Candida parapsilosis is known to cause severe and persistent outbreaks in clinical settings. Patients infected with multidrug-resistant C. parapsilosis (MDR Cp) isolates were identified in a large Turkish hospital from 2017-2020. We subsequently identified three additional patients infected with MDR Cp isolates in 2022 from the same hospital and two echinocandin-resistant (ECR) isolates from a single patient in another hospital. The increasing number of MDR and ECR isolates contradicts the general principle that the severe fitness cost associated with these phenotypes could prevent their dominance in clinical settings. Here, we employed a multidimensional approach to systematically assess the fitness costs of MDR and ECR C. parapsilosis isolates. Whole-genome sequencing revealed a novel MDR genotype infecting two patients in 2022. Despite severe in vitro defects, the levels and tolerances of the biofilms of our ECR and MDR isolates were generally comparable to those of susceptible wild-type isolates. Surprisingly, the MDR and ECR isolates showed major alterations in their cell wall components, and some of the MDR isolates consistently displayed increased tolerance to the fungicidal activities of primary human neutrophils and were more immunoevasive during exposure to primary human macrophages. Our systemic infection mouse model showed that MDR and ECR C. parapsilosis isolates had comparable fungal burden in most organs relative to susceptible isolates. Overall, we observed a notable increase in the genotypic diversity and frequency of MDR isolates and identified MDR and ECR isolates potentially capable of causing persistent outbreaks in the future.

The aerobic hyperthermophile "Fervidibacter sacchari" catabolizes diverse polysaccharides and is the only cultivated member of the class "Fervidibacteria" within the phylum Armatimonadota. It encodes 117 putative glycoside hydrolases (GHs), including two from GH family 50 (GH50). In this study, we expressed, purified, and functionally characterized one of these GH50 enzymes, Fsa16295Glu. We show that Fsa16295Glu is a β-1,3-endoglucanase with optimal activity on carboxymethyl curdlan (CM-curdlan) and only weak agarase activity, despite most GH50 enzymes being described as β-agarases. The purified enzyme has a wide temperature range of 4-95°C (optimal 80°C), making it the first characterized hyperthermophilic representative of GH50. The enzyme is also active at a broad pH range of at least 5.5-11 (optimal 6.5-10). Fsa16295Glu possesses a relatively high kcat/KM of 1.82 × 107 s-1 M-1 with CM-curdlan and degrades CM-curdlan nearly completely to sugar monomers, indicating preferential hydrolysis of glucans containing β-1,3 linkages. Finally, a phylogenetic analysis of Fsa16295Glu and all other GH50 enzymes revealed that Fsa16295Glu is distant from other characterized enzymes but phylogenetically related to enzymes from thermophilic archaea that were likely acquired horizontally from "Fervidibacteria." Given its functional and phylogenetic novelty, we propose that Fsa16295Glu represents a new enzyme subfamily, GH50_3.

Beta-glucan (BG), a polysaccharide comprised of interfacing glucose monomers joined via beta-glycosidic linkages, can be defined as a type of dietary fiber with high specificity based on its interaction with the gut microbiota. It can induce similar interindividual microbiota responses, thereby having beneficial effects on the human body. In this paper, we review the four main sources of BG (cereals, fungi, algae, and bacteria) and their differences in structure and content. The interaction of BG with gut microbiota and the resulting health effects have been highlighted, including immune enhancement, regulation of serum cholesterol and insulin levels, alleviation of obesity and improvement of cognitive disorders. Finally, the application of BG in food products and its beneficial effects on the gut microbiota of consumers were discussed. Although some of the mechanisms of action remain unclear, revealing the beneficial functions of BG from the perspective of gut microbiota can help provide theoretical support for the development of diets that target the regulation of microbiota. As dietary needs shift with the growing and aging population, there is a demand for food products that meet nutritional, safety, and tribological requirements while being cost-effective. Seniors must be given significant consideration in new product development. This study examines consumer preferences for arabinogalactan (AG) and beta-glucan (BG) hydrogels with vanilla and coffee-biscuit flavors, using consumer tests (N = 80) and an online survey (N = 852). It focuses on the gels' physical properties, such as texture and viscosity, and their impact on sensory perception. The use of two different gel-forming polysaccharides, each with a unique sensory profile, was observed to affect the sensory properties of the resulting gels and subsequently influence product acceptance. This study analyzed preferences across three age groups: young (18–39 years), middle-aged (40–59 years), and older adults (60+ years). The results showed that seniors preferred AG-based gels. Significant attributes such as the intensity of flavor and bitter taste influenced the overall liking of the gels. Texture also notably impacted preferences. The survey findings revealed statistically significant (p < 0.05) differences in preferences between older adults and younger age groups. Tailoring product development and marketing strategies based on age and sensory preferences could enhance consumer acceptance of edible gels. [ABSTRACT FROM AUTHOR]

Objective: The present review article focuses on exploring the wide dimensions of barley including botanical description, nutritional composition, phytochemistry, experimental pharmacology, and clinical studies along with its traditional uses in Unani medicine. Background: Barley (Hordeum vulgare L.) which is referred to as 'Jao' in the Unani system of medicine has a long history of utilization as a food and medicine. It belongs to family Poaceae and is considered as one of the founder crops of old-world agriculture. Methodology: A manual literature survey of classical Unani texts was conducted to collect the information available on 'Jao'. Besides, a comprehensive search of electronic databases like PubMed, Google Scholar, and Science Direct, was carried out to collect all the available information regarding its phytochemical, pharmacological and clinical studies. Result: In Unani medicine, barley is an important drug commonly used in many disorders of the respiratory system, gastrointestinal system, musculoskeletal system, skin, etc. It is used to treat obesity, diabetes, headache, pain and inflammation of the throat, acute swelling of joints and gout, chronic swellings, diarrhoea, fever, skin eruptions, hyperpigmentation of the skin, etc. Recent studies revealed that barley constitutes beta-glucan and a myriad of bioactive compounds that play numerous biological activities such as hypoglycemic, hypocholesterolemic, anti-inflammatory, antioxidant, anti-obesity, anticancer, and wound healing activities. Conclusion: Barley is a potent drug that exhibits a wide range of health benefits, most of which have been confirmed via preclinical and clinical studies. Phallus atrovolvatus, a wild edible mushroom, has attracted increasing interest for consumption due to its unique taste and beneficial health benefits. This study determined the chemical components in the so-called fruiting body during the egg and mature stages and investigated its gut microbiota-modulating activities. The egg stage contained higher total carbohydrates, dietary fiber, glucans, ash, and fat, while the total protein content was lower than in the mature stage. Two consumption forms, including cooked mushrooms and a mushroom aqueous extract from both stages, were used in this study. An in vitro gut fermentation was performed for 24 h to assess gut microbiota regulation. All mushroom-supplemented fermentations increased short-chain fatty acid (SCFA) production compared to the blank control. Furthermore, all mushroom supplementations promoted the growth of Bifidobacterium and Streptococcus. Samples from the mature stage increased the relative abundance of Clostridium sensu stricto 1, while those from the egg stage increased the Bacteroides group. The inhibition of harmful bacteria, including Escherichia-Shigella, Klebsiella, and Veillonella, was only observed for the mature body. Our findings demonstrate that P. atrovolvatus exhibits potential benefits on gut health by promoting SCFA production and the growth of beneficial bacteria, with the mature stage demonstrating superior effects compared to the egg stage. Pro‐inflammatory fungal β‐d‐glucan (BDG) polysaccharides cause respiratory pathology. However, specific immunological effects of unique BDG structures on pulmonary inflammation are understudied. We characterized the effect of four unique fungal BDGs with unique branching patterns, solubility, and molecular weights in murine airways. Scleroglucan (1 → 3)(1 → 6)‐highly branched BDG, laminarin (1 → 3)(1 → 6)‐branched BDG, curdlan (1 → 3)‐linear BDG, and pustulan (1 → 6)‐linear BDG were assessed by nuclear magnetic resonance spectroscopy. Each BDG was tested by inhalation model with C3HeB/FeJ mice and compared to saline‐exposed control mice and unexposed sentinels (n = 3–19). Studies were performed ±heat‐inactivation (1 h autoclave) to increase BDG solubility. Outcomes included bronchoalveolar lavage (BAL) differential cell counts (macrophages, neutrophils, lymphocytes, eosinophils), cytokines, serum IgE, and IgG2a (multiplex and ELISA). Ex vivo primary cells removed from lungs and plated at monolayer were stimulated (BDG, lipopolysaccharide (LPS), anti‐CD3), and cytokines compared to unstimulated cells. Right lung histology was performed. Inhalation of BDGs with distinct branching patterns exhibited varying inflammatory potency and immunogenicity. Lichen‐derived (1 → 6)‐linear pustulan was the most pro‐inflammatory BDG, increasing inflammatory infiltrate (BAL), serum IgE and IgG2a, and cytokine production. Primed lung cells responded to secondary LPS stimulation with a T‐cell‐specific response to pustulan. Glucan source and solubility should be considered in exposure and toxicological studies. The carbohydrate ganglioside GD2/GD3 cancer vaccine adjuvanted by β-glucan stimulates anti-GD2 IgG1 antibodies that strongly correlate with improved progression-free survival (PFS) and overall survival (OS) among patients with high-risk neuroblastoma. Thirty-two patients who relapsed on the vaccine (first enrollment) were re-treated on the same vaccine protocol (re-enrollment). Titers during the first enrollment peaked by week 32 at 751 ± 270 ng/mL, which plateaued despite vaccine boosts at 1.2–4.5 month intervals. After a median wash-out interval of 16.1 months from the last vaccine dose during the first enrollment to the first vaccine dose during re-enrollment, the anti-GD2 IgG1 antibody rose to a peak of 4066 ± 813 ng/mL by week 3 following re-enrollment (p < 0.0001 by the Wilcoxon matched-pairs signed-rank test). Yet, these peaks dropped sharply and continually despite repeated boosts at 1.2–4.5 month intervals, before leveling off by week 20 to the first enrollment peak levels. Despite higher antibody titers, patients experienced no pain or neuropathic side effects, which were typically associated with immunotherapy using monoclonal anti-GD2 antibodies. By the Kaplan–Meier method, PFS was estimated to be 51%, and OS was 81%. The association between IgG1 titer during re-enrollment and β-glucan receptor dectin-1 SNP rs3901533 was significant (p = 0.01). Abstract To investigate de effect of PAb gel on the bone tissue of rats submitted to Bisphosphonate-related osteonecrosis of the jaws (BRONJ). Initially, 54 animals were submitted to BRONJ model by Zoledronic Acid (ZA) (0.1 mg/kg 3x/wk for 9 wk, ip), followed by the 1st upper left molar extraction at the 8th wk. After tooth removal, the animals were divided into 3 groups, ZA that received placebo gel or PAb gel that received 1% PAb gel, inside the dental alveolus. The control Group (CONTROL) received 0.1 mg/kg of 0.9% saline and then placebo gel. Three weeks after tooth extraction, the animals were euthanized, and maxillae were colleted for macroscopic, radiographic, histological and Raman spectomery assays. Additionally, GSK3b, beta-catenin, and Runx2 mRNA expressions were determined. Blood samples were collected for the analysis of Bone-specific alkaline phosphatase (BALP) levels. PAb gel improved mucosal healing, increased the number of viable osteocytes, while it reduced the number of empty lacunae, as well as the amount of bone sequestration. Furthermore, PAb gel positively influenced the number and functionality of osteoblasts by stimulating Wnt signaling, thereby inducing bone remodeling. Additionally, PAb gel contributed to improved bone quality, as evidenced by an increase in bone mineral content, a decrease in bone solubility, and an enhancement in the quality of collagen, particularly type I collagen. PAb gel mitigated bone necrosis by stimulating of bone remodeling through Wnt signaling and concurrently improved bone quality. PAb gel emerges as a promising pharmacological tool for aiding in BRONJ therapy or potentially preventing the development of BRONJ.

Abstract Background Beta-(1,3)(1,6)-D-glucan is a complex polysaccharide, which is found in the cell wall of various fungi, yeasts, bacteria, algae, barley, and oats and has immunomodulatory, anticancer and antiviral effects. In the present study, we investigated the effect of beta-(1,3)(1,6)-D-glucan derived from yeast on the proliferation of primary NK cells and breast cancer cell lines in 2D and 3D models, and on the cytotoxicity of primary NK cells against breast cancer cell lines in 2D and 3D models. Methods In this study, we investigated the effects of different concentrations of yeast-derived beta-(1→3)(1→6)-D-glucan on the proliferation and cytotoxicity of human NK cells and breast cancer cell lines in 2D and 3D models using the XTT cell proliferation assay and the CellTiter-Glo® 2.0 assay to determine the cytotoxicity of human NK cells on breast cancer cell lines in 2D and 3D models. Results We found that the co-incubation of NK cells with beta-glucan in the absence of IL2 at 48 h significantly increased the proliferation of NK cells, whereas the co-incubation of NK cells with beta-glucan in the presence of IL2 (70 U/ml) increased the proliferation of NK cells but not significantly. Moreover, beta-glucan significantly inhibited the proliferation of breast cancer cell lines in 2D model and induced a weak, non-significant growth inhibitory effect on breast cancer multicellular tumor spheroids (3D). In addition, the cytotoxicity of NK cells against breast cancer cell lines was examined in 2D and 3D models, and beta-glucan significantly increased the cytotoxicity of NK cells against MCF-7 (in 2D). Conclusions Yeast derived beta-(1,3)(1,6)-D-glucan could contribute to the treatment of cancer by enhancing NK cell immune response as well as contributing to inhibition of breast cancer cell growth.

This review examines the totality of evidence from randomized controlled trials to answer critical questions about the satiety perception of whole-grain oats, oat bran, and isolated beta-glucan. Twenty-two randomized controlled studies were selected following stringent criteria to minimize confounding. Whole-grain oats and oat bran more effectively promoted the perception of satiety than other grain foods that were lower in fiber, higher in refined grains, or lower in viscosity. When oat products were compared with other oat products, those higher in fiber and viscosity generally resulted in increased perception of satiety. Data for isolated beta-glucan were very limited, and no clear conclusions could be drawn. Of the different indices of satiety perception (i.e. hunger, fullness, satiety, desire to eat, prospective food intake), increased fullness was found to be the most positive satiety measurement for whole-grain oat and oat bran products. However, the intake of oat products was not found to consistently reduce short-term energy intake, particularly when limited to a second-meal effect. This may be attributed to the fact that the energy intake for the entire day was typically not measured, as the impact of soluble fiber on energy intake may be delayed. [ABSTRACT FROM AUTHOR]

Nowadays, due to the high application of antibiotics, the emergence of pathogenic microorganisms resistant to antibiotics is considered a serious threat to poultry production industry. Therefore, reducing the use of antibiotics and using antibiotic alternatives is one of the main strategies to fight antibiotic resistance. Meanwhile, yeast-based prebiotics have been proposed as one of the best alternatives to antibiotics. The most important yeast-based prebiotics include hydrolyzed yeast, partially-hydrolyzed yeast, inactive yeast, yeast cell wall, yeast extract and autolyzed yeast. Yeast-based prebiotics contain functional compounds in various concentrations such as B vitamins, beta-carotene, ergosterol, ascorbic acid, polyamines, mycosins, enzymes, beta-glucans and mannan oligosaccharides, depending on the production and processing method. Yeast-based prebiotics are resistant to the acidic and alkaline conditions of the digestive system, and by inactivating pathogenic microorganisms, improving the immune system, stimulating the growth of probiotics, helping to establish beneficial microbial flora in the digestive system, and stimulating the production of volatile fatty acids, reducing intestinal acidity, improving the growth and differentiation of intestinal villi, stimulating the production of enzymes, increasing the production of antimicrobial peptides improve the production efficiency in poultry. The experimental protocol aimed to assess the influence of nucleotides and/or β-glucan on the production performance, growth-related gene expression in the broiler chickens' liver tissue, intestinal histomorphology, and some biochemical parameters. One hundred forty-four newly hatched chicks were categorized into four groups based on the supplements: a control group, a group supplemented with nucleotides (200 mg/kg diet), a group with β-glucan (1 g/kg diet), and a group with both nucleotides and β-glucan. The study's findings showed that, when compared to the control group, all dietary supplemented groups showed a substantial (P < 0.05) improvement in production parameters without having a negative impact on the general health of broilers. Additionally, the treatments positively impacted the expression of some genes associated with growth performance in the broiler chickens' liver tissue, such as the insulin-like growth factor 1 (IGF-1) and growth hormone receptor (GHR). The supplemented groups also demonstrated a significant (P < 0.05) enhancement in intestinal histomorphology, characterized by increased villi length, crypt depth, and goblet cell number. Furthermore, dietary supplementation of nucleotides and/or β-glucan led to a significant (P < 0.05) elevation in total protein and albumin levels, with no significant effect on globulin, AST, and ALT. The conclusion can be drawn that adding nucleotides along with β-glucan to the diet led to enhancements in growth performance, the expression of genes related to growth (GHR and IGF-1), intestinal histomorphology, and certain biochemical parameters (specifically total protein and albumin). Growth promoters in poultry feed have been under severe attention since antibiotics were banned for use in animal diets by the European Union. Thus, it has been important for poultry researchers to find alternatives to antibiotic growth promoters (AGPs) to boost the health and production performance of poultry. This research was conducted to evaluate the effects of adding ALTIMOS® (cell wall of Saccharomyces cerevisiae; mannan oligosaccharides [MOS] + beta-glucan [BG]) to broiler diets on productive performance, blood parameters, intestine histopathology, and cecum microbiota of broiler chicken. A total of 252 one-day-old Ross chicks were randomly selected and divided into seven treatments, with six replicates of each treatment. The treatments were the control group (0% feed additives), and groups that received 0.05, 0.125, 0.250, 0.500, 1.0, and 2.0 g MOS+BG/kg basal diet for 35 days feeding trial. The results showed that during most trial periods, the group fed the basal diet supplemented with 1.0 g MOS+BG/kg had the highest body weight and weight gain, as well as the lowest feed consumption and best feed conversion ratio, compared to other treated groups. Moreover, this group had the best productive performance in the accumulative period. The inclusion of MOS+BG at 1.0 g/kg diet showed no significant effect on carcass percent compared to the control group. In addition, the inclusion of MOS+BG at 1.0 g/kg diet resulted in the lowest count of Escherichia coli and Enterococcus in the cecum, the highest Lactobacillus bacteria count among all experimental treatments, and a higher yeast count compared to the control group. The group fed 1.0 g MOS+BG/kg ration had the lowest blood cholesterol, whereas there were no significant differences among all experimental groups in the measured liver functions. Notably, the Hemoglobin percentage in the group fed MOS+BG at 1.0 g/kg feed was the highest. In the group fed 1.0 and 2.0 g MOS+BG/kg, the intestinal villi length was longer, and the histopathology revealed mild alteration. Overall, the supplementation of 1.0 g MOS+BG/kg diet improved growth performance, blood constituents, and cecum's beneficial bacteria counts of broilers. [ABSTRACT FROM AUTHOR]

Exogenous enzymes as alternatives to feed antibiotics in poultry has become an emerging research area with the emergence of antibiotic resistance. The objective was to evaluate the effects of diet medication (antibiotics) and β-glucanase (BGase) on digesta soluble β-glucan depolymerization, carbohydrate fermentation, and performance of coccidiosis-vaccinated broiler chickens fed wheat-based diets. A total of 1,782 broilers were raised on litter floor pens, and each treatment was assigned to 1 pen in each of the 9 rooms. The 3 dietary treatments were based on wheat as the sole grain (control, control + medication and control + 0.1% BGase), and the birds were fed the respective treatments ad libitum from 0 to 33 d. Treatments were arranged in a randomized complete block design and analyzed as a one-way ANOVA. Tailoring product development and marketing strategies based on age and sensory preferences could enhance consumer acceptance of edible gels.

Introduction: Beta-glucan is one of the most important dietary fibers in cereal grains, having a great impact on human health and food quality. Wheat and barley are strategic crops and their fibers contribute toward their nutraceutical impact. However, the health of the environment is also important to ensure sustainable crop production. Hence the European decision makers have agreed to move toward organic cropping and to reduce the use of mineral fertilizers. Environment and agricultural management have a vast impact on the content of β-glucan in cereal grains. To exploit the valuable properties of β-glucan, the knowledge of factors increasing the amount of this component is crucial. We hypothesized that annual weather conditions, nitrogen fertilization, and cropping system affect the β-glucan amount in wheat and barley. Methods: For this purpose, spring barley and winter wheat flour samples from an 8-year-long period (2014-2021) were characterized. The experiment included conventional and organic cropping systems with different nitrogen rates between 0 and 150 kg ha−1. In addition, the variation of β-glucan content in different varieties was analyzed. Results and discussion: The results showed that the content of β-glucan in barley and wheat grains was affected mainly by weather conditions not by fertilization. The latter finding means that the regulation of crop nutrition is not going to impact dietary fiber content in our everyday food. Lower temperatures during tillering and higher 1,000 kernel weight and test weight showed a positive correlation with β-glucan content in barley grains, while precipitation during the grain filling period had a negative correlation. Our findings suggest that β-glucan can be obtained from low-input and organic systems as efficiently as from fertilized treatments. Rabbit meat provides a good source of high-quality animal protein with low fat content, it is considered a functional food. This study aimed to determine the growth performance, carcass traits, and chemical quality of New Zealand White rabbit meat breeding treated with different growth promoters. Therefore, 40 apparently healthy New Zealand White male rabbits, 5 weeks of age, were allotted to 4 equal experimental groups. The first group was kept as a control group, the second group included rabbits fed ad libitum supplemented with a beta-glucan 2.0 g/kg diet, The third group comprised rabbits that were injected twice intramuscular of boldenone undecylenate (5 mg/kg body weight) with 3-weeks intervals, the fourth group included rabbits that injected intramuscular of 40 µg/kg body weight estradiol day by day for 30 days. The obtained results revealed that the treated groups with growth promoters had a significant increase (P<0.05) in growth performance (total body weight gain and daily weight gain) and dressing percentage. Administration of boldenone undecylenate resulted in a significant increase (P< 0.05) in protein content in their meat compared to the control group. Rabbits injected with estradiol show a nonsignificant effect (P> 0.05) on dressing % or protein content of meat. Rabbits feed on a beta-glucan-supplemented diet show the highest carcass dressing percentage. Further studies were challenging food investigators for the rabbit meat treated with hormonal growth promoters to study their residues and their impact on consumer health. [ABSTRACT FROM AUTHOR]

Beta-glucans are polysaccharides that can be used for different applications, for example as an immunomodulator in food or feed or for managing high cholesterol levels. Certain microalgae species use beta-glucans as energy storage, accumulating them during nutrient depletion. In this study, we examined and compared beta-glucan production during nitrogen depletion in three different algae species, Phaeodactylum tricornutum, Monodopsis subterranea and Cylindrotheca fusiformis, grown in artificially illuminated flat panel airlift reactors, in order to determine the most promising microalgae species for beta-glucan production. Co-products such as fatty acids (especially eicosapentaenoic acid) and the carotenoid fucoxanthin (not produced by M. subterranea) were also considered. Biomass analysis showed that P. tricornutum cultures reached a maximal beta-glucan content of 317 ± 9 mg gDW−1, M. subterranea cultures reached 188 ± 6 mg gDW−1 and C. fusiformis cultures reached 129 ± 13 mg gDW−1. Furthermore, beta-glucan production was faster in P. tricornutum cultures. However, the maximum volumetric beta-glucan concentration reached was higher in M. subterranea cultures compared to P. tricornutum cultures as M. subterranea cultures produced more biomass during nitrogen depletion. In terms of possible co-products, P. tricornutum produced fucoxanthin and EPA, whereas M. subterranea did not produce fucoxanthin. However, M. subterranea exhibited a higher EPA content, which remained above 45 mg g−1 even after several days of nitrogen depletion. This study was directed to incorporate barley flour derived from hulled barley variety "Jau-17" in the whole wheat flour to develop chapatti. Five different samples of flour blend for chapatti production were made by adding barley (at the rate of 0, 5, 10, 15, 20 and 25%) into the whole wheat flour and their proximate analyses and other quality variables (protein, fat, ash, gluten, energy, phytates and beta-glucan contents) were determined. The same analyses were also performed for the chapattis made from the flour of these blends including the evaluation of sensory parameters for product acceptability. A significant increase in protein, ash, energy and beta-glucans content and a significant decrease in gluten, carbohydrates and phytates were observed in flour blends. A similar trend was observed in chapatti characteristics. Fat contents increased, however, in a non-significant manner in flour blends and chapati. Sensory attributes illustrated that chapatti developed from 20% barley incorporation into wheat flour had good nutritional quality and acceptability. [ABSTRACT FROM AUTHOR]

This paper explores the proximate composition of spring barley seeds and the b-glucan’s content in various two-rowed spring barley genotypes cultivated using different agricultural systems. The genotypes originated from both autochthonous and foreign sources and were grown under conventional, ecological, and no-treatment conditions. The proximate analysis of spring barley seeds was performed using near-infrared spectrometry, while the b-glucan content was determined enzymatically. The data analysis included principal component analysis and cluster analysis to identify ideal candidates for specific applications. The results indicated that the conventional cultivation system yielded higher levels of starch, sugar, protein and ash content; conversely, the ecological and no-treatment systems showed variations in nutritional parameters, with the no-treatment approach resulting in significantly higher b-glucan and fat content. In line with previous research, this study highlights the influence of genotype and agricultural practices on the barley seeds’ composition. Overall, this research contributes to a better understanding of the nutritional composition of barley seeds and offers insights into selecting specific barley genotypes for various applications in the food, feed, and brewing industries; it also underscores the potential for utilizing spring barley's b-glucan to enhance the nutritional value of various products, promoting cardiovascular and digestive health.

Strawberries are a nutrient dense food rich in vitamins, minerals, non-nutrient antioxidant phenolics, and fibers. Strawberry fiber bioactive structures are not well characterized and limited information is available about the interaction between strawberry fiber and phenolics. Therefore, we analyzed commercial strawberry pomace in order to provide a detailed carbohydrate structural characterization, and to associate structures with functions. The pomace fraction, which remained after strawberry commercial juice extraction, contained mostly insoluble (49.1 % vs. 5.6 % soluble dietary fiber) dietary fiber, with pectin, xyloglucan, xylan, β-glucan and glucomannan polysaccharides; glucose, fructose, xylose, arabinose, galactose, fucose and galacturonic acid free carbohydrates; protein (15.6 %), fat (8.34 %), and pelargonidin 3-glucoside (562 μg/g). Oligosaccharides from fucogalacto-xyloglucan, methyl-esterified rhamnogalacturonan I with branched arabinogalacto-side chains, rhamnogalacturonan II, homogalacturonan and β-glucan were detected by MALDI-TOF MS, NMR and glycosyl-linkage analysis. Previous reports suggest that these oligosaccharide and polysaccharide structures have prebiotic, bacterial pathogen anti-adhesion, and cholesterol-lowering activity, while anthocyanins are well-known antioxidants. A strawberry pomace microwave acid-extracted (10 min, 80 °C) fraction had high molar mass (2376 kDa) and viscosity (3.75 dL/g), with an extended rod shape. A random coil shape, that was reported previously to bind to phenolic compounds, was observed for other strawberry microwave-extracted fractions. These strawberry fiber structural details suggest that they can thicken foods, while the polysaccharide and polyphenol interaction indicates great potential as a multiple-function bioactive food ingredient important for gut and metabolic health.

Background & Aims: Changes in gut microbiota in metabolic dysfunction-associated steatotic liver disease (MASLD) are important drivers of disease progression towards fibrosis. Therefore, reversing microbial alterations could ameliorate MASLD progression. Oat beta-glucan, a non-digestible polysaccharide, has shown promising therapeutic effects on hyperlipidemia associated with MASLD, but its impact on gut microbiota and most importantly MASLD-related fibrosis remains unknown. Methods: We performed detailed metabolic phenotyping, including assessments of body composition, glucose tolerance, and lipid metabolism, as well as comprehensive characterization of the gut-liver axis in a western-style diet (WSD)-induced model of MASLD and assessed the effect of a beta-glucan intervention on early and advanced liver disease. Gut microbiota were modulated using broad-spectrum antibiotic treatment. Results: Oat beta-glucan supplementation did not affect WSD-induced body weight gain or glucose intolerance and the metabolic phenotype remained largely unaffected. Interestingly, oat beta-glucan dampened MASLD-related inflammation, which was associated with significantly reduced monocyte-derived macrophage infiltration and fibroinflammatory gene expression, as well as strongly reduced fibrosis development. Mechanistically, this protective effect was not mediated by changes in bile acid composition or signaling, but was dependent on gut microbiota and was lost upon broad-spectrum antibiotic treatment. Specifically, oat beta-glucan partially reversed unfavorable changes in gut microbiota, resulting in an expansion of protective taxa, including Ruminococcus, and Lactobacillus followed by reduced translocation of Toll-like receptor ligands. Conclusions: Our findings identify oat beta-glucan as a highly efficacious food supplement that dampens inflammation and fibrosis development in diet-induced MASLD. These results, along with its favorable dietary profile, suggest that it may be a cost-effective and well-tolerated approach to preventing MASLD progression and should be assessed in clinical studies. Impact and Implications: Herein, we investigated the effect of oat beta-glucan on the gut-liver axis and fibrosis development in a mouse model of metabolic dysfunction-associated steatotic liver disease (MASLD). Beta-glucan significantly reduced inflammation and fibrosis in the liver, which was associated with favorable shifts in gut microbiota that protected against bacterial translocation and activation of fibroinflammatory pathways. Together, oat beta-glucan may be a cost-effective and well-tolerated approach to prevent MASLD progression and should be assessed in clinical studies.

Introduction: Agaricus blazei mushroom is used as a food and medicine, its effective composition is beta-glucan, which is used to treat some cancers and infections, including hepatitis C.Hepatitis C is an inflammatory disease that causes liver necrosis. Caspase2 protein is one of the factors promoting cell apoptosis and plays a role in tumor suppression. The purpose of this study is to determine the expression changes of the caspase2 gene and its effects on liver cancer. Materials and Methods: In this project, raw expression data was obtained from the NCBI (National Center for Biotechnology Information) GEO (Gene Expression Omnibus) database section and using bioinformatics tools and methods and system biology such as Matlab (An abbreviation of "MATrix LABoratory), GEOR2 (Online software) and Cytoscape, the effect of consuming the desired mushroom on caspase2 gene expression was investigated. Results: It was found that the beta-glucan combination has an increasing effect on target gene expression (p-value=0.05692). Conclusion: The results show that the beta-glucan present in the mushroom can play a role as a prevention and even treatment of liver cancer by increasing the expression of caspase 2 protein by directing the damaged cell towards apoptosis.

Objective: The present study aims to investigate the role of breast cancer-susceptibility gene 1 (BRCA1) protein in theβ-Glucan (βG) molecule mediated regulation of lipopolysaccharide (LPS)-induced liver genotoxicity.Materials and Methods: In this experimental study, totally, 32 male Swiss Albino mice were randomly divided into 4equal groups: control (C), LPS-administered (LPS), βG-administered (βG) and βG-pre-administered/LPS-administered(βG+LPS). The βG was injected at the dose of 150 mg/kg/day intraperitoneally (i.p.) for 3 days. A single dose of 4 mg/kg (i.p.) LPS was administered 24 hours after the last βG injection. BRCA1 expression was determined by westernblot analysis and confirmed by quantitative immunofluorescence. Proliferating cell nuclear antigen (PCNA), nuclearfactor erythroid 2–related factor (Nrf2) and 8-OHdG protein levels were also determined by the immunofluorescenceanalysis. The alkaline comet assay was performed. superoxide dismutase (SOD), catalase (CAT) and membrane lipidperoxidation were biochemically measured, and light microscopic histology was evaluated.Results: The BRCA1 expression level was significantly decreased in the LPS group. However, in the βG+LPSgroup, expression of BRCA1 protein was over 2 folds higher than the control. After the LPS induction, the DNA strandbreaks, oxidative DNA lesions and abnormal proliferation of the liver cells were almost entirely suppressed in βG preadministratedanimals, indicating the BRCA1 mediated ubiquitination of PCNA and activation of the DNA damage repairpathways. Activation of Nrf2 in the βG+LPS group resulted in an increase in the levels of Nrf2 pathway dependentantioxidant enzymes SOD and CAT, prevented the peroxidation of membrane lipids and maintained the histologicalarchitecture of the liver.Conclusion: The results manifested that the βG is a strong inducer of the BRCA1 protein expression in the LPSinducedhepatic stress and the protein constitutes the key component of a βG mediated liver protection against anLPS-induced genotoxic and pathological damage.

As a community Web hub and database for small grains, GrainGenes strives to provide resources for researchers, students, and plant breeders to improve traits such as quality, yield, and disease resistance. Quantitative trait loci (QTL), genes, and genetic maps for quality attributes in GrainGenes represent the historical approach to mapping genes for groat percentage, test weight, protein, fat, and β-glucan content in oat (Avena spp.). Genetic maps are viewable in CMap, the comparative mapping tool that enables researchers to take advantage of highly populated consensus maps to increase the marker density around their genes-of-interest. GrainGenes hosts over 50 genome browsers and is launching an effort for community curation, including the manually curated tracks with beta-glucan QTL and significant markers found via GWAS and cloned cellulose synthase-like AsClF6 alleles.

Phallus atrovolvatus, a wild edible mushroom, has attracted increasing interest for consumption due to its unique taste and beneficial health benefits. This study determined the chemical components in the so-called fruiting body during the egg and mature stages and investigated its gut microbiota-modulating activities. The egg stage contained higher total carbohydrates, dietary fiber, glucans, ash, and fat, while the total protein content was lower than in the mature stage. Two consumption forms, including cooked mushrooms and a mushroom aqueous extract from both stages, were used in this study. An in vitro gut fermentation was performed for 24 h to assess gut microbiota regulation. All mushroom-supplemented fermentations increased short-chain fatty acid (SCFA) production compared to the blank control. Furthermore, all mushroom supplementations promoted the growth of Bifidobacterium and Streptococcus. Samples from the mature stage increased the relative abundance of Clostridium sensu stricto 1, while those from the egg stage increased the Bacteroides group. The inhibition of harmful bacteria, including Escherichia-Shigella, Klebsiella, and Veillonella, was only observed for the mature body. Our findings demonstrate that P. atrovolvatus exhibits potential benefits on gut health by promoting SCFA production and the growth of beneficial bacteria, with the mature stage demonstrating superior effects compared to the egg stage.

Simple Summary: Beta-glucan (βG) has been widely employed in animal diets, acting as a tool for intestinal health, improving immune status, and promoting enhanced performance. However, its origin can be from cereal grains, fungi, and algae, and in this context, its glycosidic linkages (1,3), (1,4), or (1,6) directly influence its functions in animal organisms. In general, it was observed that supplementing sow feed with 200 g of βG-(1,3) derived from the algae Euglena gracilis per ton, starting from the 85th day of gestation, resulted in enhancements in both colostrum production and the concentrations of immunoglobulins A, G, and M. This suggests a potential role as an immunomodulatory agent. This supplementation can become a tool to quantitatively and qualitatively enhance the colostrum provided to piglets. The effects of supplementing the diet of sows with βG-(1,3) derived from Euglena gracilis algae were assessed regarding quality and amount of colostrum as well as performance of piglets. A total of 120 sows (first (nulliparous) to sixth parity (multiparous)) from D85 of gestation until weaning were divided into two groups: the control diet group (n = 60) and the βG-(1,3) diet group (n = 60). Sows receiving βG-(1,3) exhibited an average increase of 870 g (24.9%) in colostrum production, leading to a 25.17% higher intake of colostrum by piglets. Furthermore, piglets in the βG-(1,3) group showed significantly superior weight gain of 34 g (50%) compared to the control group 18 h after birth (p < 0.05). Sows fed with βG-(1,3) produced colostrum with significantly higher concentrations of IgG (5.914 mg/mL, 16.16%) and IgM (0.378 mg/mL, 16.29%) than the control group (p < 0.05). Similarly, serum concentrations of IgG (13.86 mg/mL, 51.25%), IgA (17.16 mg/mL, 120.19%), and IgM (13.23 mg/mL, 144.78%) were significantly higher in sows fed with βG-(1,3) than in the control group (p < 0.05). Supplementing sows with βG-(1,3) derived from the Euglena gracilis algae resulted in increased colostrum production and consumption, along with greater weight gain in piglets during the first 18 h after birth. This research aimed to examine the potential alleviative effects of beta-glucan administration on fatigue, unrefreshing sleep, anxiety/depression symptoms and health-related quality of life in ME/CFS. A 36-week unicenter, randomized, double-blind, placebo-controlled trial was conducted in 65 ME/CFS patients, who were randomly allocated to one of two arms to receive four capsules each one of 250 mg beta-glucan, 3.75 µg vitamin D3, 1.05 mg vitamin B6, and 7.5 mg zinc (n = 35), or matching placebo including only microcrystalline cellulose as an excipient (n = 30) once daily. The findings showed that the beta-glucan supplementation significantly improved cognitive fatigue (assessed with FIS-40 scores) after the 36-week treatment compared to the baseline (p = 0.0338). Taken together, this study presents the novel finding that yeast-derived beta-glucan may alleviate cognitive fatigue symptoms in ME/CFS. Thus, it offers valuable scientific insights into the potential use of yeast beta-glucan as a nutritional supplement and/or functional food to prevent or reduce cognitive dysfunction in patients with ME/CFS. Abstract Introduction Stretch marks are common atrophic dermal scars with significant physical and psychological effects. Therefore, there is a need for effective cosmetics and procedures for stretch mark treatment. This study aimed to evaluate the efficacy and safety of a novel treatment for stretch marks that is made up of topical formulations containing beta-glucan combined with nanofractional radiofrequency. Methods This randomized, blinded control trial enrolled 64 Chinese women aged 20–45 years at > 6 months after delivery with obvious white or silver abdominal stretch marks. Participants were randomly allocated to group A (blank group), group B (topical product group), group C (product combined with nanofractional radiofrequency), and group D (vehicle combined with nanofractional radiofrequency). The stretch mark width, skin elasticity, skin color, skin thickness, and collagen density were noninvasively measured. Two trained assessors evaluated the severity, color, outline, and relaxation of the striae. Results Group C showed the best treatment efficacy, with no adverse effects observed during the study period. Conclusion Our findings indicate that stretch mark treatment using topical formulations containing beta-glucan, combined with nanofractional radiofrequency plus magnetic nanofractional radiofrequency, is tolerable and effective. Abstract Background Blooms of marine microalgae play a pivotal role in global carbon cycling. Such blooms entail successive blooms of specialized clades of planktonic bacteria that collectively remineralize gigatons of algal biomass on a global scale. This biomass is largely composed of distinct polysaccharides, and the microbial decomposition of these polysaccharides is therefore a process of prime importance. Results In 2020, we sampled a complete biphasic spring bloom in the German Bight over a 90-day period. Bacterioplankton metagenomes from 30 time points allowed reconstruction of 251 metagenome-assembled genomes (MAGs). Corresponding metatranscriptomes highlighted 50 particularly active MAGs of the most abundant clades, including many polysaccharide degraders. Saccharide measurements together with bacterial polysaccharide utilization loci (PUL) expression data identified β-glucans (diatom laminarin) and α-glucans as the most prominent and actively metabolized dissolved polysaccharide substrates. Both substrates were consumed throughout the bloom, with α-glucan PUL expression peaking at the beginning of the second bloom phase shortly after a peak in flagellate and the nadir in bacterial total cell counts. Conclusions We show that the amounts and composition of dissolved polysaccharides, in particular abundant storage polysaccharides, have a pronounced influence on the composition of abundant bacterioplankton members during phytoplankton blooms, some of which compete for similar polysaccharide niches. We hypothesize that besides the release of algal glycans, also recycling of bacterial glycans as a result of increased bacterial cell mortality can have a significant influence on bacterioplankton composition during phytoplankton blooms. The carbohydrate ganglioside GD2/GD3 cancer vaccine adjuvanted by β-glucan stimulates anti-GD2 IgG1 antibodies that strongly correlate with improved progression-free survival (PFS) and overall survival (OS) among patients with high-risk neuroblastoma. Thirty-two patients who relapsed on the vaccine (first enrollment) were re-treated on the same vaccine protocol (re-enrollment). Titers during the first enrollment peaked by week 32 at 751 ± 270 ng/mL, which plateaued despite vaccine boosts at 1.2–4.5 month intervals. After a median wash-out interval of 16.1 months from the last vaccine dose during the first enrollment to the first vaccine dose during re-enrollment, the anti-GD2 IgG1 antibody rose to a peak of 4066 ± 813 ng/mL by week 3 following re-enrollment (p < 0.0001 by the Wilcoxon matched-pairs signed-rank test). Yet, these peaks dropped sharply and continually despite repeated boosts at 1.2–4.5 month intervals, before leveling off by week 20 to the first enrollment peak levels. Despite higher antibody titers, patients experienced no pain or neuropathic side effects, which were typically associated with immunotherapy using monoclonal anti-GD2 antibodies. By the Kaplan–Meier method, PFS was estimated to be 51%, and OS was 81%. The association between IgG1 titer during re-enrollment and β-glucan receptor dectin-1 SNP rs3901533 was significant (p = 0.01). A longer prime–boost interval could significantly improve antibody responses in patients treated with ganglioside conjugate cancer vaccines.

Beta-glucan (β-glucan) is a natural polysaccharide produced by fungi, bacteria, and plants. Although it has been reported that β-glucan enhances innate immune memory responses, it is unclear whether different types of β-glucans display similar immune effects. To address this issue, we employed zymosan (β-1,3-glycosidic linkage) and pustulan (β-1,6-glycosidic linkage) to investigate their in vivo effects on innate memory immune responses. We examined the changes of innate memory-related markers in macrophages and natural killer (NK) cells, two immune cell types that display innate memory characteristics, at two different time points (16 h and 7 days) after β-glucan stimulation. We found that short-term (16 h) zymosan treatment significantly induced macrophages to upregulate IL15 production and increased surface IL15Rα expression on NK cells. In addition, long-term (7 days) zymosan treatment significantly induced macrophages to upregulate the expression of innate memory-related markers (e.g., TNFα, HIF1α, and mTOR) and induced NK cells to express enhanced levels of KLRG1, known as an innate memory-like marker. Our results provide support that zymosan can be an effective adjuvant to promote innate memory immune responses, providing a bridge between innate and adaptive immune cells to enhance various immune responses such as those directed against tumors. Oats are considered a functional food due to the beneficial health effects associated with their consumption and are suitable to be explored for their ability to prevent or manage chronic disease, such as hypertension. Here, we examined the cardiovascular benefits of an oat beta-glucan extract in male and female spontaneously hypertensive rats (SHRs) to unravel its sex-specific roles when used with an anti-hypertensive medication, hydrochlorothiazide. Five-week-old male and female SHRs and Wistar–Kyoto (WKY) rats were treated with oat beta-glucan and hydrochlorothiazide for 15 weeks. Twenty-week-old male and female SHRs showed high blood pressure (BP), cardiac remodeling, and cardiac dysfunction. These animals also had significantly increased levels of malondialdehyde (MDA), angiotensin II, and norepinephrine. Treatments with beta-glucan and hydrochlorothiazide were able to differentially prevent high BP, cardiac dysfunction, and alterations in malondialdehyde (MDA), angiotensin II, and norepinephrine in 20-week-old male and female SHRs. To conclude, beta-glucan alone and in combination with hydrochlorothiazide may be a promising a strategy for managing hypertension and related cardiac complications. [ABSTRACT FROM AUTHOR]

To promote the use of waxy barley bran, an underutilized resource, samples of waxy barley were divided into three parts: polished waxy barley powder (PWBP), inner bran layer powder (IBLP), and outer bran layer powder (OBLP). The color and appearance, general properties, minerals, vitamins, β-glucan, antioxidant properties, and aroma of each part were compared. In terms of appearance and color, IBLP and OBLP appeared more yellow than PWBP; general components that were more abundant in IBLP and OBLP compared with PWBP were protein, fat, and ash. IBLP and OBLP had characteristically high values of Mg and Zn, monounsaturated and polyunsaturated fatty acids, vitamin B1, total polyphenol content, H-ORAC, and DPPH. In particular, the vitamin B1 content of OBLP was approximately 10 times higher than that of PWBP, and Mg and Zn content was more than five times higher than in PWBP. The β-glucan content of IBLP and OBLP was lower than that of PWBP, but relatively high. GC–MS analysis revealed that hexanal was the aroma component common to all three samples, and the peak areas were in the order of PWBP > OBLP > IBLP. [ABSTRACT FROM AUTHOR]

This review is to summarize and analyze the currently available knowledge concerning the action of oat (Avena sativa L.) consumption on obesity, as well as possible constituents and extra- and intracellular mediators responsible for its anti-obesity effect. The oat constituents could reduce fat storage via several mediatory mechanisms - brain centers regulating appetite, gastrointestinal functions, gut bacteria, fat synthesis and metabolism and maybe via changes in oxidative processes, steroid hormones receptors and adipose tissue vascularization. Several oat constituents (starch, fiber and beta-glucan) could have anti-obesity properties, whilst one oat constituent (starch or fiber) could affect fat storage via several mechanisms of action. Introduction: Stretch marks are common atrophic dermal scars with significant physical and psychological effects. Therefore, there is a need for effective cosmetics and procedures for stretch mark treatment. This study aimed to evaluate the efficacy and safety of a novel treatment for stretch marks that is made up of topical formulations containing beta-glucan combined with nanofractional radiofrequency. Methods: This randomized, blinded control trial enrolled 64 Chinese women aged 20–45 years at > 6 months after delivery with obvious white or silver abdominal stretch marks. Participants were randomly allocated to group A (blank group), group B (topical product group), group C (product combined with nanofractional radiofrequency), and group D (vehicle combined with nanofractional radiofrequency). The stretch mark width, skin elasticity, skin color, skin thickness, and collagen density were noninvasively measured. Two trained assessors evaluated the severity, color, outline, and relaxation of the striae. Results: Group C showed the best treatment efficacy, with no adverse effects observed during the study period. Conclusion: Our findings indicate that stretch mark treatment using topical formulations containing beta-glucan, combined with nanofractional radiofrequency plus magnetic nanofractional radiofrequency, is tolerable and effective. The current study was designed to evaluate the therapeutic potential of beta‐glucan (BG), which is a key bioactive compound predominantly present in mushrooms and cereals against high‐fat‐diet (HFD)‐induced obesity, and to understand its mechanism of action. Obesity was induced in rats by supplementing the diet with HFD and BG (40 mg/kg body weight) for a period of 6 weeks. At the end of the experimental period, the body weight, as well as hyperglycemic, dyslipidemia, and obesogenic marker expressions, was assessed in the control group and in the experimental obese rats. Administration of BG to obese rats significantly reduced body weight gain and attenuated hyperglycemia, which was confirmed by the decreased blood glucose and insulin resistance. At the same time, BG mitigated dyslipidemia by altering expressions of peroxisome proliferator‐activated receptor‐gamma (PPAR‐γ), sterol regulatory element‐binding protein 1 (SREBP‐1c), fatty acid synthase (FAS), 3‐hydroxy‐3‐methylglutaryl‐CoA (HMG‐CoA) reductase, and fatty acid‐binding protein‐4 (Fab‐4) in HFD‐induced obese rats. In conclusion, this study revealed that BG is a potential candidate to ameliorate HFD‐induced obesity by modulating obesogenic marker expressions, especially by regulating the master regulator PPAR‐γ. Wheat flour noodles are sometimes fortified with β-glucan for nutritional value, but this can decrease eating quality. The contributions of β-glucan and starch molecular fine structure to physicochemical properties of wholemeal oat flour and to the texture of oat-fortified white salted noodles were investigated here. Hardness of oat-fortified noodles was controlled by the longer amylopectin chains (DP ≥ 26) and amount of longer amylose chains (DP ≥ 1000). Higher levels of β-glucan, in the range from 3.1 to 5.2%, result in increased noodle hardness. Pasting viscosities of wholemeal oat flour positively correlate with the hardness of oat-fortified noodles. The swelling power of oat flour is not correlated with either pasting viscosities of oat flour or noodle hardness. Longer amylopectin chains and the amount of longer amylose chains both control the pasting viscosities of oat flour, which in turn affect noodle texture. This provides new means, based on starch and β-glucan molecular structure, to choose oats with optimal starch structure and β-glucan content for targeted oat-fortified noodle quality.

The paper reviews a career of more than forty years researching topics in malting and brewing science. Some themes attracted particularly close attention, namely the endosperm cell walls of barley, dimethyl sulphide, flavour stability, foam and the impact of beer on health. However, the scope has been far broader than that. The underlying imperative was to pursue research that was close to application and always focussed on a specific need in the processes involved in the production of beer. © 2020 The Institute of Brewing & Distilling.
