We engage in social interaction not only in everyday life, but also in social spaces in which language learning takes place, such as in second or foreign language classrooms. Applied linguistics is increasingly applying concepts and insights from research on social interaction to topics in language learning and language teaching under the umbrella term Interactional Competence (IC). This research focuses on context-sensitive interactional competencies and abilities that language learners encounter, develop, and diversify in the learning process both in and outside of language classrooms. Another prominent line of inquiry focuses on the analysis of classroom interaction in pursuit of understanding its infrastructure and improving teaching effectiveness in line with specific pedagogical goals, using methods of analysis in Interactional Linguistics and Conversation Analysis. Language teacher training is also increasingly seeking to make select research topics in social interaction accessible for current and future language teachers. This special issue on fostering interactional competence is embedded in this broad context: It seeks to outline the many ways in which social interaction is relevant for German as a foreign/second language (DaFZ), and to spotlight some of these topics through the individual empirical contributions in this special issue. We propose a label‐free method for measuring intracellular temperature using a Raman image of a cell in the O−H stretching band. Raman spectra of cultured cells and the medium were first measured at various temperatures using a Raman microscope and the intensity ratio of the two regions of the O−H stretching band was calculated. The intensity ratio varies linearly with temperature in both the medium and cells, and the resulting calibration lines allow simultaneous visualization of both intracellular and extracellular temperatures in a label‐free manner. We applied this method to the measurement of temperature changes after the introduction of FCCP (carbonyl cyanide‐p‐trifluoromethoxyphenylhydrazone) in living cells. We observed a temperature rise in the cytoplasm and succeeded in obtaining an image of the change in intracellular temperature after the FCCP treatment. [ABSTRACT FROM AUTHOR]

We report, for the first time, utilizing a rotating ring‐disc electrode (RRDE) assembly for detecting changes in the local pH during aqueous CO2 reduction reaction (CO2RR). Using Au as a model catalyst where CO is the only product, we show that the CO oxidation peak shifts by −86±2 mV/pH during CO2RR, which can be used to directly quantify the change in the local pH near the catalyst surface during electrolysis. We then applied this methodology to investigate the role of cations in affecting the local pH during CO2RR and find that during CO2RR to CO on Au in an MHCO3 buffer (where M is an alkali metal), the experimentally measured local basicity decreased in the order Li+ > Na+ > K+ > Cs+, which agreed with an earlier theoretical prediction by Singh et al. Our results also reveal that the formation of CO is independent of the cation. In summary, RRDE is a versatile tool for detecting local pH change over a diverse range of CO2RR catalysts. Additionally, using the product itself (i.e. CO) as the local pH probe allows us to investigate CO2RR without the interference of additional probe molecules introduced to the system. Most importantly, considering that most CO2RR products have pH‐dependent oxidation, RRDE can be a powerful tool for determining the local pH and correlating the local pH to reaction selectivity. A biosensor was created for the simultaneous monitoring of endogenous H2Sn and H2S in mouse brains and exploring their roles in activation of the TRPA1 channel under two types of brain disease models: ischemia and Alzheimer's disease (AD). Based on DFT calculations and electrochemical measurements, two probes, 3,4‐bis((2‐fluoro‐5‐nitrobenzoyl)oxy)‐benzoic acid (MPS‐1) and N‐(4‐(2,5‐dinitrophenoxy) phenyl)‐5‐(1, 2‐dithiolan‐3‐yl)pentanamide (MHS‐1), were synthesized for specific recognition of H2Sn and H2S. Through co‐assembly of the two probes at the mesoporous gold film with good anti‐biofouling ability and electrocatalytic activity, this microsensor showed high selectivity for H2Sn and H2S against potential biological interferences. The biosensor can simultaneously determine the concentration of H2Sn from 0.2 to 50 μm, as well as that of H2S from 0.2 to 40 μm. Electrospray ionization mass spectrometry (ESI‐MS) is an analytical technique that measures the mass of a sample through "soft" ionization. Recent years have witnessed a rapid growth of its application in noble‐metal nanocluster (NC) analysis. ESI‐MS is able to provide the mass of a noble‐metal NC analyte for the analysis of their composition (n, m, q values in a general formula [MnLm]q), which is crucial in understanding their properties. This review attempts to present various developed techniques for the determination of the composition of noble metal NCs by ESI‐MS. Additionally, advanced applications that use ESI‐MS to further understand the reaction mechanism, complexation behavior, and structure of noble metal NCs are introduced. Zellen sind die grundlegende Funktions‐ und Struktureinheit lebender Organismen. Einzellige Gemeinschaften wie auch mehrzellige Spezies produzieren eine erstaunliche chemische Diversität, die eine große Bandbreite an unterschiedlichen Funktionen ermöglicht, dennoch hat jede Zelle zahlreiche Merkmale, die allen lebenden Organismen gemeinsam sind. Während es viele Ansätze zur Erforschung dieser chemischen Diversität gibt, sind nur wenige nicht zielgerichtet und in der Lage, die Analyse von Hunderten von verschiedenen Substanzen mit zellulärer Auflösung zu bewerkstelligen. In diesem Aufsatz diskutieren wir solche nicht zielgerichteten Ansätze, die verwendet werden, um umfassende chemische Analysen durchzuführen, Informationen für die chemische Bildgebung bereitzustellen oder Hochdurchsatz‐Profilingdaten über einzelne Zellen zu erhalten. Die Fähigkeiten der Einzelzellmessung entwickeln sich rasant bezüglich Durchsatz, Nachweisgrenzen und der Vollständigkeit der chemischen Analysen; diese Verbesserungen ermöglichen es, immer komplexere physiologische Phänomene, wie z. B. Droplet microfluidics is an enabling platform for high‐throughput screens, single‐cell studies, low‐volume chemical diagnostics, and microscale material syntheses. Analytical methods for real‐time and in situ detection of chemicals in the droplets will benefit these applications, but they remain limited. Reported herein is a novel heterogeneous chemical sensing strategy based on functionalization of the oil phase with rationally combined sensing reagents. Sub‐nanoliter oil segments containing pH‐sensitive fluorophores, ionophores, and ion‐exchangers enable highly selective and rapid fluorescence detection of physiologically important electrolytes (K+, Na+, and Cl−) and polyions (protamine) in sub‐nanoliter aqueous droplets. Electrolyte analysis in whole blood is demonstrated without suffering from optical interference from the sample matrix. Moreover, an oil phase doped with an aza‐BODIPY dye allows indication of H2O2 in the aqueous droplets, exemplifying sensing of targets beyond ionic species. [ABSTRACT FROM AUTHOR]

Current practical methods for finding the equilibrium dissociation constant, Kd, of protein–small molecule complexes have inherent sources of inaccuracy. Introduced here is "accurate constant via transient incomplete separation" (ACTIS), which appears to be free of inherent sources of inaccuracy. Conceptually, a short plug of the pre‐equilibrated protein–small molecule mixture is pressure‐propagated in a capillary, causing fast transient incomplete separation of the complex from the unbound small molecule. A superposition of signals from these two components is measured near the capillary exit and used to calculate a fraction of unbound small molecule, which, in turn, is used to calculate Kd. Herein the validity of ACTIS is proven theoretically, its accuracy is verified by computer simulation, and its practical use is demonstrated. Magic‐angle spinning (MAS) is an essential ingredient in a wide variety of solid‐state NMR experiments. The standard procedures to adjust the rotor angle are not highly accurate, resulting in a slight misadjustment of the rotor from the magic angle (θRL=tan-12) on the order of a few millidegrees. This small missetting has no significant impact on the overall spectral resolution, but is sufficient to reintroduce anisotropic interactions. Shown here is that site‐specific 1H‐15N dipolar couplings can be accurately measured in a heavily deuterated protein. This method can be applied at arbitrarily high MAS frequencies, since neither rotor synchronization nor particularly high radiofrequency field strengths are required. Group coordination is a relevant prerequisite for understanding the effectiveness of groups. But, contrary to the large number of empirical studies, only a few studies concentrate on the development of analysis methods of coordination in groups. Hence, the purpose of this paper is to give a summary of the opportunities and limitations of common methods for capturing group coordination in applied field settings and to outline how visual analytics approaches might add to the common methods. Based on two illustrated visual analytics implementation examples—1) coordination and movements of soccer players, and 2) spatio-temporal event data—the potential of visual analytics approaches is identified for studying a greater variety of types of group coordination, and to consider the multifaceted nature of group processes in order to go beyond traditional coding processes. Monoamine oxidase (MAO) has two isoforms, MAO-A and MAO-B, which show different functions, and thus selective fluorescence imaging is important for biological studies. Currently, however, specific detection of MAO-A remains a great challenge. Herein, we report a new strategy for specific imaging of MAO-A through the design of fluorogenic probes combining the characteristic structure of an inhibitor of the target enzyme along with propylamine as a recognition moiety. The high specificity of our representative probe is demonstrated by imaging MAO-A in different live cells such as SH-SY5Y (high levels of MAO-A) and HepG2 (high levels of MAO-B), and further validated by western blot analyses. The superior specificity of the probe may enable the accurate detection of MAO-A in complex biosystems. Importantly, the use of the characteristic structure of an inhibitor, as demonstrated in this work, may serve as a general strategy to design specific recognition moieties for fluorogenic probes for enzymes. Knowing the thermodynamic state of complex mixtures-liquid, gas, supercritical or two-phase-is essential to industrial chemical processes. Traditionally, phase diagrams are compiled piecemeal from individual measurements in a pressure-volume-temperature cell performed in series, where each point is subject to a long fluid equilibrium time. Herein, 1000 microfluidic chambers, each isolated by a liquid piston and set to a different pressure and temperature combination, provide the complete pressure-temperature phase diagram of a hydrocarbon mixture at once, including the thermodynamic phase envelope. Measurements closely match modeled values, with a standard deviation of 0.13 MPa between measurement and model for the dew and bubble point lines, and a difference of 0.04 MPa and 0.25 °C between measurement and model for the critical point. Herein, we develop a novel method for designing electrochemical biosensors with both current and potential signal outputs for the simultaneous determination of two species in a living system. Oxygen (O2) and pH, simple and very important species, are employed as model molecules. By designing and synthesizing a new molecule, Hemin-aminoferrocene (Hemin-Fc), we create a single electrochemical biosensor for simultaneous detection and ratiometric quantification of O2 and pH in the brain. The reduction peak current of the hemin group increases with the concentration of O2 from 1.3 to 200.6 μ m. Meanwhile, the peak potential positively shifts with decreasing pH from 8.0 to 5.5, resulting in the simultaneous determination of O2 and pH. The Fc group can serve as an internal reference for ratiometric biosensing because its current and potential signals remain almost constant with variations of O2 and pH. The developed biosensor has high temporal and spatial resolutions, as well as remarkable selectivity and accuracy, and is successfully applied in the real-time quantification of O2 and pH in the brain upon ischemia, as well as in tumor during cancer therapy. Diffusion-ordered spectroscopy (DOSY) is an effective method for the analysis of intact mixtures, but the quality of results is critically limited by resolution in the NMR dimension. A new experiment integrating diffusion weighting into the PSYCHE method for pure shift NMR spectroscopy allows DOSY spectra to be measured with ultrahigh NMR resolution at improved sensitivity. Fluorescence imaging of tyrosinase (a cancer biomarker) in living organisms is of great importance for biological studies. However, selective detection of tyrosinase remains a great challenge because current fluorescent probes that contain the 4-hydroxyphenyl moiety show similar fluorescence responses to both tyrosinase and some reactive oxygen species (ROS), thereby suffering from ROS interference. Herein, a new tyrosinase-recognition 3-hydroxybenzyloxy moiety, which exhibits distinct fluorescence responses for tyrosinase and ROS, is proposed. Using the recognition moiety, we develop a near-infrared fluorescence probe for tyrosinase activity, which effectively eliminates the interference from ROS. The high specificity of the probe was demonstrated by imaging and detecting endogenous tyrosinase activity in live cells and zebrafish and further validated by an enzyme-linked immunosorbent assay. Die Befassung mit neuen Werken gehört mittlerweile zum Standard des musiktheoretischen Unterrichts an deutschen Hochschulen, auch im sogenannten Pflichtfach Tonsatz und für sämtliche Studiengänge. Sie kann eher analytisch, eher synthetisch – unterschiedliche Erfahrungsbereiche verknüpfend – oder praktisch ausgerichtet sein. An drei Werken aus der jüngsten Geschichte des Komponierens werden die drei Zugangsweisen erprobt.

Viruses are by far the most abundant biological entities on our planet, yet existing characterization methods are limited by either their speed or lack of resolution. By applying a laboratory-built high-sensitivity flow cytometer (HSFCM) to precisely quantify the extremely weak elastically scattered light from single viral particles, we herein report the label-free analysis of viruses with a resolution comparable to that of electron microscopy and the throughput of flow cytometry. The detection of single viruses with diameters down to 27 nm is described. T7 and lambda bacteriophages, which differ in size by as little as 4 nm, could be baseline-resolved. Moreover, subtle structural differences of the same viral particles can be discriminated. Using monodisperse silica nanoparticles as the size reference standards, the virus sizes measured by the HSFCM are in agreement with the equivalent particle diameters derived from their structural dimensions. Rapid antimicrobial susceptibility testing (AST) would decrease misuse and overuse of antibiotics. The 'holy grail' of AST is a phenotype-based test that can be performed within a doctor visit. Such a test requires the ability to determine a pathogen's susceptibility after only a short antibiotic exposure. Herein, digital PCR (dPCR) was employed to test whether measuring DNA replication of the target pathogen through digital single-molecule counting would shorten the required time of antibiotic exposure. Partitioning bacterial chromosomal DNA into many small volumes during dPCR enabled AST results after short exposure times by 1) precise quantification and 2) a measurement of how antibiotics affect the states of macromolecular assembly of bacterial chromosomes. This digital AST (dAST) determined susceptibility of clinical isolates from urinary tract infections (UTIs) after 15 min of exposure for all four antibiotic classes relevant to UTIs. A critical requirement for the successful recognition of multiple analytes is the acquisition of abundant sensing information. However, for this to be achieved requires massive chemical sensors or multiplex materials, which complicates the multianalysis. Thus, there is a need to develop a strategy for the facile and efficient recognition of multiple analytes. Herein, we explore the angle-dependent structural colors of photonic crystals to provide abundant optical information, thereby generating a rainbow-color chip to realize the convenient recognition of multiple analytes. By simply using a multiangle analysis method, the monophotonic crystal matrix chip can differentially enhance fluorescence signals over broad spectral ranges, thereby resulting in abundant sensing information for highly efficient multiple analysis. Twelve saccharides with similar structures, as well as saccharides in different concentrations and mixtures, were successfully discriminated. [ABSTRACT FROM AUTHOR]

So wie man in einem U ‐ Boot Objekte oberhalb der Wasseroberfläche mit einem Periskop erfassen kann, so kann man die Elektrospray ‐ Massenspektrometrie (ESI ‐ MS) als eine Art umgekehrtes Periskop auffassen, um an der Gas/Flüssig ‐ Schnittstelle gelöste Verbindungen auf ihre chemische Zusammensetzung hin zu analysieren. Ziel ist es, Zwischenstufen in chemischen Umsetzungen, die sich anderweitig der Charakterisierung entziehen und für den Ablauf der Reaktion entscheidend sind, zu identifizieren und strukturell zu charakterisieren. Anhand von ausgewählten aktuellen Publikationen werden in diesem Kurzaufsatz die ESI ‐ MS ‐ Techniken erläutert, die derzeit zur Erforschung von Zwischenstufen in homogenen chemischen Reaktionen in Lösung eingesetzt werden. Vor dem Auge des Periskops: Die Elektrospray ‐ Massenspektrometrie (ESI ‐ MS) ist ein leistungsfähiges Instrument zur Erforschung von Reaktionsmechanismen. Dies wird anhand der Idee eines umgekehrten Periskops veranschaulicht: An der Schnittstelle zwischen Gas und Flüssigkeit erfasst die ESI ‐ MS Reaktionszwischenstufen in Lösung und gewährt somit Einblick in den Mechanismus von Mehrkomponentenreaktionen, Organokatalysen und homogenen Metallkatalysen. Motivisch-thematische Ereignisse spielen in linearen Reduktionsverfahren der Schenker-Nachfolge eine tendenziell untergeordnete Rolle. Es ist daher ein verbreitetes (Vor-)Urteil, die Ergebnisse derartiger Analysen seien irrelevant für eine individualisierende Werkbetrachtung und lägen jenseits der ästhetischen Erfahrbarkeit. Anhand von Beethovens f-Moll-Streichquartett op. 95 wird gezeigt, wie motivisch-thematische Formbeschreibungen und lineare Reduktionsverfahren einander zu ergänzen vermögen. Wesentliche Momente der thematischen Entwicklung finden im Kopfsatz von op. 95 ›hinter‹ der Satzoberfläche statt. So beherrscht der im Motto latente Konflikt des-d den Satz vom figurativen Detail bis zur Großform. Dieses thematische ›Substrat‹ wirkt formbildend in Gestalt der chromatischen Auskomponierung syntaktischer Penultimae; die vermeintlich neutralen Linienzüge erscheinen thematisch aufgeladen, so daß auch weitgespannte melodische Progressionen rezeptionsästhetisch greifbar werden. Die fraktale Struktur der Thematik und die fraktale Struktur der linearen Schichten korrelieren dabei so weitgehend, daß ein undogmatischer Rekurs auf die Schenkersche Schichtenlehre den thematisch-formbildenden Prozess transparent zu machen vermag.

Insbesondere in der deutschsprachigen Popularmusikforschung sind seit jeher soziologisch und psychologisch geprägte Ansätze vorherrschend gewesen, deren Interesse den äußerst komplexen gesellschaftlichen Funktions- und Wirkungszusammenhängen von Rock- und Popmusik gilt. Strittig ist, ob Rockmusik strukturanalytisch in den Blick zu nehmen überhaupt als dem Gegenstand angemessen gelten kann. Nicht zuletzt hieraus resultiert eine immer noch vergleichsweise starke Zurückhaltung des dezidiert musiktheoretischen Diskurses im Bereich populärer Musik. Mit der hier vorgelegten Untersuchung des Songs New York State Of Mind von Billy Joel wird der These, daß populäre Musik strukturell nicht interessiere, nicht nur prinzipiell widersprochen, mehr noch, es finden vorwiegend solche analytischen Ansätze Verwendung, die von der Beschäftigung mit traditioneller Kunstmusik her erprobt sind. Dieses Vorgehen versucht zu verdeutlichen, daß die tonalen Schemata der traditionellen Kunstmusik eine Anpassung an Idiom und Verfahrensweise populärer Musik erfahren, die als ›strukturell‹ interessant gelten darf, weil populäre Musik als ›klangliches Ereignis‹ sich nicht im Moment des ›Sounds‹ erschöpft.

Wenn von harmonischer Mehrdeutigkeit und den daraus resultierenden Schwierigkeiten für die Analyse die Rede ist, dürfte der Gedanke an Musik der Spätromantik – mit Richard Wagners Tristan als Paradigma –kaum zu unterdrücken sein. Indessen stellte schon die Harmonielehre des frühen 18. Jahrhunderts ihren Begründer Jean-Philippe Rameau vor Probleme, die nicht als Folge eines noch ungenügenden Entwicklungsstands der Theorie mißverstanden werden dürfen, sondern als Ausdruck prinzipieller, die Wahrnehmung, Auffassung und Interpretation musikalischer Zusammenhänge betreffender Gegebenheiten anzusehen sind. Anhand einiger Beispiele harmonisch ambivalenter Konstellationen unterschiedlichen Typs wird gezeigt, daß Mehrdeutigkeit in ihrer manifesten Form auf dem Konflikt von Bedeutungsebenen (historischer, perzeptueller oder sprachlicher Art) beruht, die sich nicht notwendig erst in problematischen Situationen konstituieren, sondern wenigstens zum Teil permanent vorhanden sind. Dementsprechend kann der Eindruck von Eindeutigkeit auf die Kohärenz der für die Rezeption maßgeblichen Kontexte zurückgeführt werden: Musiktheoretische Begriffe, die aus solchen ›eindeutigen‹ Sachverhalten abgeleitet sind, verlieren an Relevanz oder werden unbrauchbar, sobald das konstitutionelle Gefüge in Bewegung gerät.

Der Paradigmenwechsel von der ›sukzessiven‹ zur ›simultanen‹ Kompositionsweise im 15. Jahrhundert änderte auch das Verständnis musikalischer ›Form‹ grundlegend. Die immanent musikalischen Mittel, durch welche ›Form‹ hervorgebracht werden konnte, waren jedoch sehr begrenzt. Anhand der Missa La sol fa re mi von Josquin Desprez und ihrer engen Beziehung zur Missa Mi mi von Johannes Ockeghem wird exemplarisch dargestellt, wie Josquin durch den Rückgriff auf ›außermusikalische‹ Strategien wie zahlensymbolische Proportionierung und ›musikalische Intertextualität‹ dieser Problemstellung begegnet und eine formale Konzeption für den Messezyklus entwickelt.

Strukturanalytische Verfahrensweisen setzen die synchrone Existenz oder Vorstellbarkeit musikalischer Werke voraus. Sie zielen auf das Fixierte, im Zeitablauf Konstante, Dauernde: Material, Struktur, architektonischer oder numerischer Aufbau. Prozessual orientierte Werkanalyse sucht hingegen den Sinn der Musik in einem anderen, flexibleren Prinzip, das das fluktuale Moment einschließt: die sich in der Zeit entwickelnde Form, das Sich-Entfalten, Ausladen, Steigern und Verlaufen. Auch die amerikanische Musiktheorie versuchte Aufschlüsse über gestalterische Verfahren zu gewinnen, in denen sich der Prozeßcharakter von Musikwerken manifestiert. Anhand von vier bedeutenden Veröffentlichungen aus der Zeit zwischen 1950 und 1997 wird dies exemplarisch dargestellt. Es handelt sich um Bücher von von Rudolph Réti (The Thematic Process in Music), Wallace Berry (Structural Functions in Music), Jonathan D. Kramer (The Time of Music) und Christopher Hasty (Meter as Rhyhtm).

Musikalische Sinnlichkeit und musikalische Praxis bauen auf Formen von Reflexion und Theorie auf, und wenn diese noch so unentwickelt oder auch unbewußt sind. Das hörende, analysierende, interpretierende Subjekt ist zugleich informationsverarbeitend und informationsschaffend. Jede Analyse ist gleichzeitig auch eine bewußtseinsgeleitete, bisweilen sogar unbewußte Konstruktion ihres Gegenstandes. Die Mechaniken der Intentionalität des Hörens und des Verstehens und des Interpretierens von Musik sind unvermeidbar; man muß sie begreifen, um aktiv eingreifend mit ihnen umgehen zu können. Partikular-Kategorien haben erst im Kontext ihren Sinn, und: nie verabsolutiert. Methoden haben sich als heuristische am Gegenstand zu bewähren, und wenn sie ihm nicht genügen, muß auf sie verzichtet werden, nicht auf ihren Gegenstand. In jeder musikalischen Figur, in jeder Klangfolge ist Geschichte gespeichert, und damit auch begrifflich vermittelte Bedeutung, derer sich die Komponierenden bedienten und die sie individualisierten. Die ›Anstrengung des Begriffs‹ ist in jeder Kunstwissenschaft und für jede Beschäftigung mit Kunst, auch für die spielerische und die unterhaltsame, unverzichtbar.

Exploiting the ubiquity of cell phones for quantitative chemical sensing imposes strong demands on interfacing devices. They should be autonomous, disposable, and integrate all necessary calibration and actuation elements. In addition, a single design should couple universally to a variety of cell phones, and operate in their default configuration. Here, we demonstrate such a concept and its implementation as a quantitative glucose meter that integrates finger pumps, unidirectional valves, calibration references, and focusing optics on a disposable device configured for universal video acquisition. The first example in the literature of a compound showing anisochronous 15N atoms resulting from diastereotopicity is described. Racemic 1,3-dimethyl-2-phenyloctahydro-1 H-benzimidazole was prepared and studied by 1H, 13C and 15N NMR spectroscopy. If convenient conditions were used (monitored by theoretical calculations of 2 JN-H spin-spin coupling constants), two 15N NMR signals were observed and corresponded to the diastereotopic atoms. GIAO/density-functional calculations of chemical shifts were not only in good agreement with the experimental values but also served as prediction tools. Non-invasive separation-free protocols are attractive for analyzing complex mixtures. To increase selectivity, an analysis under kinetic control, through exploitation of the photochemical reactivity of labeling contrast agents, is described. The simple protocol is applied in optical fluorescence microscopy, where autofluorescence, light scattering, as well as spectral crowding presents limitations. Introduced herein is OPIOM (out-of-phase imaging after optical modulation), which exploits the rich kinetic signature of a photoswitching fluorescent probe to increase selectively and quantitatively its contrast. Filtering the specific contribution of the probe only requires phase-sensitive detection upon matching the photoswitching dynamics of the probe and the intensity and frequency of a modulated monochromatic light excitation. After in vitro validation, we applied OPIOM for selective imaging in mammalian cells and zebrafish, thus opening attractive perspectives for multiplexed observations in biological samples. Coated blade spray (CBS) is a technology based on solid-phase microextraction (SPME) that has been designed for the quick extraction/cleanup of analytes from complex matrices and direct desorption/ionization under ambient mass spectrometry conditions. The detection and structural characterization of the components of a mixture is a challenging task. Therefore, the development of a facile and general method that enables both the separation and the structural characterization of the components is desired. Diffusion-ordered NMR spectroscopy (DOSY) with the aid of a matrix is a promising tool for this purpose. However, because the currently existing matrices only separate limited components, the application of the DOSY technique is restricted. Herein we introduce a new versatile matrix, poly(dimethylsiloxane), which can fully separate many mixtures of different structural types by liquid-state NMR spectroscopy.With poly(dimethylsiloxane), liquid-state chromatographic NMR spectroscopy could become a general approach for the structural elucidation of mixtures of compounds. Nuclear magnetic resonance (NMR) relaxometry and diffusometry are important tools for the characterization of heterogeneous materials and porous media, with applications including medical imaging, food characterization and oil-well logging. These methods can be extremely effective in applications where high-resolution NMR is either unnecessary, impractical, or both, as is the case in the emerging field of portable chemical characterization. Here, we present a proof-of-concept experiment demonstrating the use of high-sensitivity optical magnetometers as detectors for ultra-low-field NMR relaxation and diffusion measurements. We experimentally monitored, at the single-molecule level, the competition among reverse transcription, exponential amplification (RT-LAMP), and linear degradation (restriction enzymes) starting with hepatitis C viral RNA molecules. We found significant heterogeneity in the rate of single-molecule amplification; introduction of the restriction enzymes affected both the rate and the "fate" (the binary outcome) of single-molecule amplification. While end-point digital measurements were primarily sensitive to changes in fate, the bulk real-time kinetic measurements were dominated by the rate of amplification of the earliest molecules, and were not sensitive to fate of the rest of the molecules. We show how this competition of reactions can be used for rapid HCV genotyping with either digital or bulk readout. This work advances our understanding of single-molecule dynamics in reaction networks and may help bring genotyping capabilities out of clinical labs and into limited-resource settings. Quantitative measurement of marker expression in diseased cells is still a topic of considerable interest and different methodologies are currently under intense scrutiny. This work aims at developing an in vitro diagnostic method based on the release of paramagnetic species from relaxometrically 'silent' liposomes operated by the action of a phospholipase A2 (PLA2) previously targeted to the epitope of interest. The released paramagnetic species causes an increase of the longitudinal water proton relaxation rate proportional to the number of PLA2 bound to the cell outer surface. The sensitivity of the herein proposed method, named R-ELISA, was attempted in the detection of folate receptor expression on human ovarian cancer cells by functionalizing PLA2 with folic acid. Receptor/cell number of 8.3×105 has been measured on IGROV-1 cells. The R-ELISA assay can detect nanomolar cell suspension receptor concentrations and has been validated by well-established spectrofluorimetric procedures. [ABSTRACT FROM AUTHOR]

Jüngste Fortschritte auf dem Gebiet der DNA ‐ Assemblierung und der Affinitätsbindung haben zu aufregenden neuen Entwicklungen bei Nanosensoren und ultraempfindlichen Nachweismethoden für spezifische Proteine geführt.1 – 6 Diese Sensoren und Nachweissysteme haben drei attraktive Eigenschaften gemeinsam:1, 4, 7 1) Der Proteinnachweis gelingt mittelbar durch die Detektion amplifizierter DNA, wodurch die Empfindlichkeit drastisch erhöht wird. 2) Die Zusammenlagerung der DNA wird durch Affinitätsbindung von zwei oder mehr “Sonden” zu einem einzelnen Analytmolekül ausgelöst, was eine erhöhte Spezifität bedingt. 3) Der Nachweis erfolgt in Lösung ohne die Notwendigkeit einer vorherigen Abtrennung, was die Methodik für patientennahe Anwendungen attraktiv erscheinen lässt. Wir legen in diesem Kurzaufsatz das Prinzip dar, nach dem die Zusammenlagerung von DNA ‐ Molekülen mittels Affinitätsbindung erfolgt. Dabei stellen wir neuartige Anwendungen für den Nachweis von Proteinen beispielhaft heraus. Die Konzentration macht den Unterschied: Die Bindung eines Analytmoleküls an zwei Affinitätsliganden bringt zwei komplementäre Oligonucleotide in unmittelbare Nähe, wodurch deren lokale Konzentrationen drastisch gesteigert und so die Stabilität der Hybridmoleküle erhöht wird. The article discusses a study which achieved a selective detection of individual vapors and their mixtures using a single sensing material and its localized surface plasmon resonance (LSPR) spectroscopic readout. The study used a 3D network of dispersed plasmonic nanoparticles functionalized with a soft vapor-sorbing ligand shell and coupled its LSPR response with multivariate spectral analysis.
