Using anti-neurofilament H non-phosphorylated antibodies (SMI-32) as markers for the neuronal maturation level and Y channel responsible for motion processing, we investigated early postnatal development of the primary visual areas 17 and 18 in cats aged 0, 10, 14, and 34 days and in adults. Two analyzed parameters of SMI-32-immunolabeling were used: the total proportion of SMI-32-labeling and the density of labeled neurons. (i) The developmental time course of the total proportion of SMI-32-labeling shows the general increase in the accumulation of heavy-chain neurofilaments. This parameter showed a different time course for cortical layer development; the maximal increment in the total labeling in layer V occurred between the second and fifth postnatal weeks and in layers II-III and VI after the fifth postnatal week. In addition, the delay in accumulation of SMI-32-labeling was shown in layer V of the area 17 periphery representation during the first two postnatal weeks. (ii) The density of SMI-32-labeled neurons decreased in all layers of area 18, but was increased, decreased, or had a transient peak in layers II-III, V, and VI of area 17, respectively. The transient peak is in good correspondence with some transient neurochemical features previously revealed for different classes of cortical and thalamic neurons and reflects the time course of the early development of the thalamocortical circuitry. Some similarities between the time courses for the development of SMI-32-labeling in areas 17/18 and in A- and C-laminae of the LGNd allow us to propose heterochronous postnatal development of two Y sub-channels., (© 2024 Wiley Periodicals LLC.)

In the primary visual cortex and higher-order areas, it is well known that the stimulation of areas surrounding the classical receptive field of a neuron can inhibit its responses. In the primate area MT, this surround suppression was shown to be spatially organized into high and low suppression modules. However, such an organization hasn’t been demonstrated yet in the primary visual cortex. Here, we used optical imaging of intrinsic signals to spatially evaluate surround suppression in the cat visual cortex. The magnitude of the response was measured in areas 17 and 18 for stimuli with different diameters, presented at different eccentricities. Delimited regions of the cortex were revealed by circumscribed stimulations of the visual field (cortical response field). Increasing the stimulus diameter increased the spread of cortical activation. In the cortical response field, the optimal stimulation diameter and the level of suppression were evaluated. Most pixels (3/4) exhibited surround suppression profiles. The optimal diameter, corresponding to a population of receptive fields, was smaller in area 17 (22 deg.) than in area 18 (36 deg.) in accordance with electrophysiological data. No difference in the suppression strength was observed between both areas (A17: 25%, A18: 21%). Further analysis of our data revealed the presence of surround modulation maps, organized in low and high suppression domains. We also developed a statistical method to confirm the existence of this cortical map and its neuronal origin. The organization for center/surround suppression observed here at the level of the primary visual cortex is similar to those found in higher order areas in primates (e.g. area MT) and could represent a strategy to optimize figure ground discrimination.

Abstract: Imaging studies based on intrinsic optical signals have been primarily performed by analyses of the signal amplitude as opposed to using the temporal information of the intrinsic optical signals. The present study focused on the dynamics of these signals in cat area 18, and quantitatively compared the waveforms after presentation of different stimuli across the same cortical regions. Optical imaging based on intrinsic signals was conducted on 18 hemispheres of 9 cats. For the visual stimuli, gratings with orientations that changed from horizontal (0°) to 157.5° in 22.5° steps were used. The signal time course was examined at each pixel, with the peak delay defined as the amount of time required after the stimulus onset for the intrinsic optical signal to reach its negative maximum. In the area that showed significant orientation preference to 0° and 90° but not to their 22.5° separated nearby angles, the delays were 1.92±0.22s and 1.99±0.29s (mean±SE, n=18), respectively. Delays of 2.31±0.20s and 2.28±0.25s were observed in the cortical areas that selectively responded to the orientation gratings of 45°and 135° but not their nearby angles. Statistically, the delays in areas exhibiting oblique orientation preferences were significantly longer than those showing cardinal orientation preferences. Abstract: The extraction and detection of specific responses from a large amount of background noise has been the subject of a considerable body of research in brain functional imaging, and more specifically in optical intrinsic signal imaging. Recent work by showed that by combining different conditions and using periodic stimuli, recording times can be reduced. Spectral decomposition is then used to provide amplitude and phase information locked to the stimulus. A drawback of the above method is that by focusing only on a single harmonic, response information is limited. The shape of the hemodynamic response function (HRF) and the temporal variations in the neural responses cannot be assessed. In this work it is argued that additional information about neural responses can be gathered by using higher harmonics. Moving bars were used to generate visuotopic maps on large portions of the cat visual cortex. Up to four simultaneously bars moving repetitively across the visual field at different frequencies were used to sample the HRF in the Fourier domain. The HRF profile obtained with continuous stimulation was spatially homogeneous throughout the cortex and similar to the HRF profile obtained using episodic stimulation. Furthermore, by modeling the optical response as a convolution between HRF and neuronal responses, the ratio of the second harmonic to the first provided an estimation of the receptive field size. This was further validated by measuring spatial frequency selectivity. Therefore, the use of higher harmonics opens new avenues to estimate receptive field size from temporal signals. Abstract: Previous studies have shown a noticeable phenotypic diversity for pyramidal cells among cortical areas in the cerebral cortex. Both the extent and systematic nature of this variation suggests a correlation with particular aspects of cortical processing. Nevertheless, regional variations in the morphology of inhibitory cells have not been evaluated with the same detail. In the present study we performed a 3D morphometric analysis of 120 NADPH diaphorase (NADPH-d) type I neurons in the visual cortex of a South American Hystricomorph rodent, the diurnal agouti (Dasyprocta sp.). We found significant differences in morphology of NADPH-d type I neurons among visual cortical areas: cells became progressively larger and more branched from V1 to V2 and V3. Presumably, the specialized morphology of these cells is correlated with different sampling geometry and function. The data suggest that area-specific specializations of cortical inhibitory circuitry are also present in rodents. [Copyright &y& Elsevier]

Abstract: In the visual system, neurons with similar functional properties such as orientation and direction selectivity are clustered together to form modules. Optical imaging recordings in combination with episodic paradigms have been previously used to estimate direction selectivity, a fundamental property of visual neurons. The major drawback of the episodic approach is that the extraction of the signal from various forms of physiological noise is difficult, leading to a poor estimation of direction. Recent work, based on periodic stimulation and Fourier decomposition improved the extraction of periodic stimulus responses from noise and thus, reduced the recording time considerably. Given the success of this new paradigm in mapping orientation, the present study evaluated its reliability to measure direction selectivity in the visual cortex of anesthetized cats. Here, a model that exploits the harmonics of the Fourier decomposition is proposed where the first harmonic is related to direction responses, and the second to orientation. As expected, the first harmonic was absent when a static stimulus was presented. Contrarily, the first harmonic was present when moving stimuli were presented and the amplitude was greater with random dots kinematograms than with drifting gratings. The phase of the first harmonic showed a good agreement with direction preference measured by episodic paradigm. The ratio of the first/the second harmonic amplitude, related to a direction index, was weaker in fracture. It was also weaker in areas of the ventral pathway (areas 17 and 21a) where direction selectivity is known to be reduced. These results indicate that a periodic paradigm can be easily used to measure specific parameters in optical signals, particularly in situations when short acquisition periods are needed. The purpose of the present study was to identify projections from auditory to visual cortex and their organization. Retrograde tracers were used to identify the sources of auditory cortical projections to primary visual cortex (areas 17 and 18) in adult cats. Two groups of animals were studied. In the first group, large deposits were centered on the lower visual field representation of the vertical meridian located along the area 17 and 18 border. Following tissue processing, characteristic patterns of cell body labeling were identified in extrastriate visual cortex and the visual thalamus (LGN, MIN, & LPl). In auditory cortex, of the four tonotopically-organized regions, neuronal labeling was identified in the supragranular layers of the posterior auditory field (PAF). Little to no labeling was evident in the primary auditory cortex, the anterior auditory field, the ventral posterior auditory field or in the remaining six non-tonotopically organized regions of auditory cortex. In the second group, small deposits were made into the central or peripheral visual field representations of primary visual cortex. Labeled cells were identified in PAF following deposits into regions of primary visual cortex representing peripheral, but not central, visual field representations. Furthermore, a coarse topography was identified in PAF, with neurons projecting to the upper field representation being located in the gyral portion of PAF and neurons projecting to the lower field representation located in the sulcal portion of PAF. Therefore, direct projections can be identified from tonotopically organized auditory cortex to the earliest stages of visual cortical processing. [ABSTRACT FROM AUTHOR]

Abstract: Neural mechanisms mediating perceptual grouping serve to enhance associations among stimulus elements, thereby establishing unified forms. The goals of the present study were to identify cortical areas necessary to perceptually group spatially isolated elements, and to determine if these areas are distinct from regions necessary for the discrimination of simple, solid forms. Rats were trained to discriminate horizontal and vertical lines that were either solid or composed of disjunct elements in which discrimination required perceptual grouping by proximity. Psychophysical procedures established the limits at which proximity served as a cue for grouping. Following perceptual measurements, ablations were made to selective sites within visual cortex. Lesions within area 17 or area 18A, including their interface, produced nearly complete impairment of solid line discrimination as well as perceptual grouping at all levels of proximity, whereas lesions to areas 18 or the far lateral extent of area 18A no effect on these perceptual capacities. These results indicate that grouping by proximity requires early visual processing areas, and shares cortical areas necessary for simple pattern discrimination. These results suggest that mechanisms of grouping modify primary stimulus representations, constructing a pattern of activity functional similar to that elicited by solid forms. Higher visual cortical areas are involved in the perception of complex stimuli, such as the optic flow created by self-motion. On the other hand, area 18 is thought to extract primitive visual features, feeding higher cortical areas for further processing. In this study, we applied optical imaging of intrinsic signals in the central, lower visual field of cat area 18, and reconstructed direction preference and direction selectivity maps in each hemisphere. We observed a significant overrepresentation of downward and temporal directions, in accordance with previous electrophysiological results. Cardinal orientations were not overrepresented, however. Downward directions were overrepresented at the highest direction selectivity domains. Temporal direction representation, on the other hand, decreased with direction selectivity. Our findings therefore suggest the existence of a neural substrate for the processing of optic flow in cat area 18. [ABSTRACT FROM AUTHOR]

Abstract: The development of long-range horizontal connections depends on visual experience. Previous experiments have shown that in area 17 of strabismic but not in normal cats, horizontal fibers preferentially connect cell groups driven by the same eye indicating that fibers between co-active neurons are selectively stabilized. To test whether this is a general organizing principle of intracortical long-range circuitry we extended our analyses to both intrinsic horizontal connections within area 18 and to inter-areal connections between areas 17 and 18. To this end, we visualized the functional architecture of area 18 by intrinsic signal imaging. Horizontal circuitry was labeled by injecting fluorescent latex microspheres into functionally identified domains. Additionally, domains sharing the same ocular dominance as the neurons at the injection sites were visualized by 2-deoxyglucose autoradiography to allow comprehensive labeling of functional domains in regions far from the injection sites. Quantitative analyses revealed that in strabismic cats, 72% of the retrogradely labeled neurons in area 18 and 68% of the neurons in area 17 were located in the same ocular dominance domains as the injection sites. In contrast, these numbers were 52% and 54% in normal animals. These data show that experience modifies both intrinsic connections within area 18 and inter-areal projections from area 17 to area 18 as has been previously described for intrinsic and callosal connections in area 17. This provides further evidence for the hypothesis that the correlation of activity is a major selection criterion for the stabilization of neuronal circuits during postnatal development. Abstract: Strabismus (or squint) is both a well-established model for developmental plasticity and a frequent clinical symptom. To analyze experience-dependent plasticity of functional maps in the brain we used optical imaging of intrinsic signals to visualize both orientation and ocular dominance domains in cat area 18. In strabismic animals, iso-orientation domains exhibited a pinwheel-like organization, as previously described for area 18 of normally raised animals and for area 17 of both normally raised and strabismic cats. In area 18, mean pinwheel density was similar in the experimental (2.2 pinwheel centers per mm2 cortical surface) and control animals (2.3/mm2 in normally raised animals), but significantly lower than in area 17 of both normally raised and strabismic cats (2.7–3.4/mm2). A comparison of orientation and ocular dominance domains revealed that iso-orientation domains were continuous across the borders of ocular dominance domains and tended to cross these borders at steep angles. Thus, the orientation map does not seem to be modified by experience-dependent changes in afferent activity. Together with our recent observation that strabismus does not enhance the segregation of ocular dominance domains in cat area 18, the present data indicate that the layout of functional maps in area 18 is less susceptible to experience-dependent manipulations than in area 17. Abstract: In cat visual cortex, neurons acquire progressively mature functional properties during the first postnatal months. The aim of this study was to analyze the development of astrocytes during this period. The patterns of expression of the glial fibrillary acidic protein (GFAP) as well as of two gap junction proteins expressed in astrocytes, connexin43 (Cx43) and connexin30 (Cx30), were investigated by immunohistochemistry and optical density measurements, in visual cortical areas 17 and 18 at four different ages: 2 weeks (postnatal days 12 to 15, P12–15), 1 month (P27–31), 2 months (P60–62) and beyond 1 year. Since visual experience is a key factor for neural development, the patterns of expression of these three proteins were studied both in normally-reared and monocularly deprived animals. Interestingly, the distribution of GFAP, Cx43 and Cx30 was found to change dramatically but independently of visual experience, during postnatal development, even beyond P60. During the first postnatal month, GFAP and Cx43 were mainly localized in the white matter underlying the visual cortical areas 17 and 18. Then, their distributions evolved similarly with a progressive decrease of their density in the white matter associated with an increase in the cortex. Connexin30 expression appeared only from the second postnatal month, strictly in the cortex and with a laminar distribution which was similar to that of Cx43 at the same age. In adults, a specific laminar distribution was observed, that was identical for GFAP, Cx43 and Cx30: their density was higher in layers II/III and V than in the other cortical layers. In order to investigate the neural processes underlying figure-ground segregation on the basis of motion, we studied the responses of neurones in the second visual area (V2) of marmoset monkeys to stimuli that moved against dynamic textured backgrounds. The stimuli were either “solid” bars, which were uniformly darker or lighter than the background’s average, or kinetic (“camouflaged”) bars, formed by textural elements that matched the spatial and temporal modulation of the background. Camouflaged bars were rendered visible only by the coherent motion of their textural elements. Using solid bars, we subdivided the population of marmoset V2 neurones into motion-selective (uni- and bi-directional units, 73.3% of the sample) and weakly-biased (26.7%) subpopulations. The motion selective subpopulation was further subdivided into cue-invariant neurones (units which demonstrated a similar selectivity for the direction of motion of the solid and camouflaged bars) and non-cue-invariant neurones (units which showed selectivity to the direction of motion of solid bars, but had weak or pandirectional responses to camouflaged bars). Cells with cue-invariant responses to these stimuli were as common in V2 as in the primary visual area (V1; approximately 40% of the population). In V2, neurones with cue-invariant and non-cue-invariant motion selectivity formed distinct populations in terms of classical response properties: cue-invariant neurones were characterized by a sharp axis of motion selectivity and extensive length summation, while the majority of non-cue-invariant neurones had broader motion selectivity and were end-stopped. In the light of previous studies, these different constellations of classical response properties suggest a correlation with more traditionally recognized categories of V2 units and modular compartments. The feedback effect of activity of area 21a on orientation maps of areas 17 and 18 was investigated in cats using intrinsic signal optical imaging. A spatial frequency-dependent decrease in response amplitude of orientation maps to grating stimuli was observed in areas 17 and 18 when area 21a was inactivated by local injection of GABA, or by a lesion induced by liquid nitrogen freezing. The decrease in response amplitude of orientation maps of areas 17 and 18 after the area 21a inactivation paralleled the normal response without the inactivation. Application in area 21a of bicuculline, a GABAa receptor antagonist caused an increase in response amplitude of orientation maps of area 17. The results indicate a positive feedback from high-order visual cortical area 21a to lower-order areas underlying a spatial frequency-dependent mechanism. [Copyright &y& Elsevier]

Optical imaging based on intrinsic signals was used to analyze the structure of orientation column in area 18 of cat visual cortex. Recordings were obtained in 12 adult cats during presentation of contours in various orientations. In order to quantitatively evaluate the size of the response area, a method combining both information on the amplitude of the changes and the statistical significance of the changes was proposed. The results showed that the proportion of the cortical area activated reached a peak during the presentation of horizontal or vertical contours, whereas the areas activated by oblique contours were relatively smaller. Cardinal contours (0° and 90°) activated 50.63±1.88% (mean±SE) of the cortical area in the recording site, whereas oblique contours (45° and 135°) activated only 45.25±2.02% of the area. This difference in area was statistically significant (t=2.38, d.f.=11, and P<0.05). The results demonstrated a difference in the representations of the cardinal and oblique contours in area 18 of the cat visual cortex. [Copyright &y& Elsevier]

The aim of this investigation was to understand the functions of long horizontal connections projecting from area 17 to area 18 in cats. The animals were anesthetized and prepared for recording single-cell responses to sine-wave gratings in area 18. Neuronal activity was analyzed under three conditions: prior to, during, and after inactivation of a circumscribed region of area 17. The latter was depressed with micro-injections of GABA. Cells in both areas were in close retinotopic correspondence. Cells were classified as simple and complex types. Globally, simple cells were less affected than complex units, and those which were affected shifted their optimal spatial frequency to higher values. Complex neurons were more often influenced by the interruption of area 17 input. Namely, the peaks of the tuning curves were displaced on the x-axis to a new optimal spatial frequency. This effect was obtained by a dual change: a decline in the discharge strength to the optimal spatial frequency and an enhancement to nonoptimal spatial frequency. Contrast sensitivity function disclosed similar shifts of optimal spatial frequencies. Likewise bandwith, spatial resolution, high cutoff, and low cutoff were modified to a greater extent in complex cells. It appears that there is no relationship between areas 17/18 orientation difference and the modifications observed in tuning curves to spatial frequencies. The results suggest that neurons of area 18 may carry multiple-frequency channels and that area 17 facilitates the emergence of one particular spatial frequency. [ABSTRACT FROM AUTHOR]

We examined the orientation tuning curves of 86 cells located in layer V of area 17, before, during, and after focal blockade of a small (300-μm diameter) region of near-retinotopic register in layer V of area 18 of quantitatively established orientation preference. Such focal blockade revealed three distinct populations of area 17 layer V cells-cells with decreased responses to stimuli of some orientations (21%), cells with increased responses to stimuli of some orientations (43%), and cells unaffected by the focal blockade (36%). These effects were clearcut, reproducible, and generally directly related to the known receptive field properties of the cell recorded in area 18 at the center of the zone of blockade. These effects were also analyzed in terms of alterations in orientation bandwidth in the cells in area 17 as a result of the blockade-bandwidth increases (22%) and decreases (24%) were found; however, these changes were essentially unrelated to the measured receptive field properties. Inhibitory and excitatory effects were most pronounced when the regions in areas 17 and 18 were of like ocular dominance and were of similar orientation preference. Inhibitory effects (suggesting a normally excitatory input) were most dependent upon the similarity of receptive fields; excitatory effects (suggesting a normally inhibitory input) were less heavily dependent. Focal blockade of restricted regions in layer V of area 18 was used to assess the contribution of this region to the responses to high-velocity stimuli of cells in retinotopically matched, layer V in area 17. In 40% of cases, blockade within area 18 revealed responses of area 17 cells to high-velocity stimuli to which they previously showed only poor responses. Stimulus specificity of the cells in area 17 was otherwise unaltered. All effects were reversible and repeatable. We suggest that a component of the output of layer V from area 18 normally suppresses the responses of retinotopically matched cells within area 17 to stimuli of high velocity, thereby enhancing the specificity of those cells to stimuli of low velocity [ABSTRACT FROM AUTHOR]

The numerical density of neurons and glial cells was estimated in visual area 18 of the adult human cerebral cortex and compared with that of area 17. Blocks of areas 17 and 18 came from the same brains and this allowed the comparison of 1) neuronal and glial numerical densities through the whole cortical depth with calculation of the neuron/glia ratio, 2) neuronal and glial numbers under one square millimeter of cortical surface, and 3) neuronal numerical densities in three groups of identified layers. The mean neuronal density is approximately 40000 neurons/mm in area 17 and 31500/m in area 18. The mean glial density is around 27000/mm in area 17 and 32000/mm in area 18. This gives a neuron/glia ratio of approximately 1.5 in area 17 and of 1.0 in area 18, but the total cellular density is similar in both areas. There are about 90000 neurons and 64000 glial cells under one square millimeter of cortical surface in area 17, and some 73000 neurons and 74000 glial cells in area 18. The higher neuronal density in area 17 is found through the whole depth of cortex and does not seem to be more pronounced in layer IVc of area 17 compared to layer IV in area 18 than in the groups of layers II-III and V-VI. Single-cell recording experiments were carried out to determine whether rearing kittens with surgically induced convergent strabismus (esotropia) alters the development of receptive field (RF) properties of neurons in area 18. In agreement with previous work on kittens with divergent strabismus (exotropia), there was a marked loss of binocularly driven cells in area 18 of esotropic cats. In contrast to the striate cortex of strabismic cats, the spatial properties of area 18 neurons, including receptive-field size and spatial frequency tuning, did not differ from those in normal controls. On the other hand, we found that contrast thresholds, measured at an optimal spatial frequency, were significantly elevated, and that the contrast gain in many cells was reduced in strabismic cats. These deficits were observed in both eyes, though the cells dominated by the deviating eye had a lower response amplitude at all contrasts. Furthermore, temporal frequency tuning curves were abnormal in strabismic cats in that the optimal frequencies and temporal resolutions were shifted to lower values. These effects were also bilateral. Velocity tuning, measured with a high-contrast bar stimulus, revealed that area 18 neurons in strabismic cats were unable to respond to very high velocities compared to normals. This reduced response was more severe when measured with the deviating eye in spite of the bilateral nature of the deficit. Finally, latencies to electrical stimulation of the optic chiasm or the optic radiation were significantly longer in strabismic cats. The magnitude of these effects was virtually the same for both eyes. From these observations, we conclude that the temporal properties of area 18 neurons, particularly the cells abilities to follow fast temporal modulations, are affected by raising kittens with surgically induced convergent strabismus. The spatial frequency tuning curves of neurones of area 18 depend upon the velocity of the visual stimulus. The higher the velocity the lower the spatial frequencies to which the cell is tuned. Since in area 17 the size of the cell receptive field is inversely related with the optimal spatial frequency to which the cell responds, we have investigated whether the shift of the optimal spatial frequency with the velocity corresponds to a 'change' in the receptive field size. We recorded extracellularly from neurones in area 18; for each cell we selected two gratings, one of high spatial frequency drifting at low velocity and another of low spatial frequency drifting at high velocity to which the cell gave comparable responses. The results show that the masking of the cells receptive field which abolishes the response to the high frequency low velocity grating does not prevent the cell from responding to the low frequency high velocity grating. We conclude that the size of the receptive field of neurones in area 18 depends upon the characteristics (spatial frequency and velocity) of the visual stimulus. [ABSTRACT FROM AUTHOR]

Receptive field sizes to stimulation of the ipsilateral temporal retina were studied in area 18 of adult cats at different times after complete midsagittal section of the optic chiasm. A specific postoperative evolution could thus be noticed: immediately after section, the average area of the receptive fields was reduced, as compared to control preparations, owing to the disappearance of large fields located at more than 20∘ of eccentricity. A progressive reappearance of these large fields occurred between 8 and 45 days after chiasmotomy, provided that the animal was placed in normal visual conditions during its postoperative period. No such recovery could be assessed after as long as 55 days, in animals kept in complete darkness after operation. Chiasmotomized cats also displayed a reduction of their percentage of light reactive cells with respect to controls, as expected from the suppression of the contralateral input. This percentage was at first very low and progressively increased, during postoperative recovery but again not when the animal had been kept in the dark. Finally, an increase of cells with 'diffuse responses' was observed in the late postoperative recovery stage. This latter evolution also appeared to depend upon postoperative visual experience. On the other hand, no clear indication of an interhemispheric transfer could be obtained in these experiments, even at the 17-18 boundary. [ABSTRACT FROM AUTHOR]

Responses to binocular visual stimulation were compared in cortical area 18 of normal cats and in cats in which one eye was exodeviated by surgery early in postnatal life. In contrast to normal cats where most units (58%) were binocularly activated, relatively few units (10%) in strabismic cats were activated well by stimulation of either eye. Rather individual units were driven mainly via one eye or the other but not both. In addition, there was a tendency for more units to be driven well via the unoperated eye than via the exodeviated eye. Fewer cells preferring vertically oriented than horizontally oriented stimuli were found in area 18 of strabismic cats. This trend was observed for cells driven by either the normal or deviated eye and was especially marked among the small number of binocularly activated cells. Generally binocular responses and binocular interactions were found with stimulation at corresponding retinal points. In a few striking instances, however, the receptive fields of binocular neurons were located on noncorresponding retinal points at loci which would enable the cat to correlate the two images of an external object despite the large divergent strabismus. Quantitative responses to binocular stimuli presented at varied disparities and to stimuli with varied directions of motion in depth were compared in normal and strabismic cats. Despite the large strabismus, a reduced fraction of cortical neurons displayed substantial binocular interactions. In fact, binocular facilitation was as marked in the population of cells studied in strabismic cats as it was in normal animals. The major effect of strabismus was a reduction in the strength of binocular inhibition when units were tested with sideways motion. Disparity-specific responses to motion toward or away from the organism were little affected by strabismus. The degree of binocular facilitation and binocular inhibition among the cell population was similar in normal and strabismic cats. A subpopulation of units encountered in strabismic cats showed strong disparity-specific interactions for motion toward or away from the animal without equivalent modulation for sideways moving stimuli. We have studied the effects of monocular deprivation (MD) on area 18 in the cat. Our intention was to determine whether the degree of vulnerability to MD varies regionally within area 18. Single units were recorded in animals used as normal controls (77 cells) and in MD cats (467 cells) from anterior and posterior regions of area 18 representing peripheral and central-paracentral fields, respectively. Recordings from the deprived cats were made from both hemispheres. Overall, around 27% of the cells from the MD group retained input from the deprived eye. Most of these cells (77%) were located in the hemisphere contralateral to the deprived eye. Furthermore, the majority of these cells (96%) were located in the anterior region of area 18 and had receptive fields at eccentric locations (greater than 10 deg). About one third of the cells driven by the deprived eye had diffuse receptive field borders, and most of these units were binocular. In addition, these cells were generally non-oriented. Our main result, that susceptibility to MD decreases with increasing receptive field eccentricity, may be accounted for on the basis of corresponding changes of spatial frequency selectivity of cortical cells. In cats, 131 area 18 units with receptive fields 10-50 ° from the visual axis, were recorded and tested with moving stimuli. These stimuli included light slits, dark bars, single edges, gratings and a random square pattern. 94% of the units were responsive to visual motion. 106 neurones could be classified by a screening procedure based on responses to a moving standard grating and a moving random square pattern. When testing with moving stimuli, the criteria for distinguishing between simple and complex cells in area 17 (Pettigrew et al., 1968; Bishop et al., 1971a, b, 1973; Goodwin and Henry, 1975, Sherman et al., 1976) also apply to area 18 neurones. No tests with stationary stimuli were made in distinguishing simple from complex cells. In addition to simple (50%) and complex (18%) cells, area 18 contains other cell types as hypercomplex cells (11%), cells with a radially symmetric RF (6%) and amplitude specific cells (10%). Six cells (5 %) were unclassified. A special battery of tests was designed to study the RF of the very sophisticated amplitude specific cells. Their RF appears to consist of a discharge region preceded by a trigger area and of two flanking inhibitory regions. [ABSTRACT FROM AUTHOR]

In cats, 107 area 18 neurones with identified FR type, 10-50 ° from the visual axis, were tested for the influence of direction, velocity and amplitude of movement. These three parameters are believed to be the primary parameters of a movement analysing system. 94% of the neurones were influenced by the direction of movement, all of them by the angular velocity and 16% by the amplitude of movement. For each of the primary parameters, tuning curves were established. Angular velocity influenced not only the response magnitude but also the response latency and the direction bias. By preparing response amplitude functions at different velocities the influence of movement duration was ruled out. The association of functional properties and RF organization suggests a model of information processing in area 18 of the cat. [ABSTRACT FROM AUTHOR]

Pattern vision deprivation (BD) can induce permanent deficits in global motion perception. The impact of timing and duration of BD on the maturation of the central and peripheral visual field representations in cat primary visual areas 17 and 18 remains unknown. We compared early BD, from eye opening for 2, 4, or 6 months, with late onset BD, after 2 months of normal vision, using the expression pattern of the visually driven activity reporter gene zif268 as readout. Decreasing zif268 mRNA levels between months 2 and 4 characterized the normal maturation of the (supra)granular layers of the central and peripheral visual field representations in areas 17 and 18. In general, all BD conditions had higher than normal zif268 levels. In area 17, early BD induced a delayed decrease, beginning later in peripheral than in central area 17. In contrast, the decrease occurred between months 2 and 4 throughout area 18. Lack of pattern vision stimulation during the first 4 months of life therefore has a different impact on the development of areas 17 and 18. The layers and sublayers of primary visual cortex, or V1, in primates are easily distinguishable compared to those in other cortical areas, and are especially distinct in anthropoid primates - monkeys, apes, and humans - where they also vary in histological appearance. This variation in primate-specific specialization has led to a longstanding confusion over the identity of layer 4 and its proposed sublayers in V1. As the application of different histological markers relate to the issue of defining and identifying layers and sublayers, we applied four traditional and four more recent histological markers to brain sections of V1 and adjoining secondary visual cortex (V2) in macaque monkeys, chimpanzees, and humans in order to compare identifiable layers and sublayers in both cortical areas across these species. The use of Nissl, neuronal nuclear antigen (NeuN), Gallyas myelin, cytochrome oxidase (CO), acetylcholinesterase (AChE), nonphosphorylated neurofilament H (SMI-32), parvalbumin (PV), and vesicular glutamate transporter 2 (VGLUT2) preparations support the conclusion that the most popular scheme of V1 lamination, that of Brodmann, misidentifies sublayers of layer 3 (3Bβ and 3C) as sublayers of layer 4 (4A and 4B), and that the specialized sublayer of layer 3 in monkeys, 3Bβ, is not present in humans. These differences in interpretation are important as they relate to the proposed functions of layer 4 in primate species, where layer 4 of V1 is a layer that receives and processes information from the visual thalamus, and layer 3 is a layer that transforms and distributes information to other cortical areas.
