventional) were selected from the Coastal regions of Alabama, Georgia, and Florida. Erosion rates were estimated using the USLE-Forest method and ground cover was assessed as a percentage of the area covered by bare soil, litter, light slash, heavy slash, and piles. BMP implementation (%) was based on state-specific BMP surveys. The findings show that stream management zones (SMZs) and harvest areas were less prone to soil erosion, while forest roads, skid trails, and stream crossings were more prone to erosion, regardless of the harvest type. Conventional harvest sites had an overall weighted average erosion rate of 0.35 Mg/ha/year, with an average BMP implementation of 92.70 %, while biomass harvest sites had an overall weighted average erosion rate of 0.22 Mg/ha/year, with an average BMP implementation rate of 97.91 %. Average erosion rate (Mg/ha/year) from the harvest area (biomass=0.11, conventional=0.06) and SMZs (biomass=0.05, conventional=0.02) were not significantly different (p >0.05). Conversely, average erosion rate from roads (biomass=4.17, conventional=6.44), skid trails (biomass=2.15, conventional=3.73), decks (biomass=0.39, conventional=0.72), and stream crossings (biomass=1.53, conventional=5.31) were significantly higher in conventional harvest sites (p <0.05). BMP implementation rates for roads (biomass=98.09 %, conventional=93.71 %), stream crossings (biomass=95 %, conventional=71 %), and timber harvest (biomass=96.44 %, conventional=93.33 %) were significantly higher in biomass harvest sites, while waste management BMP (biomass=89.91 %, conventional=100 %) was significantly higher in conventional harvest sites. Regarding ground cover, conventional harvest sites had significantly larger areas covered by light slash, heavy slash, and piles than biomass harvest sites, while biomass harvest sites had significantly larger areas covered by green growth. Our research shows a significant negative correlation between overall BMP implementation and erosion rates (r=-0.67, p =0.0002), as well as site size (r=-0.38 p =0.0400), indicating that larger sites tend to have lower BMP implementation and increased erosion risk. In conclusion, biomass harvest sites in our study did not yield higher erosion rates in comparison to conventional harvest sites, provided that the current BMP recommendations were implemented properly and in adequate amounts. This study was conducted in the Atwood and Tappan Lakes watersheds of the Tuscarawas basin of Ohio. The flow, total nitrogen (TN), and total phosphorus (TP) loadings were monitored with the help of local stakeholders for a few years at various locations of the watershed to develop the Soil and Water Assessment Tool (SWAT). The multi-site SWAT model calibration and validation were accomplished with a reasonable model performance. In the next step, the scenario analysis was conducted in the SWAT model using various BMPs, including vegetative filter strips, grass waterways, fertilizer reduction, crop rotation, and cover crops to evaluate their performance in reducing TN and TP from the watershed. While BMPS in many studies are decided based on researchers’ intuition, these BMPs were selected based on active consultation with the local stakeholders, who were engaged in the reduction of TN and TP loadings from the watersheds. Since the SWAT model calibration for TN and TP was not as good as the hydrologic model calibration, various scenarios of TN and TP reduction using BMPs were investigated for several years using both calibrated and uncalibrated SWAT models. We examined all the BMPs in 12 sub-watersheds of the Atwood and 10 sub-watersheds of the Tappan Lake watershed. The analysis indicated that the management practices of cover crops (rye) in combination with grass waterways with a 10% fertilizer reduction could minimize the TN and TP loading by as much as 88%, without significantly compromising the agricultural yield. However, a 10% fertilizer reduction without any BMPs could reduce TN and TP by just 9%. The cover crop (rye) including 10% fertilizer reduction with grass waterways seemed to be the most effective in reducing TN and TP, whereas the implementation of a filter strip led to a 70% reduction and was the next effective BMPs in reducing TN and TP loadings. In general, TN losses were reduced by 8% to 53%, while TP losses were reduced by 7% to 88%, depending on the BMPs used. By and large, the TN and TP reduction achieved through the calibrated model was not significantly different from the uncalibrated model, even though the reduction using the calibrated model was slightly higher for all scenarios than that of the uncalibrated model. The TN and TP loadings were highly sensitive to cattle grazing. When just 50% of the cattle were permitted to graze, the model predicted that there would be a 40% increase in total nitrogen and a 70% increase in total phosphorus in both watersheds. Our investigation revealed that monitoring the watershed at a small sub-watershed scale and calibrating the SWAT model for nitrogen and phosphorus is delicate.
