This is the third paper in the series Catalogue of fungi in China that aims to systematically and promptly publish Chinese fungal species. In this paper of the series, we focus on macrofungi from the southern border area of Xizang, a previously less concerned region. A total of 15 new species in six orders, 11 families, and 12 genera from Agaricomycetes, Basidiomycota are described. Both morphological and phylogenetic analyses support the identity of these new species and their taxonomic placements. In addition, one new combination Trechispora cryptomerioides is proposed. We hope that the third paper in the series Catalogue of fungi in China will draw more attention to reporting Chinese fungal diversity in the border areas of Xizang and other special geographic regions besides the aim of this series itself. Basidiomycetes with a wide variety of skeletons of secondary metabolites can be expected to be the source of new interesting biological compounds. During our research on basidiomycetes, two new C-29 oxygenated oleanane-type triterpenes (1 and 2) and torulosacid (3), a muurolene type sesquiterpenoid with a five-membered ether ring along with nine known compounds (4–12), were isolated from the MeOH extract of the fruiting bodies of Fuscoporia torulosa. The structures of 1–3 were determined by NMR and HREIMS analysis. Further studies on the stereochemistry of 3 were conducted using X-ray crystallographic analysis and comparison of experimental and calculated ECD spectra. In the antimicrobial assay of isolates, 1, 7, and 9 showed growth inhibitory activity against methicillin-resistant Staphylococcus aureus and other gram-positive strains. Isolation of oleanane type triterpenes from fungi including basidiomycetes, is a unique report that could lead to further isolation of new compounds and the discovery of unique biosynthetic enzymes. Medicinal macrofungi, like prized plants and animals, have dietary, therapeutic and commercial significance. This knowledge, anchored in the traditional healthcare system, warrants the identification of the most prominent species harbouring novel natural products. A traditional ethnobiological approach was adopted to collect ethnomycological data by interviewing 110 informants (95 men and 15 women) inhabiting 10 settlements in Bannu District, Pakistan. Visiting the remote nomadic sites revealed the most unique data of its type. This data was assessed and correlated with previous studies, comparing the standard numerical indices: informant consensus factor, fidelity level, use value, relative frequency of citation and Jaccard index. Tradition medicinal uses of 30 species of wild macrofungi (belonging to 19 genera and 12 families) are documented here. Podaxis pistillaris macrofungi was preference ranked with priority therapeutic uses, medicinally for stomach problems, as an antipyretic, for erotic uses, as an immune booster and as an antidiabetic; it is also nutritionally significant. Temperature and humidity are key factors for the growth of macrofungi generally in Pakistan (in spring and during the monsoon season), while the dynamic frequency of rain in the rainy season due to the impact of climate change cannot be neglected. Indigenous communities divided the ethnomycological potential of macrofungi into three main clusters: (a) edible macrofungi, for treating various disorders and as nutritional supplements; (b) non-edible macrofungi, medicinally important or just poisonous; (c) unknown or unused, without any known utility. Importantly, eight novel species with traditional uses are reported here, and the present work is the first ethnomycological study of macrofungi from Pakistan. This research aims to trace the traditional knowledge related to the naturally occurring medicinal macrofungi in the Bannu District, Khyber Pakhtunkhwa, Pakistan. Documentation of new medicinal species and novel uses of existing species reflect a healthy relationship between inhabitants of the Bannu District and wild medicinal macrofungi. Invasive fungal diseases are a major threat to human health, resulting in more than 1.5 million annual deaths worldwide. The arsenal of antifungal therapeutics remains limited and is in dire need of drugs that target additional biosynthetic pathways that are absent from humans. One such pathway involves the biosynthesis of trehalose. Trehalose is a disaccharide that is required for pathogenic fungi to survive in their human hosts. In the first step of trehalose biosynthesis, trehalose-6-phosphate synthase (Tps1) converts UDP-glucose and glucose-6-phosphate to trehalose-6-phosphate. Here, we report the structures of full-length Cryptococcus neoformans Tps1 (CnTps1) in unliganded form and in complex with uridine diphosphate and glucose-6-phosphate. Comparison of these two structures reveals significant movement toward the catalytic pocket by the N terminus upon ligand binding and identifies residues required for substrate binding, as well as residues that stabilize the tetramer. Intriguingly, an intrinsically disordered domain (IDD), which is conserved among Cryptococcal species and closely related basidiomycetes, extends from each subunit of the tetramer into the "solvent" but is not visible in density maps. We determined that the IDD is not required for C. neoformans Tps1-dependent thermotolerance and osmotic stress survival. Studies with UDP-galactose highlight the exquisite substrate specificity of CnTps1. In toto, these studies expand our knowledge of trehalose biosynthesis in Cryptococcus and highlight the potential of developing antifungal therapeutics that disrupt the synthesis of this disaccharide or the formation of a functional tetramer and the use of cryo-EM in the structural characterization of CnTps1-ligand/drug complexes. Entoloma is a relatively large genus in Agaricales, with a rich diversity of species and a wide distribution. In this study, seven new species of Entoloma belonging to the subgenus Cubospora have been identified based on morphological and phylogenetic evidence from subtropical regions of China. Morphologically, E. excavatum is characterized by the yellow, depressed, estriate pileus and medium-sized basidiospores; E. lacticolor is recognized by the white and papillate pileus, adnexed lamellae, and presence of clamp connections; E. phlebophyllum is identified by the pink-to-maroon and estriate pileus, and lamellae with lateral veins; E. rufomarginatum differs from other cuboid-spored species by the lamellae edge which is red-brown-underlined; E. subcycneum is characterized by the white pileus and carneogriseum-type cheilocystidia; E. submurrayi is recognized by the pileus margin exceeding the lamellae, 2-layered pileipellis with hyphae of different widths, and the presence of clamp connections; E. tomentosum is identified by the tomentose pileus, heterogeneous lamella edge, and versiform cheilocystidia with brown-yellow contents. Their distinct taxonomic status is confirmed by the positions of the seven new species in both the ITS + LSU and 3-locus (LSU, tef-1α, rpb2) phylogenetic trees. A 10-year-old spayed mixed breed dog presented with severe neurological signs. Computed tomography revealed a cranial mediastinal mass, osteolysis of the right second rib and second thoracic vertebra, tracheobronchial and mesenteric lymph node enlargement, pneumonia and pleural effusion. Magnetic resonance imaging detected lesions in the white matter of the right frontal lobe and left cerebral hemisphere with contrast-enhanced T1-weighted images showing demarcated enhancement. On cut section, the surface of the right cerebral frontal lobe and left cerebral hemisphere corticomedullary junctions were indistinct and the white matter was discoloured. Microscopically, multicentric granulomatous inflammation was seen in the brain, cranial mediastinal mass, masses on the right second rib, tracheobronchial and mesenteric lymph nodes, heart, kidneys, lungs and oesophagus. Necrosis and hyaline fungal structures were frequently observed in the centre of the granulomas. These fungi had septae, Y-shaped branching and were 2–3 μm in width. Sequence analysis of DNA from formalin-fixed paraffin-embedded samples identified the fungi as Schizophyllum commune. Based on these findings, this case was diagnosed as disseminated S. commune infection. This is the first report of granulomatous encephalitis caused by S. commune in a dog. [ABSTRACT FROM AUTHOR]

Macrofungi commonly referred to as Mushrooms are distributed worldwide and well known for their nutritional, medicinal, and organoleptic properties. Strain improvement in mushrooms is lagging due to paucity of efficient genome modification techniques. Thus, for advanced developments in research and commercial or economical viability and benefit, CRISPR/Cas9 (clustered regularly interspaced short palindromic repeats/CRISPR-associated nuclease 9) emerged as an efficient genome editing tool. The higher efficiency and precision of the desired genetic modification(s) are the most valuable attributes of this recent technology. The present review comprehensively summarizes various conventional methods utilized for strain improvement in mushrooms including hybridization, protoplast fusion, and di-mon mating. Furthermore, the problems associated with these techniques have been discussed besides providing the potential recluses. The significance of CRISPR/Cas9 strategies employed for improvement in various mushroom genera has been deliberated, as these strategies will paves the way forward for obtaining improved strain and effective cultivation methods for enhancing the yield and quality of the fruit bodies. DNA barcodes can provide accurate identification of plants. We used previously reported DNA primers targeting the internal transcribed spacer (ITS1) region of the nuclear ribosomal cistron, internal transcribed spacer (ITS2), and chloroplast trnL (UAA) intron to identify four trees at Bergen Community College. Two of the four trees were identified as Acer rubrum and Fagus sylvatica. However, Quercus was only identified at the genus level, and the fourth tree did not show similar identification between barcodes. Next-generation sequencing of 16S rRNA genes showed that the predominant bacterial communities in the rhizosphere mainly consisted of the Pseudomonadota, Actinomycetota, Bacteroidota, and Acidobacteriota. A. rubrum showed the most diverse bacterial community while F. sylvatica was less diverse. The genus Rhodoplanes showed the highest relative bacterial abundance in all trees. Fungal ITS sequence analysis demonstrated that the communities predominantly consisted of the Ascomycota and Basidiomycota. Quercus showed the highest fungi diversity while F. sylvatica showed the lowest. Russula showed the highest abundance of fungi genera. Average similarity values in the rhizosphere for fungi communities at the phylum level were higher than for bacteria. However, at the genus level, bacterial communities showed higher similarities than fungi. Similarity values decreased at lower taxonomical levels for both bacteria and fungi, indicating each tree has selected for specific bacterial and fungal communities. This study confirmed the distinctiveness of the microbial communities in the rhizosphere of each tree and their importance in sustaining and supporting viability and growth but also demonstrating the limitations of DNA barcoding with the primers used in this study to identify genus and species for some of the trees. Coriolopsis trogii is a basidiomycete fungus which utilizes a large array of lignin-modifying enzymes to colonize and decompose dead wood. Its extracellular enzymatic arsenal includes laccases, i.e., polyphenol oxidases of relevant interest for different industrial applications thanks to their ability to oxidize a diverse range of natural and synthetic compounds. In this work, the production of laccases in C. trogii MUT3379 was explored and improved. From an initial production of ca. 10,000 U L−1, the fermentation process was gradually optimized, reaching a final yield of ca. 200,000 U L−1. An SDS-PAGE analysis of the secretome highlighted the presence of a main protein of ca. 60 kDa showing laccase activity, which was designated as Lac3379-1 once its primary sequence was established by tandem mass spectrometry. The characterization of Lac3379-1 revealed a remarkable enzymatic stability in the presence of surfactants and solvents and a diversified activity on a broad range of substrates, positioning it as an interesting tool for diverse biotechnological applications. The high-yield and robust production process indicates C. trogii MUT3379 as a promising cell factory for laccases, offering new perspectives for industrial applications of lignin-modifying enzymes. Cordyceps militaris and Lentinula edodes are known for their high extracellular proteolytic, amylolytic, and laccase activity, which is important for bakery production. This article describes the effect of enzymes obtained from mycelial biomass of C. militaris and L. edodes on such properties of flour mixes as amylolytic and proteolytic enzymic activity during baking, technological costs, and bread quality. The research featured strains of C. militaris SRG4 and L. edodes 3790 fungi; mycelial biomass powder of these fungi on a sterile grain substrate (rice, wheat); experimental flour mixes of wheat bread and first-grade baking flour with mycelial biomass powder; bread made from the experimental flour mixes. The experimental part included standard methods used in the bakery industry. The mycelial biomass of C. militaris and L. edodes fungi proved to be a promising baking additive because it possessed hydrolytic enzymes and was rich in protein substances (32.2 ± 1.5 and 26.4 ± 2.0%, respectively), specific mushroom polysaccharides (36.7 ± 0.8 and 52.2 ± 1.2%, respectively), and carotenoids (1600 ± 40 mcg/g of C. militaris biomass). The active amylases and proteinases in the mycelial biomass powder improved the sugar-forming ability and reduced the falling number. In addition, they raised the amount of washed gluten with a slight decrease in elasticity, as well as provided an acceptable structure and viscosity of the dough at 1–4% of mycelial biomass powder. This dose resulted in an optimal bread formulation of standard quality. A bigger amount caused the crumb to darken and increased its humidity while reducing the specific volume. The bread samples with C. militaris were too acid. Mycelial biomass of C. militaris and L. edodes proved to be a good source of enzymes to be used with grain substrates in bakery production. Further research is needed to define the optimal dose and processing modes.

Pythium insidiosum is an important oomycete pathogen of mammals that causes pythiosis, an endemic disease in warm climates that stands out for its unfavorable prognosis, lethality in the affected species, and difficulties in treatment. This study evaluated in vitro anti-P. insidiosum potential of aqueous, hydroethanolic, and ethanolic extracts of indigenous wild mushrooms from southern Brazil. The extracts were prepared from Amanita gemmata, Amanita muscaria, Auricularia auricula, Gymnopilus junonius, Lactarius deliciosus, Laccaria laccata, Psilocybe cubensis, and Russula xerampelina. In vitro susceptibility assays employed the microdilution technique according to the M38-A2 protocol CLSI. The hydroethanolic and ethanolic extracts of R. xerampelina showed anti-P. insidiosum activity at minimum inhibitory concentrations ranging from 1.87--7.50 mg/mL. The other mushroom species extracts showed no inhibitory effects on growth of P. insidiosum. This is the first study to evaluate the antimicrobial activity of mushrooms on oomycetes, evidencing the antimicrobial potential of R. xerampelina on the pathogen P. insidiosum. So, the present study expands new perspectives, since the secondary metabolites produced by mushrooms can be potential targets for the development of new categories of medicines. However, considering the wide biodiversity of Brazilian mushrooms, we suggested that the search for other basidiomycetes species with anti-P. insidiosum action needs to be expanded. Molecular techniques for studying soil microorganisms allow to quantify the presence of specific functional groups from the total soil DNA and thus to assess their dynamic in response to the land use. Using this approach, a study was carried out to assess the increase the soil organic carbon (C) of arable land in the central Po Valley with three woody species (white willow, black locust, hybrid poplar) in short rotation forestry. Being fungi the most involved microorganisms in the C-cycle and representing the larger part of microbial biomass in volume, total fungi e the two most widespread main fungal degraders in the upper soil layer (Ascomycetes and Basidiomycetes) were used as indicators of the effectiveness of land use in increasing soil organic C. An innovative molecular technique (SYBR® Green I dye on the QuantStudio™ 3D digital PCR system) which allows to overcome the critical issues associated with the relatively low concentration of fungal DNA compared to that of bacteria was used. Compared to the ploughed soil, total organic carbon (TOC), microbial biomass and enzyme activities increased significantly in the woody plantations. However, TOC increased to different degrees among the woody species. White willow and black locust showed a greater increase in soil TOC than arable (+30 and 20% respectively), followed by hybrid poplar with a smaller increase of +12%. The increases in soil TOC in the three plantations were not correlated with the total woody mass, assessed 15 years after abandonment. Total fungi and Ascomycetes were highly correlated with microbial biomass and TOC, while Basidiomycetes were not. Ascomycetes, which commonly prevail in agricultural soils, were still the most abundant even after a short rotation forest cycle of 15 years and can be taken as good indicator of C sequestration activities into the soil. Soil tillage strongly reduced the content of Basidiomycetes and short rotation forestry managed to increase it at least 4-5 times over a 15-year cycle of short rotation forestry, suggesting that it can act as a qualitative indicator of C sequestration. Since Basidiomycetes have different biological and morphological characteristics than Ascomycetes, combining them (i.e., as Ascomicetes:Basidiomycetes ratio) seems to be a good option for choosing the most suitable woody species for carbon accumulation in agricultural soils.

Two genera (Chamaemyces and Hydropus) and 20 species of fungi were recorded for the first time for the Montenegrin funga (mycobiota), comprising 27 records from seven municipalities. Basic geographical and ecological data are given for all the records. Each species is shown with a photograph of the basidiomata in the field and is briefly annotated.

In this study, five species of Marasmius namely, M. bambusiniformis, M. haematocephalus, M. leveilleanus, M. midnapurensis, and M rotalis, plus Paramarasmius palmivorus are described, based on morphotaxonomic and molecular characters. Sequence data from internal transcribed spacers were used for phylogenetic analyses of the six species, supporting their identification based on macro and micromorphological characters. Dual-specificity LAMMER kinases are highly evolutionarily conserved in eukaryotes and play pivotal roles in diverse physiological processes, such as growth, differentiation, and stress responses. Although the functions of LAMMER kinase in fungal pathogens in pathogenicity and stress responses have been characterized, its role in Cryptococcus neoformans, a human fungal pathogen and a model yeast of basidiomycetes, remains elusive. In this study, we identified a LKH1 homologous gene and constructed a strain with a deleted LKH1 and a complemented strain. Similar to other fungi, the lkh1Δ mutant showed intrinsic growth defects. We observed that C. neoformans Lkh1 was involved in diverse stress responses, including oxidative stress and cell wall stress. Particularly, Lkh1 regulates DNA damage responses in Rad53-dependent and -independent manners. Furthermore, the absence of LKH1 reduced basidiospore formation. Our observations indicate that Lkh1 becomes hyperphosphorylated upon treatment with rapamycin, a TOR protein inhibitor. Notably, LKH1 deletion led to defects in melanin synthesis and capsule formation. Furthermore, we found that the deletion of LKH1 led to the avirulence of C. neoformans in a systemic cryptococcosis murine model. The genus Sidera (Hymenochaetales, Basidiomycota) comprises white-rot, mono- or dimitic fungi with poroid or hydnoid hymenophore. It has a worldwide distribution albeit with fewer species present in the Southern Hemisphere. Although recent studies revealed the existence of several new Sidera species, there are still taxonomic inconsistencies and obscure phylogenetic relationships amongst certain taxa of the genus. In this work, a large number of Sidera collections were used to obtain an updated phylogeny, based on ITS and 28S rDNA sequences by including new material from Mediterranean Europe. The monophyly of the genus was strongly supported and all species with poroid hymenophore formed a highly-supported lineage with two major subclades. In total, 23 putative species were recognised. Amongst those, five are considered to possibly represent entities new to science, but further work is required since they are represented by single specimens or environmental sequences. Examined collections originally named S. lenis from southern Europe were grouped within S. vulgaris. Similarly, several collections under various names were hereby identified as S. vulgaris, including those of the recently described species S. tibetica. Furthermore, a critical discussion (based on morphoanatomical findings) is made on the key features that could be used to distinguish S. lenis from S. vulgaris. [ABSTRACT FROM AUTHOR]

We produced shio-koji (Japanese fermented seasoning)-like seasoning using mushrooms instead of Aspergillus oryzae. Shio-koji was fermented on a small scale using 50 mushroom strains and the resulting reducing sugar content of each sample was measured. Shio-koji produced by fermentation with Ganoderma lucidum NBRC31863 and Grifola frondosa SKB003 had high reducing sugar contents of 234.3 g / L and 219.4 g / L, respectively. The formol nitrogen content was highest in shio-koji produced by fermentation with Neolentinus lepideus SKB008 (0.147%). We selected six strains of mushrooms and fermented shio-koji at a medium scale using these strains. The findings showed that G. frondosa SKB003 had the highest reducing sugar content (143.2 g / L) and that Wolfiporia cocos NBRC10024 had the highest protease activity (7.37 U / L). We therefore selected G. frondosa SKB003 and W. cocos NBRC10024 for testing, and subjected beef soaked in shio-koji produced using these strains and controls (commercial shio-koji and 7% saline solution) to sensory evaluation testing. In terms of ratings, no significant differences were detected among samples. However, noticeable differences were observed among the samples using a three-point discrimination preference method, which showed that the shio-koji produced by fermentation with G. frondosa SKB003 was rated most highly. Tofuyo (traditional fermented food of Okinawa) was produced using mushrooms instead of Monascus sp. and Aspergillus oryzae. To screen mushrooms that produce alcohol-tolerant protease, 76 strains of edible mushroom were cultivated on rice medium and total protease activity was measured in the presence of 20% ethanol. Ten strains of mushroom, such as Auricularia sp. SKB074 (4.46 U/L) and Auricularia americana SKB073 (4.03 U/L), showed higher activity than that of commercial rice-koji (1.46 U/L). Culture conditions and preliminary fermentation tests were investigated, and Auricularia auricula-judae NBRC100150 and Lepista sordida SKB010 were selected to produce tofuyo based on the taste and aroma of the prototype. Tofu was fermented using mushroom-koji of these strains in the presence of alcohol, and the texture of the produced tofuyo was evaluated using the texture profile method. Tofuyo fermented for 14 or 28 days using L. sordida SKB010 showed texture parameters comparable to those of commercial tofuyo. Tastes and aromas of tofuyo fermented by L. sordida SKB010 showed less distinctiveness, and depending on the alcohol and seasoning used, commercialization as tofuyo with new characteristics might be possible. Basidiomycetes found in forest ecosystems have been used traditionally as food and medicine for centuries, and various studies have validated many of their health benefits. In recent years, the focus in medicinal basidiomycetes has increased, and several notable trends in their application and research have been observed. In light of the emerging literatures, this review attempts to summarise and discuss evidences from the past decade on the importance of exploring basidiomycetes for their pharmacological properties. It also aims to serve as a critical reference for research works focused on the potentials of wild basidiomycetes in Malaysia, as well as from worldwide, to produce a diverse range of bioactive metabolites. Special attention is given to the low-molecular weight bioactive compounds produced in cultured mycelium and culture broth of these macrofungi. Phenol-formaldehyde (PF) resins can be impregnated and cured in situ to improve the woods dimensional stability and decay resistance. In search of renewable alternatives, the substitution of phenol by lignin cleavage products (LCP) has been discussed. However, the different chemical nature may affect the performance of the resin against fungal decay, formaldehyde emission, and equilibrium moisture content. In this study, 30 % (w/w) of the phenol in PF resins were substituted by LCP obtained from microwave-assisted pyrolysis. Scots pine sapwood was modified with the resin. The decay resistance against Rhodonia placenta, Gloeophyllum trabeum, and Trametes versicolor was determined. Additionally, effects of specimen organisation within the Petri dish, different substrates, length of leaching, and type of inoculum were studied. Further, the materials water vapor sorption properties and formaldehyde emission were determined. All modifications effectively reduced fungal decay. With 10 % weight percent gain (WPG), initial decay was detected, while 20 % WPG and 30 % WPG provided efficient protection. The substitution of phenol increases the formaldehyde emission. While further reduction in formaldehyde in the resin admixture or formaldehyde scavengers may be required, the method described herein can be used to partly replace fossil-based phenol, while maintaining good fungal resistance. Introduction: The genus Omphalotus, in particular the "Jack‐O'Lantern mushrooms" Omphalotus illudens and Omphalotus olearius, are famous for the production of the DNA‐alkylating illudins. A lesser‐known species, Omphalotus mexicanus, native to Central America, also produces cytotoxic illudins S and M, but its minor secondary metabolites are yet to be investigated. Objective: To identify, isolate, and elucidate the structure of novel secondary metabolites of the illudin family in mycelial extracts of O. mexicanus from submerse cultivation. Methodology: A fermentation of the fungus in 15 L stirred tank bioreactors is described. Mycelial extracts were separated using a combination of flash chromatography with preparative RP‐C18 high‐performance liquid chromatography (HPLC). Analysis of metabolites was done using an ultrahigh‐performance liquid chromatography ultraviolet diode array detector (UPLC‐UV‐DAD) system coupled to an electrospray ionisation quadrupole time‐of‐flight (ESI‐QTOF) mass spectrometer. Structures were elucidated using one‐dimensional (1D) and two‐dimensional (2D) nuclear magnetic resonance spectroscopy (NMR) techniques followed by comparison of experimental and simulated electronic circular dichroism (ECD) spectra to determine absolute configurations. Results: Two novel illudin derivatives, for which we propose the names omphaderol (1) and illudaneol B (2), as well as illudaneol (3) and the unusual cyclobutylcyclopentane illudosin (4), were isolated from the mycelia and characterised. Conclusion: Particularly the illudaneol derivatives with their high titers may be potential building blocks for an alternative semisynthetic route to new illudin derivatives with improved medical properties. Additionally, the findings improve the knowledge of minor illudin compounds in the mycelial extract of this fungus and may be of significance for future biosynthetic studies of the illudins. ABSTRACT: Pythium insidiosum is an important oomycete pathogen of mammals that causes pythiosis, an endemic disease in warm climates that stands out for its unfavorable prognosis, lethality in the affected species, and difficulties in treatment. This study evaluated in vitro anti-P. insidiosum potential of aqueous, hydroethanolic, and ethanolic extracts of indigenous wild mushrooms from southern Brazil. The extracts were prepared from Amanita gemmata, Amanita muscaria, Auricularia auricula, Gymnopilus junonius, Lactarius deliciosus, Laccaria laccata, Psilocybe cubensis, and Russula xerampelina. In vitro susceptibility assays employed the microdilution technique according to the M38-A2 protocol CLSI. The hydroethanolic and ethanolic extracts of R. xerampelina showed anti-P. insidiosum activity at minimum inhibitory concentrations ranging from 1.87-7.50 mg/mL. The other mushroom species extracts showed no inhibitory effects on growth of P. insidiosum. This is the first study to evaluate the antimicrobial activity of mushrooms on oomycetes, evidencing the antimicrobial potential of R. xerampelina on the pathogen P. insidiosum. So, the present study expands new perspectives, since the secondary metabolites produced by mushrooms can be potential targets for the development of new categories of medicines. However, considering the wide biodiversity of Brazilian mushrooms, we suggested that the search for other basidiomycetes species with anti-P. insidiosum action needs to be expanded.

Abstract The emergence of antibiotic resistance in pathogens is one of the major health concerns facing mankind as different bacterial strains have developed resistance to antibiotics over the period of time due to overuse and misuse of antibiotics. Besides this, ability to form biofilms is another major factor contributing to antibiotic resistance, which has necessitated the need for exploration for novel and effective compounds with ability to inhibit biofilm formation. Endophytic fungi are reported to exhibit antibacterial and anti-biofilm potential and could serve as a potent source of novel antibacterial compounds. Majority of the bioactivities have been reported from fungi belonging to phylum Ascomycota. Endophytic basidiomycetes, inspite of their profound ability to serve as a source of bioactive compounds have not been exploited extensively. In present study, an attempt was made to assess the antibacterial, anti-biofilm and biofilm dispersion potential of an endophytic basidiomycetous fungus Schizophyllum commune procured from the culture collection of our lab. Ethyl acetate extract of S. commune showed good antibacterial activity against Staphylococcus aureus, Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, Salmonella enterica and Vibrio cholerae. Minimum inhibitory concentration and minimum bactericidal concentration of the extract were in the range of 1.25-10 mg/ml against the tested bacterial pathogens. The mode of action was determined to be bactericidal which was further confirmed by time kill studies. Good anti-biofilm activity of S. commune extract was recorded against K. pneumoniae and S. enterica, which was further validated by fluorescence microscopy. The present study highlights the importance of endophytic basidiomycetes as source of therapeutic compounds.

China has a huge area of diverse landscapes and is believed to conceive incredibly high fungal diversity. To systematically and promptly report Chinese fungal species, we initiate the series of Catalogue of fungi in China here. In the first paper of this series, we focus on plant-inhabiting fungi. A total of 33 new taxa are described all over China. These taxa include two new genera, viz. Cremeoefibula and Nothopucciniastrum, 18 new species, viz. Annulohypoxylon lancangensis, Ascotaiwania coffeae, Clitocella neofallax, Coleopuccinia yunnanensis, Cremeoefibula hengduanensis, Crepidotus furcaticystidiosus, C. tomentellus, Diachea macroverrucosa, Helicogloea hangzhouensis, Hyalopsora caprearum, Nemania polymorpha, Phanerochaetella austrosinensis, Physalacria tianzhongshanensis, Setophaeosphaeria panlongensis, Subulicystidium boreale, Trechispora subaraneosa, Vikalpa dujuanhuensis, and Xylaria pteridicola, and 13 new combinations, viz. Nothopucciniastrum actinidiae, N. boehmeriae, N. coriariae, N. corni, N. coryli, N. fagi, N. kusanoi, N. hikosanense, N. hydrangeae-petiolaris, N. miyabeanum, N. styracinum, N. tiliae, and N. yoshinagae. The morphological characteristics and phylogenetic evidence are used to support the establishment of these new taxa and the accuracy of their taxonomic placements. This paper presents an annotated checklist of the Basidiomycota taxa (including lichenicolous fungi and the subdivision Pucciniomycotina) from the Azores archipelago and reviews the published records to account for their taxonomic status. The number of Basidiomycota species recorded in the Azores has increased considerably during the 20th century and now stands at 544 species. This study provides distribution data and includes changes in the nomenclature of the listed taxa. Sampling campaigns contributed to 116 new records of Basidiomycota for the Azores archipelago. In addition, there were new records for eight islands: 162 species found for the first time on São Miguel Island, 55 species new to Santa Maria Island, 33 species new to Flores Island, 15 species new to Terceira Island, 9 species new to Pico Island, 17 species new to São Jorge Island, 4 species new to Graciosa Island, and 2 species new to Corvo Island. The transformation of vegetation cover in the archipelago has been very drastic, and this is reflected in the presence of many foreign fungal species on the islands. From these data, we conclude that within Macaronesia, the diversity of Basidiomycota in the Azores is more similar to that in Madeira than in the Canary Islands. [ABSTRACT FROM AUTHOR]

Heterotrophic soil microorganisms are responsible for ~50% of the carbon dioxide released by respiration from the terrestrial biosphere each year. The respiratory response of soil microbial communities to warming, and the control mechanisms, remains uncertain, yet is critical to understanding the future land carbon (C)‐climate feedback. Individuals of nine species of fungi decomposing wood were exposed to 90 days of cooling to evaluate the medium‐term effect of temperature on respiration. Overall, the effect of temperature on respiration increased in the medium term, with no evidence of compensation. However, the increasing effect of temperature on respiration was lost after correcting for changes in biomass. These results indicate that C loss through respiration of wood‐decomposing fungi will increase beyond the direct effects of temperature on respiration, potentially promoting greater C losses from terrestrial ecosystems and a positive feedback to climate change. It is controversial how useful bioassays are for identifying the in vivo toxicity of hazardous environmental exposures. In this study, fruiting bodies of forest mushrooms (n = 46), indoor mold colonies (n = 412), fungal secondary metabolites (n = 18), xenobiotic chemicals such as biocides and detergents (n = 6), and methanol extracts of indoor dusts from urban buildings (n = 26) were screened with two different bioactivity assays: boar sperm motility inhibition (BSMI) and inhibition of cell proliferation (ICP) tests. For the forest mushrooms, the toxicity testing result was positive for 100% of poisonous-classified species, 69% of non-edible-classified species, and 18% of edible-classified species. Colonies of 21 isolates of Ascomycota mold fungal species previously isolated from water-damaged buildings proved to be toxic in the tests. Out of the fungal metabolites and xenobiotic chemicals, 94% and 100% were toxic, respectively. Out of the indoor dusts from moldy-classified houses (n = 12) and from dry, mold-free houses (n = 14), 50% and 57% were toxic, respectively. The bioassay tests, however, could not differentiate the samples from indoor dusts of moldy-classified buildings from those from the mold-free buildings. Xenobiotic chemicals and indoor dusts were more toxic in the BSMI assay than in the ICP assay, whereas the opposite results were obtained with the Ascomycota mold colonies and fungal secondary metabolites. The tests recognized unknown methanol-soluble thermoresistant substances in indoor settled dusts. Toxic indoor dusts may indicate a harmful exposure, regardless of whether the toxicity is due to xenobiotic chemicals or microbial metabolites. [ABSTRACT FROM AUTHOR]

The genus Erythrobasidium is kind of species-scarce and undersampling basidiomycetes. Currently, only six species have been accepted into the genus and the diversity still remains incompletely understood. In this study, five Erythrobasidium strains were isolated in the surface of plant leaves collected from the Baotianman Nature Reserve, Henan Province, central China. Phylogenetic analyses of the small ribosomal subunit (SSU) rRNA gene, the internal transcribed spacer (ITS) region, the D1/D2 domain of the large subunit (LSU) rRNA gene, and the translation elongation factor 1-a (TEF1-a) gene coupled with morphological studies were employed to characterize and identify these isolates. As a result of these, two new species, namely E. turpiniae sp. nov. and E. nanyangense sp. nov., were delimited and proposed based on morphological and molecular evidence. A detailed description and illustration of both new species, as well as their differences with the close relatives in the genus are provided. ctive: As a result of the study, branched polysaccharides were isolated from the basidiomycete raw materials of Ganoderma lucidum. It has been established that the isolated fractions contain branched polysaccharides in the form of complexes with melanin. Methods: After purification of polysaccharides by ion-exchange chromatography, two fractions were obtained from basidial raw materials: neutral polysaccharides GW-1 with a yield of 25.71% and anionic polysaccharides GW-2, the yield of which was 5.26%, respectively. The physicochemical properties of the obtained samples were studied by IR and UV spectroscopy. The degree of purity of the obtained fractions of branched polysaccharides was established. Using gas chromatography, one-dimensional (13C NMR, 1H NMR) and two-dimensional (COSY, TOCSY, HSQC, HMBC, NOESY) NMR spectroscopy, the composition and molecular structure of the obtained polysaccharide samples were determined. Results and Discussion: The results showed that the isolated and purified polysaccharides are branched glucans with 1,4,6- and 1,3,6-bonds between glucopyranose units. Pharmacotoxicological studies were carried out on white outbred mice and it was found that the resulting polysaccharides belong to class V, practically non-toxic compounds (LD50³ 2000 mg/kg). Conclusions: Isolated polysaccharides (GW) are promising biologically active components, on the basis of which it is possible to create drugs with hepatoprotective and antitumor activity. Substituted benzaldehydes are the most commonly used natural-occurring flavours in the world. The consumer's preference for 'natural or organic' aromas has increased the request for flavours possessing the 'natural' status. The resulting shortage of aromatic aldehydes of extractive origin, such as vanillin, veratraldehyde and piperonal, can be offset by developing a new biotechnological synthesis method. Here, we report a study on the microbiological reduction of five natural benzoic acid derivatives, namely p-anisic, vanillic, veratric, piperonylic and eudesmic acids, to produce the corresponding fragrant aldehydes. We found that different Basidiomycota strains can efficiently perform this transformation, with good chemical selectivity and tolerance to the toxicity of substrates and products. Besides confirming the carboxylic acid reductase activity of the already studied fungi Pycnoporus cinnabarinus, we discovered that other species such as Pleurotus eryngii, Pleurotus sapidus and Laetiporus sulphureus as well as the non-ligninolytic fungi Lepista nuda are valuable microorganisms for the synthesis of anisaldehyde, vanillin, veratraldehyde, piperonal and 3,4,5-trimethoxybenzaldehyde from the corresponding acids. According to our findings, we propose a reliable process for the preparation of the above-mentioned aldehydes, in natural form. A review of selected studies on fungal endophytes confirms the paucity of Basidiomycota and basal fungi, with almost 90% attributed to Ascomycota. Reasons for the low number of Basidiomycota and basal fungi, including the Chytridiomycota, Mucoromycota, and Mortierellomycota, are advanced, including isolation procedure and media, incubation period and the slow growth of basidiomycetes, the identification of non-sporulating isolates, endophyte competition, and fungus–host interactions. We compare the detection of endophytes through culture-dependent methods and culture-independent methods, the role of fungi on senescence of the host plant, and next-generation studies. Flavoring compounds are substances that directly influence the acceptance or rejection of a product. They are considered as essential components in the industrial sector due to their wide range of applications in different areas, such as food, cosmetics and pharmaceuticals. With the growing demand and concern of consumers for the acquisition of flavors from natural products, alternatives for the sustainable and low-cost production of such compounds becomes mandatory. Among these alternatives, biotechnological processes involving fungi are considered ecologically suitable and sustainable, as they tend to use conditions that are less harmful to the environment. The application of filamentous fungi such as basidiomycetes in biotechnological processes has been very promising, although it depends on the strain and growing conditions for obtaining bioaromas. The present review aims to compile reports on the potential of several basidiomycete fungi in the production of bioaromas using biotechnological methods. This review also includes the availability of nutrients and covers the new perspectives created with the application of agro-industrial residues as alternative cultivation substrates for these microorganisms. In India the waste generated from agriculture crops does not have any proper industrial use. Mushroom is a fleshy, macroscopic fungus belonging to the basidiomycetes group of Mycota kingdom. Edible mushrooms have significant diversity in their cultivation. The climate of the northern hilly region of India is suitable for Agaricus whereas the southern is fit for Volvoriella cultivation. The subtropical region offers the perfect conditions for Pleurotus cultivation. Pleurotus has many species like P. ostreatus, P. florida, P. sapidus, P. djmor, P. eous etc. Pleurotus ostreatus is suitable for tropical and subtropical region of the country. Rajasthan produces several subtropical crops like Bajra, Mustard, and Wheat, which resulting in generation of lignocellulose-rich waste. This study investigated the impact of various agro-wastes on the growth of Pleurotus ostreatus. Three combinations of agro-crop wastes were employed and examined. Pleurotus ostreatus took the least amount of time for the spawn run in the Wheat-Mustard-Bajra (T3) combination, while the Wheat-Bajra (T1) combination saw the earliest primordia initiation. The T1 combination also saw the shortest maturation time and shortest harvesting period. Furthermore, among the three highest-weight individuals from each treatment, T3 exhibited larger dimensions compared to others, with weights of 22 gm, stem length of 6 cm, and cap dimensions of 8x9cm². Maximum harvested mushroom production (636 gm), biological efficiency (84.7%) and Moisture content (91.44%) were noted in the T2 combination. The goals of sustainable development will be met and an appropriate agro-waste for mushroom cultivation will be established with the aid of this study. [ABSTRACT FROM AUTHOR]

Five species of edible basidiomycetes - Pleurotus ostreatus, Flammulina filiformis, Ganoderma lucidum, Lentinula edodes, and Polyporus squamosus - were isolated and cultivated as submerged mycelium for the biosynthesis of laccase enzymes and biomass production. The selected mushrooms were grown on different agar media to determine the average growth rate, tested on the Potato Dextrose Agar medium supplemented with guaiacol to determine the laccase enzyme index, and then grown submerged in a rotary incubator, in Erlenmeyer flasks. The development of these species was studied in a medium with sugarcane molasses and mineral salts. Bioconversion of plant material with white-rot fungi has demonstrated improvements in in vitro digestibility, however, effects on rumen fermentation parameters and methane (CH4) production, when included as a dietary ingredient, are not clear. The aim of this study was to assess changes in rumen fermentation parameters and in vitro CH4 production by dietary inclusion of ryegrass-fescue hay biodegraded with P. ostreatus (H-PO) during 14 days of solid fermentation. The experiment used 2 diets with contrasting acid detergent fibre (aADFom) content which were added with 0, 3 and 6% H-PO. Substrates were incubated 3 times with sampling at 24 h. Results were analysed by ANOVA, and compared with Tukey’s test and polynomial contrasts. Inclusion of H-PO at concentrations of 3 and 6% resulted in increased in vitro dry matter (DM) disappearance (IVDMD) by 10 and 12% respectively, whereas high concentrations of aADFom reduced IVDMD by 10% as compared to low aADFom. ABSTRACT This study was performed with the aim of determining the “natural” wood preferences of subterranean termites (Isoptera) in a field feeding experiment, giving a choice between a control (Pinus sp.) and test woods: three species of Eucalyptus. Feeding preference order of subterranean termites was Pinus sp. > Eucalyptus robusta ≅ Eucalyptus urophylla > Eucalyptus pellita by the percentage of subterranean termite-damaged wooden stakes at 30-, 45-, and 60-day exposure periods in the field. Three species of subterranean termites were identified: Heterotermes longiceps (Snyder) (Rhinotermitidae), Coptotermes gestroi (Wasman) (Rhinotermitidae), and Nasutitermes jaraguae (Holmgren) (Termitidae). This is the first record of occurrence of H. longiceps in the state of Rio de Janeiro. Unidentified fungi species of Basidiomycetes infected the wood stakes of all forest species tested; although none of them were attacked by subterranean termites.

The aim of the study was to investigate the antibacterial activity of selected strains of the genus Hericium, belonging to basidiomycetes, from the IBK Mushroom Culture Collection of the M.G. Kholodny Institute of Botany of the National Academy of Sciences of Ukraine. A total of 14 strains including H. abietis, H. cirrhatum, H. coralloides, and H. erinaceus were investigated. The strains were cultivated on a liquid glucose-peptone-yeast medium. Both the homogenised mycelium and filtrate of these fungi were evaluated against gram-positive (Bacillus subtilis, Micrococcus luteus, and Staphylococcus aureus) and gram-negative (Escherichia coli and Pseudomonas aeruginosa) bacteria by the disk diffusion and cylinder methods. The activity of the strains varied significantly. Depending on the techniques assay, species, strain, and stage of fungal development, the inhibition zone of the tested bacteria ranged from 5.3 mm to 20.0 mm. In general, the antibacterial potential of the culture filtrates of the investigated species was significantly higher than their mycelia activity. The homogenised mycelium showed potentially good results only against E. coli. The antibacterial activities of the H. abietis and H. cirrhatum species were observed for the first time. To the best of our knowledge, the ability of H. coralloides to inhibit the growth of P. aeruginosa and S. aureus has not been previously reported. The obtained results indicate the ability of the studied Hericium species to produce antibacterial metabolites with a wide and narrow spectrum of action which might have potential health benefits and could be recommended for the further analysis, isolation and identification of potentially promising antibacterial compounds in pharmacology.

Entoloma is a relatively large genus in Agaricales, with a rich diversity of species and a wide distribution. In this study, seven new species of Entoloma belonging to the subgenus Cubospora have been identified based on morphological and phylogenetic evidence from subtropical regions of China. Morphologically, E. excavatum is characterized by the yellow, depressed, estriate pileus and medium-sized basidiospores; E. lacticolor is recognized by the white and papillate pileus, adnexed lamellae, and presence of clamp connections; E. phlebophyllum is identified by the pink-to-maroon and estriate pileus, and lamellae with lateral veins; E. rufomarginatum differs from other cuboid-spored species by the lamellae edge which is red-brown-underlined; E. subcycneum is characterized by the white pileus and carneogriseum-type cheilocystidia; E. submurrayi is recognized by the pileus margin exceeding the lamellae, 2-layered pileipellis with hyphae of different widths, and the presence of clamp connections; E. tomentosum is identified by the tomentose pileus, heterogeneous lamella edge, and versiform cheilocystidia with brown-yellow contents. Their distinct taxonomic status is confirmed by the positions of the seven new species in both the ITS + LSU and 3-locus (LSU, tef-1α, rpb2) phylogenetic trees. Detailed descriptions, color photos, and a key to related species are presented.

Coriolopsis trogii is a basidiomycete fungus which utilizes a large array of lignin-modifying enzymes to colonize and decompose dead wood. Its extracellular enzymatic arsenal includes laccases, i.e., polyphenol oxidases of relevant interest for different industrial applications thanks to their ability to oxidize a diverse range of natural and synthetic compounds. In this work, the production of laccases in C. trogii MUT3379 was explored and improved. From an initial production of ca. 10,000 U L−1, the fermentation process was gradually optimized, reaching a final yield of ca. 200,000 U L−1. An SDS-PAGE analysis of the secretome highlighted the presence of a main protein of ca. 60 kDa showing laccase activity, which was designated as Lac3379-1 once its primary sequence was established by tandem mass spectrometry. The characterization of Lac3379-1 revealed a remarkable enzymatic stability in the presence of surfactants and solvents and a diversified activity on a broad range of substrates, positioning it as an interesting tool for diverse biotechnological applications. The high-yield and robust production process indicates C. trogii MUT3379 as a promising cell factory for laccases, offering new perspectives for industrial applications of lignin-modifying enzymes.

DNA barcodes can provide accurate identification of plants. We used previously reported DNA primers targeting the internal transcribed spacer (ITS1) region of the nuclear ribosomal cistron, internal transcribed spacer (ITS2), and chloroplast trnL (UAA) intron to identify four trees at Bergen Community College. Two of the four trees were identified as Acer rubrum and Fagus sylvatica. However, Quercus was only identified at the genus level, and the fourth tree did not show similar identification between barcodes. Next-generation sequencing of 16S rRNA genes showed that the predominant bacterial communities in the rhizosphere mainly consisted of the Pseudomonadota, Actinomycetota, Bacteroidota, and Acidobacteriota. A. rubrum showed the most diverse bacterial community while F. sylvatica was less diverse. The genus Rhodoplanes showed the highest relative bacterial abundance in all trees. Fungal ITS sequence analysis demonstrated that the communities predominantly consisted of the Ascomycota and Basidiomycota. Quercus showed the highest fungi diversity while F. sylvatica showed the lowest. Russula showed the highest abundance of fungi genera. Average similarity values in the rhizosphere for fungi communities at the phylum level were higher than for bacteria. However, at the genus level, bacterial communities showed higher similarities than fungi. Similarity values decreased at lower taxonomical levels for both bacteria and fungi, indicating each tree has selected for specific bacterial and fungal communities. This study confirmed the distinctiveness of the microbial communities in the rhizosphere of each tree and their importance in sustaining and supporting viability and growth but also demonstrating the limitations of DNA barcoding with the primers used in this study to identify genus and species for some of the trees. The optimization of DNA barcoding will require additional DNA sequences to enhance the resolution and identification of trees at the study site.

Bioconversion of plant material with white-rot fungi has demonstrated improvements in in vitro digestibility, however, effects on rumen fermentation parameters and methane (CH4) production, when included as a dietary ingredient, are not clear. The aim of this study was to assess changes in rumen fermentation parameters and in vitro CH4 production by dietary inclusion of ryegrass-fescue hay biodegraded with P. ostreatus (H-PO) during 14 days of solid fermentation. The experiment used 2 diets with contrasting acid detergent fibre (aADFom) content which were added with 0, 3 and 6% H-PO. Substrates were incubated 3 times with sampling at 24 h. Results were analysed by ANOVA, and compared with Tukey's test and polynomial contrasts. Inclusion of H-PO at concentrations of 3 and 6% resulted in increased in vitro dry matter (DM) disappearance (IVDMD) by 10 and 12% respectively, whereas high concentrations of aADFom reduced IVDMD by 10% as compared to low aADFom. Methane production (mg/g DM incubated) and yield (mg/g DM degraded) were reduced by increasing aADFom and P. ostreatus biodegraded hay concentrations in the diets (p < 0.001) whereas concentrations of total short-chain fatty acids were not affected by the inclusion of biodegraded hay (p = 0.080) or aADFom concentration (p = 0.076). Increasing concentrations of biodegraded hay resulted in increased acetate (p < 0.001), and reduced propionate proportions (p = 0.021) but did not affect butyrate (p = 0.233). In conclusion the use of biodegraded hay as a pre-treated ingredient improved the utilisation of low and high fibre diets by increasing the in vitro disappearance, and decreasing CH4 emissions, without increasing the propionate proportion. The use of hay biodegraded with P. ostreatus (H-PO) improves the digestibility of ruminant diets. The use of H-PO decreases methane, and increases acetate production in in vitro conditions. Culture collections (CCs) play an important role in the ex situ conservation of biological material and maintaining species and strains, which can be used for scientific and practical purposes. The Komarov Botanical Institute Basidiomycetes Culture Collection (LE-BIN) preserves a large number of original dikaryon strains of various taxonomical and ecological groups of fungi from different geographical regions. Started in the late 1950s for the investigation of Basidiomycetes' biological activity, today, in Russia, it has become a unique specialized macromycetes collection, preserving 3680 strains from 776 species of fungi. The Collection's development is aimed at ex situ conservation of fungal diversity, with an emphasis on preserving rare and endangered species, ectomycorrhizal fungi, and strains useful for biotechnology and medicine. The main methods applied in the collection for maintaining and working with cultures are described, and the results are presented. Some problems for the isolation and cultivation of species are discussed. The taxonomical structure and variety of the strains in the collection fund are analyzed, and they show that the taxonomical diversity of fungi in the LE-BIN is commensurable with the largest CCs in the world. The achievements from the ex situ conservation of the diversity of macromycetes and the main results from the screening and investigation of the collection's strains demonstrate that a number of strains can be prospective producers of enzymes (oxidoreductases and proteases), lipids, and biologically active compounds (terpenoids, phthalides, etc.) for biotechnology and medicine. The basidiomycete Serpula lacrymans is responsible for major timber devastation in houses. Basidiomycetes are known to harbour a diverse but poorly understood microbial community of bacteria, archaea, yeasts and filamentous fungi. In this study, we used amplicon‐sequencing to analyse the abundance and composition of prokaryotic communities associated with fruiting bodies of S. lacrymans and compared them to communities of surrounding material to access the 'background' community structure. Our findings indicate that bacterial genera cluster depended on sample type and that the main driver for microbial diversity is specimen, followed by sample origin. The most abundant bacterial phylum identified in the fruiting bodies was Pseudomonadota, followed by Actinomycetota and Bacteroidota. The prokaryote community of the mycelium was dominated by Actinomycetota, Halobacterota and Pseudomonadota. Actinomycetota was the most abundant phylum in both environment samples (infested timber and underground scree), followed by Bacillota in wood and Pseudomonadota in underground samples. Nocardioides, Pseudomonas, Pseudonochardia, Streptomyces and Rubrobacter spp. were among others found to comprise the core microbiome of S. lacrymans basidiocarps. This research contributes to the understanding of the holobiont S. lacrymans and gives hints to potential bacterial phyla important for its development and lifestyle. [ABSTRACT FROM AUTHOR]
