ABSTRACT All birds possess a unique venous architecture surrounding the kidneys known as the renal portal system. In veterinary medicine, this system is well known for causing a first‐pass effect when medication is administered parenterally via the leg veins, that is venous blood from the leg is filtered before entering general circulation, thus possibly compromising adequate dosage. Additionally, bilateral valves are present in these veins, and it has been hypothesized that they play a crucial role in regulating flow through the kidneys to protect them against increases in blood pressure. While this hypothesis has been acknowledged, it has not been thoroughly explored. We propose that the function of the renal portal valve extends beyond its significance for kidney function, potentially impacting general hemodynamics. Examining anatomical similarities with extant non‐avian reptiles, which lack the renal portal shunt with valve, could reveal additional functionalities of this system in birds. Given the endothermic metabolism and the energetically expensive locomotor activity of birds, the resistance of the hepatic and renal portal system might constrain the blood flow from splanchnic to non‐splanchnic blood vessels necessary for (sustained) peak performance. Therefore, diverting blood from the renal portal system using the renal portal valve as a regulatory structure might represent a key adaptation to facilitate sustained peak performance. In addition, we hypothesize that this shunt and valve represents a very early adaptation in amniotes, possibly lost in extant non‐avian reptiles but enhanced in birds, with a pivotal role in maintaining hemodynamic homeostasis to support the high metabolic rates characteristic of birds. For most quadrupeds, locomotion involves alternating movements of the fore- and hindlimbs. In birds, however, while walking generally involves alternating movements of the legs, to generate lift and thrust, the wings are moved synchronously with each other. Neural circuits in the spinal cord, referred to as central pattern generators (CPGs), are the source of the basic locomotor rhythms and patterns. Given the differences in the patterns of movement of the wings and legs, it is likely that the neuronal components and connectivity of the CPG that coordinates wing movements differ from those that coordinate leg movements. In this study, we used in vitro preparations of embryonic chicken spinal cords (E11–E14) to compare the neural responses of spinal CPGs that control and coordinate wing flapping with those that control alternating leg movements. We found that in response to N-methyl-d-aspartate (NMDA) or a combination of NMDA and serotonin (5-HT), the intact chicken spinal cord produced rhythmic outputs that were synchronous both bilaterally and between the wing and leg segments. Despite this, we found that this rhythmic output was disrupted by an antagonist of glycine receptors in the lumbosacral (legs), but not the brachial (wing) segments. Introduction: Japanese quail are of significant economic value, providing protein nutrition to humans through their reproductive activity; however, sexual dimorphism in this species remains relatively unexplored compared with other model species. Method: A total of 114 RNA sequencing datasets (18 and 96 samples for quail and chicken, respectively) were collected from existing studies to gain a comprehensive understanding of sexual dimorphism in quail. Cross-species integrated analyses were performed with transcriptome data from evolutionarily close chickens to identify sex-biased genes in the embryonic, adult brain, and gonadal tissues. Results: Our findings indicate that the expression patterns of genes involved in sex-determination mechanisms during embryonic development, as well as those of most sex-biased genes in the adult brain and gonads, are identical between quails and chickens. Similar to most birds with a ZW sex determination system, quails lacked global dosage compensation for the Z chromosome, resulting in directional outcomes that supported the hypothesis that sex is determined by the individual dosage of Z-chromosomal genes, including long non-coding RNAs located in the male hypermethylated region. Furthermore, genes, such as WNT4 and VIP, reversed their sex-biased patterns at different points in embryonic development and/or in different adult tissues, suggesting a potential hurdle in breeding and transgenic experiments involving avian sexrelated traits. Discussion: The findings of this study are expected to enhance our understanding of sexual dimorphism in birds and subsequently facilitate insights into the field of breeding and transgenesis of sex-related traits that economically benefit humans. The Diederik cuckoo, Chrysococcyx caprius, is a small Afrotropical bird in the family Cuculidae. It is taxonomically related to 13 other species within the genus Chrysococcyx and is migratory in sub‐Saharan Africa. It has a unique breeding behaviour of being a brood parasite: Breeding pairs lay their eggs in the nests of a host species and hatchlings expel the eggs of the host species. The aim of the present study was to investigate diversity in two circadian clock genes, Clock and Adcyap1, to probe for a relationship between genetic polymorphisms and their role in circannual timing and habitat selection (phenology) in intra‐African migrants. DNA extracted from blood was used for the PCR amplification and sequencing of clock genes in 30 Diederik cuckoos. Three alleles were detected for Clock with similar genotypes between individuals from the Northern and Southern breeding ranges while 10 alleles were detected for Adcyap1, having shorter alleles in the North and longer alleles in the South. Population genetic analyses, including allele frequency and zygosity analysis, showed distinctly higher frequencies for the most abundant Clock allele, containing 10 polyglutamine repeats, as well as a high degree of homozygosity. In contrast, all individuals were heterozygous for Adcyap1 and alleles from both regions showed distinct differences in abundance. Comparisons between both clock genes and phenology found several phenotypic correlations. This included evidence of a relationship between the shorter alleles and habitat selection as well as a relationship between longer alleles and timing. In both instances, evidence is provided that these effects may be sex‐specific. Given that these genes drive some of the synchronicity between environments and the life cycles of birds, they provide valuable insight into the fitness of species facing global challenges including climate change, urbanisation and expanding agricultural practices. Simple Summary: This review aimed to provide an exhaustive overview of the advancements in avian SCZ research. In the review, we summarize the latest research on trait variations in avian SCZs, including vocalization, plumage, beak, and migratory traits. In addition, we discuss the mechanisms of different types of avian SCZ movements. Finally, we outline several significant questions for future studies. A secondary contact zone (SCZ) is an area where incipient species or divergent populations may meet, mate, and hybridize. Due to the diverse patterns of interspecific hybridization, SCZs function as field labs for illuminating the on-going evolutionary processes of speciation and the establishment of reproductive isolation. Interspecific hybridization is widely present in avian populations, making them an ideal system for SCZ studies. This review exhaustively summarizes the variations in unique traits within avian SCZs (vocalization, plumage, beak, and migratory traits) and the various movement patterns of SCZs observed in previous publications. It also highlights several potential future research directions in the genomic era, such as the relationship between phenotypic and genomic differentiation in SCZs, the genomic basis of trait differentiation, SCZs shared by multiple species, and accurate predictive models for forecasting future movements under climate change and human disturbances. This study determines the effects of probiotics - Saccharomyces cerevisiae on oocysts shedding, haematology and serum proteins of broilers infected with mixed Eimeria oocysts. 120-day-old broiler chicks were randomly assigned to four groups (Groups A - D) of 30 birds each. Group A - was fed a plain diet and not infected, Group B - was fed a supplemented diet and infected, Group C - was fed a plain diet and infected, and Group D - was fed a supplemented diet and not infected. S. cerevisiae was supplemented at 0.1 %/kg of feed. At week 3, Groups B and C were infected with sporulated mixed Eimeria oocysts orally, followed by a daily faecal examination of all groups. The rate of oocyst shedding, packed cell volume (PCV), haemoglobin concentration (Hb), red blood cell (RBC) count, total white blood cell (WBC) count, differential WBC count and serum levels of total protein, albumin and globulin were determined at five days' intervals. The data collected were analyzed using a one-way analysis of variance at a 95% confidence interval (p<0.05). Group C had the significantly higher (p<0.05) number of oocysts in faeces, total WBC count and globulin levels, with the lowest PCV, Hb, RBC count, total protein and albumin levels. Group D had significantly higher (p<0.05) heterophil, eosinophil and monocyte counts. In conclusion, dietary supplementation with S. cerevisiae (1 g/kg) ameliorated the effect of Eimeria infection in broilers, by reducing the rate of oocyst shedding, anaemia and hypoproteinaemia due to the activity of the parasite. Neonicotinoids, a neuro-effective class of insecticides, are heavily applied in agricultural activities worldwide. Poultry can be exposed to neonicotinoids by several routes, but the knowledge of neonicotinoid's metabolism in poultry and its associated interspecies differences is highly limited. Hence, this study aims to investigate the species differences in metabolite formations, as well as cytochrome P450 (CYP)-dependent metabolism of four major neonicotinoid compounds, acetamiprid, imidacloprid, clothianidin, and thiamethoxam, in poultry. In vitro biotransformation assays using hepatic microsomes of chicken, ducks, geese, quails, and rats were conducted. Metabolites of neonicotinoids were then screened by LC/Q-TOF and quantified by LC/MS/MS. The results revealed an existence of interspecies differences in the formations of N-[(6-chloro-3-pyridyl) methyl] -N-methyl acetamidine (IM-1-5) of acetamiprid and dm-clothianidin of clothianidin between chicken and other species. In addition, the greatest CYP activities in the metabolism of most neonicotinoid substrates, such as acetamiprid to dm-acetamiprid, imidacloprid to hydroxylated-imidacloprid and imidacloprid-olefin, clothianidin to dm-clothianidin, and thiamethoxam to clothianidin, were found in chicken. These results suggested that the CYPs in chicken may have a greater capacity for metabolism of neonicotinoids compared to other poultry. This study further revealed that the maximum intrinsic clearance of dn-imidacloprid and dn-clothianidin in ducks may be superintended by CYP-mediated nitro-reductions of imidacloprid and clothianidin. Further studies employing CYP recombinant enzymes may be required to elucidate the specific CYP isoforms that may be involved in neonicotinoid metabolism in avian species. [ABSTRACT FROM AUTHOR]

Australia has lost vast areas of its natural vegetation through agriculture and urbanization, resulting in the area of suitable habitat for many Australian bird species being greatly diminished. Given the geographical and ecological biases in anthropogenic land use, the impact of habitat loss now and into the future may be disproportionately high for some species, threatening their long-term persistence. Such changes are occurring rapidly, and habitat loss needs to be monitored dynamically to prevent extinction. To monitor changes in available unmodified species habitat, we use species' area of feeding habitat (AFH), which represents the area of habitat within a species' range with vegetation matching their feeding requirements. We analysed the past, current and future threats of habitat modification for 467 Australian bird species by calculating AFH across three time periods: pre-colonization, current (2020), and a projected future scenario (2100). These values were used to identify species subject to substantial habitat loss due to urbanization and agriculture and delineate predictors of such losses. Most species had experienced habitat loss since colonization (n = 442, 95%), and species that had already experienced considerable habitat loss were more likely to lose habitat into the future. Species with particular habitat associations (e.g., Mallee), were also more prone to greater proportions of habitat lost. The results highlight the utility of AFH and emphasize the importance of protecting what habitat remains for the species with highly depleted geographic ranges, noting that those that have experienced the most loss of unmodified habitat are the most likely to lose more unmodified habitat under future conditions. In order to study the hybrid effect between Tibetan chickens and Avian, 1-day-old healthy Ai-Tibetan hybrid chickens and the pure breeding of Tibetan chickens were selected in the experiment, 100 feathers each. In the same feeding environment, the same nutritional standard was provided, and the growth and development, body size index, slaughter performance and meat quality were studied, and the crossbreeding effect of Avian and Tibetan chicken was comprehensively evaluated. The results showed that the growth rate of Ai-Tibetan hybrid chickens was slightly faster than that of Tibetan chick‐ ens on the whole (except 70 days of age), and the body weight at 30 and 60 days of age was significantly higher than that of Tibetan chickens (P<0.05), but there was no significant difference in the other days of age. The breast width of 60-day-old Ai-Tibetan hybrid chickens was significantly higher than that of Ti‐ betan chickens (P<0.05), and the tibia length and oblique body length of 90-day-old Ai-Tibetan hybrid chickens were significantly higher than that of Tibetan chickens (P<0.05). The thigh muscle percentage and liver weight percentage of hens in Ai-Tibetan group were significantly higher than those in other groups, but the other indexes were not significantly different. Muscle color a* of Tibetan chickens was significantly higher than that of Ai-Tibetan hybrid chickens (P<0.05), and the drip loss rate of Ti‐ betan group was much lower than that in AiTibetan hybrid chickens, and there was a signifi‐ cant difference (P<0.05). The cooking loss of the Tibetan chickens was also lower than that of AiTibetan hybrid chickens. The meat quality of Ti‐ betan chickens was better than that of Ai-Tibetan hybrid chickens. Comprehensive analysis showed that the growth rate, slaughter performance and meat quality of the hybrid offspring of Tibetan chickens and Avian had no outstanding advantages in the envi‐ ronment of high altitude, and the hybridization effect was not obvious. The aim of this review is to findout the influence of environment and diet on digestive tract in birds. Birds comprised of complex digestive system, which is thought to have a significant impact on how well they utilize the nutrition that they consume. The esophagus, stomach and intestines of herbivorous birds is larger than those of carnivorous birds thus the herbivorous birds have longer, more complex digestive tract than carnivorous birds. Lead (Pb) is a ubiquitous, non-biodegradable heavy metal contaminant with a significant impact on both human and animal health. The adverse effect of lead on health and productivity of avian species has received little attention. Alchornea laxiflora (Benth) belongs to Euphorbiaceae family and grows naturally in the Nigerian rain forest. Decoction of the leaves is usually administered traditionally to treat inflammatory and infectious diseases. The ethanol extract of Alchornea laxiflora (EaAL) leaves was used in this study to ameliorate lead-induced neurodegeneration.Seven groups of 5-week-old cockerels (n=5) were treated for 6 weeks thus: Group A - Control (water only), Group B - (100 mg/kg of EaAL daily), Group C - (200 mg/kg of EaAL daily, p.o.), Group D - (1 % lead acetate in drinking water), Group E - (1 % lead acetate in drinking water and 100 mg/kg of EaAL daily), Group F - (1 % lead acetate and 200 mg/kg of EaAL daily), Group G - (1 % lead acetate and 100 mg/kg of Vitamin C). All administrations were per os birds were euthanized on day 43 by quick cervical dislocation. Histological stains (H&E and Nissl) and Black Gold II (BGII) histochemistry were used to assess alterations in the cerebrum and cerebellum.Administration of EaAL at the two concentrations resulted in a drastic reduction in the incidence of neuropathologies observed (e.g. pyknosis and multilayering of Purkinje cells, neuronal degeneration in hippocampus cerebrum and ependymal cells, distortion of meningeal epithelial cells, etc). BGII histochemistry revealed severe demyelination caused by the administration of lead acetate, while the two doses of EaAL showed significant restoration of myelin in the cerebellum. The amelioration of demyelination observed with the use of vitamin C was considerably lower than that recorded with the use of EaAL.The use of EaAL significantly ameliorated morphological alterations and demyelination caused by the administration of lead acetate, however, caution should be exercised in the administration, as individual species idiosyncrasies may arise and the tendency to pro-oxidation at 200 mg/kg when administered alone was observed in one subject.

Migratory birds are highly vulnerable to the effects of climate change, which disrupt their timing of migration, availability of resources and even hormonal regulation. Present review summarizes the key points regarding the impacts of climate change on migratory birds and the challenges they face for long-term viability. Rising temperatures, altered precipitation patterns, and shifts in wind patterns affect migration timing, availability of food resources, and flight paths. Trophic mismatches resulting from climate change reduce reproductive success. Changes in hormone production and secretion, driven by altered day length and light intensity, affect migration and reproductive timing. Elevated corticosterone levels due to environmental stressors influence migratory decisions. Hormones regulating appetite and energy balance impact feeding behavior and fat deposition. These impacts contribute to declining population sizes and increase the vulnerability of migratory birds to extinction. Conservation efforts should prioritize protecting stopover sites, promoting international cooperation, raising public awareness, and engaging stakeholders. Research advancements, such as satellite tracking and predictive modelling, support conservation strategies. Collaborative efforts between researchers, policymakers, and conservation organizations are essential for effective conservation and management of migratory bird populations in the face of climate change. Objective Animals Studied Procedures Results Conclusions To establish normative data for selected ocular diagnostic tests and commensal conjunctival microflora and describe the incidence of ocular pathology in Chilean flamingos.A total of 41 Chilean flamingos were examined at the Blank Park Zoo in Des Moines, Iowa.In 20 flamingos, blink rate was assessed undisturbed in their exhibit, then gentle manual restraint was used to assess palpebral fissure length (PFL), aqueous tear production (phenol red thread test [PRTT] in one eye, endodontic absorbent paper point tear test [EAPPTT] in the other), intraocular pressure (IOP; rebound tonometry), and fluorescein staining. Twenty‐one other flamingos were brought to a darkened area for neuro‐ophthalmic examination, slit lamp biomicroscopy, and indirect ophthalmoscopy. Swabs from seven flamingos were used for ocular microbiome evaluation.Results are presented as mean ± standard deviation (range). Flamingos comprised 23 females/18 males, aged 11 ± 9.1 (0.7–40) years. Test results: blink rate, 3.7 ± 2 (1–9) blinks/min; PFL, 11.2 ± 1.2 (9–14) mm; IOP, 14 ± 3.2 (10–22) mmHg; EAPPT, 10.2 ± 2.8 (9–14) mm/min; PRTT, 6.8 ± 2.5 (3–13) mm/15 s. Dazzle reflex was positive in four birds examined. Pathologies included cataracts (n = 7 birds), corneal fibrosis (n = 3), endothelial pigment (n = 2), uveal cysts (n = 1), lens luxation (n = 1), and uveitis (n = 1). Ocular microbiome showed high diversity of taxa.Baseline ocular parameters and incidence of ophthalmic pathology assist veterinarians with disease screening for Chilean flamingos, while the ocular microbiome showed high diversity. Objectives Animals Studied Methods Results Conclusions To determine normal ocular parameters of the MacQueen's bustard (Chlamydotis macqueenii) and describe ophthalmic lesions in a captive bred population.Captive breeding population of 257 Macqueen's bustards.All birds were screened for ocular abnormalities using direct ophthalmoscopy. Abnormalities were photographed. Normative values for Schirmer tear test‐1 (STT‐1), applanation tonometry, aerobic and anaerobic bacterial culture, fungal culture, and transcorneal ocular ultrasonography were derived from multiple cohorts of clinically normal adult birds. Five birds with ocular pathology also underwent transcorneal ultrasonography. Statistical comparisons for normative values between OD and OS, and males and females were made using a paired t‐test or Mann–Whitney U‐test, with a significance level of p < .05.Mean tear production based on Schirmer tear test 1 (STT‐1) was 10.16 ± 4.61 mm/min (3–21 mm/min). Mean intraocular pressure (IOP) was 12.42 ± 4.94 mm Hg (5–26 mm Hg). Staphylococcus species were the most isolated bacteria from the conjunctival surfaces of normal birds (85%). Significant differences were found in transcorneal ultrasonographic measurements between males and females for axial globe length (p = .032), vitreous body depth (p = .049) and lens thickness (p = .0428). Corneal fibrosis was the most observed ocular abnormality amongst eyes with pathological changes (39%).Schirmer tear testing, tonometry and transcorneal ultrasound can easily be utilized in MacQueen's bustards and provide reproducible results. High pathogenicity avian influenza viruses (HPAIVs) cause high morbidity and mortality in poultry species. HPAIV prevalence means high numbers of infected wild birds could lead to spill over events for farmed poultry. How these pathogens survive in the environment is important for disease maintenance and potential dissemination. We evaluated the temperature-associated survival kinetics for five clade 2.3.4.4 H5Nx HPAIVs (UK field strains between 2014 and 2021) incubated at up to three temperatures for up to ten weeks. The selected temperatures represented northern European winter (4 °C) and summer (20 °C); and a southern European summer temperature (30 °C). For each clade 2.3.4.4 HPAIV, the time in days to reduce the viral infectivity by 90% at temperature T was established (DT), showing that a lower incubation temperature prolonged virus survival (stability), where DT ranged from days to weeks. The fastest loss of viral infectivity was observed at 30 °C. Extrapolation of the graphical DT plots to the x-axis intercept provided the corresponding time to extinction for viral decay. Statistical tests of the difference between the DT values and extinction times of each clade 2.3.4.4 strain at each temperature indicated that the majority displayed different survival kinetics from the other strains at 4 °C and 20 °C. Aim: The rapidly changing Arctic is ideal for investigating uncertainties in climate projections. Despite the challenges of collecting data in this region, an unprecedented large‐scale survey of shorebirds has been conducted over the last 30 years. Our study aimed to (1) develop probabilistic estimates for the change in suitable habitat for 10 Arctic shorebird species in Canada by 2075 and (2) assess the contribution of modelling decisions to the uncertainty in these estimates. Location: Arctic Canada. Methods: To evaluate uncertainty, we considered six classes of modelling decisions, yielding 216 unique projections for each species. We tested three decisions that are less commonly explored − the pool of candidate variables, a method for selecting variables, and the maximum distance of tree line dispersal, as well as the modelling algorithm, carbon emissions scenario, and global circulation model. We used a bootstrapping approach, creating a probability distribution for the proportional change in suitable habitat for each species. Results: Our findings indicated a substantial risk for 8/10 species to lose over half of their suitable breeding habitat, but this projection is much less certain than has been described previously. While much uncertainty is unexplained, we were surprised that the largest source of uncertainty among our modelling decisions was from our choice of methods for variable selection, that the other modelling decisions were relatively small sources of uncertainty, overshadowing other modelling decisions. Main Conclusions: While most scenarios predict a northward shift and significant habitat loss for Arctic‐breeding shorebirds, the Arctic Archipelago of Canada will remain an important refuge because in many other Arctic regions, there is no land farther north for these species to shift into. The degree to which Grinnellian and Eltonian niche characteristics influence species distribution may depend on latitude. Tropical regions are environmentally stable and resource-rich, whereas temperate regions are comparatively less environmentally stable (e.g., environmental filtering). Moreover, phylogenetic niche conservatism could influence distributions by inhibiting the ability for species to colonize environmentally different locations. Herein, we examine relationships between niche characteristics, passerine distributions, and phylogenetic niche conservatism across the latitudinal gradient. We used environmental and climatic variables to characterize Grinnellian niches and diets to characterize Eltonian niches. We conducted variation partitioning with retained components from ordination methods to evaluate the degree of association of Grinnellian and Eltonian niche characteristics with passerine distribution across latitudes. We examined the relationship between phylogenetic signal and niche characteristics with a phylogenetic regression. Passerine distributions were more related to environmental gradients than resources across latitudes. While niche conservatism was prevalent in Eltonian niche characteristics, phylogeny was related to Grinnellian niche characteristics in only 46% of biomes. There was no latitudinal gradient in phylogenetic niche conservatism or the degree to which Eltonian and Grinnellian niche characteristics relate to passerine distribution. Niche conservatism, albeit weak, was present for Grinnellian niche characteristics, thus potentially influencing the expansion of passerine distributions into the northern hemisphere. Anthropogenic changes to the environment expose wildlife to many pollutants. Among these, tropospheric ozone is of global concern and a highly potent prooxidant. In addition, human activities include several other implications for wildlife, e.g., changed food availability and changed distribution of pathogens in cities. These co-occurring habitat changes may interact, thereby modulating the physiological responses and costs related to anthropogenic change. For instance, many food items associated with humans (e.g., food waste and feeders for wild birds) contain relatively more ω6-than ω3-polyunsaturated fatty acids (PUFAs). Metabolites derived from ω6-PUFAs can enhance inflammation and oxidative stress towards a stimulus, whereas the opposite response is linked to ω3-derived metabolites. Hence, we hypothesized that differential intake of ω6- and ω3-PUFAs modulates the oxidative stress state of birds and thereby affects the responses towards pro-oxidants. To test this, we manipulated dietary ω6: ω3 ratios and ozone levels in a full-factorial experiment using captive zebra finches (Taeniopygia guttata). Additionally, we simulated an infection, thereby also triggering the immune system's adaptive pro-oxidant release (i.e., oxidative burst), by injecting lipopolysaccharide. Under normal air conditions, the ω3-diet birds had a lower antioxidant ratio (GSH/GSSG ratio) compared to the ω6-diet birds. When exposed to ozone, however, the diet effect disappeared. Instead, ozone exposure overall reduced the total concentration of the key antioxidant glutathione (tGSH). Moreover, the birds on the ω6-rich diet had an overall higher antioxidant capacity (OXY) compared to birds fed a ω3-rich diet. Interestingly, only the immune challenge increased oxidative damage, suggesting the oxidative burst of the immune system overrides the other pro-oxidative processes, including diet. Taken together, our results show that ozone, dietary PUFAs, and infection all affect the redox-system, but in different ways, suggesting that the underlying responses are decoupled despite that they all increase pro-oxidant exposure or generation. Despite lack of apparent cumulative effect in the independent biomarkers, the combined single effects could together reduce overall cellular functioning and efficiency over time in wild birds exposed to pathogens, ozone, and anthropogenic food sources. Maternal signals shape embryonic development, and in turn post-natal phenotypes. RNA deposition is one such method of maternal signalling and circadian rhythms are one trait thought to be maternally inherited, through this mechanism. These maternal circadian gene transcripts aid development of a functioning circadian system. There is increasing evidence that maternal signals can be modified, depending on prevailing environmental conditions to optimise offspring fitness. However, currently, it is unknown if maternal circadian gene transcripts, and consequently early embryonic gene transcription, are altered by maternal developmental conditions. Here, using avian mothers who experienced either pre-natal corticosterone exposure, and/or post-natal stress as juveniles we were able to determine the effects of the timing of stress on downstream circadian RNA deposition in offspring. We demonstrated that maternal developmental history does indeed affect transfer of offspring circadian genes, but the timing of stress was important. Avian mothers who experienced stress during the first 2 weeks of post-natal life increased maternally deposited transcript levels of two core circadian clock genes, BMAL1 and PER2. These differences in transcript levels were transient and disappeared at the point of embryonic genome transcription. Pre-natal maternal stress alone was found to elicit delayed changes in circadian gene expression. After activation of the embryonic genome, both BMAL1 and PER2 expression were significantly decreased. If both pre-natal and post-natal stress occurred, then initial maternal transcript levels of BMAL1 were significantly increased. Objective: To evaluate the feasibility of the point‐of‐care viscoelastic coagulation monitor (VCM) in Amazon parrots (Amazona spp.) and describe the parameters with fresh whole‐blood samples in healthy Amazon parrots. Design: A total of 18 Amazon parrots were enrolled. Physical examinations, a CBC, and a biochemistry profile, including bile acids, were performed on all parrots. VCM tracings were obtained at the time of venipuncture for baseline laboratory work. Measurements and Main Results: The median clot time was 2102 seconds (range: 38.6–3599 s), median clot formation time was 929 seconds (range: 21.4–1711 s), median alpha angle was 20 (range: 6–67), and the median maximum clot formation was 8.5 (range: 0–36). The median lysis index at 30 minutes (LI30) was 100 (range: 98–100), and the median lysis index at 45 minutes (LI45) was 100 (range: 90–100). Of 18 samples, alpha angles were not reported in 7 samples, LI30 was not reported in 10 samples, and LI45 was not reported in 12 samples. Of the qualitative curves, 6 reflected normal mammalian curves, and the remainder were consistent with a hypocoaguable state. Conclusions: The results were markedly variable, with the majority of VCM tracings being hypocoagulable in comparison with reference intervals established for dogs and cats. Using these protocols, the VCM is not reliable in Amazon parrots. Future areas of investigation include altering the temperature during sample analysis, the use of activators, or an exchange of clotting reagents for an extrinsic pathway activator, which may contribute to the success of this device in avian species. Meat and eggs from chicken are the major source of animal protein for the human population. The cryopreservation of poultry species is needed to guarantee sustainable production. Here, we describe the existing cryopreservation technologies for avian reproductive cells using embryonic germ cells, spermatozoa and ovarian tissues. We outline strategies to reconstitute chicken breeds from their cryopreserved embryonic germ cells using surrogate hosts and discuss the perspectives for genetic conservation and reconstitution of chicken and wild avian species using surrogate host animals. Autonomous Recording Units (ARUs) are widely used to survey for a variety of taxa. This survey method allows for high spatial and temporal coverage but will typically include identification errors that can bias estimates of occupancy. In some instances, verifying all individual detections is prohibitive. To direct verification effort, we developed a model to estimate the probability that transcribers would agree on an identification. Agreement probability was positively influenced by transcriber skill, identification confidence, species commonness and some song types. In contrast, agreement probability was lower when an acoustic signal was classified as a trill. We evaluated our model on independent data where all species detections were verified, and verification effort (time) was quantified. Our model performed well at predicting transcriber agreement on independent data (AUC = 0.71). We applied the model to randomised subsets of the independent data to compare the cost benefit of three approaches to verification under varying effort. We show how modelling probability of transcriber agreement can be used to more efficiently direct verification of species acoustic tags. Our approach could be adapted elsewhere to quantify and reduce species misidentifications in unverified passive acoustic monitoring data for either manual processing or detections from automated classifiers. The present study was conducted to investigate the presence and distribution of the endocrine cells in the pancreas of the common pheasant using immunofluorescent staining. Totally, five adult male birds were used in the present work. The tissue samples were prepared, and different antibodies were applied to detect insulin, glucagon, somatostatin, pancreatic polypeptide (PP), and neuropeptide Y (NY). The percentage of staining was estimated using image J software. The glucagon-secreting cells (Alpha cells) with 32.4% ± 12.56 intensity were the most numerous cell type which were populated throughout the large islets especially at the periphery of islets. The insulin-secreting cells (Beta cells) with 27.73% ± 8.34 intensity were accumulated in the center of the small islets. Cells containing somatostatin (Delta cells) showed 13.38% ± 5.09 intensity. Despite their lower intensity, the distribution of D cells was like to A cells throughout the islets. PP cells and NY cells were distinguished with 19.55% ± 3.88 and 18.14% ± 7.15 intensities, respectively. Their distribution was evenly in the large islets. There were two types of islets: large alpha islets which were mainly composed of alpha and delta cells, and small beta islets constituted of beta cells and a few numbers of alpha cells. In conclusion, all investigated cell types were expressed in the endocrine pancreas and distributed in two types of islets as large alpha and small beta islets. Although some molecular epidemiological surveys on avian haemosporidian protozoa have been conducted in Turkey, it is of great importance to increase the number of studies on the subject in this country, which has important bird migration routes between Europe and Africa. Hatay province, which is the intersection point of many bird migration routes in the intercontinental crossing, is a very suitable geographical location for the investigation of avian haemosporidian protozoa in a wide variety of bird species that use it as a route. Therefore, this study was planned to survey Haemoproteus spp., Plasmodium spp., and Leucocytozoon spp. in migratory birds sampled from this province. The animal material of the study consisted of Ciconia ciconia (Ciconiformes, n:45), Ichthyaetus melanocephalus (Charadriiformes, n:2), Pelecanus onocrotalus (Pelecaniformes, n:5) and Pelecanus crispus (Pelecaniformes, n:1). Microscopic examinations revealed Haemoproteus spp. gametocytes (5.66%) in one white stork and two Mediterranean gulls. On the other hand, nested PCR assay targeting the mitochondrial cytochrome b (Cytb) gene revealed the presence of Plasmodium/Haemoproteus spp. in 11.3% (6/53) of migratory birds. Leucocytozoon spp. was not detected by either microscopy or PCR. PCRpositive products were sequenced bi-directionally, and the GenBank and MalAvi databases were used for phylogenetic analysis and lineage identification of the isolates. This study revealed H-MYCAME08 lineage (OR227579, OR227580, OR227581, OR227582) in Ciconia ciconia and H-LARCRA01 [H. (Parahaemoproteus) larae] lineage (OR227577, OR227578) in Ichthyaetus melanocephalus, respectively. There are no studies investigating the haemosporidian parasites of these birds in Turkey, and studies on haemosporidian parasites of these species in different countries are also limited. Although the Haemoproteus MYCAME08 Cytb lineage has been demonstrated in ciconiform birds (Ciconidae) and the LARCRA01 lineage in charadriiform birds (Laridae), this study is unique in terms of reporting these lineages in different species in the same families. Inspired by this, we envisage that the expanding host range of Cytb lineages of haemosporidian protozoa is an indication that there is still a long way to go with avian haemosporidian protozoa. Motivation to seek social interactions is inherent to all social species. For instance, even with risk of disease transmission in a recent pandemic, humans sought out frequent in-person social interactions. In other social animals, socialization can be prioritized even over water or food consumption. Zebra finches, Taeniopygia guttata , are highly gregarious songbirds widely used in behavioural and physiological research. Songbirds, like humans, are vocal learners during development, which rely on intense auditory learning. Aside from supporting song learning, auditory learning further supports individual identification, mate choice and outcome associations in songbirds. To study auditory learning in a laboratory setting, studies often employ operant paradigms with food restriction and reinforcement and require complete social isolation, which can result in stress and other unintended physiological consequences for social species. Thus, in this work, we designed an operant behavioural method leveraging the sociality of zebra finches for goal-directed behaviours. Our approach relies on visual social reinforcement, without depriving the animals of food or social contact. Using this task, we found that visual social reinforcement was a strong motivational drive for operant behaviour. Motivation was sensitive to familiarity towards the stimulus animal and higher when engaging with a familiar versus a novel individual. We further show that this tool can be used to assess auditory discrimination learning using either songs or synthetic pure tones as stimuli. As birds gained experience in the task, they developed a strategy to maximize reward acquisition in spite of receiving more punishment, i.e. liberal response bias. Mass mortality events in wildlife can be indications of an emerging infectious disease. During the spring and summer of 2021, hundreds of dead passerines were reported across the eastern US. Birds exhibited a range of clinical signs including swollen conjunctiva, ocular discharge, ataxia, and nystagmus. As part of the diagnostic investigation, high-throughput metagenomic next-generation sequencing was performed across three molecular laboratories on samples from affected birds. Many potentially pathogenic microbes were detected, with bacteria forming the largest proportion; however, no singular agent was consistently identified, with many of the detected microbes also found in unaffected (control) birds and thus considered to be subclinical infections. Congruent results across laboratories have helped drive further investigation into alternative causes, including environmental contaminants and nutritional deficiencies. This work highlights the utility of metagenomic approaches in investigations of emerging diseases and provides a framework for future wildlife mortality events. [ABSTRACT FROM AUTHOR]

The aim of this study was to describe the gross and histopathological features of a neurological syndrome in endangered Western Australian Carnaby's black cockatoos (Zanda laitirostris) that was first observed in 2012. The syndrome, named hindlimb paralysis syndrome in Carnaby's cockatoos (CHiPS), is characterized by annual outbreaks of hindlimb paralysis with occasional loss of deep pain and cloacal tone, typically occurring between January and March. Previous limited investigations suggested a possible toxic aetiology. Full gross necropsy and histopathology examinations were performed on 17 CHiPS cases and on 11 control birds for reference. Histopathological examination was carried out on all major organs including brain, spinal cord, brachial plexus, sciatic nerve and wing and hindlimb muscles. Gross and histopathological examinations did not elucidate a definitive cause of the clinical signs seen in CHiPS cases. There were no substantial gross or histopathological changes within the brain, spinal cord, sciatic nerve or brachial plexus that could explain the hindlimb paralysis. The most noteworthy changes were seen in the hindlimb and wing muscles, with a monophasic to polyphasic myopathy present in the hindlimb muscles of 15 of the 17 CHiPS cases and in the wing muscles in 11 of those cases. The cause and significance of the myopathy is unclear and requires further investigation. Based on the above findings, the most likely differential diagnoses include neurotoxicoses (eg, organophosphate, organochlorine and carbamate) and, less likely, myotoxicosis (eg, ionophore toxicosis), nutritional myopathy (eg, vitamin E/selenium deficiency) or botulism. The amniote eggshell is a fundamental aspect of the embryo life‐support system, protecting it from UV light, microbes and mechanical damage, while regulating gas exchange and providing calcium for growth. The thickness of eggshells is highly diverse across modern birds and influences multiple eggshell functions, yet the selective pressures driving eggshell thickness have not been clearly identified. Here, we use a global dataset of avian eggshell thickness indices for 4260 (> 41%) avian species to assess trends in eggshell thickness across the phylogeny, as these indices are strongly correlated with direct measures of eggshell thickness and are non‐destructive to the sample. We analysed the dataset within a phylogenetic framework to assess the relative importance of climatic and ecological explanations for variation in eggshell thickness indices. The distribution of avian eggshell thickness indices across species was found to be primarily driven by phylogenetic relatedness, in addition to evolutionary processes that do not match a Brownian model of evolution. Across modern birds, thicker eggshells were more prevalent in species (1) with precocial young, (2) which exhibit a scavenger‐based diet, (3) which primarily feed on vertebrates or plants (excluding nectivores, seed and fruit specialists) and (4) which breed in open habitats. Thicker eggshells found in species with precocial young probably enable higher rates of calcium removal for the more advanced embryo development. Excessive light transmission through the shell damages developing embryos, while too little light transmission can impede development. Eggs in shaded habitats experience low light exposure, and thus thinner shells are more prevalent in these environments potentially to increase light transmission through the shell. Overall, variation in eggshell thickness indices appears to be driven largely by phylogeny, with certain life‐history traits linked to embryo growth rate, calcium content of their food, and the need to mitigate excessive light transmission through the shell. Wetlands in southern Ontario are at risk of degradation and alteration due to human activities. This is a concern because wetlands provide essential habitat for species from a range of taxa, such as birds and anurans (frogs and toads). One wetland-dwelling species whose decline may be linked to loss of wetland wildlife habitat is the muskrat (Ondatra zibethicus). The decline of muskrats may also be linked to declines of other species because muskrats engage in activities that could support taxa such as birds and anurans by increasing habitat heterogeneity. We investigated whether bird and anuran species richness is related to muskrat density or if it is better predicted by land cover variables that describe the wetland and surrounding area at 30 coastal wetlands on Lake Ontario. We estimated bird and anuran species richness using data from the Great Lakes Marsh Monitoring Program and the Great Lakes Coastal Wetland Monitoring Program, and muskrat density based on muskrat house counts. Our results suggest that muskrat activity did not predict richness of anurans or birds overall; however, it did predict richness for the subset of birds that nest in emergent aquatic vegetation. Our results indicate that muskrat abundance in emergent marshes may increase habitat quality for birds that nest in emergent vegetation but may not have a measurable effect on anuran diversity in these same wetlands. Urban environments have been characterized by their temporal stability of resources, which could promote stability in bird composition. Several studies have found that bird communities in urban environments persist over the years, showing a similar species composition in the short term. However, studies analyzing continuous changes in urban communities over the long term are scarce. This study aimed to analyze the stability or directional changes (instability) in bird communities along an urban gradient. Bird counts were conducted in urban, suburban, and periurban areas over 8-10 years in 2002-2019. Changes in species composition were analyzed over periods ranging from one year to the next, to changes from one year to the seventeenth. Urban bird communities were more similar between years than suburban and periurban communities. Compositional changes were greater as time lags increased, indicating directional compositional shifts. The magnitude of these changes was similar across the urban gradient. The Chimango Caracara (Milvago chimango), the Picazuro Pigeon (Patagioenas picazuro), the Rufous Hornero (Furnarius rufus), and the Red-bellied Thrush (Turdus rufiventris) significantly increased their abundances during the period, while the House Sparrow (Passer domesticus) significantly decreased its abundance. Regional changes in species abundance, urban vegetation succession, and biotic interactions could explain the changes in bird communities.

In recent years, avian medicine has advanced in the recognition of kidney diseases, but little is known about the morphology and renal morphometry of the common ostrich Struthio camelus Linnaeus, 1758. This study aimed to describe the morphometry and location of kidneys in the common ostrich. Twenty-six cadavers, aged one to seven days, which died of natural causes on a farm in the municipality of Magé, state of Rio de Janeiro, Brazil, were donated to the Department of Animal and Human Anatomy at the Universidade Federal Rural do Rio de Janeiro (UFRRJ). The specimens were identified, sexed, and fixed in a 10% aqueous solution of formaldehyde. The kidneys and renal divisions were measured using a precision digital caliper. The kidneys of S. camelus are elongated, and lobular, and have three renal divisions (cranial, middle, and caudal). The average length of the right kidney was 6.69 ± 0.83 cm, and the left kidney was 6.87 ± 0.81 cm. The body length correlated positively and significantly with the lengths of the right kidney (r = 0.8273, p = 0.0017) and left kidney (r = 0.8534, p = 0.0008) in females. The renal cranial limit was observed at the eighth lumbar vertebra (L8) in 22 kidneys (42.3%), and the caudal limit, at the eighth sacrocaudal vertebra (Sc8) in 11 kidneys (21.2%), and the second caudal vertebra (C2) in 11 kidneys (21.2%). The morphometric dimensions and skeletopy of the ostrich kidney contribute to the field of comparative avian anatomy and assist in the interpretation of imaging techniques.

Abstract The Diederik cuckoo, Chrysococcyx caprius, is a small Afrotropical bird in the family Cuculidae. It is taxonomically related to 13 other species within the genus Chrysococcyx and is migratory in sub‐Saharan Africa. It has a unique breeding behaviour of being a brood parasite: Breeding pairs lay their eggs in the nests of a host species and hatchlings expel the eggs of the host species. The aim of the present study was to investigate diversity in two circadian clock genes, Clock and Adcyap1, to probe for a relationship between genetic polymorphisms and their role in circannual timing and habitat selection (phenology) in intra‐African migrants. DNA extracted from blood was used for the PCR amplification and sequencing of clock genes in 30 Diederik cuckoos. Three alleles were detected for Clock with similar genotypes between individuals from the Northern and Southern breeding ranges while 10 alleles were detected for Adcyap1, having shorter alleles in the North and longer alleles in the South. Population genetic analyses, including allele frequency and zygosity analysis, showed distinctly higher frequencies for the most abundant Clock allele, containing 10 polyglutamine repeats, as well as a high degree of homozygosity. In contrast, all individuals were heterozygous for Adcyap1 and alleles from both regions showed distinct differences in abundance. Comparisons between both clock genes and phenology found several phenotypic correlations. This included evidence of a relationship between the shorter alleles and habitat selection as well as a relationship between longer alleles and timing. In both instances, evidence is provided that these effects may be sex‐specific. Given that these genes drive some of the synchronicity between environments and the life cycles of birds, they provide valuable insight into the fitness of species facing global challenges including climate change, urbanisation and expanding agricultural practices.

Four Indian ringneck parakeets (Psittacula krameri; syn. ringneck parrots or rose-ringed parakeets) were submitted by 2 private owners for autopsy following a history of dyspnea and death. Gross findings were varied and included thickening of the left caudal thoracic air sac, white spots throughout the liver, mild dilation of the proventriculus, coelomic effusion, splenomegaly, and pulmonary congestion and edema. Microscopically, the submitted parakeets had significant lesions in the lower respiratory tract, including necrotizing bronchitis, parabronchitis, and interstitial pneumonia with numerous syncytia containing eosinophilic intranuclear inclusions. Electron microscopy of the lungs was compatible with a herpesviral infection and Psittacid alphaherpesvirus 5 (PsAHV5) was detected via PCR and sequencing. There has been inconsistent terminology used with Psittacid alphaherpesvirus 3 and PsAHV5; we attempt here to clarify the reported history of these viruses.

Haemosporidian parasites are prevalent worldwide and can cause economic losses in poultry production. These parasites are arousing interest in Thailand and are found in many avian species. There is insufficient information on the genetic diversity of these alveolates from the largest families – Plasmodidae, Haemoprotidae and Leucocytozoidae – specifically parasitising ducks, turkeys, and geese.

Background and Aim: The pathogenesis of staphylococcal infections is mediated by virulence factors, such as enzymes, toxins, and biofilms, which increase the resistance of microorganisms to host immune system evasion. Testing and searching for standardized multi-level algorithms for the indication and differentiation of biofilms at the early stages of diagnosis will contribute to the development of preventive measures to control the critical points of technology and manage dangerous risk factors for the spread of infectious diseases. This research aimed to study the main stages of Staphylococcus aureus biofilm formation in in vitro experiments and to analyze the dynamics of respiratory syndrome development in chickens infected with these bacteria. Materials and Methods: Experimental reproduction of the infectious process was performed using laboratory models: 10-day-old White Leghorn chickens (n = 20). Before the experiments, the birds were divided into two groups according to the principle of analogs: Group I (control, n = 10): the birds were intranasally inoculated with 0.5 cm3 of 0.9% NaCl solution; Group II (experiment, n = 10): the birds were intranasally inoculated with a suspension of S. aureus bacteria, 0.5 cm3, concentration 1 billion/cm3. Results: Colonization of individual areas of the substrate under study in vitro occurred gradually from the sedimentation and adhesion of single motile planktonic cells to the attachment stage of microcolony development. Staining preparations with gentian violet due to the “metachromosia†property of this dye are a quick and fairly simple way to differentiate cells and the intercellular matrix of biofilms. Fixation with vapors of glutaraldehyde and osmium tetroxide preserves the natural architecture of biofilms under optical and scanning electron microscopy. Pure cultures of S. aureus microorganisms were isolated from the blood, lungs, small intestine, liver, kidneys, and spleen after 5–10 days during experimental infection of chickens. Clinical signs of respiratory syndrome developed within 5–6 days after infection. Acute and subacute serousfibrinous airsacculitis, characterized by edema and thickening of the membranes of the air sacs and the presence of turbid, watery, foamy contents in the cavity, was the most characteristic pathomorphological sign. The signs of acute congestive hyperemia and one-sided serous-fibrinous pneumonia developed with significant thickening of fibrinous deposits. In Garder’s gland, there was an increase in the number of secretory sections, indicating hypersecretion of the glands. In the lymphoid follicles of Meckel’s diverticulum, leukocytes, usually lymphocytes, and pseudoeosinophils were detected. Conclusion: Hydration and heteromorphism of the internal environment of biofilms determine the localization of differentiated cells in a three-dimensional matrix for protection against adverse factors. The most characteristic pathomorphological sign was the development of acute and subacute serous-fibrinous airsacculitis when reproducing the infectious process in susceptible models. There was a significant thickening of fibrinous deposits and signs of acute congestive hyperemia and one or two serous-fibrinous pneumonia developed.

Populations of the species Artemia (Brine shrimp) in saltworks have become a popularity popular field of study for aquaculture for its significance for aquatic ecosystems as being an important source of sustenance for water birds in hypersaline food webs. Besides, species in the genus Artemia are the intermediate host of severe cestode species which are associated with flamingos. This study reports on the prevalence of native Artemia parthenogenetica parasitism of Flamingolepis liguloides in the Çamaltı saltern ecosystem in the Gediz wetland between January and December 2022 in Türkiye. Infected A. parthenogenetica was sampled from April to September 2022 in salt pans where flamingo birds and salt production are available for parasitological diagnosis. The parasites were determined in the abdomen, thorax and near the gut tract of A. parthenogenetica. The highest prevalence of parasite was found in juvenile individuals as 86.67±1.45% and adults as 76.06±1.16% in May. The main intensity was varied from 1.18±0.01 to 1.92±0.06 parasites per infected host depending on the sampling months and age of the brine shrimp. The most abundant parasite infestation was recorded as 1.44±0.02 parasites per investigated host in juvenile brine shrimp in June since seasonal conditions are favorable for such infestation.

Abstract Background Central place foragers must acquire resources and return to a central location after foraging bouts. During the egg laying (hereafter laying) period, females are constrained to a nest location, thus they must familiarize themselves with resources available within their incubation ranges after nest site selection. Use of prospecting behaviors by individuals to obtain knowledge and identify profitable (e.g., resource rich) locations on the landscape can impact demographic outcomes. As such, prospecting has been used to evaluate nest site quality both before and during the reproductive period for a variety of species. Methods Using GPS data collected from female eastern wild turkeys (Meleagris gallopavo silvestris) across the southeastern United States, we evaluated if prospecting behaviors were occurring during laying and what landcover factors influenced prospecting. Specifically, we quantified areas prospected during the laying period using a cluster analysis and the return frequency (e.g., recess movements) to clustered laying patches (150-m diameter buffer around a clustered laying period location) during the incubation period. Results The average proportion of recess movements to prospected locations was 56.9%. Nest fate was positively influenced (μ of posterior distribution with 95% credible 0.19, 0.06–0.37, probability of direction = 99.8%) by the number of patches (90-m diameter buffer around a clustered laying period location) a female visited during incubation recesses. Females selected for areas closer to the nest site, secondary roads, hardwood forest, mixed pine-hardwood forest, water, and shrub/scrub, whereas they avoided pine forest and open-treeless areas. Conclusions Our findings suggest that having a diverse suite of clustered laying patches to support incubation recesses is impactful to nest fate. As such, local conditions within prospected locations during incubation may be key to successful reproductive output by wild turkeys. We suggest that prospecting could be important to other phenological periods. Furthermore, future research should evaluate how prospecting for brood-rearing locations may occur before or during the incubation period.

A diverse population of avian influenza A viruses (AIVs) are maintained in wild birds and ducks yet the zoonotic potential of AIVs in these environmental reservoirs and the host-virus interactions involved in mammalian infection are not well understood. In studies of a group of subtype H1N1 AIVs isolated from migratory wild birds during surveillance in North America, we previously identified eight amino acids in the polymerase genes PB2 and PB1 that were important for the transmissibility of these AIVs in a ferret model of human influenza virus transmission. In this current study we found that PB2 containing amino acids associated with transmissibility at 67, 152, 199, 508, and 649 and PB1 at 298, 642, and 667 were associated with more rapid viral replication kinetics, greater infectivity, more active polymerase complexes and greater kinetics of viral genome replication and transcription. Pathogenicity in the mouse model was also impacted, evident as greater weight loss and lung pathology associated with greater inflammatory lung cytokine expression. Further, these AIVs all contained the avian-type amino acids of PB2-E627, D701, G590, Q591 and T271. Therefore, our study provides novel insights into the role of the AIV polymerase complex in the zoonotic transmission of AIVs in mammals.

ABSTRACT: Direct introduction of cryopreserved embryonic gonadal germ cells (GGC) into a sterile chicken surrogate host to reconstitute a chicken breed has been demonstrated as a feasible approach for preserving and utilizing chicken genetic resources. This method is highly efficient using male gonads; however, a large number of frozen female embryonic gonads is needed to provide sufficient purified GGC for the generation of fertile surrogate female hosts. Applying this method to indigenous chicken breeds and other bird species is difficult due to small flock numbers and poor egg production in each egg laying cycle. Propagating germ cells from the frozen gonadal tissues may be a solution for the biobanking of these birds. Here, we describe a simplified method for culture of GGC from frozen embryonic 9.5 d gonads. At this developmental stage, the germ cells are autonomously shed into medium, yielding hundreds to thousands of mitosis-competent germ cells. The resulting cultures of GGC have over 90% purity, uniformly express SSEA-1 and DAZL antigens and can re-colonize recipient's gonads. The GGC recovery rate from frozen gonads are 42% to 100%, depending on length of cryopreservation and the breed or line of chickens. Entire chicken embryos can also be directly cryopreserved for later gonadal isolation and culture. This storage method is a supplementary approach to safeguard local indigenous chicken breeds bearing valuable genetic traits and should be applicable to the biobanking of many bird species.

Avian influenza viruses (AIV) pose a continuous challenge to global health and economy. While countermeasures exist to control outbreaks in poultry, the persistent circulation of AIV in wild aquatic and shorebirds presents a significant challenge to effective disease prevention efforts. PB1-F2 is a non-structural protein expressed from a second open reading frame (+1) of the polymerase basic 1 (PB1) segment. The sequence and length of the PB1-F2 protein can vary depending on the host of origin. While avian isolates typically carry full-length PB1-F2, isolates from mammals, often express truncated forms. The selective advantage of the full-length PB1-F2 in avian isolates is not fully understood. Most research on the role of PB1-F2 in influenza virus replication has been conducted in mammalian systems, where PB1-F2 interfered with the host immune response and induced apoptosis. Here, we used Low Pathogenicity (LP) AIV H7N7 expressing full-length PB1-F2 as well as a knockout mutant. We found that the full-length PB1-F2 of LPAIV prolonged survival of infected cells by limiting apoptotic cell death. Furthermore, PB1-F2 knockout LPAIV significantly decreased MHC-I expression on fibroblasts, delayed tissue healing and increased phagocytic uptake of infected cells, whereas LPAIV expressing PB1-F2 has limited effects. These findings indicate that full-length PB1-F2 enables AIV to cause prolonged infections without severely harming the avian host. Our observations may explain maintenance of AIV in the natural bird reservoir in absence of severe clinical signs.

Urbanization is resulting in habitat loss and fragmentation around the world, but its effects on biodiversity remain poorly understood. We investigated how the increase of urban cover surrounding nine Brazilian Atlantic forest reserves (14 ha - 1058 ha) affects the taxonomic, functional and phylogenetic diversity of bird communities, considering forest and non-forest species separately. Using a multi-scale local landscape approach, we estimated the percentage of urban cover in circular buffers of varying radii (200–1000 m). We calculated 10 complementary metrics of bird diversity and modelled them against the percentage of urban cover using generalized additive models. Urbanization had a negative, non-linear impact on the taxonomic diversity of bird communities, with a sharp decrease of up to 50 % in the effective number of rare, typical and dominant species in landscapes with 20–40 % of urbanization. This decrease was not restricted to forest species (urban avoiders), but also affected non-forest ones (urban utilizers and dwellers). The impact of urbanization on functional and phylogenetic diversity were less evident, suggesting that the vulnerability to urbanization has a weak phylogenetic and phenotypic signal. We conclude that urbanization impacts a broad spectrum of bird clades and traits and drastically reduce the taxonomic diversity of bird communities.

The applicability of electrical impedance tomography (EIT) in birds is unknown. This study aimed to evaluate the use of EIT in anaesthetised chickens in four recumbency positions. Four adult Hyline chickens were anaesthetised with isoflurane in oxygen, and intubated endotracheally for computed tomography (CT). A rubber belt was placed around the coelom caudal to the shoulder joint. A chicken-specific finite element (FE) model, which is essential to generate anatomically accurate functional EIT images for analysis, was constructed based on the CT images obtained at the belt level. Ten additional chickens were anaesthetised with the same protocol. An EIT electrode belt was placed at the same location. The chickens were breathing spontaneously and positioned in dorsal, ventral, right and left lateral recumbency in a randomised order. For each recumbency, raw EIT data were collected over 2 min after 13 min of stabilisation. The data were reconstructed into functional EIT images. EIT variables including tidal impedance variation (TIV), centre of ventilation right to left (COVRL) and ventral to dorsal (COVVD). right to left (RL) ratio, impedance change (ΔZ) and eight regional impedance changes including the dorsal, central-dorsal, central-ventral and ventral regions of the right and left regions were analysed. Four breathing patterns (BrP) were observed and categorised based on the expiratory curve. A linear mixed model was used to compare EIT variables between recumbencies. Fisher's exact test was used to compare the frequencies of breathing patterns for each recumbency. The ΔZ observed was synchronous to ventilation, and represented tidal volume of the cranial air sacs as confirmed by CT. Significant differences were found in CoVVD and regional impedance changes between dorsal and ventral recumbencies (P < 0.05), and in COVRL, RL ratio and regional impedance changes between right and left recumbencies (P < 0.05), which suggested a tendency for the distribution of ventilation to shift towards non-dependent air sacs. No differences were found for TIV and respiratory rate between recumbencies. Recumbency had a significant effect on the frequencies of each of the four BrPs (P = 0.001). EIT can monitor the magnitude and distribution of ventilation of the cranial air sacs in different recumbencies in anaesthetised chickens. [ABSTRACT FROM AUTHOR]

Background and Aim: The pathogenesis of staphylococcal infections is mediated by virulence factors, such as enzymes, toxins, and biofilms, which increase the resistance of microorganisms to host immune system evasion. Testing and searching for standardized multi-level algorithms for the indication and differentiation of biofilms at the early stages of diagnosis will contribute to the development of preventive measures to control the critical points of technology and manage dangerous risk factors for the spread of infectious diseases. This research aimed to study the main stages of Staphylococcus aureus biofilm formation in in vitro experiments and to analyze the dynamics of respiratory syndrome development in chickens infected with these bacteria. Materials and Methods: Experimental reproduction of the infectious process was performed using laboratory models: 10-day-old White Leghorn chickens (n = 20). Before the experiments, the birds were divided into two groups according to the principle of analogs: Group I (control, n = 10): the birds were intranasally inoculated with 0.5 cm3 of 0.9% NaCl solution; Group II (experiment, n = 10): the birds were intranasally inoculated with a suspension of S. aureus bacteria, 0.5 cm3, concentration 1 billion/cm3 . Results: Colonization of individual areas of the substrate under study in vitro occurred gradually from the sedimentation and adhesion of single motile planktonic cells to the attachment stage of microcolony development. Staining preparations with gentian violet due to the "metachromosia" property of this dye are a quick and fairly simple way to differentiate cells and the intercellular matrix of biofilms. Fixation with vapors of glutaraldehyde and osmium tetroxide preserves the natural architecture of biofilms under optical and scanning electron microscopy. Pure cultures of S. aureus microorganisms were isolated from the blood, lungs, small intestine, liver, kidneys, and spleen after 5-10 days during experimental infection of chickens. Clinical signs of respiratory syndrome developed within 5-6 days after infection. Acute and subacute serousfibrinous airsacculitis, characterized by edema and thickening of the membranes of the air sacs and the presence of turbid, watery, foamy contents in the cavity, was the most characteristic pathomorphological sign. The signs of acute congestive hyperemia and one-sided serous-fibrinous pneumonia developed with significant thickening of fibrinous deposits. In Garder's gland, there was an increase in the number of secretory sections, indicating hypersecretion of the glands. In the lymphoid follicles of Meckel's diverticulum, leukocytes, usually lymphocytes, and pseudoeosinophils were detected. Conclusions: Hydration and heteromorphism of the internal environment of biofilms determine the localization of differentiated cells in a three-dimensional matrix for protection against adverse factors. The most characteristic pathomorphological sign was the development of acute and subacute serous-fibrinous airsacculitis when reproducing the infectious process in susceptible models. There was a significant thickening of fibrinous deposits and signs of acute congestive hyperemia and one or two serous-fibrinous pneumonia developed. [ABSTRACT FROM AUTHOR]

Bird wings vary in size and morphology in terms of both size and number of feathers and the underlying skeletal anatomy. The number of primary remiges does not seem to vary much between bird species but, by contrast, the number of secondary remiges is reported to range between 6 and 40 depending on bird size. Given that the primaries are attached to the manus, and the secondaries are attached to the ulna, it was predicted that as bone lengths increased with increasing size of the bird, then feather count would increase. Data were collected for 268 species from 25 different orders, and phylogenetically controlled analysis explored the allometry between feather count and bone size. The number of primaries was typically 10 or 11 and did not vary with manus size. By contrast, the number of secondaries increased with ulna length, but only in some orders. For example, in Gruiformes, the number of secondary feathers increased concomitantly with ulna length but despite a two orders of magnitude range in body mass, almost all species in the Passeriformes had nine secondary remiges. It is unclear why, for instance, species with an ulna length of 70 mm can have between 9 and 24 secondaries depending on their order. This variation in secondary feather number can be added to variation in relative wing bone lengths, flight feather lengths, flight feather mechanical properties, and flight feather vane densities as another potential mechanism of adaptation to flight requirements. The apparent constraint of wingspan is scaling as approximately body mass1/3. Further research is needed to explore whether changes in secondary feather number relative to ulna length are accompanied by changes in feather vane width or the overlap of adjacent feathers and how this relates to wing aerodynamics.
