The circular economy (CE), a sustainability concept that promotes resource efficiency and waste reduction, has garnered significant popularity in recent years due to its potential to address pressing environmental and economic challenges. This study applies the Bayesian Mindsponge Mindspongeconomics (BMM) framework/analytic method, based on the Bayesian Mindsponge Framework (BMF), to the factors influencing young adults' pro-environmental behavior and their purchases of green products at different price levels. The findings indicate that young adults who are knowledgeable about the CE and who value environmental protection and energy conservation are more likely to engage in waste sorting, while the factors that affect their willingness to pay (WTP) more for green and energy-saving products vary at different price tiers. This study demonstrates that knowledge of the CE, daily waste sorting habits, and environmental concern positively impact young adults' WTP for products that are priced 5%, 10%, and 15% higher, respectively. Furthermore, this study also highlights the potential of educational programs and cultural influences in nurturing a generation that prioritizes environmental value. This research integrates multidisciplinary perspectives and offers practical implications for policymakers, educators, and businesses seeking to promote green literacy and foster an environmental culture among the youth, contributing to the broader goals of green transformation and sustainable development associated with the CE and the green economy, especially in the urban areas of emerging countries and beyond. Background & aimsThe stimulator of interferon genes (STING)/TANK-binding kinase 1 (TBK1) pathway is vital in mediating innate immune and inflammatory responses during oxidative/endoplasmic reticulum (ER) stress. However, it remains unknown whether macrophage thioredoxin-interacting protein (TXNIP) may regulate TBK1 function and cell death pathways during oxidative/ER stress.MethodsA mouse model of hepatic ischaemia/reperfusion injury (IRI), the primary hepatocytes, and bone marrow-derived macrophages were used in the myeloid-specific TXNIP knockout (TXNIPM-KO) and TXNIP-proficient (TXNIPFL/FL) mice.ResultsThe TXNIPM-KO mice were resistant to ischaemia/reperfusion (IR) stress-induced liver damage with reduced serum alanine aminotransferase (ALT)/aspartate aminotransferase (AST) levels, macrophage/neutrophil infiltration, and pro-inflammatory mediators compared with the TXNIPFL/FL controls. IR stress increased TXNIP, p-STING, and p-TBK1 expression in ischaemic livers. However, TXNIPM-KO inhibited STING, TBK1, interferon regulatory factor 3 (IRF3), and NF-κB activation with interferon-β (IFN-β) expression. Interestingly, TXNIPM-KO augmented nuclear factor (erythroid-derived 2)-like 2 (NRF2) activity, increased antioxidant gene expression, and reduced macrophage reactive oxygen species (ROS) production and hepatic apoptosis/necroptosis in IR-stressed livers. Mechanistically, macrophage TXNIP deficiency promoted cylindromatosis (CYLD), which colocalised and interacted with NADPH oxidase 4 (NOX4) to enhance NRF2 activity by deubiquitinating NOX4. Disruption of macrophage NRF2 or its target gene 2',5' oligoadenylate synthetase-like 1 (OASL1) enhanced Ras GTPase-activating protein-binding protein 1 (G3BP1) and TBK1-mediated inflammatory response. Notably, macrophage OASL1 deficiency induced hepatocyte apoptotic peptidase activating factor 1 (APAF1), cytochrome c, and caspase-9 activation, leading to increased caspase-3-initiated apoptosis and receptor-interacting serine/threonine-protein kinase 3 (RIPK3)-mediated necroptosis.ConclusionsMacrophage TXNIP deficiency enhances CYLD activity and activates the NRF2-OASL1 signalling, controlling IR stress-induced liver injury. The target gene OASL1 regulated by NRF2 is crucial for modulating STING-mediated TBK1 activation and Apaf1/cytochrome c/caspase-9-triggered apoptotic/necroptotic cell death pathway. Our findings underscore a novel role of macrophage TXNIP-mediated CYLD-NRF2-OASL1 axis in stress-induced liver inflammation and cell death, implying the potential therapeutic targets in liver inflammatory diseases.Lay summaryLiver inflammation and injury induced by ischaemia and reperfusion (the absence of blood flow to the liver tissue followed by the resupply of blood) is a significant cause of hepatic dysfunction and failure following liver transplantation, resection, and haemorrhagic shock. Lipid droplets (LDs) are highly dynamic intracellular organelles, which are involved in lots of biological processes. However, the dynamic morphogenesis and functions of intracellular LDs during persistent innate immune responses remain obscure. In this study, we induce long-term systemic immune activation in Drosophila through genetic manipulation. Then, the dynamic pattern of LDs is traced in the Drosophila fat body. We find that deficiency of Plin1, a key regulator of LDs’ reconfiguration, blocks LDs minimization at the initial stage of immune hyperactivation but enhances LDs breakdown at the later stage of sustained immune activation via recruiting the lipase Brummer (Bmm, homologous to human ATGL). The high wasting in LDs shortens the lifespan of flies with high-energy-cost immune hyperactivation. Therefore, these results suggest a critical function of LDs during long-term immune activation and provide a potential treatment for the resolution of persistent inflammation.

Simple Summary: A study to investigate the effect that different Wolbachia pipientis strains have on the carbohydrate and lipid metabolism, starvation resistance and feeding behavior of Drosophila melanogaster females has been performed. The data obtained demonstrate that Wolbachia most likely change their hosts' metabolism in different aspects, including the effect on energy metabolism, fitness and behavior, to ensure the host's competitive advantage over uninfected insects, which contributes to the spread of the bacteria in the population. The effect of maternally inherited endosymbiotic bacteria Wolbachia on triglyceride and carbohydrate metabolism, starvation resistance and feeding behavior of Drosophila melanogaster females was studied. Eight D. melanogaster lines of the same nuclear background were investigated; one had no infection and served as the control, and seven others were infected with different Wolbachia strains pertaining to wMel and wMelCS groups of genotypes. Most of the infected lines had a higher overall lipid content and triglyceride level than the control line and their expression of the bmm gene regulating triglyceride catabolism was reduced. The glucose content was higher in the infected lines compared to that in the control, while their trehalose levels were similar. It was also found that the Wolbachia infection reduced the level of tps1 gene expression (coding for enzyme for trehalose synthesis from glucose) and had no effect on treh gene expression (coding for trehalose degradation enzyme). The infected lines exhibited lower appetite but higher survival under starvation compared to the control. The data obtained may indicate that Wolbachia foster their hosts' energy exchange through increasing its lipid storage and glucose content to ensure the host's competitive advantage over uninfected individuals. The primary objective of this study was to explore the extensive implications and complex molecular interactions arising from the confluence of excessive glucocorticoids and RANKL on the differentiation process of BMM into osteoclasts, profoundly impacting osteoporosis development. The methodology encompassed X-ray analysis and HE staining for evaluating bone loss in mice, while immunohistochemical staining was utilized to observe phosphorylated SHP2 (p-SHP2) expression. The assessment of several phosphorylated and total protein expression levels, including NF-κB, SHP2, SYK, JAK2, TAK1, NFATC1, c-fos, and Cathepsin K, was conducted via Western blotting. Additional experiments, involving CCK8 and monoclonal proliferation assays, were undertaken to determine BMM proliferation capacity. Immunofluorescence staining facilitated the quantification of TRAP fluorescence intensity. In vivo analysis revealed that glucocorticoid surplus triggers SHP2 signaling pathway activation, accelerating osteoporosis progression. Western blot results demonstrated that SHP2 inhibition could decrease the expression of specific proteins such as p-NF-κB and p-SHP2, with minimal effects on p-SYK levels. In vitro findings indicated that glucocorticoid and RANKL interaction activates the SHP2 pathway through NF-κB and SYK pathways, enhancing expressions of p-JAK2, p-TAK1, NFATC1, c-fos, and Cathepsin K, thereby promoting BMM to osteoclast transformation. Conclusion: Excessive glucocorticoids and RANKL interaction advance osteoclast differentiation from BMM by activating the SYK/SHP2/NF-κB signaling pathway, expediting osteoporosis progression.

The main objective of the study is to analyze the spectral behaviour and antimicrobial activity of 3-(4 bromophenyl)-1-methoxy-1-methyl urea by experiment, theoretical spectroscopic, IR, Raman and UV-Vis techniques. The optimized geometrical parameters were reported on DFT/B3LYP/6-311++G (d, p) basis set of theory. Based on the potential distribution of energy, the detailed vibrational assignments of observed FT-IR and FT-Raman bands have been proposed. The distribution of vibrational modes was carried out by using normal coordinate analysis. Antimicrobial activity of BMM molecule was confirmed using molecular docking analysis. This article introduces the idea of self-instrumenting endogenous regressors in settings when the correlation between these regressors and the errors can be derived and used to bias-correct the moment conditions. The resulting bias-corrected moment conditions are less likely to be subject to the weak instrument problem and can be used on their own or in conjunction with other available moment conditions to obtain more efficient estimators. This approach can be applied to estimation of a variety of models such as spatial and dynamic panel data models. This article focuses on the latter, and proposes a new estimator for short T dynamic panels by augmenting Anderson and Hsiao (AAH) estimator with bias-corrected quadratic moment conditions in first differences which substantially improve the small sample performance of the AH estimator without sacrificing the generality of its underlying assumptions regarding the fixed effects, initial values, and heteroskedasticity of error terms. Using Monte-Carlo experiments it is shown that AAH estimator represents a substantial improvement over the AH estimator and more importantly it performs well even when compared to Arellano and Bond and Blundell and Bond (BB) estimators that are based on more restrictive assumptions, and continues to have satisfactory performance in cases where the standard GMM estimators are inconsistent. Reproduction is an energy-intensive process requiring systemic coordination. However, the inter-organ signaling mechanisms that relay nutrient status to modulate reproductive output are poorly understood. Here, we use Drosophila melanogaster as a model to establish the integrated stress response (ISR) transcription factor, Atf4, as a fat tissue metabolic sensor that instructs oogenesis. We demonstrate that Atf4 regulates lipase activity to mediate yolk lipoprotein synthesis in the fat body. Depletion of Atf4 in the fat body also blunts oogenesis recovery after amino acid deprivation and re-feeding, suggestive of a nutrient-sensing role for Atf4. We also discovered that Atf4 promotes secretion of a fat-body-derived neuropeptide, CNMamide, which modulates neural circuits that promote egg-laying behavior (ovulation). Thus, we posit that ISR signaling in fat tissue acts as a "metabolic sensor" that instructs female reproduction—directly by impacting yolk lipoprotein production and follicle maturation and systemically by regulating ovulation. Lipolysis, the key process releasing fat acids to generate energy in adipose tissues, correlates with starvation resistance. Nevertheless, its detail mechanisms remain elusive. BubR1, an essential mitotic regulator, ensures proper chromosome alignment and segregation during mitosis, but its physiological functions are largely unknown. Here, we use Drosophila adult fat body, the major lipid storage organ, to study the functions of BubR1 in lipolysis. We show that both whole body- and fat body-specific BubR1 depletions increase lipid degradation and shorten the lifespan under fasting but not feeding. Relish, the conserved regulator of IMD signaling pathway, acts as the downstream target of BubR1 to control the expression level of Bmm and modulate the lipolysis upon fasting. Thus, our study reveals new functions of BubR1 in starvation-induced lipolysis and provides new insights into the molecular mechanisms of lipolysis mediated by IMD signaling pathway.

Hedgehog (Hh) signaling has been shown to regulate multiple developmental processes, however, it is unclear how it regulates lipid metabolism. Here, we demonstrate that Hh signaling exhibits potent activity in Drosophila fat body, which is induced by both locally expressed and midgut-derived Hh proteins. Inactivation of Hh signaling increases, whereas activation of Hh signaling decreases lipid accumulation. The major lipase Brummer (Bmm) acts downstream of Smoothened (Smo) in Hh signaling to promote lipolysis, therefore, the breakdown of triacylglycerol (TAG). We identify a critical Ci binding site in bmm promoter that is responsible to mediate Bmm expression induced by Hh signaling. Genomic mutation of the Ci binding site significantly reduces the expression of Bmm and dramatically decreases the responsiveness to Ci overexpression. Together, our findings provide a model for lipolysis to be regulated by Hh signaling, raising the possibility for Hh signaling to be involved in lipid metabolic and/or lipid storage diseases. • Hh signaling is highly activated in Drosophila fat body. • Hh signaling inhibits lipid droplet accumulation. • Hh signaling promotes lipolysis through directly regulating Bmm/ATGL. • Ci upregulates Bmm/ATGL transcription through direct interaction with Bmm promoter. Enterprise Architecture (EA) has gained importance in recent years, mainly for its concept of “alignment” between the strategic and operational levels of organizations. Such alignment occurs when Information Technology (IT) is applied correctly and timely, working in synergy and harmony with strategy and the operation to achieve mutually their own goals and satisfy the organizational needs.Both the strategic and operational levels have standards that help model elements necessary to obtain desired results. In this sense, BMM and BPMN were selected because both have the support of OMG and they are fairly well known for modelling the strategic level and operational level, respectively. In addition, i* modeling goal can be used for reducing the gap between these two standards. This proposal may help both the high-level design of the information system and to the appropriate identification of the business processes that will support it.This paper presents a methodology for aligning strategy and the operation based on standards and heuristics. We have made a classification for elements of the models and, for some specific cases, an extension of the heuristics associated between them. This allows us to propose methodology, which uses above-mentioned standards and combines mappings, transformations and actions to be considered in the alignment process.

Business software systems and applications use business process management concepts to organize and automate processes. Incorporating a business process management system in existing business applications is a pretty complex task and, as well, is very expensive in terms of time and human resources. The article presents a newly conceived Framework for Dynamic Business Applications--a software platform that allows business users (domain experts) to defne at run time all business objects that they will work with, their properties, relationships, and restrictions of their business/domain model. This is very similar to what can be done with business ontologies regarding representation of entities, ideas, and events, along with their properties and relations. The Zingiberales is an ancient monocot order in the angiosperms. It has undergone radiation since the Cretaceous period. However, the relationships among primary lineages (families) are difficult to resolve. Additional support for phylogenetic relationships within this order have been obtained using plastid genome data from 76 coding genes and 4 rDNA loci from 17 species represented eight families. We reconstructed ancestral geographic areas using the Bayesian Binary Markov Chain Monte Carlo (BMM) method. Sparsity-based unmixing algorithms, exploiting the sparseness property of the abundances, have recently been proposed with promising performances. However, these algorithms are developed for the linear mixture model (LMM), which cannot effectively handle the nonlinear effects. In this paper, we extend the current sparse regression methods for the LMM to bilinear mixture models (BMMs), where the BMMs introduce additional bilinear terms in the LMM in order to model second-order photon scattering effects. To solve the abundance estimation problem for the BMMs, we propose to perform a sparsity-based abundance estimation by using two dictionaries: a linear dictionary containing all the pure endmembers and a bilinear dictionary consisting of all the possible second-order endmember interaction components. Then, the abundance values can be estimated from the sparse codes associated with the linear dictionary. Moreover, to exploit the spatial data structure where the adjacent pixels are usually homogeneous and are often mixtures of the same materials, we first employ the joint-sparsity (row-sparsity) model to enforce structured sparsity on the abundance coefficients. However, the joint-sparsity model is often a strict assumption, which might cause some aliasing artifacts for the pixels that lie on the boundaries of different materials. To deal with this problem, the low-rank-representation model, which seeks the lowest rank representation of the data, is further introduced to better capture the spatial data structure. Recently, we have cloned several Kazal-type serine protease inhibitors from the midgut of the Triatoma infestans bug. A single gene composed of multi Kazal-type domains, in tandem, encodes these inhibitors. In this work, we describe the purification and characterization of recombinant infestins 3-4 and 4, which are potent factor XIIa inhibitors (Ki=67 pM and 128 pM, respectively). We also identified the first native factor XIIa inhibitor from a hematophagous insect. The factor XIIa inhibitory activity of infestin 4 demonstrates extremely efficient anticoagulant activity, prolonging activated partial thromboplastin time by approximately 3 times. Our results suggest that infestins perform a very important role in the T. infestans midgut during meal acquisition and digestion by controlling blood coagulation by means of inhibiting thrombin and factor XIIa. In this study we examined the structure and microparticle removability of 2 types of regenerated cellulose hollow fiber filters (BMMR and its modified type, BMMU) by electron microscopy using colloidal gold solutions and viruses (HIV and JEV). The following results were obtained: 1) The meshwork structure of the BMMU wall was more suitable for filtration of microparticles than that of the BMMR. 2) Both the BMMR and the BMMU demonstrated excellent microparticle removability in gold filtrations, but the removability was higher in the BMMU than in the BMMR. 3) HIV (100–150 nm) was completely removed by the BMMRs and BMMUs when 10–5 × 10 infectious viruses were filtered. 4) In JEV (40–60 nm) filtration, however, the removability was higher in the BMMU than in the BMMR and the virus concentration was reduced to less than 10 by the BMMU. These results suggest that the BMMs, particularly the BMMU, are highly useful in eliminating microparticles, including viruses. [ABSTRACT FROM PUBLISHER]

Macrophage migration inhibitory factor (MIF) is expressed and released ubiquitously by numerous cell types and tissues. MIF is detected and constitutively expressed at the protein level both intra- and extracellularly. This chapter outlines methods for cultivating, purifying, detecting, and quantifying concentrations of MIF from murine primary derived macrophages and dendritic cell culture supernatants.

Phenotyping cells by flow cytometry is a powerful way to identify cell type and any morphological changes during cell culture. The staining procedure used in this chapter enables the characterization of mouse macrophages by a flow cytometry antibody panel which can be used for both bone marrow-derived macrophages (BMM) and macrophages derived from other tissues, such as the mouse spleen or peritoneal cavity. The surface and intracellular staining methods are versatile and can be applied to flow cytometry staining of several different cell types by changing the surface markers used with knowledge of which receptors are expressed on different cell types.

Metabolic and innate immune signaling pathways have co-evolved to elicit coordinated responses. However, dissecting the integration of these ancient signaling mechanisms remains a challenge. Using Drosophila , we uncovered a role for the innate immune transcription factor nuclear factor κB (NF-κB)/Relish in governing lipid metabolism during metabolic adaptation to fasting. We found that Relish is required to restrain fasting-induced lipolysis, and thus conserve cellular triglyceride levels during metabolic adaptation, through specific repression of ATGL/Brummer lipase gene expression in adipose (fat body). Fasting-induced changes in Brummer expression and, consequently, triglyceride metabolism are adjusted by Relish-dependent attenuation of Foxo transcriptional activation function, a critical metabolic transcription factor. Relish limits Foxo function by influencing fasting-dependent histone deacetylation and subsequent chromatin modifications within the Bmm locus.
