A characteristic feature of Alzheimer's disease (AD) is the formation of neuronal extracellular senile plaques composed of aggregates of fibrillar amyloid β (Aβ) peptides, with the Aβ1-42 peptide being the most abundant species. These Aβ peptides have been proposed to contribute to the pathophysiology of the disease; however, there are few tools available to test this hypothesis directly. In particular, there are no data that establish a dose–response relationship between Aβ peptide expression level and disease. We have generated a panel of transgenic Caenorhabditis elegans strains expressing the human Aβ1-42 peptide under the control of promoter regions of two pan-neuronal expressed genes, snb-1 and rgef-1. Phenotypic data show strong age-related defects in motility, subtle changes in chemotaxis, reduced median and maximum lifespan, changes in health span indicators, and impaired learning. The Aβ1-42 expression level of these strains differed as a function of promoter identity and transgene copy number, and the timing and severity of phenotypes mediated by Aβ1-42 were strongly positively correlated with expression level. The pan-neuronal expression of varying levels of human Aβ1-42 in a nematode model provides a new tool to investigate the in vivo toxicity of neuronal Aβ expression and the molecular and cellular mechanisms underlying AD progression in the absence of endogenous Aβ peptides. More importantly, it allows direct quantitative testing of the dose–response relationship between neuronal Aβ peptide expression and disease for the first time. The rice moth, Corcyra cephalonica (Stainton) (Lepidoptera: Pyralidae), is a serious pest of grains in storage systems resulting in immense losses but is also widely used as a factitious host for mass rearing of many important natural enemies of crop pests. Given the role of kairomones, the aim of this study was to isolate and identify potential cues from the larval body wash of C. cephalonica, which could attract its gregarious ectoparasitoid, Habrobracon hebetor (Say) (Hymenoptera: Braconidae). Gas chromatography with electroantennography (GC-EAG) and olfactory assays were used to demonstrate the attraction of female H. hebetor to different larval body volatiles. A total of 15 EAG-active compounds were discovered in the body wash of C. cephalonica larvae that triggered a response in female H. hebetor. Among them, four compounds (p-xylene, naphthalene, n-eicosane and n-tricosane) were bioassayed for the behavioural response of parasitoids and found that n-eicosane significantly attracted a higher number of parasitoids than others. Our work establishes the attraction of H. hebetor to volatile kairomone cues emanating from the factitious host larval body, which offers an opportunity for its parasitoid, H. hebetor to improve the mass rearing efficiency. It has been estimated that between 40 and 60 % of the assimilated carbon is diverted to the roots and released in the rhizosphere in form of root exudates. Root exudates thus define a complex mixture of low and high molecular weight compounds, including carbohydrates, amino acids, organic, and proteins, but also a broad spectrum of specialized molecules, such as flavonoids, glucosinolates, terpenoids, or alkaloids. Root exudates favour soil mineral nutrition, can bind to soil aggregate and in turn modify soil physico-chemical properties, but also mediate plant-plant, plant-microbe, and plant-animal interactions belowground. With this review, we aim to highlight how chemical ecologists have approached the study of root exudates-mediated interactions between plants and their biotic and abiotic surroundings. We do so by presenting a series of study cases for, on one hand, showcasing different methodologies that have been developed to test the activity of different root exudates, and, on the other hand, to show the broad array of interactions mediated by root exudates. Ultimately, we aim to spur further research and collaborations between chemists and ecologists studying belowground chemically-mediated interactions, so as to tackle essential challenges in terms of food security and climate change in the near future.

• Behavioural tests using the CPZ model focus mainly on motor, anxiety and cognition but not other common clinical symptoms in MS like pain or fatigue. • Most histological correlations associated with behavioural deficits were found to be inappropriate. • CPZ-feeding alone may be a useful tool to model some behavioural aspects of demyelinating diseases but it should not be regarded as a model of MS per se. The feeding of cuprizone (CPZ) to animals has been extensively used to model the processes of demyelination and remyelination, with many papers adopting a narrative linked to demyelinating conditions like multiple sclerosis (MS), the aetiology of which is unknown. However, no current animal model faithfully replicates the myriad of symptoms seen in the clinical condition of MS. CPZ ingestion causes mitochondrial and endoplasmic reticulum stress and subsequent apoptosis of oligodendrocytes leads to central nervous system demyelination and glial cell activation. Although there are a wide variety of behavioural tests available for characterizing the functional deficits in animal models of disease, including that of CPZ-induced deficits, they have focused on a narrow subset of outcomes such as motor performance, cognition, and anxiety. The literature has not been systematically reviewed in relation to these or other symptoms associated with clinical MS. This paper reviews these tests and makes recommendations as to which are the most important in order to better understand the role of this model in examining aspects of demyelinating diseases like MS. The cowpea aphid, Aphis craccivora, is a major insect pest of cowpea in Africa. Volatile organic compounds (VOCs) mediate plant–arthropod interactions that could be used in the management of insect pests. In this study, we established the VOC profile involved in the interaction between A. craccivora and four cowpea cultivars, namely Ex‐Luanda, Katumani 80, Machakos 66 and Ken Kunde 1. Behavioural assays were conducted to study host preference and gas chromatography‐mass spectrometry (GC/MS) for chemical analysis of volatiles. In preference assays, alate A. craccivora had no significant preference for any of the four cowpea cultivars tested. However, in the olfactometer assays, the aphids showed a significant preference for odours from cultivar Ex‐Luanda compared to Katumani 80. Machakos 66 and Ken Kunde 1 elicited neutral responses. In pairwise comparisons, alate A. craccivora did not distinguish between odours of respective cowpea cultivars. GC/MS analysis identified 23 compounds in the volatiles of the four cowpea cultivars. Not all compounds were detected in all cowpea cultivars, and the detected compounds amounts varied in each cultivar. Of these, only four compounds (hexanal, (E)‐2‐hexenal, 1‐octen‐3‐ol and p‐xylene) were emitted in significantly different quantities in the four cultivars. A blend of hexanal and (E)‐2‐hexenal added to cowpea cultivar Ex‐Luanda decreased its attractiveness to A. craccivora compared to the control. Our findings showed differential attractiveness of VOCs of cowpea cultivars to A. craccivora, suggesting that VOCs could be used in the management of A. craccivora. [ABSTRACT FROM AUTHOR]

Consistent inter-individual differences in behaviour have been previously reported in adult shelter cats. In this study, we aimed to assess whether repeatable individual differences in behaviours exhibited by shelter cats in different situations were interrelated, forming behavioural syndromes. We tested 31 adult cats in five different behavioural tests, repeated three times each: a struggle test where an experimenter restrained the cat, a separation/confinement test where the cat spent 2 min in a pet carrier, a mouse test where the cat was presented with a live mouse in a jar, and two tests where the cat reacted to an unfamiliar human who remained either passive or actively approached the cat. Individual differences in behaviour were consistent (repeatable) across repeated trials for each of the tests. We also found associations between some of the behaviours shown in the different tests, several of which appeared to be due to differences in human-oriented behaviours. This study is the first to assess the presence of behavioural syndromes using repeated behavioural tests in different situations common in the daily life of a cat, and which may prove useful in improving the match between prospective owner and cat in shelter adoption programmes.

BACKGROUND: The response of virgin females of the legume pest Acanthoscelides obtectus (Coleoptera: Bruchidae) to headspace extracts of volatiles collected from flowers of a nectar plant, Daucus carota, was investigated using behaviour (four‐arm olfactometry) and coupled gas chromatography–electroantennography (GC‐EAG). RESULTS: Odours from inflorescences were significantly more attractive to virgin female beetles than clean air. Similarly, a sample of volatile organic compounds (VOCs) collected by air entrainment (dynamic headspace collection) was more attractive to beetles than a solvent control. In coupled GC‐EAG experiments with beetle antennae and the VOC extract, six components showed EAG activity. Using coupled GC–mass spectrometry (GC–MS) and GC peak enhancement with authentic standards, the components were identified as α‐pinene (S:R 16:1), sabinene, myrcene, limonene (S:R 1:3), terpinolene and (S)‐bornyl acetate. Females preferred the synthetic blend of D. carota EAG‐active volatiles to the solvent control in bioassays. When compared directly, odours of D. carota inflorescences elicited stronger positive behaviour than the synthetic blend. CONCLUSION: This is the first report of behaviourally active volatiles linked to pollen location for A. obtectus, and development of the six‐component blend is being pursued, which could underpin the design of semiochemical‐based field management approaches against this major pest of stored products. © 2018 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Animal personality has become a major topic in animal behaviour. Much recent progress has come from the use of the open-field test, which is routinely used to separate individuals into fast and slow explorers. However, there is no standard way to conduct the test and it is unclear whether the test really measures exploration. Here, we addressed these issues by combining an open-field test with a battery of independent assays intended to assess the convergent and discriminant validity of the behavioural traits suggested to measure exploration in the open-field test. Our study subjects were common mynas, Acridotheres tristis , introduced to Australia. The analyses confirmed that the open-field test allows individuals to be separated according to their propensity to explore, mainly through metrics related to spatial and object examination of the novel cage. However, other metrics classically used to describe exploratory behaviour, such as the latency to enter the novel space, reflected shyness rather than exploration. The open-field test can therefore be a powerful tool to investigate personality, but only if the biological meanings of the metrics derived from the test are properly validated with independent behavioural assays. Municipal wastewater treatment plant (WWTP) effluent is a substantial source of pollution in aquatic habitats that can impact organisms across multiple levels of biological organization. Even though wastewater effluent is discharged continuously all year long, its impacts across seasons, specifically during winter, have largely been neglected in ecotoxicological research. Seasonal differences are of particular interest, as temperature-driven metabolic changes in aquatic organisms can significantly alter their ability to respond to chemical stressors. In this study, we examined the effects of multiple levels of wastewater effluent exposure (0, 25, or 50% treated effluent) on the physiological and behavioural responses of adult fathead minnow (Pimephales promelas) at temperatures simulating either summer (20 °C) or winter (4 °C) conditions. At 20 °C, wastewater exposure posed a metabolic cost to fish, demonstrated by higher standard metabolic rate and was associated with increased haematocrit and a reduction in boldness. In contrast, fish exposed to wastewater at 4 °C experienced no change in metabolic rate but performed fewer social interactions with their conspecifics. Taken together, our results demonstrate that wastewater exposure can lead to metabolic and behavioural disruptions, and such disruptions vary in magnitude and direction depending on temperature. Our findings highlight the importance of studying the interactions between stressors, while also underscoring the importance of research during colder periods of the year to broaden and deepen our understanding of the impacts of wastewater contamination in aquatic ecosystems. Abstract: In this study, we examined variation in stress-responsiveness between 10 full sibling families of Atlantic salmon from a commercial breeding programme. Behavioural observations (locomotor activity during acute confinement and feeding response after placement in a novel environment) and cortisol release into the water were used as non-invasive markers of stress-responsiveness. Locomotor activity during acute stress was highly correlated with cortisol release rate, while feeding response did not correlate with either of the other two markers. Locomotor activity and cortisol release rate differed significantly between families, whereas feeding response did not. These results provide further support for hypothalamus-pituitary-interrenal responsiveness being a heritable (genetic) trait. Furthermore, results from an infectious pancreas necrosis virus (IPNV) challenge (carried out within the breeding programme from which our experimental fish originated) indicated a positive correlation between stress-responsiveness and mortality at the family level. In summary, the behavioural measure of locomotor activity during acute confinement provides a simple alternative measure of stress-responsiveness to physiological measures of cortisol, and families characterised by high stress-responsiveness showed increased susceptibility to IPNV. Abstract: The present study was conducted to clarify the interference of selenomethionine (SeMet) on methylmercury (MeHg) toxicity through the evaluation of changes in biomarkers of exposure and effect in rats exposed to MeHg and co-exposed to MeHg and SeMet. Male Wistar rats received two intraperitoneally (i.p.) administrations, either MeHg (1.5mg/kg body weight), SeMet alone (1mg/kg body weight) or combined MeHg and SeMet, followed by 3 weeks of rat urine collection and neurobehavioural assays. The effects of different administrations were investigated by the quantification of total mercury in kidney and brain, analysis of urinary porphyrins, determination of hepatic GSH and evaluation of motor activity functions (rearing and ambulation). MeHg exposure resulted in a significant increase of urinary porphyrins during the 3 weeks of rat urine collection, where as it caused a significant decrease in motor activity only at the first day after cessation of rat exposure. Additionally, SeMet co-exposure was able to normalize the porphyrins excretion, and a tendency to restore rat motor activity was observed, on the first day after cessation of exposure. Brain and kidney mercury levels increased significantly in rats exposed to MeHg; however, in co-exposed rats to SeMet no significant changes in Hg levels were found as compared to rats exposed to MeHg alone. Hence, the present study shows that urinary porphyrins are sensitive and persistent indicators of MeHg toxicity and demonstrates for the first time that SeMet reduces its formation. Finally, these results confirm that the mechanism of interaction between SeMet and MeHg cannot be explained by the reduction of Hg levels in target organs and suggestions are made to clarify the interference of SeMet on MeHg toxicity. Simple Summary: An important activity of modern animal shelters is the development of successful adoption programmes. In this regard, there is a need for reliable tests of individual differences in behaviour to help match the "personality" of potential adoptees with the lifestyle and needs of prospective owners; a companion animal for an elderly person remaining at home requires a different match than a pet for someone who will be away most of the day; a pet kept exclusively indoors in a small apartment requires a different match than an indoor/outdoor pet. In the present study, we repeatedly tested 31 mixed-breed adult cats of both sexes and a wide range of ages in five behavioural tests at a shelter in Mexico City, Mexico. The tests were designed to be easily implemented by shelter staff, and were short and low cost and intended to simulate common situations in a pet cat's everyday life. We found consistent (stable) individual differences in the cats' behaviour on all five tests, as well as correlations between their behaviour across tests. This suggests that such tests may contribute to reliably characterizing the "personality" of individual cats and so help increase the rate of successful adoptions.
