Distribution patterns of benthic dinoflagellates that are widely observed in tropical and temperate waters and have toxic potential are changing in response to ocean climate change. Although there have been no outbreaks associated with the genus Coolia affecting humans, it is crucial to understand their changing distribution and clearly identify the species in the study area to prepare for potential toxic events. In this study, five strains of Coolia species were isolated from macroalgae samples collected from Jeju Island and the eastern coastal waters of Korea. Through morphological and molecular analysis of these isolates, one strain was identified as Coolia palmyrensis , marking the first report of this species in Korea, and four strains as C. malayensis. One of the C. malayensis strains was isolated in Pohang on the eastern coast of Korea, where it had not been previously reported. From 2021 to 2023, monitoring of Jeju Island using a quantitative polymerase chain reaction (PCR) assay revealed that C. palmyrensis cells occurred mostly in autumn, with a maximum density of 242 cells g-1, and overwintering populations were observed in 2023. However, C. malayensis cells were not observed in this area. Additionally, C. malayensis was observed in Pohang and Ulsan, located further north than Jeju Island with maximum cell densities of 537 and 201 cells g-1, respectively. These data and our decade of monitoring experience confirmed the introduction and establishment of C. palmyrensis and habitat shift of C. malayensis in Korean coastal waters. The genus Gambierdiscus produces an array of bioactive hydrophilic and lipophilic secondary metabolites that range in mode of action and toxicity. In this study, the metabolite fingerprint was mapped for thirteen Gambierdiscus, five Coolia and two Fukuyoa species (34 isolates) by assessing the production of 56 characterised secondary metabolites. Gambierdiscus polynesiensis was the only species to produce Pacific-ciguatoxin-3B (P-CTX3B), P-CTX3C, iso-P-CTX3B/C, P-CTX4A, P-CTX4B and iso-P-CTX4A/B. G. australes produced maitotoxin-1 (MTX-1) and MTX-5, G. cheloniae produced MTX-6 and G. honu produced MTX-7. Ubiquitous production of 44-methylgambierone was observed amongst all the Gambierdiscus isolates, with nine species also producing gambierone. Additional gambierone analogues, including anhydrogambierone (tentatively described herein), were also detected in all Gambierdiscus species, two Coolia and two Fukuyoa species. Gambieroxide was detected in G. lewisii and G. pacificus and gambieric acid A was detected in ten Gambierdiscus species, with G. australes (CAWD381) being the only isolate to produce gambieric acids A–D. This study has demonstrated that the isolates tested to date produce the known CTXs or MTXs, but not both, and highlighted several species that produced 'unknown' compounds displaying characteristics of cyclic polyethers, which will be the focus of future compound discovery efforts. The genus Amphidinium Clap. & J. Lachm. comprises a high diversity of planktonic and benthic (epiphytic and sand-dwelling) dinoflagellates from marine and freshwater ecosystems. High morphological plasticity and vaguely defined genus characteristics (e.g., a small epicone size) have complicated the clear delineation of species boundaries. Although six Amphidinium morphospecies have been reported from Mexican coastal waters, species identifications are uncertain and not generally supported by molecular phylogenetic data. In this study, seven isolates of Amphidinium from diverse benthic coastal locations on the NE Pacific, Gulf of California, and southern Gulf of Mexico were subjected to critical morphological analysis using photonic and scanning electron microscopy. The phylogenetic reconstruction was based on nuclear-encoded, partial large-subunit (LSU) rDNA and internal transcribed spacer I and II (ITS1 and ITS2) sequences. The revised phylogenetic analysis was consistent with the traditional subdivision of the genus Amphidinium into two sister groups: Herdmanii and Operculatum clades. This study provided the first confirmed records of A. theodorei and A. massartii from coastal waters of Mexico. The molecular phylogenetic evidence indicated that the morphologically described A. cf. carterae from Baja California was in fact more closely allied with A. eilatiensis sequences. A few Amphidinium species are known to form toxigenic (i.e., fish-killing) harmful algal blooms worldwide, and therefore knowledge on species diversity and biogeography is critical in developing effective strategies for evaluating the potential emerging threat in Mexican coastal waters.

This study aimed to assess the harmful effects of C. cf. canariensis phylogroup II (PII) and C. malayensis strains through bioassays. Experimental exposures (24, 48, and 72 h) of Artemia salina nauplii to Coolia species (330–54,531 cells mL−1) were performed independently. When a concentration-dependent response was achieved, additional experiments were carried out to evaluate the cell-free medium toxicity. The two Coolia species were harmful to Artemia nauplii, inducing significant mortality and sublethal responses. Coolia cf. canariensis PII was the most toxic species, inducing significant lethality at lower concentrations and shorter exposure times, followed by C. malayensis. Only the survival curves achieved after 24 and 48 h of exposure to C. cf. canariensis PII fitted to a concentration–response curve with valid LC50s of 18,064 and 19,968 cells mL−1, respectively. Moreover, extracellular compounds (i.e., culture filtrates) of C. cf. canariensis PII induced significant mortality to nauplii after 48 and 72 h. The toxicity of C. cf. canariensis PII was demonstrated for the first time using bioassays, and it was surprisingly higher than that of the C. malayensis strain, which was previously demonstrated to induce biological activity at the cellular and subcellular levels. Our findings highlight the harmful and lethal effects induced by Coolia cells and the importance of bioassays for toxicity assessments.

Coolia species are epiphytic and benthic dinoflagellates with a cosmopolitan distribution in tropical and subtropical areas. In the austral summer of 2016, during a survey in Bahía Calderilla, a dinoflagellate of the genus Coolia was detected in macroalgae samples, and a clonal culture was established. Subsequently, the cultured cells were observed by scanning electron microscopy (SEM) and identified as C. malayensis based on their morphological characteristics. Phylogenetic analyses based on the LSU rDNA D1/D2 regions confirmed that strain D005‐1 corresponded to C. malayensis and clustered with strains isolated from New Zealand, Mexico, and Asia Pacific countries. Although the strain D005‐1 culture did not contain yessotoxin (YTX), cooliatoxin, 44‐methyl gambierone, or its analogs in detectable amounts by LC–MS/MS, more research is needed to evaluate its toxicity and to determine the possible impact of C. malayensis in northern Chilean waters. The genus Amphidinium has been the subject of recent attention due to the production of polyketide metabolites. Some of these compounds have shown significant bioactivities and could be related to species interactions in the natural benthic microenvironment. Among these compounds, amphidinols (AMs) are suspected to be related to fish kills and probably implicated in ciguatera symptoms associated with the occurrence of benthic harmful algal blooms (bHABs). Here, we present the first report of a variety of AMs produced by cultured strains from several species from the Mexican Pacific, the Gulf of California, and the Gulf of Mexico. Through ultra-high performance liquid chromatography coupled with tandem mass spectrometry (UHPLC-MS/MS), ten previously known AMs (AM02, -04, -05, -06, -07, -09, -11, -14, -15, and -17), four recently reported AMs (N7, N8/N9, N12, and N13), and three new variants (U1, U2, and U3) were identified. Of the twelve analyzed Amphidinium cultures, five were not AM producers, and the cell quotas of the remaining seven strains ranged from close to nondetectable to a maximum of 1694 fg cell−1, with many intermediate levels in between. The cultures from the Mexican North Pacific coast produced AMs in a higher quantity and variety than those from worldwide locations. This is the first study of AMs from Mexican Amphidinium strains, and our results confirm the relevance of continuing the investigation of the genus bioactive metabolites. [ABSTRACT FROM AUTHOR]

A harmful benthic Prorocentrum concavum bloom was recorded in August 2018 in Xincun Bay, China, which is the location of a national seagrass nature reserve. Annual ecological surveys have been conducted to study the population dynamics of P. concavum in the benthic community and water column. Seasonal variations in benthic P. concavum abundance were found and the abundances on seagrass and macroalgae in the wet season were 2.5 and 2.82 times higher, respectively, than those in the dry season, although the differences were not statistically significant. The abundance of P. concavum in the water column differed significantly between seasons. The maximum abundances of benthic and planktonic P. concavum were (1.7 ± 0.59) × 106 cells (100 cm2)−1 on Thalassia hemperichii in July and 2.0 × 104 ± 4.7 × 103 cells L−1 in June, respectively. High spatial heterogeneity in P. concavum abundance was observed among five sampling sites. Abundances were significantly higher in seagrass beds than those in macroalgae beds, mangroves, and coral reefs. The abundance of P. concavum at site A (in a seagrass bed and close to a cage-culture area) was 5.6 times higher than that at site D (seagrass bed and distant from the cage-culture area). Planktonic P. concavum showed a similar spatial distribution and presented a maximum density at site A. Moreover, the abundance of benthic P. concavum also showed heterogeneity on host substrates, and the abundance on T. hemperichii was significantly higher than that on sediment. Based on a Spearman’s test, temperature, dissolved organic phosphorus, and dissolved organic nitrogen were the three important factors driving the spatiotemporal distribution of benthic P. concavum in Xincun Bay. Planktonic P. concavum were derived from cells on the substrates and were influenced by concentrations of dissolved oxygen. In conclusion, seagrass beds may be a reservoir of harmful benthic algal blooms in Xincun Bay and the dense cage-culture area provides sufficient organic nutrients for the growth and reproduction of benthic dinoflagellates.

• O. cf. siamensis had a colder and shorter thermal niche than O. cf. ovata. • Some clones of O. cf. siamensis have a fast growing strategy, not O. cf. ovata. • Acclimation to temperature relies on xanthophyll cycle pigments or lipid content. • Toxicity decreases with low temperatures and is maximal under optimal growth ones. • When both species are present, the toxic risk may increase with the global warming. Reports of the benthic dinoflagellate Ostreopsis spp. have been increasing in the last decades, especially in temperate areas. In a context of global warming, evidences of the effects of increasing sea temperatures on its physiology and its distribution are still lacking and need to be investigated. In this study, the influence of temperature on growth, ecophysiology and toxicity was assessed for several strains of O. cf. siamensis from the Bay of Biscay (NE Atlantic) and O. cf. ovata from NW Mediterranean Sea. Cultures were acclimated to temperatures ranging from 14.5 °C to 32 °C in order to study the whole range of each strain-specific thermal niche. Acclimation was successful for temperatures ranging from 14.5 °C to 25 °C for O. cf. siamensis and from 19 °C to 32 °C for O. cf. ovata, with the highest growth rates measured at 22 °C (0.54–1.06 d−1) and 28 °C (0.52–0.75 d−1), respectively. The analysis of cellular content of pigments and lipids revealed some aspects of thermal acclimation processes in Ostreopsis cells. Specific capacities of O. cf. siamensis to cope with stress of cold temperatures were linked with the activation of a xanthophyll cycle based on diadinoxanthin. Lipids (neutral reserve lipids and polar ones) also revealed species-specific variations, with increases in cellular content noted under extreme temperature conditions. Variations in toxicity were assessed through the Artemia franciscana bioassay. For both species, a decrease in toxicity was observed when temperature dropped under the optimal temperature for growth. No PLTX-like compounds were detected in O. cf. siamensis strains. Thus, the main part of the lethal effect observed on A. franciscana was dependent on currently unknown compounds. From a multiclonal approach, this work allowed for defining specificities in the thermal niche and acclimation strategies of O. cf. siamensis and O. cf. ovata towards temperature. Potential impacts of climate change on the toxic risk associated with Ostreopsis blooms in both NW Mediterranean Sea and NE Atlantic coast is further discussed, taking into account variations in the geographic distribution, growth abilities and toxicity of each species. Ciguatera poisoning is mainly caused by the consumption of reef fish that have accumulated ciguatoxins (CTXs) produced by the benthic dinoflagellates Gambierdiscus and Fukuyoa. China has a long history of problems with ciguatera, but research on ciguatera causative organisms is very limited, especially in the Beibu Gulf, where coral reefs have been degraded significantly and CTXs in reef fish have exceeded food safety guidelines. Here, five strains of Gambierdiscus spp. were collected from Weizhou Island, a ciguatera hotspot in the Beibu Gulf, and identified by light and scanning electron microscopy and phylogenetic analyses based on large and small subunit rDNA sequences. Strains showed typical morphological characteristics of Gambierdiscus caribaeus, exhibiting a smooth thecal surface, rectangular-shaped 2′, almost symmetric 4″, and a large and broad posterior intercalary plate. They clustered in the phylogenetic tree with G. caribaeus from other locations. Therefore, these five strains belonged to G. caribaeus, a globally distributed Gambierdiscus species. Toxicity was determined through the mouse neuroblastoma assay and ranged from 0 to 5.40 fg CTX3C eq cell−1. The low level of toxicity of G. caribaeus in Weizhou Island, with CTX-contaminated fish above the regulatory level in the previous study, suggests that the long-term presence of low toxicity G. caribaeus might lead to the bioaccumulation of CTXs in fish, which can reach dangerous CTX levels. Alternatively, other highly-toxic, non-sampled strains could be present in these waters. This is the first report on toxic Gambierdiscus from the Beibu Gulf and Chinese waters and will provide a basis for further research determining effective strategies for ciguatera management in the area.

Highlights • No clear intra-seasonal variation of benthic dinoflagellate abundances in Guadeloupe and in Martinique. • Benthic dinoflagellates showed a preferential association with certain macrophytes. • Halophila stipulacea, hosted the highest Gambierdiscus spp. abundances. • Sargassum spp. hosted the lowest number of benthic dinoflagellates. Abstract Epibenthic dinoflagellates were monitored monthly over an 18 month period in Guadeloupe and Martinique (Lesser Antilles, Caribbean Sea). These islands are located in the second most affected ciguatera fish poisoning (CFP) region of the world. Guadeloupe presented five times more total epibenthic dinoflagellates and two times less abundant Gambierdiscus spp. compared to Martinique, although the area of frequent CFP outbreaks covers Guadeloupe and not Martinique. Results did not show any clear seasonal variations of benthic dinoflagellates abundances. Temperature and salinity were not driving parameters in the evolution of total benthic dinoflagellate abundances. Preferential associations were found between macrophyte species and epibenthic dinoflagellates. The Phaeophyceae Dictyota spp. hosted the highest abundances of total epibenthic dinoflagellates, composed mainly of Ostrepsis and Prorocentrum genera. The seagrass Halophila stipulacea hosted the highest abundances of Gambierdiscus spp. and Sinophysis spp. whilst the highest abundance of Coolia was determined on Galaxaura spp. The pelagic Sargassum spp. hosted the lowest abundances of benthic dinoflagellates including the genus Gambierdiscus. Marine benthic dinoflagellate toxins, potent bioactive compounds with wide-ranging presence in marine ecosystems, have surged in response to global climate change and human activities, prompting an urgent and imperative inquiry. This study conducts an in-depth review of contemporary research concerning these toxins, employing meticulous bibliometric analysis. Leveraging a dataset of 736 relevant literatures sourced from the Web of Science (spanning from 2000 to May 2023), our analysis delves comprehensively into the scientific discourse surrounding these toxic compounds. Employing tools such as VOSviewer, co-citation analysis, co-occurrence analysis, and cluster analysis, our study yields nuanced insights into the intricate characteristics and trajectories of the field. The co-citation analysis underscores the pivotal role played by benthic and epiphytic dinoflagellates like Ostreopsis and Gambierdiscus in shaping prevailing research trends. Our study identifies four distinct research directions, encompassing the domains of ecology, toxicology, toxin production, and taxonomy. Moreover, it traces the evolutionary journey of research stages, marking the transition from a focus on taxonomy to an emphasis on unraveling molecular mechanisms. The culmination of our comprehensive analysis yields three pertinent research recommendations: a call for widescale global studies, the advancement of rapid toxin monitoring techniques, and a deeper exploration of the factors influencing toxin synthesis and toxicity. Abstract The genus Gambierdiscus produces ciguatera toxins, which has led to extensive studies of this genus. Epiphytic dinoflagellate cells were isolated from coralline macroalgae collected from the coastal waters of Jeju Island, Korea, and two clonal cultures were established. The morphology of these cells was examined using light and scanning electron microscopy, and the sequences of the small subunit (SSU), large subunit (LSU), and internal transcribed spacer (ITS) region of rDNA were determined. The phylogenetic trees based on SSU, LSU (D1-D3), and LSU (D8-D10) rDNA showed the two Korean isolates forming a clade with unidentified strains named Gambierdiscus sp. type 2. This clade showed a clear divergence from the two closest clades containing of the G. caribaeus and G. carpenteri strains. Morphologically, the Korean isolates had an anteroposteriorly compressed cell shape having a smooth theca ornamentation. Growth of the Korean isolates ceased when water temperatures were <20 °C or >30 °C with an optimum at 26 °C. This temperature preference may account for why Gambierdiscus sp. type 2 has only been isolated from the temperate waters of Korea and Japan. However, the two most closely related species G. caribaeus and G. carpenteri have broader temperature ranges tolerating as high as 34–36 °C and are globally distributed. Based on these results, the Korean isolates are proposed as a new species in the genus Gambierdiscus and named Gambierdiscus jejuensis sp. nov. The genus Gambierdiscus is a group of marine epiphytic-benthic dinoflagellates that produce the toxins that cause ciguatera fish poisoning (CFP). To date, 15 species and six ribotypes of Gambierdiscus have been identified, and multiple species commonly co-occur within a single site or epiphyte community. Toxicity can vary by species; thus, it is important to be able to differentiate among species for research and monitoring purposes. Species comprising the Gambierdiscus genus have very similar morphological characteristics and are difficult or impossible to distinguish using light microscopy. DNA sequencing has been an important tool in the definition of Gambierdiscus species, but it can be time-consuming and expensive. To provide an alternative approach, a new polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) protocol was developed for efficient and accurate identification of Gambierdiscus strains isolated from the Caribbean Sea. The assay targeted the D1-D2 hypervariable regions of the large subunit ribosomal RNA (LSU rDNA) gene and used a single restriction enzyme, BsrI. This method produced distinct RFLP banding patterns for the six species of Gambierdiscus reported from the Gulf of Mexico and Caribbean Sea and also distinguished them from four Pacific endemic species. This method was successfully used to type 472 clonal isolates of Gambierdiscus from the US Virgin Islands and east coast of Mexico. The dinoflagellate genus Bysmatrum encompasses five epibenthic or tide-pool species and has been characterized by separated anterior intercalary plates. In the present study, we obtained six strains of Bysmatrum from the South China Sea and French Atlantic coast by isolating single cells/cysts from plankton and sediment samples. All strains were examined with light microscopy and scanning electron microscopy. Based on morphological observations, three strains were identified as Bysmatrum subsalsum, characterized by the elongated and rectangular first and a hexagonal second anterior intercalary plate. They differ from each other in the number of sulcal lists and the configuration of the first anterior intercalary plate. One strain was identified as Bysmatrum gregarium and the other two as Bysmatrum granulosum. The cyst-theca relationship of B. subsalsum from the French Atlantic was established by incubation of the cyst, and the geochemical composition of the cyst wall was measured through micro-Fourier transform infrared spectroscopy. Bysmatrum subsalsum from Malaysia shows a bright red stigma in the sulcal area under light microscopy, which was confirmed with transmission electron microscopy: it was identified as a type B eyespot. Small subunit ribosomal DNA (SSU rDNA), partial large subunit ribosomal DNA (LSU rDNA) and internal transcribed spacer (ITS) sequences were obtained from all six strains. The maximum likelihood and Bayesian inference analysis based on concatenated SSU, ITS and LSU sequences revealed that Bysmatrum is monophyletic and nested within Peridiniales. Our strains of B. subsalsum form a new ribotype in the molecular phylogeny (designated as ribotype B). The genetic distance based on ITS sequences among Bysmatrum species ranged from 0.34 to 0.47 and those genetic distances at the intraspecific level of B. subsalsum could reach 0.41, supporting the possibility of hidden crypticity within B. subsalsum. Coolia are marine benthic dinoflagellates which are globally distributed and potentially toxic. This study provides the first investigation of species diversity and toxicity assessment of Coolia in Hong Kong waters. Fifty-one strains of four Coolia species, including C . malayensis , C . canariensis , C . tropicalis , and C . palmyrensis , were isolated from twelve sub-tidal habitats, and identified phylogenetically using 28S rDNA sequences. Exposure experiments (48-hour) demonstrated that the algal lysates extracted from the four Coolia species exhibited different toxic effects on the lethality and abnormality of two invertebrate larvae, i.e., brine shrimp Artemia franciscana and sea urchin Heliocidaris crassispina . Heliocidaris crassispina was more sensitive to the toxic effects of Coolia species than A . franciscana . Toxicity tests from both larvae revealed that C . malayensis was generally more toxic, and caused higher mortality rates when compared with the other three species. SUMMARY A new species of benthic marine dinoflagellate, Pyramidodinium spinulosum Horiguchi, Moriya, Pinto & Terada is described from the deep (36 m) seafloor off Mageshima Island, Kagoshima Prefecture, Japan in the subtropical region of the northwest Pacific. The life cycle of the dinoflagellate consists of a dominant, attached, dome-shaped, vegetative form and short-lasting, motile cell. Asexual reproduction takes place by the formation of two motile cells within each non-motile cell. The released motile cells swim only for a short period and transform directly into the dome-shaped vegetative form. The duration of the cell cycle varies and can be extremely long, ranging 5-38 days under culture conditions. The non-motile cell is enclosed by a cell wall and its surface is covered with many (80 -130) spines of various length. The dinoflagellate is photosynthetic and contains many (more than 50) discoidal chloroplasts. Phylogenetic analysis reveals that the dinoflagellate is closely related to the type species of the genus Pyramidodinium, P. atrofuscum which also possesses a dominant, attached, non-motile form. However, P. spinulosum can be clearly distinguished from P. atrofuscum by the cell shape (dome-shaped vs. pyramid-shaped) and surface ornamentation (spines vs. wart-like processes) of the non-motile form. Based on these morphological differences together with molecular evidence, it was concluded that this organism from a deep water sand sample should be described as a second species of the genus Pyramidodinium, P. spinulosum. Satellite-derived sea surface temperature data from the MODIS Aqua instrument were used to project rates of growth for four Gambierdiscus and one Fukuyoa species, dinoflagellates associated with ciguatera fish poisoning. Dinoflagellate growth rates were modelled using experimental temperature vs. growth equations and projected bottom temperatures with light penetration and bathymetry masks. Daily projected growth rates for each species were used to calculate monthly, yearly and multiyear averages between 2003 and 2013. The resulting projections were then used to characterize patterns of regional Gambierdiscus/Fukuyoa abundance and compare these to ciguatera fish poisoning incidences in the Greater Caribbean Region. Model output indicated the highest growth potential was in the shelf waters of the Caribbean Sea, with moderate growth in the Bahamas, southern Florida and Gulf of Mexico; the lowest growth potential was in the northern Gulf of Mexico and along the U.S. south Atlantic coast. Mean projected growth rates generally coincided with distribution of ciguatera fish poisoning incidences in the region, with some exceptions in the southwestern Caribbean. The results of this study indicate spatial differences in Gambierdiscus and Fukuyoa growth likely play a prominent role in governing the occurrence of ciguatera fish poisoning in the greater Caribbean. Species of the benthic dinoflagellate genus Gambierdiscus are the putative producer of the toxins causing ciguatera fish poisoning (CFP) in tropical and subtropical areas. Although CFP events and distribution of G. caribaeus have been reported in Thailand, little is known about the conditions that enhance the growth of Gambierdiscus in Thailand. Flence, this study was aimed at determining the effects of temperature and salinity and their interactions on the growth of Thai G. caribaeus. The strain tested showed the highest cell yield in IMK/2 medium. Based on growth rate (p), Thai G. caribaeus grew in the temperature range of 20°C-35°C and the salinity range of 20M0. The suboptimal condition (> 80% of the maximal growth rate) for the strain tested was 20.5°C-30.5°C and salinity of 25.0-37.5. The optimal condition was 25°C and salinity of 30. Furthermore, the Thai G. caribaeus revealed high toxicity to mice. Our results suggest that tolerance of up to 35°C may allow these organisms to be distributed in tropical areas, including Thailand, where the water temperature often exceeds 30°C. Projected water temperatures at six sites in the Gulf of Mexico and Caribbean Sea were used to forecast potential effects of climate change on the growth, abundance and distribution of Gambierdiscus and Fukuyoa species, dinoflagellates associated with ciguatera fish poisoning (CFP). Data from six sites in the Greater Caribbean were used to create statistically downscaled projections of water temperature using an ensemble of eleven global climate models and simulation RCP6.0 from the WCRP Coupled Model Intercomparison Project Phase 5 (CMIP5). Growth rates of five dinoflagellate species were estimated through the end of the 21st century using experimentally derived temperature vs. growth relationships for multiple strains of each species. The projected growth rates suggest the distribution and abundance of CFP-associated dinoflagellate species will shift substantially through 2099. Rising water temperatures are projected to increase the abundance and diversity of Gambierdiscus and Fukuyoa species in the Gulf of Mexico and along the U.S. southeast Atlantic coast. In the Caribbean Sea, where the highest average temperatures correlate with the highest rates of CFP, it is projected that Gambierdiscus caribaeus , Gambierdiscus belizeanus and Fukuyoa ruetzleri will become increasingly dominant. Conversely, the lower temperature-adapted species Gambierdiscus carolinianus and Gambierdiscus ribotype 2 are likely to become less prevalent in the Caribbean Sea and are expected to expand their ranges in the northern Gulf of Mexico and farther into the western Atlantic. The risks associated with CFP are also expected to change regionally, with higher incidence rates in the Gulf of Mexico and U.S. southeast Atlantic coast, with stable or slightly lower risks in the Caribbean Sea. The benthic dinoflagellates in the genus Gambierdiscus produce toxins that bioaccumulate in tropical and sub-tropical fish causing ciguatera fish poisoning ( CFP). Other co-occurring genera such as Coolia have also been implicated in causing CFP. Little is known about the diversity of the two genera Gambierdiscus and Coolia along the Thai coasts. The results of morphological analyses based on observation under light microscopy and scanning electron microcopy showed that strains of Gambierdiscus from Thailand displayed the typical Gambierdiscus plate formula: Po, 4′, 0a, 6″, 6c,?s, 5′′′, 0p and 2′′′′. Morphological examination of Thai Gambierdiscus enabled it to be identified as Gambierdiscus caribaeus: round and anterior-posteriorly compressed cell shape, broad 2′′′′ plate, rectangular 2′ plate, and symmetrical 3″ plate. The phylogenetic analyses based on the large subunit ( LSU) rDNA D8/ D10 sequences of Gambierdiscus from Thailand confirmed the morphological identification. The thecal plate formula for all of the Coolia isolates from Thailand was Po, 4′, 0a, 6″,?c,?s, 5′′′, 0p and 2′′′′. Most, but not all, of these isolates could be identified morphologically as Coolia malayensis. An LSU rDNA D1/ D2 phylogenetic analysis confirmed identity of C. malayensis isolates identified morphologically. The remaining unidentified isolates fell in the C. tropicalis clade. Species of the dinoflagellate genus Gambierdiscus are the cause of Ciguatera Fish Poisoning, common in tropical areas. Nevertheless, until recently this syndrome was not reported in the NE Atlantic Ocean. A new photosynthetic dinoflagellate species, Gambierdiscus silvae sp. nov. is described based on samples taken from tide pools on rocky shores of the Canary Islands (NE Atlantic Ocean). Its morphology was studied by light and scanning electron microscopy. The new species is anterioposteriorly compressed, lenticular in shape with an epitheca slightly higher than the hypotheca. It is round in apical view and has a thick smooth theca with many scattered pores. Plate formula is Po, 4′, 0a, 6″, 6c, 6s?, 5′″, 0p, 2″″. Plate 2′ is hatchet-shaped and Plate 2″″ is very wide and the largest of the hypotheca. Phylogenies inferred from the large subunit nuclear rRNA showed that three G. silvae strains clustered in a well supported sister clade to G. polynesiensis , distinct from the other species. G. australes was observed for the first time in the Atlantic, together with G. excentricus already reported from these islands. The toxic benthic dinoflagellate genus Ostreopsis has been connected to the production of palytoxin and its analogs in many tropical and temperate areas. Although the type species, O. siamensis, was originally described from the Gulf of Thailand in 1901, little is known about the species composition and distribution of the genus Ostreopsis in Thailand. In this study, a total of 64 Ostreopsis strains isolated from the Andaman Sea as well as the Gulf of Thailand were investigated by analyzing the nucleotide sequences of the LSU rDNA D1/D2, D8/D10 and ITS-5.8S rDNA regions. Phylogenetic analyses (BI and ML) resulted in some of the strains being assigned to previously described clades, O. cf. ovata and Ostreopsis sp. 6, and revealed the existence of a novel clade named Ostreopsis sp. 7, which exhibited large genetic distances from the other clades. Among O. cf. ovata, several strains from Thailand were formed into a new subclade, the Thailand subclade, whereas a few strains belonged to the South China Sea subclade. Morphometric characteristics such as the cell sizes of the two O. cf. ovata subclades and those of Ostreopsis sp. 7 were not significantly different from each other (p > 0.05). Their characteristics were similar but slightly different from those of O. ovata and were significantly different from those of Ostreopsis sp. 6 (p < 0.05). Toxicities of Ostreopsis from Thailand were evaluated using mouse bioassay. Strains of Ostreopsis sp. 6 and Ostreopsis sp. 7 tested were highly toxic, while the two subclades of O. cf. ovata strains seemed to be nontoxic. This study suggests that toxic Ostreopsis sp. 7 is distributed in the Andaman Sea, whereas the two subclades of O. cf. ovata and toxic Ostreopsis sp. 6 are distributed in the Gulf of Thailand. The toxic benthic dinoflagellate Ostreopsis cf. ovata has considerably expanded its distribution range in the last decade, posing risks to human health. Several aspects of this species are still poorly known. We studied ultrastructural features of cultivated and natural populations of Ostreopsis cf. ovata from the Gulf of Naples (Mediterranean Sea) using confocal laser scanning, and scanning and transmission electron microscopy. New information on the morphology and location of several sulcal plates was gained and a new plate designation is suggested that better fits the one applied to other Gonyaulacales. The microtubular component of the cytoskeleton, revealed using an anti-β-tubulin antibody, consisted of a cortical layer of microtubules arranged asymmetrically in the episome and in the hyposome, complemented by a complex inner microtubular system running from the sulcal area towards the internal part of the cell. The conspicuous canal was delimited by two thick, burin-shaped lobes ending in a tubular ventral opening. The canal was surrounded by mucocysts discharging their content into it. A similar structure has been reported in other benthic and planktonic dinoflagellates and may be interpreted as an example of convergent evolution in species producing large amounts of mucus. Abstract: Ostreopsis ovata is a benthic dinoflagellate that produces palytoxin and ovatoxins. Blooms of O. ovata causing human health problems and mortality of benthic fauna have been reported from many tropical and temperate marine waters. In the present study we examined the combined effects of temperature and different nutrient conditions on the biochemical composition, growth, toxicity and carbohydrate production of an O. ovata strain originating from the Tyrrhenian Sea. O. ovata cultures with N:P ratios of 1.6, 16 and 160 (N deficient, NP sufficient and P deficient, respectively) were grown at 20°C and 30°C. Biomass accumulation, growth rates, cell volumes, biochemical composition, cell toxicity and carbohydrate production in each treatment were studied. Results indicated that under nutrient sufficiency O. ovata biomass accumulation increased significantly compared to N and P deficiency and also that N limitation severely affected growth. The highest growth rates were recorded at 30°C. Cellular contents and the atomic ratios of C, N and P were higher in the cells grown at 20°C than in those grown at 30°C. O. ovata cell volumes increased at 20°C. N deficiency significantly increased cell toxicity. Toxicity per cell was higher at 20°C, but per carbon was highest at 30°C. The highest carbohydrate production was found in conditions of N deficiency and at the lower temperature. Our study suggests that temperature increases due to global warming and nutrient enrichment of coastal waters stimulate the proliferation of O. ovata, particularly for the strains that have become adapted to warm temperate waters. Abstract: Little is known about how the growth of individual Gambierdiscus species responds to environmental factors. This study examined the effects of temperature (15–34°C), salinity (15–41) and irradiance (2–664μmolphotonsm−2 s−1) on growth of Gambierdiscus: G. australes, G. belizeanus, G. caribaeus, G. carolinianus, G. carpenteri, G. pacificus and G. ruetzleri and one putative new species, Gambierdiscus ribotype 2. Depending on species, temperatures where maximum growth occurred varied between 26.5 and 31.1°C. The upper and lower thermal limits for all species were between 31–34°C and 15–21°C, respectively. The shapes of the temperature vs. growth curves indicated that even small differences of 1–2°C notably affected growth potentials. Salinities where maximum growth occurred varied between 24.7 and 35, while the lowest salinities supporting growth ranged from <14 to 20.9. These data indicated that Gambierdiscus species are more tolerant of lower salinities than is generally appreciated. Growth of all species began to decline markedly as salinities exceed 35.1–39.4. The highest salinity tested in this study (41), however, was lethal to only one species, Gambierdiscus ribotype 2. The combined salinity data indicated that differences in salinity regimes may affect relative species abundances and distributions, particularly when salinities are <20 and >35. All eight Gambierdiscus species were adapted to relatively low light conditions, exhibiting growth maxima at 50–230μmolphotonsm−2 s−1 and requiring only 6–17μmolphotonsm−2 s−1 to maintain growth. These low light requirements indicate that Gambierdiscus growth can occur up to 150m depth in tropical waters, with optimal light regimes often extending to 75m. The combined temperature, salinity and light requirements of Gambierdiscus can be used to define latitudinal ranges and species-specific habitats, as well as to inform predictive models. This study reports some recent phylogeographical considerations on the genus Ostreopsis distribution worldwide, with particular attention to the Mediterranean Sea, and new recent advances on the quali-quantitiative detection of Ostreopsis species along coastal areas of the Mediterranean Sea based on the PCR and quantitative real time PCR (qrt-PCR) assays. It was found that O. cf. ovata is widely dispersed throughout tropical and warm temperate coastal areas. In the Atlantic/Mediterranean region it represents a panmictic population that is highly divergent from Indo-Pacific populations. Furthermore, we demonstrated that the developed qrt-PCR assay is specific, robust and high sample throughput for the quantification of the toxic O. cf. ovata in the environmental samples. SUMMARY A new genus of sand-dwelling photosynthetic dinoflagellate, Testudodinium Horiguchi, Tamura, Katsumata et A. Yamaguchi is proposed based on Testudodinium testudo (Herdman) Horiguchi, Tamura, Katsumata, et A. Yamaguchi comb. nov. (Basionym: Amphidinium testudo Herdman) and a new species in this new genus, Testudodinium maedaense Katsumata et Horiguchi sp. nov. is described. Amphidinium corrugatum is also transferred to this genus, making a new combination T. corrugatum (Larsen et Patterson) Horiguchi, Tamura et A. Yamaguchi. These three species are similar to the members of the genus Amphidinium in having an extremely small episome and a dorsoventrally flattened cell body. They are, however, distinguished from the genus Amphidinium seusu stricto by the possession of a distinct longitudinal furrow in the middle of ventral side of the episome. Phylogenetic trees based on small subunit (SSU) rDNA revealed that all three of these Testudodinium species formed a robust clade and, although statistical support is not high, the tree suggests Testudodinium clade is not closely related to Amphidinium seusu stricto clade. Abstract: Ostreopsis cf. ovata is an epiphytic/benthic dinoflagellate that produces palytoxin-like compounds (putative palytoxin, ovatoxin-a, -b, -c, d and -e). Here we report on effects of nitrogen (N) and phosphorous (P) limited conditions on cell growth, cell size, biovolume, and toxin production of an O. cf. ovata strain isolated from the Adriatic Sea (Italy). Experiments were carried out in batch cultures using nitrate (NO3 −) and phosphate (PO4 3−) as nutrient sources, and testing N:P ratios of 16, 5, 92 (control, N-limited and P-limited conditions, respectively). Residual N and P in the medium, cell yield, toxin concentrations, and toxin composition were analyzed throughout the growth. Two distinct cell size classes were identified and named Class 1 (small cells) and Class 2 (large cells), whose relative contribution under control condition was about 30 and 70%, respectively. N-limitation affected cell size, with significantly higher abundance (16%) of small cells being recorded under N stress than under control and P stress conditions. Conversely, P-limitation induced an increase of cell volume all over the growth cycle. Nutrient limitations affected growth rates and reduced final cell yields of 2.2-fold and 1.8-fold for N- and P-limited treatments vs control, respectively. Under all tested conditions O. cf. ovata showed the same qualitative profile, leading to a slight different contribution of each toxin to the total toxin content. On overall, toxins showed increasing concentrations from early to late stationary growth phase; particularly under control condition total toxin content increased from 13 to 24pgcell−1. Nutrient limitations affected toxin production, which resulted significantly lower than control in late stationary phase, especially under N-deficiency: a 53% and 40% decrease in toxin cell content was observed under N- and P-limited conditions, respectively. Abstract: Ostreopsis ovata Fukuyo is an epiphytic, toxic dinoflagellate, inhabiting tropical and sub-tropical waters worldwide and also in certain temperate waters such as the Mediterranean Sea. Toxic blooms of O. ovata have been reported in SE Brazil in 1998/99 and 2001/02 and the French–Italian Riviera in 2005 and 2006. These blooms had negative effects on human health and aquatic life. Chemical analyses have indicated that O. ovata cells produce palytoxin, a very strong toxin, only second in toxicity to botulism. Increase in water temperature by several degrees has been suggested as the reason for triggering these blooms. Four laboratory experiments were performed with O. ovata isolated from Tyrrhenian Sea, Italy to determine the effects of water temperature and co-occurring algae on the cell growth and/or the toxicity of O. ovata. The cells were grown under different temperatures ranging from 16°C to 30°C, and cell densities, growth rates and the cell toxicities were studied. Results indicated high water temperatures (26–30°C) increased the growth rate and biomass accumulation of O. ovata. In mixed cultures of O. ovata with other co-occurring algae, biomass decreased due to grazing by ciliates. Cell toxicity on the other hand was highest at lower temperatures, i.e., between 20 and 22°C. The present study suggests that sea surface temperature increases resulted by global warming could play a crucial role inducing the geographical expansion and biomass accumulation by blooms of O. ovata. Aim Ostreopsis is a benthic and epiphytic dinoflagellate producing potent toxins widespread in tropical and warm temperate coastal areas world-wide. We tested the hypothesis that as it is benthic, it would show distinct biogeographical patterns in comparison with planktonic species. Here, we analyse sequence variability in ribosomal DNA markers to provide the first phylogeographical study of this toxic benthic dinoflagellate. Location Mediterranean Sea, Atlantic Ocean, Pacific Ocean. Methods Ribosomal DNA sequence data from partial nuclear LSU (D1/D2 domains) and 5.8S genes and non-coding internal transcribed spacer (ITS) regions were obtained from 82 isolates of Ostreopsis species, collected at 26 localities throughout the world. Molecular sequence data were analysed using maximum parsimony, maximum likelihood and Bayesian methods for phylogenetic inference. A statistical parsimony network was obtained based on concatenated LSU and 5.8S rDNA–ITS region sequences of the Mediterranean/Atlantic Ostreopsis cf. ovata isolates to infer haplotype distribution over their geographical range. Light epifluorescence microscopy analyses were performed on cultured and field Ostreopsis material for taxonomic identification, while laboratory experiments for encystment induction were carried out on selected O. cf. ovata isolates. Toxin assays of Ostreopsis species isolates were carried out using the haemolytic-based method. Results Analyses based on single and concatenated ribosomal genes gave substantially similar results. The rDNA phylogeny revealed different clades corresponding to different species within the genus Ostreopsis. In the species O. cf. ovata, different genetic lineages were correlated with macrogeographical distribution. A network of haplotypes inferred from the Atlantic and Mediterranean isolates of O. cf. ovata revealed that these two areas might host a single panmictic population. The Atlantic/Mediterranean population of O. cf. ovata was differentiated considerably from the Indo-Pacific populations. Other species of Ostreopsis were found, but they turned out to be restricted to just one of the two main warm-water oceanic basins, the Mediterranean/Atlantic and the Indo-Pacific. Main conclusions Ostreopsis cf. ovata was found to be widely dispersed throughout the coastal areas of tropical and some warm temperate seas. In the Atlantic/Mediterranean region it may constitute a panmictic population that is highly distinct from Indo-Pacific populations. Ostreopsis cf. siamensis was found only in the Mediterranean Sea, and strains identified as Ostreopsis lenticularis and Ostreopsis labens were found only in the Indo-Pacific region. Ciguatera poisoning is mainly caused by the consumption of reef fish that have accumulated ciguatoxins (CTXs) produced by the benthic dinoflagellates Gambierdiscus and Fukuyoa. China has a long history of problems with ciguatera, but research on ciguatera causative organisms is very limited, especially in the Beibu Gulf, where coral reefs have been degraded significantly and CTXs in reef fish have exceeded food safety guidelines. Here, five strains of Gambierdiscus spp. were collected from Weizhou Island, a ciguatera hotspot in the Beibu Gulf, and identified by light and scanning electron microscopy and phylogenetic analyses based on large and small subunit rDNA sequences. Strains showed typical morphological characteristics of Gambierdiscus caribaeus, exhibiting a smooth thecal surface, rectangular-shaped 2′, almost symmetric 4″, and a large and broad posterior intercalary plate. They clustered in the phylogenetic tree with G. caribaeus from other locations. Therefore, these five strains belonged to G. caribaeus, a globally distributed Gambierdiscus species. Toxicity was determined through the mouse neuroblastoma assay and ranged from 0 to 5.40 fg CTX3C eq cell−1. The low level of toxicity of G. caribaeus in Weizhou Island, with CTX-contaminated fish above the regulatory level in the previous study, suggests that the long-term presence of low toxicity G. caribaeus might lead to the bioaccumulation of CTXs in fish, which can reach dangerous CTX levels. Alternatively, other highly-toxic, non-sampled strains could be present in these waters. This is the first report on toxic Gambierdiscus from the Beibu Gulf and Chinese waters and will provide a basis for further research determining effective strategies for ciguatera management in the area. This study indicates that bilaterally flattened, armored, benthic dinoflagellates are more diverse in morphology than previously known. A new species, Plagiodinium belizeanum Faust et Balech gen. et sp. nov., is described in floating detritus from Twin Cays, Belize, mangrove habitats. Plagiodinium belizeanum cells are small, with dimensions of 26.5-30.5 μm in length, 20-24.5 ???m in width, and 6.5-8.5 μm in depth. Cells are ablong and bilaterally compressed with a posteriorly located, spherical nucleus, many chloroplasts, and spherical starch granules. The epitheca descends ventrally, is cap-shaped, and is composed of five plates and a very small platelet provisionally named P??? situated in the center. The epitheca is narrowly oval in opical view with a pointed truncated ventral side and a rounded dorsal side. The cingulum is composed of five plates. The hypotheca is constructed of five posteriorly elongated postcingular plates and one antapical plate. The sulcus is very short and narrow, comprised of five small plates. The thecal plate arrangement of P. belizeanum is P[SUB0] 5', O′, 5C, 1′′, 5S. No lists are present. Thecal plates have a smooth surface with small and irregularly scattered pores. The intercalary bond is smooth on the outer cell surface and broadly striated on its inner surface. We conclude that P. belizeanum represents a new, benthic, peridinioid, armored genus, Plagiodinium gen. nov. Ciguatera fish poisoning (CFP) is prevalent around the tropical and sub-tropical latitudes of the world and impacts many Pacific island communities intrinsically linked to the reef system for sustenance and trade. While the genus Gambierdiscus has been linked with CFP, it is commonly found on tropical reef systems in microalgal assemblages with other genera of toxin-producing, epiphytic and/or benthic dinoflagellates – Amphidinium, Coolia, Fukuyoa, Ostreopsis and Prorocentrum. Identifying a biomarker compound that can be used for the early detection of Gambierdiscus blooms, specifically in a mixed microalgal community, is paramount in enabling the development of management and mitigation strategies. Following on from the recent structural elucidation of 44-methylgambierone, its potential to contribute to CFP intoxication events and applicability as a biomarker compound for Gambierdiscus spp. was investigated. The acute toxicity of this secondary metabolite was determined by intraperitoneal injection using mice, which showed it to be of low toxicity, with an LD 50 between 20 and 38 mg kg−1. The production of 44-methylgambierone by 252 marine microalgal isolates consisting of 90 species from 32 genera across seven classes, was assessed by liquid chromatography-tandem mass spectrometry. It was discovered that the production of this secondary metabolite was ubiquitous to the eight Gambierdiscus species tested, however not all isolates of G. carpenteri , and some species/isolates of Coolia and Fukuyoa. l Ostreopsis sp. 1 and sp. 6 were the only Ostreopsis species found in Korea. l The significant genetic differentiation of a total of 141 Ostreopsis sp.1 isolates from Korea was detected. l Only the Jeju Island Ostreopsis sp. 1 populations showed a signal in agreement with population equilibrium. Species belonging to the toxic dinoflagellate genus Ostreopsis are widespread, occurring from tropical to temperate waters. As mainly benthic/epiphytic species, they would be expected to show distinct geographical patterns. In this study, ribosomal DNA (rDNA) sequences from partial nuclear LSU D8-D10, 5.8S, and ITS regions were determined for 169 isolates of Ostreopsis species collected from three coastal sites (i.e., Jeju Island, Chuja Island, and Pohang) within Korea. The phylogenetic tree inferred from the LSU rDNA D8-D10 sequences showed that Korean Ostreopsis species corresponded to either Ostreopsis sp. 1 or sp. 6, with Ostreopsis sp. 1 being relatively predominant regarding their distribution. While Ostreopsis sp. 1 occurred throughout all the three sampling sites within Korea, Ostreopsis sp. 6 was confined to the northern part of Jeju Island. When further investigated, the genetic diversity of Ostreopsis sp. 1 in Korea based on ITS sequences showed a total of 21 haplotypes. The presumed ancestral haplotype H3, was also present in the Japanese and Russian populations of Ostreopsis sp. 1. Although the overall demographic history of all the Korean populations of Ostreopsis sp. 1 could not be clearly identified, probably due to a mixture of different regional demographic patterns within Korea, each Ostreopsis sp. 1 population showed a characteristic demographic pattern at a regional scale. While the Jeju Island Ostreopsis sp. 1 population showed a signal in agreement with population equilibrium, the Chuja Island and Pohang Ostreopsis sp. 1 populations showed distribution patterns that are expected in a sudden population expansion model. The results from this study provide a basis for a better understanding of the distribution and genetic structure of the Asian Ostreopsis sp. 1 populations.
