Chocolate agar (CA) is an enriched medium for the isolation and identification of fastidious bacteria. Defibrinated blood is used to manufacture CA, but this expensive product is not always affordable for companies in developing countries. Blood powder (BP) is potentially a cheaper alternative, although its pre-treatment using autoclaving can impair the quality of the media. Therefore, optimization of BP as a substitute for defibrinated blood for CA manufacture deserves further research. CA was manufactured with irradiated BP (dehydrated bovine blood powder) and its physical and microbiological characteristics were compared with those of conventional CA and CA prepared with autoclaved BP. Each medium was seeded with 20–200 CFU of target bacteria using the spiral pouring method. Finally, another medium was prepared using BP pre-treated by grinding and gamma irradiation and its performance assessed. Compared to conventional CA, the medium containing ground and irradiated BP provided a similar CFU count for both fastidious (Neisseria, Haemophilus, Campylobacter, and Streptococcus) and non-fastidious (Moraxella, Staphylococcus, Enterococcus, Klebsiella, and Pseudomonas) species, unlike the medium prepared with BP subjected only to irradiation, which provided a lower growth of fastidious species. Morphology and characteristics of all bacterial colonies were very similar in conventional CA and the new medium, the number of Pseudomonas CFU being higher in the latter. The medium prepared with ground plus irradiated vs. irradiated BP more closely resembled conventional CA, having a browner background. The new CA medium prepared with ground and gamma irradiation-sterilized BP has comparable productivity properties to conventional CA. This article describes the use of recycled glass sand in the production of autoclaved products. Traditional autoclaved bricks consist of crystalline sand, lime and water. The conducted research aimed at the complete elimination of quartz sand in favor of glass sand. This work focuses on porosity as the functional property of the materials. The aim of this article is to determine the number and structure of the pores of autoclaved bricks. Two types of research were carried out: (a) non-destructive, i.e., computed tomography examination as a pictorial and quantitative method and (b) mercury porosimetry as a quantitative method, i.e., a test that exposes the porous skeleton of the material for destruction. The tests showed the presence of pores with a size in the range of 0.1 ÷ 100 μm, and the volume of voids in the material was determined at the level of about 20% for the sample modified with glass sand (GS) and for the reference sample made of traditional silicate brick. In order to complete the research on the internal structure of autoclaved bricks, microstructure studies were performed using a scanning electron microscope (SEM). The tests showed the presence of tobermorite in the reference sample (with 90% QS-quartz sand) and the presence of natrolite and gyrolite in the sample modified by glass sand (90% GS). Biomaterials designed for tissue engineering should be nontoxic and nonimmunogenic and should achieve their intended functions. Treated dentin matrix (TDM), a bioactive extracellular matrix, is promising for tooth regeneration. However, the effect of sterilization on the surface properties of allogenous TDM in the animal model is unclear. The biological characteristics and influences of dental pulp stem cells (DPSCs) with autoclaved TDM (a-TDM) were studied using scanning electron microscopy, immunofluorescence microscopy, immunohistochemistry, and reverse transcription polymerase chain reaction in vitro. In addition, a-TDM was implanted in a mouse model for 6 weeks and was a substrate with DPSCs for tooth reconstruction in a goat animal model in vivo. Allogenous a-TDM induced and supported DPSCs to develop new dentin pulp–like tissues, enamel dental pulp, and cementum periodontal complexes. Immunohistochemistry data showed that the markers dentin sialoprotein, βⅢ-tubulin, dentin matrix protein 1, amelogenin, VIII factors, type I collagen, cementum-derived attachment protein, and scleraxis transcription factor were positive stained in toothlike tissue. Allogenous a-TDM served as an effective scaffold enabling DPSCs to proliferate and differentiate into a broad array of resident cells including odontoblasts, fibroblasts, vascular cells, and neural endings. The use of environmental and sustainable ornamental flower production practices with renewable resources has drawn worldwide interest. One of these renewable resources is vermicompost (earthworm castings). In recent years, increasing demand for improving environmental quality have focused on the importance of Plant Growth Promotion Bacteria (PGPBs) in agriculture. Vermicomposts also help microbial agents function effectively in soil. In this study, a total of six treatments [A: PGPB formulation, B: Not autoclaved vermicompost, C: Autoclaved vermicompost, D: Not autoclaved vermicompost+PGPBs, E: Autoclaved vermicompost+PGPBs, F: Control (untreated bacteria and vermicompost)] were tested for their effects on the plant growth and development parameters in gladiolus (Gladiolus grandiflorus L. ‘Red Beauty’) in greenhouse condition. Vermicompost was added to the related pots by dissolving in water. After the addition of vermicompost, PGPB formulation was given immediately to related pots. All the treatments were applied to soil once in three leaf stage, close to the plant root zone. Parameters in terms of yield and quality attributes of plant and corm were determined and analyzed. The treatment A increased in plant height of gladiolus of 24.55% rate. The earliest times to flowering was determined in E application (100.48 day), which also increased in corm diameter with rate of 17.41% and number of corms and cormels with rate of 151.83% according to F application. Results indicated that the treatment E promoted overall better performance as compared to other treatments diameter of flowers for number of leaves per plant, number of florets per spike, stem diameter, spike length, fresh and dry weight of flowers, the number and diameter of corm. Autoclaved vermicompost can be good choice in gladiolus cultivation but it should be enriched with PGPB.

The assessment of water-extractable organic matter using an autoclave can provide useful information on physical, chemical, and biological changes within the soil. The present study used virgin forest soils from Chini Forest Reserve, Langkawi Island, and Kenyir Forest Reserve (Malaysia), extracted using different extraction methods. The dissolved organic carbon (DOC), total dissolved nitrogen (TDN), total dissolved phosphorus (TDP), and ammonium-nitrate content were higher in the autoclave treatments, up to 3.0, 1.3, 1.2, and 1.4 times more than by natural extraction (extracted for 24 h at room temperature). Overall, the highest extractable DOC, TDN, TDP, ammonium and nitrate could be seen under autoclaved conditions 121 °C 2×, up to 146.74 mg C/L, 8.97 mg N/L, 0.23 mg P/L, 5.43 mg N mg/L and 3.47 N mg/L, respectively. The soil extracts became slightly acidic with a higher temperature and longer duration. Similar trends were observed in the humic and nonhumic substances, where different types of soil extract treatments influenced the concentrations of the fractions. Different soil extraction methods can provide further details, thus widening the application of soil extracts, especially in microbes.

This paper describes the use of glass sand in the production of autoclaved bricks. Traditional autoclaved materials consist of SiO2, CaO, and H2O. The purpose of the tests is to analyze the possibility of using glass sand in autoclaved materials and to determine their properties and durability. Depending on the structure, building materials can have porosities ranging from 0% (glass, metals) to over 90% (thermal insulation materials such as aerated concrete). Porosity of materials is directly related to the strength of materials and their density, and further to the thermal and acoustic insulation properties of products used especially for external wall construction, i.e., bricks, concrete, and aerated concrete. This type of silicate brick is formed at a temperature of 203 °C, therefore the dominant phase forming the microstructure is tobermorite, in contrast to the C-S-H phase, which dominates in concretes and which is characterized by a larger specific surface. The nature of pores, their number, appearance and arrangement in the material can be studied using computer techniques (SEM, XRD, computed tomography, porosimetry). Computed tomography (micro-CT analysis) showed that the number of voids in the material modified by glass sand is about 20% in relation to the weight of the product. The density of the product with glass sand was determined to be 2.2 kg/dm3.

This paper adopts an experimental approach on the use of rice husk ash as a partial replacement of cement in the production of concrete and the consequent effects on density, compressive strength and split tensile strength of the formulated product. Rice husk ash is used as replacement of cement in different dosages of 0%, 2.5%,5%, 7.5%, 10%, 12.5%, and 15%. Results showed that 10% replacement of cement with rice husk ash is optimum. At 10% replacement of cement with RHA, the density is increased by 5.02%, the compressive strength by 22.22% and the split tensile strength by 20.45% in comparison with control aerated concrete. Both agriculture wastes, black rice husk ash (BRHA) and bagasse ash (BA), were used as sand replacement to prepare autoclaved aerated concrete (AAC) product at various temperatures and times. The compressive strengths of AAC were increased with the further autoclaving temperatures and times, except dry density. BRHA and BA have influence on the mechanical properties of AAC with low dry density for all conditions, approximately 6–54%, due to the high Na 2 O and K 2 O content and the fineness of both ashes compared to sand. The appearance of relative high Al 2 O 3 of BA compared to BRHA leads to the additional strength gain of AAC sample containing BA, because of the formation of tobermorite through katoite. The aim of this study was to evaluate the effects of two species of poultry (cockerel and Japanese quail), two levels of enzyme (0 and 0.5 g/kg), and 6 processing methods (control (C), gamma irradiation 25 (GI25), and 50 kGy (GI50), autoclaving (AU), fermentation without lactobacillus (F), and fermentation with lactobacillus (FL)) on the nitrogen-corrected apparent metabolizable energy (AMEn) of barley. In this experiment, each processing method was tested at two levels of enzyme (0 and 0.5 g/kg) in six replications with two cockerels or four quails per replication. The results showed that the AMEn values by barley processing methods were: C-11.45 and 12.27; GI25-11.50 and 11.30; GI50-11.49 and 11.29; AU-11.70 and 11.46; F- 12.90 and 12.30; FL-1295 and 12.51 MJ/kg in cockerels and quails, respectively. The AMEn (11.49 MJ/kg) of enzyme 0 was significantly lower than that of enzyme 0.5 (12.35 MJ/kg). There was a significant difference in the AMEn of processed barley between cockerels and quails. This paper presents the methodology used for manufacturing the Zircaloy-4 tubes required for the Zircaloy-4 mock-up dissolver assembly. The evolution of the microstructure at different stages of production of Zircaloy-4 tubes was characterized using scanning electron microscopy (SEM). Microstructural evolution follows the sequence as dendritic structure (as-cast ingot) → Widmanstätten structure (β-quenched) → bimodal grain size (hot extruded) → heterogeneous deformed structure (pilgered) → partially recrystallized structure (final annealed). High strength and low ductility were obtained for the Zircaloy-4 tubes at pilgered condition due to grain size refinement and work hardening during the pilgering process. Compared with the pilgering stage, the Zircaloy-4 tubes in the final annealed condition exhibited moderate strength and high ductility due to the partially recrystallized microstructure. Autoclaving was carried out to improve the corrosion properties of the pilgered Zircaloy-4 tubes in boiling 11.5 M nitric acid. When exposed to boiling 11.5 M nitric acid for 1000 h, a lowest corrosion rate of 0.003 mpy was obtained for autoclaved Zircaloy-4 tubes. Laser Raman Spectroscopy (LRS) analysis confirmed that the origin of passivity of pilgered and autoclaved Zircaloy-4 tubes was due to the presence of a protective passive film composed of ZrO2. NaOH group exhibited significantly decreased CP level and in vitro DM digestibility, increased crude ash (CA) level and unchanged in vitro CP digestibility, comparing with control group. These results suggest that NaOH treatment affected the nutritional quality of SM adversely. HCl and formic acids groups showed significantly increased CP level and in vitro digestibilities of DM and CP, and decreased CA level, showing that acid treatment can improve nutritional quality of SM: formic acid treatment may be more effective because of the greater values in CP level and digestibilities and decreased crude fibre level which was not observed in HCl group (P<0.05). The results obtained here suggest acid, especially formic acid, treatment is promising to improve the nutritional quality of SM but autoclaving and NaOH treatments. [ABSTRACT FROM AUTHOR]

This paper investigated the hydration properties of steel slag under autoclaved condition (216 °C and 2 MPa). The results show that under autoclaved condition, besides CS and CS, f-CaO and the minerals containing MgO can react sufficiently. The morphologies of Ca(OH) produced by f-CaO are quite different from those produced by CS and CS. The Ca(OH) produced by f-CaO has a larger specific surface area or volume than that produced by CS and CS. The mean Ca-Si ratio of the C-S-H gel produced by steel slag is between 1.7 and 1.9, which is a little smaller than that produced by cement. The reaction of f-CaO tends to cause greater expansion than that of the minerals containing MgO. [ABSTRACT FROM AUTHOR]

Abstract: Previous studies have been carried out on calcined phosphogypsum (PG) for making the building materials. The present study was focused on autoclaved PG and its use in making load-bearing wall bricks. Autoclaved PG was prepared from original waste PG with steam pre-treatment. The crystalline phase, morphology, and thermal characteristics of original waste PG and autoclaved PG were investigated by XRD, SEM, and SDT. Then bricks of the size of Chinese standard brick were prepared from different types of PG in the PG-fly ash–lime–sand system. Results showed that the compressive strength of bricks from autoclaved PG by lower-pressure steam of 0.12MPa, 120°C for 16h was much higher. The flexural strength and compressive strength of the bricks could reach 4.0MPa and 15.0MPa, respectively. The durability of the bricks was investigated by 15 freezing–thawing cycles at temperatures from −20°C to 20°C, and the weight loss was only 0.029% after all of cycles. Hemihydrates (CaSO4 ·0.5H2O) were dehydrated products from dihydrates in original PG with lower-pressure steam treatment, and hemihydrates were susceptible to absorbing the humidity and were transformed into densified re-crystallization gypsum (CaSO4 ·2H2O) that contributed to the final strength of bricks. Microstructural characteristics of bricks were investigated by XRD and SEM. Tobermorite was the significant hydrated product, which contributed to the strength of bricks.
