Aflatoxins, produced by aflatoxigenic Aspergillus section Flavi fungi, commonly occur in groundnut and maize grown in Mozambique and have long been associated with high prevalence of liver cancer, stunting, and restricted access to lucrative international markets. Effective aflatoxin control options in the country are limited and not adequately explored. Biocontrol products based on atoxigenic strains of A. flavus provide viable aflatoxin mitigation measures but require development for Mozambique. Four hundred and sixty-eight (468) and 558 groundnut and maize farmers, respectively, voluntarily evaluated the effectiveness of two biocontrol products (Aflasafe MWMZ01 and Aflasafe MZ02), each containing as active ingredients four distinct atoxigenic isolates of A. flavus belonging to native vegetative compatibility groups (VCGs), at preventing aflatoxin contamination and displacement of aflatoxigenic fungi for 2 years in various agro-ecologies. Most groundnut and maize treated with the biocontrol products were below maximum levels for food in the European Union (EU; 85%; p < 0.01) and the United States (US; 99%; p < 0.01). In contrast, most non-treated maize and groundnut (ranging from 38 to 70%; p = 0.05) were above the EU and US maximum allowable levels for food. Aflatoxin reductions ranged from 78 to 98% (p < 0.01) in treated groundnut, and from 61 to 93% (p < 0.01) in treated maize. Toxigenic fungi were almost completely displaced from soils and crops by the applied atoxigenic active ingredients. This study revealed that the atoxigenic based biocontrol technology is effective in Mozambique at displacing aflatoxigenic fungi and reducing aflatoxin accumulation in both groundnut and maize but a combination with other management tools is encouraged for better retention of crop quality along the value chain. The advancement of fungal biocontrol agents depends on replacing cereal grains with low-cost agro-industrial byproducts for their economical mass production and development of stable formulations. We propose an innovative approach to develop a rice flour-based formulation of the beneficial biocontrol agent Trichoderma asperelloides CMAA1584 designed to simulate a micro-bioreactor within the concept of full biorefinery process, affording in situ conidiation, extended shelf-life, and effective control of Sclerotinia sclerotiorum, a devastating pathogen of several dicot agricultural crops worldwide. Rice flour is an inexpensive and underexplored byproduct derived from broken rice after milling, capable of sustaining high yields of conidial production through our optimized fermentation-formulation route. Conidial yield was mainly influenced by nitrogen content (0.1% w/w) added to the rice meal coupled with the fermentor type. Hydrolyzed yeast was the best nitrogen source yielding 2.6 × 109 colony-forming units (CFU)/g within 14 days. Subsequently, GControl, GLecithin, GBreak-Thru, GBentonite, and GOrganic compost+Break-Thru formulations were obtained by extrusion followed by air-drying and further assessed for their potential to induce secondary sporulation in situ, storage stability, and efficacy against Sclerotinia. GControl, GBreak-Thru, GBentonite, and GOrganic compost+Break-Thru stood out with the highest number of CFU after sporulation upon re-hydration on water-agar medium. Shelf-life of formulations GControl and GBentonite remained consistent for > 3 months at ambient temperature, while in GBentonite and GOrganic compost+Break-Thru formulations remained viable for 24 months during refrigerated storage. Formulations exhibited similar efficacy in suppressing the myceliogenic germination of Sclerotinia irrespective of their concentration tested (5 × 104 to 5 × 106 CFU/g of soil), resulting in 79.2 to 93.7% relative inhibition. Noteworthily, all 24-month-old formulations kept under cold storage successfully suppressed sclerotia. This work provides an environmentally friendly bioprocess method using rice flour as the main feedstock to develop waste-free granular formulations of Trichoderma conidia that are effective in suppressing Sclerotinia while also improving biopesticide shelf-life. The use of arbuscular mycorrhizal fungi (AMF) offers promising benefits to agriculture in the Amazon regions, where soils are characteristically acidic and nutrient-poor. The purpose of this research was to investigate the potential effects of two recently described species of AMF (Nanoglomus plukenetiae and Rhizoglomus variabile) native to the Peruvian Amazon for improving the plant growth of Plukenetia volubilis (inka nut or sacha inchi) and protecting the roots against soil pathogens. Two assays were simultaneously conducted under greenhouse conditions in Peru. The first focused on evaluating the biofertilizer effect of AMF inoculation, while the second examined the bioprotective effect against the root knot nematode, Meloidogyne incognita. Overall, the results showed that AMF inoculation of P. volubilis seedlings positively improved their development, particularly their biomass, height, and the leaf nutrient contents. When seedlings were exposed to M. incognita, plant growth was also noticeably higher for AMF-inoculated plants than those without AMF inoculation. Nematode reproduction was significantly suppressed by the presence of AMF, in particular R. variabile, and especially when inoculated prior to nematode exposure. The dual AMF inoculation did not necessarily lead to improved crop growth but notably improved P and K leaf contents. The findings provide strong justification for the development of products based on AMF as agro-inputs to catalyze nutrient use and uptake and protect crops against pests and diseases, especially those that are locally adapted to local crops and cropping conditions. In sub-Saharan Africa (SSA), many crops are contaminated by Aspergillus section Flavi fungi with highly toxic, carcinogenic aflatoxins. This contamination has severe negative impacts on health, trade, income, and development sectors, hindering progress toward various objectives of most Sustainable Development Goals (SDGs), including SDG 2 – Zero Hunger and SDG 3 – Good Health and Wellbeing. Farmers, industries, and governments need sound aflatoxin management strategies to effectively limit aflatoxin contamination throughout the crop value chain. One effective technology is biocontrol using native atoxigenic isolates of A. flavus coated on a carrier that is applied on growing crops. Atoxigenic A. flavus competitively displaces aflatoxin producers in the field, and this form of bioprotection results in reduced aflatoxin in crops. Over 15 years ago, field tests in Nigeria using a manually manufactured biocontrol product showed promising results. However, it became evident that scaling up the manufacturing process was essential to make this bioprotectant widely accessible to millions of farmers and achieve tangible impact in the context of sustainable food systems. The objective of this paper is to document the evolution of biocontrol manufacturing from small-scale, lab-based production to industrial manufacturing at different scales. The improvements in product formulation, and manufacturing processes and design are highlighted to better address “fit-to-scale” product demand in different countries. Industrializing the manufacturing process coupled with incentivization, commercialization strategies, and effective partnerships has allowed the manufacturing of thousands of tons of biocontrol products. This has enabled production of over a million tons of aflatoxin-safe maize, groundnut, and sorghum, contributing to enhanced food safety and security in several countries in SSA. The key lesson learnt is that for any input-based technology to mitigate aflatoxin (or any food safety issue), their large-scale manufacturing and commercialization is crucial for achieving tangible results. Only widespread adoption of any technology can address the great challenge posed by aflatoxins, a critical step toward meeting several SDGs. The urgency to combat aflatoxin contamination is increasing as its impacts are intensifying in several regions across the globe.

This community service program aims to enhance the understanding of Vocational High School (SMK) students in sustainable agriculture, particularly in producing bacterial consortiums of Biofertilizer and Bioprotectant. Situation analysis reveals that SMK students face challenges in accessing training, limited resources and facilities, and a lack of awareness regarding the importance of sustainable farming practices. The program presents strategic solutions, beginning with developing a comprehensive learning module on sustainable agriculture and utilizing bacterial consortiums, biofertilizers, and bioprotectants. This module is designed to assist students in comprehending the concepts, benefits, and practical applications of sustainable farming. The solution also includes practical field training, especially in the propagation of bacterial consortiums, utilizing IoT temperature sensor DS18B20 to monitor the propagation process. The anticipated outcome of this program is an improvement in sustainable agricultural literacy among SMK students. The expectation is that they will better understand the concepts of sustainable agriculture and the implementation of bacterial consortiums, biofertilizers, and bioprotectants. Additionally, students are expected to acquire practical skills in the propagation of bacterial consortiums. Thus, the program aims to positively contribute to developing students' skills and encourage active participation in sustainable agriculture.

Soybean (Glycine max (L.) Merril) is a food and industrial crop and a base for agricultural production. Adding organic material can increase plant growth and produce high yields. One of the organic materials that can be used is palm oil mill solid waste, specifically solid decanter cake (DC). However, the results showed that DC could only substitute 50 per cent of chemical fertilisers. DC must be combined with other materials, specifically Plant Growth plant-promoting rhizobacteria (PGPR), to maximise its benefits. PGPR is a group of bacteria that live in the plant rhizosphere and actively function as biofertilisers, biostimulants, and bioprotectants. Combining DC and PGPR is necessary because they have a mutually beneficial relationship, as DC is an organic material that can provide the nutrients PGPR bacteria need. This research will evaluate whether the collaboration of DC and PGPR can increase soybean growth and yield in ultisols using a Randomized Block Design. Combining decanter cake (DC) with a PGPR concentration can enhance soybean growth and yield. Soybean plants treated with 15-ton ha-1 + 50% PGPR DC tend to have the highest number of leaves, plant height, and number of branches, but DC 10-ton, ha-1 + 30% PGPR, has an equivalent effect. The treatment of 10 tons per hectare of decanter cake with 30% PGPR concentration tended to have the highest number of pods and yield.

Natural and environmental-friendly approaches to control pests and diseases in horticultural production systems are showing an increasing trend. Biological alternatives, such as botanical extracts and basic substances, show promise in the reduction of the necessity for conventional plant protectants. In ornamentals, less is known about the usage, behaviour and potential of botanicals and basic substances for plant protection. In two trials, we investigated the effect of a weekly foliar spray of an aqueous extract of nettle (Urtica sp.; dried material 15 g · L–1) on two aphids, Macrosiphum euphorbiae and Aulacorthum solani. Experiments were conducted in the greenhouse with Ranunculus asiaticus as hostplant. The Urtica tea and azadirachtin (NeemAzal-T/S) as a standard used in common horticultural production were compared with the water control. Both previously infested and noninfested plants were built up to examine the effect on aphid population growth and migration. As a result, we showed that both treatments, Urtica tea and azadirachtin, reduce the aphid density significantly in comparison with the water control, although the effect of the Urtica tea was not as substantial as that of azadirachtin. Contrarily, treatments could not prevent aphid migration on previously noninfested plants. The study demonstrates that the basic substance Urtica tea can be suitable for the reduction of aphid pests in ornamental plant production, and thus, reduce the amount of synthetic plant protectants in horticulture.

Nanotechnology is a promising way to enhance the stability of Bacillus thuringiensis (Bt) insecticidal proteins under environmental conditions. In this work, two emulsions were prepared through the Pickering emulsion technique, stabilized by Cu2+-SQDs/S-CN nanocomposites and by GO nanosheets. In addition, a pH-sensitive polymer was incorporated into these emulsions, allowing the Bt protein, Cry1Ab, to be released in an alkaline pH environment, as it occurs in the lepidopteran pests' gut. The effectiveness of these two nanomaterials in protecting Cry1Ab from degradation, and therefore enhancing its pesticidal activity, was assessed by exposing samples of the purified unprotected protein and encapsulated protein to high-intensity UV light and 40°C temperature treatments. The UV treatment results were evaluated using SDS-PAGE analysis and pointed out that Cry1Ab could be structurally protected by the emulsions. The bioassays with first instar larvae of the lepidopteran pest Ostrinia nubilalis confirm the nanomaterial protection to UV and temperature treatments, i.e., decreasing about half the degradation rate and increasing up to 12-fold the residual activity after UV treatment. Our results indicate that encapsulation could be an effective strategy to improve the effectiveness of Cry1Ab under environmental conditions. Plant pathogens and pests can cause significant losses in crop yields, affecting food security and the global economy. Many traditional chemical pesticides are used to combat these organisms. This can lead to the development of pesticide-resistant strains of pathogens/insects and negatively impact the environment. The development of new bioprotectants, which are less harmful to the environment and less likely to lead to pesticide-resistance, appears as a sustainable strategy to increase plant immunity. Natural Rhamnolipids (RL-Nat) are a class of biosurfactants with bioprotectant properties that are produced by an opportunistic human pathogen bacterium. RL-Nat can act as plant resistance inducers against a wide variety of pathogens. Recently, a series of bioinspired synthetic mono-RLs produced by green chemistry were also reported as phytoprotectants. Here, we explored their capacity to generate novel colloidal systems that might be used to encapsulate bioactive hydrophobic compounds to enhance their performance as plant bioprotectants. The synthetic mono-RLs showed good surfactant properties and emulsification power providing stable nanoemulsions capable of acting as bio-carriers with good wettability. Synthetic RLs-stabilized nanoemulsions were more effective than RLs suspensions at inducing plant immunity, without causing deleterious effects. These nanoemulsions were innocuous to native substrate microbiota and beneficial soilborne microbes, making them promising safe bio-carriers for crop protection. [ABSTRACT FROM AUTHOR]

The 21st century has brought new challenges to the agri‐food industry due to population growth, global warming, and greater public awareness of environmental issues. Ensuring global food security for future generations is crucial. However, pests, weeds, and diseases still significantly contribute to crop losses, and the availability of effective conventional synthetic pesticides is decreasing. To address this, new and diverse pest management tools are needed. One pest management tool showing potential for invertebrate pest management is the exploitation of volatile organic compounds (VOCs)—in particular, the compounds 1‐octen‐3‐ol and 3‐octanone. This review aims to explore the extent to which 1‐octen‐3‐ol and 3‐octanone show potential in the future management of invertebrate crop and animal pests. A significant increase in the rate of publication of literature on the use of 1‐octen‐3‐ol and 3‐octanone in crop protection since 2018 is identified by this review, therefore, showing the potential importance of these compounds for use in future pest management. This review also identifies key interactions between naturally occurring biosynthesised 1‐octen‐3‐ol and 3‐octanone, and a range of invertebrate targets. Many of these interactions with key crop pests are sourced from the taxonomic families Lamiaceae, Fabaceae, and Trichomaceae. However, analysis of the practical application of these sources in an integrated pest management programme identifies clear limitations with the use of naturally occurring biosynthesised 1‐octen‐3‐ol and 3‐octanone. Rather, future focus should be placed on the development and exploitation of synthesised nature identical 1‐octen‐3‐ol and 3‐octanone for use as a biopesticide product. Overall, 1‐octen‐3‐ol and 3‐octanone show potential for exploitation in future crop protection, being abundant in source and diversity of invertebrate interactions. However, their use as a naturally occurring biosynthesised chemical is likely not practical for direct implementation in crop protection. Enzymatic transformation in the Nitrogen (N) and phosphorus (P) cycles in soil can only be carried out by microbes. The latest approach in coffee cultivation is to utilize soil microbes to enhance plant growth, also to reduce the attack of the nematode Pratylenchus coffeae on coffee roots. This exploratory study aimed to observe the attack of P. coffeae on coffee tree, and the presence of N-fixing bacteria (NFB) and P- solubilizing microbes (PSM) in the coffee rhizosphere which have the potency to be used as biofertilizer and bioprotectants in coffee plantation. The study was conducted in Arabica and Robusta coffee plantation of PT Perkebunan Kalibendo, East Java; on immature plants (IP) and mature plants (MP). This exploration explained that the IP and MP of arabica and robusta coffee plantations were attacked by P. coffeae. The NFB and PSM were successfully isolated from the rhizosphere of both coffee plantations types. The population of NFB Azotobacter in IP was lower than in MP, but the PSM population in IP rhizosphere was not different from that in MP. Based on morphological and biochemical characterization, three isolates of Azotobacter bacteria, six species of P- solubilizing bacteria and six species of phosphate-solubilizing fungi were obtained. This exploration confirmed that the microbes involved in the N and P cycle colonized the rhizosphere of coffee which was attacked by P. coffeae. Further research is recommended to observe the effectiveness of microbes as biological fertilizers and bioprotectants for coffee plants. [ABSTRACT FROM AUTHOR]

Pineapple (Ananas comosus L. Merr) is a plant that can live well on nutrient-poor lands such as peatlands. However, plant resistance to nutrient-poor conditions is caused by symbiosis with endophytic bacteria capable of plant growth-promoting bacteria (PGPB) through biostimulant and bioprotective mechanisms. This study aims to characterize the potential of endophytic bacteria from pineapple roots from peatlands as biostimulants and bioprotective agents. Thirteen endophytic bacteria from pineapple root were characterized by their ability as biostimulants through phosphate dissolution tests using Pikovskaya media qualitatively and quantitatively, and the production of indole-3-acetic acid (IAA) upon additional 0.1% tryptophan. Furthermore, bacteria as bioprotectors were characterized by qualitative production of chitinase enzyme and antifungal tests against Fusarium proliferatum using dual culture techniques. The results showed the presence of 2 positive bacteria in all tests, namely the ANAP3 and ANAP5 isolates. ANPA3 bacteria show the highest activity in IAA production (26.3 ppm), and the highest antifungal activity in inhibiting F. proliferatum, with an inhibitory index reaching 52.6%. Meanwhile, ANAP5 is an endophytic bacterium with the highest phosphate dissolving activity, with a dissolved phosphate value of 253.5 ppm. This finding shows that some endophytic bacteria from pineapple roots on peatlands can be biostimulants and bioprotectors that can be developed as PGPB. [ABSTRACT FROM AUTHOR]

Fusarium wilt of bananas caused by Fusarium oxysporum f. sp. cubense Tropical Race 4 (Foc TR4) poses the most serious threat to banana production globally. The disease has been managed using chemical fungicides, yet the control levels are still unsatisfactory. This study investigated the antifungal activities of tea tree (Melaleuca alternifolia) essential oil (TTO) and hydrosol (TTH) against Foc TR4 and their bioactive components. The potential of TTO and TTH in inhibiting the growth of Foc TR4 was evaluated in vitro using agar well diffusion and spore germination assays. Compared to the chemical fungicide, TTO effectively suppressed the mycelial growth of Foc TR4 at 69%. Both the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of TTO and TTH were established at 0.2 µg/µL and 50% v/v, respectively, suggesting the fungicidal nature of the plant extracts. The disease control efficacies were also demonstrated by a (p ≤ 0.05) delayed Fusarium wilt symptom development in the susceptible banana plants with reduced LSI dan RDI scores from 70% to around 20–30%. A GC/MS analysis of TTO identified terpinen-4-ol, eucalyptol, and α-terpineol as the major components. In contrast, an LC/MS analysis of TTH identified different compounds, including dihydro-jasmonic acid and methyl ester. Non-Saccharomyces wine yeasts are not only proposed to improve the sensory profile of wine but also for several distinctive promising features. Among them, biocontrol action at different steps of the wine production chain could be a suitable strategy to reduce the use of sulfur dioxide. In this work, the activity of a selected strain of Metschnikowia pulcherrima was evaluated as inoculum in cold clarification with the aim to reduce SO2 and improve the aromatic profile of the wine. Fermentation processes were carried out at the winery level for two consecutive vintages using a pied de cuve as the starter inoculum coming from indigenous Saccharomyces cerevisiae strains. M. pulcherrima revealed an effective bio-protectant action during the pre-fermentative stage even if the timely and appropriate starter inoculum in the two years permitted the effective control of wild yeasts during the fermentation also in the control trials. In general, the main oenological characters did not show differences if compared with an un-inoculated trial, while the inoculum of M. pucherrima in cold clarification determined an enhancement of ethyl hexanoate, isobutanol, acetaldehyde, and geraniol even if they are considered in different amounts for each year. Indeed, the analytical and sensory profiles of wines were also influenced by the vintage and variation pied the cuve population. Nonetheless, the overall results indicated that M. pulcherrima led to biocontrol action and an improvement of the aromatic and sensory profile of the wine. [ABSTRACT FROM AUTHOR]

Microbial crop protection products based on Trichoderma have the ability to display multifunctional roles in plant protection, such as pathogen parasitism, enhance nutrient availability and stimulate plant growth, and these traits can be used to enhance the overall agronomic performance of a variety of crops. In the current study, we explored the multifunctional potential of two indigenous Brazilian strains of Trichoderma (T. asperelloides CMAA 1584 and T. lentiforme CMAA 1585) for their capability of controlling Sclerotinia sclerotiorum, a key plant pathogen of cotton, and for their ability of growth promotion in cotton plants (Gossypium hirsutum). Both strains were able to solubilize mineral phosphorus (CaHPO4), to release volatile organic compounds that impaired the mycelial growth of S. sclerotiorum, and to promote the growth of cotton plants under greenhouse conditions. In dual culture, Trichoderma strains reduced the growth rate and the number of sclerotia formed by S. sclerotiorum. By treating sclerotia with conidial suspensions of these Trichoderma strains, a strong inhibition of the myceliogenic germination was observed, as a result of the marked mycoparasitic activity exerted on the sclerotia. The parasitism over S. sclerotiorum was more effectivewith T. asperelloides CMAA 1584, whilst the biostimulant effects on cotton growth were more pronounced with T. lentiforme CMAA 1585, which also showed a higher capacity of phosphate solubilization. Thus, T. asperelloides CMAA 1584 displays higher efficiency in controlling S. sclerotiorum, while T. lentiforme CMAA 1585 is more suitable as a biostimulant due to its ability to promote growth in cotton plants. Overall, these Trichoderma strains may be used in mixture to provide both pathogen control and promotion of plant growth, and this strategy will support growers in minimizing the use of synthetic fertilizers and fungicides against white mold in cotton crops. Mango (Mangifera indica L) is one of the most important tropical fruits worldwide; however, being a climacteric fruit, its postharvest deterioration is rapid, which implies that the presence of anthracnose caused by Colletotrichum gloesporioides and the long storage periods for its commercialization affect the shelf life of the fruit. The objective of this research was to evaluate the effect of three concentrations of a shellac-based wax and three concentrations of a biological coating (bioprotectant) made with Bacillus spp. and Streptomyces spp. as coatings for the preservation of physicochemical and microbiological quality parameters in Keitt mangoes stored for 35 days at 7 ºC ± 3. Mangoes with 0.5% bioprotectant and 30% wax showed an incidence of 40% up to day 35, which was significantly lower than that reported for the control group (80%). In the physicochemical parameters, the 30% wax preserved firmness, maintained a weight loss percentage of less than 10% during 35 days and presented the lowest pH values and the highest percentages of citric acid as of day 21 of storage, and the 0.5% bioprotectant in the variables of pH, total soluble solids and titratable acidity were similar to those reported in the control group; firmness was not affected during the days of storage. Maize is an important cereal crop which is severely affected by Spodoptera frugiperda. The study aims to identify endophytic bacteria of maize root and leaf apoplastic fluid with bioprotective traits against S. frugiperda and plant growth promoting properties. Among 15 bacterial endophytic isolates, two strains—namely, RAF5 and LAF5—were selected and identified as Alcaligenes sp. MZ895490 and Bacillus amyloliquefaciens MZ895491, respectively. The bioprotective potential of B. amyloliquefaciens was evaluated through bioassays. In a no-choice bioassay, second instar larvae of S. frugiperda fed on B. amyloliquefaciens treated leaves (B+) recorded comparatively lesser growth (1.10 ± 0.19 mg mg−1 day−1) and consumptive (7.16 ± 3.48 mg mg−1 day−1) rates. In larval dip and choice bioassay, the same trend was observed. In detached leaf experiment, leaf feeding deterrence of S. frugiperda was found to be greater due to inoculation with B. amyloliquefaciens than Alcaligenes sp. The phenolics content of B. amyloliquefaciens inoculated plant was also found to be greater (3.06 ± 0.09 mg gallic acid g−1). However, plant biomass production was more in Alcaligenes sp inoculated treatment. Fusarium wilt of bananas caused by Fusarium oxysporum f. sp. cubense Tropical Race 4 (Foc TR4) poses the most serious threat to banana production globally. The disease has been managed using chemical fungicides, yet the control levels are still unsatisfactory. This study investigated the antifungal activities of tea tree (Melaleuca alternifolia) essential oil (TTO) and hydrosol (TTH) against Foc TR4 and their bioactive components. The potential of TTO and TTH in inhibiting the growth of Foc TR4 was evaluated in vitro using agar well diffusion and spore germination assays. Compared to the chemical fungicide, TTO effectively suppressed the mycelial growth of Foc TR4 at 69%. Both the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) of TTO and TTH were established at 0.2 µg/µL and 50% v/v, respectively, suggesting the fungicidal nature of the plant extracts. The disease control efficacies were also demonstrated by a (p ≤ 0.05) delayed Fusarium wilt symptom development in the susceptible banana plants with reduced LSI dan RDI scores from 70% to around 20–30%. A GC/MS analysis of TTO identified terpinen-4-ol, eucalyptol, and α-terpineol as the major components. In contrast, an LC/MS analysis of TTH identified different compounds, including dihydro-jasmonic acid and methyl ester. Our findings indicate the potential of tea tree extracts as natural alternatives to chemical fungicides to control Foc TR4.

Fungal spoilage led to a considerable economic loss of foodstuff which ultimately affects public health due to mycotoxins production. Moreover, the consumption of commercial antifungal drugs creates side effects and develops antifungal resistance. To overcome these challenges, the current work was aimed to investigate novel antifungal cyclic dipeptide (CDP) from Lactobacillus coryniformis (Loigolactobacillus coryniformis) BCH-4. CDPs have flexible, cyclic, and stable conformation. The proline-based CDPs provide additional structural compatibility and bio-functional values. Keeping in view, high-performance liquid chromatography (HPLC) was performed to explore cyclo(L-Leu-L-Pro) from L. coryniformis BCH-4. The HPLC detected concentration (135 ± 7.07 mg/mL) exhibited in vitro antifungal activity of 5.66 ± 0.57 mm (inhibitory zone) against Aspergillus flavus. Based on these results, cyclo(L-Leu-L-Pro) was used as a bioprotectant for selected food samples (grapes, lemon, cashew nuts, and almonds). A significant impact of cyclo(L-Leu-L-Pro) was observed in contrast with MRS broth (control) and cell-free supernatant. In silico molecular docking analysis of this CDP was carried out against FAD glucose dehydrogenase, dihydrofolate reductase, and urate oxidase of A. flavus as target proteins. Among these proteins, FAD glucose dehydrogenase exerted strong interactions with cyclo(L-Leu-L-Pro) having S-score of – 8.21. The results evaluated that the detected CDP has strong interactions with selected proteins, causing excellent growth inhibition of A. flavus. Non-Saccharomyces wine yeasts are not only proposed to improve the sensory profile of wine but also for several distinctive promising features. Among them, biocontrol action at different steps of the wine production chain could be a suitable strategy to reduce the use of sulfur dioxide. In this work, the activity of a selected strain of Metschnikowia pulcherrima was evaluated as inoculum in cold clarification with the aim to reduce SO2 and improve the aromatic profile of the wine. Fermentation processes were carried out at the winery level for two consecutive vintages using a pied de cuve as the starter inoculum coming from indigenous Saccharomyces cerevisiae strains. M. pulcherrima revealed an effective bio-protectant action during the pre-fermentative stage even if the timely and appropriate starter inoculum in the two years permitted the effective control of wild yeasts during the fermentation also in the control trials. In general, the main oenological characters did not show differences if compared with an un-inoculated trial, while the inoculum of M. pucherrima in cold clarification determined an enhancement of ethyl hexanoate, isobutanol, acetaldehyde, and geraniol even if they are considered in different amounts for each year. Indeed, the analytical and sensory profiles of wines were also influenced by the vintage and variation pied the cuve population. Nonetheless, the overall results indicated that M. pulcherrima led to biocontrol action and an improvement of the aromatic and sensory profile of the wine.

Methanol, a by-product associated with plant metabolism, is a substrate for pink pigmented facultative methylotrophs (PPFMs) of phyllosphere. The symbiotic interaction of PPFMs has many desirable effects on plant growth and disease resistance. The present study investigated the potential of native PPFMs for mitigating biotic stress and plant growth promotion in ginger. PPFMs were isolated from ginger phyllosphere by leaf imprint technique and screened against major fungal phytopathogens of ginger viz. Macrophomina phaseolina, Sclerotium rolfsii, Pythium myriotylum, Colletotrichum gloeosporioides and Fusarium oxysporum. Among the 60 PPFMs, IISRGPPFM13 was selected for its highly inhibitory activity against the target pathogens. The isolate was useful for mineral solubility, production of IAA, siderophores and hydrolytic enzymes like cellulase, pectinase, lipase, amylase and chitinase. On in planta experiments revealed that IISRGPPFM13 considerably increased plant growth parameters when the bacterium was applied as soil drenching cum foliar spraying. Methanol utilization potential of the isolate was confirmed by mxaF gene analysis where the sequence showing 95.51% identity towards Methylobacterium platani and M. iners. Further, 16S rRNA gene sequence showing 98.73% identity with M. komagatae 002-079 T (AB252201). This is the first report of its kind that a genus of Methylobacterium with biostimulant potential isolated from the phyllosphere of ginger. [ABSTRACT FROM AUTHOR]

Biostimulants and bioprotectants are derived from natural sources and can enhance crop growth and protect crops from pests and pathogens, respectively. They have attracted much attention in the past few decades and contribute to a more sustainable and eco-friendly agricultural system. Despite not having been explored extensively, plant extracts and their component secondary metabolites, including phenolic compounds have been shown to have biostimulant effects on plants, including enhancement of growth attributes and yield, as well as bioprotectant effects, including antimicrobial, insecticidal, herbicidal and nematicidal effects. Medicinal and aromatic plants are widely distributed all over the world and are abundant sources of phenolic compounds. This paper reviews the characterisation of phenolic compounds and extracts from medicinal and aromatic plants, including a brief overview of their extraction, phytochemical screening and methods of analysis. The second part of the review highlights the potential for use of phenolic compounds and extracts as biostimulants and bioprotectants in agriculture as well as some of the challenges related to their use.

Plant bioactive compounds provide novel straightforward approaches to control plant diseases. Rosemary (Salvia rosmarinus)-derived extracts carry many prominent pharmacological activities, including antimicrobial and antioxidant, mainly due to its phenolic compounds, rosmarinic acid (RA), carnosic acid and carnosol. However, the effects of these extracts on plant diseases are still unknown, which constrains its potential application as bioprotectant in the agricultural production. In this study we demonstrate the antiviral effect of the aqueous rosemary extract (ARE) against tobacco necrosis virus strain A (TNVA) in ARE-treated tobacco (Nicotiana tabacum) plants. Our results show that ARE-treatment enhances plant defense response, contributing to reduce virus replication and systemic movement in tobacco plants. RA, the main phenolic compound detected in this extract, is one of the main inducers of TNVA control. The ARE-induced protection in TNVA-infected plants was characterized by the expression of H 2 O 2 scavengers and defense-related genes, involving salicylic acid- and jasmonic acid-regulated pathways. Furthermore, treatment with ARE in lemon (Citrus limon) and soybean (Glycine max) leaves protects the plants against Xanthomonas citri subsp. citri and Diaporthe phaseolorum var. meridionalis , respectively. Additionally, ARE treatment also promotes growth and development, suggesting a biostimulant activity in soybean. • Garlic-based compounds have strong antibacterial, antifungal and, allelopathic characteristics. • Garlic extract has shown to improve the crop quality and soil conditions under biotic and abiotic stress conditions. • Vegetable seeds primed with garlic extract revealed noticeable differences in germination indices and seedling growth (particularly root growth and fresh weights) in a dose-dependent manner. • Garlic allelochemicals also act as a plant biostimulant to enhance auxin biosynthesis and its transportation, resulting in a promoted root growth. During the last century, advances in recognizing plant-derived chemicals have led to potential biological substances of significant implications in medicine and agriculture. Identification of garlic-derived allicin has since proved the potential antimicrobial role of garlic against a myriad of microbes. Similarly, during the past few decades, attention is given to the allelopathic roles of garlic and understand the mechanism underlying the bioactivity of garlic-derived compounds in the fields of crop production and protection. In this review, we try to assess the potential role of garlic as medicinal and allelopathic crop and compile the studies that concluded the biological functions of garlic in various fields of researches, in particular, the roles in agriculture or plant development. Attention is drawn to clarify the role of garlic-based compounds as antimicrobial, allelopathic and, general human health. Based on the literatures, will discuss about the futuristic possibility of garlic extracts as plant bioprotectant to increase the production and ensure crop safety against phytopathogens. The review thereby encompasses these issues: 1. The biostimulants, and their role in agriculture. 2. The medicinal importance of garlic. 3. The allelopathic influence of garlic. 4. Chemical composition of garlic, and 5. The hypothesized use of garlic extracts as next generation biostimulant. A total of 70 bacteria were isolated from the rhizosphere of potato cv. Hartapel that grew at an altitude of 700 m above sea level on the island of Buru-Maluku. Of these isolates, 36 isolates were capable of producing IAA, GA, Siderophore and phosphate solubilization. Among the selective isolates, isolate HB8 produced the highest amount of IAA (5.816 mg l -1 ), while isolate HB32 produced the highest amount of GA (6.879 mg l -1 ). Isolate HB18 had the best ability in producing salicylate type siderophore (4.214 mg l -1 ) and isolate HB3 showed the highest phosphate solubilization (14.237 mg l -1 ). There were three isolates (HB3, HB8 and HB31) positively produced HCN. All 36 isolate showed physiological characters which revealed the potential use for biostimulant, biofertilizer and bioprotectant against soil borne pathogens. [ABSTRACT FROM AUTHOR]

Abstract: Neutron scattering techniques represent a powerful tool for characterizing both the structure and dynamical properties of bio-systems, for example, proteins and membranes interacting with their solvents. In this paper, Elastic Neutron Scattering (ENS) data collected at the Institut Laue-Langevin (Grenoble, France) on dry and D2O hydrated lysozyme by varying hydration level are presented, and compared with previously published data on the same protein system, also with the addition of bio-protectants. The data have been collected with three different spectrometers, i.e. IN13, IN10 and IN4. This set of ENS data gives direct access to the temperature behavior of both (i) the Mean Square Displacement (MSD) and (ii) the characteristic system relaxation time. As a result, an explicative hypothesis on the relationship between the so-called “protein dynamical transition” (PDT) and the “fragile-to-strong dynamical crossover” (FSC) is formulated. Furthermore, by taking into proper account the effect of the finite instrumental energy resolution of the used spectrometers, the vibrational MSD of dry and hydrated lysozyme is calculated. The vibrational MSD of the lysozyme in the dry state resulted to be higher than the one in the hydrated state; the latter reaches the former at a temperature value of T=220K that corresponds to the temperature at which the FSC occurs. As a result, a cage effect resulting from the hydration water on the protein surface is hypothesized and subsequently linked to the FSC. [Copyright &y& Elsevier]

Future space missions will likely include plants to provide fresh foods and bioregenerative life support capabilities. Current spacecraft such as the International Space Station (ISS) operate at 1 atm (101 kPa) pressure, but future missions will likely use reduced pressures to minimize gas leakage and facilitate rapid egress (space walks). Plants for these missions must be able to tolerate and grow reliably at these reduced pressures. We grew two lettuce cultivars, 'Flandria' a green bibb-type and 'Outredgeous,' a red, loose-leaf type, under three pressures: 96 kPa (ambient control), 67 kPa (2/3 atm), and 33 kPa (1/3 atm) for 21 days in rockwool using recirculating nutrient film technique hydroponics. Each treatment was repeated three times using a different hypobaric chamber each time. A daily light integral of 17.2 Moles Photosynthetically Active Radiation per day was provided with metal halide lamps set to deliver 300 µmol m -2 s  -1 photosynthetic photon flux (PPF) for a 16 h photoperiod at 22 °C. Oxygen was maintained at 21 kPa (equal to 21% at 1 atm) and CO 2 at 0.12 kPa (equal to 1200 ppm at 1 atm). Leaf area for 'Outredgeous' was reduced 20% and 38% at 67 kPa and 33 kPa respectively; shoot fresh mass was reduced 22% and 41% at 67 kPa and 33 kPa respectively when compared to control plants at 96 kPa. These trends were not statistically significant at P ≥ 0.05. Leaf area for 'Flandria' showed no difference between 96 and 67 kPa but was reduced 31% at 33 kPa; shoot fresh mass was reduced 6% and 27% at 66 kPa and 33 kPa respectively compared to 96 kPa. There were 10% and 25% increases in anthocyanin concentration at 66 kPa and 33 kPa compared to 96 kPa, potentially increasing the bioprotective capacity of the plant. Previous studies with other cultivars of lettuce showed slight change in growth across this range of pressures, suggesting responses may vary among genotypes, hypobaric exposure treatments, and / or environmental conditions. New Inelastic Neutron Scattering findings on homologues disaccharides (C12H22O11)/H2O mixtures are presented. The comparison among the spectra of trehalose, maltose and sucrose/H2O mixtures, besides evidencing a different destructuring effectiveness on the H2O hydrogen bond network, and hence different cryoprotectant properties, shows a higher ‘crystallinity’ degree for the trehalose/H2O system which accounts for its higher ‘rigidity’.
